Description
a)海藻及び水を含有し、少なくとも5wt%の乾燥固形分(DS)含有量を有する、バイオマスを提供するステップと、
b)バイオマスを浸出プロセスにかけて、海藻の内部に存在する水を浸出させるステップと、浸出された海藻を含む浸出されたバイオマスを得るステップと、
c)任意選択で、浸出されたバイオマスを最大で40wt%の水分レベルまで乾燥させて、乾燥した浸出されたバイオマスを得るステップと、
d)浸出されたバイオマスを塩水で調理して、調理されたバイオマスを得るステップと、
e)任意選択で、調理されたバイオマスを洗浄及び/又は乾燥させるステップと、
f)ステップd)又はe)の調理されたバイオマスを海藻ベースの粉末に変換するステップと、
を含む。
特に、本発明の粉末は、食品の機能性(栄養、感覚、及び物理化学)に関連する輸送、拡散、及び溶解現象の最適化を可能にし得る。更に、当該製品は、特定の流動挙動、テクスチャ、及び外観を有するように容易に設計され得る。したがって、当該食品機能性を最適化する本発明の粉末の能力は、食品構造の設計に非常に有益であることがあり、これは、古典的なニーズ(例えば、テクスチャ及び口当たり)と共に、異なる生理学的応答をトリガするために調節された消化を含むウェルネス及び健康に対する影響を強化し得る。
測定方法
・Cl-量は、AgNO3による電位差滴定(Metrohm)によって測定した。200~300mgの試料(W試料)を、250mLビーカー内の150mLの浸透水に添加した。試料の均質な分散が達成されるまで、試料を撹拌した。4~5滴の発煙硝酸を試料に添加した。滴定は、ポテンショメータ(682 Titroprocessor、Metrohm)及び複合電極Ag/AgNO3を用いて行った。塩化物のwt%は、式:%Cl-=V×C×M[Cl]×100/W試料で直接計算することができ、式中、M[Cl]は35.5g/molであり、Vは使用されるAgNO3(mL)溶液の体積であり、Cはその濃度、すなわち0.1Nである。
・AIMは、250mLビーカー内の150mLの浸透水中に0.5gの試料(Wsample)を分散させることによって測定した。1.5mLの濃硫酸を加えた。ビーカーをプラスチックホイルで覆って蒸発を防止し、沸騰温度で2時間ベンマリーで加熱した。分散体を4000 rpm(3250gに相当)で10分間遠心分離した。
AP25フィルタ及び結晶皿の総質量(Wフィルタ+皿)を決定した。酸性分散液を濾過し、(pH紙で確認して)そのpHが中性を維持するまで50℃の浸透水ですすいだ。約500 mLの水を使用した。
試料を有するフィルタを室温で一晩乾燥させ、60℃のオーブン内で1日更に乾燥させ、試料、フィルタ及び皿の総重量を決定した(W最終)。AIM(%)=[(W最終-Wフィルタ+皿)/W試料]×100.
・AIAを以下のように測定し、2.000(2)グラム(W試料)の試料をシリカ又は白金るつぼ上に置き、500℃のホットプレート上で約1時間燃焼し、その後550℃の炉内に16時間置いた。得られた灰を、10mLの濃HCl及び20mLの脱塩水を含有する溶液に添加した。灰を含有する溶液を約30分間80℃に加熱し、続いてWhatman N°40(無灰フィルタ)を用いて濾過した。灰を含むフィルタを、試料中にCl-が検出されなくなるまで水ですすいだ。試料中のCl-の存在をAgNO3で確認した(AgClの沈殿はCl-の存在を意味する)。
第2のシリカ又は白金るつぼを550℃のオーブンに10分間入れ、次いでデシケータ内で室温に冷却した。続いて、るつぼを、水を含まない環境で計量(Wるつぼ)した。灰を含むフィルタをるつぼ上に置き、室温から500℃まで、少なくとも1時間にわたって、ホットプレート上で徐々に加熱した。次に、るつぼを炉に移し、800℃で16時間加熱した。デシケータ内で室温に冷却した後、水を含まない環境でるつぼの重量を再度測定した(Wるつぼ+灰)。AIA(%)=[(Wるつぼ+灰 - Wるつぼ)/W試料]×100。
・D50,D90,D10及びD[4,3]:粒径分布を決定する方法は、米国薬局方(USP40)の方法<429>に準拠し、ISO規格13320-1に基づく。試料粉末を最初に振動ホッパー内に注ぎ、Mastersizer 3000(Malvern)を定期的に流す。空気分散装置を使用して、検出器が十分な信号対雑音比に到達し、複数の散乱を回避するために、1~15%の光を遮って粉末粒子をレーザビームによって吹きつけた。異なる角度で粒子によって散乱された光は、多素子検出器によって測定される。赤色及び青色光を使用して、Mie理論に結び付けると、体積サイズ分布の計算が可能になり、粒子は球体と見なされ、したがって同等の球サイズが決定された。得られたサイズ分布から、10、50及び90%での累積体積分率が決定され、それぞれD10、D50及びD90を得た。メジアン径D50は、粉末の粒径の考えを提供するが、D10及びD90は、より微細及びより粗い粒径を定量化することを可能にする。
・CIELAB L*、a*、及びb*は、International Commission on Illumination(Commission Internationale d’Eclairage)によって指定される最も完全な色空間を表す。これは、人間の目に見える全ての色を記述し、参照として使用されるデバイスに依存しないモデルとして機能するように作成された。試料のL*値とb*値は、試料を比色計の(約半分充填された)ガラスセルに配置することによって取得される。使用した比色計は、Minolta CR400 Colorimeterであった。

再構成された脱脂乳を水性媒体として使用した。粉末形態の脱脂乳は、Isigny-Ste-Mere(Isigny,フランス)によって提供された。脱脂乳は、室温で4時間撹拌しながら、粉末脱脂乳を10%w/wで超純水(18.2MΩ.cm抵抗率)に溶解して再構成された。特に、1000gの再構成脱脂乳を調製するために、108.66gの脱脂乳粉末(DS=92.03wt%)を、891.34gの超純水に溶解した。様々な海藻ベースの粉末の分散体を、再構成した脱脂乳中に様々な割合(0.1~1%w/w乾燥物基準)で調製した。海藻ベースの粉末を好適な最終比例で秤量し、5wt%スクロース(再水和を促進するため)と完全に混合し、磁気撹拌(500rpm)下で、再構成した脱脂乳中にゆっくりと分散させた。撹拌を室温で30分間維持した。続いて、試料を500rpmで撹拌しながら約30分間80℃に加熱し、この温度で更に3分間保持した。
貯蔵弾性率G’の測定
レオロジー測定は、上面と下面の両方にクロスハッチングが施された50mmのplate-and-plate形状を備えたMCR 302制御応力レオメータ(Anton Paar Physica)を使用して行われました。レオメータはまた、ペルチエ温度制御装置も備えている。間隙を1mmで固定した。試料の縁部上のパラフィン油の薄層によって試料を被覆して、測定中の蒸発を回避した。動的振動又は粘弾性測定値を選択して、各配合システムのゲル化速度及びテクスチャ化特性を評価した。これらの測定のために、試料を、80℃で予熱したMCR 302プレート上に注ぎ、80℃から10℃まで温度掃引試験(2℃/分)に供し、続いて0.4Hzの周波数で15分間の時間掃引実験に供して、再編成(構造再配置)により、システムが10℃でのこの検討された時間後に平衡状態に達することを確実にした。続いて、試料は、0.2%に固定された線形粘弾性領域(LVE)の一定の剪断歪みで100~0.01Hzの周波数掃引に供された。粘弾性測定がLVEドメインで確実に実行されるように、0.4Hzで歪み掃引実験が0.01%から100%まで実施された。
全てのこれらのレオロジー実験において、各測定は、少なくとも2回実施された。
データ処理:G’
この特許において考慮されるG’値は、10℃で0.4Hzの機械的スペクトル(周波数掃引試験)から収集された。実際に、機械的スペクトルは、得られたゲルの実際の構造的挙動を表すため、このG’値を最も適切なパラメータとして使用することが好適であるように見えた。
様々な濃度の全ての調査された試料について得られたG’値に基づいて、データを説明するために、べき法則関係(式1を参照)を使用した。C*は、それ以下ではゲル様挙動が存在しない最低濃度を表し、又は臨界ゲル化濃度を暗示的に表すことに留意されたい。Cは海藻ベースの粉末濃度(乾燥物基準)であり、nは、フィッティングモデルの指数値を表し、k及びk’は、フィッティングモデルの定数係数である。

試料を比較するために、式2~4を使用した。

式中、pは並進シフト係数である。p=1の場合、試料Aは試料Bと同様のゲル強度を示すことを意味する。p>1の場合、試料Bは試料Aよりも高いG’を表示することを意味する。p<1の場合、試料Bは試料Aよりも低いG’を表示することを意味する。
データ処理:C0
C0の決定のために、以下のステップを考慮した。
(i)上記の機械的スペクトルから収集された貯蔵弾性率G’値を、海藻ベースの粉末濃度C(%、DS)の関数として対数スケールでプロットした(図1を参照)。
図1において、破線及び実線は、それぞれ、実験データ(生データ)及び推定データに対するべき法則の式3及び1のフィッティングを表す。図1で使用されるデータは、それぞれ、実施例1及び比較例1に属する。
(ii)文献に記載されているアプローチに従う(例えばAgoda-Tandjawa、G.、Dieude-Fauvel、E.、Girault、R.&Baudez、J.-C.(2013)。Chemical Engineering Journal,228,799-805)式G’=kCnは、線形回帰を使用して、G’=k’(C-C0)nの形式に数学的に変換された。この第2の式では、k’は倍率を表し、C0は、それ以下ではゲル様挙動が得られない濃度を表す。G’=kCn=k’(C-C0)nの条件に従って、全ての調査された海藻ベースの粉末について線形回帰を実施し、両方の指数(n)値は同一であり、C>C0であることに留意されたい。
上記のフィッティングモデルを使用して決定されたC0の検証は、上述のものと同様の条件で全ての海藻ベースの粉末のレオロジー挙動を評価して、ゲル様挙動を証明することによって検証された。
実施例1:カッパフィカス属アルベレッチー種(Kappaphycus alvarezii)ベースの粉末
実施例1を繰り返したが、海藻はキリンサイ属スピノサム種(Eucheuma spinosum)であり、塩水は250g/LのKClを含み、調理したバイオマスは水中で3回洗浄した。得られた海藻ベースの粉末の特性を表4に示す。
本発明の粉末と市販の海藻粉末との比較
実施例2のコットーニィ(Cottonii)粉末は、乳製品デザート(クリーミー及びゲル化テクスチャ)において試験されている。
クリーミーデザートのレシピ
以下のプロセス及びレシピを使用した。
プロセスステップ
1.全ての乾燥成分を予めブレンドし、冷たいミルク及びクリームに分散させた。
2.水和30分
3.予備加熱63℃
4.80Barでの均質化
5.95℃で2分間の低温殺菌
6.135℃で15秒間の滅菌
7.75℃で予備冷却
8.10℃で冷却
9.10℃で4時間の間、「緩衝槽」に保管する
10.ポット内の充填
成分
粘度を、Brookfield RV粘度計及びHelipath(商標)システムを用いて10℃のデザート試料に対して測定した。Brookfield Helipath(商標)は、Brookfield Viscometerをゆっくりと下げ、その結果、その回転する特殊なTバー型スピンドルがデザート試料を通る螺旋状経路を描くように設計された。回転速度を5rpmに設定し、6回の回転値の平均を計算することにより、相対センチポアズ値で7日目及び21日目に粘度を測定した。図2は、2つのクリーミーデザート試料の粘度測定値が類似していることを示し、本発明の粉末が市販の品質改良剤の機能性に匹敵する能力を有することを示した。
官能評価により、本発明の粉末を使用したクリーミーデザートは、滑らかで光沢のあるテクスチャ及びクリーミーな口当たりを有する、基準に近い非常に良好なクリーミーなテクスチャを示すことが示された。
ゲル化デザートのレシピ
ゲル化デザートは、以下のプロセスで作製され、表Bに示される成分を使用して、実施例2の粉末及び実施例1と同様のプロセスを用いてスピノサム(spinosum)から作製された粉末を使用して作製された。
プロセス
1.全ての乾燥成分を予めブレンドし、冷たいミルク及びクリームに分散させた。
2.水和30分
3.予備加熱63℃
4.80Barでの均質化
5.95℃で2分間の低温殺菌
6.135℃で15秒間の滅菌
7.75℃で予備冷却
8.70℃で4時間「緩衝槽」に充填する
9.ポット内の充填
成分
TA.XTplusテクスチャアナライザプラスは、ゲル化乳製品デザートのグラムにおける力、ゲル強度プロファイルを測定するために用いられている。以下の試験パラメータが使用され、直径1インチの円筒型プローブと、5℃の試験試料温度と、3つの試験試料と、1mm/秒の試験速度と、20mmの距離である。
図3は、各デザートトライアルで行われた3つの測定の平均をプロットし、本発明の粉末で作製された試料のゲル強度が、市販の品質改良剤が使用されたものとほぼ同じであることを示している。
官能評価により、本発明の粉末に基づくゲル化デザートは、市販のものと同様の滑らかな表面、クリーミーな口当たり及び中程度の硬さを有する非常に良好なテクスチャを有することが示された。
Claims (15)
- 巨視的な多細胞性の海洋藻類の少なくとも80wt%の乾燥基準の粒子を含む海藻ベースの粉末であって、再構成した脱脂乳の溶液の総重量に対して前記粉末の0.3wt%を含む再構成した脱脂乳の溶液で測定して、少なくとも30Paの貯蔵弾性率(G’)を有し、G’は前記溶液を10℃で0.4Hzの周波数掃引試験に供することによって決定し、前記脱脂乳は、室温で4時間撹拌しながら、18.2MΩ.cm抵抗率を有する超純水に10%w/wで粉末脱脂乳を溶解して再構成されており、前記粉末が、最大で0.1wt%の臨界ゲル化濃度(C0)を有する、海藻ベースの粉末。
- 前記粉末の総重量に基づいて、最大で25wt%の含水量を有する、請求項1に記載の粉末。
- 前記G’が少なくとも50Paである、請求項1又は2に記載の粉末。
- 前記C0が、0.001~0.100wt%である、請求項1~3のいずれか一項に記載の粉末。
- 前記粉末が、少なくとも50のCIELAB L*値を有する、請求項1~4のいずれか一項に記載の粉末。
- 前記粉末が、前記粉末の総重量に基づいて、少なくとも1wt%の食物繊維成分を含む、請求項1~5のいずれか一項に記載の粉末。
- 前記粉末が、前記粉末の総重量に基づいて、少なくとも0.1wt%のタンパク質成分を含む、請求項1~6のいずれか一項に記載の粉末。
- 前記粉末が、前記粉末の総重量に対して最大で20wt%のCl-含有量を有する、請求項1~7のいずれか一項に記載の粉末。
- 前記粉末が、前記粉末の総重量に対して最大で50wt%の量の酸不溶性材料(AIM)を有する、請求項1~8のいずれか一項に記載の粉末。
- 前記粉末が、前記粉末の総重量に対して最大で5.0wt%の量の酸不溶性灰(AIA)を有する、請求項1~9のいずれか一項に記載の粉末。
- 前記海藻が、アマノリ属の一種(Porphyra sp.)、ダルス(Palmaria Palmata)、キリンサイ属スピノサム種(Eucheuma spinosum)、キリンサイ(Eucheuma denticulatum)、キリンサイ属の一種(Eucheuma sp.)、キリンサイ属コットーニィ種(Eucheuma cottonii(カッパフィカス属アルベレッチー種(Kappaphycus alvarezii)としても知られる))、オオキリンサイ(Kappaphycus striatus)、オオキリンサイ属の一種(Kappaphycus sp.)、ツノマタ属トチヤカ種(Chondrus crispus)、アイリッシュモス(Irish moss)、Fucus crispus、ツノマタ属の一種(Chondrus sp)、Sarcothalia crispata、アカバギンナンソウ属laminaroides(Mazzaella laminaroides)、アカバギンナンソウ属の一種(Mazzaella sp.)、スギノリ属acicularis(Chondracanthus acicularis)、スギノリ属chamissoi(Chondracanthus chamissoi)、スギノリ属の一種(Chondracanthus sp.)、スギノリ属pistilla(Gigartina pistilla)、スギノリ属mammillosa(Gigartina mammillosa)、スギノリ属skottsbergii(Gigartina skottsbergii)、スギノリ属の一種(Gigartina sp.)、オゴノリ属の一種(Gracilaria sp)、テングサ属の一種(Gelidium sp.)、イボノリ属stellatus(Mastocarpus stellatus)及びこれらの混合物からなる海藻の群から選択される、請求項1~10のいずれか一項に記載の粉末。
- 請求項1~11のいずれか一項に記載の前記粉末と、追加の化合物とを含む組成物であって、前記化合物が粉末状又は非粉末状形態であり、前記製品が、グアーガム、キサンタンガム、ローカストビーンガム、カッシアガム、タラガム、コンニャクガム、アルギン酸、寒天、カラギーナン、β 1,3グルカン、デンプン、及びこれらの組み合わせからなる群から選択される、組成物。
- 請求項1~12のいずれか一項に記載の前記粉末又は前記組成物を含む食品製品、飼料製品、パーソナルケア製品、医薬製品又は工業製品。
- 前記製品が乳製品である、請求項13に記載の製品。
- 請求項1~11のいずれか一項に記載の前記海藻ベースの粉末を製造する方法であって、
a.海藻及び水を含有し、少なくとも5wt%の乾燥固形分(DS)含有量を有する、バイオマスを提供するステップと、
b.前記バイオマスを浸出プロセスにかけて、前記海藻の内部に存在する前記水を浸出させるステップと、浸出された海藻を含む浸出されたバイオマスを得るステップと、
c.任意選択で、前記浸出されたバイオマスを最大で40wt%の水分レベルまで乾燥させて、乾燥した浸出されたバイオマスを得るステップと、
d.前記浸出されたバイオマスを塩水で調理して、調理されたバイオマスを得るステップと、
e.任意選択で、前記調理されたバイオマスを洗浄及び/又は乾燥させるステップと、
f.ステップd)又はe)の前記調理されたバイオマスを海藻ベースの粉末に変換するステップと、
を含む、方法。
