Description
本出願は、2017年5月16日に出願された米国出願第62/506,892号の優先権を主張する。先の出願の開示は、本出願の開示の一部とみなされ、その全体が本出願に組み込まれる。
本明細書は、抗体中和(例えば、麻疹ウイルスワクチンからの血清による抗体中和)に対する感受性が低下したウイルス(例えば麻疹ウイルス又はアデノウイルス)を作製及び使用するための方法及び材料に関する。
麻疹ウイルス(MV)は、5歳未満の子供を中心に毎年多くの死亡を引き起こす。ワクチン接種を受けていない子供は、麻疹及び麻疹に関連する死亡のリスクが最も高い。特に、移行性の抗麻疹抗体価が非防御レベルまで低下しているが、まだ幼すぎて9~12ヶ月の小児に推奨される現在の麻疹ワクチンを接種できない小児は、麻疹及び麻疹に関連する死亡のリスクが高くなる可能性がある。
本明細書は、核酸、ポリペプチド、並びに核酸及び/又はポリペプチドを含むウイルスを提供する。本明細書はまた、ウイルスを使用してがん患者を処置したり、乳児にワクチン接種を行って乳児をMV感染から防御する方法を提供する。例えば、本明細書は、MV ヘマグルチニン(H)ポリペプチド、MV Hポリペプチドをコードする核酸、CDV Fポリペプチド、CDV Fポリペプチドをコードする核酸配列、並びにかかる核酸及び/又はポリペプチドを含むウイルスを提供する。例えば、本明細書は、改変型Hポリペプチドをコードする核酸及び改変型Fポリペプチドをコードする核酸を含む組換えウイルス(例えば、MV又はアデノウイルス(Ad))を提供する。かかる組換えウイルスは、抗体中和に対する感受性の低下、膜融合を引き起こす能力の低下、及び/又は複製適合性(replicative fitness)の減少を示し得る。本明細書に記載の組換えウイルスは、野生型ウイルスのように効率的に細胞(例えば、Vero細胞及びHeLa細胞などのヒト細胞)中で増殖し得る。本明細書に記載のウイルスを使用して、がん患者を処置し、又はウイルスが抗体中和に対する感受性の低下を示すように乳児をワクチン接種し得る。ある場合には、本明細書に記載の組換えウイルスを使用して、既存の麻疹免疫を有する患者においてがんを処置し得る。ある場合には、本明細書に記載の組換えウイルスを使用して、中和抗麻疹抗体(例えば、経胎盤的に獲得された中和抗麻疹抗体)を有する子供をワクチン接種し得る。
麻疹ウイルス(MV#1)を、以下のアミノ酸置換(配列番号1に関して):H17S、D149N、S189P、G211S、E235G、N238D、S240N、L249P、L276G、V280I、N282K、G302R、E303G、Y310C、Q311R、Q334H、A359T、K364N、R377Q、M378K、P397L、N405S、D416N、T420A、V421A、L423P、F476L、N481Y、K488E、G491D、H495R、D505T、R533G、S546G、R547G、F552V、V562T、D574A、K576R、I594L、G603E、T609N、G613E、及びT614Aを有する改変型Hタンパク質(配列番号3)を有するように作製した。この改変型Hタンパク質(配列番号3)は、野生型MVi/Madrid.SPA/50.10 (遺伝子型H1)株の麻疹ウイルスヘマグルチニンタンパク質(配列番号9)に19個の点変異を導入することにより作製した。その19個の点変異は、S189P、E235G、N238D、L249P、G302R、Y310C、Q311R、R377Q、M378K、D416N、N481Y、K488E、G491E、H495R、D505T、R533G、S546G、R547G、及びF552Vである。
この実施例2では、MVはMeVと称し、MV#1はMeVΔ7、Δ7又はΔ8と称し、MV#2はMeV#1と称し、MV#3はウイルス3と称し、MV#4はMeV#4と称し、MV#5はMeV#2と称する。これらのいくつかの合成は、実施例2で再び説明し、実施例1で提示されたデータの一部は実施例2でも提示する。さらに、MeVΔ7を使用してΔ8ウイルスを生成した。
Vero細胞(CCL-81, ATCC)、安定にトランスフェクトされたVeroヒト(Vero/hSLAM)(Ono et al., J. Virol., 75(9):4399-401 (2001))及びイヌ(Vero/dogSLAM)(von Messling et al., J. Virol., 77(23):12579-91 (2003))SLAMを5%(vol./vol.)の熱不活化ウシ胎仔血清(FBS)(Gibco)及び0.5mg/mLのジェネティシン(G418; Corning)(Vero/hSLAM)又は1mg/mLのゼオシン(ThermoFisher, Walthman MA)(Vero/dSLAM)を添加したダルベッコの修正最小必須培地(DMEM)(HyClone, GE Healthcare Life Science)で増殖させた。チャイニーズハムスター卵巣(CHO)細胞、CHO-CD46(Nakamura et al., Nat. Biotechnol., 22(3):331-6 (2004))、CHO-SLAM(Tatsuo et al., Nature, 406:893-6 (2000))、及びCHO-N4(Liu et al., J. Virol., 88(4):2195-204 (2014))を記載のとおりに培養した。ベビーハムスター腎臓細胞(BHK)を、DMEM-10%FBSで維持した。ウイルスは他で説明されているように増殖させた(Munoz-Alia et al., J. Virol., 91(11):e00209-17 (2017);Munoz-Alia et al., PLoS One, 13(2):e0192245 (2018))。
組換えMeVは、p(+)MeVvac2(EGFP)Nプラスミドに含まれるMoraten/Schwartzワクチン株の分子cDNAクローンに基づく(del Valle et al., J. Virol., 81(19):10597-605 (2007))。このプラスミドでは、強化型緑色蛍光タンパク質(EGFP)をN遺伝子の上流に挿入した。細菌内での増幅時のプラスミド不安定性を回避するために、2つのアプローチに従って段階的にプラスミド骨格をpSMART(登録商標)LCkanベクター(Lucigen, Middleton, WI)に置き換えた。最初のアプローチでは、SacII及びNotI制限酵素を含むマルチクローニングサイトをベクターに付加した。次に、最適なT7プロモーターとそれに続くハンマーヘッド型リボザイム(HHrbz)(図7)を、フォワードプライマーに直接配列を挿入することによりウイルスゲノムの上流に挿入し、P遺伝子の開始に位置する特有の内部制限部位SacIIまでMeVゲノムを増幅した。次に、p(+)MVvac2(EGFP)NプラスミドのSacII-NotI断片を、同様に消化されたpSMART(登録商標)LCkanベクターに挿入した。2番目のアプローチでは、ヒト伸長因子1αコアプロモーター、キメライントロン、T7 RNAポリメラーゼプロモーター、HHrbz及びクローニングサイトを含むカセットを合成し、ベクターに連結した。すべてのプラスミドの増殖は、30℃で増幅させた大腸菌Stbl2TM細胞(Invitrogen, 10268019)で行った。
細胞(6ウェルプレートで5×105/ウェル)にFugene HD(Promega)を使用して、ワクチン株MeV-Fをコードする(1μg)pCGプラスミドと適切なMeV-HをコードするpCGを同時トランスフェクトした。Hema-Quik染色(Fisher Scientific 123-745)の24時間後に融合活性を評価した。
細胞を洗浄し、Versene(Gibco)を使用して剥離し、すぐにフィコエリトリン結合抗体抗SLAM(FAB1642P; R&D Systems)、抗CD46(FAB2005P; R&D Systems)、及び抗nectin-4(FAB2659P; R&D Systems)、又は対照アイソタイプ抗体(IC0041P; R&D Systems)と共にインキュベートした。4℃で1時間インキュベートした後、細胞を再度洗浄し、FACSCantoフローサイトメトリシステム(BD Bioscience)で蛍光を測定した。細胞あたりの受容体の数は、較正ビーズ(BD QuantiBrite; BD Biosciences)を参照して推定した。
CD46エクトドメイン(残基35~328)のコード配列をPCRによりpGEM-CD46ベクター(Sino Biologicals Inc., HG12239-G)から増幅し、In-Fusionクローニングキット(Clontech)を使用してマウスIgκ鎖リーダー配列及びFc領域の5'末端にある3Cプロテアーゼ切断配列とインフレームでpFUSEベクター(pfc1-hg1e3; Invivogen)に挿入した。CD46-Fc、SLAM-Fc、及びnectin-4-Fc(Munoz-Alia et al., PLoS One, 13(2):e0192245 (2018))組換えタンパク質はExpi293細胞(Gibco)で発現し、他で記載されているように培養上清から精製した(Munoz-Alia et al., PLoS One, 13(2):e0192245 (2018))。組換え可溶性MeV-Hの発現及び精製は、他で記載されているように行った(Munoz-Alia et al., J. Virol., 91(11):e00209-17 (2017))。受容体-FcのMeV-Hへの結合は、他で記載されている酵素結合免疫吸着アッセイによって決定した(Munoz-Alia et al., PLoS One, 13(2):e0192245 (2018))。450nmでの吸光度をInfinite M200Proマイクロプレートリーダー(Tecan)で測定した。Prismソフトウェア(GraphPad)を使用してデータを分析し、一部位結合飽和モードに調整して、半飽和濃度(見かけのKd値[解離定数])を決定した。報告された値は優れた適合性を示した(R2>0.99)。
ウイルス調製物をDTTの存在下で加熱し、4~12%のビス-トリスポリアクリルアミドゲルに分画し、PDVF膜にトランスファーした。次に、ブロットを、コンジュゲート二次ウサギ抗体(ThermoFisher, #31642)でプローブした抗GFP、抗MeV-Hcyt、抗MeV-N及び抗MeV-Fで分析した。ブロットは、SuperSignal Wester Pico化学発光基質(ThermoFisher)で明らかにし、ChemiDocイメージングシステム(BIO-RAD)で分析した。
ウイルス中和アッセイは、他で説明されている蛍光ベースのプラーク減少微量中和(PRMN)アッセイに基づいて実施した(Munoz-Alia et al., J. Virol., 91(11):e00209-17 (2017);Munoz-Alia et al., PLoS One, 13(2):e0192245 (2018);及びMunoz-Alia et al., Virus Res., 236:30-43 (2017))。各アッセイは、アッセイごとに4回重複して異なる日に少なくとも2回繰り返した。GraphPadソフトウェア(Prism 7)を使用してS字型用量反応(可変勾配)にデータをフィッティングした後、50%阻害濃度(IC50)を計算した。
MeV-Hステルスのモデルは、プログラムPhyre2を使用して90%を超える信頼度で生成された(Kelley et al., Nat. Protoc., 10(6):845-58 (2015))。次に、GlyProサーバ(http://www.glycosciences.de)を使用したin silicoグリコシル化のために構造を提出した。これは、無秩序な領域の部分である、N168及びN187を含む予測されるすべてのN-グリコシル化部位で複雑な五分岐N-グリカンモデルを生成した。PyMOLソフトウェア(http://pymol.org)を使用してMeV-H/CD46結晶学的共構造(PDB 3INB)のCD46受容体を重ね合わせ、操作した。
統計学的有意性は、適切な統計学的検定に従ってGraphPad Prism 7で計算した。
MeV-Hへの抗原ドリフトのモデリング
MeV-Hには7つの主要な抗原部位があり、これらの部位のうち4つまでの複数の破壊はポリクローナル抗体の中和を無効にしない(Munoz-Alia et al., J. Virol., 91(11):e00209-17 (2017);Lech et al., PLoS One, 8(1):e52306 (2013);及びMunoz-Alia et al., PLoS One, 13(2):e0192245 (2018))。B細胞の免疫優位性の欠如により、すべての抗原部位の除去により、中和不可能な変異体が生成される可能性がある。これを調査するために、MeV-Hについて記載されているすべてのエピトープを体系的に破壊した。実験計画は、遺伝子型H1のMeV-H背景に自発的中和mAbエスケープ変異体選択を組み込むことに基づいている。この特定の株は、最も抗原的に高度なMeV-Hの1つであるという以前の観察に基づいて選択され(Munoz-Alia et al., PLoS One, 13(2):e0192245 (2018))、これは、それ以外の場合は剛性のMeV-Hタンパク質の変更を最小限にし得る(Fulton et al., Cell Rep., 11(9):1331-8 (2015))。nAb結合領域のリストを作成し、それらの領域の破壊を、ここではΔ7という名前(実施例1ではMV#1としても知られる)の単一のMeV-Hと合わせた(表3)。
抗原部位III(受容体結合部位、RBS)の変異は野生型向性(SLAM及びnectin-4)と互換性がないため、いくつかのアミノ酸置換:N481Y(Lecouturier et al., J. Virol., 70(7):4200-4 (1996))、H495R(Okada et al., J. Virol., 83(17):8713-21 (2009))、及びS546G(Shibahara et al., J. Gen. Virol., 75:3511-6 (1994))を介して受容体特異性をCD46にスイッチする計画とした。受容体依存性融合活性におけるアミノ酸置換の影響を評価するために、ワクチン由来のMeV-Fと組み合わせてMeV-H変異体の一過性発現を行った。明らかな合胞体形成がなくても細胞間伝播が起こる可能性があるため、対応する組換えMeVは逆遺伝学によってもレスキューした(Langedijk et al., J. Virol., 85(21):11242-54 (2011))。結果を図8に示す。MeV-Hワクチン株(A)を使用した場合、SLAM、CD46、又はnectin-4のいずれかを発現するCHO細胞でウイルスの侵入と細胞融合の両方が観察された。同様に、MeV-H H1は、SLAM及びnectin-4発現細胞でのウイルス侵入と合胞体形成を可能にした。後者の背景にN481Y、H495R、又はH495R/S546Gを導入しても、一過性トランスフェクションアッセイで見られるように、CD46依存性の融合活性は有意に増加しなかった。しかし、N481Y及びH495R/S546G変異体の合胞体形成がない場合、ウイルスの侵入が観察された。N481Y変異体にH495Rを含めると、CD46依存性融合のレベルがMeV-H Aで観察されるレベルにまで回復した。この変異体にS546Gを追加すると、CD46依存性融合がほぼ2倍に増強され、同様の増加がN481Y/S546G変異体で観察された。それにもかかわらず、トリプル変異体(N481Y/H495R/S546G)が使用された場合にのみCD46依存性増強感染が観察されたため、この組み合わせをnAbエスケープ変異の背景として選択した。
表3は、MeV-H球状ドメインエスケープウイルスを生成するための基礎として最初に使用した。この情報とトリプルCD46向性置換を一緒に使用して、Δ7ウイルスを直接操作し、これまでに説明された7つの操作上の非重複抗原部位をすべて破壊する(Φ、Ia、Ib、IIa、IIb、III、及びIV)(図9A)。導入された変異の数が各部位に特異的な他のnAbの中和を無効にするのに十分かどうかを判断するために、MeV-HA、H1及びΔ7を有するウイルスの中和感受性を30種のmAbのパネルに対して決定した。結果を図9Bに要約し、これは、Aウイルスが試験した30種のnAbすべてで中和されたが、この数はH1ウイルスでは18種に減少したことを示す。反対に、Δ7ウイルスはnAb BH030によってのみ中和された。
MeV-HのB細胞エピトープ破壊がその抗原性に影響するかどうかを判断するために、以下を実施した。マウスにMeV-Hをコードするプラスミドの流体力学的注射を与え、1ヶ月後に抗体反応を評価した(図11A)。中和アッセイにより、MeV-HΔ7で免疫したマウスはMeV-HAよりも中和抗体の力価が低いことが示されたが、おそらく変動のために統計的有意性に達しなかった(図11B)。それにもかかわらず、MeV-HΔ8は検出可能なレベルを生成することができなかった(図11C)。MeV-HΔ8がΔ8ウイルス自体の中和活性を調べることにより、新しいエピトープに対する非交差反応性抗体を生成しているかどうかを評価するために、以下を実施した。図11Cに示す結果は、MeV-H Aとは異なるが共通のエピトープを有することについて、Δ8ウイルスが、抗MeV- H H1及びΔ7によるのと同様に、同型抗体、すなわちMeV-HΔ8による免疫化によって引き起こされる抗体によって中和されたことを示した。MeV-HA免疫によって誘導された中和抗体は、Δ8ウイルスに対する中和活性を発揮しなかった。次に、複数の抗原部位を除去すると交差中和が無効になり、ウイルスは抗MeV-Hポリクローナル反応から回避することができる。
異なる動物モデルは、異なる抗体レパートリーを示す可能性がある(Nachbagauer et al., Nat. Immunol., 18(4):464-73 (2017))。MeV-H A免疫後のウサギで生じた抗体の呼気を中和分析により評価した。ウイルスΔ8は中和の傾向を示したが(図12A)、ワクチンウイルスと比較してND50力価の8倍(3 log2)の減少を示した。4倍の差(2 log2又は抗原単位(Smith et al., Science, 305(5682):371-6 (2004))以上の場合、ヒト季節性インフルエンザワクチン(Russell et al., Vaccine, 26(Suppl 4):D31-4 (2008);及びGarten et al., Science, 325(5937):197-201 (2009))のアップデートを保証するため、ウイルスΔ8はワクチンウイルスと抗原的に有意に異なると考えられた。一方、親前駆体が(H1)であったため、即時前駆体ウイルス(Δ7)は抗原的に区別できなかった。K471E変異はΔ7をΔ8ウイルスと区別し、抗原変異をもたらしたため、抗原の違いはすべてのmAbエスケープ変異体選択の組み合わせの結果であり、優性変異体の存在ではないことを確認したいという要望があった。したがって、他の遺伝子型特異的MeV-H遺伝子タンパク質を有する組換えMeVの以前のパネルを試験した(Munoz-Alia et al., J. Virol., 91(11):e00209-17 (2017);及びMunoz-Alia et al., PLoS One, 13(2):e0192245 (2018))。特に興味深いのは、MeV C2がK471E変異を有していたためである。しかし、PRMN力価の差は全体にわたって2倍未満であり、したがって重要ではないとみなされた。全体として、これらの結果は、麻疹ウイルスの抗原変異には、増分成分とパルス成分の両方があることを示唆している。すなわち、抗原の閾値を超えた場合、アミノ酸置換は累積的な効果を有する可能性がある。
麻疹免疫ヒト血清中のMeV-Fに対する中和抗体は、MeV-Hの蓄積的抗原置換の効果を緩衝し得る。ウイルス中和に対する2つのMeV糖タンパク質の寄与に関する洞察をさらに得るために、2つのアプローチを使用した:1)MeV-H及びMeV-F特異的抗体の枯渇(図13A)、並びに2)3つの異なる糖タンパク質交換を伴うウイルスキメラの同質遺伝子セットの中和感受性の研究。このエンベロープ交換ウイルスは、HとFを除くMeVに由来するすべての遺伝子を有し、それらを関連するが非交差反応性のイヌジステンパーウイルス(CDV)H及びFと単一及び二重のいずれかで交換した(Miest et al., Mol. Ther., 19(10):1813-20 (2011);及びZhang et al., Virology, 482:218-24 (2015))。したがって、CDV由来の遺伝子とのMeV-H及びMeV-Fタンパク質遺伝子の二重スイッチングによりMeV#2が生成され、MeV-H又はMeV-Fのいずれかでの単一スイッチにより、それぞれMeV#3及びMeV#4が生成された(図13B)。使用されたCDV-Hタンパク質遺伝子は、ヒト血清によるCDVの潜在的な交差中和を低減することが示されたため(Munoz-Alia et al., J. Virol., 91(11):e00209-17 (2017);Zhang et al., Virology, 482:218-24 (2015))、Ondersterpoortワクチン株を意図的に意図したY537D置換を有していた。親MeV(MeV#1)と3つのキメラ(MeV#2、MeV#3、MeV#4)はすべて、Vero細胞で区別できない合胞体を形成し、このことは異型の補完性を実証している(図13B)。
次に、両方のMeVエンベロープ糖タンパク質に対する抗体反応の幅が、MeVの抗原的に静的(staticity)であると仮定した。この仮説に対処するために、完全に抗原的に異なるウイルスの代用として、異型CDV-F(このウイルスを以後ステルスと称する)と組み合わせてΔ8ウイルスのレスキューを進めた。MeVレスキューシステムの堅牢性が改善されるまで、MeVステルスは得られなかった。親の組換えMeV Moratenワクチンのレスキューとは異なり、MeVステルスは、複数の独立したレスキューの試みの後に観察された単一のGFP陽性細胞から分離及び拡大された。5回の半盲検継代後、ウイルスは細胞単層を介して伝播した(図7)。ウイルスをさらに増殖させてウイルスストックを生成し、サンガー配列決定及びウェスタンブロット分析のために試験した。精製ビリオンのイムノブロッティングにより、ステルスが同型MeV-Fタンパク質を欠損し、それ以外の点ではワクチン株と同様のタンパク質含有物を示すことが実証された(図14B)。さらに、配列決定の結果は、糖タンパク質をコードする配列のいずれにも補償的変異を示さず、このことは、CDV-Fと組み合わせたMeV-HΔ8をコードするステルスウイルスの生存率をさらに確認した。
インフルエンザAウイルスでは、受容体結合アビディティと抗原変異は密接に関連しており(Hensley et al., Science, 326:734-6 (2009);及びLi et al., J. Virol., 87(17):9904-10 (2013))、ウイルスの適合性(fitness)の損失を補うことができる(Kosik et al., PLoS Pathog., 14(1):e1006796 (2018))。nAbエスケープ変異が存在する場合に受容体特異性が影響を受けるかどうかに対処するために、MeV受容体を単独で発現するCHO細胞に感染させた。予想外に、CD46及びnectin-4の密接な構造的及び機能的相互作用を考えると(図15)、ステルスウイルスは非常に効率的なCD46依存性融合を生成することが証明されたが、nectin-4を発現するCHO細胞では何も示さなかった(図16A)。細胞表面の分子数がそれら2つに匹敵し、SLAMを発現するCHO細胞の分子数(約20,000)の10倍多かったため、受容体密度の違いは、nectin-4の使用に対するCD46の識別の潜在的な説明として除外した(図16B)。CHO細胞のパネルにステルスと同じMeV-HΔ8をコードするΔ8ウイルスを感染させた場合も同様の結果が得られ、これは原因としてMeV-HΔ8とCDV-Fの間のアロステリック相互作用に反対するものである。しかし、MeVマトリックスタンパク質(MeV-M)とのさらなる相互作用は、依然として受容体結合相互作用に影響を与える可能性がある。次に、一過性トランスフェクションに基づく融合アッセイを、ウイルスの状況で観察された結果と並行するかどうかを研究した。このアプローチは、MeV-FをMeV-H A又はMeV-HΔ8と組み合わせて使用することであった。MeV-HΔ8は、CD46依存性融合に有意な影響を与えることなく、nectin-4を識別したことが示された(図16C)。同様のCD46依存性融合活性は、ヒト及びアフリカミドリザル細胞の両方で観察された(図17)。ELISAにより様々な組換えMeV-Hタンパク質(A、H1及びΔ8)の受容体結合解離定数を調べた(図18)。対照として使用された抗FLAG抗体とは異なり、組換え細胞受容体はMeV-Hタンパク質に対して異なる結合アビディティを示した(図16D)。MeV-HΔ8は、MeV-H A及びH1よりもCD46に約4000倍良好に結合し、見かけのKdは190pMで、MeV-H Aは819μMであった(Kdは不明であった)。nectin-4の結合値は3つの受容体すべてで最悪であり、使用した最高濃度では飽和に達しなかった。組換えMeV-Hタンパク質に対するnectin-4の見かけのKdに有意差は見られなかったが、MeV-HΔ8のBmax値は、MeV-H A及びH1と比較しておよそ半分低下した(それぞれ1.56、1.14、及び0.74)。予想どおり、MeV-HΔ8はSLAM-Fcへの無視できるほどの結合を示した。MeV-H H1は、MeV-H AよりもSLAM-Fcへの結合が低く、Kdは10.43μM及び2.68μMであったが、Bmax値も低かった(それぞれ2.09及び1.19)。まとめると、これらの結果は、MeV-HΔ8が、nectin-4については低下するがCD46とのアビディティ相互作用の増加を介して、nectin-4よりもCD46の使用を区別することを示している。

Claims (15)
- 麻疹ウイルスHポリペプチドをコードする核酸であって、該麻疹ウイルスHポリペプチドが、配列番号9に以下のアミノ酸置換:S189P、E235G、N238D、L249P、G302R、Y310C、Q311R、R377Q、M378K、D416N、N481Y、K488E、G491E、H495R、D505T、R533G、S546G、R547G、及びF552Vの19個を有する配列を含む、核酸、及び
麻疹ウイルスFポリペプチド以外のモルビリウイルスFポリペプチドをコードする核酸
を含む組換えウイルス。 - 前記ウイルスが麻疹ウイルスである、請求項1に記載のウイルス。
- 前記ウイルスがアデノウイルスである、請求項1~2のいずれか1項に記載のウイルス。
- 前記コードされる麻疹ウイルスHポリペプチド及び前記モルビリウイルスFポリペプチドが前記組換えウイルスのエンベロープに組み込まれている、請求項1~3のいずれか1項に記載のウイルス。
- 前記モルビリウイルスFポリペプチドがイヌジステンパーウイルスFポリペプチドである、請求項1~4のいずれか1項に記載のウイルス。
- 前記ウイルスがCD46依存性細胞侵入を示す、請求項1~5のいずれか1項に記載のウイルス。
- 前記ウイルスが、野生型ウイルスと比較してNectin-4依存性細胞侵入の低下を示す、請求項6に記載のウイルス。
- 前記ウイルスが、麻疹ウイルスFポリペプチドを欠損する、前記麻疹ウイルスFポリペプチドをコードする核酸を欠損する、又は前記麻疹ウイルスFポリペプチド及び前記麻疹ウイルスFポリペプチドをコードする核酸の両方を欠損する、請求項1~7のいずれか1項に記載のウイルス。
- 前記ウイルスが、野生型麻疹ウイルスHポリペプチドを欠損する、前記野生型麻疹ウイルスHポリペプチドをコードする核酸を欠損する、又は前記野生型麻疹ウイルスHポリペプチド及び前記野生型麻疹ウイルスHポリペプチドをコードする核酸の両方を欠損する、請求項1~8のいずれか1項に記載のウイルス。
- 請求項1~9のいずれか1項に記載のウイルスを含む、哺乳動物において生存腫瘍細胞の数を低減するための組成物。
- 前記哺乳動物がヒトである、請求項10に記載の組成物。
- 請求項1~9のいずれか1項に記載のウイルスを含む、哺乳動物において麻疹ウイルスに対する免疫反応を刺激するための組成物。
- 麻疹ウイルスHポリペプチドをコードする核酸であって、該麻疹ウイルスHポリペプチドが、配列番号9に以下のアミノ酸置換:S189P、E235G、N238D、L249P、G302R、Y310C、Q311R、R377Q、M378K、D416N、N481Y、K488E、G491E、H495R、D505T、R533G、S546G、R547G、及びF552Vの19個を有する配列を含む、核酸、及び
麻疹ウイルスFポリペプチド以外のモルビリウイルスFポリペプチドをコードする核酸
を含む核酸構築物。 - 前記核酸構築物がウイルスベクターである、請求項13に記載の核酸構築物。
