Claims (1)
- 【特許請求の範囲】 1.タンパク質溶解インヒビターを検出するための固相イムノアッセイであって : a) 適当な支持体に共有結合したチューブリンタンパク質またはチューブリン −様ペプチドを、プロテアーゼインヒビターと一緒にタンパク質溶解活性を含む 溶液と接触させ; b)選択したプロテアーゼに対するインヒビター活性を、支持体と支持体に連結 したチューブリンタンパク質の遊離末端を特異的に認識する標識モノクローナル 抗体を含む溶液体とを接触させることにより検出する、ことを含んで成る、上記 固相イムノアッセイ。 2.チューブリンタンパク質またはチューブリン−様ペプチドがN−末端を介し て支持体に共有結合している、請求の範囲第1項に記載のイムノアッセイ。 3.チューブリンタンパク質またはチューブリン−様ペプチドが結合している支 持体が、ポリマー性材料から作られたマイクロタイタープレートのウェルである 、請求の範囲第1、2または3項のいずれかに記載のイムノアッセイ。 4.マイクロタイタープレートのウェルのポリマー性材料が、ポリエチレン、ポ リスチレンまたはポリ塩化ビニルであり、そしてチューブリンが上記支持体にカ ルボジイミド誘導体、N−ヒドロキシスクシンイミド誘導体およびスルホスクシ ンイミド誘導体から選択される活性化試薬により該支持体に結合される、請求の 範囲第3項に記載のイムノアッセイ。 5.活性化試薬がビス(スルホスクシニジミル)スベレートである、請求の範囲 第4項に記載のイムノアッセイ。 6.標識モノクローナル抗体が、モノクローナル抗体および標識抗−抗 体を含んで成る系であり、ここで第一抗体が結合したチューブリンの遊離末端を 特異的に認識し、一方標識抗−抗体が第一抗体に特異的である、請求の範囲第1 ないし第5項のいずれか1項に記載のイムノアッセイ。 7.抗−抗体がペルオキシダーゼ酵素を用いて標識されている、請求の範囲第6 項に記載のイムノアッセイ。 8.モノクローナル抗体がα−チューブリンのC−末端を特異的に認識する、請 求の範囲第7または8項に記載のイムノアッセイ。 9.支持体に連結されたチューブリンタンパク質がα−チューブリンである、請 求の範囲第1ないし第8項のいずれかに記載のイムノアッセイ。 10.以下の配列番号1: のアミノ酸配列を有するペプチド。 11.請求の範囲第1ないし第8項のいずれか1項に記載の固相イムノアッセイ での請求の範囲第10項に記載のペプチドの使用。 12.表面にN−末端を介して共有結合しているチューブリンタンパク質または チューブリン−様ペプチドを有する分析用支持体、およびチューブリンタンパク 質のC−末端を特異的に認識する標識モノクローナル抗体を含む溶液を含んで成 る分析用キット。 13.支持体に結合しているチューブリンがα−チューブリンである、請求の範 囲第12項に記載の分析用キット。 14.標識モノクローナル抗体が、モノクローナル抗体および標識抗−抗体を含 んで成る系であり、ここで第一抗体がα−チューブリンのC−末端を特異的に認 識し、一方標識抗−抗体が第一抗体に特異的である、請求の範囲第12項に記載 の分析用キット。
