【発明の詳細な説明】
セロトニン様レセプター活性を有する新規のポリペプチド、これらのポリペプチ
ドをコードする核酸、および使用
本発明は、新規のポリペプチドおよびそれらの発現を可能にする遺伝物質に関
する。より具体的には本発明はセロトニン様レセプター活性を有する新規のポリ
ペプチドに関する。
セロトニンは、睡眠、食欲、運動、性的活動、および血管収縮のような多種多
様の行動機能を誘導および調節することが可能なニューロモジュレーターである
。セロトニン活性は、セロトニン様レセプターもしくは5−HT(5−ヒドロキ
シトリプタミンとして)レセプターと表示されるレセプターとの相互作用により
媒介されることが認められている。分子生物学的研究ならびに薬理学的研究によ
り5−HTレセプターの多大な数のサブタイプの存在が明らかにされている。今
日までに記載されている5−HTレセプターは、イオンチャネルに関連するレセ
プターの一族(5−HT3レセプター)か、あるいはG蛋白質と相互作用しそし
て7回貫通膜ドメインを保持するレセプターの一族のいずれかに属する。その上
アミノ酸配列の分析により、G蛋白質と相互作用する5−HTレセプターは2つ
の明瞭な群に再分割することができることが示されており、それらは、哺乳類の
サブタイプ5HT1A、5HT1B、および5HT1Dを含む5HT1レセプタ
ー類ならびに3つのドロソフィラの5HTレセプター類と、そしてサブタイプ5
HT2および5HT1Cを含む5HT2レセプター類との2つである。
これらのレセプターばかりが現存する5HTレセプターであることは恐らくあ
りえず、それは、薬理学的研究により5HT4レセプター類、ならびにサブタイ
プ5HT1に関連する幾つかのレセプター類(「5HT1−様」レセプター類)
のような他のサブタイプが明らかにされているためである。その上、追加的な分
子生物学的研究によってはサブタイプ5HT1B/ID内の不均質性も証明され
ている。
本発明は、セロトニン様レセプター活性を有する新規のポリペプチドの成果を
開示するもである。G蛋白質と相互作用するレセプターの一族に属しながらも、
これらの新規のポリペプチドは構造的見地および薬理学的見地の両方から既述の
セロトニン様レセプター(5HT1、5HT2、5HT3、および5HT4)と
は異なる。より具体的には本発明は、5HT5bと表示されるこれらの新規のポ
リペプチド、および更にそれらの発現もしくは同定を可能にする遺伝物質の単離
および特性決定の成果である。
従って本発明の第一主題は、配列番号2のペプチド配列もしくは後者の誘導体
の全てもしくは一部分を含むポリペプチド類である。
本発明の目的のためには、用語「誘導体」は、配列番号2のペプチド配列の遺
伝的および/または化学的特性の修飾により取得されるいずれかの分子を意味す
る。遺伝的および/または化学的特性の修飾は、一つもしくは複数の残基のいず
れかの突然変異、置換、欠失、添加、および/または修飾を意味することが理解
される。このような誘導体を、具体的には、リガンド(一つもしくは複数)に対
するペプチドの親和性を増加させる、産生レベルを改善する、プロテアーゼに対
する耐性を増加させる、活性を増加および/または改変する、もしくはそれに新
規の薬物
動態学的および/または生物学的特性を付与する目的のような様々な目的のため
に作成することができる。添加により得られる誘導体の内では、一つの末端に連
結される追加的異型部分を有するキメラポリペプチドを例として挙げることがで
きる。用語「誘導体」は更に、他の細胞源、そして具体的にはヒト起源もしくは
他の生物体の細胞を基にし、かつ同じ種類の活性を保持する配列番号2のポリペ
プチドと相同なポリペプチドも含む。このような相同ポリペプチドは、実施例に
記載されるハイブリッド形成実験により取得することができる。
本発明のポリペプチドはセロトニンに結合する能力を保持するポリペプチドで
あることが好ましい。それらがセロトニン様レセプター活性を有するポリペプチ
ドであることが更により好ましい。好ましい態様に従うと、本発明のポリペプチ
ドは更に配列番号2の全ペプチド配列を認識する抗体により認識されることが可
能である。
本発明の具体的な態様は、配列番号2の全ペプチド配列を含むポリペプチド5
HT5bにより表される。実施例に示されるように、このポリペプチドを機能的
なセロトニン様レセプターを形成するための様々な種類の細胞において発現する
ことができる。
本発明のポリペプチドは後述するような細胞宿主によるヌクレオチド配列の発
現によるか、当業者に知られる技術を使用する配列番号2に基づく化学的合成に
よるか、あるいはこれらの技術の組み合わせにより取得することができる。
今後、先に特定される本発明のポリペプチドを5HT5bポリペプチドと表示
する。
本発明の主題は更に、5HT5bポリペプチドをコードするいずれの
ヌクレオチド配列でもある。より好ましくはこのような配列は、
(a)配列番号1のヌクレオチド配列もしくはその相補鎖の全てもしくは一部
分、
(b)配列(a)とハイブリダイズし、かつ先に特定されるポリペプチドをコ
ードするいずれかの配列、ならびに
(c)遺伝子コードの縮重の結果として配列(a)および(b)から取得され
る配列、
から選択される。
本発明の様々なヌクレオチド配列は人工起源もしくはそうではないものである
ことができる。それらは、ゲノム、cDNA、もしくはRNA配列、ハイブリッ
ド配列、あるいは合成もしくは半合成配列であることができる。これらの配列は
、例えば配列番号1の配列を基にして得られるプローブによりDNAライブラリ
ー(cDNAライブラリー、ゲノムDNAライブラリー)をスクリーニングする
ことにより取得することができる。このようなライブラリーは、当業者に知られ
る分子生物学の標準技術により様々な起源の細胞から調製することができる。本
発明のヌクレオチド配列はまた、具体的にはホスホルアミダイト法に従う化学合
成によるか、あるいは別法ではライブラリーをスクリーニングすることにより取
得される配列の化学的もしくは酵素的修飾を含む混合法により調製することもで
きる。
本発明のヌクレオチド配列を、既述の5HT5bポリペプチドの産生用に使用
することができる。この場合、前記ポリペプチドをコードする部分は、一般的に
は細胞宿主内でのその発現を可能にするシグナルの調節下に置かれる。これらの
シグナル(プロモーターおよびターミネータ
ーなど)の選択は、用いられる細胞宿主に従って変えることができる。この目的
のためには、本発明のヌクレオチド配列は、自律複製ベクターもしくは組込みベ
クターであることができるベクターの一部分を形成することができる。より具体
的には、自律複製ベクターは選択される宿主内で自律的に複製する配列を用いて
調製することができる。組込みベクターに関しては、これらは、例えば相同組換
えによりそのベクターの組込みを可能にする宿主ゲノムの所定の領域と相同な配
列を用いて調製することができる。組換え法による本発明の5HT5bポリペプ
チドの産生に利用可能な細胞宿主は、真核生物もしくは原核生物のいずれかの宿
主である。適切な真核生物宿主の中では、動物細胞、イースト、もしくは真菌類
を挙げることができる。具体的には、イーストに関しては、サッカロミセス(S accharomyces
)、クルイベロミセス(Kluyveromyces
)、ピキア(Pichia)、シュワニオミセス(Schwanniomyce s
)、もしくはハンセヌラ(Hansenula)属のイーストを挙げることが
できる。動物細胞に関しては、COS、CHO、C127、およびNIH−3T
3細胞などを挙げることができる。真菌類の中では、アスペルギルス エスエス
ピー(Aspergillus ssp.)、もしくはトリコデルマ エスエス
ピー(Trichoderma ssp.)をより具体的に挙げることができる
。原核生物宿主としては、以下に示す細菌、すなわち大腸菌(E.coli)、
バチルス(Bacillus)、もしくはストレプトミセス(Streptom yces
)を使用することが好ましい。
本発明のヌクレオチド配列はまた、遺伝子療法という状況で用いることができ
るアンチセンス配列の産生か、あるいは生物学的試料中のセロ
トニン様レセプターの発現のハイブリッド形成実験による検出ならびに遺伝子異
常(多型性、突然変異)もしくは異常発現の証明を可能にするプローブの産生の
いずれかのために、薬剤学的分野において利用可能でもある。
アンチセンスオリゴヌクレオチドによる所定の遺伝子の発現の阻害は、遺伝子
の活性を調節する有望な手法であることが判明している。アンチセンスオリゴヌ
クレオチドはmRNAに相補的であり、そしてそのため後者と特異的にハイブリ
ッド形成して蛋白質への翻訳を阻害することが可能な小オリゴヌクレオチドであ
る。従って本発明の主題は、先に特定される5HT5bポリペプチドの産生を少
なくとも部分的に阻害することが可能なアンチセンスオリゴヌクレオチドである
。本発明はまた、5HT5bポリペプチドの産生を少なくとも部分的に阻害する
ことが可能なアンチセンス配列にも関する。これらのアンチセンス配列は、細胞
性mRNAに相補的な標的細胞転写物中に産生される。このようなオリゴヌクレ
オチドもしくは配列は、先に特定されるヌクレオチド配列の全てもしくは一部分
からなることができる。これらは一般的には本発明のペプチドをコードする配列
に相補的な配列もしくは配列断片である。このようなオリゴヌクレオチドは、フ
ラグメント化もしくは化学合成などにより配列番号1の配列から取得することが
できる。
先に記載するように、本発明はまた、本発明の5HT5bポリペプチドをコー
ドする先に特定されるヌクレオチド配列もしくは対応するmRNAとハイブリッ
ド形成することが可能な、合成もしくはそうではないヌクレオチドプローブを産
生することを可能にもする。このようなプローブは、5HT5bセロトニン様レ
セプターの発現を検出するためか、
あるいは別法では遺伝子異常(間違ったスプライシング、多型性、および点突然
変異など)を証明するための診断用の道具としてインビトロで用いることができ
る。セロトニンの多大な活性を考慮すると、本発明のプローブはそのため神経学
的、心臓血管学的、もしくは精神医学的疾患を5HT5bレセプターに関連する
ものとして同定することが可能である。これらのプローブはまた、他の細胞源、
および好ましくはヒト起源の細胞から、先に特定される5HT5bポリペプチド
をコードする相同核酸配列を証明および単離するのに用いることもできる。本発
明のプローブは一般的には少なくとも10の塩基を含み、そしてそれらは配列番
号1もしくはその相補鎖の全配列と同程度の塩基を含むことができる。これらの
プローブはそれらの使用前にラベル化することが好ましい。この目的のためには
当業者に知られる様々な技術を利用することができる(放射能ラベル化もしくは
酵素的ラベル化など)。これらのプローブを用いることができるハイブリッド形
成条件は以下の一般的なクローニング技術ならびに実施例において記載される。
本発明の他の主題は、先に特定される5HT5bポリペプチドを表面に発現す
ることが可能な組換え細胞に関する。これらの細胞は、本発明のポリペプチドを
コードする先に特定されるヌクレオチド配列を組み込ませ、そしてその後に前記
細胞を前記配列の発現のための条件下で培養することにより取得することができ
る。
本発明に従う組換え細胞は、真核生物もしくは原核生物細胞のいずれかである
ことができる。適切な真核生物細胞の中では、動物細胞、イースト、もしくは真
菌類を挙げることができる。具体的には、イーストに関しては、サッカロミセス
(Saccharomyces)、クルイベ
ロミセス(Kluyveromyces)、ピキア(Pichia)、シュワニ
オミセス(Schwanniomyces)、もしくはハンセヌラ(Hanse nula
)属のイーストを挙げることができる。動物細胞に関しては、COS、
CHO、C127、およびNIH−3T3細胞などを挙げることができる。真菌

。このようにして取得される細胞を使用して、リガンドもしくは本発明のポリペ
プチドの活性のモジュレーターとして振る舞う様々な分子の能力を測定すること
ができる。従ってより具体的にはこれらを、リガンドもしくは本発明のポリペプ
チドの活性のモジュレーターを、そしてより具体的にはセロトニン作用薬もしく
は拮抗薬を証明および単離するための方法に用いることができる。
従って本発明の他の主題は、本発明の5HT5bポリペプチドのリガンドの証
明および/または単離のための方法に関し、この方法に従うと以下に示される段
階が実施されるが、それらは、
− 恐らく未同定である、ある分子もしくは様々な分子を含む混合物を、本発
明のポリペプチドをその表面で発現する既述の組換え細胞と、前記本発明のポリ
ペプチドと前記分子との間の相互作用(後者は前記ポリペプチドに対する親和性
を保持する場合には)を可能にする条件下で接触させる段階、および
− 前記本発明のポリペプチドに結合した分子を検出および/または単離する
段階、
である。
ある特別な態様においては、本発明のこの方法は、5HT5bポリペプチドに
ついてのセロトニン作用薬および拮抗薬を証明および/または単離するのに適す
る。
本発明の他の主題は、本発明の5HT5bポリペプチドのモジュレーターを証
明および/または単離するための方法に関し、この方法に従うと以下に示される
段階が実施されるが、それらは、
− 恐らく未同定である、ある分子もしくは様々な分子を含む混合物を、本発
明のポリペプチドをその表面に発現する既述の組換え細胞と、5HTの存在下、
前記本発明のポリペプチドと5HTとの間の相互作用を可能にする条件下で接触
させる段階、および
− 前記本発明のポリペプチドに関して5HTの活性をモジュレートすること
が可能な分子を検出および/または単離する段階、
である。
本発明の他の主題は、医療用産物として既述の方法に従って同定および/また
は取得されるリガンドもしくはモジュレーターの使用に関する。このようなリガ
ンドもしくはモジュレーターは実際のところ、5HT5bレセプターに関連する
所定の神経学的、心臓血管学的、もしくは精神医学的疾患を治療することを可能
にすることができる。
本発明はまた、本発明の5HT5bポリペプチドに作用する少なくとも一つの
分子を活性成分として含むいずれの医療用産物にも関する。この分子は、既述の
方法に従って同定および/または単離されるリガンド
もしくはモジュレーターであることが好ましい。
本発明の他の利点は、以下に示され、そして説明的かつ非制限的であるとして
見なすべき実施例を読む際に明白になるであろう。図面の説明文 表1
:5HT5bレセプターの薬理学的特性。この結果は、5HT5bレセプタ
ーを発現するCos−7細胞の膜に対する[125I]LSDの結合についての競
合実験を表す。IC50値(結合した[125I]LSDの50%を置換するのに必
要なリガンド濃度に相当する)を実験的に算出し、そして以下に示す式:Ki=
IC50/(1+C/Kd)[式中、Cは[125I]LSDの濃度(150pM)
であり、そしてKdは[125I]LSDの解離定数である]に従ってKiに変換
した。括弧内の数値は実施した個々の実験数に相当し、各点は三重反復試験で実

制限酵素類は、New England Biolabs社(BioIags
)、Bethesda Research Laboratories社(BR
L)、もしくはAmersham社から供給され、そしてその供給社の推奨に従
って用いる。
連結反応のためには、DNA断片をアガロースもしくはアクリルアミドゲル電
気泳動によりサイズに従って分離し、フェノールもしくはフェノール/クロロホ
ルム混合物で抽出し、エタノールで沈殿させ、そしてその後に供給社の推奨に従
ってファージT4 DNAリガーゼ(BioIabs社)の存在下でインキュベ
ートする。
5’突出端の充填(filling−in)は、供給社の推奨に従って大腸菌
(E. coli)のDNAポリメラーゼI(Biolabs
社)のクレノウ(Klenow)断片を用いて実施する。3’突出端の破壊は、
製造元の推奨に従って用いられるファージT4のDNAポリメラーゼ(Biol
abs社)の存在下で実施する。5’突出端の破壊は、S1ヌクレアーゼでの制
御下処理により実施する。
合成オリゴデオキシヌクレオチドによりインビトロで誘導される突然変異誘発
は、Taylor et al.[Nucleic Acids Res.13
(1985)8749−8764]により開発された方法に従い、Amersh
am社により配布されるキットを用いて実施する。
いわゆるPCR[Polymerase−catalyzed Chain R

us社)を製造元の説明書に従って用いて実施する。
ヌクレオチド配列の検証はSanger et al.[Proc.Natl
.Acad.Sci.USA、74(1977)5463−5467]により開
発された方法により、Amersham社により配布されるキットを用いて実施
する。
ハイブリッド形成実験用には、緊縮性の通常条件は一般的には以下に示すもの
であり、すなわち、ハイブリッド形成:65℃下、5×デンハート溶液(Den
hart’s)の存在下、3×SCC;洗浄:65℃
下、0.5×SSC、である。
1. 5HT5bレセプターの単離
様々な既知のセロトニン様レセプターとの間の配列比較により、具体的には第
IIIおよび第VIドメインのような所定の可能な貫膜領域内に幾分かの保存が
示される。新規のレセプターを証明および単離する目的で、これらの2つの領域
に相当する3つの縮重オリゴヌクレオチドを調製し、そしてその後にマウス脳の
RNA調製の際の一連のPCR反応に用いた。縮重オリゴヌクレオチドの配列を
以下に示す。

PCR反応は以下に示す様式で実施し、すなわち、5μgの成体マウス脳のR
NAを500ngのオリゴヌクレオチド(i)および200単位のMMLV逆転
写酵素(BRL社)の存在下での逆転写反応に供した。その後にこの反応の産物
の半分を5単位のTaqポリメラーゼ(Cetus社)、ならびに1μgのオリ
ゴヌクレオチド(i)およびオリゴヌクレオチド(ii)の存在下で30回の増
幅周期に供した。その後にこの反応の産物の1/20をオリゴヌクレオチド(i
)および(iii)
の存在下で30回の追加的増幅周期に供した。このようにして得られる産物を酵
素Bam HIおよびHind IIIで消化し、プラスミドBluescri
pt(Stratagene社)の対応部位0内に挿入し、そして配列決定を行
った。このようにして得られ、かつセロトニン様レセプターとの幾分かの相同性
を有する断片の内の一つをランダムプライミング(Feinberg and
Vogelstein、Analytical Biochemistry 1 32
(1984)6)によりラベル化し、そしてそれをブローブとして用いてU
niZapファージ(Stratagene社)内に構築されたマウス脳cDN
Aライブラリーをスクリーニングした。得られた陽性ファージ中では、それらの
内の一つでλNSと称され、かつプラスミドpNSにより保持されるものは2.
1kbの挿入断片を含んでいた。このファージを単離し、そしてその後にその挿
入断片をプラスミドBluescript内に導入した。この断片の配列を、合
成オリゴヌクレオチドによるジデオキシヌクレオチド技術を用いて2本の鎖につ
いて決定した。
このようにして得られた配列は配列番号1の配列に相当する。このことにより
、単離されたcDNAは370アミノ酸の読み枠を保持することが示される。そ
の上、疎水性の分析により、この蛋白質がG蛋白質に結合するレセプターの一族
の一員に生じる特性である7つの疎水性ドメインを保持することが明らかにされ
た。しかもN−末端は1つのN−グリコシル化部位を含み、そしてその推定され
る細胞質ドメインはCおよびA蛋白質キナーゼによるリン酸化用の共通部位を含
む。
5HT5bレセプターをコードするゲノムクローンも、プローブとしての配列
番号1の配列によりゲノムライブラリーをスクリーニングする
ことにより単離した。このライブラリーは、マウス胎仔の幹細胞のゲノムDNA
の部分的Sau 3A消化、およびその後のファージ ラムダーEMBL3内へ
の挿入により予め取得してあった。
得られる断片をプラスミドBluescript内にサブクローン化し、そし
て部分的配列決定を行った。これらの配列の分析により、第3細胞質ループの中
央に位置するイントロンの存在が明らかにされた。
2. 配列相同性の検討
先に単離された5HT5bレセプターの配列を、G蛋白質と結合する以下のレ
セプター:5HT1B、5HT1D、5HT5A、5HT1A、5HT−dro
2A、5HT−drol、α2、D2、β1、D1、H2、5HT1C、および
5HT2の配列と比較した。これらの実験により、有望な貫膜ドメインおよび所
定のループ内の所定の相同性が明らかになったが、末端領域もしくは第三細胞質
ループ内には相同性が全く存在しなかった。表2は、保存領域内の%相同性値を
示す。
3. Cos−7細胞内での5HT5bレセプターの発現および薬理学的特性決 定
実施例1において単離されたcDNA断片を真核生物発現ベクター内に挿入し
、これを使用してCos−7細胞をトランスフェクトした。その後には、得られ
たトランスフェクト化細胞の膜を調製し、そして所定のラベル化セロトニン様リ
ガンドに結合する能力についてのテストを行った。
5HT5bレセプターをコードする2.1kbのcDNAをEcoRI−Xh o
Iの形態でプラスミドpNSから単離し、そしてその後にベクターp513
の対応部位に挿入した。このベクターp513はベ
クタ−pSG5[Green et al.、Nucl.Acids Res.
16(1988)369]から、多重クローニング部位の添加により取得され
る。このようにして取得され、かつp513NSと表示される組換えベクターを
その後に用いて(10cmのデッシュ当たり20μg)、リン酸カルシウムの存
在下でCos−7細胞をトランスフェクトした。
トランスフェクト後48時間目にその組換え細胞を回収し、そしてその膜をA
mlaikyおよびCaron[J.Biol.Chem.260(1985)
1983]により記載される技術に従って調製する。その後に飽和結合および競
合実験を、様々な放射ラベル化リガンドの存在下でこれらの膜について実施した
(表1を参照せよ)。このためには膜試料(10−20μgの蛋白質)を10分
間、37℃で、最終容量250μlの50mM トリス−HCl緩衝液(pH7
.4)中でリガンドの存在下でインキュベートした。その後にこの反応を、ホワ
ットマン(Whatman)のGF/Cガラス繊維濾紙上での吸引濾過により停
止させ、そして4mlの50mM トリス−HCl緩衝液(pH7.4)で4回
すすいだ。非特異的結合を10μMの5HTの存在下で決定した。放射活性をγ
計測器で測定した。
得られる結果により、[125I]LSDは、Kd=470pMおよびBmax=1
70fモル/mg膜蛋白質の飽和結合部位を有することが示される(図1)。そ
の上、対照実験においては、[125I]LSDはプラスミドp513でトランス
フェクトさせたCos−7細胞とは結合しなかったことが示される。
このレセプターの薬理学的特性を決定するために、膜に結合した
[125I]LSDを様々なセロトニン様薬物(表1)の存在下で置換した。これ
らの様々な薬物により、以下に示す順序の置換効率が示され、それは、
エルゴタミン>5−CT>メチセルギド>5HT>RU24969>8−OH−
