9−シスレチノイドおよびその新規な中間体の合成方法
発明の技術分野
本発明は、9−シスレチノイド化合物の合成およびとりわけ9−シスレチノイ
ン酸およびその関連化合物ならびにその新規中間体の合成に関する。
発明の背景
細胞の増殖および分化を仲介するうえでの全トランスレチノイン酸、13−シ
スレチノイン酸および合成レチノイン酸類似体などのレチノイドの役割は、皮膚
疾患、例えば乾癬および座瘡の処置を制御するための薬理学的な有利性、ならび
に化学療法および化学的予防法などの腫瘍学的な応用において関心を呼び起こし
ている。レチノイドの活性の分子的な基礎の解明における有意な進歩は現在、改
善された治療指数を有するレチノイド化合物の設計の可能性を提供している。
現在までに、レチノイン酸の幾つかの受容体が同定された。これらの受容体は
、リガンド依存性転写因子(ligand dependent transcription factors)として機
能する細胞内受容体のスーパーファミリーの成員である。現在、これらの受容体
は2つのサブファミリー、レチノイン酸受容体(RARs)およびレチノイドX
受容体(RXRs)に分類されている。これらのサブファミリーの分類は主とし
て、アミノ酸構造における相違、様々な天然に存在するおよび合成のレチノイド
に対する応答性および異なる標的遺伝子の発現を調節する能力に基づく。各RA
RおよびRXRサブファミリーは3つの別個のイソ型(RARα,RARβおよ
びRARγ、およびRXRα、RXRβおよびRXRγと呼ぶ)を有する。複数
のレチノイド受容体の発見は、別個のサブファミリーおよびそのイソ型の機能的
な特性の問題を提起する。
近年、9−シスRAおよび9−10シスオレフィン立体化学を有する他のレチ
ノイドがRARsおよびRXRsに結合し、これらを介して遺伝子の発現を調節
することができることが認められた(ヘイマン(Heyman)ら、Cell、68:39
7(1992))。しかしながら、現在まで、当該技術分野はこのような化合物
の低収率、非選択的で高価な製造法しか提供しなかった。例えば、文献は9−
10シスオレフィンレチノイドを製造する非選択的な方法(例えば、ロブソンら
(Robson)ら、J.Am.Chem.Soc.、77:4111(1955)、マツイ(Mat
sui)ら、J.Viatminol.、4:178(1958)、ドイツ特許第DE1068
719号およびアウレル(Aurell)ら、Tetrahedron Lett.、31:5791(
1990)参照)、ならびに13−14オレフィン結合の立体選択的な調製方法
(例えば、パテンデン(Pattenden)ら、J.Chem.Soc.(C)、1984(19
68)およびメイヤー(Mayer)ら、Experientia、34:1105(1978)
参照)を提供した。
さらに、アセチレンエステルへの銅酸塩の共役付加(conjugate addition)を介
してアルキル置換レチノイドおよびトリメチルシリル置換レチノイドの調製方法
を提供しているが、該方法は過多な工程を使用しており、重要なことにはアリル
アルコールのアルデヒドへの酸化を使用し、それによって、得られたレチノイド
に存在する9−10シスオレフィン結合の完全性を破壊する(エルンスト(Erns
t)ら、J.Org.Chem.、52:398(1987))。
従って、シス配置の9−10オレフィン結合を有するレチノイド化合物を選択
的で低費用の仕方で調製するのが望ましい。
発明の概要
本発明は、9−シスレチノイド、好ましくは、9−シスレチノイン酸の選択的
で低費用の合成方法を提供する。とりわけ、本発明は9−シスレチノイドを製造
するための、
(a)塩基の存在下でシクロヘキセニルケトンを還元してシクロヘキサジエニン
を生成し、
(b)該シクロヘキサジエニンを親電子性ニトリル源と縮合してシクロヘキサジ
エニンニトリルを生成し、
(c)該シクロヘキサジエニンニトリルに1,4共役様式にてアルキル基を付加
してシクロヘキサトリエンニトリルを生成し、
(d)該シクロヘキサトリエンニトリルを対応シクロヘキサトリエンアルデヒド
に還元し、ついで
(e)ホスホネートまたはホスフィン塩への塩基の付加によって生成したカルボ
アニオン等価物で該シクロヘキサトリエンアルデヒドを伸長して、レチノイドエ
ステル立体異性体の混合物(ここで炭素9−10間のオレフィン結合はシス配置
であり、炭素11−12および13−14間のオレフィン結合はトランス配置ま
たはシス配置のいずれであってもよい)からなる反応生成物を生成することを特
徴とする方法を提供する。その後、該レチノイドエステルを加水分解してレチノ
イン酸立体異性体の混合物を生成してよく、該混合物を今度は分離して個々のレ
チノイン酸立体異性体、好ましくは9−シスレチノイン酸を提供することができ
る。
また、9−シスレチノイドを製造するための、
(a)塩基および親電子性ニトリル源の存在下でシクロヘキセニルケトンを還元
してシクロヘキサジエニンニトリルを生成し、
(b)1,4共役様式にてアルキル基を該シクロヘキサジエニンニトリルに付加
してシクロヘキサトリエンニトリルを生成し、
(c)該シクロヘキサトリエンニトリルを還元して対応シクロヘキサトリエンア
ルデヒドにし、ついで
(d)ホスホネートまたはホスフィン塩に塩基を付加することによって生成した
カルボアニオン等価物で該シクロヘキサトリエンアルデヒドを伸長して、レチノ
イドエステル立体異性体の混合物(ここで炭素9−10間のオレフィン結合はシ
ス配置であり、炭素11−12および13−14間のオレフィン結合はトランス
配置またはシス配置のいずれであってもよい)からなる反応生成物を生成するこ
とを特徴とする方法をも提供する。
さらに本発明は、塩基および親電子性ニトリル源の存在下でシクロヘキセニル
ケトンを還元してシクロヘキサジエニンニトリルを生成することを特徴とするシ
クロヘキサジエニンニトリルの製造方法をも提供する。
さらに、本発明は、
(a)シクロヘキサジエニンを親電子性ニトリル源で縮合してシクロヘキサジエ
ニンニトリルを生成し、ついで
(b)1,4共役様式にてアルキル基を該シクロヘキサジエニンに付加し、9−
10オレフィン結合がシス配置である9−シスアルキルシソイド(cisoid)ニトリ
ルを生成する
ことを特徴とする9−シスアルキルシソイドニトリルの製造方法を提供する。
さらにまた、本発明は上記方法に従って製造された9−シスレチノイドを提供
する。このような9−シスレチノイドは、9−シスレチノイン酸、9,11−ジ
シスレチノイン酸、9,13−ジシスレチノイン酸および9,11,13−トリシ
スレチノイン酸を包含するのが好ましい。これらおよび本発明を特徴付ける様々
な他の利点、新規な特徴は、本明細書に添付および一部を形成する請求の範囲に
特に指摘している。しかしながら、本発明の好ましい態様を例示し記した付随の
図面および記載事項を、本発明、その利点およびその使用によって得られたもの
のより良い理解のために参照するとよい。
発明の態様の詳細な説明
本発明の第一の態様に従い、本発明者らはレチノイドの合成方法を開発した。
この方法の一連の段階を以下に示す。
一連の上記プロセスにおいて、R1およびR2は各々独立に、水素または1〜5
の炭素原子を有する低級直鎖または分枝鎖アルキルを示してよく;
R3は水素または1〜6の炭素原子を有する低級分枝鎖または直鎖アルキル、
アルケンまたはアルキンを示してよく;
R4およびR5は各々独立に水素または1〜12の炭素原子を有する低級分枝鎖
または直鎖アルキル、アルケン、アリールまたはアラルキルを示してよく;
R6は1〜5の炭素原子を有する低級直鎖または分枝鎖アルキル、アルケンま
たはアルキンを示し;
R7は水素または製薬学的に許容し得る塩を示し;
構造式における炭素11−12および13−14間の破線はトランスまたはシ
ス配置のいずれであってもよいオレフィン結合を示す。
本明細書で使用する製薬学的に許容し得る塩には、ピリジン、アンモニウム、
ピペラジン、ジエチルアミン、ニコチンアミド、ギ酸、尿素、ナトリウム、カリ
ウム、カルシウム、マグネシウム、亜鉛、リチウム、メチルアミノ、トリエチル
アミノ、ジメチルアミノおよびトリス(ヒドロキシメチル)アミノメタンが含ま
れるが、これらに限られるわけではない。さらに他の製薬学的に許容し得る塩は
当業者によく知られている。
該方法の一連のプロセスは、リチウムジイソプロピルアミド(LDA)などの
塩基およびジエチルクロロホスフェートなどのクロロジホスフェートの存在下で
、β−イオノン(アルドリッチ・ケミカル(Aldrich Chemica1)、ミルウォーキ
ー(Milwaukee)、ウィスコンシンから入手可能)などのシクロヘキセニルケトン
(1)の還元から開始し、4−(1−[2,6,6−トリメチルシクロヘキセン−
1−イル])−(ブト−3−エン−1−イン)などのシクロヘキサジエニン(2
)を生成する。塩基としてのLDAに加えて、本発明の方法のこの段階において
有用な塩基の好ましい以下に限られるものではない他の例には、水素化ナトリウ
ム(NaH)、水素化カリウム(KH)、n−ブチルリチウム(n−BuLi)
、s−ブチルリチウム(s−BuLi)、t−ブチルリチウム(t−BuLi)
、ナトリウムアミド(NaNH2)およびリチウム、カリウムまたはナトリウム
ヘ
キサメチルジシルアジドが挙げられる。
第2段階は、塩基の存在下で親電子性ニトリル源(シアン酸フェニルまたは臭
化シアンなど)を利用した、5−(1−[2,6,6−トリメチル−1−シクロヘ
キセン−1−イル])−ペント−2−イン−4−エンニトリルなどのシクロヘキ
サジエニンニトリル(3)への該シクロヘキサジエニン(2)の縮合からなる。
この反応系列の第3段階は、1,4共役様式にてアルキル基をシクロヘキサジ
エニンニトリル(3)に付加し、シス配置の9−10二重結合を有する(2Z,
4E)−3−メチル−5−(1−[2,6,6−トリメチル−1−シクロヘキセン
−1−イル])−2,4−ペンタジエニトリルなどの本質的に純粋なシクロヘキサ
トリエンニトリル(4)を生成することに関する。この点において、該方法のこ
の態様に従ったアルキル基の付加は、当業者によく知られた多くの技術および方
法を使用してなされてよいが、好ましくは、該付加反応は非極性溶媒、最も好ま
しくは、ヘキサンまたはヘプタンにて行われる。
第4段階では、該シクロヘキサトリエンニトリル(4)を、ヘキサン中での標
準的なDIBAL(水素化アルミニウムジイソブチル)還元により、対応シクロ
ヘキサトリエンアルデヒド(5)((2Z,4E)−3−メチル−5−(1−[2
,6,6−トリメチル−1−シクロヘキセン−1−イル])−2,4−ペンタジエ
ンナール)に還元する。
この反応系列の第5段階は、主にエチル−3,7−ジメチル−9−(2,6,6
−トリメチル−1−シクロヘキセン−1−イル)−2−トランス−4−トランス
−6−シス−8−トランス−ノナテトラエノエートを含むレチノイン酸エステル
(レチノイドエステル)(6)の立体異性体の混合物を生成するため、ジエチル
−3−エトキシカルボニル−2−メチルプロプ−2−エニルホスホネートなどの
ホスホネートまたはホスフィン塩(例えば、トリフェニルホスフィン)に塩基(
例えばn−BuLi)を付加することによって形成されたカルボアニオン等価物
でシクロヘキサトリエンアルデヒド(5)を伸長することに関する。この点で、
該レチノイン酸エステル立体異性体(6)はトランス配置対シス配置にて13−
14オレフィン結合の15:1を越える優勢(prevalence)を示す。本発明の方法
のこ
の態様において有用である塩基の例には、以下に限られるものではないが、DM
PUの存在下で、水素化ナトリウム(NaH)、水素化カリウム(KH)、n−
ブチルリチウム(n−BuLi)、s−ブチルリチウム(s−BuLi)、t−
ブチルリチウム(t−BuLi)およびナトリウムアミド(NaNH2)を含む
。
該プロセスの第6で最終の段階は、レチノイン酸エステル(6)をメタノール
性水酸化物の水溶液(MeOH/KOH)で加水分解して、レチノイン酸異性体
(7)を得ることに関する。レチノイド異性体(7)の例には、制限はないが、
主要成分の3,7−ジメチル−9−(2,6,6−トリメチル−1−シクロヘキセ
ニル)−2−トランス−4−トランス−6−シス−8−トランス−ノナ−テトラ
エン酸(9シスレチノイン酸)および検出可能な量の3,7−ジメチル−9−(
2,6,6−トリメチル−1−シクロヘキセニル)−2−シス−4−トランス−6
−シス−8−トランス−ノナ−テトラエン酸(9,13ジシスレチノイン酸)お
よび3,7−ジメチル−9−(2,6,6−トリメチル−1−シクロヘキセニル)
−2−トランス−4−シス−6−シス−8−トランス−ノナ−テトラエン酸(9
,11ジシスレチノイン酸)が含まれる。加水分解後、所望のレチノイン酸異性
体(7)をメタノール性水酸化物水溶液から選択的に結晶化するかまたは、別の
やり方として、準分取逆相(ODS)高性能液体クロマトクラフィー(HPLC
)によって精製することができる。
本発明の範囲内において上記方法にある種の修飾をなしうることが当業者によ
り理解されるであろう。例えば、第1段階および第2段階(すなわち、シクロヘ
キサジエニン(2)の縮合)は、塩基の存在下での水の代わりに、塩基の存在下
での、リチウムアニオンおよびシアン化フェニルなどの親電子性ニトリル源で還
元シクロヘキセニルケトン(1)をクエンチする(quenching)ことにより、1段
階で1ポットの反応に組み合わせ、シクロヘキサジエニンニトリル(3)を直接
生成することができる。
さらに、本発明の合成は上記に示したレチノイドの合成以上の広範囲の応用が
できることを当業者は理解するであろう。例えば、段階2および3のそれぞれの
縮合および付加は、シス配置にて選択的に生成された9−10オレフィン結合を
有する様々なアルキルシソイドニトリルおよびその誘導体(例えば、アミン、酸
、アルデヒド、エステル)を形成するために使用されてよい。
上記の合成方法は、重要なことに費用を含め、現在利用できるものに比べて多
くの利点を提供する。この点において、β−シクロシトラール(該物質は、本発
明の方法において利用する出発物質より平均で少なくとも20〜40倍高額であ
る)などの現存の方法において利用される一層典型的な出発物質に対して本発明
の方法はβ−イオノンなどの市販の高価でなく、容易に利用できる出発物質を利
用する。加えて、該合成方法における第3段階(すなわち、1,4共役様式にて
アルキル基をシクロヘキサジエニンニトリル(3)に付加してシクロヘキサトリ
エンニトリル(4)を形成する)ではシス配置での9−10二重結合の選択的な
導入ならびにアルキル、アルケン、アリール、アラルキルなどの広範な種々の置
換基を炭素番号9にて導入することを可能にする。さらに、該方法を通じて使用
する試薬は標準的なもので、容易に入手することができ、危険性が報告されてい
ない高価ではない有機または無機化学物質である。
実施例1
本発明のレチノイドの合成は、以下の9−シスレチノイン酸合成スキームによ
り例示される。
4−(1−[2,6,6−トリメチルシクロヘキセン
−1−イル])−(ブト−3−エン−1−イン)(2):
該中間体4−(1−[2,6,6−トリメチルシクロヘキセン−1−イル])−(
ブト−3−エン−1−イン)(2)をβ−イオノン(1)(アルドリッチ・ケミ
カルズ社から利用できる)から以前に記載された方法に従って調製した(ネギシ
(E.I.Negishi)ら、J. Org. Chem.、45:2526(1980)(その開
示内容を参照のため本明細書に包含する))。
5−(2,6,6−トリメチル−1−シクロヘキセン−
1−イル)−ペント−2−イン−4−エンニトリル(3):
100mL容の炎で乾燥させたフラスコにテトラヒドロフラン(THF)(3
0mL)中の中間体4−(1−[2,6,6−トリメチルシクロヘキセン−1−イ
ル])−(ブト−3−エン−1−イン)(2)(1.045g、6.0ミリモル)
を入れ、−78℃に冷却した。ヘキサン中のn−BuLiの溶液(2.25M;2
.75mL、6.15ミリモル)をゆっくり添加し、僅かに黒ずんだ混合物を10
分間撹拌した。シアン化フェニル(フェノールと臭化シアンからムレイ(R.E.
Murray)ら、Synthesis、150(1980)(参照のためここに開示内容を
包含する)の方法に従って調製したもの(750μL、6.6ミリモル)を加え
、該混合物を室温(例えば約20〜25℃)にまで温めた。水酸化ナトリウム(
6Nの溶液を2mL)を添加し、該混合物を酢酸エチル(50mL)で抽出した
。該有機層をさらにNa0H(2×10mL)、水(3×10mL)および食塩
水(2×100L)で洗浄し、MgSO4で乾燥させ、該溶剤を蒸発させた。得
られた残渣物は実質的に純粋であり(TLCによる)、幾つかのベースライン物
質を除去するため溶離剤としてヘキサンを使用してシリカゲルの短いパッドを通
して濾過した。該溶剤を蒸発させ、わずかに黄色の油状物を1.088g(91
%の収率)得た。Rf0.75(ヘキサン)、IR(生味)2933、2320

(2Z,4E)−3−メチル−5−(2,6,6−トリメチル−
1−シクロヘキセン−1−イル)−2,4−ペンタジエニトリル(4):
50mL容の炎で乾燥させた丸底フラスコに無水ヨウ化銅(1.09g、5.7
0ミリモル)およびTHF(20mL)を入れ、0℃に冷却した。メチルリチウ
ム(Et2O中の1.4M溶液を4.28mL)を該溶液が無色透明になるまで、
ゆっくりと10分間かけて添加した。該銅酸塩溶液をドライアイス−アセトン浴
を使用して−78℃に冷却し、THF(10mL)中の5−(2,6,6−トリメ
チル−1−シクロヘキセン−1−イル)−ペント−2−イン−4−エンニトリル
3(544mg、2.73ミリモル)の溶液を滴下した。ウエストミジェ(West
mijze)ら、Tetrahedron letters、3327(1979)および
Synthesis、454(1978)(その開示内容は参照のため本明細書に包含さ
れる)を参照。該混合物をこの温度にて45分間撹拌し、飽和塩化アンモニウム
溶液(10mL)を添加した。ついで、該混合物を室温に温めた。EtOAc(
50mL)を添加し、2%のNaOH(2×10mL)で洗浄し、ついで飽和N
H4Cl(2×10mL)、水(2×10mL)および食塩水(2×10mL)
で洗浄した。該有機層をMgSO4で乾燥させ、該溶剤を蒸発させた。得られた
残渣物は実質的に純粋であり(TLCによる)、幾つかのベースライン物質を除
去するため溶離剤としてヘキサン:EtOAc(9:1)を使用してシリカゲル
の短いパッドを通して濾過した。該溶剤を蒸発させ、わずかに黄色−オレンジ色
の油状物の538mg(92%の収率)を得た。Rf0.75(ヘキサン:Et

(2Z,4E)−3−メチル−5−(2,6,6−トリメチル−
1−シクロヘキセン−1−イル)−2,4−ペンタジエンナール(5):
25mL容の炎で乾燥させた丸底フラスコに(2Z,4E)−2,4−ペンタ
ジエニトリル−3−メチル−5−(2,6,6−トリメチル−1−シクロヘキセン
−1−イル)(4)(94.2mg、0.438ミリモル)およびヘキサン(5.0
mL)を入れ、−78℃に冷却した。水酸化アルミニウムジイソブチル(ヘキサ
ン中の1.0M溶液を480μL、0.480ミリモル)を内部反応温度が−70
℃を超えないような速度で滴下した。該混合物を5分間撹拌したところ、薄層ク
ロマトグラフィー(TLC)分析(ヘキサン:EtOAc(9:1))は該反応
が完了した(出発物質のRf0.75;生成物のRf0.70)ことを示した。ロ
シェル塩の飽和溶液(1.0mL)を添加し、該混合物を室温まで温めた。Et
OAc(10mL)を添加し、該混合物を水(2×10mL)および食塩水(2
×10mL)を用いて洗浄した。該有機層をMgSO4で乾燥させ、該溶剤を蒸
発させ
た。得られた残渣物は実質的に純粋であり(TLCによる)、90.5mg、9
5%の収率であった;IR(生味)2958、2928、2866、1668(

エチル−3,7−ジメチル−9−(2,6,6−
トリメチル−1−シクロヘキセン−1−イル)−2−ト ランス−4−トランス−6−シス−8−トランス−ノナテトラエノエート(6)
無水THF(5.0mL)に溶かしたジエチル−3−エトキシカルボニル−2
−メチルプロプ−2−エニルホスホネート(354mg、1.33ミリモル)の
溶液を0℃に冷却し、無水DMPU(0.5mL)およびヘキサンに溶かしたn
−BuLi(2.35M溶液を0.56mL、1.33ミリモル)を添加した。該
混合物を同温で20分間撹拌し、ついで−78℃に冷却した。THF(3.0m
L)中に(2Z,4E)−3−メチル−5−(2,6,6−トリメチル−1−シク
ロヘキセン−1−イル)−2,4−ペンタジエンナール(5)(241mg)1.
115ミリモル)を溶かした溶液をゆっくり添加し、該反応混合物を−78℃で
さらに60分間撹拌した。TLCにてモニターされたように反応が完了したとき
に該混合物を0℃まで加温した。飽和塩化アンモニウム溶液(5mL)を添加し
、EtOAc(3×10mL)を使用して該混合物を抽出した。該有機層を水(
2×5mL)および食塩水(10mL)で洗浄し、MgSO4で乾燥させ、濃縮
した。得られた残渣物を、短いシルカゲルクロマトグラフィー(sgc)カラム
上で精製して、13−トランス異性体:13−シス異性体の〜15:1の割合で
所望のエステルの296mg(78%の収率)を得た。IR(生味):2928

トリメチル−1−シクロヘキセニル)−2−トランス−
4−トランス−6−シス−8−トランス−ノナ−テトラエン酸(7)
エタノール(10mL)中のエチル−3,7−ジメチル−9−(2,6,6−ト
リメチル−1−シクロヘキセン−1−イル)−2−トランス−4−トランス−6
−シス−8−トランス−ノナテトラエノエート(6)(275mg)8.25ミ
リモル)の溶液を1NKOH(10mL)で70℃にて3時間処理し、室温にま
で冷却し、10%(容量:容量)のHClで酸性にして、EtOAc(2×20
mL)で抽出した。該有機層を水(2×5mL)および食塩水(2×5mL)で
洗浄し、MgSO4で乾燥させ、該溶剤を蒸発させた。残渣物をエタノール:水
(材料の1g当たり〜20ml)の9:1混合物の最小容量で溶かすことにより
