Description
Claims (1)
- 【特許請求の範囲】 1.骨髄始原細胞およびリンパ系始原細胞について富化される造血細胞を含有す る組成物を得る方法であって: ヒト造血細胞の混合物を、リンパ系細胞、骨髄細胞、および樹状突起細胞に分 化し得るが、実質的に赤芽球細胞には分化しない細胞を含有する富化画分に分離 する工程を包含する、方法。 2.リンパ系始原細胞について富化される造血細胞を含有する組成物を得る方法 であって: ヒト造血細胞の混合物を、リンパ系細胞に分化し得るが、実質的に骨髄細胞お よび赤芽球細胞には分化しない細胞を含有する富化画分に分離する工程を包含す る、方法。 3.樹状突起細胞について富化される造血細胞を含有する組成物を得る方法であ って: ヒト造血細胞の混合物を、樹状突起細胞に分化し得るが、実質的に骨髄細胞お よび赤芽球細胞には分化しない細胞を含有する富化画分に分離する工程を包含す る、方法。 4.骨髄始原細胞およびリンパ系始原細胞について富化される造血細胞を含有す る組成物を得る方法であって: ヒト造血細胞の混合物を、細胞表面マーカーCD34およびCD45RAを発現するが、 CD19を発現しない細胞を含有する富化画分に分離し、CD34+CD45RA+細胞を得る工 程を包含する、方法。 5.請求項4に記載の方法であって、 前記細胞を、細胞表面マーカーCD10を発現しない富化画分に分離し、CD34+CD4 5RA+CD10-細胞を得る工程をさらに包含する、方法。 6.樹状突起細胞前駆体について富化される造血細胞を含有する組成物を得る方 法であって: ヒト造血細胞の混合物を、細胞表面マーカーCD34、CD45RA、およびCD10を発現 する細胞を含有する富化画分に分離し、CD34+CD45RA+CD10+細胞を得る工程を包 含する、方法。 7.前記富化細胞が、細胞表面マーカーCD19を発現しない、請求項6に記載の方 法。 8.ヒト造血骨髄始原細胞およびヒト造血リンパ系始原細胞について富化される 造血細胞を含有する組成物であって、該細胞が、リンパ系細胞および骨髄細胞に 分化し得るが、実質的に赤芽球細胞には分化しない、組成物。 9.ヒト造血リンパ系始原細胞およびヒト造血樹状突起始原細胞について富化さ れる造血細胞を含有する組成物であって、該細胞が、リンパ系細胞および樹状突 起細胞に分化し得るが、実質的に赤芽球細胞および骨髄細胞には分化しない、組 成物。 10.ヒト造血樹状突起始原細胞について富化される造血細胞を含有する組成物 であって、該細胞が、樹状突起細胞に分化し得るが、実質的に赤芽球細胞および 骨髄細胞には分化しない、組成物。 11.細胞表面マーカーCD34およびCD45RAを発現するが、CD19を発現しないヒト 造血細胞について富化されるヒト造血細胞の組成物。 12.請求項11に記載の組成物であって、前記富化細胞が細胞表面マーカーCD 10を発現しない、組成物。 13.細胞表面マーカーCD34、CD45RA、およびCD10を発現するヒト造血細胞につ いて富化されるヒト造血細胞の組成物。 14.請求項13に記載の組成物であって、前記富化細胞が、細胞表面マーカー CD19を発現しない、組成物。 15.骨髄始原細胞および/またはリンパ系始原細胞の増殖および/または分化 に関与する生物学的反応修飾物質を得る方法であって、 細胞表面マーカーCD34およびCD45RAを発現するヒト造血細胞の増殖および/ま たは分化に対する効果についてサンプルを試験する工程を包含する、方法。 16.樹状突起始原細胞の増殖および/または分化に関与する生物学的反応修飾 物質を得る方法であって、 細胞表面マーカーCD34、CD45RAおよびCD10を発現するヒト造血細胞の増殖およ び/または分化に対する効果についてサンプルを試験する工程を包含する、方法 。 17.リンパ系始原細胞の増殖および/または分化に関与する生物学的反応修飾 物質を得る方法であって、 細胞表面マーカーCD34、CD45RA、およびCD10を発現するヒト造血細胞の増殖お よび/または分化に対する効果についてサンプルを試験する工程を包含する、方 法。 18.請求項17に記載の方法であって、前記富化細胞が細胞表面マーカーCD19 を発現しない、方法。 19.被験体のリンパ系細胞および骨髄細胞を一時的に再定住させる方法であっ て、ヒト造血骨髄始原細胞およびヒト造血リンパ系始原細胞について富化される 造血細胞を含有する治療有効量の組成物を被験体に投与する工程を包含し、該始 原細胞が、リンパ系細胞および骨髄細胞に分化し得るが、実質的に赤芽球細胞に は分化しない、方法。 20.請求項19に記載の方法であって、前記富化細胞が、細胞表面マーカーCD 34およびCD45RAを発現するが、CD19を発現しない、方法。 21.被験体のリンパ系細胞を一時的に再定住させる方法であって、ヒト造血リ ンパ系始原細胞について富化される造血細胞を含有する治療有効量の組成物を被 験体に投与する工程を包含し、該始原細胞が、リンパ系細胞に分化し得るが、実 質的に赤芽球細胞および骨髄細胞には分化しない、方法。 22.請求項21に記載の方法であって、前記富化細胞が、細胞表面マーカーCD 34、CD45RAおよびCD10を発現する、方法。 23.請求項22に記載の方法であって、前記富化細胞が、細胞表面マーカーCD 19を発現しない、方法。 24.被験体の樹状突起細胞を一時的に再定住させる方法であって、ヒト造血樹 状突起始原細胞について富化される造血細胞を含有する治療有効量の組成物を被 験体に投与する工程を包含し、該始原細胞が、樹状突起細胞に分化し得るが、実 質的に赤芽球細胞および骨髄細胞には分化しない、方法。 25.請求項24に記載の方法であって、前記富化細胞が、細胞表面マーカーCD 34、CD45RAおよびCD10を発現する、方法。 26.請求項25に記載の方法であって、前記富化細胞が、細胞表面マーカーCD 19を発現しない、方法。 27.請求項9または10に記載のヒト造血始原細胞集団を含有する組成物であ って、組換えDNA分子を含有する、組成物。 28.組換えDNA分子を含有する請求項9または10に記載のヒト造血始原細胞 集団を含有する組成物であって、該組換えDNA分子が抗原特異的領域からなるポ リペプチドをコードし、そして成熟細胞で発現される場合、シグナルを伝達し得 る、組成物。 29.請求項28に記載の組成物であって、前記組換えDNA分子がT細胞レセプ ターをコードする、組成物。 30.請求項29に記載の組成物であって、前記T細胞レセプターがαおよびβ 鎖を含む、組成物。 31.請求項28に記載の組成物であって、前記組換えDNA分子が、機能的なシ グナル伝達領域および抗体に由来する抗原結合部位を含有するキメラタンパク質 をコードする、組成物。
