Claims (1)
- 【特許請求の範囲】 1.生物学的試料中のHIV-I交差反応性乳癌関連抗原を検出する方法であって、 該抗原を含むと疑われる生物学的試料を、 以下のファミリーの細胞膜タンパク質、p160、p80、p45、およびp24、 ならびに以下のファミリーのクロマチンタンパク質、p24、 の全てのメンバーを認識する抗体に曝す工程、および 該抗体と該タンパク質とで形成される免疫複合体の存在を検出する工程であっ て、該細胞膜タンパク質およびクロマチンタンパク質が健常な生物学的試料中で 検出され得ない工程、を包含し、 ここで、該方法が該生物学的試料がHIV-I抗原を含まないことを保証するコン トロールを包含する、方法。 2.HIV-I交差反応性の婦人科の癌の関連抗原を検出するための請求項1に記載 の方法であって、 該抗原を含むと疑われる生物学的試料を、 以下のファミリーの細胞膜タンパク質、p120、p41、およびp24、 ならびに以下のファミリーのクロマチンタンパク質、p24、 の全てのメンバーを認識する抗体に曝す工程、および 該抗体と該タンパク質とで形成される免疫複合体の存在を検出する工程であっ て、該細胞膜タンパク質およびクロマチンタンパク質が健常な生物学的試料中で 検出され得ない工程、を包含する方法。 3.前記抗体が標識されている、請求項1または2に記載の方法。 4.前記標識が、酵素、蛍光色素、放射性同位元素、および発光物質からなる群 より選択される、請求項3に記載の方法。 5.前記検出の工程が、酵素反応、蛍光、発光、または放射活性によるものであ る、請求項1または2に記載の方法。 6.前記生物学的試料が、体分泌物、体液、および組織標本からなる群より選択 される、請求項1または2に記載の方法。 7.前記生物学的試料が、前記抗体に曝す前に、ゲル電気泳動により分離される 、請求項1または2に記載の方法。 8.前記抗体がモノクローナル抗体5023である、請求項1または2に記載の方法 。 9.卵巣癌、子宮頚癌、子宮内膜癌、および外陰癌からなる群より選択される婦 人科の癌ならびに乳癌の治療のための、患者への薬学的に有効な投与形態でのモ ノクローナル抗体5023の使用。 10.前記モノクローナル抗体が放射活性リガンドまたは細胞障害性リガンドと 結合されている、請求項9の使用。 11.乳癌細胞および婦人科の癌の細胞の増殖を阻害するHIV交差反応性抗原に 結合するモノクローナル抗体5023を含む抗原−抗体複合体であって、 HIV抗原自体ではないがHIV交差反応性抗原である抗原に結合するモノクローナ ル抗体5023を含む、抗原−抗体複合体。 12.前記抗原が、以下の細胞膜タンパク質、p160、p80、p45、またはp24、あ るいは、クロマチンタンパク質、p24、の1つである、請求項11に記載の複合 体。 13.固定化形態である、請求項11に記載の複合体。 14.ニトロセルロース膜上に固定化されている、請求項12に記載の複合体。 15.乳癌および婦人科の癌を診断する方法であって、該癌と関連する遺伝配列 を含むと疑われる生物学的試料を、HIV-Iに相同なレトロウイルス配列の存在に ついてアッセイする工程であって、ここで、細胞膜タンパク質およびクロマチン タンパク質が健常な生物学的試料中に検出され得ない、工程を包含し、該方法が 、調査中の該生物学的試料がHIV-I抗原自体を含まないことを保証するコントロ ールを包含する、方法。 16.前記配列がPCR増幅により決定される、請求項15に記載の方法。 17.前記PCR増幅がHIV-I配列由来のプライマーにより生じる、請求項15に記 載の方法。 18.前記プライマーが、HIV-I gp120の可変領域、またはgp41から選択される 、請求項17に記載の方法。 19.前記プライマーが、配列番号2および3ならびに配列番号4および5から なる群より選択される、請求項18に記載の方法。 20.乳癌細胞および婦人科の癌の細胞の増殖を阻害するためのオリゴヌクレオ チドの使用であって、HIV-I交差反応性の癌抗原をコードする癌細胞のオリゴヌ クレオチド配列を同定する工程、該癌細胞配列に対するアンチセンスオリゴヌク レオチドを設計する工程、および該細胞を該オリゴヌクレオチドに曝す工程、を 包含し、ここで、細胞膜タンパク質およびクロマチンタンパク質が健常な生物学 的試料中に検出され得ない、方法。 21.前記オリゴヌクレオチドが、配列番号6、配列番号7、および配列番号8 からなる群より選択される、請求項20に記載の方法。 22.乳癌細胞および婦人科の癌の細胞の増殖を阻害する組成物であって、HIV- I交差反応性の抗原をコードする癌細胞配列のアンチセンスである、配列番号6 、配列番号7、および配列番号8からなる群より選択されるオリゴヌクレオチド を含む、組成物。
