Restoring the immunocompetent capacity of T helper cells in HIV-infected patients
Background of the Invention
The present invention relates to IL-IL for administering a drug to a patient infected by the immunodeficiency virus.

Summary of the Invention

Detailed description
The present invention provides an effective means to enhance IL-2 production in T helper cells.
Wherein the level is human immunodeficiency virus
Excessive levels of IL-10 associated with lentivirus infection known as [HIV]
Level that is inhibited by
This chronic and often lethal virus infection results in an impairment in the T helper cell response.
Characterized by equilibrium. This IL-10 comprises a subset of T helper cells
Produced by many different cells. Maintain IL-2 production in HIV-infected patients
It is beneficial. IL-2 is responsible for T cell proliferation and is key to the state of the immune system
It is an indicator.
Decreased IL-2 production is indicated by administration of an appropriate amount of antagonist specific for IL-10.
It is stopped by giving. An antagonist of IL-10 can be expressed using standard mutagenesis methods.
It can be produced by mutagenizing the -10 amino acid sequence. Such a way

Briefly, methods for obtaining αIL-10 antibodies are based on humoral responses in mammals.
Administering an antigen in an amount sufficient to elicit the following. Antibody is mammalian serum
Or collected from depleted lymphocytes, immortalized, and secretes desired antibodies
The desired cells are isolated and cultured for harvest of the desired antibody. To IL-10

Those methods require a suitable source of IL-10 as antigen. Antigen is damaged
Not IL-10 or immunoreactive peptide. Recombinant expression of IL-10 is an antigen

Can be used to obtain an intact or immunoreactive portion of IL-10.
Antibodies for use in the present invention are preferably cognate for the patient,
Thus, additional immunological problems are minimized. Immunocompromised individuals respond to non-autoantibodies
And low reactivity, and therefore non-self derived from cells of the same species
Antibodies are also useful. Different species of antibodies are useful, but possible opposite
To control the immune response
Measures should be taken. For example, if a humanized rat antibody is
Immune response can be minimized.
Antibodies for use in the present invention are typically neutralizing antibodies, and are preferably
Or higher than or greater than the affinity of IL-10 for its natural receptor
It will have near-close coupling constants. It against their corresponding receptors
Antibodies with binding constants that are 100-fold lower than these cytokins are not preferred.
Binding comparisons are performed using standard equilibrium methods. The basic technique is Chapter 25 o

For example, it is suspended in a phosphate buffer. Pharmaceutically acceptable excipients such as lecithin,

If the antagonists are antibodies, they will hit 1 ml of serum for each antibody.
Provides about 1 to 150 μg and preferably 10 to 100 μg of circulating levels of αIL-10
Administered in amounts. The antibody has a half-life of 2-7 days, and repeated administrations
IL-10 levels
Required if the level is less than or equal to Total αIL-10 applied per dose
The amount is between 1 and 10 mg / kg of body weight for the individual antibody.
The method of the invention enhances IL-2 production, and the level approaches 100% normal.
Or more. 50% increase in IL-2 production prior to treatment
Seems to be good. The treatment is performed using T-helper cells in any conventional assay.
