真菌酵素及び細菌酵素を用いる下水処理用組成物及び方法
発明の分野
本発明は、真菌酵素及び細菌酵素によるセルロース分解に関する。酵素のこの
組合わせは、下水処理、特に浄化槽に有効である。
発明の背景
微生物による下水処理は、当該技術において既知である。多くの下水処理セン
ター及び個々の浄化槽は、汚水の分解に微生物を用いるものである。一般に、下
水は水、有機廃棄物(炭水化物、脂肪及びタンパク質を含む)及び紙製品からの
セルロースを含む。セルロースは、生(未処理)下水中の固形分の約15%まで
を表すことがある。
典型的には、下水の非セルロース有機廃棄物成分はセルロース成分より容易に
分解される。有機廃棄物を構成する炭水化物、脂肪及びタンパク質は、選択され
た細菌の細胞の外に遊離した細胞外酵素によってかなり容易に消化される。しか
しながら、セルロースの分解は依然として多くの下水処理方式における課題であ
る。
セルロースのグルコースへの分解プロセスは、段階的である。まず、セルロー
スがエンドグルカナーゼの作用により加水分解され、セルロースの無定形領域に
沿って結合が破壊される。この酵素反応は、β(1>4)結合の切断を行ってセ
ロビオースを生成し、β(1>4)エキソグルカナーゼの作用により分子の非還
元端から除去される。この後、セロビオースがβ(1>4)グルコシダーゼによ
ってグルコースに加水分解される。即ち、セルロースのグルコースへの分解は、

発明の要約
前述の課題は、バシラス種(Bacillus spp.)培養物を真菌セルラーゼと組合わ
せて含む下水処理組成物の発見で解決した。バシラス種からの細菌培養物によっ
て生産された細胞外酵素と真菌セルラーゼの組合わせは相乗的セルロース分解を
もたらす。結果から、セルロースを含む下水を本発明の組成物と接触させるとセ
ルロース分解の結果としてグルコース生成の顕著な増大が示される。本組成物は
、セルロース分解の増大ほかに炭水化物、脂肪及びタンパク質を分解することが
できる幅広い成分の分解系である。本組成物が天然に存在する微生物からの酵素
を含むことから、浄化槽添加剤として特に有効である。
本発明は、また、炭水化物、タンパク質、脂肪及びセルロース並びにその混合
物を分解する、そのバシラス種培養物及び真菌セルラーゼの新規な使用方法を提
供するものである。
好適実施態様の詳細な説明
バシラス種は、Williams & Wilkins社によって出版されたBergey's Manual of
Determinative Bacteriologyの第8版、1986年の1105〜1139頁に同定されてい
る既知の天然に存在する細菌である。好ましいバシラス種としては、B.サチリス
(subtilis)、B.リケニホルミス(licheniformis)、B.メガテリウム(megaterium)
及びその混合物が挙げられる。細菌培養物は、B.サチリス、B.リケニホルミス及
びB.メガテリウムの混合物であることが更に好ましい。当業者に既知であるよう
に、細菌培養物は培養物が貯蔵される期間を延長するために胞子として調製され
る。貯蔵に便利のよいように、バシラス種培養物は胞子として組成物中に存在さ
せることが好ましい。胞子は、下水のような栄養物に曝されると酵素生産生物に
なる。胞子が汚水に曝されると細菌になり、炭水化物、脂肪及びタンパク質を分
解するのに特に有効な細胞外酵素を生産する。本組成物中に用いられるバシラス
種の胞子数は、処理されるべき下水の種類、下水処理施設の大きさ、下水を組成
物で処理する頻度等によってかなり変動させてもよい。
本明細書に用いられる組成物の有効成分は、細菌培養物及び真菌酵素として定
義される。典型的な浄化槽添加剤として調製された組成物用細菌培養物の濃度範
囲は、好ましくは少なくとも約104胞子/g組成物(有効成分)を使用し、胞
子の上限濃度はたいていコストによってのみ制限される。更に好ましくは少なく
とも106胞子/g組成物(有効成分)、最も好ましくは106〜108胞子/g
組成物(有効成分)が組成物に用いられる。
セルラーゼは、真菌アスペルギルス・ニガー(Aspergillus niger)から単離さ
れる。その酵素はその真菌培養物から既知の手段によって抽出され、例えば、No
vo Nordisk、コネチカット州;Sigma Chemical、セントルイス、ミズーリ州;及
びGeorge A.Jeffrey's Company、サレム、バージニア州から広く市販されてい
る。その真菌が好気性でありかつ下水処理が主に浸水式嫌気性環境であることか
ら、セルラーゼ酵素は本発明に用いられる真菌から単離されることが好ましい。
バシラス種菌は、通性嫌気性であり、下水処理の典型的な嫌気性条件で発育する
。
細菌培養物の胞子数のように、本組成物に使用した真菌セルラーゼ酵素の比活
性及び量は広く可変であり、本組成物を使用するシステムの酵素要求に従って調
節される。例えば、特に高含量のセルロースを有する廃棄物システムに関しては
、多量のセルロース酵素が好ましい。浄化槽添加剤としての使用の場合、用いら
れる真菌酵素の活性は好ましくは少なくとも約1000CU/g組成物の有効成
分である(酵素の濃度の上限はたいていコストによってのみ制限される)。浄化
槽添加剤を処方する際の経済的理由で、酵素範囲は更に好ましくは1500〜2
500CU/g組成物の有効成分、最も好ましくは1500〜2000CU/g
組成物の有効成分である。
細菌培養物の真菌酵素に対する割合は、かなり変動させてもよい。細菌培養物
:真菌酵素の比は、有効成分の好ましくは約10:90〜約99.99:0.01
重量%である。当業者に既知であるように、処理されるべき材料の種類、胞子数
及び原料の比活性等によって調節される。
本組成物は、また、処理施設への胞子及び真菌酵素の貯蔵又は供給を容易にす
る任意の充填剤及び添加剤を含めてもよい。用いられる充填剤としては、アルカ
リ金属塩(例えば、NaCl、NaSO4、CaCO3、その混合物等)、不活性
製剤(例えば、ミルオーガナイト)、その混合物等が含まれるが、決してこれら
に限定されない。
本組成物は、当業者に既知の手段によって液体又は粉末として調製される。
前述のように、本発明の組成物に使用された酵素は天然に存在する生物によっ
て生産される。即ち、本組成物は、下水の典型的な成分(例えば、炭水化物、タ
ンパク質、脂肪及びセルロース)の幅広い成分の分解が所望される多くの産業用
に有効である。
下記実施例の項で示されるように、バシラス種酵素と真菌セルラーゼの組合わ
せは相乗的であることがわかった。組合わせて用いられる少量の細菌と真菌酵素
は、多量の単独真菌酵素が用いられる場合よりセルロースを分解するのに有効で
あることがわかった。本発明の組合わせは、幅広い成分を含む下水処理及びセル
ロースが廃棄物である産業用に特に有効であるセルロースからグルコースを生成
するという手段を与える。
本発明の組成物の酵素作用は、広範囲のpHにわたって生じることができる。
最適には、処理されるべき培養液のpH範囲は約4〜約10であり、更に好まし
くはpH6〜8である。処理されるべき培養液の温度範囲はかなり変動させるこ
とができるが、最適酵素作用は温度範囲好ましくは約10〜約45℃、更に好ま
しくは20〜35℃で生じる。セルロースの分解は、また、バシラス種から分離
されかつ真菌起原のセルラーゼと組合わせた酵素で起こることができる。当業者
に既知であるように、真菌は好気性微生物でありバシラス種は通性嫌気性微生物
であるので、組成物を調製する際に基質環境の酸素含量を考慮しなけければなら
ない。
当業者に既知であるように、本組成物の用量、使用頻度及び有効成分の濃度は
相互に依存する可変量であり、処理されるべき環境、分解されるべき粒子の濃度
、前の微生物の使用等によっても広く変動するであろう。これらの可変量に対す
る調整は、当業者に既知の通常の手順により達成される。例えば、浄化槽添加剤
としての使用の場合(浄化槽は典型的には約1000ガロンの容量を有する)、
本組成物の有効成分の有効量は少なくとも約10g、更に好ましくは少なくとも
約100g(使用量の上限は主にコストによって制限される)、最も好ましくは
150〜1000gである。
下記の実施例によって本発明を更に具体的に説明するが、これらに限定されな
い。
実施例
セルロース分解はグルコース生成により測定され、Aibba,S.,K.Kitai & T.I
manaka,Applied Environmental Microbiology,Vol.46,pp.1059-1065(1983
)に記載されているジニトロサリチル酸法(DNS)よって求めた。
実施例に記載された組成物は、バシラス・サチリス、バシラス・リケニホルミ
ス及びバシラス・メガテリウムから単離された胞子及びアスペルギルス・ニガー
から単離された真菌セルラーゼを使用した。胞子及びセルラーゼは共に、George
A.Jeffreys Company から入手した。培養物の計数は、108胞子/g組成物の
有効成分であった。セルラーゼの比活性は、1600CU/g組成物の有効成分
であった。(供給会社から入手した実際のセルラーゼ酵素活性は約128,00
0CU/gであった。)ミルオーガナイトは、Milwaukee Metropolitan Sewage
District、ミルウォーキー、ウィスコンシン州から購入した。
実施例1及び2並びに比較例1及び2
合成の下水は、タンパク質5%、脂肪5%、セルロース5%及び残りは蒸留水
で調製した。合成の廃棄物を、各試験組成物用の35ml試験管に入れた。
本発明の組成物A
実施例1
組成物Aを、最終セルラーゼ濃度0.050%(64CU/g)に廃棄物で1
0%に希釈した。48時間後、グルコース生成(もって、セルロース分解のレベ
ルを示す)をDNS法を用いて下記表1に示されるように4.83g/1tとし
て測定した。
実施例2
組成物Aを、最終セルラーゼ濃度0.0125%(16CU/g)に廃棄物で
2.5%に希釈した。47時間後、グルコース生成をDNSにより下記表1に示
されるように1.31g/1tとして測定した。
比較例1
合成廃棄物にセルラーゼのみを0.5%の濃度(640CU/g)で加えた以
外は、実施例1を繰り返した。48時間後、グルコース生成をDNSにより表1
に
示されるように1.41g/1tとして測定した。
比較例2
組成物Aが真菌セルラーゼを含まずかつCaCO3とNaSO4を等価量のミル
オーガナイトで置き換えた以外は、実施例1を繰り返した。48時間後、グルコ
ース生成をDNSにより表1に示されるように1.31g/1tとして測定した
。
表1は、実施例1及び2並びに比較例1及び2で得られたデータの纏めである
。真菌セルラーゼを培養物に加えると、グルコース生成の顕著な増大が検出され
た。実施例1及び2が示すように、セルラーゼが添加されると、実施例1及び2
のセルラーゼ濃度が単独酵素より約10倍少ないが、単独酵素より多いか又は同
じ量のグルコースが本組成物により生成された。従って、単独酵素と真菌酵素及
び細菌酵素の組合わせとの間のこれらの主な差は、細菌酵素と真菌酵素の相乗作
用によるものであった。同様の結果が生下水を用いて見られた(表2)。
本発明の組成物B
実施例3
生下水(Ridgewood排水処理工場、リッジウッド、ニュージャージー州から入手
した)を35ml試験管に入れ、組成物Bを最終セルラーゼ濃度0.0125%(
16CU/g)として2.5%に希釈した。48時間後、グルコース生成(もっ
て、セルラーゼ分解レベルを示す)をDNSを用いて下記表2に示されるように
0.291g/1tとして測定した。
組成物B又は酵素単独を存在させずにグルコース生成を測定する対照も実験し
た。表2に示されるように、試料をDNS法によって分析した際にグルコースが
検出されなかった。
比較例3
生下水にセルラーゼのみを0.5%の濃度(640CU/g)で加えた以外は
、実施例3を繰り返した。48時間後、グルコース生成をDNSにより表2に示
さ
れるように0.128g/1tとして測定した。
比較例4
組成物Bが真菌セルラーゼを含まずかつCaCO3とNaSO4をミルオーガナ
イトで置き換えた以外は、実施例3を繰り返した。48時間後、グルコース生成
をDNSにより表2に示されるように0.029g/1tとして測定した。
以上、本発明を具体的な好適実施態様について記載してきた。上記の詳細な説

び置換を行うことができる。DETAILED DESCRIPTION OF THE INVENTION Compositions and Methods for Sewage Treatment Using Fungal and Bacterial Enzymes FIELD OF THE INVENTION The present invention relates to cellulolysis by fungal and bacterial enzymes. The carbohydrates, fats and proteins that make up organic waste are fairly easily digested by extracellular enzymes released outside the cells of selected bacteria. However, cellulose degradation remains a challenge in many sewage treatment systems. The process of breaking down cellulose into glucose is gradual. First, cellulose is hydrolyzed by the action of endoglucanase, and bonds are broken along the amorphous region of cellulose. This enzymatic reaction cleaves the β (1> 4) bond to produce cellobiose, which is removed from the non-reducing end of the molecule by the action of β (1> 4) exoglucanase. After this, cellobiose is hydrolyzed to glucose by β (1> 4) glucosidase. That is, the breakdown of cellulose into glucose involves a complex of enzymes. It is believed that sufficient quantities of these enzymes are not produced by natural bacteria. The lack of sufficient enzymatic activity is especially apparent in septic tanks, where cellulose precipitates are a problem. There is a need for new methods to reduce cellulose precipitates in raw sewage. SUMMARY OF THE INVENTION The foregoing problems have been solved by the discovery of a sewage treatment composition comprising Bacillus spp. Culture in combination with a fungal cellulase. The combination of extracellular enzymes and fungal cellulases produced by bacterial cultures from Bacillus sp. Results in synergistic cellulolysis. The results show a significant increase in glucose production as a result of cellulolysis when sewage containing cellulose is contacted with the composition of the invention. The composition is a broad component degradative system capable of degrading carbohydrates, fats and proteins as well as increasing cellulolysis. This composition is particularly effective as a septic tank additive because it contains enzymes from naturally occurring microorganisms. The present invention also provides novel uses of the Bacillus seed culture and fungal cellulases for degrading carbohydrates, proteins, fats and celluloses and mixtures thereof. Detailed Description of the Preferred Embodiments Bacillus spp. Is a known naturally occurring bacterium identified in the 8th edition of Bergey's Manual of Determinative Bacteriology, published by Williams & Wilkins, p. .
