This application relates to a medical patch used for the treatment and prevention of angina, heart failure, and other cardiac diseases. Conventionally, nitroglycerin has been used as a coronary vasodilator for the treatment and prevention of angina pectoris attacks and other cardiac diseases, and has been administered in a timely manner as intravenous injections, sublingual tablets, ointments, etc. Supervenous injections, which have a short duration of effect, can only be performed by a qualified person.
Furthermore, the latter has drawbacks such as being troublesome in use. Some drugs are known that can be applied transdermally by applying them to the skin, but they generally have complex structures and are rarely satisfactory. In view of the above, the inventors conducted various studies and discovered that a fiber layer composed of water-absorbing fibers and non-water-absorbing fibers as a carrier for nitroglycerin has the property of being able to adjust the release rate of nitroglycerin.
The present idea is to provide a fiber layer composed of a mixed fiber of water-absorbent fibers and non-water-absorbent fibers on the adhesive-coated surface of the base material, and to impregnate this with the required amount of nitroglycerin. By changing the content ratio of water-absorbing fibers and non-water-absorbing fibers that make up the above-mentioned fiber layer, the duration of the effect of nitroglycerin can be appropriately adjusted, making it possible to use it effectively for a long time, and to easily use it when needed. It can be used for In the following, an example will be described. A rubber-based, acrylic-based, silicone-based, or other adhesive layer 2 is applied to a required portion of the upper surface of a base material 1 formed into an appropriate shape using a material such as a synthetic resin film or a metal foil laminated synthetic resin film. A fiber layer 3 having a special structure composed of water-absorbing fibers 31 and non-water-absorbing fibers 32, with the adhesive layer on the peripheral surface of the surface of the base material where the adhesive layer is present. is provided, and the fiber layer is impregnated with a required amount of a predetermined amount of nitroglycerin-containing solution 4. The upper surface of this nitroglycerin-containing fiber layer is coated with an impermeable coating 5 such as a synthetic resin film, a metal foil laminated synthetic resin film, etc., which has releasability to the adhesive layer.
The peripheral edge portion of the adhesive layer is adhered to the adhesive layer exposed on the circumferential surface of the base material 1, and the nitroglycerin-impregnated fiber layer is hermetically covered. Water absorbent fibers constituting the fiber layer include, for example, viscose rayon, copper ammonia rayon, polyvinyl alcohol fiber, cotton, etc., and non-water absorbent fibers include synthetic fibers such as polyester, polyethylene, and polypropylene. There is. The ratio (weight) of both these fibers is water absorbent fiber: non-water absorbent fiber = 1:10 to 10:1, particularly preferably 1:
10-12:10. These mixed fibers can be used in the form of nonwoven fabrics, woven fabrics, other fabrics, fleeces, slivers, and other forms of suitable shapes. Alternatively, non-woven fabrics, woven fabrics, other fabrics, fleeces, slivers, etc. made of water-absorbing fibers and non-water-absorbing fibers may be combined in the above-mentioned proportions, and woven fabrics, etc. may be formed by combining the above-mentioned separately spun fabrics, etc. It may be woven by combining threads in the above proportions. The patch 6 shown in Fig. 1 is made of aluminum foil with polyethylene film laminated on both sides and has a diameter of about 50 mm.
An annular rubber adhesive layer 2 with a width of about 5 mm is formed on the periphery of a base material 1 with a diameter of
Non-woven fabric approximately 0.5 mm thick and approximately 40 mm in diameter (weight approximately 160 mm) composed of 80% (by weight) and 20% (by weight) rayon.
g/m 2 ) and impregnated with 0.5 g of polyethylene glycol solution containing 2% nitroglycerin.
A coating 5 of aluminum foil laminated polyethylene film was adhered and sealed to the adhesive layer on the periphery of the base material, and contained 10 mg of nitroglycerin. One or more appropriate numbers of these patches can be sold commercially by sealing them in a bag made of aluminum foil, aluminum foil laminated polyethylene film, synthetic resin film, or the like. The above patch was applied to a mongrel dog weighing about 7 kg after shearing, and the concentration of nitroglycerin in the blood of the mongrel dog was measured over time. The results are shown in the following table.

[Table] The present invention has the above structure, and as the content ratio of water-absorbing fibers in the fiber layer impregnated with nitroglycerin increases, the retention effect on nitroglycerin increases, and the amount released per unit time increases. On the other hand, as the content ratio of non-water-absorbing fibers increases, the retention effect on nitroglycerin decreases, resulting in a larger release amount per unit time and a shorter release time. Therefore, by changing the content ratio of water-absorbing fibers and non-water-absorbing fibers, the amount of nitroglycerin retained, the amount released per unit time, and the release time can be easily adjusted, and the releasable coating can be peeled off. Coupled with the fact that it can be easily applied by itself, an appropriate amount of nitroglycerin can be efficiently absorbed through the skin, allowing prompt treatment in response to the onset of symptoms. In addition, if during application, it is necessary to discontinue use due to side effects such as headaches, this can be easily prevented by peeling it off the skin, making it extremely effective for the treatment and prevention of cardiac diseases such as angina pectoris.
