【発明の詳細な説明】インターロイキン−6の生産を阻害するステファニア テトランドラの抽出物 発明の分野
本発明は、インターロイキン−6(interleukin−6、以下、IL−6と略称
)の生産を阻害するステファニア テトランドラ エス ムーア(Stephania te trandra
S.Moore)の抽出物、この抽出物の製造方法およびIL−6の過剰生産
によって惹起される免疫疾患の治療に有用な前記抽出物を含む医薬組成物に関す
る。
発明の背景
大韓民国の南部地方や済州島に見られるステファニア ジャポニカ ミアース
(Stephania japonica Miers)およびシノメニウム アクタム レド エ ウィ
ルス(Sinomenium acutum rehd et Wils)(メニスペルマケアエ(Menispermace
ae))は、鎮痛剤および抗炎症剤として長い間用いられてきた。一方、大韓民国
に見られないエス.テトランドラ エス.ムーア(メニスペルマケアエ)は、中
国などで神経痛および関節炎の治療剤として伝統的に用いられてきた。特に、ア
ルカロイドであるテトランドリンは抗炎症剤および抗高血圧剤として用いられて
きた。エス.テトランドラ エス.ムーアは、食作用阻止効果および酸化防止効
果を有し(Seow,W.k.,et al.,Int.Archs Allergy Appl.Immun.,85,404(1988))
、臨床および実験的な硅肺症モデルにおいて効果を示すと報告され(Li,Q.,et a
l.,Chinese J.Tuberc.Resp.Dis.,4,321(1981);Xu,X.,et al.,Eoltoxicol.Enviro
n.Safety,7,306(1983);およびLiu,B.,et al.,Ecotoxicol.Environ.Safety,7,3
23(1983))、ヒト単球によって分泌されるインターロイキン−1および腫瘍壊死
因子−αの生産を阻害する能力を有することが知られている(Seow,W.K.,et.,Cl
in.Exp.Immunol.,75,47(1989);およびFerrante,A.,et al.,Clin.Exp.Immunol.,80
,232(1990))。テトランドリンおよびその誘導体は、脳機能を活発にするもの
と報
告され(Tsumura & CO,WPI Acc.No.:92-231935(1992))、マラリア治療剤および
毛髪成長促進剤として開発された(Sunstar KK,WPI Acc.No.:89-117236(1989))
。
周知のように、IL−6は細胞の成長、分化および活性化に関与する調節因子
である。これは種々の器官細胞によって生産および分泌され、人体の防御機構に
重要な役割をする(Hirano,T.,et al.,Immunol.Today,11,443(1990))。
単球の培養液から初めて発見されたIL−6はB細胞による抗体生産を誘導す
ると報告された(Muraguchi,A.,et al.,J.Immunol.,127,412(1981))。IL−
6のcDNAがヒラノら(Hirano,T.at al.)によって成功裡にクローニングさ
れて以降(Nature,324,73(1986))、IL−6は、B細胞ハイブリドーマ(hybri
doma)および形質細胞腫に対して成長因子として作用し(Snick,V.J.,et al.,Pr
oc.Natl.Acad.Sci.U.S.A.,83,9679(1986))、また造血作用に関与する因子とし
ての役割をすると報告された(Koike,K.,et.,J.Exp.Med.,168,879(1988))。さ
らに、IL−6は、T細胞の活性化および成長を刺激し(Lotz,M.,et al.,J.Exp
.Med.,167,1253(1988))、肝細胞の急性段階反応(acute phase response)を誘
発し(Geiger,T.,et al.,Eur.J.Immunol.,18,717(1988))、神経系での細胞分化
を調節し(Satoh,T.,Mol.Cell.Biol.,8,3546(1988))、ケラチノサイトの成長を
刺激し、骨の新陳代謝を調節し、メサンギウム細胞の成長を刺激し、黒色腫およ
び乳がん細胞の成長を阻害するなどの機能を有すると報告された。
すでに、報告されているように、IL−6の生産を適切に調節しないと種々の
疾病が生じる。報告された疾病の例としては慢性関節リウマチ(Hirano,T.,et a
l.,Eur.J.Immunol.,18,1797(1988))、肝硬変(Deviere,J.,et al.,Clin.Exp.Im
munol.,77,221(1989))、乾せん(Grossman,R.M.,et al.,Proc.Natl.Acad.Sci.U
.S.A.,86,6367(1989))、多発性骨髄腫(Bataille,R.,et al.,J.Clin.Invest.,8 4
,2008(1989))、心臓粘液腫(cardiac myxoma)、AIDS(Miles,S.A.,et al
.,Proc.Natl.Acad.Sci.U.S.A,87,4068(1990))および他の自己免疫疾患がある。
かかる観察は、人体内の免疫体系の恒常性維持および疾病の治療や予防のために

L−6生産の調節が重要であるという事実を支持している。
したがって、インターロイキンの生産を調節するための多数の研究法が提案さ
れてきた。たとえば、IL−6またはIL−6受容体に対する抗体を用いてIL
−6の過度な分泌によって骨髄腫にかかった患者の骨髄球の増殖を阻害した(Su
zuki,H.,Eur.J.Immuno.,22,1989(1992))。しかし、IL−6の生産を特異的に
阻害する物質または方法は今のところ報告されおらず、したがって、IL−6の
生産を特異的に阻害できる阻害剤の発見が必要とされていた。
発明の概要
したがって、本発明の目的は、IL−6の生産を特異的に阻害するエス.テト
ランドラ エス.ムーアの根から得られた抽出物を提供することである。
本発明の他の目的は、IL−6の過剰生産によって惹起される免疫疾患の治療
に有効な量の該抽出物を含有する医薬組成物を提供することである。

発明の詳細な説明
本明細書で言及した全ての文献はここに引用して本明細書の記載とする。
本発明によって、ステファニア テトランドラ エス.ムーア(Stephania te trandra
S.Moore)の根からの抽出物は、IL−6の生産を特異的に阻害する能
力を有し、したがって、IL−6の過剰生産によって惹起される種々の免疫疾患
の治療に有用であるということが見出された。
エス.テトランドラ エス.ムーアの前記抽出物は、種々の溶媒、たとえば、
メタノール、エタノール、ヘキサン、CH2Cl2またはこれらの混合物、生理食
塩水、蒸留水などを用いて製造できる。特に、下記にさらに詳細に説明されるエ
ス.テトランドラ エス.ムーアの抽出物A、B、CおよびDは下記の好ましい
態様によって製造できる。
乾燥したエス.テトランドラ エス.ムーア根1kgにメタノール1ないし3l
、好ましくは2lを加え、混合物を50ないし70℃の温度で6ないし12時間
、または室温で少なくとも24時間撹拌した後、濾過する。前記手順を好ましく
は
3回繰り返し、濾液を集めて減圧下、たとえば、7mmHgのもとで濃縮して抽出
物Aを得る。
前記抽出物A100gをメタノール200ないし400ml、好ましくは250
mlとヘキサン200ないし400ml、好ましくは250mlとに分配する。これか
らメタノール分画を分離した後、減圧濃縮する。水酸化アンモニウムまたは水酸
化ナトリウムを用いて残留物をpH9ないし11の範囲に調整する。生成物を蒸
留水とCH2Cl2との混合液(1:1(v/v))400ないし800ml、好ま
しくは500mlに分配する。これからCH2Cl2分画、すなわちアルカロイド分
画を分離した後、減圧濃縮して抽出物Bを得る。
一方、乾燥したエス.テトランドラ エス.ムーア根1kgを粉砕し、篩いにか
けた後、蒸留水または生理食塩水に50ないし200mg/ml、好ましくは100
mg/mlの濃度で懸濁する。この懸濁液を80ないし100℃、好ましくは95℃
で4ないし12時間、好ましくは6時間加熱する。加熱した懸濁液を濾過し、濾
液を減圧濃縮して抽出物Cを得る。
他の方法としては、乾燥したエス.テトランドラ エス.ムーア根1kgを蒸留
水1ないし3l、好ましくは2lと混合し、80ないし100℃、好ましくは9
5℃で4ないし15時間、好ましくは12時間加熱する。加熱した混合物を濾過
し、濾液を減圧濃縮する。残留物を−70ないし−90℃、好ましくは−80℃
で2ないし10時間、好ましくは8時間貯蔵した後、4ないし8時間、好ましく
は6時間凍結乾燥して粉末状の抽出物Cを得る。
また、抽出物Dは乾燥したエス.テトランドラ エス.ムーア根1kgにエタノ
ール1ないし3l、好ましくは2lを加え、混合物を60ないし90℃で6ない
し12時間、または室温で24時間以上撹拌する手順を含む抽出過程を好ましく
は3回繰り返して集めた濾液を減圧濃縮することによって得られる。
前記抽出物A、B、CおよびDはそれぞれ抗炎症効果を示し、コラーゲンの合
成および反応性酸素類(reactive oxygen species)の生産を阻害し、血清中の
GOTおよびGPT濃度を減少させる。したがって、これらはそれぞれ単独で、
または混合物として慢性関節リウマチ、肝硬変、乾せん、多発性骨髄腫、心臓粘
液腫、硅肺症およびAIDSのようなIL−6の過剰生産によって惹起される免
疫疾患治療用の医薬組成物に使用され得る。ここで好ましくは、抽出物Aは炎症
性疾患、関節炎および繊維化疾患の治療用;抽出物Bは関節炎および自己免疫肝
硬変の治療用;抽出物Cは硅肺症および肝の繊維化疾患の治療用;抽出物Dは肝
硬変の治療用に使用され得る。IL−6の過剰生産による免疫疾患の治療に有用
な本発明の医薬組成物は、活性成分であるエス.テトランドラ エス.ムーアの
抽出物と共に薬剤学的に許容可能な賦形剤、担体または希釈剤を含むことができ
る。医薬製剤は通常の方法によって製造できる。
組成物の製造において、活性成分は担体に混合、希釈するか、またはカプセル
、におい袋(sachet)または他の容器形態の担体に封入することが好ましい。担
体が希釈剤として働く場合、これは活性成分の溶剤、賦形剤または媒質として作
用する固体、半固体または液体物質であり得る。したがって、この組成物は錠剤
、丸剤、粉末、香粉、エリキシル、懸濁剤、乳化剤、液剤、シロップ、エアロゾ
ール、軟質および硬質ゼラチンカプセル、滅菌注射剤、滅菌包装粉末などのよう
な形態であり得る。
適当な担体、賦形剤および希釈剤の例としては、ラクトース、デキストロース
、スクロース、ソルビトール、マンニトール、スターチ、アカシアゴム、アルギ
ン酸塩、ゼラチン、リン酸カルシウム、ケイ酸カルシウム、セルロース、メチル
セルロース、微細結晶性セルロース、ポリビニルピロリドン、水、ヒドロキシ安
息香酸メチル、ヒドロキシ安息香酸プロピル、タルク、ステアリン酸マグネシウ
ムおよび鉱油などがある。該組成物は潤滑剤、湿潤剤、香味剤、乳化剤および防
腐剤などをさらに含むことができる。本発明の組成物は、患者に投与してから、
活性成分を迅速に、または持続的に、または遅延放出させるため、当分野で周知
の任意の方法によって調剤できる。
前記医薬組成物は、経口、経皮、皮下、静脈内および筋肉内投与をはじめ、種
々の経路を通じて投与できる。通常の活性成分の1日投与量は約1ないし500
μg/kg体重、好ましくは30ないし300μg/kg体重の範囲であり、一回また
は数回にかけて投与できる。しかし、実際に投与する活性成分の量は治療すべき
症状、投与経路、患者の年齢および体重、および患者の症状を含む種々の関連因
子を考慮して決定しなければならないことは勿論であり、したがって、前述した
投与量は本発明を限定しない。
下記製造例および実施例は本発明の範囲を限定することではなく、本発明をさ
らに詳細に説明するためのもので、特に言及しない限り、実施例で用いられた実
験方法は下記参照実施例によって実施できる。
また、以下固体混合物中の固体、液体中の液体および液体中の固体に対する百
分率は特に言及しない限り、それぞれ重量/重量、体積/体積および重量/体積
に基づいたものである。
製造例:エス.テトランドラ エス.ムーアの抽出物の製造
よく乾燥したエス.テトランドラ エス.ムーア根約4.0kgを細かく切り刻
み、メタノール約51で二日間抽出した。この抽出手順を3回繰り返して集めた
抽出物を減圧濃縮して5.6%収率でメタノール抽出物約224g(抽出物A)
を得た。
抽出物A200gを90%メタノール500mlとn−ヘキサン500mlの混合
物に分配した後、90%メタノール層を分離して減圧濃縮してメタノールを除い
た。残留物を0.1M MH4OHでpH10に調整して蒸留水:CH2Cl2(1:
1(v/v))混合物600mlに分配させた。次いで、CH2Cl2層、すなわち
、アルカロイド分画を分離し、減圧濃縮して0.6%収率で抽出物B25gを得
た。
一方、硅肺症治療試験に用いるための抽出物Cは次のように製造した。よく乾
燥したエス.テトランドラ エス.ムーア根1kgを粉砕し、篩い(60メッシュ
)にかけた後、蒸留水に100mg/mlの濃度で懸濁させた。この懸濁液を100
℃で6時間加熱して濾過した。濾液を減圧濃縮して8%収率でエス.テトランド
ラ エス.ムーアの水抽出物(抽出物C)80gを得た後、これを−20℃で貯
蔵
した。
肝硬変治療試験に用いるための抽出物Cを製造するために、よく乾燥したエス
.テトランドラ エス.ムーア根1113.5gを蒸留水21と共に冷却装置が
備えられた31丸底フラスコに入れ、混合物を95℃で12時間加熱した後濾過
した。濾液を回転真空蒸発器(Buchi451)を使って減圧濃縮し、ディープフリー
ザー(SANYO、Japan)で−84℃で3時間冷却した後、凍結乾燥器(EYELA
,Japan)を使って4時間凍結乾燥して5.1%収率で粉末状抽出物C56.5
5gを得た。
また、よく乾燥したエス.テトランドラ エス.ムーア根500gをエタノー
ル約1.51を用いて室温で3日間抽出した。この抽出手順を3回繰り返して集
めた抽出物を減圧濃縮して2.6%収率でエス.テトランドラ エス.ムーアの
エタノール抽出物13gを得た。
参照例1:検定用細胞の分離
(1)ヒト単球、マクロファージおよび好中球の分離
正常なヒト末梢血液をヘパリンで処理し、同量のハンクス細胞培養用塩類溶液
(Hank's balanced salt solution(HBSS):Mg2+およびCa2+を含有しない)
で希釈した。希釈した血液を、密度1.119のフィコル−ヒパク(Ficoll-Hypa
que,Sigma,St.Louis,MO,U.S.A.)層上に積み重ねた密度1.077のフィコル−
ヒパク層を含む遠沈管に入れ、700xgで30分間遠心して密度1.077の
フィコル−ヒパク層と血漿層との間の層から単球を、そして密度1.077のフ
ィコル−ヒパク層と密度1.119のフィコル−ヒパク層との間の層から好中球
を得た。分離された細胞を4℃HBSS(Ca2+およびMg2+を含有しない)で
2回洗浄し、10%牛胎児血清(FBS,Hyclone,Logan,UT,U.S.A.)を含むRPM
I1640培地(Gibco,Grand Island,NY,U.S.A.)に懸濁させた。懸濁液を24
−ウェル培養平板(Costar,Cambridge,MA,U.S.A.)のウェルに入れ、37℃で2
時間培養して単球、マクロファージおよび好中球を得た。
(2)繊維芽細胞の分離
ファンらの方法(Phan S.H.,et al.,J.Clin.Invest.,76,241(1985))の変形を
用いてラットから繊維芽細胞を次のように分離した。
ラットをエーテルで麻酔させ、無菌作業台で肺臓を取り出した。肺臓を2ない
し4mmの大きさに細かく切り刻み、コラゲナーゼおよび0.5%トリプシンを
含むリン酸塩緩衝食塩水(PBS)に懸濁させて37℃で2時間組織を消化した
。懸濁液を滅菌したガーゼに通して濾過し、消化されなかった組織などを除いた
。分離した細胞はPBSで2回または3回洗浄し、10%牛胎児血清(FBS,Hycl
one,Logan,UT,U.S.A.)を含むRPMI1640培地(Gibco,Grand Island,NY,U
.S.A.)を懸濁させた。懸濁液を培養平板のウェルに入れ、5%CO2培養器(Lu
naire Environ,Inc.,Pennsylvania,U.S.A.)を用いて37℃で1ないし2日間培
養した。平板をRPMI1640培地で洗浄して平板に付着していない細胞を除
いた。新鮮な培地を平板に加え、密集層が形成されるまで培養を続けた。5回以
下の継代培養を経た細胞を下記試験に用いた。
NIH3T3繊維芽細胞(ATCC CRL 1658)を10%FBSを含むRPMI1
640培地で前述した条件下で培養した。
(3)慢性関節リウマチ患者から滑膜細胞の分離
慢性関節リウマチ患者の滑膜組織を冷PBSで3回洗浄し、滅菌はさみを使っ
て2mmの大きさに細かく切った後、これをコラゲナーゼA(5mg/ml,BM,India
napolis,IN,U.S.A.)とDNaseタイプI(0.15mg/ml,Sigma)を含むDME
M(Sigma,U.S.A.)に懸濁し、5%CO2培養器を用いて37℃で2時間培養し
た。その後、ここに0.5%トリプシン−0.2%EDTAを加えて30分間培養
を続けた。消化された組織をPBSで2回、およびDMEMで1回洗浄し、分離
した細胞を10%FBSを含有したDMEM(DMEM−10%FBS)に懸濁
させて一週間培養した。
その後、トリプシン−EDTAで滑液付着細胞を分離し、DMEMで洗浄した
後、DMEM−5%FBSに105細胞/mlの濃度で懸濁させた。懸濁液を24
−ウェル培養プレートのウェルに1ml/ウェルの量で加え、37℃で24時間培
養した。生成した培養液を次の実験に用いるために−20℃で貯蔵し、その一部
は継代培養して凍結状態で液体窒素タンクに貯蔵した。
(4)エス.テトランドラ エス.ムーアの抽出物を用いた細胞の処理
エス.テトランドラ エス.ムーアの抽出物を前記手順で得られた細胞5x1
05/mlに種々の濃度で加え、細胞を5%CO2培養器で37℃で1時間前培養し
た。その後、シリカ(100μg/ml)1mlおよび2%FBSを含むRPMI1
640培地1mlをそこに加え、細胞を前記と同一の条件下で48時間培養した。
培養上澄液を集め、1,500rpmで10分間遠心分離して細胞およびシリカを除
いた。得られた上澄液をPBSに対して透析し、0.2μm濾過注射器で濾過した
後、濾液を−20℃で貯蔵した。
参照例2:エス.テトランドラ エス.ムーアの抽出物の細胞毒性検定
エス.テトランドラ エス.ムーアの抽出物の細胞毒性は下記手順によって決
定した。
参照例1(1)で得られた単球およびマクロファージ5x105細胞/mlずつ
をそれぞれ参照例1(4)の手順に従って、製造例で得られた抽出物AおよびB
0.1、1および10μg/mlずつで処理し、同一条件下で培養した。アリー(Al
ley,M.C.)らの方法(Cancer Res.,48,589(1988))によって、培養液を1ml/ウ
ェルの量で培養プレートのウェルに入れ、3−4,5−ジメチルチアゾール−2
,5−ジフェニルテトラゾリウムブロミド(MTT,Sigma)0.5mgを各ウェル
に加えた。37℃で4時間培養した後、培養液を遠心分離して上澄液を除いた。
酸性化されたイソプロパノール0.04N HCl(イソプロパノール溶液)10
0μlずつを各ウェルの細胞に加えて生きている細胞によって生産されたホルマ
ザンを溶離し、ELISAリーダー(Titertek multiskan Mcc/340)を
用いて540nmでの光学密度(O.D.)を測定した(図1)。
図1は、抽出物AまたはBで処理しなかった対照群の光学密度を100%と見
なす場合、抽出物AまたはBの濃度に対する試料の光学密度の相対値を示す。エ
ス.テトランドラ エス.ムーア抽出物の毒性によって単球およびマクロファー
ジの生存率が減少するとホルマザンの生産も減少し、これによって光学密度が減
小する。抽出物Aで処理した試料は、抽出物Aの濃度が10μg/mlに及ぶまで
、対照群と比べて大きい差を示さず、抽出物Bで処理された試料も同じような結
果を示す。したがって、抽出物AおよびBは10μg/ml以下の濃度では細胞毒
性を示さないことを確認し、以降、すべての試験は前記濃度範囲内で行った。抽
出物AおよびB共に100μg/mlの濃度では細胞毒性を示した。
一方、抽出物CおよびDの細胞毒性はラットを使って検査した。抽出物Cおよ
びD40mgをそれぞれ週2回17週間ラットに経口投与した結果、ラットに対す
る毒性(死亡、体重減少など)がないことが判明した。
実施例1:エス.テトランドラ エス.ムーアの抽出物によるヒト単球および マクロファージでのIL−6生産の阻害
参照例1(1)で得られた単球/マクロファージを抽出物AまたはB0.1な
いし10μg/mlと共に1時間培養し、シリカ100μg/mlで48時間処理した
。培養液を遠心分離して上澄液を得た後、これをPBSに対して透析した。上澄
液中のIL−6の活性はIL−6の依存性B9ハイブリドーマ菌株を用いて測定
した。
2U/mlの組み換えヒトIL−6を加えた10%FBSを含むRPMI164
0培地でB9菌株(Dr.Kishimoto,T.,Osaka University,Japan)を培養し、細胞
を無血清培地で3回洗浄した。細胞を10%FBSを含むRPMI1640培地
に5x104細胞/mlの濃度で懸濁し、懸濁液を96−ウェル培養プレートのウ
ェルに100μl/ウェルの量で入れた。その後、プレートを37℃で5%CO2
下で68時間培養した。3H−チミジン0.5μCiを50μl/ウェルの量でウェ
ルに加え、培養を4時間続けた。培養が終わると、多重細胞採取機(Inotech)
を使って細胞をガラス繊維濾過器に集め、結合された3H−チミジンの量を液体
シンチレーション計測器(Beckman)で測定した。
図2は、抽出物AまたはBで処理しなかった対照群の3H−チミジン結合量を
100%と見なす場合、抽出物AまたはBの濃度による3H−チミジン結合量の
相対値を示す。図2から理解されるように、単球/マクロファージでのIL−6
の生産は、エス.テトランドラ エス.ムーアの抽出物AまたはBの濃度に依存
して阻害され、エス.テトランドラ エス.ムーアの抽出物AまたはBの濃度が
10μg/mlである時50%阻害された。
実施例2:エス.テトランドラ エス.ムーアの抽出物によるラット肺マクロ ファージでのIL−6生産の阻害
ラットをケタミンで麻酔し、気管支に滅菌された細い管を挿入し、30ml注射
器を使ってRPMI1640培地10mlを3回繰り返して注入および吸入するこ
とによって肺マクロファージを得た。得られた細胞を400xgで5分間遠心分
離し、10%FBSを含むRPMI1640培地50mlに懸濁させた後、37℃
で2時間培養して培養プレートに付着させた。プレートをPBSで2回洗浄して
肺胞リンパ球(浮き上がる細胞)を除いて肺マクロファージを得た。
肺マクロファージを24−ウェル培養プレートのウェルに2x105細胞/ウ
ェルの量で入れ、シリカ100μg/mlおよび抽出物A10μg/mlで3日間処理
した。培養液を遠心分離して上澄液を得た後、これをPBSに対して透析した。
透析液中のIL−6活性度は実施例1と同様の方法によってIL−6依存性B9
ハイブリドーマ菌株を用いて測定した。
その結果、ラット肺マクロファージでのIL−6の生産も抽出物Aによって阻
害されることが観察された(図3)。図3で、培地は何らの処理もしなかった対
照群、Siはシリカで刺激した試料、そしてSi+EXTはシリカで刺激し、抽出
物Aで処理した試料をそれぞれ示す。
実施例3:抽出物CによるIL−6生産の阻害
実験的な硅肺症モデルを作るために、処理群当り各5匹のスプラグ−ダウリー
ラット(体重約150g)の気管支をむき出しにした後、0.5mlのPBSに溶
解させたシリカ500mgを注入した。
シリカを注入してから1週間後、前記ラットに一週間に2回ずつ17週間抽出
物C40mgを経口投与するか、またはマウスIL−6抗体(MIL−6Ab、Im
munex,Seatle,U.S.A.)250μgを静脈内にまたは腹腔内に注射した。実施例1
と同様な方法で、血清でのIL−6活性度(図4)および参照例1の2で得られ
た肺繊維芽細胞培養液でのIL−6活性度(図5)を測定した。
図4および図5から分かるように、IL−6の活性はいずれの場合でも抽出物
Cによって阻害され、これはエス.テトランドラ エス.ムーアの抽出物が動物
けい肺症モデルに対して阻害効果を有することを示す。図4および図5で、正常
はPBSで処理した対照群、siはシリカで刺激した試料、si+EXT.Cはシリ
カで刺激した後抽出物Cで処理した試料、si+MIL−6はシリカで刺激し後M
IL−6Abで処理した試料をそれぞれ示す。
実施例4:エス.テトランドラ エス.ムーアの抽出物によるIL−6遺伝子 発現の阻害
エス.テトランドラ エス.ムーアの抽出物がIL−6遺伝子の発現を阻害す
るかの可否を確認するため、IL−6の過剰生産によって惹起された慢性関節リ
ウマチ患者から得られた滑膜細胞(Hirano,T.,et al.,Eur.J.Immunol.,18,1797(
1988))に対する前記抽出物の効果を次のように測定した。
参照例1(3)で分離した滑膜細胞を培養プレートのウェル6個に1.5x1
06細胞/ウェルの量で入れ、5%CO2下で24時間培養してウェルに付着させ
た。抽出物Alμg/mlまたは10μg/mlをウェルに加え、プレートを37℃で
5%CO2下で3日間培養した。
培養が終わると、培養液を遠心分離して上澄液を除き、沈殿した細胞をPBS
で洗浄した後、変性溶液(4M チオシアン酸グアニジニウム、25mMクエン
酸ナトリウム、pH7.0、0.1M2−メルカプトエタノール、0.5%サルコシ
ン)500μlをウェルに入れ、ピペッティングして破砕した。この溶液を管に
移し、2Mクエン酸ナトリウム(pH4.0)50μlおよび水−飽和性フェノー
ル(water-saturated phenol)500μlを加えてよく混合した。その後、2倍
容量のクロロホルムを混合物に加え、生成した混合物を氷上に10分間放置した
後12,000rpmで遠心分離して上澄液を得た。上澄液にイソプロピルアルコー
ル1mlを入れて−20℃で2時間放置した後、12,000rpmで20分間遠心分
離して沈殿したペレットを得た。ペレットを70%メタノールで洗浄し、乾燥し
た後、0.1%ジエチルピロカボネート水溶液20μlに最終濃度10μg/mlに
なるように溶解させた。
このようにして得られたRNAから単鎖cDNAを合成するために、M−ML
V逆転写酵素(Promega,U.S.A.)を用いて次のように逆転写反応を行った。反応
管に5倍濃度の緩衝液(250mMトリス−HCl、pH8.3、375mM K
Cl、15mM MgCl2、50mM DTT)5μl、5μM dNTP混合物(
dATP、dCTP、dGTPおよびdTTPがそれぞれ5μM)2μl、プラ
イマーNotI−dT(5′−dCGCCGGCG(T)18−3′)1μl(0.
2μg)、蒸留水1μl、前記細胞RNA10μlおよびM−MLV逆転写酵素(P
romega,U.S.A.)1μl(200U)を入れ、生成した溶液をよく混合した後、4
2℃で30分間反応させた。この溶液を90℃で5分間加熱して反応を終えた。
前記生成溶液を用いてcDNAを増幅するために、ポリメラーゼ連鎖反応(P
CR)を行った。
前記逆転写反応生成溶液20μl、10倍濃度のPCR緩衝液(100mM ト
リス−HCl,pH8.3、400mM KCl,10mM DTT,15mM MgC
l2,5μg/mlBSA)8μl、5′−末端プライマー(5′−ATGAACTC
CTTCTCCACAAGCGC−3′)1μl(20pmol)、3′−末端プラ
イマー(5′−GAAGAGCCCTCAGGCTGGACTG−3′)1μl
(20pmol)、蒸留水69μlおよびTaq DNAポリメラーゼ(promega,U.S
.
A.)1μl(2.5U)をよく混合し、混合物を95℃で5分間処理して他の望ま
しくない酵素を阻害した。95℃で1.5分;55℃で1分;72℃で1.5分で
構成された温度サイクルを30回繰り返すことによってPCRを行い、反応混合
物を最後に95℃で1.5分;55℃で1分;および72℃で5分間反応させた
。
PCR産物10μlを1.0%アガロースゲルで100Vで30分間電気泳動さ
せた。ゲルEtBr溶液で10分間染色し、蒸留水で洗浄した後、写真を取った
(図6)。図6から分かるように、対照群として用いられた一定に発現されるア
デニンホスホリボシル転移酵素(APRT)RNAの発現は、抽出物Bの影響を
受けないが、IL−6RNAの発現は抽出物Bの濃度10μg/mlで著しく阻害
される。この結果はエス.テトランドラ エス.ムーアの抽出物がIL−6遺伝
子の発現を阻害できるということを示す。図6で、M列は標準DNAサイズマー
カーで、10、1および0は抽出物Bの濃度(μg/ml)を示す。
実施例5:エス.テトランドラ エス.ムーアの抽出物による滑膜細胞の増殖 阻害
参照例1(4)と同様の手順によって、ヒト単球/マクロファージを0.1な
いし100μg/ml範囲の濃度で抽出物Aで処理して培養した。培養液をPBS
に対して透析し、透析液を滑膜細胞1x104細胞に加えた後、細胞を5日間培
養した。培養液に3H−チミジンを加えた後、細胞を4時間さらに培養し、液体
シンチレーション計数器を使って細胞に結合された3H−チミジンの量を測定し
た(図7)。図7で分かるように、抽出物Aは10μg/mlの濃度で滑膜細胞の
増殖を著しく阻害した。
実施例6:エス.テトランドラ エス.ムーアの抽出物によるコラーゲン合成 の阻害
IL−6は繊維化を引き起こすサイトカインとして知られており、ラット繊維
芽細胞でコラーゲン合成を誘導する(Kang,H.S.et al.,Korean J.Immunol.,14,1
93(1992))。IL−6の前記作用を阻害するエス.テトランドラ エス.ムーア
の抽出物の能力を確認するため、ラット肺繊維芽細胞および肺組織でのコラーゲ
ン合成に対する抽出物の阻害効果を測定した。ラット肺繊維芽細胞の培養液中に
生成したコラーゲンの量は間接ELISA方法で測定し、肺組織の培養液中に生
成したコラーゲンの量はヒドロキシプロリンの濃度を測定すると共に内部対照群
の標準曲線を用いてコラーゲンの量を計算することによって決定した。
ラット肺繊維芽細胞培養液中に合成されたコラーゲンの量を測定するため、内
部対照群としてコラーゲン(Sigma、Type I)をペプシン1mg/mlを含む1M酢
酸に溶かし、この溶液をコーティング緩衝液(0.05M炭酸塩、pH9.6)で
1μgないし16pg範囲の濃度で5倍ずつ連続希釈した。希釈した溶液を平底マ
イクロタイタープレート(Dynatech,Cantilly,VA,U.S.A.,Immulon2)のウェルに
100μl/ウェルの量で加えた。
一方、参照例1(2)で得たラット肺繊維芽細胞の培養上澄液1mlをスピード
バック乾燥器(Savant,Hicksville,NY,U.S.A.)を使って10ないし20倍濃縮
し、コーティング緩衝液(0.1M NaHCO3,0.02% NaN3;Na2C
O3でpHを9.6に調整)100μlに溶かし、この溶液を100μl/ウェルの
量でウェルに加えた後、4℃で一晩コーティングした。
プレートを洗浄緩衝液(PBS,0.05ツイン20、pH7.4)で3回洗浄
し、1%牛血清アルブミン(BSA,Sigma)を100μl/ウェルの量でウェル
に加えた。プレートを室温で2時間培養してコーティングされない部分をブロッ
クした。プレートを前記と同じ緩衝液で4回洗浄し、希釈緩衝液(0.05M
Tris−HCl,1mM MgCl2・6H2O、0.15M NaCl,0.02% Na
N3、1%BSA,0.05%ツイン20、pH8.1)で1、000倍希釈したア
ルカリ性ホスファターぜ結合されたウサギ抗−ヤギIgG(Cappel,Durham,NC,U.
S.A.)を100μl/ウェルの量でウェルに加えた。
プレートを37℃で2時間培養した後、前記と同じ緩衝液で3回洗浄した。基
質緩衝液(0.05M NaHCO3、10mM MgCl2・6H2O、pH9.8)で
1mg/mlの濃度に希釈したp−ニトロフェニルリン酸100μl/ウェルをウェ
ルに加え、ELISAリーダーを使って405nmで培養液のO.D.を測定した。
生産されたコラーゲンの量はこのO.D.値と内部対照群のO.D.値から計算した
。その結果、抽出物Aの濃度10μl/mlで37℃で1時間前処理し、シリカ1
00μl/mlで48時間処理したラット肺繊維芽細胞の培養液では産生されたコ
ラーゲンの量が著しく減少したことが観察された(図8)。図8で、si+PBS
はシリカで刺激した試料を示し、si+EXT.Aは抽出物Aで処理してシリカで
刺激した試料を示す。
また、実施例3の手順によって、ラット肺組織で合成されたコラーゲンの量を
測定するために、ラットの気管支をむき出しにした後、シリカ500mgを注入し
、このラットに週2回ずつ17週間1%DMSOに溶かした抽出物C40mgまた
は1%DMSOのみを経口投与させた後、次のようにヒドロキシプロリンの量を
測定した。
ラット肺組織0.1ないし0.2gをPBS1mlと混合した後、パイレックス管
(Corning,Rochester,NY,U.S.A.)の中で粉砕した。生成した組織抽出物を超音
波機(Heat system,W−380)で破砕し、ここにヒドロクロン酸(hydrochron
ic acid)1mlを加え、この混合物を120℃乾燥器で一晩乾燥した。生成物を
冷凍器で冷凍し、凍結乾燥器(Labconco)で凍結乾燥し、蒸留水1mlを加えて完
全に溶解させた。生成した溶液50μlを微量遠心分離管に加え、そこに蒸留水
50μlを加えて溶液を希釈した。内部対照群として、トランス−γ−ヒドロキ
シ−L−プロリン(Sigma)を20μgないし150pg範囲の濃度に希釈し、それ
ぞれの希釈液100μlずつを微量遠心分離管に加えた。
クロラミン−T(ナトリウムN−クロロ−P−トルエンスルホンアミド)1.
41gをn−プロパノール10mlと蒸留水10mlに溶解させて製造した溶液0.
9mlを前記管に加え、これを室温で20分間放置した後、この混合液に、p−ジ
メチルアミノベンズアルデヒド15gをn−プロパノール62mlに溶解させてか
ら60%過塩素酸26mlを総容量100mlになるように加えて作ったアルデヒド
/過塩素酸溶液1mlを加えてよく混合した。微量遠心分離管を65℃水浴に15
分間入れて発色させ、650nmで試料のO.D.を測定し、内部対照群の標準曲線
を用いて試料中のヒドロキシプロリン量を計算した。
図9から分かるように、正常なラット肺組織(正常)で生産されたコラーゲン
の量を100%と見なす場合、シリカのみで処理したか(si)、シリカおよびジ
メチルスルホキシドで処理した(si+DMSO)ラット肺組織で合成されたコラ
ーゲンの量は著しく多かったが、シリカ、DMSOおよび抽出物Cで処理したラ
ット肺組織(si+EXT.C)では、産生したコラーゲンの量が50%減少した
。この結果はエス.テトランドラ エス.ムーアの抽出物が抗繊維化活性を有す
ることを示す。
実施例7:エス.テトランドラ エス.ムーアの抽出物による反応性酸素類の 生産阻害
炎症反応は、種々の刺激剤で刺激された免疫細胞からIL−6のような種々の
サイトカインの分泌;前記サイトカインで刺激された他の免疫細胞によるホスホ
リパーゼA2、リソソーム酵素、反応性酸素類などの生産;および前記生成物に
よって誘発される組織の損傷を含む一連の反応として知られている(Pruzanski,
W.and Vadas,P.,Immunol.Today,12,143(1991))。炎症反応を遮断するエス.テ
トランドラ エス.ムーアの抽出物の能力は、反応性酸素類、たとえば、H2O2
およびO2 -の生産に対する阻害活性を測定することによって評価した。
H2O2の量は、96−ウェルマイクロプレートを使った微量検定法によって次
のように測定した。RPMI1640培地の入っている各ウェルに好中球5x1
05細胞を加え、セイヨウワサビペルオキシダーゼ(500μg/ml,type II,Si
gma)25μlおよびフェノールレッド(1mg/ml)75μlを各ウェル加えた。
その後、細胞を抽出物A10、20、および50μg/mlで1時間処理し、10- 7
Mホルボールミリスタートアセタート(PMA)で刺激した後、37℃で60
分間反応させた。培養が終わった後、3M NaOHを25μl/ウェルの量でウ
ェルに加えて反応を中止し、ELISAリーダー(Dynatech Lab.Inc.)を使っ
て620nmでO.D.を測定してフェノールの酸化による色の変化を測定し、H2
O2
の量は希釈したH2O2(Sigma)を用いて作成した標準曲線によって決定した。
生産されたO2 -の量を測定するため、RPMI1640培地に1x106細胞
/800μlの濃度で懸濁した好中球を24−ウェルプレートのウェルの一部に
加え、空いているウェルにはスーパーオキシドジスムターゼ(SOD,Sigma)
10μg/mlを加えた。プレートを37℃で2分間放置し、チトクロムC(3mg
/ml,Sigma)を100μl/ウェルの濃度でウェルに加えた。細胞を10、20
および50μg/mlの濃度の抽出物Aで1時間処理し、刺激剤として10-7M
PMAを導入して37℃で20分間反応させた。1mM N−エチルマレイミド
(Sigma)をウェルに加えて反応を中止させ、培養液を1,600xgで10分間
遠心分離して上澄液を得た。チトクロムCの還元によって惹起された上澄液の色
の変化をUV−Vis分光光度計(Kontron Instrument,Milano,Italy)を使って
550nmで測定した。生成したO2 -の量をチトクロムCの吸光計数(E550nm
=1.83x104mM-1cm-1)を使って20分間1x106細胞でチトクロムCの
還元を阻害できるSODの濃度で表した。表Iから分かるように、抽出物Aは濃
度50μg/mlgでH2O2生産を50%阻害し、O2 -の生産を25%阻害する。
この結果は抽出物Aが炎症反応に対して強い阻害活性を有することを示す。
一方、濃度10μg/mlの抽出物Aを用いて37℃で1時間処理した後、シリ
カ100μg/mlで刺激したヒト単球/マクロファージ5x105細胞によって生
産されたH2O2およびO2 -の量を測定するために前記と同様な手順を繰り返した
。図10から分かるように、シリカで刺激し、抽出物Aで処理した単球/マクロ
ファージ(si+EXT.A)は、シリカのみで刺激した対照群(si+MED)に
比べてH2O2およびO2 -の量が著しく減少した。
実施例8:エス.テトランドラ エス.ムーア抽出物による肝硬変の阻害
肝硬変(肝硬化)は肝全体の繊維化、繊維性中隔による肝実質の完全な破壊お
よび再生性結節の形成を特徴とする。肝硬変は大部分慢性肝炎または慢性アルコ
ール中毒から由来するが、その正確な原因は明らかではない。肝硬変患者の場合
、炎症および繊維化に関与するIL−6のようなサイトカインの量が増大した状
態であり、したがって、エス.テトランドラ エス.ムーアの抽出物による肝硬
変の阻害はIL−6に対する阻害活性によって測定できる。
4週齢の雄スプラグダウリーラット(Nakataukasa,H.,et al.,J.Clin.Invest.
,85,1833-1843(1990))に実験的に肝硬変を誘発させるために、ラットに1.0ml
/100g体重のCCl4溶液(50%CCl4+50%とうもろこし油)を1週
間に2回腹腔内に注射し、抽出物A、B、CまたはDをそれぞれ0.2mlずつ1
週間に2回CCl4の注射時に経口投与した。試験開始13週後、各ラットをエ
ーテルで麻酔し、心臓から血液試料を取って血清グルタミン酸−オキサロ酢酸ト
ランスアミナーゼ(sGOT)および血清グルタミン酸−ピルビン酸トランスア
ミナーゼ(sGPT)値を測定した(図11)。
図11から分かるように、対照群として用いられたCCl4、DMSOおよびP
BSで処理したラットから得られた血液試料と比べる際、抽出物AまたはCで処
理したラットから得られた血液試料のsGOT値は減少しなかったが、抽出物D
またはBで処理したラットから得られた試料の場合、それぞれ20%および40
%減少する。また、抽出物Bで処理したラットから得られた血液試料のsGPT
は60%以上減少した。
前記ラットから分離された肝の病理組織学的検査のため、肝を10%中性ホル
マリン水溶液に固定し、4mm厚さに展開した後、パラフィンに埋め込んだ。埋め
込まれた組織を5mm厚さに切断し、ヘマトキシリンエオシンおよびマソン3色染
色法で染色した後、顕微鏡で観察した(図12)。
図12から理解されるように、正常の肝(A)に比べてCCl4のみを投与した
ラットの肝(B)においては厚くなった繊維性バンドを有する肝小葉の結節形成
が明らかであった。CCl4および抽出物Bを投与したラットの肝(F)、CCl4
および抽出物Cを投与したラットの肝(E)およびCCl4および抽出物Dを投与
したラットの肝(D)では肝硬変の徴候が見られたが、肝小葉の結節を取り囲ん
でいる繊維性バンドはCCl4のみで処理したラットから得られた肝に比べて減少
した。CCl4および抽出物Aを投与したラットの肝(C)では前記D、Eおよび
Fのものと比べて肝硬変に対する阻害効果が低かった。
下記製剤例は例示するのみで、いずれの点でも本発明の範囲を制限するもので
はない。
