【発明の詳細な説明】
フレーバー付けられた製品及びその製法技術分野及び公知技術
本発明は、フレーバー、及び特に、より充実したかつより濃厚な特性を与える
ために所望される製品のフレーバー付けのそれ、即ち、一般に脂肪、特にバター
及びチーズを含有する製品に関連している型の官能特性、及び香辛性で塩気(”
風味のある”)型の官能特性に関する。
この目的と共に、本発明は、フレーバー付け成分としての次のものの使用に関
する:
a)疎水性アミノ酸残基1個及び少なくとも1個の酸性アミノ酸残基を有するト
リペプチド1種又は数種及び/又は
b)N−ラクトイル−X[ここで、Xはアミノ酸残基を表わす]型の構造を有す
るアミノ酸誘導体1種又は数種。
ここで、”疎水性アミノ酸残基”とは、疎水性であると一般に認められる慣用
のアミノ酸、つまり、アラニン(Ala)、メチオニン(Met)、バリン(V
al)、ロイシン(Leu)、プロリン(Pro)、フェニルアラニン(Phe
)、チロシン(Tyr)、イソロイシン(Ile)及びトリプトファン(Trp
)の1種の残基のことである。
同様に、ここで、”酸性アミノ酸残基”とは、グルタミン酸(Glu)からの
残基又はアスパラギン酸(Asp)からの残基のことである。
したがって、本発明により使用されるトリペプチドは、前記の疎水性残基の1
個及びGlu及びAspからの少なくとも1個の酸性残基から形成される。第3
のアミノ酸残基は再び酸性残基であってよいか又は、親水性アミノ酸、つまり、
システイン(Cys)、グリシン(Gly)、グルタミン(Glu)、セリン(
Ser)、トレオニン(Thr)及びアスパラギン(Asn)の1種、又は塩基
性アミノ酸、つまり、リシン(Lys)、アルギニン(Arg)、オルニチン(
Orn)及びヒスチジン(His)の1種の残基から選択することができる。
前記の”アミノ酸残基”という用語は、当業者に慣用の意味で、それは、トリ
ペプチド鎖に相当するアミノ酸によって与えられる揃いの原子に関することであ
る。今後、これらの残基は、ポリペプチド鎖の構造を定義するために慣用されて
いる、前記の括弧で示された3文字の略語(例えば、Eur.J.Bioch.1984、138、9
〜37参照)によって表わす。
本発明により、N−ラクトイル−X[ここで、Xはアミノ酸残基を表わす]型
の構造を有するアミノ酸誘導体は、トリペプチドの代わりに、又はそれに付加的
に、フレーバー付け成分として使用され得る。従って、本発明は、N−ラクトイ
ル又は2−ヒドロキシプロパノイル基(今後これを、略語”Lac”で示す)、
及び前記の慣用のアミノ酸の1個の残基を含有する化合物に関係する。従って、
及び明確な例を得るために、これらの本発明による誘導体の1つは、誘導体(S
)−Lac−Metであり、その構造は次のとおりである:
前記から明確であるように、不斉炭素原子が本発明による化合物の構造中に存
在するので、これらは、個々に、又は異性体の混合物として、本発明の目的のた
めに満足して全て使用され得る数種の立体配置異性体の形で生じ得る。それらの
立体配置は、本発明によるトリペプチド及びラクトイル誘導体を製造するために
使用される出発アミノ酸のそれによっても典型的に左右される。
実際に、前記のトリペプチド及びアミノ酸誘導体は、それらが彼ら自身の味を
実際に欠けているという事実にも拘らず、それらが添加される製品に極めて高く
評価される官能特性、とりわけ、全く著しい丸味、クリーム性及び実質性を与え
ることができる、極めて有用なフレーバー付け成分であることを見いだした。こ
の故に、これらは添加される製品の舌ざわり又は”口当たり”を改善することが
でき、これらのトリペプチド及びアミノ酸誘導体が、”低−脂肪”又は”低−カ
ロリー”消費製品(これは脂肪が少ない製品)にフレーバー付けをしようとする
フレーバー付け組成物の製造のために使用される場合に、又はそれらが直接これ
らの製品に添加される場合に、その結果は特に有用である。特に、それらが共に
配合される製品に特別な味を与えることができる揮発性物質を基礎とする組成物
から基本的に成る、他の慣用のフレーバー付け物質と組み合わせた場合に、それ
らはこれらの揮発性物質の官能効果を増すばかりでなく、牛乳から得られる天然
濃縮物及び同様の製品を使用して頻繁に製造される、慣用のバター又はチーズ型
の食料品及びフレーバー付け組成物中の脂肪の存在によって与えられる型の充実
性も与え得ることが判明した。
また、本発明によるトリペプチド及びアミノ酸誘導体は、”うまみ”型のその
効果で周知の、及び例えばオリエンタル型の調理で広く使用されるフレーバー付
け成分、グルタミン酸1ナトリウム(MSG)を使用して得られ得る型の官能効
果を生じさせることもできることが認められた。それらは、実際に、それらが添
加され、かつ少なくとも部分的にMSGに代わる製品中のうまみ特性を与える又
は強化することに貢献することができ、それらの効果は、後者よりもストックら
しさが少なく、かつより塩分が少なく、従って、若干の型の適用にはより有用で
ある。
ところで、この結果は、これより以前の文献に照らして全く予期外である。実
際に、若干のトリペプチドは、例えばチーズ、醤油又は再び魚加水分解物の天然
組成物であると知られているという事実にも拘らず、それらは大抵、疎水性基を
有しないか、2又は3個含有するトリペプチドであり、その場合には、それらの
味は有用な使用を見いだすには苦すぎる。更に、これらの天然製品中に広く確認
されるトリペプチドの官能効果は、しばしば、それらのトリペプチドが官能的に
重要であると分かった場合ですら、MSGのそれよりもずっと少なく感じている
。
従って、我々が知るかぎりでは、M.Nogushi et al.、J.Agr.Food Chem.、1975
、23、49だけが、魚加水分解物中に、単一の疎水性基、つまり、Glu−Asp
−Val、(Glu、Ile)−Asp、Glu−Gly−Ala及びIle−
Glu−Gluを含有するトリペプチドを確認した。しかしながら、これらの著
者によって述べられた、本質的に酸性である他のトリペプチドは、MSGのそれ
に似ているが、さほど強くない味を有すると記載されており、疎水性基を有する
前記のトリペプチドは、重要ではないと感じられ、かつ苦い又は単調な味を有す
ると記載された。更に、酸性トリペプチドの立場と異なり、フレーバー付け目的
の
ために使用するための後者の可能な重要性に関する提案はなされなかった。
更に、疎水性基1個及び少なくとも1個の酸性基を有する数種のトリペプチド
は、以前に蛋白質合成の研究の場合に引用されたが、知るかぎりでは、そのよう
なトリペプチドは、消費製品に又は再びそれらの口当たりを改善する、又はそれ
らの官能的側面を変えるための慣用の他のフレーバー付け成分を含有するフレー
バー付け組成物に添加されるべきであるということは提案されなかった。
Lac−X型のアミノ酸誘導体に関する限り、我々の知識では、Lac−Ph
e、Lac−Ile、Lac−Ala、Lac−Val及びLac−Glyだけ
が文献に引用されており、かつこれはその研究に関しては全くフレーバーの分野
の外である。従って、以前に引用されたそれらの化合物の官能特性の記載はない
。本発明の説明
従って、本発明の目的は、前記のようなトリペプチド及び/又はアミノ酸誘導
体をフレーバー付け成分として使用する食料品及び他の食用製品のフレーバー付
け法である。
このように、本発明による有利な実施態様によれば、前記のようなトリペプチ
ド1種又は数種及び/又はアミノ酸誘導体1種又は数種を、人間又は動物によっ
て食べられる製品に、又は製薬学的製品に、前記の製品の口当たりを与える、改
善する又は増すために充分な量で添加する。
本発明の他の有利な実施態様によれば、フレーバー付け剤として使用されるト
リペプチドは、2つの酸性基を有する。後記の例から判るように、1つの疎水性
基及び2つの酸性基を有するそれらのトリペプチド、及びより特別に、酸性基が
グルタミル基であるそれらは、全く著しい官能特性を有するフレーバー付け組成
物及びフレーバー付けされた製品を与え、かつその丸味性、クリーム性及び一般
に彼らが消費される際に口中に引き起す感覚を増加させる。
更に、官能的観点から特に有用な結果は、使用されるトリペプチドが、Glu
−Glu−Leu、γGlu−Glu−Leu又はGlu(Glu−Leu)、
Leu−Glu−Glu、Glu−Asp−Phe、Glu−Glu−Ile、
Asp−Glu−Leu及びGlu−Leu−Gluから選択される際に得られ
たということも確認された。特に、Glu−Glu−Leuは、特にこのペプチ
ドが、MSGが典型的に使用されるそれらに比較してモル量で使用される場合に
、MSGの特徴である型のフレーバー特性を与えることに極めて効果的であると
実証された。
一般に、本発明によるトリペプチドは、MSGのそれに強さで比較可能なイン
パクトを有し、一方、あま
り著しくない塩気ストックフレーバーを与えることが判明した。
本発明により使用され得るアミノ酸誘導体について、最良の結果はLac−G
lu、Lac−Ala及びLac−Met、特に後者で得られるということが判
った。
前記のトリペプチド及び/又はアミノ酸誘導体を含有する、本発明による使用
から得られるフレーバー付けされた製品も、本発明の1目的物である。それらは
、前記のトリペプチド及び/又はLac−X型の誘導体が直接添加された食料品
及び飲料のような完成製品の形であってよい。本発明による化合物は、それ自体
が前記の完成製品中に配合されるように意図されるフレーバー付け組成物の製造
のために使用することもできる。これらのフレーバー付け組成物は典型的に、そ
の効果が、慣用の溶剤又は添加剤と同様に、特に、一般的使用のマイクロカプセ
ル封入剤中で、その粘稠度又は口当たりよりはむしろ食料品の味に影響しうる他
のフレーバー付け成分を含有する。本発明によるフレーバー付け化合物は、世界
知的所有権機構(WO)第95/34222号明細書及び世界知的所有権機構(
WO)第95/10927号明細書(その内容は参照に包含される)に記載され
た型のフレーバー付け組成物と組み合わせて使用することもできる。
こうして得られるフレーバー付け組成物は、人間及
び動物によって食べられ得る極めて多様な製品の口当たりを改善又は増加するた
めに使用することができる。低脂肪マーガリン及びバター、同様に、ヨーグルト
、アイスクリーム、チーズ、牛乳及び牛乳を基礎とする製品又は飲料を含む牛乳
誘導物、更に、即席食品、マヨネーズ、スープ、ソース、甘味及び風味ビスケッ
ト、チョコレート、甘味物及びカラメル、スナック、特にカリカリした物、デザ
ート及び飲み物、特にトロピカルフルーツ型の物、又は再び動物の飼料について
の例として参照され得る。
当然、及び先に記載したように、そのようにフレーバー付けされた製品は、本
発明によるトリペプチド及び/又はアミノ酸誘導体を直接それらの製品に添加す
ることによって得ることができる。
一般に、全ての低脂肪食料製品は、これらのトリペプチド又はアミノ酸誘導体
、又はそれらを含有するフレーバー付け組成物を使用して、粘稠度及び口当たり
の観点から改善することができる。更に、後記の例によって示されているように
、本発明によるペプチド及びアミノ酸誘導体は、既に前記のクリーミイ型効果及
び口当たりと平行して、又はそれとは無関係で、それらが配合される組成物又は
食料品のフレーバー特徴を変えることもできる。従って、本発明による化合物は
、風味型、例えば、香辛料入りの、塩気のある、及び刺激の強い食料の全ての適
用に有用であると認められ
た。従って、これらは、特に、調理済み食品又はスナック及び他の塩味食料をフ
レーバー付けるために、かつこの型の食料品のために意図されたフレーバー及び
フレーバー付け組成物の製造のために好適である。
本発明によるフレーバー付け成分が、低脂肪製品のフレーバー付けのために特
に有用であるという事実にも拘らず、それらは、それらがより充実した、かつク
リーミーな粘稠度を与える普通の脱脂−還元食料品にフレーバー付けするために
、又は脂肪及び/又はMSGの存在と関連して官能特性を強調するためにも使用
され得ることは言うまでもない。これは、特に、肉型、チーズ又はバター及びマ
ーガリン、前記のトリペプチド及びLac−X型の誘導体が意図する目的のため
に特に有用かつ効果的であると実証された製品の製造の場合に当てはまる。
本発明によるトリペプチド及びアミノ酸誘導体が、前記の様々な製品に添加さ
れ得る濃度は、それらの製品の特性及び得たいと所望されるフレーバー付け効果
に依存する。従って、それらは極めて広い範囲で変化し得る。例えば、これらの
化合物が、それ自体、食料品及び飲料中に配合されるために意図されるフレーバ
ー付け組成物の製造のために使用される場合には、これらは典型的に、組成物の
重量に対して、0.01〜1重量%の割合で、又は丁度10重量%まで、それら
組成物に添加される。本発明による化合物が、フレー
バー付けされるべき食料品及び飲料に直接添加される場合には、それらは、食料
品又は飲料の重量に対して、0.001〜0.5重量%の割合で、又は丁度1重
量%で使用されうる。勿論、前記の下限又は上限濃度が一定の適用において実際
の実験を通じて適切であると実証し得るので、これらの値は、例として挙げられ
得るにすぎない。また、当業者はそれらの特性に関連してそれらを選択すること
ができる。
本発明によるフレーバー付け組成物が、特に、前記のような2種又はそれ以上
のトリペプチドの混合物、前記のようなLac−X型の2種又はそれ以上の誘導
体の混合物、又は再び、それらのトリペプチドの1種又は数種とそれらの誘導体
の1種又は数種との混合物を含有することが言及されなければならない。実際に
、これらの混合物が、特に濃厚なかつ均一な官能効果を得ることを可能にさせる
ということが判明した。更に、これらの化合物が特に良好にアミノ酸と結合し、
フレーバー付け組成物に充実性を与える、後者の官能効果を補強し、本発明によ
るトリペプチド及びLac−X構造の化合物は付加的に充実性と豊満性を与える
ことも判明した。
従って、本発明は、食料品上で脂肪又はMSGのような成分の存在する典型的
な官能特性を改善又は与えることができるフレーバー付け法及び特にフレーバー
付け組成物を与える。特に、本発明により得られるこ
れらの食料品及び飲料のクリーミーな粘稠度及びより充実した口当たりにおける
改善が特徴である。これらは、本発明によりフレーバー付けされなかった相応す
る食料品よりも、もっとインパクトのある、口中でより持続する味を、口当たり
の増加と共に有する。本発明を実施する方法
前記のトリペプチド及びアミノ酸誘導体は、慣用反応を利用する方法を用いて
商業的に得られる製品から製造することができる。これらの反応は、後記のよう
に、本質的に化学型であるか、又は適当な基質から出発する酵素型であってよい
。
化学型の方法のうち、例えば、PyBOP(登録商標)(ベンゾトリアゾリル
オキシ−トリス[ピロリジノ]−ホスホニウムヘキサフルオロホスフェート;J.
Coste et al.、Tetrahedron Lett.1990、31、205及び1991、47、259参照)のよ
うな通例のペプチドカップリング試薬を用いる、溶液中でのペプチド合成の慣用
法が行なわれた。下の図式は、トリペプチドLeu−Glu−Gluの特別な場
合に適用される、本発明によるトリペプチドを製造するために使用される合成の
ための計略を説明している:第I図式
この方法におけるトリペプチドの製造には、4段階、2カップリング段階及び
2脱保護段階を包含する。食料品をこれらの製品が意図される使用に供する場合
には、それらを、いかなる有機副産物も、いかなる塩も共に完全に含有しない、
純粋な状態で得ることが必要であり、それというのも後者はペプチドの味及び官
能効果を変え得るからである。この要求は、最終脱保護反応としての水添分解の
選択を指定した。
第I図式に説明されているように、2つの好適なL立体配置のアミノ酸は、好
適な溶剤中の、PyBOP(登録商標)の存在で、一緒にカップリングされる。
こうして得られる保護されたジペプチドは、トリフル
オロ酢酸(TFA)を用いるアシドリシスによって脱保護され、かつ次いで、第
3のN−保護アミノ酸とカップリングして保護されたトリペプチドを生成させる
。ジクロロメタン、エタノール及び水の混合物から成る溶剤中での水添分解によ
る後者の脱保護により、所望のトリペプチドを純粋な状態で生じる。一般的方法
A.カップリング反応
ジクロロメタン(2ml/ミリモル)中のN−保護アミノ酸(1当量)、アミ
ノエステル又はペプチドエステル塩(1当量)及びPyBOP(登録商標)(1
当量)の溶液又は懸濁液を、ジイソプロピルエチルアミン(DIEA)と共に、
1〜2時間、室温で撹拌しながら処理した。反応を、薄層クロマトグラフィー(
TLC)によって追跡した。混合物を酢酸エチル(ジクロロメタンの容量の50
倍)中に注ぎ、かつ得られた溶液を次のように処理した:有機相を5%KHSO
4、5%NaHCO3の水溶液及び食塩水で洗浄し、Na2SO4上で乾燥させ、
濾過し、かつ溶剤を真空下で蒸発させた。
こうして得られた生成物を、適当な溶離剤を用いてシリカゲル上での”フラッ
シュ”クロマトグラフィーによって精製した。
保護された出発アミノ酸を、Novabiochem、Bachem又はPropeptideから購入し
、かつそのまま使用した。P
yBOP(登録商標)(給源:Nova)を、ジクロロメタン中に溶かし、ミリポア
水で洗浄し、Na2SO4上で乾燥させ、かつ使用する前に真空下で濃縮によって
取り出した。
B.中間的脱保護
トリフルオル酢酸(TFA)及びジクロロメタンの1:1混合物中のN−t−
ブチルオキシカルボニル−ペプチジルエステルの溶液(5ml/ミリモル)を、
20分間、室温で撹拌した。その溶液を真空下で蒸発させ、痕跡量の残留トリフ
ルオロ酢酸を、高真空下でジエチルエーテル中で、共−蒸発によって蒸発させた
。この方法で得られたペプチドエステル塩を、そのまま、Aに記載の方法におけ
る第2カップリング反応で使用した。
C.最終的脱保護
ジクロロメタン/エタノール/水(10:12:4)混合物中のN−ベンジル
オキシカルボニル−保護ペプチド及び活性炭素上の10%Pd(保護基の10重
量%)の懸濁液(26ml/ミリモル)を、室温で4日間、水素気圧下で撹拌し
た。Celite(登録商標)上でパラジウムを濾別した後に、有機溶剤を真空下で濃
縮させ、ガスを含まない湧き水(50mm/ミリモル)を加えた。水溶液を酢酸
エチルで2回、及びエーテルで2回洗浄した。有機溶剤を真空下で蒸発除去し、
水溶液を凍結乾燥させた。
前記の方法により、一般に60%以上の総収率を有する所望のトリペプチドを
得ることができた。
全てのペプチドを、陽子及び13C NMR(核磁気共鳴)により、COSY
及びHMQC型の相関を用いて分析した。また、これらを、MS(質量分析法)
により、化学的イオン化技法(DCI−MS、NH3)又は電気噴霧技法(ES
I−MS/MS)を用いて、及びHPLC(高性能液体クロマトグラフィー)に
よって分析した。分光分析のための条件
HPLC
クロマトグラムは、Spectra Physics又はBeckman装置中で、30℃で、214
nmUV検出器、Nucleosil(登録商標)RP−18.5μm、100Å、25
cm×0.40cmカラムを用いて得られた;溶剤A:水中の0.1%TFA;
溶剤B:水/アセトニトリル3:7混合物中の0.1%TFA;Bに関する勾配
:0%(0〜5分)、64%まで線形増加(5〜40分);流れ1ml/分。
NMR
Bruker AMX-360及びARX-400を用いて得たスペクトル。
DCI−MS
Finnigan MAT-4500装置を使用して、イオン化ガスとしてのアンモニア及び7
0eVフィラメントを用い
て得たスペクトル。
ESI−MS/MS
550〜1650m/z範囲での検量のためのミオグロビンを用いる、VG bio
tech BIO-Q又はFinnigan TSQ 700型の分光器;アリコート10μlを、給源中に
直接注入し、かつ溶離を、アセトニトリル、0.1%水性TFA、メトキシエタ
ノール及びイソプロパノール均等量(equivalent volumes)を使用して、7μl(
VG Viotech)の流れで、又は0.1%酢酸を含有する水/メタノール3:1混
合物を用い、10μl/分(Finnigan)で行なった。
次のトリペプチドをこの方法で製造した:

Lac−X構造のアミノ酸誘導体を、トリペプチドのために前記したそれと同
様の方法で製造し、かつこの場合には、PyBOP(登録商標)1当量及びジイ
ソプロピルエチルアミン過剰量の存在で、好適な保護アミノ酸と(+)−(S)
乳酸(Fluka)との間の単一
カップリング反応が要求された。中間保護されたペプチドエステルを精製し、か
つ保護基をCに記載のように除去した後に、所望のN−(S)−ラクトイル−X
(ここでXはアミノ酸残基である)を単離した。
次の中間体及び最終生成物を製造した:
13.N-(S)-ラクトイル-Glu
中間体ジベンジルエステル、つまりN−(S)−ラクトイル−Glu(OBz
l)−OBzlの分析特性は次のようであった:

この化合物の合成は、他の類似物のそれとはやや異なった。氷浴を使用して2
5℃を保ちながら、ジクロロメタン及びジイソプロピルエチルアミン中で、Py
BOP(登録商標)の存在で、L−メチオニンメチルエステル塩酸塩(Aldrich)
と、(+)−(S)−乳酸とをカップリング反応させた後に、混合物を酢酸エチ
ル中で希釈し、かつAで記載したように処理した。真空蒸発後の残査を、酢酸エ
チルを使用してシリカゲル上で濾過し、トリピロリジノホスフィンオキサイド(
PyBOP(登録商標)反応の副産物)を除去した。濾液を濃縮させ、溶離剤と
して、ヘキサン/酢酸エチル35:65、次いでヘキサン/酢酸エチル3:7及
び最後に酢酸エチルを使用して、シリカゲル上でフラッシュクロマトグラフィー
によって精製した。クロマトグラフィーによる純粋なフラクション(TLC)を
集め、濃縮させて、次の分析特性を有する中間体メチルエステル、つまり、N(
S)−ラクトイル−Met
−OCH3を得た:
次いで、中間体メチルエステル(10ミリモル)を、水(20ml)及びエタ
ノール(10ml)の混合物中の水酸化ナトリウム(21ミリモル)を使用して
鹸化した。エタノールを真空下で蒸発させ、水相を水(100ml)で希釈し、
エーテルで2回抽出した。水相のpHを5.5に調整し、かつ凍結乾燥させた。
Dowex(登録商標)(50Wx8(Fluka)樹脂上で脱塩させた後に、所望のN−(S)−
ラクトイル−Metが粘性の黄色油状物の形で得られた。所望の生成物を結晶性
粉末の形で得るために、後者のpHを0.2N Na O Hで5.5に調整し、
分析特性は次のようであった:
ところで、本発明を次の実施例により詳説するが、
例中、略語は当業者に慣用のものである。これらの例は、本発明を実施する典型
的な方法を表わすが、これは特に、挙げられた成分の相対的又は絶対的割合に関
して、限定的に解されるものではない。例1 フレーバー付け組成物の製造
