Description
Claims (10)
- 1.少なくとも1個のステムーループ構造を有する選択されたRNAとハイブリ ダイズしうるオリゴヌクレオチドを製造する方法であって、a.該ステムールー プ構造と相補的なオリゴヌクレオチドを選択する;b.選択されたオリゴヌクレ オチドが該ステムーループ構造とシュードーハーフーノットを形成する能力を決 定する;及びc.選択されたオリゴヌクレオチドが該ステムーループ構造とシュ ードーハーフーノットを形成することができると決定された場合に、選択された オリゴヌクレオチドを合成する; ことを含む方法。
- 2.ステムーループ構造がHIVTARエレメントまたはrev応答性エレメン トである請求項1の方法。
- 3.該合成オリゴヌクレオチドが、その5′末端、3′末端、糖の2′位、リン 酸または複素環に反応性部分を連結している請求項1の方法。
- 4.該ステムーループ構造に相補的な複数のオリゴヌクレオチドが選択され、該 複数のそれぞれについて該ステムーループ構造とシュードーハーフーノットを形 成する能力を決定し、そして、オリゴヌクレオチドが該シュードーハーフーノッ ト構造を形成する最大の能力を有するように合成される請求項1の方法。
- 5.少なくとも1個のステムーループ構造を有する選択されたRNAとハイブリ ダイズしうるオリゴヌクレオチドを製造する方法であって、a.該ステムールー プ構造と相補的であり、選択されたRNAとシュードーハーフーノットを形成し うると予想されるオリゴヌクレオチド配列を選択する;及び b.該配列を有するオリゴヌクレオチドを合成する;ことを含む方法。
- 6.少なくとも1個のステムーループ構造を有する選択されたRNAの発現を調 節する方法であって、 a.該ステムーループ構造と相補的なオリゴヌクレオチドを選択する;b.選択 されたオリゴヌクレオチドが該ステムーループ構造とシュードーハーフーノット を形成する能力を決定する;及びc.選択されたオリゴヌクレオチドが該ステム ーループ構造とシュードーハーフーノットを形成することができると決定された 場合に、RNAを選択されたオリゴヌクレオチドと接触させる; ことを含む方法。
- 7.少なくとも1個のステムーループ構造を有する選択されたRNAの発現を調 節する方法であって、 a.該ステムーループ構造と相補的であり、選択されたRNAとシュードーハー フーノットを形成しうると予想されるオリゴヌクレオチド配列を選択する;及び b.オリゴヌクレオチドをRNAと接触させる;ことを含む方法。
- 8.オリゴヌクレオチドのRNAとハイブリダイズする能力を改良する方法であ って、 a.該RNAの少なくとも1個のステムーループ構造と相補的な配列を有するオ リゴヌクレオチドを同定する; b.選択されたオリゴヌクレオチドが該ステムーループ構造とシュードーハーフ ーノットを形成する能力を決定する;及びc.該オリゴヌクレオチドとして、選 択されたRNAとシュードーハーフーノットを形成しうると予想されるものを選 択する;ことを含む方法。
- 9.複数のオリゴヌクレオチドが同定されている請求項8の方法。
- 10.少なくとも1個のステムーループ構造を有する選択されたRNAとハイブ リダイズしうるオリゴヌクレオチドのための配列を選択する方法であって、a. ステムーループ構造と相補的な配列を有するオリゴヌクレオチドを選択する;及 び b.選択されたRNAとシュートーハーフーノットを形成しうると予想される相 補的なオリゴヌクレオチドを選択する:ことを含む方法。
