Claims (1)
- 【特許請求の範囲】 1. CMV病の疑いのあるヒトの血液標本中にヒトサイトメガロウイルスの後 期構造蛋白質をコードするmRNAの存在を検出することを特徴とする症状のあ るCMV病の診断方法であって、以下の段階、 −標的配列と特異的に反応できるプライマー対と適切な増幅試薬を使用して上記 mRNA内の標的配列を増幅すること、 −増幅した核酸を含む場合がある標本と増幅した配列の一部に相補的な配列を有 する標識核酸プローブを反応させること、 −増幅した配列とプローブの間で形成されるハイブリッドを検出すること、 を含む方法。 2. 上記mRNAがpp67のmRNAである請求項1に記載の方法。 3. 転写をベースとした増幅技術を使用して、mRNAを増幅する請求項1又 は2に記載の方法。 4. 上記増幅技術がNASBAである請求項3に記載の方法。 5. HCMVのpp67をコードする核酸配列の一部に対応 するオリゴヌクレオチドであって、10−35ヌクレオチドの長さであり、以下 の配列からなる群から選択される配列の少なくとも10ヌクレオチドの断片を含 むオリゴヌクレオチド。 6. プロモーター配列に結合した請求項5に記載のオリゴヌクレオチド。 7. HCMVのpp67配列内に位置する標的配列の増幅用プライマーセット であって、 以下の核酸配列から本質的になる1つのプライマーと 以下の核酸配列から本質的になる第2のプライマー を含むプライマーセツト。 8. HCMVのpp67配列内に位置する標的配列の増幅用プライマーセット であって、 以下の核酸配列から本質的になる1つのプライマーと 以下の核酸配列から本質的になる第2のプライマー を含むプライマーセット。 9. 請求項2の方法におけるプライマーとしての、請求項5に記載のオリゴヌ クレオチドのいずれかの使用。 10. 請求項2の方法におけるプローブとしての、検出可能な標識で標識され た以下の配列から本質的になるオリゴヌクレオチドの使用。 11. 請求項7及び/又は8に記載の、プライマーの1つ又は複数のセット、 及び プライマーの上記セットにより規定される増幅した核酸配列の少なくとも一部に 実質的に相補的な核酸配列を含む、検出可能な標識で標識されたオリゴヌクレオ チド、 を含むHCMV病の診断用テストキット。
