Description
本出願は、米国特許法第119条(e)の下で、2013年11月15日に提出された米国特許仮出願第61/904,786号および2013年2月13日に提出された第61/764,297号に対する優先権を主張するものであり、それらの出願のすべては参照により全体として本明細書に組み入れられる。
異所性組織ミネラル化は、慢性関節疾患および急性致死性新生児症候群を含めた、多くのヒト疾患と関連している。不要な組織石灰化を阻止するために、組織ミネラル化を促進する因子および組織ミネラル化を阻害する因子を緊密にバランスのとれた状態にしておかなければならない。異所性石灰化と関連した疾患のヒト同類および動物モデルについての遺伝子解析により、異所性組織ミネラル化の重要な調節因子として細胞外の無機ピロホスフェート(PPi)および無機ホスフェート(Pi)のバランスが同定された(Terkeltaub, 2001, Am. J. Phys. Cell Phys., 281:C1-C11(非特許文献1))。3つの細胞外酵素である組織非特異的アルカリホスファターゼ(TNAP)、進行性硬直症タンパク質(ANK)、およびエクトヌクレオチドピロホスファターゼ/ホスホジエステラーゼ-1(NPP1)の作用により、哺乳類におけるPiおよびPPiの濃度がそれぞれ1~3mMおよび2~3μMに緊密に制御される。PPiはバイオミネラル化の調節因子であり、非晶質リン酸カルシウムからの塩基性リン酸カルシウムの形成を阻害する。

別様に定義されていない限り、本明細書において用いられるすべての技術的および科学的な用語は、本発明が属する技術分野における当業者によって一般に理解されるものと同じ意味を有する。本明細書において記載されるものと同様のまたは同等の任意の方法および材料を、本発明の実践または試験において用いることができるが、好ましい方法および材料が記載される。
ヒトNPP4についての現在の高分解能構造決定により、NPP1との詳細な構造-機能比較研究が可能となった。これらの膜結合型細胞表面酵素は、細胞外プリン作動性シグナルの代謝ならびにヌクレオチド再取り込みに関与している。両方の外酵素は、ヌクレオチド含有基質の標的化を説明する、2個の結合した亜鉛イオンに近接した疎水性の狭い溝を保有し、そこでは、ヌクレオチド塩基が溝内で結合しかつ加水分解によりヌクレオチドモノホスフェートが産物の1つとして産出される。アデニンは両酵素にとって最も好ましい塩基タイプであり、かつ結合しているAMP分子産物とそれぞれとの共結晶構造により、結合部位のその領域におけるそれらの機能的類似性が強調される。両酵素は、Ap3Aを加水分解し得るが、ATPに対して驚くほど異なる応答を呈することが見出された。
本発明は、NPP1またはその変異体のレベルまたは活性を増加させるNPP1活性化因子組成物および方法を含む。様々な態様において、本発明のNPP1活性化因子組成物および治療の方法は、NPP1ポリペプチド、NPP1 mRNA、NPP1酵素活性、NPP1基質結合活性、その変異体、またはそれらの組み合わせの量を増加させる。様々な態様において、病的な石灰化または骨化の減少が治療成果を向上させ得る疾患および障害には、IIAC、OPLL、低リン酸血症性くる病、変形性関節症、および、アテローム性動脈硬化プラークの石灰化が含まれるが、それらに限定されるわけではない。

NPP1およびNPP4は、個別の膜内ドメインを有して細胞表面に局在する膜貫通型タンパク質である。例えば、NPP1はII型配向にあり、一方でNPP4はI型配向にある。対照的に、NPP2は、プレプロタンパク質として合成され、タンパク質分解プロセシングの後、フーリンによる細胞外ドメインの切断後に可溶性タンパク質として分泌される(Jansen et al., 2005, J. Cell Sci. 118:3081-3089)。バキュロウイルス内で可溶性組換えタンパク質としてNPP4を発現させるために、該タンパク質構築物から細胞質ドメインおよび膜貫通ドメインを除いた。対照的に、可溶性細胞外タンパク質としてNPP1を発現させるために、NPP1の膜貫通ドメインをNPP2の膜貫通ドメインと交換し、それにより、バキュロウイルス培養物の細胞外液中に可溶性組換えNPP1の蓄積がもたらされた。

タンパク質をニッケルアフィニティーカラムによって精製し、かつイミダゾールを用いた溶出後に、C末ヒスチジンタグを、タバコエッチ病ウイルス(TEV)プロテアーゼを用いて該タンパク質から切り除いた。ヒスチジンタグの切断後に、ニッケルカラムでの第2ラウンドの精製を実施して、C末ヒスチジンタグ、および第1ラウンドの精製中にニッケルカラムと非特異的に結び付いた夾雑タンパク質を除去した。可溶性NPP1は素通り中に溶出し、かつ回収され、かつスピン濃縮によって濃縮される。精製全体により、1Lの細胞培養物あたりおよそ2mgの純タンパク質がもたらされる(図3~4)。同様の一般的精製スキームが、NPPファミリーのいくつかのメンバーについての生化学的、生物物理学的、および生理学的研究のために記載されている(Albright et al., 2012, Blood 120:4432-4440;Saunders et al., 2011, J. Biol. Chem. 18:994-1004;Saunders et al., 2008, Mol. Cancer Ther. 7:3352-3362)。当技術分野およびタンパク質精製の科学において経験のある者に公知であるように、IgGのFcドメインを含有するタンパク質の精製も、プロテインAまたはプロテインGカラムへの結合によって達成され得る。
NPP1はグリコシル化タンパク質であり、昆虫細胞の糖類部分は、哺乳類宿主からの強い免疫原性反応を誘導すると予想される。動物を組換えNPP1で処理することによって免疫反応を誘導する可能性を低下させるために、哺乳類発現系を用いて、昆虫細胞グリコシル化パターンを哺乳類グリコシル化パターンで置き換えた。同一のNPP1構築物を哺乳類発現ベクター中にクローン化し、その後にHEK293細胞への安定なトランスフェクションが続くことによって、HEK293哺乳類腎臓細胞株内でタンパク質を発現させた。His抗体に対する免疫ブロットによって、安定なクローンを同定した。強く発現するクローンを拡張し、本明細書における他の箇所で記載されるバキュロウイルス精製スキームにおいて記載されるように、培養培地を回収しかつ処理した。タンパク質の全収率は、培養培地1Lあたりおよそ1.5mgであり、かつ試料の純度は95%超であった(図4)。
NPP1の細胞外可溶性ドメインが酵素的に活性のあることを証明するために、NPP1についての定常状態のミカエリス-メンテン酵素定数を、ATPを基質として用いて決定した。加えて、NPPファミリーメンバー間の基質特異性を例証するために、NPP1のATP加水分解を、NPP1と38%の配列同一性を有するタンパク質であるNPP4と直接比較した。NPP1がATPを切断したことを証明するために、酵素反応についてのHPLC解析を用い、かつ該反応の基質および産物の素性を、ATP、AMP、およびADP標準物質を用いることによって確認した(図5)。ATP基質はNPP1の存在下で経時的に分解し、酵素産物AMPの蓄積を有する(図5)。種々の濃度のATP基質を用いて、NPP1に関する初速度をATPの存在下で導き出し、かつデータを曲線に適合させて、酵素速度定数を導き出した(図5C)。NPP4とNPP1との間の相当な配列同一性にもかかわらず、NPP4はATP加水分解活性を有さず、一方でNPP1はATPをAMPおよびPPiにすぐに加水分解した(図5C)。生理的pHにおいて、NPP1の反応速度定数は、Km=144μMおよびkcat=7.8s-1である。
NPP1およびNPP4によるATP切断を測定するために、および産物同定のために用いられたHPLCプロトコールを、文献から改変した(Stocchi et al., 1985, Anal. Biochem. 146:118-124)。50mM Tris pH8.0、140mM NaCl、5mM KCl、1mM MgCl2、および1mM CaCl2緩衝液中に種々の濃度のATPを含有する反応を、0.2~1μM NPP1の添加によって開始させ、かつ等量の3Mギ酸または0.5N KOHによって種々の時点で抑え、かつ氷酢酸によってpH6に再酸性化した。抑えられた反応液を体系的に希釈し、HPLCシステム(Waters, Milford MA)にロードし、かつ基質および産物を254または259nmにおけるUV吸光度によってモニターした。基質および産物を、0%~10%(または20%)のメタノール勾配を有する15mM酢酸アンモニウムpH6.0溶液を用いて、5μm 250×4.6mm HPLCカラムのC18(Higgins Analytical, Mountain View, CA)で分離した。産物および基質を、それらの対応するピークの統合および式に従って定量化した。
式中、[基質]0は、初回基質濃度である。式で用いられるAMP、ADP、およびATPの吸光係数は、15.4mM-1cm-1であった。254nmでモニターした場合、反応と同じ日に流した基質および産物の標準物質を用いて、統合された産物/基質ピーク面積を濃度に変換した。
TTWマウスは、Institute of Cancer Research系統マウス(ICR, Japan)の同胞交配の中で発見され、多発性進行性異常石灰化を発症しかつ最終的に重度の奇形および関節強直で死亡する。これらのマウスは、特異的な脊柱および関節の異常により、OPLLおよび変形性関節症の両方についての確立された動物モデルとして働く。異所性組織ミネラル化表現型を説明する遺伝子欠損は、NPP1をおよそ350個のアミノ酸トランケートする、NPP1における位置568での未成熟終止コドン(グリシンから終止へ)にあることが突き止められている。C57BL/6J-Enpp1asj/Grsr、つまりNPP1asjマウスであるTTWマウスの変種は、バリンのアラニンへの点変異をもたらす、エクソン7、位置737におけるNPP1タンパク質コード領域に点変異を有し、かつ12週目に脊柱の関節炎、および7ヶ月齢までにこわばってかつ曲げることができなくなる多くの関節の変形性関節症を呈する。これらのマウスモデルは、変形性関節症を含めた異所性石灰化のヒト疾患を模倣する貴重な試薬であり、かつ本研究の仮説を検証するための適切な動物モデルである。
ttwマウスにおける血清PPi濃度を正常化するNPP1腹腔内(IP)投与レベルを確立する。初回薬物動態実験を実施して、インビボでPPiレベルに影響を及ぼすのに必要なNPP1の適切な経路および濃度を確立する。野生型(C57bl/6)動物に、0.03mg/kgで始まる濃度で、単回用量のNPP1を投与する(例えば、i.p.、i.v.など)。ヒト血清中の生理的NPP1濃度はかなり低いため(100~300ρM)、この初回濃度が選出される。水性緩衝液中2.15mg/mlという哺乳類NPP1の濃度は、生理的条件に極めて近い(50mM Tris pH8.0、150mM NaCl、0.8mM ZnCl2、0.4mM CaCl2、0.4mM MgCl2)。例えば、40gのマウスに、約0.5μlの濃縮ストックを注射する。血清PPi濃度に直接影響を及ぼす投与レベルに到達するように、必要に応じてNPP1投与量を調整する。
本明細書における他の箇所で記載されるように、TTWおよびC57BL/6J-Enpp1asj/GrsrJマウスを用いて、異所性石灰化のマウスモデルに対する組換えNPP1の効果を判定する。4組の繁殖ペアを確立して、それぞれの遺伝的コロニーを作り上げる。進行性の身体的障害の発生により、ホモ接合型雌は一腹の仔を維持し得る能力が損なわれるため、ヘテロ接合型雌をホモ接合型雄と繁殖させる。動物を3週齢目にイェール動物資源センター(Yale Animal Resource Center)によって遺伝子型決定し、かつ一度離乳させたら、動物をそれらの遺伝子型および年齢に従ってケージに分離する。厳密な記録管理を維持して、動物が実験前に正しく同定されることを確保する。6週齢目に、マウスを約6匹のマウスのコホートに分離し、かつ本明細書における他の箇所で記載されるように、血清PPi濃度を正常化することが観察された最低濃度で始まる、増加する濃度のNPP1で処理する。
NPP4を50mM Tris pH8.0、150mM NaCl、0.8mM ZnCl2、0.4mM CaCl2、0.4mM MgCl2中に交換し、かつA280を用いてタンパク質濃度を算出した。6mg/ml(0.14mM)のNPP4とウェル溶液(200mMクエン酸二アンモニウム、17.5%~19.5%(w/v)PEG 3350)とを1:1比で混合し、かつ600μlを上回るウェル溶液に2μlの液滴を密封チャンバー内で懸濁することによる懸滴(hanging drop)法によって最良の回折結晶を得た。タンパク質結晶は、典型的に4~6日間以内に出現し、翌週の間ゆっくりと増大し続け、最高500um×150um×50umの最終寸法に達した。結晶を、上記ウェル溶液と、0.6mM ZnCl2、5mMリガンド(存在している場合)、および最高20%~25%までの最終濃度のグリセロールの5%(v/v)増分とからなる一連の混合物中を迅速に通過させ、次いで直ちに液体窒素中で瞬間冷凍することによって、低温保護を達成した。NPP4のアポ結晶は得るのが難しく、浸漬によるリガンドとのNPP4の構造を得る努力を阻んだ。ATPまたは切断可能なATP類似体(Sigma M7510)との共結晶化は、AMP産物複合体をもたらし、結晶化の間のゆっくりとした加水分解を反映した。切断不能なATP類似体(Sigma M6517)との共結晶化の試みは、認識可能な結合がないことを示した。
マウスNPP1(rcsbコード4B56)をMOE(Molecular Operating Environment, Chemical Computing Group Inc., Montreal, Canada)にロードし、およびA鎖を欠失させ、かつB鎖には残基K169~E905を保持させた。アスパラギン結合型グリコシル化部位を切り抜き、かつメチル基でキャッピングした。水を欠失させ、および原子タイプを固定しかつProtonate 3Dでプロトン付加した。亜鉛イオンを、それらのリガンドの距離および形状に関して動かないようにした。金属電荷を+1でモデル化した。このタンパク質をよりよいものにする大きなチャネルに沿った拡張立体構造におけるホスフェート部分とともに、フェニルアラニン239およびチロシン322から形成された裂け目の間にアデニン核を置くことによって、ATPを手作業で位置付けした。該タンパク質をリガンドから4.5Åの距離に拘束し、かつリガンドの拡張Huckel処理とともにAMBER12(AMBER12:EHT)を用いて、システム全体に対して最小化を実施した。
グリコシル剥離およびキャッピング、ならびにATPの配置を含めて、NPP1と同様の様式でNPP4をモデル化した。アデニンの配置に関して、NPP4における対応する残基は、フェニルアラニン71およびチロシン154である。タンパク質のプロトン付加、拘束、および最小化を類似の形式で実施し、AMBER12:EHT力場処理を用いた。
ヒトNPP1およびNPP4についての定常状態の酵素活性を、吸光度(Ap3A基質)またはHPLC(ATP基質)のいずれかによって判定した。NPP4に対するヌクレオチドモノホスフェート(NMP)の親和性を、405nmにおける吸光度変化によってモニターされる、定常状態のpNP-TMP(p-ニトロフェニル5'チミジンモノホスフェート)切断速度の[NMP]依存度から概算した(Saunders et al., 2008, Mol. Cancer Ther. 7:3352-3362)。[NPP4]は5nMであり、かつ[pNP-TMP]は20mMであった。直角双曲線に対する、ヌクレオチド濃度依存的NPP4切断活性の最良適合から、IC50値(すなわち、最大活性の半分を呈するヌクレオチド濃度)を決定した。IC50は、混合した阻害に対する加重平均親和性を反映する(Saunders et al., 2008, Mol. Cancer Ther. 7:3352-3362)。
Albright et al., 2012, Blood 120:4432-4440に記載されているように、血小板の調製および血小板凝集能測定を実施した。
本明細書において記載されるように、病的な石灰化および/または骨化を伴う疾患および障害に対する有用な治療法であり得る、可溶性形態のヌクレオチドピロホスファターゼ/ホスホジエステラーゼ-1(NPP-1)および変異NPP-4を開発した。NPP1機能不全の疾患状態におけるこの酵素の生理的活性についての直接実証も本明細書において記載され、それによって、異所性石灰化の障害を選択することにおけるNPP1酵素補充療法の実用性が確立される。
高度に相同なNPPファミリーメンバーによって呈される基質判別の程度を判定するために、ヒトNPP4およびNPP1の、それらの推定上インビボ基質であるAp3AおよびATPに対する、定常状態の酵素速度をそれぞれ測定した(図5)。ヒトNPP4は、触媒ドメインにわたってNPP1と40%の配列同一性を共有し、かつマウスNPP1の構造は決定されており(Jansen et al., 2012, Structure 20:1948-1959)、原子レベルでの基質判別の構造的起源を同定する機会が提供されている。ヒトおよびマウスのNPP1は79%同一であり、マウスNPP1構造上へのヒト配列の配列マッピングにより、すべての配列差は、基質結合部位および活性部位の外側にあることが示されている。NPP1のミカエリス定数はNPP4のものよりも>30倍緊密であるものの、NPP4およびNPP1は、同等の最大代謝回転数(kcat約7~8s-1)でAp3Aを加水分解する。対照的に、NPP4がATPをAMPおよびPPiに加水分解する速度は、NPP1による加水分解と比べて無視できるほどである。
NPP4基質特異性の分子基盤を理解し、かつNPP1に対するその類似性および相違に関する詳細な洞察を得るために、X線結晶学を用いて、アポ形態およびAMP結合形態の両方のヒトNPP4の高分解能3次元構造を、それぞれ1.50Åおよび1.54Å分解能において決定した(表1および図6A)。次いで、これらを、2.70Å分解能におけるmNPP1-AMPを含めた、最近決定されたマウスNPP1複合体の構造と比較して、それぞれについての観察された基質特異性に関与する構造特質を規定した。NPP4およびNPP1の二金属性(bimetallo)触媒ドメインは、2個の結合した亜鉛イオンを含有し、かつ同様の全体的折り畳みを共有し、かつ保存された触媒メカニズムを採用して、基質上の同じ位置で加水分解し、ヌクレオチドモノホスフェート産物をもたらす。これらの酵素のいずれかによるAp3AまたはATPの加水分解により、AMP産物分子が産出される。したがって、hNPP4-AMPおよびmNPP1-AMP構造は、産物複合体である。NPP4の触媒ドメイン全体と、NPP1、NPP2、および細菌NPPのものとの重ね合わせにより、それぞれ1.54Å、1.43Å、および1.43Åというrmsd値がもたらされる。ヌクレオチド結合を好む、NPP4において観察される構造特質は、細菌酵素において大部分が保存されている。
NPP4は、末端に5'-ヌクレオチド基を有するホスホジエステル基質を標的とし、アデニン環に対して最大の選好性を呈示する。ヌクレオチド含有基質に対するこの特異性は、一方の側におけるTyr154の環面および他方におけるPhe71の先端からなる、およそ6.8Å幅の、タンパク質表面上にあるあらかじめ形成された疎水性の溝の結果である(図7A~7B)。その中で結合しているヌクレオチド塩基は、それぞれから約3.4Åにある、チロシン環との好ましいπ-πスタッキング相互作用、およびフェニルアラニンとのファンデルワールス(VDW)相互作用を経験する。溝の後壁は、ヌクレオチド塩基のちょうど手の届かないところにあり、該タンパク質とのいかなる直接的な水素結合相互作用も不可能となっており、数個の水介在性水素結合のみが、AMP環の縁部とNPP4との間に観察される。
NPP4のアポ構造は、AMP結合型構造と本質的に同一である(図7C)。空の疎水性の溝内にまたは触媒残基において変化は観察されないため、結合部位はあらかじめ形成されており、かつ基質結合があっても誘導性適合調整を受けないと考えられる。アポ構造は、キレート様相互作用でZn1において結合している、結晶化条件由来のクエン酸陰イオンを有する。Asn91は、著しく動く唯一の活性部位残基であり、クエン酸分子のための余地を与えるように旋回する。NPP4-AMP複合体において、Asn91は、Zn1付近で結合しているホスフェート基に水素結合を付与する。Asn91の旋回能により、反応の触媒中間体を介したホスフェート酸素との水素結合相互作用を維持することが可能となり得、またはリガンドの侵入もしくは退出を容易にする柔軟性が与えられ得る。
NPP4およびNPP1の溝領域内でどのようにAMPが結合しているかについての全体的類似性が図7Dに図解されている。NPP1は、NPP4のTyr154およびPhe71に対応する保存された残基を含有し、したがってまた5'-ヌクレオチド末端を有する基質を標的とする。AMPのホスフェート基は、両酵素においてZn1付近で結合する。AMP位置において観察される小さな相違は、実在しているのかどうか、またはNPP1-AMP構造についての大幅により低い分解能(NPP4-AMPに関する1.54Åに対して、2.70A)を単に反映しているのかどうかは未知である。
NPPはAPスーパーファミリーのメンバーであり、触媒が生じる活性部位の中心における重要な構造特質、残基、および構造様式の多くを共有する。図8は、NPPに関する一般的反応メカニズムを図示しており、というのはそれが、対応する結晶構造または各工程に対する結合シミュレーションモデルとともに、Ap3AのNPP4加水分解に適用されるからである。
NPP1はATPをAMPおよびPPiにすぐに切断し、一方でNPP4は、ごくゆっくりとそうするのみである。両酵素とも、アデニン環に対する選好性を有してヌクレオチド含有基質を標的とし、かつ同様の形式でAMPに結合するため(図7D)、このずれの手掛かりは結合ポケット内の他の場所にあるはずである。NPP1はATPをAMPおよびPPiに効率的に加水分解するため、ATPは、AMPに関して観察されるのと同じ配向でNPP1に結合する可能性が高い。したがって、2個のホスフェート基をNPP1-AMP共結晶配位に付加することによって、ATPをNPP1活性部位内にモデル化し(4B56)(図9A、9C、9E)、次いで、記載されるように複合体をエネルギー最小化に供した。NPP1の結合部位とNPP4との重ね合わせにより、このATP基質の4.5Å内のほとんどの側鎖間の良好な重なりが明らかとなっているが、著しい相違が末端γ-ホスフェート付近で生じている。NPP1において、タンパク質コアへのPhe516(マウスによる番号付け)の付与により、ATPのγ-ホスフェートに対してより多くの空間が創出され、それは、リジン立爪と称される3個のリジン残基(Lys237、Lys260、およびLys510)によって電荷安定化していると考えられる。これらのリジンのうちの2個(Lys260およびLys510)は、結合ポケットの上縁を裏打ちし、かつNPP1-AMP構造において観察されるように、γ-ホスフェートの非存在下で不規則なままである。それらのシミュレーションにより、新たな基質がNPP1結合部に入るとき、これらのリジンはγ-ホスフェートに静電的に引き入れられかつこの過程で規則正しくなることが図解されている。結合したATPのγ-ホスフェートは、Tyr433、Lys260、およびLys510から構成される誘導性適合蓋の下にあるように見え、そこでそれは3個のリジンによって電荷安定化されているはずである(図9A)。PPi産物が放出されると、Lys260およびLys510は、新たな基質がこの部位に引き入れられるまで再び不規則になるはずである。
NPP1によるAp3Aの加水分解は、NPP1が生理的条件下で血小板凝集において役割を果たし得るかどうかという疑問を起こさせる。この酵素はAp3A基質に対して活性があることを考慮すると、血管腔内のNPP1の相対的存在量が、凝血におけるNPP1の役割を決定し得る。NPP1は、脳毛細血管内皮上に存在しているが、他の場所の毛細血管上にはないことが報告されている。加えて、NPP1は、形質細胞、骨芽細胞、軟骨細胞、ならびに骨芽細胞および軟骨細胞から排出される基質小胞(MV)の膜表面に存在している。NPP1は、10~30ng/mL(つまり100~300pM)という非常に低い濃度で血管系内に可溶性タンパク質としても存在している。
ヒトNPP4(rcsbコード4LR2)をMOE(Molecular Operating Environment, Chemical Computing Group Inc., Montreal, Canada)にロードし、およびアスパラギン結合型グリコシル化部位を切り抜きかつメチル基でキャッピングした。水を欠失させ、および原子タイプを固定しかつProtonate 3Dでプロトン付加した。亜鉛イオンを、それらのリガンドの距離および形状に関して動かないようにした。金属電荷を+1でモデル化した。このタンパク質をよりよいものにする大きなチャネルに沿った拡張立体構造におけるホスフェート部分とともに、フェニルアラニン239およびチロシン322から形成された裂け目の間にアデニン核を置くことによって、Ap3Aを手作業で位置付けした。Ap3A分子の残りは、溶液中に遊離させたままにした。該タンパク質をリガンドから4.5Åの距離に拘束し、かつリガンドの拡張Huckel処理とともにAMBER12(AMBER12:EHT)を用いて、システム全体に対して最小化を実施した。ドッキングされたAp3A分子(図11)は、第2のアデニン結合部位の位置を明らかにした。この結合部位は、該タンパク質表面における狭い溝によって形成され、それは、ある特定の態様において、ポケットの表面を裏打ちする残基の変更によって排除され得る。
NPP4タンパク質におけるAp3Aの位置を直接決定するために、NPP4の不活性変異体をAp3Aとともに結晶化した(図12)。この構造を用いて、これらの酵素の血栓形成促進効果を低下させるように設計されたNPP4およびNPP1の変異を精密化し得る。

Claims (16)
- エクトヌクレオチドピロホスファターゼ/ホスホジエステラーゼ-1(ENPP1)活性が減少している対象において軟組織の病的石灰化の進行を減少させ、または予防するのに用いるための組成物であって、該組成物が、可溶性エクトヌクレオチドピロホスファターゼ/ホスホジエステラーゼ-1(ENPP1)ポリペプチドを含み、ここで、該可溶性ENPP1ポリペプチドがIgG Fcドメインを含み、かつ、該可溶性ENPP1ポリペプチドが4~20の連続したアスパラギン酸残基を欠如する、組成物。
- 前記可溶性ENPP1ポリペプチドが、哺乳類細胞内で発現されたポリペプチドの分泌産物である、請求項1記載の組成物。
- 前記哺乳類細胞内で発現されたポリペプチドが、ENPP1膜貫通ドメインの残基77~98を欠如し、かつ、シグナル配列を含む、請求項2記載の組成物。
