Description

サイトメガロウイルス(CMV)は、一般に生後間もなくに獲得され、永続的で生涯にわたる感染を確立するβヘルペスウイルスである。CMV血清陽性率は米国成人の約50%であり、健常人のCD8+T細胞およびNK細胞によってよく制御されるため、感染症は通常無症候性である。CMV血清陽性は、ヘテロ二量体活性化受容体CD94-NKG2Cを発現するNK細胞の割合の増加、および健常成人におけるNKG2ChighCD57+NK細胞の増加と関連する(Lopez-Verges et al., Proc Natl Acad Sci U S A. 2011;108(36):14725-14732)。CMVに曝露されていない個体はCMV「ナイーブ」である。
本明細書で使用される場合、「適応型NK細胞」は、単一の適応型NK細胞、複数の適応型NK細胞、および/または適応型NK細胞を含む単離細胞集団を含む。
いくつかの実施形態において、適応型NK細胞の産生および/または製造をインビトロで行う。インビトロでの製造には、細胞分化、増殖、濃縮、および/または単離が含まれ得る。
いくつかの実施形態において、適応型NK細胞の製造をインビボで行う。インビボで製造した適応型NK細胞を、それを製造した同じ対象において、または異なる対象において、例えば適応型NK細胞製剤の同種異系適用で使用し得る。いくつかの実施形態において、インビボで製造した適応型NK細胞を、対象から取り出し、その後対象に再投与し得る。

ナチュラルキラー(NK)細胞は、造血細胞移植(HCT)後に早期に再構成し、HCT後の転帰に影響を及ぼす可能性がある主要リンパ球集団である。しかし、NK細胞の移植片対白血病(GvL)活性は、HCT後の最初の1年を通してNK細胞の機能的成熟が遅延されることによって制限される。再構成ドナーNK細胞の未成熟表現型は、生体外での腫瘍細胞株および原発性急性骨髄性白血病(AML)芽球に応答したNK細胞媒介性細胞障害性およびインターフェロン(IFN)-γ産生における有意な障害に関連する。全体的に見て、HCT後早期に再構成するドナーNK細胞の表現型および機能の未熟性は、それらの臨床的有用性を制限する。
移植手順
骨髄機能廃絶(MA)前治療は、悪性血液疾患の患者366人に使用され、シクロホスファミド(60mg/kg×2)および全身照射(13.2Gy、165cGy、1日2回×4日)からなった。一部の患者では、このレジメンは、フルダラビン(-8日目~-6日目に25mg/m2/日)、およびミコフェノール酸モフェチル(-3日目~+30日目に12時間ごとに1g)も含んだ。全ての患者はまた、-3日目に開始し、HCT後180日まで継続してシクロスポリンAも受けた。強度軽減前治療(RIC)は、308人の患者に使用され、シクロホスファミド(50mg/kg)およびフルダラビン(200mg/m2)および全身照射(2Gy)からなった。前治療後、骨髄、末梢血、または臍帯血由来の幹細胞(単一または二重)を注入した。表Iは、レシピエントCMV状態(血清陰性、再活性化を伴わない血清陽性、および再活性化を伴う血清陽性)によって層別化されたHCT患者の人口統計を記載する。
前治療の前に、全てのレシピエントを、酵素結合免疫吸着法を用いる血清学的検査によってCMV曝露について評価した。CMV IgG抗体レベルが10.0 ELISA単位毎ミリリットル(EU/mL)より大きい(>)ことは血清陽性とみなした。移植後、全てのレシピエントは、移植後+100日まで、pp65抗原血症(2006年以前)または定量的リアルタイムポリメラーゼ連鎖反応(PCR)(2006年以降)のいずれかにより、毎週CMV再活性化についてのスクリーニングを受けた。CMV予防は、100日までの高用量のアシクロビル(静脈内へ8時間ごとに500mg/m2[10~12mg/kg]、または経口で1日5回の800mg[18mg/kg小児])を含んだ。CMV再活性化は、CMV抗原血症(≧2 pp65陽性細胞/50,000)、DNAemia(定量的リアルタイムPCRによる≧500コピー)、または血液、体液、または組織からのCMVの培養として定義され、ガンシクロビルまたはフォスカーネットで治療された。
ミネソタ大学の血液および骨髄移植プログラムでは、患者の特性および転帰に関する全てのデータが前向きに収集された。ミネソタ大学の施設審査委員会は全てのプロトコルを承認し、全ての患者(および/またはその法定後見人)はヘルシンキ宣言に従ってインフォームドコンセントを提供した。
HCTレシピエント由来の末梢血単核細胞(PBMC)を末梢血サンプルから密度勾配遠心分離によって単離し、LSR II(カリフォルニア州サンノゼのBD Biosciences)を用いて蛍光標識細胞分取(FACS)により分析した。CMVを再活性化したレシピエント由来のPBMCを、ウイルス診断時、抗ウイルス療法の2週間後、4週間後、および8週間後、ならびに移植後6ヶ月および1年に収集した。CMV血清陰性であるか、またはウイルス再活性化を伴わないCMV血清陽性であったレシピエントでは、移植後100日目、6ヶ月目、および1年目にPBMCを収集した。表現型分析のために、以下の蛍光コンジュゲート抗体を使用した:エネルギー結合色素(Energy Coupled Dye)(ECD)-コンジュゲート抗CD3(カリフォルニア州ブレアのBeckman Coulter, Inc.;IM2705U)、PECy7結合コンジュゲート抗CD56(カリフォルニア州サンディエゴのBioLegend;318318)、パシフィックブルー(Pacific Blue)コンジュゲート抗CD57(カリフォルニア州サンディエゴのBioLegend;322316)、およびPEコンジュゲートNKG2C(ミネソタ州ミネアポリスのR&D Systems;FAB138P-025)。RICレシピエントとMAレシピエントの間の適応型NK細胞のパーセンテージおよび絶対計数の統計的比較のために、GraphPadを用いて計算した対応のない両側t検定を用いた。エラーバーはSEMを表す。GraphPadを使用して、28人のCMV血清陽性レシピエントにおける絶対単球数およびリンパ球数と適応型NK細胞増殖との間の相関に関するR2値および関連するp値を算出した。
カプラン-マイヤー曲線を用いて、HCT後1年間の無病生存(DFS)の確率を推定し(Kaplan et al.J. Am. Stat.Assoc.1958; 53:457-481.)、比較のためにログランク検定を用いた。調整した生存曲線を、層別コックスモデルに基づいて計算した(Chang et al.J. Chronic Dis.1982; 35:669-674)。コックス回帰を用いて因子のDFSに対する独立した効果を調べ、比例ハザードをマルチンゲール残差を用いて調べた。再発の累積発生率を、非再発死亡率(NRM)を競合リスクとして扱って評価した。ファインアンドグレイ比例ハザードモデル(The Fine and Gray proportional hazards model)(Fine et al.J. Am. Stat.Assoc.1999; 94:496-509)を使用して、再発に対するCMV再活性化の独立した効果を求め、調整再発曲線を算出した。対象の主要な共変量は、時間依存性共変量として扱ったHCT後のCMV再活性化、および前治療レジメンの強度であった。潜在的交絡因子には、ドナータイプ、診断、移植年(<2008年対≧2008年)、移植片対宿主病(GvHD)予防、性別、疾患リスク、および以前の自家移植が含まれた。HCT時の疾患リスクを、ASBMT RFI 2006リスクスコアリングスキーマ(ASBMT RFI 2006 risk scoring schema)(asbmt.orgでワールドワイドウェブで入手可能)に基づいて標準リスクまたは高リスクに分類した。分散は、比較しているグループ間で類似していた。再帰的分割を用いて、再発と関連する適応型CD56dimCD57+NKG2C+NK細胞のパーセンテージおよび絶対数の最適カットポイントを決定した。全ての臨床分析は、SASバージョン9.3(ノースカロライナ州ケーリーのSAS Institute)を使用して実施した。
5人の健常なCMV血清陽性ドナーから収集したバフィーコートをMemorial Blood Bank(ミネソタ州ミネアポリス)から得た。末梢血単核細胞(PBMC)をFicoll-Paque(イギリスのバッキンガムシャーのリトル・チャルフォントのGE Healthcare)を用いて密度勾配遠心分離によって単離し、10%ウシ胎児血清(マサチューセッツ州ウォルサムのGibco, LifeTechnologies, Thermo Fisher Scientific Inc.)を補充したRPMI培地で5時間2:1(エフェクター:標的)比でK562細胞と共に培養した。GolgiStopおよびGolgiPlugタンパク質輸送阻害剤(カリフォルニア州サンノゼのBD Biosciences)をアッセイに1時間加えた。NK細胞サブセットの機能解析には以下の抗体を使用した:BV785コンジュゲート抗CD3(カリフォルニア州サンディエゴのBioLegend;318318)、PECy7コンジュゲート抗CD56(カリフォルニア州サンディエゴのBioLegend;359620)、PE-CF594コンジュゲート抗CD57(カリフォルニア州サンディエゴのBioLegend;359620)、PEコンジュゲートNKG2C(ミネソタ州ミネアポリスのR&D Systems;FAB138P-025)、PerCP-Cy5.5コンジュゲート抗CD107a(カリフォルニア州サンディエゴのBioLegend;328616)、およびBV605コンジュゲートIFN-γ(カリフォルニア州サンディエゴのBioLegend;502536)。K562細胞株は、ATCC(バージニア州マナサス)から購入し、マイコプラズマ汚染について毎月スクリーニングした。実験は2回独立して行った。有意性を判定するために、GraphPadで両側の対応のあるt検定を使用した。エラーバーはSEMを表す。
CMVを再活性化するRICレシピエントでHCT後の再発リスクが低い。
2001年から2013年の間にミネソタ大学で治療された急性骨髄性白血病(AML)(n=313)、急性リンパ芽球性白血病(ALL)(n=187)、骨髄異形成症候群(MDS)(n=85)、非ホジキンリンパ腫(NHL)(n=42)、慢性骨髄性白血病(CML)(n=28)、ホジキン病(n=14)、および多発性骨髄腫(n=5)を有する674人の同種異系HCTレシピエントで1年の再発リスクおよびDFSを分析した。516人の患者が標準リスクとして分類され、148人の患者が高リスクとして分類され、36人の患者が以前の自家移植を受けていた。37人の患者が骨髄を受け、166人の患者が末梢血幹細胞を受け、471人の患者が臍帯血移植を受けた。コホート全体を、レシピエントCMV血清状態(CMV血清陰性[n=270]対再活性化を伴わないCMV血清陽性[n=214]対再活性化を伴うCMV血清陽性[n=190])によって、および前治療レジメン(強度軽減[n=308]対骨髄機能廃絶[n=366])によって層別化した。疾患タイプおよび治療関連変数は、CMV血清状態によって層別化された群間でバランスがとれていた(表I)。
RIC HCT後のCMV再活性化、再発防止、およびDFS改善との間の関連性をさらに評価するために、HCT後のドナー由来末梢血NK細胞の表現型を分析した。CMV血清陰性レシピエントではなくCMV血清陽性レシピエントにおけるCD56dimCD57+NKG2C+NK細胞の頻度の増加は、6ヶ月および1年で観察された(図2A、左パネルおよび中央パネル)。CMVを再活性化したレシピエントは、最も高い割合のCD56dimCD57+NKG2C+NK細胞を示した。さらなる分析により、CD56dimCD57+NKG2C+NK細胞の頻度が、再活性化後4週間(8.58%対4.64%、p=0.02)、8週間(7.77%対3.84%、p=0.02)、および6ヶ月(14.80%対6.29%、p=0.01)でMAレシピエントに対してRICレシピエントで有意に高かったことが明らかになった(図2A、右パネル)。同様に、CMV血清陰性レシピエントと比較してCMV血清陽性レシピエントにおけるCD56dimCD57+NKG2C+NK細胞の絶対数がより大きいことに対する関連性が、6ヶ月および1年で観察された(図2B、左パネルおよび中央パネル)。CD56dimCD57+NKG2C+NK細胞の絶対数は、CMVを再活性化したCMV血清陽性レシピエントにおいて最高であり、また、移植後6ヶ月ではMAレシピエントに対してRICレシピエントで有意に高かった(22.2対10.44細胞/μl、p=0.04)。絶対数において同様の傾向が1年目に認められた。RICレシピエントにおいてCD56dimCD57+NKG2C+NK細胞増殖率がより高かったことは、100日目の急性GvHD率の差によって説明されなかった。なぜなら、これらの比率は、RIC前処置レジメン(43%[38~43%])とMA前処置レジメン(42%[36~48%])との間で有意差がなかったからである(p=0.46)。生存者の91%が100日目に≧90%のドナーキメラ現象を有し、6カ月目の生存者の93%が≧90%のドナーキメラ現象を有したため、100日目以降に増殖したCD56dimCD57+NKG2C+NK細胞は、ドナー起源の細胞から分化した可能性が高い。したがって、CMV再活性化に応答するドナー由来CD56dimCD57+NKG2C+NK細胞の増殖は、RICレシピエントの間でのみ、再発リスクの低下およびDFSの改善と相関していた。
HCT後6ヶ月(n=68)に、レシピエントにおける絶対CD56dimCD57+NKG2C+NK細胞数をCMV血清状態または再活性化とは独立して分析した。この時点で、ほぼ全ての患者が、移植された90%のドナーより大きい(>)。CD56dimCD57+NKG2C+NK細胞の絶対数に対する最適カットポイントを決定するために、再帰的分割を用いた。CD56dimCD57+NKG2C+NK細胞数(>2.5細胞/μl、n=54)に基づく増殖群のレシピエントは、2年再発率がより高い(46%[10~82%])非増殖群(0.1~2.5細胞/μl、n=14)と比較して2年再発率が低い(16%[6~26%]、p=0.06)傾向にあった。(表III)。したがって、これらのデータは、移植後早期にCMV再活性化に応答して増殖するCD56dimCD57+NKG2C+NK細胞が再発を防ぐことを示唆している。
CMV感染を模倣するインビトロ研究は、単球がNKG2Cを発現するNK細胞の増殖を促進する役割を果たすことを示している(Rolle et al.J Clin Invest.2014; 124:5305-5316)。したがって、RICレシピエントは、MAレシピエントと比較して、CD56dimCD57+NKG2C+NK細胞の優先的増殖を説明する、ウイルス診断時におけるより高い絶対単球数(AMC)を有し得る(図2)。HCTコホート内には、CMVの再活性化を経験した28人のレシピエント(16人のRICおよび12人のMA)が存在し、これらのレシピエントについて、ウイルス診断時のAMCおよび絶対リンパ球数(ALC)ならびに6ヶ月および/または1年でのNK細胞表現型データが利用可能であった。ウイルス診断時では、平均AMCは、MAレシピエントと比較してRICにおいてより高い傾向があった(0.71対0.54×109細胞/L、p=0.08)。対照的に、平均ALCは、RICレシピエントとMAレシピエントとの間で類似していた(0.65対0.61×109細胞/L、p=0.39)。CMV再活性化の時点でレシピエントの血液中に存在する単球の数とその後の適応型NK細胞の増殖との間に関連性が存在するかどうかを判断するために、各レシピエントのウイルス診断時におけるAMC値を、6ヶ月(1年での表現型が利用できなかった場合)または1年のいずれかでのCD56dimCD57+NKG2C+NK細胞の絶対数およびパーセンテージに対してプロットした。ウイルス診断時におけるAMCと、6ヶ月または1年でのCD56dimCD57+NKG2C+NK細胞の絶対数(図3A、p=0.02)および相対パーセンテージ(図3B、p=0.01)の両方との間の有意な正の相関が観察された。このような相関は、ウイルス診断時のALCとCD56dimCD57+NKG2C+NK細胞の増殖との間には観察されなかった(図3C、図D)。したがって、CMV再活性化の時点で単球数がより大きいことは、RICレシピエントにおけるCD56dimCD57+NKG2C+NK細胞の優先的増殖を説明し得る。
CD56dimCD57+NKG2C+NK細胞が、他のNK細胞サブセットと比較して白血病標的に対する高められたエフェクター機能を媒介するかどうかを決定するために、健常なCMV血清陽性血液ドナーから末梢血単核細胞(PBMC)を単離し、K562骨髄性白血病細胞と共に2:1の比で培養した。腫瘍壊死因子(TNF)およびIFN-γ産生を、未成熟CD56brightNK細胞、初期成熟CD56dimCD57-NKG2C-NK細胞、後期成熟CD56dimCD57+NKG2C-NK細胞、および適応型CD56dimCD57+NKG2C+NK細胞において分析した。分析した全ての他のサブセットと比較して、CD56dimCD57+NKG2C+NK細胞は、K562細胞に応答してTNFおよびIFN-γ産生の両方の頻度が高かった(図4A、図4B)。CD107a表面発現によって測定されるNK細胞脱顆粒については、有意な差異は観察されなかった(図示せず)。したがって、CD56dimCD57+NKG2C+NK細胞は、腫瘍標的の直接的な認識の際に炎症性サイトカイン産生の増強を介して直接HCT後の再発防止に寄与する可能性が高い。
方法
OP9ネイティブおよびOP9-DL1細胞
空のMigR1レトロウイルスベクター(OP9ネイティブ)またはデルタ様1遺伝子を含むMigR1レトロウイルスベクター(OP9-DL1)のいずれかを形質導入したOP9間質細胞(Schmitt et al.Immunity.2002;17(6):749-756)を、MEMα培地+20%ウシ胎児血清(FBS)中で、24ウェルプレート中4×105細胞/ウェルの濃度で2日間培養して細胞接着させた。次いでプレートを2,000cGyで照射して細胞増殖を停止させた。
単核細胞を密度勾配遠心分離によって全臍帯血から単離した。T細胞およびB細胞は、それぞれ抗CD3および抗CD19マイクロビーズを用いた正の磁気選別によって除去した。次いで、CD3/CD19欠失細胞を、照射したOP9細胞に5×105細胞/ウェルの濃度で加え、RPMI培地+10%FBS中で、1ng/mL IL-15または10ng/mL IL-15のいずれかを補充し、50ng/mL IL-21有りまたは無しで、14日間培養した。
臍帯血由来NK細胞を、培養前、ならびに培養の7日後および14日後に、以下の細胞外抗体および細胞内抗体を用いる蛍光標識細胞分取(FACS)によって分析した:BV785コンジュゲート抗CD3(カリフォルニア州サンディエゴのBioLegend)、PE-Cy7コンジュゲート抗CD56(カリフォルニア州サンディエゴのBioLegend)、BV605コンジュゲート抗CD57(カリフォルニア州サンディエゴのBioLegend)、APCコンジュゲートSYK(カリフォルニア州サンディエゴのeBioscience)、およびPEコンジュゲート抗PLZF(カリフォルニア州サンノゼのBD Biosciences)。
最終分化標準CD56dimCD57+NK細胞および適応型CD56dimSYK-NK細胞の増殖を支持するインビトロ培養系の確立
Notchシグナル伝達経路は、エフェクターCD8+T細胞の分化に必須の役割を果たし、エフェクター機能の獲得を制御する(Backer et al.Nat Immunol.2014;15(12):1143-1151)。Notchシグナル伝達が、最終分化および適応型NK細胞サブセットの分化および増殖を促進する役割を果たし得るかどうかを決定するために、CD3/CD19欠失単核細胞を、低用量(1ng/mL)のIL-15または高用量(10ng/mL)のIL-15の存在下で、IL-21(50ng/mL)有りまたは無しで、ネイティブOP9間質細胞またはNotchリガンドDL1を安定に形質導入されたOP9間質細胞上で臍帯血から培養した。細胞を7日目および14日目に採取し、CD56+NK細胞を、CD57およびSYKの発現について蛍光標識細胞分取(FACS)によって分析した。培養前に、平均2.13%±0.44のCD56+NK細胞がCD57を発現した。低用量IL-15(0.59%±0.19)または高用量IL-15(0.70%±0.23)のいずれかでの7日目のOP9ネイティブ培養物でCD57+NK細胞の増殖は観察されなかった。同様に、低用量IL-15(0.24%±0.11)または高用量IL-15(0.22%±0.10)のいずれかでの14日目のOP9ネイティブ培養物でCD57+NK細胞の増殖は観察されなかった。低用量IL-15(0.91%±0.34)または高用量IL-15(1.30%±0.31)のいずれかでの7日目のOP9-DL1培養物でCD57+NK細胞の増殖は観察されなかった。同様に、低用量IL-15(0.93%±0.40)または高用量IL-15(0.86%±0.44)のいずれかでの14日目のOP9-DL1培養物でCD57+NK細胞の増殖は観察されなかった。低用量IL-15プラスIL-21(0.71%±0.24)または高用量IL-15プラスIL-21(1.05%±0.29)のいずれかでの7日目のOP9ネイティブ培養物でCD57+NK細胞の増殖は観察されなかった。同様に、低用量IL-15プラスIL-21(0.18%±0.06)または高用量IL-15プラスIL-21(2.74%±1.90)のいずれかでの14日目のOP9ネイティブ培養物でCD57+NK細胞の増殖は観察されなかった。低用量IL-15プラスIL-21(2.06%±0.66)での7日目のOP9-DL1培養物でCD57+NK細胞の増殖は観察されなかった。高用量IL-15プラスIL-21(5.31%±2.10)での7日目のOP9-DL1培養物でCD57+NK細胞の中程度の増殖が観察された。低用量IL-15プラスIL-21(1.38%±0.56)での14日目のOP9-DL1培養物でCD57+NK細胞の増殖は観察されなかった。高用量IL-15プラスIL-21(12.67%±5.11)での14日目のOP9-DL1培養物でCD57+NK細胞のロバストな増殖が観察された。
転写因子前骨髄球性白血病ジンクフィンガー(PLZF)の転写サイレンシングは、CMV感染に応答して増殖する適応型NK細胞の特徴である(Schlums et al.Immunity.2015;42(3):443-456)。単離直後の単核細胞由来およびインビトロで14日間培養された細胞由来のNK細胞において、PLZFの細胞内染色を行った。臍帯血からの単離直後のCD56dimSYK-NK細胞は、主にPLZF陽性であった(99.3%±0.10)。高用量IL-15を用いた培養は、CD56dimCD57-SYK+NK細胞(9.93%±0.75)、CD56dimCD57+SYK+NK細胞(11.47%±2.18)、CD56dimCD57-SYK-NK細胞(7.05%±1.31)、およびCD56dimCD57+SYK+NK細胞(6.67±1.05)におけるPLZF発現の中程度の低下をもたらした。しかし、IL-21の添加は、CD56dimCD57-SYK+NK細胞(14.92%±1.80)およびCD56dimCD57+SYK+NK細胞(12.13%±2.43)と比較して、CD56dimCD57-SYK-NK細胞(35.82%±7.02)およびCD56dimCD57+SYK-NK細胞(66.0%±8.86)におけるPLZF発現の顕著な低下をもたらした。したがって、IL-21は、CD56dimSYK-NK細胞におけるPLZF発現を抑制し、それにより、これらの細胞は適応特性を獲得するようになる(図6)。
実施例2に記載のように培養した臍帯血由来NK細胞は、細胞傷害性受容体2B4、低親和性Fc受容体CD16、およびキラー免疫グロブリン様受容体(KIR)の発現によって示されるように、細胞傷害性のコンピテントである。追加のデータ(図示せず)は、これらの細胞が、ウイルス感染細胞、新生細胞、および/または自家活性化免疫細胞に応答して脱顆粒する能力を示す、高レベルのグランザイムおよびパーフォリンを発現することを示す。
インビトロで培養したCMV血清陽性ドナー由来NK細胞へのラパマイシンの添加は、適応型NK細胞の分化プログラムを促進した(図7A、図7B)。機能アッセイにおいて、ラパマイシンで処理したNK細胞は、脱顆粒および腫瘍壊死因子(TNF)産生の増強を示した(図7C、図7D)。したがって、エクスビボNK細胞増殖方法へのラパマイシンの添加は、養子免疫療法の前にNK細胞分化を増強し、固有の代謝特性を有する成熟NK細胞サブセットについて富化し、NK細胞の細胞障害性およびサイトカイン産生を増強する新規な戦略を示唆する。
成人CMV血清陽性ドナーの末梢血由来の適応型NK細胞は、高用量のIL-15、IL-21、およびCD16刺激によりインビトロで増殖され得る。健常なCMV血清陽性ドナーからのCD3/CD19欠失PBMCをCellTrace色素で標識し、高用量IL-15(10ng/mL)のみ、IL-15プラスIL-21(50ng/mL)、またはIL-15プラスIL-21プラス抗CD16抗体(1:1000)と共に10日間培養した。図8Aは、代表的なCMV血清陽性ドナーからのCD57、NKG2C、SYK、およびCellTraceの発現を示す。図8Bは、NKG2Cを発現するNK細胞のパーセンテージの累積データを示す。図8Cは、4人のCMV血清陽性ドナーからのSYK欠失NK細胞のパーセンテージを示す。
NKG2C発現を誘導するために、NK細胞を、実施例2に記載のように、可溶性および/または膜結合性炎症性サイトカイン、例えば、IL-1、TNF-α、IL-6、IL-8、およびIFN-γ;膜結合性HLA-E;および膜結合性HLA-E提示CMVペプチドのうちの1つ以上と共に、7日間および14日間培養する。
TIGIT発現が低い適応型NK細胞は、骨髄由来サプレッサー細胞に抵抗性である。

ナチュラルキラー細胞は自然免疫系のリンパ球である。T細胞およびB細胞とは異なり、これらはクローン様式で生殖系列再構成抗原特異的受容体を発現しない。これらは細胞傷害性T細胞と同様の殺滅メカニズムを共有しているが、NK細胞は活性化受容体および抑制性受容体のファミリーを介して標的を認識する。これらの受容体の間のバランスは、NK細胞の最終的な機能を決定する。NK細胞が健康な細胞と形質転換された細胞または感染した細胞を区別する主なパラダイムは、「自己喪失」仮説によって説明される。損傷した細胞に対するMHCクラスIのダウンレギュレーション、またはキラー免疫グロブリン様受容体(KIR)の抑制性サブグループと自己細胞上のそれぞれのヒト白血球抗原(HLA)リガンドとの間のミスマッチは、標的細胞をNK細胞殺滅に感受性にする。NK細胞はまた、以前の抗原曝露を必要とすることなく、腫瘍細胞を認識し殺す能力を有し、これは、癌治療のためのエフェクターとしてのNK細胞の開発を容易にする性質である。しかし、T細胞と同様に、NK細胞の抗腫瘍活性は、腫瘍内微小環境に存在する抑制性因子によって制限され、これにより、免疫機能が弱められ、予後不良となる。新興の研究は、T細胞およびNK細胞上の抑制性受容体、例えば細胞傷害性Tリンパ球関連4(CTLA-4)、プログラム細胞死1(PD-1)、およびT細胞IgおよびITIMドメイン(TIGIT)が抗腫瘍反応を抑制できることを示す。
MDSCは、T細胞およびNK細胞の増殖およびNK細胞の機能を抑制する。
MDSCとNK細胞サブセットとの間の相互作用を調べるために、健常ドナーからの末梢血単核細胞をIL-6およびGM-CSFと共に1週間培養し、続いてCD33+細胞を富化させることによってMDSCを作製した(図16A)。新鮮な単球をこれらの実験の骨髄細胞対照として用いた。精製したT細胞およびNK細胞を、単球またはMDSCと様々な比率で共培養し、3~4日間の培養後に増殖について評価した。単球は増殖にほとんど影響しなかったが、MDSCは、2.7±1.6倍のT細胞の増殖の抑制(p=0.005)を誘導し、NK細胞に対して同様の効果(1.5±0.27倍抑制、p=0.006)を有した(図9A)。同様に、CD16の関与は、新鮮な単球で培養したNK細胞と比較してMDSCによって有意に抑制されたNK細胞脱顆粒およびIFN-γ産生を刺激した(それぞれ、2.4±1.4倍の抑制、p=0.0001、および2.6±1.5倍の抑制、p=0.003)(図9B)。
適応型NK細胞が普通型NK細胞と比較してMDSC抑制に抵抗し得るかどうかを調べるために、精製ポリクローナルNK細胞を単球またはMDSCと2:1の比で5日間共培養し、CD16刺激後の脱顆粒、増殖、およびサイトカイン産生について調べた。普通型および適応型NK細胞は、同様の量のCD16を発現し(図16B)、適応型NK細胞を、CD57+NKG2C+FcεRγ-であるCMV血清陽性ドナー由来NK細胞として定義した(Schlums et al.Immunity.2015;42(3):443-56)。IL-15の存在下で単独で、または単球との共培養で培養した場合、同様のNK細胞活性が観察された(脱顆粒:44±14%対49±12,IFNγ:28±14.0%対27±9.0%)(図10A)。単球対照と比較して、MDSCは、普通型NK細胞の集団内で、CD107a(52.4±2.4%対33.5±3.1%、p=0.0007)、IFN-γ(31.3±3.8%対13.8±3.0%、p=0.009)、TNF(29.1±2.1%対13.6±3.3%、p=0.007)、および増殖(49.0±2.9%対23.0±3.5%、p=0.001)(Ki67によって測定)の有意な抑制を媒介した。しかし、適応型NK細胞は、同じMDSC集団に対して抵抗性であった(図10A)。さらに、MDSCの存在下での普通型NK細胞の脱顆粒およびIFNγは、トランスウェルによって分離されたときに完全に回復した(図10B)。したがって、CMV感染は、MDSC抑制に抵抗性である適応型NK細胞の集団を生じる。
健常な供血者由来の精製NK細胞を、IL-15(10ng/mL)単独の存在下もしくは非存在下で、またはIL-12(10ng/mL)およびIL-18(100ng/mL)または抗CD16(1μg/mL)の追加刺激とともに、染色前に一晩培養した。TIGIT発現は刺激なしでは低く、IL-15単独ではわずかにアップレギュレートされた。抗CD16による追加刺激は、TIGIT発現をさらに増加させた。しかし、DNAM-1は、ベースラインの発現レベルが高いために、さらに増加させることができなかった(図11A)。これらのポリクローナル活性化NK細胞上のTIGITに対する染色パターンは、二峰性の発現を示した(図11A)。この現象をさらに調べるために、TIGITおよび他の抑制性受容体の適応型および普通型NK細胞での発現を調べた。DNAM-1、CD96、NKp44、NKp46、PD-1、Tim3、またはNKG2Aでは、適応型NK細胞と普通型NK細胞との間に発現の差はなかった(図11B、図16C)。対照的に、TIGIT発現は、単球(TIGIT MFI:1595±407対2196±461、p=0.008)またはMDSC(TIGIT MFI:1680±336対2556±403、p=0.008)と共培養したかどうかにかかわらず、普通型NK細胞に対して適応型NK細胞で有意に少なかった。(図11C)。普通型および適応型NK細胞は、単球またはMDSCとの共培養の前後で同様のレベルでTIGITおよびDNAM-1を共発現したが(NK単独:18%±10%対14%±11.5%、NK+単球:86%±8%対83%±9%、NK+MDSC:84%±9%対82%±6%、図11D)、適応型NK細胞のTIGITの発現は依然として低かった。
単球、MDSC、およびNK細胞をCellTracker Blueで標識し、一晩チャンバースライドで共培養した。細胞を抗CD155(緑色)および抗TIGIT(赤色)で染色し、サイズによって区別した。予想通り、NK細胞上のTIGITは、MDSC上のCD155と共存した(図12A)。TIGITがNK細胞のMDSC依存性調節に関与するかどうかを評価するために、健常な給血者由来のポリクローナルNK細胞を、5日間、IL-15(10ng/mL)の存在下で、2:1の比率で、自家MDSCまたは新鮮な単離単球と共培養した。IFN-γ産生を、CD16刺激の6時間後に、低いTIGIT発現に対する高いTIGIT発現の差異に基づいて、MDSCと共培養した普通型NK細胞において評価した。これらのデータは、低いTIGIT発現を有するNK細胞が、高いTIGIT発現を有するNK細胞と比較して有意により多くのIFN-γを産生することを示す(36.2%対19.9%、p=0.0005、図16D)。次に、TIGITの関与がNK細胞のMDSC抑制を引き起こす原因であるかどうかを調べた。抗TIGIT抗体の機能を、これまでに記載されているように(Warren et al.Int Immunol.2001;13(8):1043-52)正常なNK細胞を用いるP815アッセイで試験した。抗CD158b対照の存在がNK細胞の細胞障害性を阻害したが、NK細胞機能は抗TIGITの存在下で影響を受けず(図18A)、アゴニスト機能の欠如を示した。
単球およびMDSC単独におけるTIGITリガンドCD155およびCD112の発現をさらに調べた。MDSCは、単球でほとんど発現しないのと比較して高レベルのCD155を発現した(MFI:675±124対107±23、p=0.015)。さらに、CD112発現は、単球と比較してMDSCにおいて有意に高かった(MFI:1714±331対865±196、p=0.015)(図13A)。MDSC誘導性の普通型NK細胞の抑制のメカニズムをさらに調べるために、スーパーオキシド、アルギナーゼ、ROS、TGF-β、およびiNOSを含むMDSCによって利用される経路を、MDSC生成の終わりに一晩遮断した。スーパーオキシド、アルギナーゼ、TGF-β、またはiNOSを遮断したとき、CD155の発現に実質的な差は見られなかったが、カタラーゼによるROS産生の阻害は、MDSC上のCD155の発現の有意な減少をもたらした(55%±23減少、p=0.03)(図13B)。
普通型NK細胞の機能および増殖に対するTIGITの関与の強力な抑制効果を考慮して、MDSCと共培養したNK細胞におけるTIGITとのCD16誘導性シグナル伝達相互作用を分析した。対照単球と共培養した場合と比較して、MDSCと共培養したNK細胞は、ERK1/2(MFI:1356±143対696±202、p=0.03)およびZAP70/Syk(MFI:159±14対109±14、p=0.03)のリン酸化の低下を示した。さらに、TIGITの遮断またはROSの阻害は、ZAP70/SykおよびERK1/2のリン酸化を増加させた(p=0.03、図14A、図14B)。TIGITまたはROSを遮断することがMDSCと培養されたNK細胞の機能を回復できるかどうかを細胞障害性アッセイにおいて調べるために、単球またはMDSCのいずれかと共に培養したNK細胞を、TIGIT遮断またはカタラーゼで前処理し、洗浄した後、51Cr標識K562細胞とインキュベートした。NK細胞の細胞傷害性は、単球との共培養と比較して、MDSCとの共培養の後に有意に減少した(図14C)。抗TIGITもカタラーゼも単独で培養したNK細胞には何ら影響を及ぼさなかったが、TIGIT遮断およびROS阻害の両方は、MDSCによって媒介される抑制効果を完全に逆転させた(図14C)。さらに、NK細胞およびMDSC共培養物におけるカタラーゼ処理と組み合わされたTIGITの遮断は、pZAP70/SykおよびpERK1/2またはNK細胞の細胞傷害性のいずれに対しても相加効果を有しなかった。
サイトカインによって生じたMDSCの接触を介した抑制メカニズムを同定し、このメカニズムがインビボでの生理的セッティングにおいてオペラントであったかどうかを調べた。CMV血清陽性MDS患者および健常ドナー(HD)からのPBMCを、適応型NK細胞およびMDSCの頻度について分析した。CMV+MDS患者の血液中で適応型NK細胞の頻度が高かったが(n=10,17%±15%対7%±5%)、総NK細胞頻度はHDと比較して有意に低かった(n=8、1.3%±1.2対8%±7%)。単球型MDSC(mMDSC)をCD45+Lin-CD11b+CD33+HLA-DR-/lowCD14+として、顆粒球型MDSC(gMDSC)をCD45+Lin-CD11b+CD33+CD15+として定義した。健常な給血者と比較して、MDS患者の血液中で、mMDSC(1.6±0.2対11.4±9.2、p=0.02)およびgMDSC(0.02±0.02対1.84±1.6、p=0.01)の両方の頻度の有意な増加が観察された(図15A)。さらに、MDS-MDSCは、MDS-単球と比較してCD155の発現を増加させた(図15B)。MDS患者由来のPBMCを、普通型および適応型NK細胞上のTIGITの発現について評価した。MDS患者において、TIGITの発現は、普通型NK細胞と比較して適応型NK細胞で有意に低かった(MFI:347±189対660±311、p=0.002;図15C)。MDS患者のCMV誘導性適応型NK細胞は、MDS-普通型NK細胞と比較して、IL-15およびCD16刺激による活性化後に有意により優れた機能を示した。さらに、適応型NK細胞は、健常ドナーのNK細胞と同様の脱顆粒およびIFNγ産生を示した(図15D)。普通型NK細胞におけるTIGITシグナル伝達の遮断は、普通型NK細胞の機能的低反応性を救済したが、低レベルのTIGIT発現を有する適応型NK細胞に対しては相加的効果はほとんどなかった(図15D)。
進行中の癌を有する患者におけるNK細胞およびT細胞免疫療法の潜在的な治療効果を記載する免疫療法の文献が急増している。この急増は、T細胞PD-1/PD-L1および/またはCTLA-4経路に対するチェックポイント遮断による予期しないほど強力な臨床結果によるものである。まとめると、癌治療に対する新規アプローチは、抗原特異的応答およびチェックポイント遮断に重点を置いて、戦略的変化を経ている。標的に対するNK細胞応答は、活性化受容体および抑制性受容体を介したシグナルの釣り合いによって決定されるが、同様のチェックポイント遮断メカニズムはあまり理解されていない。本研究において、TIGIT/CD155は、普通型NK細胞のMDSC誘導性抑制の根底にある重要な軸であると同定された。CMV誘導性適応型NK細胞は、低レベルのTIGITを発現し、MDSC抑制に感受性ではなかった。サイトカイン曝露による正常血液から生じたMDSCの機能は、癌(MDS)患者で誘導されたMDSCの機能と同等であり、これらの知見の生理学的関連性および潜在的なトランスレーショナルな役割を強調する。本開示によって与えられるように、そして理論に拘束されることを望むものではないが、NK細胞のTIGIT誘導性免疫抑制を克服するための少なくとも2つの方法がある。第1に、普通型NK細胞のCD16シグナル伝達を正常レベルに回復させるTIGIT遮断である。第2に、CMV曝露後のTIGIT抵抗性適応型NK細胞の増殖である。ここでは、NK細胞のサブセットを、単離して、または単離せず、増殖させ、必要に応じて調整して、TIGITの発現レベルを低く維持することにより腫瘍誘導性免疫抑制に対する抵抗性を発揮し得ることが示されている。
患者および健常ドナー
健常な対象から正常末梢血を得た。骨髄異形成症候群患者(MDS、n=15)からの凍結末梢血単核細胞(PBMC)を、International Blood and Marrow Transplant ResearchのNational Marrow Donor Program (NMDP)/Centerから得た。全ての健常およびMDS患者ドナーは、CMV血清陽性であった。全てのサンプルは識別を取り消され、使用はヘルシンキ宣言に従ってミネソタ大学とNMDP施設審査委員会によって承認された。
Ficoll勾配遠心分離(Ficoll-Paque Plus,イギリスのリトル・チャルフォントのGE Healthcare)後に、MDS患者および健常な給血者からのPBMCを採取した。その後、10%熱不活性化FBS、IL-6(10ng/mL、ミズーリ州セントルイスのSigma-Aldrich)、およびGM-CSF(10ng/mL、R&D Systems)を含有するRPMI培地中に2×106/mLの運命で1週間細胞を播種し、培養3日目に新しくして、MDSCを得た(Koehn et al.Blood.2015;126(13):1621-8)。次に、HLA-DR+細胞を抗ヒトHLA-DRマイクロビーズ(ドイツのベルギッシュ・グラートバッハのMiltenyi Biotech)を用いて単離し、その後、抗CD33マイクロビーズ(ドイツのベルギッシュ・グラートバッハのMiltenyi Biotech)を用いてHLA-DR-画分からMDSCを精製した。単球のMDSCを全ての示された実験で使用した(≧85%CD14+HLA-DR-)。NK細胞およびT細胞を、負の欠失(EasySep Human NK Cell Enrichment Kit,カナダのバンクーバーのStemcell Technologies)またはCD3マイクロビーズ(ドイツのベルギッシュ・グラートバッハのMiltenyi Biotech)によって一晩休止させたPBMCから単離した。対照単球を、抗CD33マイクロビーズを用いて一晩休止させたPBMCから単離した。
精製単球またはサイトカイン誘導性MDSCを、10%FBSを補充したRPMI培地(ミネソタ州ミネアポリスのGibco)(以下、培地と称する)でCellTraceバイオレット色素(5μM、カリフォルニア州カールスバッドのInvitrogen)で標識された自家T細胞またはNK細胞(1×105個)と1:1~1:16の比率で96穴U底プレートに2連で播種した。T細胞を抗CD3/CD28活性化ビーズ(40ビーズ/ウェル)およびIL-15(1ng/mL)で刺激し、またはNK細胞についてはIL-15(10ng/mL)単独で刺激し、3~5日間培養した。細胞をLSRIIフローサイトメーター(BD Biosciences)により得、データをFlowJo(オレゴン州アシュランドのTree Star)により分析した。
様々な時点について単球またはMDSCと培養され、様々な刺激を受けた精製NK細胞またはMDS PBMC(図17)を染色緩衝液(0.5%ヒトAB血清含有PBS)に再懸濁し、蛍光色素結合抗体で染色した(表IV)。培養NK細胞によるCD107a、Ki67、IFN-γ、およびTNFα産生の検出は、製造業者の指示書に従ってFoxp3/転写因子固定/浸透化キット(Foxp3/Transcription Factor Fixation/Permeabilization kit)(カリフォルニア州サンディエゴのeBioscience, Inc.)を用いて行った。固定および細胞外染色の前に、NK細胞をブレフェルジンAおよびモネンシン(GolgiPlugおよびGolgiStop;ニュージャージー州フランクリンレイクのBD Bioscience)で37℃で6時間処理した。以下の抗原に対する抗体でMDSCを染色して、NK細胞との共培養の前の純度および表現型を決定した:CD11b、HLA-DR、CD33、CD14、CD15、CD66b、CD56、CD3、CD19(lin-)、および固定可能死細胞マーカー(fixabale dead cell marker)(表IV)。染色前のいくつかの実験において、MDSCを、10μg/mLのTGFβ用中和抗体(R&D systems)、200IU/mLのROSスカベンジャーカタラーゼ(ミズーリ州セントルイスのSigma-Aldrich)またはスーパーオキシドジスムターゼ(Sigma-Aldrich)、500μmol/Lのアルギナーゼ阻害剤N(ω)-ヒドロキシ-ノル-l-アルギニン(nor-NOHA;Calbiochem)、またはiNOS阻害剤NG-モノメチル-l-アルギニン(L-NMMA;ミズーリ州セントルイスのSigma-Aldrich)を含有する、MDSCの様々な抑制メカニズムを標的とする試薬の存在下で一晩単独で培養し、CD155(PVR)について染色した。全ての細胞をLSRIIによって得、FlowJoによって分析した。適応型および普通型NK細胞をゲーティングし、図17のゲーティング戦略に従って同定した。
健常な給血者由来の精製NK細胞を、IL-15(10ng/mL)の存在下で2:1の比率で自家MDSCまたは新鮮な単離単球と5日間共培養し、24ウェルプレートに、接触させて、またはトランスウェルインサート(0.4μmの孔)(ニューヨーク州コーニングのCorning)で分離して可溶性因子交換のみを可能にして、播種した。細胞を抗CD16で6時間刺激した後、脱顆粒およびIFNγ産生について、上記のように染色した。
MDSCおよび単球を、20分間、CellTracker Blue(14μM、カリフォルニア州カールスバッドのInvitrogen)で予め標識した。その後、それらをIL-15(10ng/mL)で一晩予備活性化したNK細胞と共に40分間共培養した。混合した細胞をポリリシンで前処理したカバーガラス上に30分間載せ、3%BSAでブロックし、2%パラホルムアルデヒド中で37℃で30分間固定した。固定後、細胞を抗TIGITおよび抗PVR(CD155)で室温で2時間染色し、次いで共焦点顕微鏡法の前に蛍光標識した二次抗体で1時間染色した。
健常な供血者由来の精製NK細胞を、5日間、IL-15(10ng/mL)の存在下、TIGITに対する遮断抗体(10μg/mL)またはカタラーゼ(200IU/mL)の存在下または非存在下で、自家MDSCまたは新鮮な単離単球と2:1の比率で共培養した。次いで、細胞を洗浄し、4時間休止させ、抗CD16アゴニスト抗体で10分間および30分間刺激した。その後、それぞれZap-70およびERK1/2リン酸化の分析を行った。細胞をBD固定緩衝液および透過緩衝液IIIで固定して透過性にし、製造業者(ニュージャージー州フランクリンレイクのBD Bioscience)の指示書に従ってpZap-70(pY319)/Syk(pY352)およびpERK1/2(pT202/pY204)について染色した。
TIGITに対する遮断抗体(10μg/mL)またはROSスカベンジャーカタラーゼ(200IU/mL)の存在下または非存在下での単球またはMDSCとの5日間の共培養の後、NK細胞の細胞傷害性を、K562(バージニア州マナサスのATCC)細胞に対する5:1~2.5:1のエフェクター:標的比でのクロム(51Cr)放出アッセイ(4時間)により分析した。51Cr放出をγシンチレーションカウンター(マサチューセッツ州ウォルサムのPerkin Elmer)によって測定し、特異的標的溶解を測定した。
MDS患者および健常ドナーからのPBMCを一晩休止させ、MDSCを上記のように単離した。NK細胞および単球を、以前に記載されているように、一晩休止させた健常なPBMCから単離した。NK細胞を、5日間、IL-15(10ng/mL)の存在下、単球または同種異系MDS MDSCと2:1の比率で共培養した。抗CD16で6時間刺激した後、TIGITに対する遮断抗体の存在下または非存在下で脱顆粒、増殖、ならびにIFN-γおよびTNFα産生についてNK細胞を評価した。
全てのデータを、上記のソフトウェアで最初に分析し、Prism Version 6ソフトウェア(カリフォルニア州ラホヤのGraphPad Software)によって要約した。全てのデータを最初に正規分布について検定した。その後、(図の凡例に示されているように)スチューデントのt検定またはノンパラメトリックなマンホイットニーU検定によって群間の差異を分析した。インビトロアッセイについては、平均およびSEMとして、MDS患者由来の細胞を用いて行った実験については、平均およびSDとして、データを提示した。代表的なヒストグラムまたは画像を、平均値に基づいて選択した。
成熟樹状細胞のCMVペプチドのプールによるパルス化が、適応型NK細胞の増殖を誘導する。
100ng/mLのGM-CSFおよび20ng/mLのIL-4を補充したCellGro培地(2.5%ヒトAB血清)中で単球(2×106/mL)を培養して、未成熟樹状細胞への単球の成熟を誘導した。培養5日後、未成熟樹状細胞を回収し、スピンダウンし、GM-CSF(100ng/mL)およびIL-4(20ng/mL)(未成熟樹状細胞の場合)、またはGM-CSF(100ng/mL)、IL-4(20ng/mL)、ポリイノシン酸:ポリシチジル酸(Poly I:C)(20μg/mL)、リポ多糖(LPS)(10ng/mL)、およびIFN-γ(1000IU/mL)(成熟樹状細胞の場合)を補充した新鮮なCellGro培地(2.5%ヒトAB血清)に1×106細胞/mLで播種した。選択した成熟樹状細胞培養物に、CMV pp65ペプチドプール(10μg/mL)(カタログ番号11549、メリーランド州ジャーマンタウンのNational Institutes of Health AIDS Reagent Program;プール中のペプチドのリストは、aidsreagent.org/pdfs/11549_TAB_002.pdfでワールドワイドウェブで入手できる)またはHIV PTE Gagペプチドプール(10μg/mL)(カタログ番号12437、メリーランド州ジャーマンタウンのNational Institutes of Health AIDS Reagent Program;プール中のペプチドのリストは、aidsreagent.org/support_docs/11554_Lot21164_Solubility-MW-Purity.docxでワールドワイドウェブで入手できる)をさらに補充したか、または補充しなかった。自家PBMCを5日目に解凍し、37℃で10%FBSを補充したRPMI中で一晩休止させた。
自家単球およびIL-15の存在下で培養した場合、CMV血清陽性ドナーからのNK細胞は、CD45RA-CD45RO+表現型に偏っている。
類別された健常なCMV血清陰性および血清陽性ドナー由来のCD3/CD19欠失末梢血単核細胞を10ng/mLのIL-15と培養した。CMV血清陰性およびCMV血清陽性ドナー由来の培養前および培養後のCD3-CD56+CD45RA-CD45RO+NK細胞のパーセンテージを、FACSを使用して測定した。
適応型NK細胞はTreg媒介性抑制に抵抗性である。
調節性T細胞(Treg)を以下の方法を用いてインビトロで作製した:CMV血清陽性ドナーから密度勾配遠心分離によってPBMCを単離した。次に、CD4+T細胞を磁気ビーズ分離によって全PBMCから単離した。次いで、CD4+T細胞を選別して、CD4+CD25hiCD127lowTreg集団を単離した。次いで、選別したTregを、照射したK562-mbIL-21フィーダー株、抗CD3/CD28ビーズ、および300U/ml IL-2と共に21日間培養液中で増殖させた。次いで、50U/mlのIL-2を補充した培地中で6日間、自家または同種異系NK細胞およびHLA-DR+抗原提示細胞と1:1、1:2、1:4、または1:8の比率で増殖させたTregを培養した。機能的実験のために、NK細胞を、1μg/mlの抗CD16アゴニスト抗体、5ng/mlのIL-12、および50ng/mlのIL-18で5時間刺激した。
Claims (12)
- 培地中に存在する、適応型NK細胞の富化された集団または単離された集団であって、
(a)前記培地が、
(i-a)IL-15及びラパマイシン(さらに任意にIL-21及びCD16シグナル伝達の活性化因子の少なくとも1つを含んでいてもよい);
(ii-a)CMVペプチド補充成熟樹状細胞;又は

- TIGIT阻害剤が、TIGITに対する抗体を含む、請求項2記載の富化された集団または単離された集団。
- 前記適応型NK細胞が、CD57+、SYK-、FcεRγ-、EAT-2-、CD45RO+、およびCD45RA-のうちの少なくとも1つである、請求項1記載の富化された集団または単離された集団。
- (i)前記適応型NK細胞が、培養前の前記NK細胞の集団と比較してPLZFの発現の減少を示す、
(ii)前記適応型NK細胞が、培養前の前記NK細胞の集団と比較して抗腫瘍免疫活性の増強を示す、または
(iii)前記適応型NK細胞が、培養前の前記NK細胞の集団と比較して、細胞傷害性の増加、サイトカイン産生の増加、持続性の増加、およびT調節細胞に対する抵抗性の増加のうちの1つ以上を示す、請求項1記載の富化された集団または単離された集団。 - 前記適応型NK細胞が、
(i)CD56dimであり、またNKG2C+、CD57+、およびTIGITlowのうちの1つ以上である、
(ii)CD3-、CD56+であり、またCD57+、NKG2C+、SYK-、FcεRγ-、EAT-2-、CD56dim、TIGITlow、CD45RO+、およびCD45RA-のうちの少なくとも1つである、
(iii)CD56dimであり、またNKG2C+である、または
(iv)標準NK細胞と比較してPLZFおよびPD-1のうちの少なくとも1つの発現の減少を示す、請求項1記載の富化された集団または単離された集団。 - 前記適応型NK細胞が、
(i)標準NK細胞と比較して抗腫瘍免疫活性の増強を示す、
(ii)骨髄由来サプレッサー細胞(MDSC)が誘発する免疫応答の抑制を克服することができる、
(iii)Tregが誘発する免疫応答の抑制を克服することができる、または
(iv)標準NK細胞と比較して長命である、請求項1記載の富化された集団または単離された集団。 - 前記培地が、(i)CD155阻害剤、TIGIT阻害剤、および反応性酸素種(ROS)産生阻害剤のうちの少なくとも1つ、ならびに/または(ii)薬剤的に許容できる担体、をさらに含む、請求項1記載の富化された集団または単離された集団。
- (i)ROS産生阻害剤が、カタラーゼを含む、または(ii)ROS産生阻害剤もしくは前記CD155阻害剤が、骨髄由来サプレッサー細胞(MDSC)上のCD155の発現を減少させるのに十分な量で存在する、請求項9記載の富化された集団または単離された集団。
- 請求項1~10のいずれか1項に記載の富化された集団または単離された集団を含む、癌、前癌状態またはウイルスを治療または予防するための、治療剤又は予防剤。
- 前記対象が骨髄由来サプレッサー細胞(MDSC)を含む、請求項11記載の治療剤又は予防剤。
