発明の名称
遊離および結合両被検物質を有するサンプル中の
総被検物質濃度の測定方法発明の分野
本発明は、サンプル中の被検物質の量を定量する方法に関し、さらに詳しくは
遊離および結合両被検物質を含有するサンプル中の総被検物質濃度を定量する方
法に関する。発明の背景
アッセイとくにイムノアッセイは、細菌性、ウイルス性および寄生虫性疾患な
らびに感染性疾患たとえば世界中の主要な保健問題となっているエイズのような
疾患の診断手段として広く使用されている。
イムノアッセイは一般的に、血清または他の生物学的液体中の被検物質の検出
および/またはその量の定量に用いられ、それは原理的にはサンプル中に存在す
る可能性がある特定の被検物質に対する特異的結合物質たとえば抗体の結合を基
盤とするものである。残念ながら被検物質の検出は被検物質が内因性結合物質−
被検物質複合体の形成によって遮蔽されるため、内因性結合物質の存在によって
妨害されることが多い。たとえばヒト免疫不全ウイルス(HIV)-1p24コア蛋白質
のような被検物質は、血清、血漿等の中ではウイルス感染の結果として産生され
たヒト抗体と結合して平衡状態で存在する。抗体による遮蔽(抗原−抗体複合体
の形成による)は、酵素結合免疫定量法(ELIZA)および放射免疫検定法(RIA)
のような慣用方法では、結合状態の被検物質の検出を妨害する。したがって、
遊離の被検物質のみが検出できる。しかしながら、遊離被検物質の量の定量は、
様々な因子たとえば被検物質の濃度、ならびに抗体の濃度および親和性によって
影響を受けることになる。
現在の方法では被検物質の総濃度の検出が不可能なことは、総被検物質の定量
が重要な場合に、重大な問題を提起しているのである。たとえば、HIV−感染個
体の血清または血漿中HIV抗原レベルの測定は、血清変換前におよび予後のため
に抗原または感染ウイルスの存在の決定に重要であり、また抗ウイルス剤療法の
モニタリングの手段として有用である。生後1年以内の小児におけるHIV感染は
、残存する母体由来の抗体によりHIV診断の標準的血清学的アッセイが不可能な
ことから診断を確定できない。すなわち、疾患の発症過程、制御および予後にお
けるHIVの役割を理解するためには、HIV−感染個体における血清抗原およびウイ
ルス抗原産生の信頼できる測定がきわめて重要である。
被検物質の検出および/または定量を改良するためにアッセイ系の感度および
特異性を増大させる努力はこれまで以下に示すように試みられてきた。
1986年の8月5日付にて、Neurathに対して発行された米国特許第4,604,348号
には、抗原または抗体が免疫複合体の形態で存在するサンプル中の抗原または抗
体を検出する方法が記載されている。この方法は解離緩衝液の存在下に蛋白質吸
着固体支持体へ抗原または抗体を不可逆的に結合させるものである。免疫複合体
に由来する抗原または抗体が固体支持体に吸着されたならば、その存在はRIAま
たはELIZAによって測定できる。
同じくNeurathら、Proc.Natl.Acad.Sci.USA,79:4415−
4419(1982年7月)には、抗体から抗原を分離するための固相法が記載されてい
る。とくに、免疫複合体は血清からポリエチレングリコールによって沈殿させ、
NaSCNで解離させ、ニトロセルロースまたはポリスチレン支持体上に吸着させる
。検出はついでRIAによって行われる。
1985年9月18日に公告された欧州特許出願公告第0 155 104号には、被検物質
が天然の蛋白質バインダーに部分的に結合して存在し、遊離の被検物質と被検物
質誘導体が特異的バインダーとの反応で競合する遊離被検物質アッセイが記載さ
れている。天然の蛋白質バインダーへの被検物質誘導体の結合は低下させるが被
検物質の結合は低下させない判別遮断剤が使用される。
1987年10月27日付にてVodianらに対して発行された米国特許第4,703,001号に
は、被検物質の実質的なその抗体からの遊離および/または他の血清蛋白質の大
部分の変性のためのpH依存性カオトロピック剤を使用して、血清被検物質を検出
するイムノアッセイが記載されている。
Nishanianら、J.Infect.Dis.,162:21−28(1990年7月), von Sydowら
、Br.Med.J.,296:238−240(1988)、ならびにKageyamaら、J.Virol.Meth
.,22:125−131(1988)には、サンプルを酸性のpHで前処理して免疫複合体を
解離させ抗体は抗原の免疫反応性とほとんどまたは全く相互作用しないように変
性させることにより、血清または血漿サンプル中の(免疫複合体中に存在した)
HIV−抗原の検出を促進する方法が記載されている。
これらの方法は、検出不能または検出能の劣る被検物質の検出を容易にするも
のではあるが、これらは総被検物質濃度の測定法を提
供するものではない。発明の要約
本発明は、サンプル中の総被検物質濃度を定量するにあたり、
(a)サンプル中の遊離被検物質の濃度をアッセイし、
(b)そのサンプルが予めそのサンプルに添加された既知量の被検物質を含有
する場合のサンプル中の遊離被検物質の濃度をアッセイし、この測定を少なくと
も1回行い、
(c)工程(a)および(b)において得られたアッセイ濃度から総被検物質
濃度を定量する、
各工程からなる方法に関する。
他の実施態様においては、本発明は、サンプル中のHIV抗原濃度を定量するに
あたり、
(a)サンプル中の遊離HIV抗原の濃度をアッセイし、
(b)そのサンプルが予めそのサンプルに添加された既知量の被検物質を含有
する場合のサンプル中の遊離HIV抗原の濃度をアッセイし、この測定を少なくと
も1回行い、ついで
(c)工程(a)および(b)において得られたアッセイ濃度から総HIV抗原
濃度を定量する、
各工程からなる方法に関する。発明の詳細な説明
本発明の方法は、
(a)サンプル中の遊離被検物質の濃度をアッセイし、
(b)そのサンプルが予めそのサンプルに添加された既知量の被検物質を含有
する場合のサンプル中の遊離被検物質の濃度をアッセイし、この測定を少なくと
も1回行い、
(c)工程(a)および(b)において得られたアッセイ濃度から総被検物質
濃度を定量する、
各工程からなるサンプル中の総被検物質濃度の定量に使用することができる。
本発明の方法においては、血漿、血清、尿等のような生物学的サンプル中の遊
離被検物質の検出および/または定量に使用される任意のアッセイが使用可能で
あり、被検物質を遊離型および結合型の両者で含有する生物学的サンプル中の総
被検物質濃度の迅速かつ正確な定量値が提供される。被検物質が遊離型で存在し
ない場合には、それは上述のような慣用の技術を用いて遊離させることができる
。本発明の方法を用いて評価できる被検物質の例には、ウイルス蛋白質、細菌蛋
白質、ホルモン、薬剤等がある。とくに、血液中に保持された感染性物質、たと
えば、HIV、肝炎ウイルス、エプシュタイン−バールウイルス、サイトメガロウ
イルス、HTLV−1、HIV−II等を挙げることができる。
使用できる市販のアッセイの例には、低pHと熱の組合せを用いて血清および血
漿中抗原/抗体複合体を解離するために設計されたDu Pont HIV-1 Acid Disrupt
ion Difference Immune Complex Kitと併用されるDu Pont HIV-1 p24 Core Prof
ile ELIZAがある。
総被検物質濃度を定量する本発明の方法の重要な態様には、スパイクの不存在
下においては1回、サンプルに予め添加された既知量の被検物質(「スパイク」
)の存在下において少なくとも1回、少なくとも2回の遊離被検物質のアッセイ
が包含される。以下に示すように、被検物質「スパイク」の使用が、遊離型であ
れ結合型であれ、被検物質の総濃度の定量を可能にする。
一般的には、サンプル中の遊離被検物質濃度は、「そのまま」(スパイクを使
用しないで)アッセイにおいて測定される。ついで同じアッセイが、スパイクを
用いて少なくとも2回目として行われる。換言すれば、第2のアッセイにおける
サンプルは、「そのまま」の被検物質のみでなく、サンプル中に存在するすべて
の被検物質に加えて、さらに、既知量の被検物質(「スパイク」)をも含有する
。スパイクの不存在下における被検物質濃度の測定に加えて、少なくとも1つの
スパイクを使用しなければならない。所望により、3種またはそれ以上のスパイ
クを使用することもできる。
最初のサンプル中における総被検物質濃度はついで、得られた測定値から以下
の計算によって定量することができる。
総被検物質濃度=スパイク非添加サンプル測定値×スパイク値/
(スパイク添加サンプル測定値−スパイク非添加サンプル測定値)
2種のスパイク(たとえば、1つは高レベル、1つは低レベルのスパイク)が
使用される場合には、総被検物質濃度の計算式は、
スパイク非添加サンプル測定値×(高スパイク値−低スパイク値)/
(高スパイク添加サンプル測定値−低スパイク添加サンプル測定値)となる。
3種以上のスパイクが用いられる場合には、これらの計算式は用いられたスパ
イクの数により、必要なように改変される。
以下の実施例は本発明の実際を例示するものであって、本発明の限定と解する
べきではない。実施例1
A部:抗原および抗体陽性基準サンプルの調製
濃度194pg/mlのHIV-1 p24抗原を含有する22種の抗原および抗体
陽性基準サンプルはそれぞれ、44μlのHIV-1抗体陽性、抗原陰性血清/血漿を、
375μlの正常ヒト血清/血漿、および2000pg/mlのp24抗原含有HIV-1ウイルス溶
解物451μlと混合して調製した。
B部:サンプルの評価
22種の基準サンプルそれぞれについて、スパイクの不存在下および少なくとも
1種のスパイクの存在下にアッセイした。この評価には2つの異なるスパイクの
レベル、約200および400pg/mlを使用した。サンプルのアッセイには、Du Pont
HIV-1 p24 Core Profile ELISA抗原アッセイを上述のDu Pont HIV-1 p24 Acid D
isruption Differance(ADD)キットと併用して使用した。ADDキットは、血清お
よび血漿中の抗原/抗体複合体を、低pHと熱の組合せを用いて分裂させるために
使用された。サンプルを1.5Mグリシン試薬によってpH1.85の酸性とし、37℃で
1時間インキュベートした。1時間後に、サンプルを1.5Mのトリス、pH11によ
って中和して、Du Pont HIV-1 p24 Core Profile ELISAでアッセイした。
Du Pont HIV-1 p24 Core Profile ELISA抗原アッセイでは、マイクロタイター
プレートのウエルに固定化された抗−HIV p24マウスモノクローナル抗体が用い
られる。固定化されたモノクローナル抗体はついで、サンプル中のウイルスの溶
解により放出されるHIV-1 p24抗原を捕獲する。捕獲された抗原は、HIV-1 p24コ
ア抗原に対するビオチン化ポリクローナル抗体と複合体を形成し、ストレプトア
ビジン−HRP(西洋ワサビペルオキシダーゼ)共役体でプローブされる。複合体
はついでオルトフタレンジアミン塩酸塩(OPD)とインキュベートして検出され
る。これは捕獲されたHIV-1 p24コア抗原の量に正比例する黄色を発色する。各
ウエルの吸光度をマイクロプレ
ートリーダーによって測定し、HIV-1 p24コア抗原標準曲線の吸光度に対して検
量する。
1.試薬
グリシン試薬
これはサンプルを約pH2.0の酸性にするのに用いられる。
%組成
グリシン 11.3%
塩酸 9.6%
蒸留水 79.1%
トリス試薬
これは分裂後サンプルの中和に用いられる1.5Mトリス、pH11である。
%組成
トリス塩基 17.0%
蒸留水 83.0%
2.サンプルプレパレーション
ブランクマイクロタイタープレートのウエルに以下の成分:
a)10μlの5%トリトンX-100
b) 90μlのサンプル
c) 10μlの陰性血清(非スパイク)、約2000pg/mlのHIV-1 p24抗原(低ス
パイク)または約4000pg/mlのHIV-1 p24抗原(高スパイク)を添加した。
スパイクのレベルの実際の値は90μlのサンプルを90μlの陰性血清に置換して
アッセイしたことに留意すべきである。
3.アッセイ
90μlの1.5Mグリシン、pH1.85、および90μlのサンプルまたは標準液を1時
間、37℃においてインキュベートした。1時間後に、90μiの1.5Mトリス、pH11
を各ウエルに添加し、室温で10分間インキュベートした。このサンプル混合物(
150μl)をついでHIV-1 p24Core Profile ELISAプレートに移し、37℃で2時間
インキュベートした。ついでマイクロタイタープレートウエルを洗浄し、各ウエ
ルに100μlのビオチン化検出抗体を加え、37℃で1時間インキュベートした。つ
いでウエルを洗浄し、1:25希釈のストレプトアビジン−HRP接合体を加え、37
℃で15分間インキュベートし、ついでウエルを再度洗浄した。OPD基質を室温で3
0分間添加し、反応はキット停止溶液(4N硫酸)で停止し、490〜650nmにおけ
る吸光度を測定した。
4.定量
すべてのサンプルについての値は、HIV-1 p24コア抗原標準曲線からの内挿に
よって定量した。スパイクの実際の値は、サンプル1〜11については228pg/ml
(低スパイク)および448pg/ml(高スパイク)、サンプル12〜22については234
pg/ml(低スパイク)および438pg/ml(高スパイク)と定量された。上に注釈
したように、スパイクレベルの実際の値は90μlのサンプルを90μlの陰性血清に
置換してアッセイした。
総被検物質濃度はそれぞれ以下の組合せ用いて定量された。
a)非スパイク添加サンプルと低スパイク
b)非スパイク添加サンプルと高スパイク;および
c)非スパイク添加サンプルと低および高両スパイク
単一スパイクでの総抗原濃度の定量は次のように行われた。
非スパイク添加サンプル測定値×スパイク値/(スパイク添加サンプル測定
値−非スパイク添加サンプル測定値)
両スパイクを組合せて用いた総抗原濃度の定量は次のように行われた。
非スパイク添加サンプル測定値×(高スパイク値−低スパイク値)/(高ス
パイク添加サンプル測定値−低スパイク添加サンプル測定値)
表1に示したデータと上述の実際のスパイク値を用いたサンプル1の総抗原濃
度の定量を以下に掲げる。残りの21の基準サンプルについても、同様に定量を
行った。
これらの計算の結果はすべての22のサンプルについて以下の表2にまとめる
。
5.結果
結果は以下の表1および2に示す。表1に示したデータは、両スパイク、低お
よび高スパイクの不存在下、存在下におけるアッセイでの各サンプルについての
HIV-1 p24抗原レベルの測定値である。
表2に示す結果は、表1に掲げたデータと上述のようにして測定
された各スパイクの実際の値から定量された総抗原濃度である。実際に測定され
た値と定量された総抗原濃度との間の比較も示す。
これらのデータは、内因性結合物質たとえば抗体の存在は、アッセイで直接測
定した場合、多くのサンプルで著しく不正確な値を生じることを示している。
本発明の方法は、被検物質を遊離型および結合型で含有するサンプル中の総被
検物質濃度をより正確かつ精密に定量する手段を提供することによってこの問題
を克服するものである。
Claims (1)
- 【特許請求の範囲】 1.サンプル中の総被検物質濃度を測定するにあたり、 (a)サンプル中の遊離被検物質の濃度をアッセイし、 (b)そのサンプルが予めそのサンプルに添加された既知量の被検物質を含有 する場合のサンプル中の遊離被検物質の濃度をアッセイし、この測定を少なくと も1回行い、ついで (c)工程(a)および(b)において得られたアッセイ濃度から総被検物質 濃度を定量する 各工程からなる方法。 2.サンプル中の総HIV抗原濃度を定量するにあたり、 (a)サンプル中の遊離HIV抗原の濃度をアッセイし、 (b)そのサンプルが予めそのサンプルに添加された既知量の被検物質を含有 する場合のサンプル中の遊離HIV抗原の濃度をアッセイし、この測定を少なくと も1回行い、ついで (c)工程(a)および(b)において得られたアッセイ濃度から総HIV抗原 濃度を定量する 各工程からなる方法。 3.HIV−抗原はHIV-1 p24抗原である請求項2に記載の方法。
