Description
(a)極性アミノ酸残基と非極性アミノ酸残基が交互に配置された、4~34個のアミノ酸残基からなるアミノ酸配列を含むペプチド、または、
(b)7位の非極性アミノ酸残基を中心として、N末端方向およびC末端方向に、非極性アミノ酸と極性アミノ酸とが交互に、且つ、対称の位置に配置される、13個のアミノ酸残基からなるアミノ酸配列を含むペプチド、であることを特徴とする。
前記非極性アミノ酸残基が、アラニン残基、バリン残基、ロイシン残基、イソロイシン残基、メチオニン残基、フェニルアラニン残基、トリプトファン残基、プロリン残基、および、グリシン残基からなる群から選択されるアミノ酸残基であることを特徴とする。
Uchida.J Pharmacol Sci 2005に記載の方法(図1)に基づいてTrinitrobenzene sulphonic acid(TNBS)惹起性の大腸潰瘍モデルラットを作製し、本発明の医薬組成物の潰瘍修復効果を検討した。
7週齢雄性SDラットを麻酔下に開腹後、リングピンセットにより大腸の一部を固定し、管腔内にTNBS+ethano1溶液0.2mLを注入して限局性潰瘍モデルを作製した。2日目に、本発明の自己集合性ペプチド溶液(PuraMatrix(商標)、スリー・ディー・マトリックス社、東京)(n=6)、または生理食塩水(n=7)を大腸内視鏡観察下に潰瘍部に適用し、2日目、4日目、7日目に内視鏡で潰瘍サイズを観察した。7日目に屠殺し、大腸の潰瘍面積、腸重量(g/3cm)、組織所見を評価した(図2、図3)。
ラットの体重の変化はPuraMatrix適用群と対照群で同等であった(図4)
内視鏡での評価:PuraMatrix適用群では、対照群に比べて潰瘍サイズが縮小していた(図5)。
屠殺後の評価:潰瘍面積はPuraMatrix適用群の方が対照群に比べ有意に縮小していた(P=0.024)。腸重量もPuraMatrix適用群の方が対照群に比べ低下傾向にあった(図6)。
以上の結果から、本発明の医薬組成物が炎症性腸疾患における腸管の難治性潰瘍の治療に有効であることが示された。
実施例1と同様の手順で大腸潰瘍モデルラットを作製し、本発明の医薬組成物の潰瘍修復効果を分子レベルから検討した。
実施例1と同様の手順で大腸潰瘍モデルラットを作製し、本発明の自己集合性ペプチド溶液(PuraMatrix(PM):n=6)または生理食塩水(Control:n=7)を潰瘍部位に投与し、7日目に潰瘍部位の組織を採取した。採取した組織における各種サイトカイン量を、PCRにより測定した。
結果を図7~10に示す。測定した各種サイトカインのうち、炎症性サイトカインであるIL-1a、IL-6に関しては、本発明の医薬組成物を投与した群において発現の有意な低下がみられた(図7)。また、膜たんぱく質であり腸管保護作用を有するClaudin 1に関しては、本発明の医薬組成物を投与した群において発現の有意な上昇がみられた(図9)。
以上の結果から、本発明の医薬組成物が炎症性腸疾患における腸管の難治性潰瘍の治療に有効であることが、分子レベルにおいても示された。

- 潰瘍性大腸炎またはクローン病に起因する腸管における潰瘍または瘻孔の治療または予防のための医薬組成物であって、
