抗体
本発明は、特異的部位で結合できる抗体、このような抗体の部位特異的結合方
法、特異的部位で結合している抗体、および治療と診断におけるこのような抗体
の使用に関する。
抗体は、疾患を誘発する外来性物質に対する哺乳動物の免疫応答の重要な成分
である。抗体は、多くの方法のいずれかを用いて生体外で製造され、そのような
抗体、特にモノクローナル抗体やその断片は、診断および治療用物質として有用
であることがわかった。抗体の有用性は、そのユニークな抗原特異性、すなわち
病原性抗原または腫瘍関連抗原のような独立した化学的分子を認識しこれに結合
する能力に由来する。その有用性の別の側面は、その多様性、すなわち非常に多
種類の独立した遺伝的に規定された抗体(モノクローナル抗体)を作成する哺乳
動物の能力(および、現在はファージディスプレイ(phage display)のような
過程)である。抗体の有用性の最後の側面は、その「定常」領域を介して相互作
用する能力である。この最後の側面は、他のセットの性質、例えばエフェクター
細胞、補体または他の結合分子(例えばプロテインA)との相互作用のようなイ
ソタイプに共通の性質を決定する。
抗体または断片の有用性は、成分単独では自然には存在しない性質を結合体(
conjugate)に持ち込む1以上の追加の分子的残基(molecular moieties)(以後
、物質と呼ぶ)を、直接または間接に化学的に結合することによって増強される
。そのような性質には、色素や放射性核種のようなレポーター機能、酵素的機能
、第2の結合機能(例えば、ビオチン−アビジン)、薬物(例えば、アドリアマ
イシン)、細胞毒性機能(例えば、リシン)、キレート物質、または広範な分子
的残基のいずれかと抗体が反応できるようにする化学的結合残基(chemical link
age moiety)がある。すなわち、抗体結合体は、抗体と、該抗体に直接または間
接に抗体に結合した活性物質(物質)を含んでなる。例えば、活性物質が放射性
核種の時、放射性核種は抗体に直接結合するか、または例えばTMTのようなキ
レート物質または例えばブロモアセチルのようなさらなるタンパク反応性基
(架橋物質)により抗体に結合するキレート物質を介して、抗体に間接的に結合
してもよい。同様に、活性物質は、酵素−プロドラッグ治療に使用するための分
子的キメラでもよく、そのようなキメラは、癌細胞のような哺乳動物細胞中で活
性化される転写制御DNA配列、転写制御DNA配列に機能的に結合しており、
投与されるプロドラッグの癌細胞に毒性のある物質への変換を触媒することがで
きる異種酵素をコードするDNA配列を含んでなる。この分子的キメラは、抗体
に直接結合しているが、またはウイルスベクターまたはリポソーム内に含有され
、そのウイルスベクターまたはリポソームが次に抗体に結合してもよい(例えば
、ヨーロッパ特許出願第90309430.8号を参照)。
抗体やその断片への分子の化学的付加または架橋について多くの方法が記載さ
れており、そのような反応は一般に結合反応(conjutgation reaction)と呼ば
れる。結合反応は、抗体中に天然に存在する化学的官能基を利用する。例えば、
共通のアプローチの1つは、一級アミン(主に、リジン残基のεアミノ基)を標
的にする。このアプローチの他の共通の例は、チロシン残基のヨード化(例えば
、125I)である。このアプローチの欠点は、ランダムであることである。すな
わち、リジンまたはチロシン残基は、分子構造の全体に存在するため、これらの
残基が伝達を補助すべき天然の性質(例えば、抗原認識、補体反応性またはエフ
ェクター細胞相互作用)が犠牲になることがある。結合体の挙動は、個別のユニ
ークな成分(そのいくつかは、全く無効であるかまたは有害である)の挙動の平
均である。このランダムなアプローチは、抗原認識に影響することも知られてい
る。
部位特異的結合(すなわち、抗体構造内の特異的アミノ酸残基への結合)は、
得られる結合体は異なる生成物の混合物ではないという利点を導く。抗体が結合
体に導入する性質(例えば、抗原反応性または薬物動態安定性)は、明確な化学
的構造に帰因する。さらに、抗原結合性を有する領域から空間的に離れているよ
うな部位を選択できる。例えば、抗体の可変領域には抗原結合部位を含まれ、重
鎖のCH2ドメインにはエフェクター(FcR)および補体(C1q)相互作用
残基が含まれることが知られている。従って、重鎖のCH3ドメインへの付加を
選択することにより、これらの機能を犠牲にすることを避けることができる。
部位特異的結合方法の2つの例が記載されている。抗体の糖鎖部分(carbohyd
rate portion)に対する抗体の結合が詳述されている(O'Shannessy and Quarles
,J.Immunol.Met 99:153-161,1987)。ヒトのIgG抗体は、CH2ドメイン
のasn297に1つの共通のグリコシル化部位を有する。(なお、本明細書に
記載のアミノ酸はすべて、EU指数に従って番号が付けられていることに留意さ
れたい。Kabatら、「免疫学的興味のあるタンパクの配列」(Sequences of prot
eins of immunological interest)、第5版、NIH publication No.91-3242
,1991。共通のイソタイプを、G1、G2、G4などと呼ぶ)。グリコシル化結
合方法は、抗原またはエフェクター相互作用に必要となるかも知れない糖鎖を変
化させる危険性を有する(Lundら、Molec.Immunol 27: 1145,1990;Isaacsら
、J.Immunol.1-18: 3062,1992)。
抗体への部位特異的結合の他の例では、部位特異的突然変異誘発による抗体上
の遊離チオールの形成が関与する。抗体は天然のシステイン残基を有し、そのチ
オール基はジスルフィド結合により結合している。天然に存在するシステイン残
基の位置は種間で高度に保存されており、これらの残基が、抗体の構造と機能に
とって必須であることを示している。抗体は、遊離のスルフヒドリル基を天然に
は含有しない。そのチオール基が酸化されないか、または天然のスルフヒドリル
結合が犠牲にならないシステインを含有するように、抗体を作成することは理論
的に魅力的である。システイン残基のスルフヒドリル基は、マレイミド、ハロゲ
ン化アルキルおよびアリール、α−ハロアシルおよびピリジルジスルフィドのよ
うなスルフヒドリルに特異的な、多くの結合化学による結合のために使用するこ
とができる。しかし、天然にはないシステイン残基を有するように設計されてい
る変種モノクローナル抗体は、結合のために利用できる遊離のチオール基を事実
上有するということはない。(抗体は2つの同じ重鎖と2つの同じ軽鎖から構成
され、遺伝子の作成方法および発現方法に依存して、1つの鎖の残基の遺伝子的
置換により、完全なH2−L2構造に2つの新しい残基をできることがあるため
;さらに、2つの残基は、その空間的配置と近傍の状況のため化学的に同一では
ないため)。作成されたチオールの1つの既知の例では(Bodmerら、米国特許第
5,219,996号、およびLyonsら、Protein Eng.3: 703,1990)、抗体構
造内の別個の「凹型」分子ポケット(直径0.13〜0.5mmの小分子のみが近
づくことができ、同じかまたは異なる抗体鎖内にジスルフィド結合を形成するこ
とができないポケット)にシステイン残基が導入された時のみ、利用可能な遊離
のチオールが観察された。モノクローナル抗体の「凸型の」または「平面的な」
表面に導入されたシステイン残基は、遊離のチオールを有さないことがわかった
。従って当業者は、表面システイン残基は、同じおよび/または別の抗体鎖内に
ジスルフィド結合を形成し、そのため抗体の巨大分子構造や機能をゆがめると結
論できるであろう。
公表されている文献は、作成した表面チオールは抗体の構造と機能を変化させ
ることを示している。例えば、ser444→cys変種の抗体産生細胞の粗上
清中にモノマーとダイマー(IgG−IgG)の両方とも観察された(Shopes,
J.Immunol.148: 2918,1992)。これに関連して、ser444→cys変種
を利用して、ダイマー抗体の産生方法が記載された(Caronら、J.Exp.Med.17
6: 1191-1195,1992)。これらの例の両方のダイマーIgGが、インビトロでエ
フェクター機能が増強していることがわかった。これらの実験および前述のBdom
erらの実験において、ダイマーが操作した部位で形成したか、またはどの程度正
常なジスルフィド結合が修飾されているかを示す、生理学的証拠は提供されてい
ない。最後に、ser119→cys変種を作成することによる「つながれた」
抗体が記載されている(Shopes,Mol.Immunol.30: 603,1993)。この変種は
、ダイマーおよび鎖間構造変種すなわち「つながれた」抗体を生成したと報告さ
れている。構造異常物の混合物は、規定された化学的物質を得るという目的を犠
牲にする。
従って、抗体の表面に露出したシステイン残基を含有するモノクローナル抗体
(特に、ser442→cys重鎖変種を含有するもの)が、部位特異的結合を
受けることを見いだしたことは、驚きであり予想外であった。
従って本発明は、システイン残基が物質に結合できるように、抗体の表面に露
出しているシステイン残基を含んでなるモノクローナル抗体を提供する。ここで
、該抗体は免疫化学的機能を有し、また免疫化学的機能の用語は、主に抗体の結
合能力を意味するが、エフェクター機能(もし存在するなら)をも包含する。抗
体の可変領域が抗原結合部位を含有することを考えると、結合の好適な部位は、
抗原結合に関与しない可変領域の表面上の表面残基(例えばsSv重鎖−軽鎖リ
ンカーペプチド)、並びに軽鎖の定常領域、重鎖のCH1、CH2、およびCH
3ドメインを包含する抗体の定常領域の表面残基であり、ヒンジ領域も含まれる
。すなわち、抗原結合に関与しない抗体表面のすべての残基、特にCDRの一部
でないものが、物質への結合に適している。抗体がエフェクター機能を有する場
合は、結合の好適な部位は、エフェクター機能を含むことが知られているCH2
ドメイン以外は同じである。システイン残基は実質的に還元型である。より好ま
しくは、還元システイン残基は、重鎖のCH3ドメインにあり、好ましくはCH
3ドメイン内の442位にある。還元システイン残基の別の好適な位置は、重鎖
−軽鎖リンカーペプチドである。新規のcys442抗体は、その産生細胞によ
り、モノマーIgG型と凝集型の両方を示すように発現されることが可能である
。凝集の存在は、BodmerらとShopesの研究で観察されたように、cys422変
種が表面変種であることを示唆するが、驚くべきことにほとんどの抗体は凝集型
ではなかった。モノマーIgGは、例えばゲル濾過クロマトグラフィーにより容
易に精製可能であり、モノマー型は長期間の保存でも安定であった。モノマーI
gGは、遊離のチオールを有さないことがわかった(表1)。理論に拘泥するつ
もりはないが、チオールはまず、天然に存在する付加物(例えば、グルタチオン
)により阻止(すなわち、保護)されると我々は考えている。
我々は、加工されたチオール(engineered thiol)は、天然のジスルフィド結合
を還元しない制御された条件下で還元されることを発見した。例えば、天然のジ
スルフィド結合を還元することができない低濃度の還元剤のような穏やかな条件
が、加工されたチオールの還元に適していることがわかっている。還元抗体は、
pH8で長期間保存された時でもモノマー型を維持する。(pHと酸素をうまく
操作することによりチオールは反応性になることは公知である)。すなわち、制
御された還元により、加工された抗体は、特に加工されたチオールで特異的に部
位特異的化学的付加が可能になるようにできる。
従って本発明はまた、抗体の表面に露出したシステイン残基を含んでなるモノ
クローナル抗体であって、制御された還元により物質への部位特異的化学的結合
が可能になり、システイン残基は抗体の免疫化学的機能を妨害しない部位に導入
される抗体に向けられている。
本発明の抗体は、好ましくはモノクローナル抗体またはその断片であり、抗体
という用語は、抗体と抗体断片の両方を包含する。本発明の抗体は任意の種から
得られる。抗体は、可変領域が1つの抗体から得られ、定常領域は別の抗体(例
えば、ヒトの抗体)から得られるキメラ抗体でもよい。すなわち、キメラ抗体は
、種/種キメラまたはクラス/クラスキメラでもよい。このようなキメラ抗体は
、抗原結合性を改善するために、またはエフェクター機能を改善するために、1
つまたはそれ以上のさらなる修飾を受けても良い。改変抗体の別の型は、定常領
域とCDR以外の可変領域がヒトのフレームワークに移される複合抗体を包含す
るヒト化抗体である。結合能を回復するために必要であれば、フレームワークま
たはこのような抗体の定常領域内の追加のアミノ酸を修飾してもよい。すなわち
、本発明で使用される抗体は、アミノ酸配列が天然に存在するものではない任意
の改変抗体を包含する。しかし、CDRを移植された抗体が最も好ましい。本発
明の抗体は、例えばG1、G2、G4のような異なるイソタイプを含む。抗体の
例は、J.Cell Biol 125(2)437-446,1994年4月、およびProc.Natl.Acad.Sc
i.87,3542-3546,1990年5月、に記載の40KD抗体(CO/17.1.A)
であり、好ましくはヒト化抗−40KD抗体であり、特にG4イソタイプのヒト
化抗−40KDである。抗−40KD抗体の具体例は、323/A3であり、好
ましくはヒト化323/A3であり、特に好ましくは323/A3 IgG4で
ある。
抗体の別の例は、A.Tomasettiら、ヨーロッパ生化学会連合(Federation of
European Biochemical Societies)第317巻、143-146、1993年2月、に開示
されている抗−葉酸受容体抗体であり、好ましくはヒト化抗−葉酸抗体および特
にG1イソタイプのヒト化抗−葉酸抗体である。抗−葉酸抗体の具体例は、MO
V18であり、好ましくはヒト化MOV18 IgG1である。抗体のさらなる
例には、抗−CEA、抗−ムチン(mucin)、抗−20/200KD、抗−ガン
グリオシド、抗−ジゴキシン、抗−CD4、抗−CD23、抗−CDw52、お
よびより具体的には、ヒト化抗−CDw52抗体であるCampath−1Hが
ある。Campath−1H(登録商標)のCDRを含む抗体鎖DNA配列は、
EPO328404に開示されており、その開示内容は参考のため本明細書に引
用される。(Page,M.J.,and Sydernham,M.A.,チャイニーズハムスター卵巣
細胞でのヒト化モノクローナル抗体Campath−1Hの高レベル発現、Biot
ech.9: 64-68,1991)。
本発明で使用される抗体断片には、Fab、F(ab)2、Fv、および組換えDNA
技術で得られるような合成ペプチド配列からなる断片がある。
本発明で使用されるモノクローナル抗体は、当該分野で公知の任意の方法で調
製されるか、またはさらに詳しくはGB9022547.5に記載のものがある
。精製は、EP−A−91917891に記載のように行われる。
断片は、文献、例えば「抗体、実験室マニュアル(Antibodies,a laboratory
manual)」、E.Harlow and D.Lane編、コールドスプリングハーバーラボラト
リー(Cold Spring Harbor Laboratory)、1988に記載のような任意の方法、ま
たは分子遺伝学的方法により調製される。
本発明はまた、システインが直接または間接的に物質に結合している抗体を提
供する。物質が抗体に間接的に結合している時、それは、例えばキレート物質の
ような1つまたはそれ以上の結合残基を介して、抗体に結合される。1つまたは
それ以上の結合残基を介して抗体に結合されるそのような物質は、「2官能性物
質」として知られている。そのような結合残基は、例えば、抗体のようなタンパ
ク内のチオール官能基に共有結合することができるマレイミドまたはブロモアセ
チルのような官能性化学的残基でもよい。結合残基はまた、物質と抗体結合残基
の間の橋として機能する化学的スペーサー(例えば、p−ベンジル基)を利用し
てもよい。
結合物質の例には、色素、放射性核種、酵素、薬物、細胞毒、およびビオチン
/アビジンがある。薬物や細胞毒の具体的な例は、それぞれアドリアマイシンお
よびリシンである。キレート物質の具体的な例には、以下のものが含まれる:

物質が放射性核種の場合、これは直接または間接的な方法で抗体の遊離のチオ
ールに結合される。例えば、Schawartz法(Schawartz,A.,and Steinstrasser
, A.,J.Nucl.Med.18:721,1987)に類似の方法を修飾して、99mTcを抗
体に結合させる(ここでは、天然のジスルフィドを還元する必要はないであろう
)。金属性放射性核種(90Y、186Re、177Lu、111Inおよび67Cu)は、
間接的プレラベル法により結合される(ここで放射性核種をまず2官能基キレー
ト物質に付加し、次に抗体に結合させる)か、または間接的ポストラベル法によ
り結
合させる(ここではあらかじめ形成したキレート物質−抗体結合体に放射性核種
を結合させる)。本発明のキレート物質は、キレート物質を抗体のチオール基に
結合させることができる任意のヘテロ−またはホモ−2官能性架橋リンカー(す
なわち、化学的スペーサー)(M.McCallら、Bioconjutate Chem.1,222-236,
1990)、またはPierce「免疫技術のカタログとハンドブック」(″Immuno Techn
ology Catalog and Handbook″)、1992、E8〜E39頁およびParker、Chem.Soc.
Rev.[1990],19,271-291に記載の架橋物質とそこに記載の方法を用いて、抗
体に結合させてもよい。
架橋リンカーと組合せるキレート物質の例としては、以下のものがある:
・ブロモアセチル−DOTA(2−[p−(ブロモアセトアミド)ベンジル−
1,4,7,10−テトラアザシクロードデカン−N,N',N'',N'''−四酢
酸);
・ブロモアセチル−TRITA(2−[p−(ブロモアセトアミド)ベンジル
]−1,4,7,10−テトラアザシクロトリデカン−N,N',N'',N'''−
四酢酸);
・ブロモアセチル−TMT(2−[p−(ブロモアセトアミド)ベンジル]−
6,6''−ビス[N,N'',N''−テトラ(カルボキシメチル)アミノメチル)

ルピリジン)。
2官能性キレート物質架橋リンカーの組合せであるブロモアセチル−DOTA
(BADとも呼ぶ)は、M.J.McCall,H.Diril.and C.F.Meares,Bioconjuga
te Chem.1:222-226,1990)に記載されている。
さらに詳しくは、本発明で使用されるキレート物質は、DOTAまたはTMT
であり、タンパク反応性基(架橋リンカー)は、ブロモアセチルまたはマレイミ
ドであり、最も好ましくはブロモアセチルである。
本発明で使用されるキレート物質は、任意の既知の方法により調製される(例
えば、M.McCallら、前述)。
本発明において使用される放射性核種には、インビボの放射免疫療法および/
または標的細胞または標的組織のイメージングに適したものがある。放射免疫療
法のためには、細胞性DNAを破壊するために高線量のエネルギーを標的部位に
与える必要がある。αおよびβ放射性放射性核種は、適当なエネルギー範囲の放
射線を出す。しかし、α−放射性物質は、短命であるか、または崩壊して有害な
娘産物になる。従って放射免疫療法に最適の放射性核種は、普通β−放射性物質
である。イメージングのためには、放射線は、体組織との相互作用ができるだけ
少なく、外部から検出できるように強いシグナルを出す必要がある。従って、イ
メージングにはガンマ放射線放出性放射性核種が最も適している。イメージング
と放射免疫療法の両方のために、放射性核種は、投与と標的部位への結合の間の
経過時間後に活性を有し検出するのに適当な半減期を有していなければならない
。放射能標識抗体は、血流中から内皮孔を介して標的の細胞外液に移動する必要
がある。大きな抗体または抗体/キレート物質複合体は、拡散が遅く、放射性核
種の半減期は数時間〜数日が好ましい。本発明の特定の側面において、放射性核
種は、195mPt、57Ni、57Co、105Ag、68Cu、52Mn、52Fe、111I
n、113mIn、99mTC、67Ga、169Yb、166Tm、167Tm、146Gd、157D
y、95mNb、103Ru、97Ru、99Ru、101mRh、201Tl、203Hg、197H
g、203Pb、99Rh、48Cr、57Co、125I、131I、35S、153Sm、88Y、90
Y、186Re、188Re、211At、212Bi、212Pbおよび177Luよりなる群
から選択される。
本発明のより具体的な例では、放射性核種は、111In、67Cu、186Re、18 8
Re、177Lu、99mTc、131I、88Y、90Y、211At、212Bi、212Pb、57
Co、153Sm、88Y90Yおよび177Luよりなる群から選択される。
本発明のより具体的な例では、放射性核種は、177Lu、153Sm、90Yおよび111
Inである。本発明はまた、直接または間接に放射性核種に結合した本発明
の抗体を含んでなる放射標識抗体、特にDOTAまたはTMTを介して90Yまた
は177Luで標識されたキレート物質−抗体結合体を提供する。本発明はまた、
特異的部位で結合されることができる抗体の産生方法、および本発明の抗体の部
位特異的結合方法を提供する。
本発明の別の側面において、治療と診断における本発明の結合した抗体の使用
が提供される。特に、色素、放射性核種酵素、薬物および細胞毒を用いて検出で
きるか、或いは治療できる症状の診断および/または治療のための、本発明の抗
体の使用を提供する。これらの抗体複合体は、リンパ腫や白血病、特に小細胞肺
癌や非小細胞肺癌、前立腺癌ならびに卵巣癌のような癌の治療に有用である。本
発明の最も好適な側面において、癌や関連する転移のイメージングおよび/また
は治療に使用される本発明の抗体複合体を提供する。これらはまた、免疫抑制物
質として使用され、および特に重症の血管炎、リウマチ様関節炎、全身性ループ
ス、また多発性硬化症、移植片対宿主疾患、乾癬、若年性糖尿病、シェーグレン
症候群、甲状腺疾患、重症筋無力症、移植拒絶および喘息などの自己免疫疾患な
どのT細胞介在性疾患の治療に、使用される。
本発明はまた、前記のいずれかの疾患の治療またはイメージングのための薬剤
の製造における前記結合した抗体の使用を提供する。
本発明の別の側面によれば、本発明の結合した抗体複合体を用いた治療および
診断ができる症状の治療法であって、治療的有効量の抗体複合体をそのような治
療の必要な哺乳動物に投与することを具備した方法を提供する。特に、リンパ腫
や白血病、特に小細胞肺癌や非小細胞肺癌、前立腺癌ならびに卵巣癌のような癌
の治療法、および特に癌や関連疾患の治療法が提供される。これらはまた、重症
の血管炎、リウマチ様関節炎、全身性ループス、また多発性硬化症、移植片対宿
主疾患、乾癬、若年性糖尿病、シェーグレン症候群、甲状腺疾患、重症筋無力症
、移植拒絶および喘息などの自己免疫疾患などのT細胞介在性疾患の治療方法に
使用される。
また本発明において、結合したモノクローナル抗体またはその断片および1つ
またはそれ以上の薬剤学的に許容される賦形剤とからなる、本発明の結合した抗
体を含有する薬剤学的に許容される組成物が提供される。
このような組成物は、結合した抗体以外に、おそらくは他の抗体もしくは抗生
物質のような他の物質との混合物である、生理学的に許容される希釈物または担
体を含む。適当な担体には、リン酸緩衝化生理食塩水、リン酸緩衝化生理食塩水
グルコースおよび緩衝化生理食塩水などがあるが、これらに限定されない。投与
経路は、通常静脈内、筋肉内、皮下、および腹腔内注射または送達などの非経口
投与である。
放射免疫療法の点から、本発明の放射性核種に結合した抗体を含有する組成物
の投与量は、治療される症状および治療の受容者により異なるが、成人患者では
約1〜100mgの範囲であり、好ましくは1〜10mg、最も好ましくは5mgが、
通常注入物として投与される。1日10mg投与した数週間後または数ケ月後に第
2回目の繰り返し投与が好ましい。
放射免疫療法の点から、このような組成物の投与量は、治療される症状および
治療の受容者により異なるが、成人患者では1〜約100mgの範囲であり、好ま
しくは1〜10mg、最も好ましくは5mgである。

すなわち、本発明の結合したモノクローナル抗体が、容器中で通常凍結乾燥型で
、単独または目的の型の細胞に特異的な追加の抗体とともに提供される。色素、
放射性核種、酵素、薬物、細胞毒、キレート物質およびビオチン/アビジンに結
合した抗体は、緩衝液(例えば、トリス、リン酸塩、炭酸塩など)、安定剤、殺
菌剤、不活性タンパク(例えば、血清アルブミン)などおよび使用説明書のセッ

-label(ICN))含有)中で、37℃で48〜72時間インキュベートした。遠
心分離して上清を採取し、透析し、濃縮(セントリコン(Centricon)、アミコ
ン(Amicon)、ビバリー、マサチューセッツ州)した。抗体をプロテインAクロ
マトグラフィー(HPLCダイナマックスハイドロポア−プロテインA(HPLCDy
namax Hydropore-protein A)ミニカラム[ライニン(Rainin)、ウォボーン(W
oburn)、マサチューセッツ州])で精製し、1M酢酸で溶出し、限外ろ過(例
えば、PM30膜(アミコン(Amicon)))し、さらにHPLCゲル濾過(例え
ば、S−5200Aジオール、YMC、ウィルミントン、ノースカロライナ州)

4℃で一晩行なった。細胞を遠心分離し、冷結合緩衝液で3回洗浄し、放射能を
測定した。点は、3重測定の平均±標準誤差(棒)を示す。2つの抗体は、35S
でin situで生合成的に同じ比活性(0.3mCi/nmole)まで標識し、前記
のように精製した(35S−リガンド)。平衡競合は平行して行い、ここで各リガ

実施例1.cys置換による変種モノクローナル抗体の産生
(a) Campath−1Hと抗−ジゴキシン変種。従来法の分子遺伝学的
方法により、種々のイソタイプの重鎖にcys442を導入した。例えば、ヒト
IgG2、IgG4、Campath−1Hおよび抗−ジゴキシン(いずれもI
gG1)の遺伝的作製体は、ウェルカムラボラトリーズ(Wellcome Laboratorie
s)内で得た。CH3領域の末端部分を、制限酵素Nsil(5’)およびEc
oRI(3’)で切断し、各切断部位で連結したアニーリングした2本鎖オリゴ
ヌクレオチドで置換した。抗原の特異性は、可変領域とCH1領域の各置換によ
り、定常領域に導入した。Pinkら、Biochem.J.117:33-47,1970が最初に報告
したG4配列に一致するようにするため、ser228→proをIgG4に導
入した。G4mcと呼ぶIgG4 cys442変種を、CH2領域中のこれら
の残基を変化させることによりさらに修飾した:leu235→ala、gly
237→ala、およびglu318→ala。これらの後者の変化は、このよ
うな変化がFcガンマ受容体と補体C1qとの抗体の相互作用を低下させるよう

(b) 323/A3変種。マウス抗体323/A3は、腺腫の治療の同定に
有用なヒト上皮組織上のエピトープと反応する(Edwardsら、Cancer Res.46: 1
306-1317,1986)。323/A3の可変領域内の相補性決定領域をまず「ヒト化
」し、ヒトIgG1イソタイプに移植した。cys変種を調製するために、cD
NA発現作製体を、Campath−1H IgG4 cys442変種の定常
領域をコードするcDNAにフレーム内に連結した。ヒト化323/A3 Ig
G4 cys442変種を、NSO細胞中で発現し、常法に従って精製した。
(c) 323/A3 sFv断片。ヒト化した323/A3の単一鎖のsF
v断片を、通常のPCR法とクローニング法により作製した。cys残基置換を
リンカー領域に導入して、cys変種作製体を作成した。例えば、通常のリンカ
ー領域(gly4ser 3つの繰り返し)を、部位特異的突然変異誘発により
gly4sergly2cys2sergly4serを含有するように修飾し
た。変種を大腸菌(E.coli)中で発現し、親和性クロマトグラフィーにより精
製した。
(d) Mov−18変種。Mov−18は、主に卵巣癌組織上で発現される
葉酸結合タンパクと反応する(Miottiら、Intl.J.Cancer 39,297-303,1987
))。ヒトIgG1イソタイプcDNAを、逆転写酵素を用いて公共の供給源の
mRNAライブラリーからクローン化した。Mov−18の可変領域をヒト化し
、ヒトG1定常領域に連結させた。cys442を部位特異的突然変異誘発によ
り、重鎖cDNAに導入した。ヒト化Mov−18 IgG1 cys442を
NSO細胞中で発現させ、常法に従って精製した。
実施例2.変種モノクローナル抗体の還元
抗体溶液を、脱気した緩衝液(例えば、100mMリン酸ナトリウムまたはリン
酸トリメチルアンモニウム、pH>8.0、好ましくはpH8.0〜8.5、濃
度は例えば100μM(15mg/ml))中で調製した。ある量の還元剤を抗体溶
液と混合して、所望の程度の還元を達成する。例えば、Reduce-Imm(登録商標)
(ピアス(Perce))のような固相還元剤の50%ゲルスラリー5部を、抗体溶
液中に撹拌して入れる。Reduce-Imm(登録商標)ゲルの還元能力は、製造業者の
情報から30μgモル/ml充填ゲルであると仮定した。50%スラリーの250μ
lを、100μM抗体溶液1mlに添加して、30倍以上過剰の還元剤を有する混
合物を作成した(完全剤モル数/抗体モル数)。抗体−還元剤混合物を、室温で
1時間攪拌し、遠心分離し、溶液に、実施例3に記載のようにチオール測定、p
H低下、精製または結合のような追加の操作を行なった。
あるいは抗体タンパクを、メルカプトエチルアミンのような可溶性還元剤で還
元した。例えばタンパクを、0.1Mリン酸ナトリウム(pH6.0)、5mME
DTA中で200〜300μMに濃縮した。メルカプトエチルアミンを、最終的
にタンパク濃度の10倍過剰になるように加え、室温で1時間静かに攪拌した。
次に還元タンパクを還元剤から分離し、ゲル濾過のような従来法により結合のた
めに調製した。タンパク溶液は通常、0.1Mリン酸テトラメチルアンモニウム
(pH8.2)、25μM DTPA中で150×gで2分間プレ平衡化させた
Bio Spin 30 カラム(バイオラッドラボラトリーズ(Bio-Rad Laboratories))
でゲル濾過した。
実施例3.還元変種抗体の部位特異的結合
還元ser442→cys変種。Campath−1H G4mcを、2−[
p−(ブロモアセトアミド)ベンジル]−1,4,7,10−テトラアザシクロ
ドデカン−N,N',N'',N'''−四酢酸(ブロモアセチル−DOTA)に結合
させた。ブロモアセチル−DOTAは、2官能性残基であり、第一の側面は57C
oであらかじめ標識されたキレート物質であり、第2の側面は遊離のスルフヒド
リルに共有結合できる反応性基である。2官能性キレート物質とそれを57Coで
標識する方法は、既に記載されている(Mearsら、Analytical Biochem.142: 68
-78,1984)。簡単に説明すると、ブロモアセチルDOTAの溶液を、57Coで
トレース標識し、次に10倍モル過剰を抗体溶液に加えた。結合反応は、37℃で
2時間行い、製造業者の薦める方法に従ってBio Spin 30 カラム(バイオラッド
(Bio-Rad))でゲル濾過により、反応物を分離して反応を停止させた。2官能
性キレート物質(例えば、ブロモアセチルTMT)はまた、放射標識の前に使用
した。結合反応の温度と時間は、最大の部位特異的結合が得られるように変化さ
せた。例えば、リン酸テトラメチルアンモニウム(pH8.2)、25μM D
TPA中150〜250μMの濃度の還元タンパクを、金属を含まない反応バイ
アル中で10倍過剰のブロモアセチルTMTに加えた。反応を室温で24時間行
い、結合体は前記のようにゲル濾過で単離した。
実施例4.結合体のマウス生体分布
還元ser442→cys変種であるCampath−1H G4mcを、プ
レ標識することなく、前述のように部位特異的方法によりブロモアセチルDOT
Aに結合させた。比較のために、Lym−1について既に記載されている(この
方法は、McCallら、Bioconjugate Chem.1: 222-226,1990の方法に従い、2−
イミノチアランを用いて、リジン残基のεアミノ基にチオール基をランダムに導
入する)ように非還元Campath−1H G4mcにチオール基を導入して
、ランダム結合体を調製した。イミノチオール化したCampath−1H G
4mcを、部位特異的結合体と同程度(抗体当たりのキレート物質)にブロモア
セチル−DOTAに結合させた。プラスチック容器中の90YCl3を以下の試薬
と混合して、結合体を放射性にした:8倍量のモノクローナル抗体結合体(>2
5mg/ml、0.1M酢酸アンモニウム、pH6.7)、2倍量の90Y、および最
終濃度10μMの非放射性イットリウム。まず、非放射性イットリウムを加え、
次に90Yと結合体を加えた。キレート化は、室温で90分間行なった。Camp
ath−1H抗原を発現する皮下の癌を有するマウス(CHO−10/D4)に
、10μg(2μCi)を静脈内注射した。所定の時間にマウスを屠殺し、組織を
切り出し、重量を測定し、放射能を測定した。結果を下記表IIに示し、組織1g

実施例5
還元ser442→cys変種であるCampath−1HをTMTに結合さ
せ、これを図4に示すように重鎖中のcys442残基に置換基を介して共有結
合させた。反応を制御して、抗体1分子当たり平均1〜2キレート物質を含有す
る結合体を産生させてもよい。結合体を、金属のない条件下でゲル濾過により未
反応の2官能性キレート物質を含まないように精製すると、緩衝化した金属のな
い環境で安定である。
免疫結合体キレート化.入手できる最高の金属のない試薬を用いて、金属のな
いプラスチック中で結合体を放射標識した。担体を含まない90YCl3を、デュ
ポン/ニューイングランドヌクレア(Dupont/New England Nuclear)、アマーシ
ャム(Amersham)および他の供給源から購入した。比活性は典型的には、10〜
30μlの0.05N HCl中5mCi、比活性5.6×105Ci/gであった。使
用前に、90YCl3を0.1部の6M酢酸アンモニウムでpHを約5.8に緩衝
化した。キレート化は、以下の試薬を連続的に加えて、室温で最高90分間イン

Biochem.142: 68-78,1984に記載のように薄層クロマトグラフィーにより追跡
した。90%を超える放射性金属がルーチンにキレート化された。(キレート化

に結合した90Yの画分はHPLCゲル濾過、薄層クロマトグラフィーまたはSD
