Description
本願は、2018年8月1日に出願されたPCT出願番号PCT/CN2018/097912号の優先権を主張し、その全体が本明細書に記載されているかのように参照により本明細書に組み込まれる。
本開示は、その様々な態様の中で、5,6,7,4’-テトラメトキシフラボン(「テトラ-O-メチルスクテラレイン」または「TMS」)、3,5,6,7,3’,4’-ヘキサメトキシフラボン(「ヘキサメトキシクェルセタゲチン(hexamethoxyquercetogetin)」または「HMQ」)、またはそれらの混合物を含む組成物、および甘味料または甘味増強剤としてのそれらの使用を提供する。
非カロリー高甘味度甘味料の使用は、小児肥満、II型糖尿病、および関連する疾患に対する健康上の懸念のために増加している。したがって、例えば、グラニュー糖(スクロース)または高フルクトースコーンシロップ(HFCS)など、従来の甘味料よりも高い甘味を有する甘味料が求められている。しかしながら、多くの非カロリーまたは低カロリー甘味料は、不快なオフフレーバーを有し得るかつ/または予期せぬ、そして決して望ましくない甘味プロファイルを有し得る。
第1の態様では、本開示は、テトラ-O-メチルスクテラレイン、ヘキサメトキシクェルセタゲチン、またはそれらの混合物を含む組成物を提供する。いくつかの実施形態では、組成物は、0.001~20ppmの範囲の濃度で4w/v%のスクロースベースに添加された場合、4w/v%のスクロースベースの甘味を増強する。いくつかの実施形態では、組成物は、約2ppmの濃度で4w/v%のスクロースベースに添加される。

b.オレンジ油の蒸留残渣のシクロヘキサン溶液に等量のメタノールを添加し、液/液抽出を行う工程;
c.工程(b)で得られたオレンジ油の脱脂された蒸留残渣を含むメタノール画分を収集し、収集したメタノール画分を乾燥させる工程;
d.収集したメタノール画分に酢酸エチルを添加し、得られた溶液をシリカゲルカラム(フラッシュカラム、120g、I.D.36.6×H207)に負荷する工程;
e.シリカゲルカラム(120g、I.D.36.6×H207)をシクロヘキサン(2000mL)で溶出することで開始し、1:1のシクロヘキサン:酢酸エチル(1500mL)の比で溶出を終了し、最後の2つの画分を別々に収集して乾燥させて、グラジエント溶出を実行する工程、
f.収集した最後から2番目の画分を分取C18(2)カラムに適用し、テトラ-O-メチルスクテラレインおよびヘキサメトキシクェルセタゲチンを溶出させる工程;および
g.収集した最後の画分を分取FPF(2)カラムに適用し、テトラ-O-メチルスクテラレインを溶出させる工程
を含む方法によってオレンジ油の抽出物から精製される。
a.蒸留残渣をエタノールに溶解する工程;
b.エタノール溶液を濾過し、濾過したエタノール溶液を収集する工程;および
c.濾過したエタノール溶液を濃縮して、テトラ-O-メチルスクテラレインとヘキサメトキシクェルセタゲチンとの混合物を得る工程
を含む方法によってオレンジ油の蒸留残渣から得られる。
a.オレンジ油の蒸留残渣をエタノールに溶解する工程;
b.エタノール溶液をシリカゲルカラムに添加する工程;
c.シリカゲルカラムをシクロヘキサンで溶出することで開始し、4:1のシクロヘキサン:エタノールの比で溶出を終了し、副画分を収集して、グラジエント溶出を実行する工程;
d.収集した副画分を、逆相カラムを備えた分取HPLCに適用する工程;および
e.ポリメトキシフラボンを逆相カラムから溶出させる工程
を含む方法によってオレンジ油の抽出物から精製される。
健康的である、すなわち、非カロリーであり、非発癌性であり、かつ糖尿病患者にとって理想的である、代替的な(高甘味度)甘味料および甘味増強剤は、従来のカロリー甘味料のレベルを低下させることを可能にし、したがって、同等の甘さの従来のカロリー甘味料よりも少ないカロリーを提供するため、必要とされている。さらに、異味、例えば、苦味または金属味を含まない甘味増強剤が求められている。
いくつかの態様では、本開示は、前述の態様およびその実施形態による組成物を含む配合物を提供する。これらの配合物では、かかる組成物は、非晶質固体、結晶性固体、粉末、タブレット、液体、キューブ、グレーズ、またはコーティング、粒状物、担体/粒子に付着またはコーティングされたカプセル化の形態、湿潤もしくは乾燥、またはそれらの任意の組み合わせを含むが、これらに限定されない、任意の適切な形態をとり得る。
特定の態様では、本開示は、消耗品の甘味を伝達、増強、または改善するための、前述の態様および実施形態による組成物の使用を提供する。
実施例1:本明細書に提示されたいくつかの態様によるポリメトキシフラボンを含む組成物の精製
HMQおよびTMSの精製:オレンジ油の蒸留残渣60gを、200mLのシクロヘキサンに溶解した。等量のメタノールを添加し、液液抽出を行った。メタノール画分を収集し、「抽出物1」と名付けた。
HMQ、TMS、HMQとTMSとの混合物を含むポリメトキシフラボンの精製:オレンジ油の蒸留残渣100gを、50(w/w)%の水性エタノール1900gに添加し、固液抽出を行った。抽出した溶液を濾過し、減圧下で乾燥させた。乾燥した抽出物は、ポリメトキシフラボンの混合物を含んでいた。
G1.甘味付けまたは甘味増強用組成物であって、テトラ-O-メチルスクテラレイン、ヘキサメトキシクェルセタゲチン、またはそれらの任意の混合物を含み、天然に存在しない組成物である、組成物。

- 前記組成物が、固体の形態であって、甘味料、甘味増強剤、または香味料として使用するための粉末濃縮物である、請求項1から4までのいずれか1項記載の組成物。
- 1種以上の追加の甘味料をさらに含む、請求項1から5までのいずれか1項記載の組成物。
- 請求項1から6までのいずれか1項記載の組成物を含む、卓上用甘味料組成物。
- 請求項1から6までのいずれか1項記載の組成物を含む、包装された食品または飲料製品。
- 前記組成物を消耗品に導入することにより前記消耗品の甘味を伝達、増強、または改善するための請求項1から6までのいずれか1項記載の組成物の使用。
- 請求項1から6までのいずれか1項記載の組成物を消耗品に導入することを含む、消耗品の甘味を伝達、増強、または改善する方法。
