コングルチニンフラグメント、その調整と使用技術分野
本発明は、組み換えDNA技術によるコングルチニンの交差反応物質の調製に関
する。
背景
フングルチニンは、いくつかの細胞外糖タンパク質レセプターと類似構造を有す
るウシ血清タンパク質である。完全な分子は、ジスルフィド結合された三重体が
会合して4価の巨大分子を形成している多量体構造を示す。フングルチニンは、
免疫複合体と結合し得、そして反すう動物の免疫応答での役割をなし得る、カル
シウムイオン依存性のレクチンとして記載されている。しかしながら、タンパク
質の生理学的な厳密な機能は明らかにされていない。これまでの構造研究により
、非常に限定されたアミノ末端配列データに基づいて、コングルチニンがコラー
ゲン様の分子であることが示されている。
糖に特異的に結合する化合物を研究することは、非常に興味深い。糖は特定の分
子または分子のファミリーと結合し得る。S、 au■胚タンパク質Aは、免疫
複合体の単離での使用が見いだされた。その他のレクチンもまた、特定の多糖類

する。
本発明のオリゴペプチドは、少な(とも8個のアミノ酸、通常は、少なくとも約
1211Nのアミノ酸、そして約180個を越えないアミノ酸、さらに一般的に
は約120個を越えないアミノ酸を有する。このオリゴペプチドは、以下の配列

可変領域が各々の示されたアミノ酸によって置換され得るだけでなく、G−X−
Yのモチーフの中のXおよびY位がプロリンまたはヒドロ半シブロリンによって
、および同類の置換物(conservative 5ubstitu口ons
)によって置換され得る。

N−近位領域では、約アミノ酸20位からアミノ酸180位までの領域、さらに
詳細には、アミノ酸25位から60位および60位から178位の領域が特に興
味深い。残基27位から55位までの領域が超可変性を有し、一方60位から1
78位までの領域は可変性が低いことがわかった。超可変領域は、トリプレット
のグリシン部位での変異は10%を越えないで、残りの部位では大きく変化し得
る。通常は、G−X−YモチーフのYは、N−近位領域においてプロリンまたは
ヒドロキシプロリンとしての、アミノ酸を少なくとも約25%、さらに一般的に
は少なくとも50%を有する。オリゴペプチド中の繰り返しの回数は、通常は少
なくとも約3回、そして約55回よりは多くなく、一般的には、約4回から51
回、さらに一般的には約6回から40回の範囲にある。
N−近位領域は、配列の可変性の原因である2つの断続がある、約49回から5
1回のG−X−Yの繰り返しを有することにより特徴付けられる:断続の1つは
C38であり、Gが考えられ、そしてもう1つは、G以外のアミノ酸が考えられ
る部位のG 105である。12個のヒドロキシ−プロリン残基(このうち11
個は不変部位)が、G−X−YのモチーフにおけるY位に見い出され、コラーゲ
ン様の分子の特徴を示す。この配列は、少なくとも3つの潜在的な炭水化物認識
部位を有し、このうち2つはコラーゲン様の可変ドメイン中に(〇−結合された
)、および1つは、C−近位領域中(N−結合された)にある。さらに、N−近
位領域には、R−G−D、種々の細胞の表面に見られるレセプターのインテグリ
ンのスーパーファミリーをL?!、mする通常の認識配列が含まれる。
分子全体の構成からすると、フングルチニンは、補体タンパク質clq、マンノ
ース−結含タンパクlit (MBP)および肺胞表面活性物質関連タンパク質
(PSAP)に類似性を有している。
C−近位領域、特に232位−333位の残基は、MBPおよびPSAPの炭水
化物認識ドメインに顕著な相同性を有する。
フングルチニンの配列相同性は、ニワトリおよびラットの肝細胞のl結合の動物
性レクチン、ヒトのアンアログリコプロティンレセプター、ヒトリンパ球Fcイ
ブンロンレセプター、ラット軟骨プロテオグリカンのマトリックスタンパク質、
hrco ha a ere rina液素性の可溶解性レクチン、マウスリン
パ節帰巣レセプターおよびヒト内皮白血球付着分子(ELAM−1)のレクチン
ドメインにも広がる。全部で15のタンパク質が並んでいる保存部位には、フン
グルチニン配列の237位j09位。
323位および331位の4つのシスティン、および298位のP、307位の
E 、 319位のN、293位のWおよび318位のWが含まれる。
炭水化物結合部位および/または2価のカチオン配位のような潜在的な機能的に
重要な領域は、保存されているようだ。
当該ペプチドの大きさによって、ペプチドは種々の方法で調製され得る。アミノ
酸が約60個より少ないアミノ酸配列では、このペプチドは、現在では入手可能
な自動合成装置、例えば、ABI 43OAペプチド合成装置を使用して合成さ
れ得る。
アミノ酸が約25個よりも多いアミノ酸配列では、組み換えDNA技術が使用さ
れ得る。当該ペプチドをコードするこの配列は、合成でXll製され、天然に存
在する遺伝子から単離され、ゲノム遺伝子またはc DNAのフラグメントとし
て、またはそれらの組み合せとして得られ得る。さらに、分泌のために、この配
列は、生成物を培養上清へ分泌させる/グナル配列をコードする配列を有するコ
ーディング鎖の5−末端に連結させられ得る。多くの発現ベクターが、文献に記
載されており、入手可能であるかまたは容易に調製され得る。発現カセットは原
核細胞および真核細胞用が入手可能である。グリコンレーンヨンのような、当該
ペプチドのプロセッシングが所望される場所では、通常、昆虫、菌類または哺乳
類宿主、および、そのような宿主において真核性のプロセッシングに関連する、
多糖類側鎖を備えるためのプロセッシングに機能するベクターを用いる、真核細
胞用のベクターが用いられる。
当該組成物には、種々の方法により応用が見い出され得る。
C−近位領域からのフラグメントは、糖のレクチンへの結合を阻害するために使
用され、または種々の糖と結合させて、適切な炭水化物構造を有する分子を単離
するため、およびそのような分子を精製をするため等に使用され得る。この分子
は、これら自身であり、ポリソームの一部であり、または細胞と結合して存在し
得る。当該組成物は、相補的炭水化物リガンドの存在を示す特徴付けのために使
用され得る。N−近位配列は、これらのレセプターを有する細胞と結合し得るた
めのレセプターのインテグリンスーパーファミリーへの結合における用法が見い
だされ得る。さらに、超可変額域は、末梢血単核細胞(Reid、 Bioch
em、 Sac、 Trans、 ll:1−12 (1983))のCIQお
よび軟骨細胞のコラーゲ7 (Mollenhauer and Yon De
rMark、 EMBOJl:45−5o、(1983):およびMol 1e
nhauerら、ニ域に結合する細胞を同定する、細胞と結合するための分子認
識部位として働き得る。
当該化合物はまた、この化合物が結合する表面膜レセプターを有する細胞を活性
化するために使用され得る。当該化合物を細胞の活性化に十分な量、例えば1が
ら100■g/■lを、細胞を含む培地に加えることによって、活性化状態に関
連する増殖およびタンパク質の発現を含み、細胞は、活性化される。
以下の実施例は、発明の例証として提供され、限定としては提供されていない。

コングルチニンを、地方の屠殺場からのランの血液から個々に単離した。酵母細
胞壁を処理したタンパク質(Strangら、(1986)前述)を、さらに5
ephaery+ 400で精製した。精製タンパク質の相同性は、5OS−P
AGEによって顕著になり、43にのソングルバンドが観察された。細菌のコラ
ゲナーゼによるコングルチニンの短時間消化によって生じた非コラーゲンのc。
OH−領域のための配列情報は、トリプシンペプチド(図IA−V中のT)およ
び引き続きのコラゲナーゼ消化(図IA−1■)によって生産された、cL2お
よびcL3のフラグメントの特徴付けによって得られた。C00H−領域(CL
I)の配列分析は、完全なフングルチニンのりジン特異性エンドペプチダーゼ処
理によって生じた(図IA−III中のLP)、オーバーラフピングペプチドの
特徴付けによって完了した。
コングルチニンのCOOH−ドメインの改変されていないコングルチニンを、2
5 raM Tris−HCI、10mM CaCl2、p)!7.4中、37
°Cで、15がら3o分間、細菌のコラゲナーゼ(フングルチニン1mgに対し
てコラゲナーゼ25ユニツト)で短時間消化した。生成されたペプチドは、逆相
c4カラムを使用してHPLCで精製した。単離されたCoo)I−末端ペプチ
ドを、N)12−末端アミノ酸配列分析または他のタンパク質分解酵素によって
さらに分画する前に、4−ビニルピリジンで改変した。

リシル結合は、リジン特異性エンドペプチダーゼを使用しr選択1cMWした。
酵素ハ、0.2 M NHaHCOx、pH8,3中で、アルキル化CLIに対
して1:50の比率(v/v) テ、37°(j?4時間使用した。C00H−
末端ドメインのトリプシン消化は、0.2MNHa)ICO3、pH1,9中、
37″で、6時間、基質に対する比率がl:Ioo <v/v)の酵素で、同様
に実施した。
コラゲナーゼドメインでは、大きなペプチドフラグメントが、化学的な方法、そ
の後のタンパク質分解酵素によりまたは罵2の化学的な方法による二次的な開裂
によって生じた。
ランのタンパク質をヒドロ亭ジルアミンで処EJして、llAlおよびHA2
(図IA−1を生成した。3番目のフラグメンl−[fA3は、観察されるよう
にNS2の高可変部位から得られた非開裂タンパク質、あるいはヒドロ亭ジルア
ミンによる不完全な開裂による。HAIとHA2との間のオーバーラッピング配
列は、HA3または完全なコングルチニンから得られた、ペプチドLP2の特徴
付け(図I A−111およびIB−11)によって決定した。同様の目的のた
めに、それぞれ図I B−mおよび−■に示されるように、CNBr開裂のペプ
チド(CBx)および以−ヨードソ安息香酸ペプチド(IBA)を使用した。
LPハ、リジン特異性エンドペプチダーゼ(m)によっテ開裂されたペプチドを
示し;cLは、細菌のコラゲナーゼ消化(■)によって得られたペプチドを示す
。37℃、 30分間の消化テcL1 (167−333)を得た。短時間の消
化(15−20分間)では、トリプシン消化(V)に使用された長いペプチド(
164−333)が生じた。長時間の消化(60分以上)により、CLIは、破
壊され短いペプチドCL2を生じた。さらに長時間の消化(12時間)では、主
にCL3ペプチドを生じた。図18において、■は、完全なコングルチニンのア
ミノ末端配列分析を示す。下線■、■、■およびVは、コラゲナーゼドメインを
配列分析するために使用されたオーバーラフピングペプチドを示す。■は、サー
モリシンによるLP2−火消化(subdigesLion)およびキモトリプ
シンまたはΣ、 aureus V8によるLP3の一次消化を示す。
Thは、完全なフングルチニン由来のLP2のサーモリシンペプチド;Cは、H
A3由来のLP3のキモトリプシンペプチド;sPは、H^3由来のLP3のS
、 aureus V8ペプチドを示す。mは、H4SのCNBr開裂から回収
したCNBrヘブチド(Caw)を示す。5P−Calは、Σ aureus
V8によるCawの一次消化によって生産された。■は、完全なフングルチニン
のWでの開裂によって得られるヨードソ安、豐香酸ペプチド(IBA)を示す。
■は、アルギニン特異性酵素によるタンパク質の消化によって得られた4つのペ
プチドを示す。Txa、Txbおよびryは、完全なフングルチニンに由来した
。Txcは、lA3に由来した。Leeら、Arch、Biochem、Biυ
立v−241:577−5f19 (1985); MahoneyおよびHe
rmondson、 11順堕」工注」18:3810−3814 (1979
)。
5P−Cal (lA3から得た)を、始めにIAIとlA2との間のオーバー
ラッピング配列を決定するために使用した。5P−CBlは、しかしながら、N
52を現さなかった。残基37−54の間の5P−CBIと配列相同性を有する
、Txc(lA3由来)もまた、852を現わさなかった。完全なフングルチニ
ンから得られたIBAを、続いて分析した。このペプチドは、残基43−56の
間の5P−CBIとの配列相同性および残基53−71に沿ってlA2と同じ配
列を現した。これは、C52およびG53を現したが、N52は現さなかった。
DS2は、化学開裂反応および/または精製工程の間に生じ得るNの脱アミノ化
生成物なので、LP2は、その後のサーモリシンによるLP2の一次消化によっ
て、完全なフングルチニンから生じた。
m1crobore C1gカラムによって分離されたTh1a、 Th1bお
よび丁hlcは、残基23−42の間の配列相同性を示すが、アミノ酸残基31
.34.37および40ての可変部位を示した。一連のThlペプチドもまた、
残基31.33.34および40を除いて、5P−CBIと配列相同性を示した
。5P−CB+は、エドマン分解サイクルで多数のPTl(−アミノ酸を示した
。ペプチドTh2は、残基49.51および52を除いて、43−52の間の残
基で、ペプチドTxb、 Txc、 5P−CBlおよびIBAが配列している
。このTh2ペプチドは、ヒドロキシルアミン開裂部位N52およびGS3を確
定し、さらにIAIと1(A2との間の配列のオーパーラ、ブを明確にしている
。ペプチドTh3は、アミノ末端残基Fを有し、26サイクルから配列分析され
た。このペプチドは、アミノ末端領域HA2と同一の配列を示した。Tt+4は
、Vをアミ/末端残基として有し、P61に代わる[61以外はTh3と同じ配
列を示した。LP2ペプチドのRP−HPLCによる回収は、予測されたよりも
低かった。これはおそらく、lA2−含有LP2が使用された条件下で回収され
なかったことを示す。
上記の結果は、LP2に存在する超可変配列の広がりを示した。
ヘフチドLP3もまた、可変配列を示した。アルギニンプロテアーゼ処理された
完全なコングルチニンから得られたペプチド(Ty)は、lA2中のS82、に
84およびN93と比較して、TF中のlA2、P84およびT93以外は、)
lA2(残基8O−98)のアミノ末端領域と同一の配列を示した。キモトリプ
シンペプチドCIと+IA2、LP3、ryおよびSF3との比較により、残基
93、ioo、103および110の変化が示された。ペプチドC2a、 Ca
bおよびC2cは、配列相同性を有したが、この領域で超可変性を示す残基12
1.123.124および+26では異なっていた。C2とSF3 (H^3由
来)との比較は、135位での可変残基を示した。さらに可変配列は、C3a、
C3b、 C3cおよびSF3 (l(A3由来)の間に観察された。
N−近位領域の配列の変化および超可変性は、次のページに示す。
当該組成物は、種々の方法でN−近位領域との結合に関連する、特にフングルチ
ニンのN−近位領域の超可変配列の1つの配列に関する、特定の配列を同定する
ために使用され得る。従って、個々のコラーゲン様の配列のレセプターが、単離
され得る。さらに、フングルチニンの保存C−近位領域に結合する種々の炭水化
物が単離され得る。従って、当該組成物は、アフィニティクロマトグラフィーに
、相補性のリガンド結合部位をフングルチニン等との結合から離すために使用さ
れ得る。当該組成物の他の使用には、細胞クロマトグラフィー(アフィニティ細
胞分1tl)、細胞活性の調節等が含まれる。より小さいフラグメントを使用す
ることによって、同一タンパク貫での機能の多重性を防ぐことができるので、個
々の特性が単離および同定され得る。

Claims (8)
- 1.ウシコングルチニンのコラーゲン領域またはレクチン領域内に含まれる少な くとも8個の、そして180個を越えないアミノ酸配列を含有するペプチド。
- 2.前記ペプチド配列が、コラーゲン領域の超可変領域内に含まれる、請求項1 に記載のペプチド。
- 3.前記ペプチド配列が、コラーゲン領域の可変領域内に含まれる、請求項1に 記載のペプチド。
- 4.前記ペプチド配列が、レクチン領域内に含まれる、請求項1に記載のペプチ ド。
- 5.ウシコングルチニンと特異的に結合する糖類を有する化合物を単離する方法 であって、該方法が、該化合物を支持体に結合された請求項4に記載のペプチド 配列に結合する工程; 該支持体に結合された化合物を結合されていない化合物から分離する工程;そし て 結合された化合物を単離する工程を包含する、方法。
- 6.ウシコングルチニンのフラグメントに特異的に結合する表面膜レセプターを 有する細胞を単離する方法であって、該方法が、 細胞混合物を支持体に結合された請求項4に記載のペプチド配列に結合する工程 ; 該支持体に結合された細胞を結合されていない細胞から分離する工程;そして 結合された細胞を単離する工程を包含する、方法。
- 7.前記細胞が造血細胞である、請求項6に記載の方法。
- 8.ウシコングルチニンのエピトープと反応する表面膜レセプターを有する細胞 を活性化する方法であって、該方法が、該表面膜レセプターを有する細胞を該エ ピトープを有する請求項1に記載のペプチドに結合する工程を包含し、その結果 、該ペプチドが該表面膜レセプターに結合し、そして該細胞が活性化される、方 法。
