Abstract
Description
Claims (27)
- 1.受容細胞中へのトランスフェクションの後、ATCC40889の同定特性 を有するヒト免疫不全ウイルス1型(HIV−1)ウイルス株MN−ST1の活 性培養物を産出できる分子クローンの実質的に純粋な製剤。
- 2.ATCC40890の同定特性を有する(HIV−1)ウィルス株BA−L のエンベロープとrevをコードしている配列を含むDNAの実質的に純粋な製 剤。
- 3.MN−ST1のエンベロープ(env)タンパク質をコードしているDNA 断片。
- 4.表IIIに記載の配列を持つ請求項8記載のDNA断片。
- 5.BA−Lのenvタンパク質をコードしているDNA断片。
- 6.表IIIに記載の配列を持つ請求項5記載のDNA配列。
- 7.精製したMN−STlenvタンパク質。
- 8.表IIに記載した配列を持つ請求項7記載のタンパク質。
- 9.精製したBA−Lタンパク質。
- 10.表IIIに示した配列を持つ請求項9記載のタンパク質。
- 11.i)請求項3記載のDNA断片;およびii)ベクター からなるDNA構造。
- 12.更に、revタンパク質をコードしているDNA断片とrev−応答領域 からなる請求項11記載のDNA構造。
- 13.i)請求項5記載のDNA断片;およびii)ベクター からなるDNA構造。
- 14.更に、revタンパク質をコードしているDNA断片とrev−応答領域 からなる請求項13記載のDNA構造。
- 15.組換え的に生産されるMN−STlenvタンパク質。
- 16.組換え的に生産されるBA−Lenvタンパク質。
- 17.請求項11または13に記載の上記組換えDNA構成でもって、上記組換 えDNA分子中にコードしてあるウイルスタンパク質の発現をさせるように、安 定に形質転換してある宿主細胞。
- 18.組換えHIV−1ウイルス株MN−ST1タンパク質を製造する方法であ って、上記ウイルスタンパク質を発現させて単離するように、請求項17記載の 上記の宿主細胞を培養することからなる方法。
- 19.HIV−1感染に対する哺乳動物用のワクチンであって、上記感染に対す る免疫化を誘導するのに充分な量の請求項1記載の上記MN−ST1ウイルス株 の非感染性抗原部分、および医薬的に許容できる担体からなるワクチン。
- 20.HIV−1感染に対する哺乳動物用のワクチンであって、上記感染に対す る免疫化を誘導するのに充分な量の請求項2記載の上記BA−Lウイルス株の非 感染性抗原部分、および医薬的に許容できる担体からなるワクチン。
- 21.更に助剤を含有する請求項19または請求項20記載のワクチン。
- 22.HIV−1感染に対する哺乳動物用のワクチンであって、上記感染に対す る免疫化を誘導するのに充分な量のMN−ST1ウイルス株envタンパク質の 少なくとも5個のアミノ酸、および医薬的に許容できる担体からなるワクチン。
- 23.HIV−1感染に対する哺乳動物用のワクチンであって、上記感染に対す る免疫化を誘導するのに充分な量のBA−Lウイルス株envタンパク質の少な くとも5個のアミノ酸、および医薬的に許容できる担体からなるワクチン。
- 24.上記タンパク質が組換え的に生産されたタンパク質である請求項22また は23記載のワクチン。
- 25.HIV−1感染に対する候補ワクチンを試験する方法であって、上記ワク チンと請求項1記載のMN−ST1ウイルス株を試験哺乳動物に投与し、次いで 上記感染の存在または不存在を検出する段階からなる方法。
- 26.HIV−1活性に影響を与える能力について医薬をスクリーニングする方 法であって、請求項17記載の宿主細胞を、上記ウイルスの上記活性が影響され 得るような条件下で上記医薬と接触させることからなる方法。
- 27.生理学的試料中のHIV−1の検出のための生物検定法であって: i)MN−ST1またはBA−LウイルスからのenVタンパク質の少なくとも 5個のアミノ酸で表面を塗布し; ii)上記表面を上記試料で接触せしめ;次いでiii)上記タンパク質と上記 試料中に存在する上記タンパク質に特異的な抗体との間で形成した複合体の存在 または不存在を検出する段階からなる方法。
