ヒトの腫瘍に附帯した新規な抗原に
対する新規なモノクローナル抗体
発明の技術分野
本発明は、新規なモノクローナル抗体及び新規な抗原、並びにヒトの癌腫細胞と
反応性を有するこの新規なモノクローナル抗体の調製法及び用途に関する。より
具体的には、本発明のモノクローナル抗体は、結腸及び肺の癌腫を含む種々のヒ
トの腫瘍と附帯した新規な細胞表面の抗原との反応性を有するものである。
本発明のモノクローナル抗体は、生体内及び生体外での悪性癌腫の検査の如き臨
床的診断目的のいずれにも適している。更に、本発明の抗体は、例えば、腫瘍細
胞と反応させたり、化学療法剤、毒素、免疫学的応答変更因子及び放射性同位元
素(これらに限定されるものではない)を含む抗腫瘍効果を有する種々の薬剤の
標的−選択的キャリアーとしての結合体において治療的用途に適している。また
、本発明の抗原も治療的及び診断的目的に適している。
発明の背景
毎年、癌腫によって数百万人が死亡している。例えば、肺の癌腫は、男性におけ
る癌による死亡原因の大多数を占めており、女性における癌による死亡の最大原
因である乳癌を追い抜いている。
癌腫の殆どのケースでは、疾患の早期の時点で徹底的に除去しなければ、化学療
法及び放射線療法によっても治癒しない。従って、黒色腫及び肉腫の如き他の悪
性腫瘍と同様に、乳癌層、結腸癌腫、卵巣癌腫及び肺癌腫の診断及び治療方法が
非常に必要とされているのである。
癌腫に附帯した抗原と反応性を有するモノクローナル抗体は公知である〔例えば

公知のモノクローナル抗体の殆どは多くのタイプのヒトの癌腫と反応性を有する
が、僅かな抗体は、例えば、肺、乳房、卵巣、結腸、胃または膵臓といった身体
の特定の器官由来の癌腫と反応する。標的の抗原は、通常、糖蛋白または糖脂質
である〔例えば、ヘルストロム(Hellstrom)らによるCancer
Re5earch、 46:3917−23 (1986) ;及びフィフク(
Fink)らによるProg、 Cl1n、 Pathol、。
9:I21−33 (1984)を参照のこと〕。例えば、特定のタイプの癌腫
上に存在する糖蛋白抗原と反応性を有するモノクローナル抗体として、米国特許
第4.737.579号(非小細胞肺癌腫に対するモノクローナル抗体);米国
特許第4.753.894号(ヒト乳癌に対するモノクローナル抗体):米国特
許第4.579.827号(ヒト消化器系癌に対するモノクローナル抗体);及
び米国特許第4.713.352号(ヒト腎臓癌腫に対するモノクローナル抗体
)がある。あるモノクローナル抗体は、粘液と思われる高分子量の抗原と反応す
る。
例えば、モノクローナル抗体872.3は、多くの異なる癌腫上で選択的に発現
する分子量1.000kdより大きい腫瘍−附帯胎児腫瘍性糖蛋白抗原を認識す
るものと思われる。かくして、B7z3は、乳癌層の84%、結腸癌腫の94%
、卵巣癌腫の100%及び非小細胞肺癌腫の96%と反応することが明らかにさ
れてきた〔ジョンストン(Johnston)によるActa Cytol、、
l (5):537−56 (1987)及びシュロム(Schlom)らに
対して付与された米国特許第4.612.282号を参照のこと〕。同じく、モ
ノクローナル抗体KC−4は、結腸、前立腺、肺及び乳癌層の如き多くの癌腫上
で発現される約400〜500kdの蛋白質抗原を認識するC米国特許第4.7
08.930号を参照のこと〕。
一定の腫瘍細胞に附帯していると考えられている糖脂質抗原と反応性を育するモ
ノクローナル抗体も開示されている。例えば、ヤング(Young)らによるJ
、 Bxp、 Med、、 150:1008−19 (1979)は、アジア
c+ (asialo) GMt 、キルステン(Kirsten) vウス肉
腫ウィルスによって形質転換されたBALB/c3T3細胞のマーカーとして定
着した細胞表面グリコスフィンゴリピド抗原に特異的な2つのモノクローナル抗
体の調製を開示している。二−ブ(Kniep)らによるJ、 In+muno

更に、特定のタイプの癌腫細胞に見い出された糖脂質抗原と反応性を育するモノ
クローナル抗体として、ローゼン(Rosen)らによるCancer Re5
earch、 44:2052−61 (1984) (ヒト小細胞肺癌腫に対
するモノクローナル抗体);バルキ(VarkDらによる第4.579.827
号(ヒト結腸腺癌層に対するモノクローナル抗体)に開示されたものがある。ま
た、ヒト非小細胞肺癌腫、乳癌腫及び結腸癌腫の表面上で発現される糖質抗原を
認識するL6モノクローナル抗体を開示しているヘルストロム(Hellstr
om) ラG::ヨるProc、 Nat’ 1. Acad、 Sci、 I
JsA、 83ニア059−63 (1986)も参照のこと。
種々の腫瘍細胞上に見出される抗原に対して反応性を発揮する更なるモノクロー
ナル抗体が非常に必要とされている。これは、同じ腫瘍塊に向けられた異なるモ
ノクローナル抗体の組み合わせを用いることを診断及び治療においてしばしば必
要としている殆どの腫瘍の抗原異質性のためである。その上、正常な組織との反
応性を有さないかまたは極めて微弱であると同時に、広範囲な腫瘍との高度な反
応性を発揮するモノクローナル抗体は一般的ではない。そのような抗体は明らか
に作用である。
従って、種々の腫瘍によって高濃度に発現される抗原と反応性を存するモノクロ
ーナル抗体は、より進んだ癌治療方法の開発のためだけでなく、癌の早期発見、
癌の転移のより正確な特定、癌患者の免疫学的監視にも有用なものとなるだろう
。また、新規な細胞表面分子に対するモノクローナル抗体は、癌ワクチンの形で
免疫原を調製する価値があり、しかも、例えば、ホルモンまたは成長因子の受容
体としてまたは細胞内及び細胞間連絡に他の状態で含まれる分子として重要な細
胞性作用も持っているかもしれない分子を更に特定するのに使用できると思われ
る。その抗原ですらそれ単独で酵素的または成長因子的活性を育しているかも知
れないのである。
本発明は、ヒトの腫瘍細胞、特に肺及び結腸癌腫由来の細胞に附帯した細胞表面
糖蛋白抗原(C1抗原)の決定基に特異的であるモノクローナル抗体(CI)を
提供するものである。従って、本発明の抗体は、抗体C1によって特定されたC
1抗原を発現する腫瘍の診断及び治療に有用であり得る。本発明のC1抗体は、
クラスIgG及びIgG1サブクラスに属するものであり、正常なヒト細胞には
有意な反応性を示さない。
本発明の抗体は、ヒト肺組織及び他のヒト組織における悪性状態の存在を測定す
る生体外での診断方法において用いてもよい。
その方法は、抗体C1との反応性を有する66.000ダルトンの01抗原糖蛋
白の特質を有する抗原の存在について、組織を試験する方法を含んでいる。例え
ば、組織を、CI抗原の特質を存する細胞附帯抗原の決定基を特定する本発明の
CIモノクローナル抗体、この抗体の機能的等価物またはフラグメントと接触さ
せてもよく、前記抗体と抗原決定基のいかなる相互作用も検出される。かかる方
法の1つは、そのような細胞を含む疑わしい検体中での癌腫細胞の存在を測定す
ることを含んでいる。
検体は、その検体中に存在することができる他のタイプの細胞からそのような細
胞を区別することができるモノクローナル抗体と接触される。該接触は、抗体が
そのような細胞と結合できる条件下で行われる。接触後、抗体の該細胞への結合
があったかなかったかが決定される。この結合は、検体中の癌腫細胞の存在また
は不存在に関連している。一般に、検体は該モノクローナル抗体についての標識
化された特定の結合パートナ−と接触される。この標識は検出可能なシグナルを
発することができるものである。
また、該モノクローナル抗体それ自身を標識することも可能である。
もう1つの診断法は、検出可能なシグナルを発する試剤で標識化された本発明の
精製した抗体または該抗体のフラグメントを患者に投与することによって、生体
内で腫瘍の位置を確認することを含む。次いで、その位置は、外部シンチグラフ
ィー、xmi層撮影法または放射性核スキャン法を使用して探知される。この方
法は、疾患の程度に関して癌患者の状態を推し量ったり、治療効果をモニターし
たりするよりよい方法をも提供する。
本発明は、また、Ct抗体及び類似の抗体が腫瘍細胞表面で高濃度に発現するC
I抗原と反応することができるので、治療への応用をも含む。本発明のモノクロ
ーナル抗体を腫瘍治療のための組成物のl1i1!に使用することもできる。該
組成物は、薬学的に許容し得る非経口賦形剤と共に治療学的に有効量の該抗体を
含む。
また、本発明の抗体は、化学療法剤、毒素、免疫学的応答変更因子及び放射性同
位元素(これらに限定されるものではない)を含む抗腫瘍効果を有する種々の作
用剤のキャリアーとして、免疫複合体で使用することもできる。
本発明は、また、分子量か約66.000ダルトンであり、しかもアミノ末端ア
ミノ酸配列
X−E−L−T−[−L−H−T−N−D−V−H−S−R−L−E−Q−T−
S−X(Xは未確認アミノ酸を表す)を有することによって特徴付けられる新規
なCI抗原及びその等価物であって、抗体CI及びこの抗原と結合する抗体のク
ラスによって特定されるものをも含む。
本発明は、一定の腫瘍に対して免疫になるワクチンとして精製されまたはクロー
ン化されたCI抗原を使用する方法を含む。
発明の詳細な説明
ここに記載された本発明がより十分に理解されるように、以下に詳細に説明する
。
本発明は、ヒト腫瘍細胞、特に肺及び結腸の癌腫由来の細胞上に位置する抗原(
CI抗原)と特異的に反応するC1で示される新規なモノクローナル抗体、C1
モノクローナル抗体の調製法及び該抗体を使用する診断法及び治療法に関する。
該C1抗体は一定の範囲の腫瘍とは反応するものの、正常なヒトの組織または黒
色腫、リンパ腫の如き他のタイプの腫瘍との反応性は実質的に示さない。
本発明は、更に、C1抗原で示されかつヒトの肺または結腸の腫瘍に主として附
帯した新規な細胞表面糖蛋白抗原及び該C1抗原を使用する方法に関する。
本発明のモノクローナル抗体は、ハイブリドーマ融合技術またはEBV−不滅化
法(immortalization)テクノロジーを利用する技術によって調
製され得る。
ハイブリドーマ融合技術は、コーラ−(Kohler)及びミルスタイン(Mi
lstein)によって初めて導入された〔コーラ−及びミルこれらの技術は、
動物(例えば、マウス)の中での望ましい免疫欠乏反応(つまり、抗原の生成)
を引き起こすために動物内にある免疫原(例えば、精製した抗原または抗原を保
持している細胞または細胞抽出物)を注入することを含む。例えば、胸膜滲出液
由来のヒト肺癌腫細胞、外植されたヒト非小細胞肺癌腫由来の培養細胞(NSC
LC) 、または正常胎児の肺由来の細胞またはそのような細胞由来の溶解産物
を免疫原として使用することができる。後記の実施例1において、H3059で
示される結腸のヒト腺癌由来の膜試料及び結腸癌腫セルライン3347由来の細
胞が免疫原として使用されている。例えば、その膜試料を、マウスの体内に注入
し、充分な時間が経過した後、該マウスを犠牲にして0抗体−生成リンパ球を得
ている。抗体−生成細胞は、リンパ結節、膵臓及び感作された動物の末梢血から
誘導してもよい。膵臓細胞が好ましい。マウスリンパ球は高い割合で以下に説明
するマウスのミエローマと安定な融合を行う。ラット、ウサギ及びカエルの体細
胞を使用することも可能である。望ましい免疫グロブリンをコードする膵臓細胞
染色体は、一般にポリエチレングリコール(PEG)の如き融合剤の存在下で、
膵臓細胞をミエローマ細胞と融合させることによって不滅化される。いかなるミ
エローマセルラインも標準的技術に従って融合パートナ−として使用することが
できる。例えば、P3−NS 1/1−Ag4−1、P3−X63−Ag3.6
53またはSp2/○−Ag14ミエローマラインがある。これらのミエローマ
ラインはメリーランド州のロックビルにあるアメリカン・タイプ・カルチャー・
コレクション(ATCC)から入手することができる。
次いで、目的とするハイブリドーマを含む、得られた細胞を、HAT培地の如き
選択培地で生育させる。融合しなかった親ミエローマまたはリンパ球細胞は最終
的にそこで死滅する。ハイブリドーマ細胞だけが生き残り、限界希釈条件下で成
長することかでき、分離したクローンが取得できる。該ハイブリドーマの上溝を
、例えば、免疫化に用いた抗原を使用する免疫学的試験に付して、望ましい特異
性を有する抗体をふるい分ける。次いで、陽性のクローンが限界希釈条件下でサ
ブクローン化され、生成したモノクローナル抗体が単離され得る。モノクローナ
ル抗体から他の蛋白及び他の不純物を除くために、モノクローナル抗体の単離及
び精製には多様な慣用的方法がある。モノクローナル抗体の精製に通常使用され
る方法には、硫酸アンモニウム沈澱法、イオン交換クロマトグラフィー法、及び
アフィニティクロマトグラフィー法がある〔例えば、シラ(Zo la)ら、M
onoclonal Hybridoma Antibodies:Techn

方法で生成したハイブリドーマは、当該分野で公知の技術を使用して生体外また
は生体内(腹水中)で増殖され得る〔一般的には、フィフク(Fink)らによ
る上記文献の123頁の図6−1を参照〕。
一般に、個々のセルラインは、例えば、実験室の培養容器中で生体外で増殖して
もよく、単独の特異的モノクローナル抗体を高濃度で含有する培地が、デカンテ
ーション、濾過または遠心分離によって採取され得る。また、モノクローナル抗
体の収量は、初めの融合に、0抗体及びミエローマ細胞を供給するために用いら
れたタイプの組織適合動物の体内に該ハイブリドーマのサンプルを注入すること
によって向上させ得る。融合細胞ハイブリッドによって生成した特異的モノクロ
ーナル抗体を分泌する腫瘍が、注入された動物の体内で増える。腹水または血清
の如き該動物の体液が高濃度でモノクローナル抗体を提供する。上記の文献でコ
ール(Cole)らが議論しているように、ヒトハイブリドーマまたはEBV−
ハイブリドーマを使用する場合は、マウスのような動物の体内に注入される異種
移植体による拒絶反応を避けることが必要である。免疫不全のまたはヌードのマ
ウスを用いてもよく、または、まず、ハイブリドーマを、生体外で培養された固
形の皮下腫瘍として、照射ヌードマウスの体内に継代接種し、次いで、大量の特
異的ヒトモノクローナル抗体を分泌する腹水腫瘍が大きくなった、初めに接種し
た照射ヌードマウスに、腹腔内注入してもよい(コールらによる上記の文献を参
照のこと〕。
マウスの抗体で治療した患者はヒト抗マウス抗体を生じているので、一定の治療
への適用には、キメラ(マウス−ヒト)モノクローナル抗体またはヒトモノクロ
ーナル抗体がマウスの抗体に対して好ましいかも知れない〔シャウラ−(Sha
wler)らによるJ。

抗原と反応性を有するキメラマウス−ヒトモノクローナル抗体は、例えば、キメ
ラ抗体の調製のために最近発展した技術によって調製することができる。

ト起源の新規な抗体は適当な生物学的機能を有するC1抗原から作ることができ
る。例えば、ヒトモノクローナル抗体は、例えば、本発明のCI抗原の如き抗原
を使用して、生体外で該抗原に対してヒトリンパ球を感作させ、次いで、ポアベ
ック(Borrebaeck)らにより説明されているように、マウスまたはヒ
トのリンパ球で感作された該抗原感作リンパ球をEBV−形質転換またはハイブ

好ましい態様によれば、CIで示される本発明の抗体は、後記の実施例1に記載
しているように、免疫原として結腸腺癌滲出細胞由来の膜及び結腸癌腫セルライ
ン3347由来の細胞を使用するハイブリドーマ技術を介して生成される。CI
抗体を生成するC1ハイブリドーマは、メリーランド州ロックビルのATCCに
寄託されており、次のように特定されている。
CI 受託番号No、: HB 9803該C1抗体は、IgG1サブクラスに
属する。この抗体は、腫瘍細胞、特に結腸及び肺癌腫由来の細胞に非常に強力な
反応性を発揮する。該CI抗体は、リンパ腫セルライン、CEMSMOLT−4
、B細胞リンパ腫うインP3HR−1、及び黒色腫細胞とは検出できる程度の結
合をしない。
更に、本発明の抗体は、主要な器官、例えば、腎臓、膵臓、肝臓、皮膚、肺、乳
房、結腸、脳、甲状腺、心臓、リンパ結節または卵巣由来の線維芽細胞、内皮ま
たは上皮細胞の如き正常なヒトの組織とは、免疫組織学的に検出できるいかなる
結合をも生じない。また、該抗体は末梢血白血球とも反応しない。このように、
この抗体は、腫瘍細胞の一定の範囲に対するその特異性において及び正常細胞に
比較して腫瘍細胞に高度に特異性である点において、公知の大半の抗腫瘍抗体〔

本発明は、上記で説明したC】抗体及び該抗体の活性結合領域を含有する、Fa
b、F (ab)*及びFvフラグメントの如きいかなるフラグメントも包含す
るものである。かかるフラグメントは、当該分野で充分に確立されている技術を
用いて、C1抗体から作ることができる〔例えば、ローゼアクラス(Rouss

更に、本発明は、CI抗体と同じ抗原決定基に結合することができ、かっ、該決
定基で結合するためにC1抗体と競い合うことができる抗体を包含するものであ
る。これらには、C1抗体と同じ抗原特異性を有するものの、種の起源、アイソ
タイプ、結合親和性または生物学的機能(例えば、細胞障害作用)において相違
する抗体が含まれる。例えば、本発明の抗体のクラス、アイソタイプ及び他の突

これらの抗イデイオタイプ抗体は、免疫原としてCI抗体を使用して作ることが

本発明のモノクローナル抗体によって認識されるC1抗原は、腫瘍細胞、特に結
腸及び肺の癌腫由来の細胞に独特な新規な細胞表面糖蛋白抗原を含む。CI抗原
は、ポリアクリルアミドゲル電気泳動でのイムノブレシピテーションに付した場
合、約66.000ダルトンの分子量を有する。
該新規なCI糖蛋白抗原のアミノ末端アミノ酸配列は次の通りである。

本発明のモノクローナル抗体は、また、生体外及び生体内の両方において、該C
I抗体が特異的に反応するC1抗原を保持するヒト癌腫を検出する診断への応用
に有用である。生体外での診断法は、当該技術分野において公知であり〔例えば
、ロスによる上記文献及びクブチック(Kupchik)による上記文献を参照
のこと〕、腫瘍細胞(例えば、ヒト組織、細胞または切除された腫瘍検体)の免
疫組織学的検出方法または腫瘍附帯抗原(例えば、血液サンプルまたは他の生物
学的分泌物)の血清学的検出法を含むものである。
免疫組織学的技術は、腫瘍組織検体の如き生物学的検体を本発明の抗体と接触さ
せ、次いで、その抗原と複合体を形成した抗体の検体の存在を検出することを含
んでいる。検体とのかかる抗体−抗原複合体の形成は、組織中での腫瘍細胞の存
在を示している。
検体上での抗体の検出は、免疫ペルオキシダーゼ発色法、アビジン−ビオチン(

えば、免疫ペルオキシダーゼ発色法は、以下の実施例3に記載しているように、
肺及び結腸の癌腫の01抗体に反応性かあること、及び正常なヒト組織検体の抗
体には反応性かないことを証明するのに使用された。
血清学的診断技術は、癌に罹っていると考えられる患者の血清または他の分泌物
の中に分泌されたかまたは「放たれた」腫瘍附帯抗原の検出及び定量を含む。こ
のような抗原は、放射性免疫測定法(RI A)または酵素結合免疫吸着測定法
(ELISA)の如き当該技術分野で公知の技術を利用して、体液中で検出され
得る。これらの方法では、「放たれた」抗原と反応性を有する抗体が、分泌物サ

本発明の抗体は、また、ヒトの腫瘍を検出するための生体内診断法への応用にも
有用である。そのような取り組みの1つに、検出可能なシグナルを発する適当な
イメージング剤で標識化した抗体を使用する腫瘍イメージング技術による生体内
での腫瘍の検出法がある。そのような試剤で抗体を標識化するイメージング剤及
び操作はよく知られている〔例えば、ウエンセル(Wensel)及び化した抗
体は、放射性核スキャン法の如き技術によって検出することができる〔例えば、
ブラッドウェル(Bradwell)ら。

本発明のCI抗体を使用して、多くの生体内での治療的応用が可能である。腫瘍
細胞を標的にするのに単独で使用してもよいのに加えて、該抗体をヒトの癌を治
療する適当な治療剤と結合させて使用してもよい。例えば、該抗体を、治療剤を
癌の部位に送達するために治療薬または毒素と結合または連結させて使用しても
よい。そのような治療剤を抗体に結合させる技術は、よく知られ−56,Ala
n R,Li5s、Inc、、 (1985) ;ヘルストロム(He l I
s trom)ら。

体は、細胞を生体外でそれにさらした場合、簡単に内在化しないので、例えば、
直接化学結合または組み換えDNA技術を使用して抗体を一定の酵素と結合させ
ることによって、化学療法剤を腫瘍細胞に向かわせるのが好ましいかも知れない
。そのような結合体か腫瘍に集まると、該酵素は、該結合体が腫瘍細胞と結合し
たあとに投与されるある不活性(非毒性)プロドラッグを活性な抗癌剤に変化さ
せることができるものであまた、該抗体は、それが腫瘍部位に集まると、幾つか
の細胞径(cell diameters)を殺す高エネルギー放射線源、例え
ば、1211の如き放射性同位元素と結合し得る〔例えば、オーダー(0rde
r)ら、Monoclonal Antibodies for Cancer
Detection and Therapy。
ブラッドウィンら編、 pp、 303−16. Academic Pres
s (1985)を参照のこと〕。いまひとつの態様に従えば、該CI抗体に、
米国特許出願第4.676、980号にセーガル(Segal)によって記載さ
れた腫瘍細胞の治療のための抗体異質結合体(antibody hetero
conj−ugate )を形成させるために第2の抗体を結合させてもよい。
本発明のCI抗体についての他の治療への応用としては、補体の存在下であるか
、または抗体−薬剤結合体または抗体−毒素結合体の一部としてかのいずれかで
、癌患者の骨髄由来の腫瘍細胞を除去する用途がある。このアプローチに従えば
、自己由来の骨髄は、患者に戻された該抗体と骨髄による治療によって、半ビポ
で除去され得る〔例えば、ラムゼイ(Ram5ay)ら、 J、 C11n。
Immunol、、 8 (2):81−88 (1988)を参照のこと〕。
更にまた、先に説明したように、本発明のキメラなまたは他の組み換えCI抗体
は、治療に使用することができる。例えば、抗腫瘍活性を有する第2蛋白、例え
ば、リンフ才力インまたはオンコスタチン(oncostatin)の少なくと
も1つの機能的に活性な部分に結合したCI抗体の少なくとも抗原結合領域を含
む融合蛋白は、生体内でヒトの腫瘍を治療するのに使用することができる。
更に、C1の抗原結合領域がヒトFC領域、例えば、IgG1と結合しているキ
メラC1抗体を、抗体依存細胞性の細胞障害作用または補体媒介細胞障害作用を
促進するのに使用することができる。更にまた、当該技術分野で公知の組み換え
技術を、該抗体の結合特異性の1つがCI抗体の結合特異性である二重特異性抗
体を構築するのに使用することができる〔例えば、米国特許第4,474、89
3号を参照のこと〕。
最後に、CI抗体の抗イデイオタイプ抗体を、活性腫瘍免疫法及び腫瘍治療法に
おいて使用することができる〔例えば、ヘルストロム(Hellstrom)

従って、本発明は、医薬組成物、ヒトの腫瘍の治療のための組み合わせ及びヒト
の腫瘍の治療方法を包含する。例えば、本発明は、薬学的に有効量のCt抗体及
び薬学的に許容し得るキャリアーを含むヒトの腫瘍を治療するための医薬組成物
を包含する。該組成物は、未修飾CI抗体、治療剤(例えば、薬剤、毒素、酵素
または第2の抗体)と結合したCI抗体及び組み換え体(キメラまたは二重特異
性CI)の形でのCI抗体のいずれのC1抗体を含んでもよい。該組成物は、更
に、癌腫を治療するための他の抗体または結合体(例えば、抗体カクテル)を含
んでもよい。
本発明の抗体組成物は、静脈内、腹腔内、経口、リンパ内投与または腫瘍内への
直接投与を含む通常の様式を使用して投与することかでき、これらに限定される
ものではない。静脈内投与か好ましい。
本発明の抗体組成物は、溶液剤または懸濁剤、錠剤、ビル剤、散剤、坐剤、重合
体マイクロカプセル剤またはマイクロ小胞剤、リポソーム剤、及び注射または注
入可能な溶液剤を含む様々な服用形態をとり得る。好ましい形態は投与及び治療
への応用の様式に依存する。
また、本発明の抗体組成物は、好ましくは、ヒト血清アルブミン;イオン交換体
;アルミナ;レシチンニリン酸塩、グリシン、ソルビン酸の如き緩衝物質;及び
塩または硫酸プロタミンの如き電解質の如き当該技術分野で公知の通常の薬学的
に許容されるキャリアー及びアジュバントも含む。
本発明の組成物の投与及び投薬の最も有効な様式は、疾患の重さ及び経過、患者
の健康状態及び治療に対する応答、及び担当医師の判断に依存する。従って、該
組成物の投薬量は個々の患者に応じて定められるべきである。さもなければ、本
発明の抗体組成物の有効な投薬量は、約1〜約5000 mg/m”としてもよ
い。
抗原C1として示される本発明の新規な抗原も、治療的応用に使用することがで
きる。該抗原は、腫瘍から精製することも、組み換えDNA技術によって作るこ
ともできる〔ブラウン(Brown)らによる係属中の米国特許出願第827.

8号。これらは参照文献として本明細書に含まれる。〕。C1抗原をコードする
遺伝子は、CI抗原のmRNAをまず豊富化する方法によってクローン化するこ
とかできる。このような方法によって、未完成鎖上のC1抗原決定基を認識する
抗体とのイムノアフィニティーク口マトグラフィーによって、ポリソーム(mR
NAリポソームと未完成ポリペプチド鎖からなる)を精製することができる。該
mRNAは、例えば、C1抗体でのイムノブレシビテーションによって単離され
、そのcDNAが適当な発現ベクター中でクローン化される。
また、ある発現ベクターを使用してcDNAライブラリーをふるい分けするのに
、CI抗体またはCI抗原に対する抗血清を使用できるかも知れない。精製また
はクローン化されたC1抗原は、免疫原として単独で投与しても、適当な免疫学
的アジュバントと一緒に投与してもよい。
精製またはクローン化されたC1抗原は、本発明の方法において、一定の腫瘍に
対して免疫するワクチンとして使用してもよい。
そのようなワクチンの調製操作は、当該技術分野で公知であるローン化された腫
瘍附帯抗原、例えば、C1抗原の発現のために 、構築されるのである。該組み
換えウィルスに感染した細胞は、宿主不適合性抗原及び免疫ウィルス性蛋白とと
もに、該細胞の表面で腫瘍抗原を発現するであろう。このことは、腫瘍拒絶にお
いて中心的役割を演する細胞性免疫の誘導に有利である。好適なウィルス、例え
ば、ライニス(Wyeth)痘癒ワクチン〔ニューヨーク市健康株協会(New
York C1ty Board of Health 5train) )
のプラーク精製ウィルスから誘導したワクシニアウィルスを、ワクシニアウィル
スr7.5K」プロモーター〔ツー(Hu)らによるムViro1..62:1
76−180(1988) )制御下、C1抗原をコードする配列を含む組み換
えウィルスを構築するのに使用する。次いで、その組み換えウィルスを、癌を防
ぐワクチンとして静脈内に投与してもよい。
ここに記載した発明がより深く理解されるように、以下の実施例を記載する。こ
れら実施例は説明だけを目的としたものであり、いかなる意味においても、本発
明の範囲を限定するものと解釈されないことが理解されるべきである。
実施例I
CIモノクローナル抗体の調製
本発明のモノクローナル抗体を、以前にイエ−らにより上記のProc、Nat
’1. Acad、 Sci、(USA)、 (1979)に記載された/%イ
ブリドーマ融合技術を使用して調製した。以下の実施例で使用した全てのセルラ
インは、固形の腫瘍または滲出液のいずれかとして、ヒトから得た腫瘍のサンプ
ルからワシントン州シアトルのオンコジーンで発育させたものである。即ち、H
3O59で示されるヒトの結腸腺癌の膜試料を使用して4回、及び結腸癌腫セル
ライン3347由来の細胞を使用して3回、3力月齢B A L B / cマ
ウスを免疫した。該膜試料は以下のようにして調製した。腫瘍は腹水滲出液から
得た。10m1の溶解緩衝液(lysig buffer) (19,8mlの
脱イオン水+0.1mlの0.2 M NaHCO,+ O,l mlの0.2
MPMSF(プロテアーゼ抑制剤)エタノール溶液〕をその細胞に加え、25〜
50回氷の上で粉砕した。次いで、該混合物を15m1試験管に移し、0°C,
2000XGで2分間回転した。上溝を分離し、5mlのセルロース管に移した
。細胞溶解を確認するため、ペレットを顕微鏡でチェックした。次いで、上溝を
限外遠心分離器で、4°C,37000XGて15分間遠心分離に付した。次い
で、上清を吸引し、ペレットを少なくとも1mlのPBS中に再分散した。該分
散体を5mlの試験管に移し、氷上で1分間音波処理した。i : ioo希釈
液を作り、ブラッドフォード(Bradford)の操作〔ブラッドフォード、
Analytic Biochemistry、72:248−254(19
76))を使用して蛋白濃度を測定した。膜試料(ま、レブコ(Revco)冷
凍装置(−70°C)で0.2mlのアリコートとして冷凍保存した。
以下のようにして、マウスに7回注入した。
第1注入、100μg膜試料+100μgムラミルジペプチド(MDP)150
μm+50μm不完全フロインドアジュバントを4か所に皮下注射し;第2注入
:100μgの膜試料を4か所に皮下注射し;第3注入:100μgの膜試料を
腹腔内注射し:第4注入: iooμgの膜試料を腹腔内注射及び4か所に皮に
皮下注射し;
第6注入:107個の3347細胞を腹腔内注射及び4カ1所に皮下注射し;
第7注入=106個の3347細胞を腹腔内注射及び4カA所に皮下注射した。
最終の免疫化から3日後、その膵臓を取り出して膵臓細胞を培地中に分散させた
。次いで、ポリエチレングリコール(PEG)ヲ使用シテ、該牌UA細胞ヲAT
CCCRL 1580. P3 X 63− Ag8.653細胞ト融合サセ、
イエーラニよる上記のProc、Nat’1. Acad、 Sci。
(USA)、に記載されているように、HAT選択培地中のマイクロ1ノブター
ウエル中で成育させた。該混合物を接種して単一の融合細胞またはクローンから
発生する低密度培養物を生成させた。

983)に記載された分析法と類似のELISA分析法を使用して、結腸癌セル
ライン3347の直接結合活性についてふるい分けした。この分析法に従えば、
その抗原(ふるい分けされる抗体はこの抗原と反応性である)をマイクロタイタ
ブレート上に固定化し、次いで、ハイブリドーマの上清とインキュベートする。
もし、上溝が目的とする抗体を含有しているなら、その抗体は固定化抗原と結合
し、抗免疫グロブリン抗体−酵素結合体及び光学密度で測定可能な変換を起こす
該酵素に対する基質との添加によって検出されるであろう。
この例として、96ウ工ル組織培養プレート(テンプ1ルノジのコースタ−1M
A)中に結腸癌細胞を配分し、加湿37°Cインキュベーター(5%CO□)内
で一夜インキユベートした。次1.zで、新しく調製した100μmの1.0%
グルタルアルデヒドで該細胞を固着して、最終的なウェル濃度を0.5%とし、
室温で15分間インキュベートした後、IX PBSで3回洗浄した。次に、該
細胞を、PBS中5%BSAで1時間ブロックし、再びPBSで3回洗浄した。
次いで、ハイブリドーマ培養物からの上溝を100μm/ウェルで加え、該ウェ
ルを室温で1時間インキュベートし、該細胞をPBSで3回洗浄した。次に、0
.1%BSA及びPBSで希釈したヤギ抗マウスホースラディツシュペルオキシ
ダーゼ(Zymed、 CA)を加え、100μl/ウエルの濃度にした。室温
で1時間または37°Cで30分間、該反応混合物をインキュベートし、次いで
、該細胞をPBSで3回洗浄した。次いで、0−フェニレンジアミン(OPD)
を100μm/ウェルで加え、暗所にて室温で5〜45分間インキュベートした
。該細胞に結合した抗体を、ウェル中で10〜20分間内に起きた色の変化(こ
よって検出した。100μl/ウエルのH,So、を加えて該反応を停止させ、
ダイナチック(アレキサンドリア、VA)マイクロエリザ・オートリーダーで4
92nmの吸収を読み取った。
次に、カバーグラスに付着し、かつ、フルオレセインで発色した3347細胞を
使用して、該抗体を蛍光分析法で試験した。殺菌したカバーガラスのついた24
ウ工ル組織培養プレートに、各ウェルに付き1〜2X10’個の密度で培養した
3347細胞を配分し、75〜90%の集合体(conf 1uency)に成
長させた。細胞を3%バラホルムアルデヒドで5分間を要して固定し、結合緩衝
液で洗浄した。結合緩衝液でl:2に希釈した250μmの抗体含有上溝液を各
ウェルに添加し、室温または37℃で30分間インキュベートした。次いで、細
胞を3回洗浄した。250μlのFITC結合ヤギ抗マウスIgG抗体(バーリ
ンガムのTAGOlCA)の最適希釈液を各ウェルに加え、前記のようにインキ
ュベートした。次いで、洗浄工程を繰り返し、カバーグラスを除去して顕微鏡の
スライドに載せた。蛍光顕微鏡を使用して発色パターンを読み取った。
ELISA及び蛍光分析法の両方で結腸癌腫セルラインについて陽性のウェルか
ら得た上溝を、後記の実施例■で記載したように、3347細胞ペレツト、結腸
癌腫組織及び正常な腎臓、肝臓、及び膵臓組織について、免疫組織学的技術によ
って更に試験した。
完全細胞または精製した可溶性抗原または細胞抽出物を、固定化抗原として使用
してELISA分析法を行うことができるということに留意すべきである。可溶
性抗原または細胞抽出物を抗原として使用した場合は、まず最初に、該抗原をP
BS中50μl/ウェルでプレート上に配分し、分析を始める前に該プレートを
室温で一夜インキユベートした。完全細胞を抗原として使用する場合は、新鮮な
ものを使用しても、固定した後に使用してもよい。
いずれの場合においても、まず、細胞を培地中の100μl/ウエルに10’個
の割合で配分し、−夜または37°Cのインキュベーター(5%Cod)内で集
合体が成育するまでインキュベートした。
かくして、結腸癌セルラインと結合しかつ正常組織と結合しない抗体を生成した
ハイブリドーマを選別し、クローン化し、生体外で増大させ、更に抗体特異性を
試験した。ヒト結腸癌と反応性を存する抗体を生成したそれらハイブリドーマを
、再クローン化し、増大させ、初回感作された(Pristane−prime
d) 3力月齢BA L B / cマウスに注入した。そこで、それらは腹水
腫瘍として生育した。
この操作に続いて、ハイブリドーマセルラインを得、クローン化し、腹水腫瘍と
して生育させるためにマウスに注入した。上記の如く、該ハイブリドーマはAT
CCに寄託している。腹水中に分泌された抗体を、蛋白A−または蛋白G−セフ
ァローズ上で精製するか〔例えば、エイ(Ey)らのImmunochemis
try、 15:429−436 (1978)を参照のこと〕、または、セフ
ァクリル(5ephacryl)S−300でのゲル濾過によって精製した。精
製したCI抗体を更に特徴付けるために使用した。
実施例■
CIモノクローナル抗体の特徴付は
アイソタイプの確定
Clハイブリドーマによって生成した免疫グロブリンのクラスを決定するために
、以下の技術を利用した。
a)オクレルロ二−免疫拡散法
C1ハイブリドーマ細胞の上溝のアリコートを、25%寒天プレートの中央ウェ
ルに配置した。単一特異性ウサギ抗マウスIgアイソタイプ抗体(サザン・バイ
オテクノロジー、バーリンガム、AL)を外側のウェルに配置し、該プレートを
37℃で24〜48時間インキュベートした。次いで、沈降線を読み取った。
b)ELISAアイソタイピング
ダイナチックイムロン96−ウェルプレートに、ヤギ抗マウスIg抗体をjμm
/ml濃度でPBS中50μl/ウェルで被覆し、カバーをしたまま4℃で一夜
放置した。該プレートをPBS/ツイーン20,0.05%で洗浄した。プレー
トを洗浄後、CI /%イブリドーマの上溝を加え、室温で1時間インキュベー
トした。ウシ血清アルブミン(BSA)を含有するPBS/ツイーン20で洗浄
後、ペルオキシダーゼ(Zymed)と連結した単一特異性ヤギ抗マウスIg−
HRPアイソタイプ抗体と37℃で2時間インキュベートした。洗浄後、該プレ
ートを0.1Mクエン酸塩緩衝液(pH4,5)中の1mg/m1o−フェニレ
ンジアミン及び0.03%H10,でインキュベートした。ダイナチックELI
SAプレート・リーダー上で、490及び630nmの光学密度を測定した。
これらの操作に基づき、C1モノクローナル抗体はIgG1のアイソタイプであ
ることを決定した。
非イオン性界面活性剤での浸透の前または後に細胞に結合している抗体を測定す
ることによって、抗原の細胞レベル下(5ubce−11ular)の位置を決
定した。完全培養細胞の細胞表面に結合している抗体を、ヘルストロムらによる
Cancer Re5earch、 46:3817−3923 (1986)
に記載されているように、蛍光励起細胞分離捕集装置(FAC3)を使用して直
接蛍光によって同定した。即ち、FAC3細胞分離捕集装置を使用して結合分析
を行うために、l×IO−の培養細胞をIMDM培地〔グランドアイランドのギ
ブコ(Gibco) 、NYE中の15%胎仔ウシ血清(FBS)中に分取して
、総容量を試験管当たり500μlとした。該細胞をセロフユージ(Serof
uge)で1.5分間遠心分離し、上溝を分離した。lOμm/mlで、かつ、
紅藻素で標識されたモノクローナル抗体100μlを各試験管に添加し、次いで
、その内容物を混合し、氷上で30分間インキュベートした。セロフユージで1
.5分間遠心分離することによって、該反応混合物をlolの15%FBS/I
MDMで3回洗浄した (3回目の洗浄後に各試験管をプロットした)。各ペレ
ットを500μmのPBSに再分散した。各サンプルをクールターエビクラス(
Coulter Epics) CF A CS ニかけ、平均蛍光強度(MF
I)を測定した。MFMから、線形蛍光当量(linear fluore−s
cence equivalent ) (L F E)を決定した。陰性コン
トロールのLFEで割った各試料のLFEから、特異性によって発色した細胞対
コントロール抗体の明度の比率(1,0=蛍光の差なし、zO=蛍光が2倍明る
い、等々)を得た。
結合データを以下の表1に示した。CI抗体をFITCと結合させる試みがうま
(いかないときは、蛍光性結合剤として紅藻素を使用した。

ナル抗体は、肺及び結腸癌腫セルラインと反応するが、TまたはBリンパ腫ライ
ンとも正常な末梢血白血球とも反応しない。3つの黒色腫ラインのうち2つにつ
いての弱い反応性も観察された。
免疫組織学
ガーリジエス(Garrigues)らの[nt、 J、 Cancer、 2
9:511−15(1982)により修正された、rmmunochemist
ry、 pp、104−69. JohnWiley & 5ons、 New
York (1979)に記載されたL−A、スターンベルが−(Stern
berger)のPAP技術を、凍結した組織切片の免疫組織学的研究に用いた
。手術でこれらの試験に使用する組織を得、液体窒素中で予備冷却したイソペン
タンを使用して、切除から4時間以内に凍結した。その後、該組織を使用するま
で液体窒素中または一70°Cで保存した。凍結切片を調製し、風乾し、アセト
ンで処理し、再乾燥した(ガーリジエスらの上記文献を参照のこと)。組織学的
評価に使用する切片をヘマトキシリンで発色した。非特異性バックグランドを減
少させるため、PBSで115に希釈した正常ヒト血清で該切片を予備インキュ
ベートした(ガーリジエスらの上記文献を参照のこと)。10%正常ヒト血清及
び3%ウサギ血清の溶液で、マウス抗体、ウサギ抗マウスIgG1及びマウスP
APを希釈した。ウサギ抗マウスrgGcジャレッツビル(Jar−ettsv
ille)のスターンベルガーーメイヤー免疫化学会社、MD)は1150希釈
で使用した。特別に精製したP A P 2 mg/mlを含有するマウスペル
オキシダーゼ−抗ペルオキシダーゼ複合体(PAP、スターンベルガーーメイヤ
ー免疫化学会社)は】/80希釈で使用した。
発色操作は、例えば、CI抗体の如き特異的抗体またはコントロール抗体のいず
れかでひと続きの切片(serial 5ecHons)を2.5時間処理し、
該切片を1150に希釈したウサギ抗マウスTgGと室温で30分間インキュベ
ートし、次いで、該切片をl/80に希釈したマウスPAP複合体に室温で30
分間曝すことにより行った。抗体で各々の処理を行ったあと、そのスライドをP
BS中で2回洗浄した。
免疫組織化学的反応は、新たに調製した0、 05 M ) IJス緩衝液(p
H7,6)中の0.5%3,3°−ジアミノベンジジン・4塩酸塩(セントルイ
スのシグマ化学会社、MO)及び0.01%H20zを水中の1%Q s Oa
に20分間曝すことによって、発色を強くした。該切片を水で濯ぎ、アルコール
中で脱水し、キシレン中でクリヤにし、そして、スライド上に載せた。他の切片
もヘマトキシリンで発色した。
該スライドを暗号を付してそれぞれ評価し、暗号化したサンプルを別々の観察者
がチェックした。典型的なスライドは、微分干渉コントラストオブチクラス〔ゼ
イスーノマルスキー(Zeiss−Nomarski) )を使用して写真に撮
った。抗体発色の程度は、0(非反応性)、+(若干陽性の細胞が僅かにある)
、++ (少なくとも1/3の細胞が陽性) 、+++ (殆どの細胞が陽性
)、++++ (全ての細胞が強く陽性)として評価した。十と0の発色間の差
は、十と士十の発色間の差よりも明確ではないので、++またはそれより高い程
度の発色が、「陽性Jと考えられる。
新生細胞及びストロマ細胞の両方が、腫瘍サンプル中に観察された。ストロマ細
胞は全くかまたは腫瘍細胞よりも極めて弱い程度しか発色しないので、記録され
た発色は腫瘍細胞のものである。
以下の表2は、モノクローナル抗体を使用した、種々の腫瘍及び正常組織検体の
免疫組織学的発色を表したものである。表から明らかなように、CI抗体はヒト
結腸癌腫及び肺癌腫と反応するが、乳癌腫または黒色腫由来の細胞とは反応せず
、試験した卵巣癌腫サンプルだけが陽性であった。CI抗体は、試験したいかな
る正常ヒト組織とも反応性を示さなかった。

実施例■
C1抗体によって認識されるCI抗原の精製結腸癌腫3347細胞、及び肺癌腫
2964細胞からCI抗原を単離し、イムノアフイニテイクロマトグラフイーで
部分的に精製した。SDS −PAGEによって精製して均−性にし、5DS−
ポリアクリルアミドゲルから、膜上への電気溶出(electroeiutio
n )または電気プロット(electroblotting) lこよって回
収した。
電気泳動のあと、クーマシー・ブリリアント・ブル−で5DS−ポリアクリルア
ミドゲルを発色させ、次し1で、余分な発色剤を取り除いた。発色したCI抗原
ノくンド(Mr=6. 6000)をカミソリの刃で切り取り、電気溶出に付し
た。
によって記載されているように、ミニ−トランスプロット・エレクトロフォレチ
ック・トランスファー・セル(Mini−TransblotElectrop
horetic Transfer Ce1l ) (/<イオラ・ソド(Bi
orad)研究所、リッチモンド、CA)を使用して、イモピロン膜(Immo
b−ilon membrane) (ベッドフォードのミリボア(Mi 1l
ipore)社、MA)上へ電気プロットすることによっても、SDS〜ポリア
クリルアミドゲルから、CI抗原を回収した。酸膜をクーマシー・ブリリアント
・ブルーで発色し、余分な発色剤を取り除き、そして、次のアミノ末端配列分析
のために、発色したCI抗原バンド(Mr=66.000)をカミソリの刃で切
り取った。
配列分析
33p[1101の3347細胞由来の抗原、49pmolの2964細胞由来
の抗原、及び6 pmolの2964細胞由来の抗原と共に、Ct抗原の3つの
処理品について自動エドマン分解を行った。
CI抗原のアミノ末端配列は次の通りであった。

免疫学的特徴付は
ウェスタン・プロット分析
トービン(Towbin)らによりProc、 Nat’1. Acad、 S
ci、(USA)。
76:4350−4354 (1988)に記載されているように、イムノアフ
ィニチイ精製CI抗原を、5DS−PAGE (10%アクリルアミド)に付し
、ニトロセルロース膜〔キーン(Keene)のシュライバー(Schleic
her) ・アンド・シュエル(Schell) 、NH)上に電気ブトットし
た。第2抗体としてのアルカリフォスファターゼ結合ウサギ抗マウスIgG(ラ
イルのICNバイオメディカルズ、IL)及び色素体としての5−ブロモ−4−
クロロ−3−インドリルホスフェート・p−トルイジン塩とp−ニトロブルーテ
トラゾリウム・クロライド〔バイオラッド(Biorad)研究所〕を使用して
、CI抗原を免疫検出した。免疫検出(Lmmunodetection)は、
Mr=6’6.000での主要なバンドがCI抗体で特異的に発色することを示
した。
放射線免疫沈澱(Immunoprecipitation)10%の透析した
胎児ウシ血清を追加したRPM11640培地(グルコース不含有RPMI 1
640)中で、37℃で4時間インキュベートすることにより、3347細胞を
3H−グルコサミンで代謝的に標識化した。20mM)リス−HCl緩衝液、p
H7,5,100mMNaC1,1mMEDTA、0.5%NP−40、PMS
F (] Ou 1/ml)アプロチニン(10tt 1/ml)で該細胞ペレ
ットを抽出した。CI抗体で4°Cで1時間細胞溶解産物をインキュベートする
こによって、C1抗原を免疫沈澱した。該抗原−抗体複合体をヤギ抗マウスIg
G及びパンソルビン〔カルバイオケム(Calbiochem) 、サンディエ
ゴ、CA)で沈澱させた。還元条件下及び非還元条件下で、洗浄した免疫沈澱物
をSDS −PAGEによって分析し、EN’HANCE”を該ゲルにしみこま
せた後、蛍光間接撮影で目に見えるようにした。
J、 Exp、 Med、、 134:242−264 (1971)でビテッ
タ(Vitetta)らが記載しているラクトペルオキシダーゼ法によって、全
て肺の腺癌から誘導された2964細胞、2707細胞、CH,T2細胞、及び
2981細胞を1251で表面標識化した。CI抗原は、CI抗体、ヤギ抗マウ
スIgG及びパンソルビンで118 I−標識化細胞溶解産物から免疫沈澱した
。還元条件下で、SDS −PAGEによって該免疫沈澱物を分析し、オートラ
ジオグラフィーで目に見えるようにした。
CI抗体は約66.000〜68,000のMrでCI抗原を特異的に沈澱させ
た。これらのデータは、CIモノクローナル抗体によって認識される抗原決定基
が、約66.000のMrで独特の一本鎖糖蛋白上に位置していることを示して
いる。該CI抗原は、様々な腫瘍細胞、特に肺癌腫瘍及び結腸癌腫瘍に附帯して
いるのである。
先に記載したように、本発明の精神及び範囲から逸脱することなしに、本発明に
ついて多くの修飾及び変更を加えることが可能である。ここに記載した具体的な
態様は実施例として与えただけで、本発明は、請求の範囲の文言によってのみ限
定されるものである。

Claims (35)
- 1.ハイプリドーマセルラインATCCNo.HB9803によって産生するモ ノクローナル抗体であって、ヒト腫瘍細胞の細胞表面糖蛋白抗原上の決定基に結 合する抗体、及び前記抗体の機能的等価物、結合フラグメント及び免疫複合体。
- 2.前記腫瘍細胞が、癌腫細胞である請求項1記載のモノクローナル抗体。
- 3.前記癌腫細胞が、肺及び結腸の癌腫細胞からなる群から選ばれたものである 請求項2記載のモノクローナル抗体。
- 4.検出可能なシグナルを生ずることのできる標識と結合した請求項1記載のモ ノクローナル抗体。
- 5.標識が、放射性核種、酵素、蛍光剤及び発色基からなる群から選ばれたもの である請求項4記載のモノクローナル抗体。
- 6.ハイプリドーマセルラインATCCNo.HB9803によって産生するモ ノクローナル抗体であって、ヒト腫瘍細胞の細胞表面糖蛋白抗原上の細胞決定基 に結合し、前記抗原がポリアクリルアミドゲル電気泳動で測定したときに分子量 が約66,000ダルトンであり、かつ、以下のアミノ末端アミノ酸配列を有す ることによって特徴付けられる抗原である抗体、及び前記抗体の機能的等価物、 結合フラグメント及び免疫複合体。 ▲数式、化学式、表等があります▼ (Xは未確認アミノ酸を表す)
- 7.治療学的に有効量の請求項6の抗体を薬学的に許容し得る非経口賦形剤と共 に含む腫瘍治療用組成物。

- 9.クラスIgGに属する請求項8記載のモノクローナル抗体。

- 13.ハイプリドーマセルラインATCCNo.HB9803によって産生する モノクローナル抗体からなる請求項12記載のモノクローナル抗体。

- 15.マウス−ヒト抗体である請求項12記載のモノクローナル抗体。
- 16.薬学的に許容し得る非経口賦形剤と共に治療学的に有効量の請求項12記 載の抗体を含む腫瘍治療用組成物。
- 17.ヒトの腫瘍を検出するための免疫学的検定法であって、a)ヒト腫瘍細胞 に附帯した細胞表面糖蛋白抗原と反応性を有するモノクローナル抗体を、腫瘍細 胞のサンプルと混合すること、ここで、前記抗原はポリアクリルアミドゲル電気 泳動で測定したときに分子量が約66,000ダルトンであり、かつ、以下のア ミノ末端アミノ酸配列を有することによって特徴付けられる抗原であり、また、 前記抗体は検出を可能にするために標識されている ▲数式、化学式、表等があります▼ (Xは未確認アミノ酸を表す) ;及び b)前記抗原が附帯した腫瘍細胞に結合した前記標識されたモノクローナル抗体 を測定すること、 を含む方法。
- 18.前記モノクローナル抗体が、ハイプリドーマセルラインATCCNo.H B9803によって産生する抗体である請求項17記載の免疫学的検定法。
- 19.前記抗体が、放射性核種、酵素、蛍光剤及び発色基からなる群から選ばれ た標識で標識化されている請求項17記載の免疫学的検定法。
- 20.腫瘍を検出する方法であって、請求項1、6、8または12記載のモノク ローナル抗体をヒト組織または分泌物サンプルと接触させ、次いで、前記抗体と 前記サンプル中の全ての抗原的に相当する腫瘍細胞またはその抗原決定基との相 互作用を検出する方法。
- 21.前記腫瘍細胞が肺癌腫細胞であり、ヒト組織が肺組織である請求項20記 載の方法。
- 22.前記腫瘍細胞が結腸癌腫細胞であり、ヒト組織が結腸組織である請求項2 0記載の方法。
- 23.前記モノクローナル抗体と前記腫瘍細胞との相互作用が、免疫組織学的発 色法によって検出される請求項20記載の方法。
- 24.生体内においてヒトの腫瘍を位置確認する方法であって、a)請求項1、 6、8または12のモノクローナル抗体を精製すること: b)前記抗体を放射線標識化すること;c)適当なキャリアーでヒトの患者に前 記抗体を投与すること;及び d)該モノクローナル抗体を、外部シンチグラフィー、X線断層撮影法または放 射性核スキャン法によって位置確認すること、を含む方法。
- 25.腫瘍の治療のための免疫療法であって、a)請求項1、6、8または12 のモノクローナル抗体を精製すること; b)前記モノクローナル抗体を細胞毒、毒素、または放射性薬品と結合させるこ と;及び c)適当なキャリアーでヒトの腫瘍患者に前記結合モノクローナル抗体を投与す ること、 を含む方法。
- 26.前記モノクローナル抗体が、抗−イディオタイプ抗体である請求項25記 載の方法。
- 27.モノクローナル抗体を産生する連続セルラインであって、前記モノクロー ナル抗体が、ヒト癌腫細胞の細胞表面糖蛋白抗原上の決定基と結合する能力によ って特徴付けられるモノクローナル抗体であり、癌腫細胞またはその免疫原決定 基で免疫されたマウス及びマウスミエローマ細胞から誘導されたリンパ球のハイ プリドーマを含む連続セルライン。
- 28.モノクローナル抗体を産生する連続セルラインであって、前記モノクロー ナル抗体が、ヒト癌腫細胞の細胞表面糖蛋白抗原上の決定基と結合する能力によ って特徴付けられるモノクローナル抗体であり、癌腫細胞を有するヒト及びミエ ローマ細胞から誘導されたリンパ球のハイプリドーマを含む連続セルライン。
- 29.ヒト癌腫細胞の細胞表面糖蛋白抗原上の決定基と結合することのできる、 ハイプリドーマセルラインATCCNo.HB9803モノクローナル抗体。
