改良された抗インフルエンザ活性を有する修飾オリゴヌクレオチド
本願は、1992年7月2日出願の出願番号07/909,069の一部継続出願である。同
時に1992年7月23日出願の出願番号07/918,239の一部継続出願でもある。また、
1991年5月10日出願の出願番号07/698,568の一部継続出願でもある。
発明の背景産業上の利用分野
本発明は、アンチセンスオリゴヌクレオチドに関する。より具体的には、本発
明はインフルエンザウイルスの複製または増殖を阻害する能力のある修飾オリゴ
ヌクレオチドに関する。関連技術の要約
インフルエンザAウイルスは、RNAのマイナスストランドから成るセグメントゲ
ノムを有する膜封入ウイルスである。A型およびB型では一本鎖RNAの8つのセグ
メントに、またC型では7つにセグメントに10個のインフルエンザウイルス遺伝
子が存在する。セグメントの大きさは様々で(長さが890〜2341ヌクレオチド)
、それぞれが異なるmRNA合成の鋳型となっている。インフルエンザウイルスビ
リオンには、これらの鋳型からmRNAを合成するのに必要なウイルス特異なRNAポ
リメラーゼがあり、この特異なポリメラーゼがないと、インフルエンザウイルス
RNAのマイナスストランドは感染力を持たない。mRNAの転写は、インフルエンザ
ウイルスmRNAポリメラーゼが細胞mRNAまたはmRNA前駆体の5'末端から12〜15ヌク
レオチドを取ったときに始まり、借用したオリゴヌクレオチドをプライマーとし
て使用する。一般に、このプロセスにより作られたmRNAは1つの蛋白しかコード
しない。M RNAおよびNS RNAは、スプライシングされたmRNAもコードし、その結
果これらの2つのセグメントのそれそれについて異なる2種類の蛋白が生成され
る。
インフルエンザウイルスはヒトおよび動物(たとえばブタ、鳥類、ウマ)に感
染し、急性呼吸器疾患の原因になる。インフルエンザウイルスに有効なワクチン
を生み出そうとこれまで数多くの試みが成されてきた。しかし、特に長期的に見
た場合には、完璧に成功した例はまったくない。これは、少なくとも一部はイン
フルエンザウイルスのゲノムがセグメントゲノムであるという特徴に起因し、こ
の特徴のためにセグメントを再構築することによって、多数の形のウイルスが存
在できる。たとえば、動物とヒトのインフルエンザウイルスの間でRNAセグメン
トが交換されうることが示唆されているが、これによって両集団に新たな抗原サ
ブタイプが導入されることになる。したがって、長期にわたるワクチン接種の方
法は、
新しいサブタイプが発生する(抗原「シフト」が起こる)ために失敗している。
さらに、ウイルスの表面蛋白の血球凝集素(ヘマグルチニン)およびノイラミニ
ダーゼは、常に抗原がわずかずつ変化している(抗原「ドリフト」)。このよう
にきわめて多様であるという事実から、特定のインフルエンザウイルスに対して
開発された特異的免疫が新しい変異株に対する保護に役立たない理由が説明でき
る。そこで、これに代わる抗ウイルス戦略が必要となる。B型およびC型のインフ
ルエンザウイルスはA型に比べて臨床上の疾患をもたらすことは少ないが、化学
的抗ウイルス剤はこれらのウイルスが原因で生じる感染を抑制するのに役立つは
ずである。
したがって、インフルエンザウイルスの複製および増殖を阻止する能力のある
アンチセンスオリゴヌクレオチドの開発には大きな関心が寄せられる。このよう
なアンチセンスオリゴヌタレオチドは、インフルエンザウイルスの複数の株に存
在するインフルエンザウイルスの保存領域にハイブリダイズするよう設計できる
ので、様々な株のインフルエンザに対する幅広い防御能を提供できるものとして
有望である。
アグラワルらによる米国特許第5,194,428号では、本明細書でも参考文献とし
て引用しているが、インフルエンザPB1ポリメラーゼ遺伝子にハイブリダイズす
ることによりインフルエンザの複製を阻止する能力のある一定の修飾ヌクレオチ
ド間結合を有するオリゴヌクレオチドを開
示している。カウサートら(WO92/03454(1992年))は、インフルエンザポリメ
ラーゼ1、2、3、または血球凝集素、核蛋白、ノイラミニダーゼ、マトリックス
蛋白、あるいは非構造的蛋白遺伝子に、もしくは様々なスプライシング結合部位
やパッケージング配列にハイブリダイズすることによって、インフルエンザウイ
ルスの増殖を阻止するアンチセンスオリゴヌクレオチドを開示している。
ペダーソンらによる米国特許第5,149,797号および第5,XXX,XXX号(出願番号0
7/839,472、1992年12月24日)は、本明細でも参考として引用するが、リボヌク
レアーゼH不活性化セグメントに隣接してリボヌクレアーゼH活性化セグメント
を有するキメラ混合リン酸バックボーンのオリゴヌクレオチドを開示している。
したがって、アンチセンスオリゴヌクレオチドは抗インフルエンザ治療薬とし
て有望と思われる。しかし、このような薬剤のいずれを使用する場合でも、イン
フルエンザウイルスの複製または増殖阻止効力がさらに大きい改良された薬剤が
今後もなお必要とされる。このような改良されたアンチセンスオリゴヌクレオチ
ドは、抗インフルエンザ治療薬として使用するだけでなく、インフルエンザウイ
ルスゲノムのどの領域が幅広い交差株阻害のための最もよい候補となるかを調べ
るのにも有用であろう。
発明の概要
本発明は、これまでに知られているオリゴヌクレオチドに比べてインフルエン
ザウイルスの複製または増殖の阻止能がより大きな修飾オリゴヌクレオチドを提
供する。これらの修飾オリゴヌクレオチドは、細胞内のインフルエンザウイルス
核酸にハイブリダイズするためにそのようなインフルエンザウイルスの必須核酸
に対して十分に相補的な核酸配列を有することを特徴とする。好ましい実施例で
は、必須の核酸はPB1、2、3のポリメラーゼ遺伝子、または血球凝集素、核蛋白
、ノイラミニダーゼ、マトリックス蛋白、あるいはインフルエンザの非構造的蛋
白遺伝子、もしくは様々なスプライシング結合部またはパッケージング配列の一
部である。
様々な実施例で、本発明による修飾オリゴヌクレオチドは1ないし複数のタイ
プの修飾ヌクレオチド間結合を有する。好ましい実施例では、修飾ヌクレオチド
間結合の少なくとも一部は、ホスホロチオエート結合またはホスホロジチオエー
ト結合である。特定の好ましい実施例では、ホスホロチオエートヌクレオチド間
結合を、他の修飾結合、すなわちホスホジエステルでない結合を有する修飾オリ
ゴヌクレオチド中に含んでいる。これらのその他の修飾ヌクレオチド間結合は、
アルキルホスホネート結合またはアルキルホスホノチオエートであることが望ま
しい。これらの修飾ヌクレオチド間結合は、オリゴヌクレオチドの一端もしくは
両端、あるいはその近傍に
存在することが最も望ましい。
本発明に基づく修飾オリゴヌクレオチドのさらに別の好ましい実施例では、1
ないし複数の修飾ヌクレオシドを有するオリゴヌクレオチドに、ホスホロチオエ
ートまたはホスホロジチオエートのヌクレオチド間結合が存在する。この修飾ヌ
クレオシドは2'−Oアルキルヌクレオシドであることが好ましい。修飾ヌクレオ
シドの少なくとも一部がオリゴヌクレオチドの一端もしくは両端、あるいはその
近傍に存在するのが最も好ましい。
本発明に基づくさらに別の好ましい実施例では、一端もしくは両端にエキソヌ
クレアーゼ抵抗性賦与キャップ構造を有するオリゴヌクレオチドに、ホスホロチ
オエートまたはホスホロジチオエートのヌクレオチド間結合が存在する。このキ
ャップ構造は少なくとも分子の3'末端に存在するのが好ましい。このキャップ構
造は、本発明に基づく修飾オリゴヌクレオチドのあらゆる実施例の一端もしくは
両端にも存在することがあり、少なくともオリゴヌクレオチドの3'末端に存在す
るのが好ましい。
本発明に基づく修飾オリゴヌクレオチドのさらに別の好ましい実施例では、一
端もしくは両端あるいはその近傍にヌクレオチドによる自己相補的領域を含有す
る自己安定型のオリゴヌクレオチドで、ホスホロチオエートまたはホスホロジチ
オエートのヌクレオチド間結合が存在し、できれば少なくとも3'末端もしくはそ
の近傍に存在するのが好ましく、それによりオリゴヌクレオチドはヘ
アピン状構造を形成する。
本発明に基づくこれらの修飾オリゴヌクレオチドは、インフルエンザの複製ま
たは増殖を阻止するのに、既存のオリゴヌクレオチドまたは同一遺伝子にハイブ
リダイズする修飾オリゴヌクレオチドのいずれよりも大きな効力をもたらす。本
発明に基づくこれらの修飾オリゴヌクレオチドは、いずれもオリゴヌクレオチド
の糖または塩基に対する他の修飾も任意選択として含めることができ、また、や
はり任意選択で、ホスホロチオエート、ホスホロジチオエート、アルキルホスホ
ネートまたはアルキルホスホノチオエートの各結合以外の追加のヌクレオチド間
結合を有するようにすることができる。

好ましい実施例の詳細な説明
本発明は、抗インフルエンザ活性を有するアンチセンスオリゴヌクレオチドに
関する。インフルエンザに対するin vivo活性を有する修飾オリゴヌクレオチド
のことを、ここでは抗インフルエンザ修飾オリゴヌクレオチドという。本発明は
、これまでに知られているオリゴヌクレオチドまたは同一遺伝子にハイブリダイ
ズする修飾オリゴヌクレオチドに比べてインフルエンザウイルスの複製または増
殖の阻止能がより大きな修飾オリゴヌクレオチドを提供する。本発明に基づく修
飾オリゴヌクレオチドは、以下の好ましい実施例それぞれでさらに詳しく述べる
ような特異的な好ましい特徴を持っている。これらの特徴に加えて、本発明に基
づく修飾オリゴヌクレオチドは、
任意選択で追加のリボヌクレオチド、2'-置換リボヌクレオチド、またはデオキ
シリボヌクレオチドモノマー、あるいはそのいずれも持つことができ、これらの
いずれもこれまでの技術で知られているヌクレオチド間結合のいずれをも含むこ
とができ、5'-3'結合を介して結合することができる。このような修飾オリゴヌ
クレオチドは、任意選択でホスホジエステル、ホスホトリエステル、ホスホルア
ミデート、シロキサン、炭酸エステル、カルボキシメチルエステル、アセトアミ
デート、カルバメート、チオエーテル、架橋ホスホルアミデート、架橋メチレン
ホスホネート、架橋ホスホロチオエートまたはスルホンヌクレオチド間結合のい
ずれかまたはすべてを有することが好ましい。当業者であれば、これらのヌクレ
オチド間結合のいずれかを有するオリゴヌクレオチドの合成はこの分野に精通し
た者はよく知っていることを認めるであろうし、これについてはウルマンとペイ
マンによるChemical Reviews 90巻543〜584頁(1990年)およびシュナイダーと
バンナーによるTetrahedron Lett.31巻335頁(1990年)の文献に示されている
。本発明に基づく修飾オリゴヌクレオチドは、全部で約6から約100のモノマー
を含有することが望ましい。このような修飾オリゴヌクレオチドは、任意選択で
修飾核酸塩基または糖、あるいはその両者を含有することができ、またジアミン
、コレステリルまたはその他の親油性基などの追加置換基を持つことができる。
本発明に基づく修飾オリゴヌクレオチドの様々な好ましい実施例を、以下の表
Iに示す。これらの実施例はいずれもインフルエンザPB1ポリメラーゼ遺伝子の
同一領域からのヌクレオチド配列を有するが、当業者であれば、インフルエンザ
ウイルスの他の必須核酸配列と相補的なヌクレオチド配列を有するオリゴヌクレ
オチドの抗インフルエンザ活性は、そのオリゴヌクレオチドに本発明に基づく修
飾オリゴヌクレオチドの好ましい実施例の構造的特性を組み込むことによってさ
らに強化することも可能であることがわかるだろう。本発明の目的では、相補的
とは、生理学的条件下で必須核酸配列にハイブリダイズする配列を有することを
意味する。インフルエンザウイルスの必須核酸配列とは、インフルエンザウイル
スの複製または増殖に必要な核酸配列のことをいう。このような必須核酸配列は
、インフルエンザウイルスの既知のどの株からのものでもよい。たとえば、この
ようなオリゴヌクレオチドは、本明細書でも参考として引用している米国特許第
5,194,428号の表Iに示されているように、インフルエンザPB1ポリメラーゼ遺伝
子(ポリメラーゼ1)からの別の配列を有することもできる。実際、インフルエ
ンザPB1(ポリメラーゼ1)遺伝子のどの配列[SEQ.ID No.1]でも(図1参照
)、本発明に基づく修飾オリゴヌクレオチドの基礎となる。この代わりに、他の
インフルエンザ配列または遺伝子からの配列を使用することもできる(表II参照
)。実際的な問題としては、本発明
に基づく修飾オリゴヌクレオチドの好ましい実施例の構造的特性は、細胞中でイ
ンフルエンザウイルスのいずれかの必須核酸配列とハイブリダイズする核酸配列
を有するどのアンチセンスオリゴヌクレオチドの抗インフルエンザ活性も増強す
るはずである。
本発明に基づく修飾オリゴヌクレオチドの好ましい実施例それそれについて、
以下に個別にさらに詳しく説明する。
最初の好ましい実施例では、本発明に基づく抗インフルエンザ修飾オリゴヌク
レオチドは、ホスホロチオエートまたはホスホロジチオエートのヌクレオチド間
結合によって結合されたヌクレオチドの領域(「ホスホロチオエートまたはホス
ホロジチオエート領域」)を1ないし複数有し、さらにアルキルホスホネートヌ
クレオチド間結合により結合されたヌクレオチドの領域(「アルキルホスホネー
ト領域」)を1ないし複数有する混合バックボーンキメラオリゴヌクレオチドの
形である。この実施例では、少なくとも1つのアルキルホスホネート領域は、オ
リゴヌクレオチドの5'末端または3'末端、あるいはその両者、もしくはそれらの
近傍でヌクレオチドを有していることが好ましい。本発明の目的では、「オリゴ
ヌク
レオチドの5'末端または3'末端もしくはその近傍」というときには、オリゴヌク
レオチドの5'末端または3'末端から約5ヌクレオチド以内に少なくとも1個のヌ
クレオチドを有することをいう。アルキルホスホネート領域は、アルキルホスホ
ネート結合により結合された約2から約10の隣接するヌクレオチトから成るの
が好ましい。ホスホロジオエートまたはホスホロジチオエート領域は、ホスホロ
チオエート結合またはホスホロジチオエート結合によって連結される最低3から
最高約100までの隣接するヌクレオチドから成るのが好ましい。本発明のこの
実施例に基づくこのインフルエンザ修飾オリゴヌクレオチドの例を、表IにCMPD
Aとして示す。
本発明のこの実施例に基づく抗インフルエンザ修飾オリゴヌクレオチドは、固
相法によって、あるいはその代わりにホスホロチオエート領域についてはH−ホ
スホネート化学品および硫黄酸化、アルキルホスホネート領域についてはアルキ
ルホスホンアミデート化学品によって合成できる。好ましいH−ホスホネート法
についてはアグラワルらが米国特許第5,149,798号で明らかにしており、この教
示を参考として本明細書に引用する。アルキルホスホンアミダイト化学品は、ア
グラワルとグッドチャイルドによりTetrahedron Lett.28巻3539-3542頁(1987
年)に述べられているようによく知られた技術である。ホスホロジチオエート含
有オリゴヌタレオチドの合成についても、本明細書でも引用する米国特許第5,15
1,510号に明
らかにされているようによく知られた技術である(その他、たとえばマーシャル
とカルサーズによるScience 259巻1564〜1570頁(1993年)およびそこでの引用
文献などを参照のこと)。
2番目の好ましい実施例では、本発明に基づく抗インフルエンザ修飾オリゴヌ
クレオチドは、ホスホロチオエートまたはホスホロジチオエートのヌクレオチド
間結合によって結合されたヌクレオチドの領域(「ホスホロチオエートまたはホ
スホロジチオエート領域」)を1ないし複数有し、さらにアルキルホスホノチオ
エートまたはアリールホスホノチオエートのヌクレオチド間結合により結合され
たヌクレオチドの領域(「アルキルホスホノチオエート領域」)を1ないし複数
有する混合バックボーンキメラオリゴヌクレオチドの形である。この実施例では
、少なくとも1つのアルキルホスホノチオエート領域は、オリゴヌクレオチドの
5'末端または3'末端、あるいはその両者、もしくはその近傍にヌクレオチドを有
することが好ましい。アルキルホスホノチオエート領域は、アルキルホスホノチ
オエート結合により結合された約2から約10の隣接するヌクレオチドから成る
のが好ましい。ホスホロチオエートまたはホスホロジチオエート領域は、ホスホ
ロチオエート結合またはホスホロジチオエート結合によって接続される最低3か
ら最高約100までの隣接するヌクレオチドから成るのが好ましい。本発明のこ
の実施例に基づくこのインフルエンザ修飾オリゴ
ヌクレオチドの例を、表IにCMPD B、CMPD EおよびCMPD Fとして示す。
本発明のこの実施例に基づく抗インフルエンザ修飾オリゴヌクレオチドは、固
相法によって合成するか、あるいはその代わりに合成すべき領域それそれについ
ての化学物質によって合成する。ホスホロチオエート領域またはホスホロジチオ
エート領域は、最初の実施例について述べたようにして合成する。アルキルホス
ホノチオエート領域は、2またはそれ以上のヌクレオシドをアルキルホスファイ
ト結合によりカップリングさせてから、アルキルホスファイト結合を酸化的にチ
オレート化してアルキルホスホノチオエート結合をもたらすことにより合成する
。この合成手順については、実験例1で詳しく述べる。
3番目の好ましい実施例では、本発明に基づく抗インフルエンザ修飾オリゴヌ
クレオチドは、デオキシリボヌクレオチドの領域(「デオキシリボヌクレオチド
領域」)およびリボヌクレオチドまたは2'-置換リボヌクレオチドの領域(「リ
ボヌクレオチド領域」)を有するハイブリッドオリゴヌクレオチドの形である。
約1からおよそすべてにわたるヌクレオチド間結合がホスホロチオエート結合ま
たはホスホロジチオエート結合であることが好ましい。2'-置換リボヌクレオチ
ドとして望ましいのは、ハロ、アミノ、アルキル、アリールまたはより低級のア
ルキル(1〜6炭素原子)で置換されたリボヌクレオチド、
中でも2'-OMe-リボヌクレオチドである。できれば、リボヌクレオチド領域の少
なくとも一部はオリゴヌクレオチドの5'末端または3'末端、あるいはその両者も
しくはその近傍に存在することが好ましい。さらに、リボヌクレオチド領域は、
それそれ約2から、できれば約4から約100までの隣接するリボヌクレオチド
または2'-置換オリゴヌクレオチドあるいはその両者から成ることが最も好まし
い。デオキシリボヌクレオチド領域は任意選択であり、また存在する場合には約
1から約100の隣接するデオキシリボヌクレオチドを有する事ができる。本発
明のこの実施例に基づく抗インフルエンザ修飾オリゴヌクレオチドの例は、表I
ではCMPD C、CMPD D、CMPD G、CMPD H、CMPD K、CMPD MおよびCMPD Nとして示さ
れる。
本発明のこの実施例に基づく抗インフルエンザ修飾オリゴヌクレオチドは、固
相法によって合成されるのが標準的であり、できればデオキシリボヌクレオチド
領域についてはデオキシヌクレオチドH−ホスホネートを使い、またリボヌクレ
オチド領域についてはリボヌクレオチドまたは2'-置換リボヌクレオチドH−ホ
スホネートを使ったH−ホスホネート法により合成するのが好ましい。
4番目の好ましい実施例では、本発明に基づく抗インフルエンザ修飾オリゴヌ
クレオチドは、5'末端または3'末端あるいはその両者にオリゴヌクレオチドに対
するエキソヌクレーゼ抵抗性を賦与するキャップ構造を有するオリゴヌクレオチ
ドの形である。このような修飾オリゴ
ヌクレオチドは、1からおよそすべてまでの修飾(非ホスホジエステル)ヌクレ
オチド間結合も有することが好ましい。好ましいキャップ構造としては、図4に
示すようなものや、低級アルキル(C1〜C12)基またはアルコール基がある。好
ましい修飾ヌクレオチド間結合としては、ホスホトリエステル、ホスホルアミデ
ート、シロキサン、炭酸エステル、カルボキシメチルエステル、アセトアミデー
ト、カルバメート、チオエーテル、架橋ホスホルアミデート、架橋メチレンホス
ホネート、架橋ホスホロチオエート、スルホン、ホスホロチオエートおよびホス
ホロジチオエートの各結合がある。
本発明のこの実施例に基づく抗インフルエンザ修飾オリゴヌクレオチドは、現
在の技術でよく知られている手法に従って合成される(たとえば、ウルマンとペ
イマンによるChemical Review 90巻543〜584頁(1990年);シュナイダーとバン
ナーによるTetrahedron Lett.31巻335頁(1990年)を参照)。3'末端にキャッ
プ構造を有するオリゴヌクレオチドについては、キャップ構造を可逆的に固相担
体に付着させてから、合成スキームにおける最初のヌクレオチドモノマーと連結
させる。5'末端にキャップ構造を有するオリゴヌクレオチドについては、合成ス
キームの最後のヌクレオチドモノマーを付加した後でオリゴヌクレオチドの末端
にキャップ構造を連結させる。
5番目の実施例では、抗インフルエンザ修飾オリゴヌクレオチドを、オリゴヌ
クレオチドと分子内でハイブリ
ッドし、エキソヌクレアーゼ抵抗性のヘアピン状構造を形成するような自己相補
的領域を持たせてることにより自己安定型状態にする。本発明のこの実施例に基
づく抗インフルエンザ修飾オリゴヌクレオチドは、一般に、インフルエンザ・ハ
イブリダイジング領域と自己相補的領域との2つの領域を有することを特徴とす
る。インフルエンザ・ハイブリダイジング領域は、インフルエンザウイルスの必
須核酸配列と相補的なヌクレオチド配列を有している。この領域は、約6から約
100ヌクレオチドであるのが好ましい。本発明のこの実施例の1つの形を図2
に示す。この形では、インフルエンザ・ハイブリダイジング領域は長方形がつな
がったもので示してあり、自己相補的領域は円がつながったもので示してある。
標的インフルエンザのメッセンジャーRAN分子中の相補的核酸配列は、ダイヤ形
をつなげたもので示してある。ヌクレオチド間の水素結合は点で表してある。オ
リゴヌクレオチドは安定状態にある。すなわち、標的ハイブリダイジング領域と
自己相補的領域の間の塩基対によって、または自己相補的領域内の相補的配列の
間の塩基対によって、あるいはこの両者によってエキソヌクレオチド減成に対す
る抵抗性が賦与されている。このオリゴヌクレオチドが相補的核酸配列を有する
インフルエンザ核酸分子に出会うと、オリゴヌクレオチドのインフルエンザ・ハ
イブリダイジング領域とオリゴヌクレオチドの自己相補的領域との間の塩基対が
切断され、オリゴヌクレオチド
のインフルエンザ・ハイブリダイジング領域と核酸分子の相補的核酸配列との間
の塩基対で置き換えられる。このように塩基対が切断され置換されるのは、標的
核酸配列と標的ハイブリダイジング領域との間のハイブリッドによって形成され
た分子間塩基対構造のほうが、自己相補的オリゴヌクレオチドによって形成され
る分子内塩基対構造よりも熱力学的にさらに安定しているからである。
本発明の本実施例に基づくオリゴヌクレオチドの第2の形は、第1の形と同じ
ように働くが、自己相補的塩基対としては異なる構造を形成する。この代替形態
は、図3に示すようなハンマー様構造を形作る。この形態では、自己相補的領域
にはその自己相補的領域内の他のオリゴヌクレオチド配列と塩基対を作ることの
できるオリゴヌクレオチド配列が入っている。自己相補的領域には、インフルエ
ンザ・ハイブリダイジング領域に相補的なオリゴヌクレオチト配列もある。
本発明に基づく自己安定型オリゴヌクレオチドの第2の重要な意味を持つ領域
は、自己相補的領域である。自己相補的領域には、オリオヌクレオチド内の他の
オリゴヌクレオチド配列と相補的なオリゴヌクレオチド配列がある。これらの他
のオリゴヌクレオチド配列は、インフルエンザ・ハイブリダイジング領域内また
は自己相補的領域内にあり、あるいは両領域にまたがることもある。相補的配列
は塩基対を作り、図2に示すようなヘアピン構造、あるいは図3に示すようなハ
ンマー様構造を形成
する。ヘアピン構造またはハンマー様構造はいずれも、ヘアピン構造の場合には
図2に示すように塩基対を作っていないヌクレオチドによるループを持つことが
でき、ハンマー様構造の場合には図3に示すようにそのようなループがまったく
ないこともある。自己相補的領域を含む分子内ハイブリダイゼーションにより形
成される塩基対の数は様々であるが、3'末端にエンドヌクレアーゼが接近できな
いように二本鎖構造を維持するのに充分な数でなければならない。一般に、この
ような二本鎖構造を維持するには4対以上の塩基対が必要になる。好ましい実施
例では、自己安定型オリゴヌクレオチドに約10の分子内塩基対が形成されてお
り、10の塩基対は連続していて、3'-末端の大部分のヌクレオチドを含んでい
る。もちろん分子内塩基対はオリゴヌクレオチドのすべてのヌクレオチドを含む
ほど広がることもできる。約50以下のヌクレオチドから成る自己相補的領域を
含むことが好ましい。
本実施例に基づくオリゴヌクレオチドは、第4の実施例について述べるように
、1からほとんどすべての修飾ヌクレオチド間結合を有することがある。できれ
ばインフルエンザ・ハイブリダイジング領域または自己相補的領域の少なくとも
一方に、また最も好ましくはこの両方に、ホスホロチオエート結合またはホスホ
ロジチオエート結合あるいはその両者によって連鎖された約2からほぼすべての
ヌクレオチドが入っていることが望ましい。
本発明の本実施例に基づく抗インフルエンザ修飾オリゴヌクレオチドの例を、
表IにCMPD Oとして示す。
当業者であれば、上記の種々の好ましい実施例の特性を組み合わせて、よりい
っそう大きな抗インフルエンザ能を有する追加の実施例を実現できることがわか
るだろう。したがって、本発明は、キメラ特性、ハイブリッド特性、キャップ構
造、自己安定特性といったいずれもここで述べた特性のあらゆる可能な組み合わ
せを使った抗インフルエンザ修飾オリゴヌクレオチドを考えている。
本発明の好ましい実施例のいすれも、同一のインフルエンザmRNAにハイブリダ
イズする現在の技術におけるアンチセンスオリゴヌクレオチドに比べてインフル
エンザの複製または増殖を阻止する能力が大きい。たとえば、米国特許第5,194,
428号は、インフルエンザウイルスの複製を阻止する修飾オリゴヌクレオチドに
ついて明らかにしている。その修飾オリゴヌクレオチドの中には、インフルエン
ザPB1(ポリメラーゼ1)遺伝子にハイブリダイズするオリゴヌクレオチドホスホ
ロチオエートがある。本研究では、インフルエンザPB1ポリメラーゼ遺伝子にハ
イブリダイズするオリゴヌクレオチドホスホロチオエートのインフルエンザウイ
ルス複製阻止能を、本発明の種々の好ましい実施例と比較して調べた。そこで調
べた本発明の実施例のいずれでも、抗インフルエンザ活性の効力は驚くほど向上
しており、これは同一遺伝子上の同一部位に結合するオリゴヌクレオチドホスホ
ロチオエー
トと相関している。下記の表IIIに示すように、本発明に基づく抗インフルエン
ザ修飾オリゴヌクレオチドを用いた場合には、50%阻害濃度(IC50)は2倍から
15倍近くになった。
このようなオリゴヌクレオチドは、様々な目的に活用できる。まず第1に、ど
のインフルエンザ遺伝子のどの部位が、インフルエンザウイルスの複数の株に対
して幅
広い有効性を示すアンチセンスオリゴヌクレオチヂの最もよい基盤となるかを調
べる試験で利用できる。第2に、どの構造特性あるいはその組み合わせが、in v
itroおよびin vivoでインフルエンザに対して最も大きな効果をもたらすかを明
らかにするのに利用できる。さらに、このようなオリゴヌクレオチドはインフル
エンザ感染の治療薬としても有用である。このような治療では、オリゴヌクレオ
チトは腹腔内投与、鼻腔内投与、経口投与または直腸投与ができる。このような
オリゴヌクレオチドは、できれば体重1kgあたり約1mgから約50mgの濃度で投
与するのが好ましい。
以下に挙げる例は、本発明の好ましい実施例を個別にさらに詳しく示すための
もので、これらに限定しようとするものではない。
実験例1
メチルホスホネート領域およびホスホロチオエート領域
を有するキメラオリゴヌクレオチドの合成
メチルホスホネートおよびホスホロチオエート領域を有するキメラオリゴヌク
レオチドを、メチルホスホネート領域についてはメチルホスホンアミダイトを、
ホスホロチオエート領域についてはヌクレオシドH−ホスホネートを使って合成
した。
メチルホスホンアミダイトの台成は次のようにした。
窒素下で15℃エーテル中でメチルジクロロホスフィ
ン(51ml)とジイソプロピルアミン(102mmol)を反応させて、メチルクロロ-N,
N-ジイソプロピルアミノホスフィンを合成した。濾過して塩を除き、溶媒を蒸発
させた後、1Hおよび31P NMRで特徴づけられる油としてメチルクロロ-N,N-ジイソ
プロピルアミノホスフィン(48mmol、理論値の95%)を得、これは−20℃で8週
間以上にわたって安定であった。この成生物をN,N-ジイソプロピルエチルアミン
を含有するジクロロメタン中で通常の保護をしたヌクレオシドと室温で10〜20分
間反応させた。水性の後処理をし、−30℃から−40℃でペンタンを使って酢酸エ
チルから沈殿させると、白色固体成生物が80〜90%の収率で得られた。この生成
物は、CH2Cl2:EtOAc:Et3N(9:9:2)でシリカの薄層クロマトグラフィー(tlc
)で精製し、1Hおよび31P NMRで調べた。
これらの成生物を、標準ホスホルアミダイト試薬に用いたのと同一の条件およ
びプログラムにより自動化されたDNA合成装置で使用した。ヌクレオチドを33mg/
mlの濃度でアセトニトリルに溶解し、テトラゾールで活性化した。カップリング
時間を1分間として、あらかじめパックしたCPG担体で合成を行った。
ジメトキシトリチル定量分析によってカップリング効率を追跡したところ、ホ
スホルアミダイトを使って並行実施していた対照合成と同じであることがわかっ
た。
カップリングの後、成生物を脱トリチル化し、室温で2時間かけてNH4OHを使
って担体から切り離し、エチレン
ジアミン:エタノール(1:1)を使って室温で4時間脱ブロックした。この塩基
処理によって、HPLCで定量分析したモデル試験において、ヌクレオシド間ホスホ
ネート基の約1%が減成した。
H−ホスホネート合成は、本明細書でも参考として引用している米国特許第5,
149,798号に例示されている方法で行った。それからH−ホスホネートを二硫化
炭素/ピリジン/トリエチルアミン(9:9:1 vol/vol)中で0.2M硫黄で酸化して
ホスホロチオエートに変換した。
実験例2
メチルホスホノチオエート領域およびホスホロチオ
エート領域を有するキメラオリゴヌクレオチドの合成
いずれかの末端に隣接する4つのヌクレオチドメチルホスホノチオエート領域
を、また中央部には隣接する12のオリゴヌクレオチドホスホロチオエート領域
を有する修飾オリゴヌクレオチドを調製するにあたっては、8マイクロモル規模
で以下の手順を用いた。第1モノマーは、制御細孔ガラス(CPG)固体担体にあ
らかじめ結合させておいた。第2のモノマーは、デオキシヌクレオチドメチルホ
スホンアミダイトであるが、標準的なアミダイトカップリングサイクルを使って
第1モノマーとカップリングさせた(たとえば、アグラワルとグッドチャイルド
によるTetrahedron Lett.28巻3539〜3542頁(1987年)を参照)。別々のサイク
ルで、さらに3つのデオキシヌ
クレオチドメチルホスホンアミダイトを次々とカップリングさせて鎖を延ばした
。それから、アセトニトリル中の1%ボカージュ(Beaucage)試薬(3H-1,2-ベ
ンゾジチオール-2-オン)を使って室温で5分間かけて酸化的チオレーションを
行い、CPG-結合ペンタヌクレオチドメチルホスホノチオエートを生成させた。続
く12のモノマーは、本明細書で参考のため引用しているアグラワルとザメクニ
ックによる米国特許第5,149,798号のH−ホスホネート法により順次追加した。
それからH−ホスホネートを二硫化炭素/ピリジン/トリエチルアミン(9:9:1
vol/vol)中で0.2Mの硫黄で酸化してホスホロチオエートに変換した。最後
の4モノマーは、デオキシヌクレオチドメチルホスホノチオエートであるが、こ
れは追加してから前記の方法で酸化的チオレーションを行った。こうしてできた
オリゴヌクレオチドを、0.5mlエチレンジアミン中で室温で30分間、脱保
護してから、時々撹拌しながら6時間室温で保存した。最後にこの混合液を濾過
し、真空状態で蒸発させて固体塊を得て、この塊を水に溶解し、Sep Pak C18で
脱塩した。
実験例3
メチルホスホノチオエート結合を有する
オリゴヌクレオチドの核酸減成に対する抵抗性
3’末端に2〜4の隣接するデオキシヌクレオチドメチルホスホネートを有す
るか、さもなければデオキシヌクレ
オチドホスホジエステルをすべて有するオリゴヌクレオチドの3'エキソヌクレオ
チド減成に対する相対的抵抗性を、オリゴヌクレオチドホスホジエステルとの比
較で調べた。オリゴヌクレオチドそれそれについて、0.4A260単位のオリゴ
ヌクレオチドを凍結乾燥させ、0.5mlの緩衝液(10mM Tris、10mM MgCl2、p
H8.5)に溶解してから、5μl(1.5ミリ単位)のヘビ毒ホスホジエステラーゼ
と混和した。混合液を温度を調節したセルで37℃でインキュベートし、A260の
経時変化をプロットした。濃色性の増大をオリゴヌクレオチド減成の指標とした
。結果を下の表IVに示す。
この結果から、3'末端近くにメチルホスホノチオエート結合を有するオリゴヌ
クレオチドは、このような結合を持たないオリゴヌクレオチドに比べてはるかに
安定していることがわかる。さらに、メチルホスホノチオエート結合の数が増え
るほど、オリゴヌクレオチドの安定性は増大した(4結合>>3結合>>2結合
)。
実験例4
デオキシリボヌクレオチド領域および2'-OMe-リボ
ヌクレオチド領域を有するハイブリッド
オリゴヌクレオチドホスホロチオエートの合成
ハイブリッドオリゴヌクレオチドホスホロチオエートを、本明細書でも参考と
して引用している米国特許第5,
149,798号で述べられているH−ホスホネート法を利用して、自動合成装置(870
0型、ミリポア社、マサチューセッツ州ミルフォード)を使い、CPG上で5〜6μモ
ル規模で合成した。
デオキシリボヌクレオシドH−ホスホネートは、ミリポア社から入手した。2'
-OMeリボヌクレオチドH−ホスホネートは、標準的な手順で合成した。2'-OMeヌ
クレオシドを含有するオリゴヌクレオチドのセグメントは、2'-OMeリボヌクレオ
シドH−ホスホネートを使って希望するサイクルだけ組み立てた。同様に、デオ
キシリボヌクレオシドを含有するオリゴヌクレオチドのセグメントも、デオキシ
ヌクレオシドH−ホスホネートを使って希望するサイクルだけ組み立てた。組立
後、CPG結合オリゴヌクレオチドH−ホスホネートを例2で述べたように硫黄で
酸化し、ホスホロチオエート結合を生成した。それからオリゴヌクレオチドを濃
NH4OH中で40℃で48時間かけて脱保護した。
粗オリゴヌクレオチド(約500A260単位)をC18逆相媒体上で逆相低圧クロマト
グラフィーで分析した。80%酢酸水溶液で処理してDMT基を除いてから、オリゴ
ヌクレオチドを蒸留水で透析し、凍結乾燥した。
実験例5
ハイブリッドオリゴヌクレオチドホスホロチオエート
の相対的ヌクレアーゼ抵抗性
様々なハイブリッドオリゴヌクレオチドホスホロチオエートの相対的ヌクレア
ーゼ抵抗性について調べるために、オリゴヌクレオチドをヘビ毒ホスホジエステ
ラーゼ(SVPD)で処理した。2'-OMe-RNA領域を持たないオリゴ、または5'末端に
3つの、3'末端に4つの隣接する2'-OMeリボヌクレオチドを有するオリゴ、ある
いはすべてに2'-OMeヌクレオチドを有する約0.2A260単位のオリゴを、500μlの
緩衝液(40mM NH4CO3、pH4.0+20mM MgCl2)に溶解し、0.1単位のSVPDと混和し
た。混合液を37℃で420分間インキュベートした。0分後、200分後、420分後に
、165μlを取り出してイオン交換HPLCを使って分析した。すべてに2'-OMe-リボ
ヌクレオチドを有するオリゴヌクレオチドは、SVPDに対する抵抗性がきわめて強
かったのに対して、2'-OMe-リボヌクレオチドを持たないオリゴヌクレオチドは
ほぼ完全に消化されてしまし、また5'末端および3'末端に2'-OMe-RNA領域を有す
るオリゴヌクレオチドは50%消化された。オリゴヌクレオチドホスホジエステル
は、10分の1の濃度のSVPDで1分間で約80%消化された。
これらの結果から、オリゴヌタレオチドホスホロチオエートに2'-OMeリボヌク
レオチドが存在すると、エキソヌクレオチド分解消化に対する抵抗性が増強され
ること、またこの抵抗性の増強は、2'-OMeリボヌクレオチドの使用比率が大きい
ほど高まることがわかる。リボヌクレオチド、他の2'-置換リボヌクレオチドお
よび2'-OMeリボヌ
クレオチドの特徴および挙動は似ているので、これらの結果から、リボヌクレオ
チド、2'-置換リボヌクレオチド、あるいはリボヌクレオチドおよび2'-置換リボ
ヌクレオチドを混ぜて有するハイブリッドオリゴヌクレオチドホスホロチオエー
トまたはホスホロジチオエートあるいはその両者でも、同様にヌクレアーゼ抵抗
性が増強されることがわかる。
実験例6
3'キャップ構造を有する
オリゴヌクレオチドホスホロチオエートの合成
オリゴヌクレオシドホスホロチオエートは、制御細孔ガラス(CPG)上でH−
ホスホネート化学品を用いて8700型自動合成装置(ミリゲン-バイオサーチ社、
マサチューセッツ州バーリントン)で合成してから、二硫化炭素/ピリジン/ト
リエチルアミン(9:9:1 vol/vol)中で0.2M硫黄で酸化した。合成は5〜10
マイクロモル規模で行った。オリゴヌクレオシドホスホロチオエートは、低圧イ
オン交換クロマトグラフィー(DEAE-セルロース、DE-50ワットマン)で精製して
から、逆相クロマトグラフイー(C18)および透析を行った。5'-キャップ構造オ
リゴヌクレオシドホスホロチオエートは、必要な配列を組立た後で、N-Fmoc-O'-
DMTr-3-アミノ-1,2-プロパンジオール-H-ホスホネートで最後のカップリングを
行って調製した。それから5'-キャップ構造オリゴヌクレオシドH-ホ
スホネートを硫黄で酸化した。3'-キャップ構造オリゴヌクレオシドホスホロチ
オエートはN-Fmoc-O'-DMTr-3-アミノ-1,2-プロパンジオール-CPG上に組立てられ
、硫黄により酸化された。これらの手順を組み合わせて利用して、3',5'-キャッ
プ構造オリゴヌクレオシドホスホロチオエートを調製した。
この代わりに、他の3'または5'キャップ構造を持つオリゴヌクレオシド(たと
えば図4を参照)を、キャップ調製手順で、N-Fmoc-O'-DMTr-3-アミノ-1,2-プロ
パンジオール-H-ホスホネートまたはCPGの代わりにホスホネートまたはCPGで誘
導されたキャップ構造を用いることによって調製できる。同様に、オリゴヌクレ
オチドホスホロチオエート以外のキャップされた修飾オリゴヌクレオチドは、適
当な合成手順にキャップ調製手順を付け加えることによって同じような方式で調
製する。
実験例7
終末キャップ構造を有するオリゴ
ヌクレオチドホスホロチオエートのin vivoでの安定性
雄CDC2F1マウス(平均体重20g)に、放射標識したオリゴヌクレオチドを200マ
イクロリットルの生理食塩水に溶解したものを30mg/kgの用量で静注または腹腔
内注した。キャップ構造のオリゴヌクレオチドおよびキャップ構造になっていな
いオリゴヌクレオチドをそれそれ3匹ずつのマウスに投与した。各動物から投与
の24時間後まで個
体別に尿を採取し、0.5%SDS溶液、10mM 20mM Tris Cl(pH7.6)、10mM EDTA中の
プロテイナーゼK(2mg/ml、最終濃度)によって37℃で1時間抽出してから、フ
ェノルクロロホルム抽出およびエタノール沈殿を行った。回収したオリゴヌクレ
オチドをPAGE(20%ポリアクリルアミド/7M尿素)で分析してからオートラジオ
グラフィーを行った。ケージのすすぎ水からの放射性物質も測定して尿溢流につ
いて調べた。
投与の24時間後に、キャップ構造のものもそうでないものも、オリゴヌクレオ
シドホスホロチオエートの約30%が排泄された。排泄されたキャップ構造ではな
いオリゴヌクレオシドホスホロチオエートおよび5'-キャップ構造オリゴヌクレ
オシドホスホロチオエートは、図5に示すようにかなり減成していた。排泄され
た3'-キャップ構造および3',5'-キャッップ構造のオリゴヌクレオシドホスホロ
チオエートは、これとは対照的に実質的にはまったく減成していなかった。この
ことから、尿中に排泄されたオリゴヌクレオシドホスホロチオエートのin vivo
における減成は、オリゴヌクレオチドの3'ヒドロキシル基にキャップを付加する
ことによって阻害される3'-エキソヌクレアーゼ活性によって媒介されることが
わかる。
実験例8
自己安定型オリゴヌクレオチドホスホジエステルの
ヌクレアーゼ抵抗性
本試験に使用した対照オリゴヌクレオチドは、自己相補的領域を持たないオリ
ゴデオキシヌクレオチドホスホジエステルであった。被験化合物は、10ヌクレ
オチドの3'自己相補的領域を有する点を除いてはこれとまったく同じであった。
サイズの効果についてコントロールするために、3'末端に10のマッチしないヌク
レオチド(T10)を有する点を除いては最初の対照オリゴヌクレオチドとまった
く同じであるさらに別の対照オリゴデオキシヌクレオチドホスホジエステルを使
用した。
オリゴヌクレオチドの3'エキソヌクレオチド減成に対する相対的抵抗性につい
て調べた。オリゴヌクレオチドそれぞれについて、オリゴヌクレオチド0.4A260
単位を凍結乾燥し、0.5mlの緩衝液(10mM Tris、10mM MgCl2、pH8.5)に溶解し
、5μl(1.5ミリ単位)のヘビ毒ホスホジエステラーゼ(SVPD)と混和した。
混合液を温度を調節したセルで37℃でインキュベートし、A260の経時変化をプロ
ットした。濃色性の増大をオリゴヌクレオチド変性の指標とした。
これらの実験の結果を以下の表Vに示す。この結果から、本発明に基づく自己
安定型オリゴヌクレオチドホスホジエステルは、オリゴヌクレオチドホスホジエ
ステルと比べても、また非相補的テールを有するオリゴヌクレオチドホスホジエ
ステルと比べても、3'エキソヌクレオチド減成に対してはるかに抵抗性が大きい
ことがわかる。
前記の試験に加えて、オリゴヌクレオチドをDNAポリメ
ラーゼI 3'-エキソヌクレアーゼで消化させることも行った。図6に示すよう
に、非自己安定型オリゴヌクレオチドは、30分で完全に消化されたが、10のヌク
レオチド自己相補的領域を有する自己安定型オリゴヌクレオチドは、30分たって
も部分的にしか消化されていなかった。
実験例9
自己安定型オリゴヌクレオチド
ホスホロチオエートのヌクレアーゼ抵抗性
自己安定型および非自己安定型のオリゴヌクレオチドホスホロチオエートの相
対的ヌクレアーゼ抵抗性を調べるために、DNAポリメラーゼI 3'-エキソヌクレ
アーゼ活性の定量分析を使用した。これは、SVPDによるオリゴヌクレオチドホス
ホロチオエートの減成が緩慢なためであ
る。
オリゴヌクレオチドはすべて5-末端にγ-32P-ATPおよびキナーゼで標識をした
。5'-標識オリゴヌクレオチド40pmoleを20μlの緩衝液(40mM Tris HCl、pH8.0
、10mM MgCl2、5mM DTT、50mM KCl、50μg/ml BSA)に溶解した溶液に、DNAポリ
メラーゼI 5単位を加え、37℃でインキュベートした。0分後、30分後、60分
後、120分後に4μlをとり、停止溶液(98%ホルムアミド、10mM EDTA、0.1%
キシレンシアノール、0.1%ブロモフェノールブルー)と混和した。この試料を1
5%アクリルアミドケル(尿素)で分析し、オートラジオグラフィーを行った。
結果を図7に示す。自己相補的領域を持たないか、あるいは(オリゴヌクレオ
チドホスホジエステルについての実験例8で述べたように)3'末端にマッチして
いないヌクレオチド(T10)のみを有するオリゴヌクレオチドホスホロチオエー
トは、4時間以内にほぼ50%消化された。10ヌクレオチド自己相補的領域を有
するオリゴヌクレオチドホスホロチオェートは、4時間後にも減成していなかっ
た。6または4ヌクレオチド自己相補的領域を有するオリゴヌクレオチドホスホ
ロチオエートも、安定していることがわかった。これらの結果から、自己安定型
オリゴヌクレオチドホスホロチオエートは、非自己安定型のオリゴヌクレオチド
ホスホロチオエートに比べて核酸減成に対する抵抗性がはるかに強いことがわか
る。
実験例10
修飾オリゴヌクレオチドの抗インフルエンザ活性
MDCKイヌ腎細胞を、非必須アミノ酸(GIBCO BRL、ニューヨーク州グランドア
イランド)を含み、5%ウシ胎仔血清(ハイクロン・ラボラトリーズ社、ユタ州
ロゲン)、0.1% NaHCO3を加えた最少必須培地(MEM)に、96ウェル組織培養プ
レート(コーニング社、ニューヨーク州コーニング)を用い4×105細胞/mlの濃
度で0.2ml/ウェルずつ播種した。細胞を1晩インキュベートして、細胞の単層が
できるようにした。それから増殖培地を取り除き、あらかじめ決めておいた濃度
のオリゴヌクレオチド0.1mlを、0.18% NaHCO3および50μg/mlのゲンタマイシン
を含有する無血清MEMに入れた。各化合物のそれそれ4ウェルずつについてこれ
を実施した。そのうち1ウェルは毒性対照で、3ウェルは抗ウイルス試験用であ
った。3つの細胞対照ウェルおよび6つのウイルス対照ウェルに、0.18% NaHCO3
および50μg/mlのゲンタマイシンを含有する無血清MEM 0.1mlを加えた。オリゴ
ヌクレオチド化合物を加えて10分以内に、インフルエンザA/NWS/33(H1N1)ウイ
ルスを含む、0.18% NaHCO3、20μg/mlのトリプシン、2μg/mlのEDTAおよび50
