Description

SET結合タンパク質1(SETBP1)は、1500超のアミノ酸の大きなAT-フック転写因子をコードし、がんタンパク質(oncoprotein)SETの相互作用因子を同定するためのスクリーニングにおいて最初にクローニングされたが、心臓、脳、腎臓、肝臓、肺、膵臓、脾臓、および骨格筋を含む広く種々の組織において発現される(Minakuchi et al., Eur. J. Biochem. 268(5):1340-1351 (2001))。過剰発現またはミスセンス変異を経てのその異常な活性化は、近年、原発性急性骨髄性白血病(AML)(Cristobal et al., Blood 115(3):615-625 (2010))、慢性骨髄性白血病急性転化(CML-BC)(Oakley et al., Blood 119(25):6099-6108 (2012))、非定型慢性骨髄性白血病(aCML)(Piazza et al., Nat. Genet. 45(1):18-24 (2013))、慢性骨髄単球性白血病(CMML)、二次性AML(sAML)(Makashima et al., Nat. Genet. 45(8):942-946 (2013))、若年性骨髄単球性白血病(JMML)(Sakaguchi et al., Nat. Genet. 45(8):937-941 (2013))、および骨髄異形成症候群(MDS)(Damm et al., Leukemia 27(6):1401-1403 (2013); Fernandez-Mercado et al., Br. J. Haematol. 163(2):235-239 (2013));およびHou et al., Am. J. Hematol. 89(2):181-186 (2014))を含む種々のヒト骨髄系新生物において頻繁に起こることが見出されている。
核外移出におけるその旧来の役割(Fukuda et al., Nature 390(6657):308-311 (1997); Ossareh-Nazari et al., Science 278(5335):141-144 (1997); Hutten et al., Trends Cell. Biol. 17(4):193-201 (2007);およびTurner et al., Curr. Med. Chem. 15(26):2648-2655 (2008))とは対照的に、Xpo1(CRM1としても公知)が、Hoxa9、Hoxa10、およびMybを含む重要な標的の転写を活性化するにあたって、Setbp1およびSetbp1のミスセンス変異体の本質的な補因子であり得ることは、本出願の発明者らによって観察された。Setbp1/Setbp1ミスセンス変異体とXpo1との間の相互作用を担うモチーフも同定された。このモチーフは、SETBP1活性化を有する新生物を処置するための治療標的を表す。さらに、このモチーフを含むSetbp1ペプチドフラグメント(本明細書で「ポリペプチド25-20」といわれる)と相互作用し得る低分子に関する化合物スクリーニングを行うことによって、Setbp1またはそのミスセンス変異体によって不死化された骨髄系前駆細胞の増殖を阻害し、これら細胞においてSetbp1標的遺伝子転写をも低減し得る化合物が、同定された。SETBP1活性化を有する骨髄系新生物および他のがん(例えば、固形腫瘍のがん)を処置するためのこれら化合物の同定および治療上の使用が、本明細書で記載される。
本発明の一局面は、被験体において骨髄系新生物または固形腫瘍を処置するための方法であって、上記方法は、必要性のある被験体に、治療有効量の式(1)の化合物:

定義
用語「薬学的に受容可能なキャリア」とは、本明細書で使用される場合、水、エタノール、ポリオール(例えば、グリセロール、プロピレングリコール、および液体ポリエチレングリコールなど)、これらの適切な混合物、ならびに植物性油、コーティング、等張化剤および吸収遅延剤、リポソーム、市販の洗浄剤(cleanser)などが挙げられるが、これらに限定されない、任意のおよび全ての溶媒、または分散媒を含む。補助的な生体活性成分がまた、このようなキャリアの中に組み込まれ得る。
Setbp1およびXPO1は、HEK293T細胞において互いと物理的に会合することが観察された。Setbp1が、がん遺伝子Hoxa9およびHoxa10を活性化し得る転写調節因子であり、このような活性化が、骨髄系前駆細胞のSetbp1活性化誘導性形質転換におそらく必須であることは、以前に示された(Oakley et al., Blood 119(25):6099-6108 (2012))。SETBP1媒介性転写調節に決定的に関与する潜在的な補因子を同定しようとする試みにおいて、本発明者らは、XPO1が、3×FLAGタグ化野生型Setbp1を発現させるDNA構築物でトランスフェクトしたHEK293T細胞の核抽出物から、FLAGタグ特異的M2抗体を使用して効率的に免疫沈降され得ることを見出した。同様に、XPO1はまた、ヒト白血病患者において同定された再発性ミスセンス変異(D868N)を有するSetbp1変異体であるSetbp1(D/N)によってプルダウンされ得る(図1A)。さらに、両方のFLAGタグ化Setbp1タンパク質はまた、これらの核抽出物からXPO1特異的抗体で効率的に免疫沈降され得る(図1A)。これらの結果は、XPO1が、複合体形成においてSetbp1およびSetbp1(D/N)の両方と会合し得ることを示唆する。
XPO1の正常な機能を阻害することなく、SETBP1とXPO1との間の相互作用を遮断し得る低分子を同定するアプローチとして、NES2の周りのSetbp1領域と相互作用し得る化合物をスクリーニングした。共免疫沈降によって、Setbp1タンパク質のa.a.805~a.a.1020にまたがりかつNES2を含む、Setbp1タンパク質の細菌合成したフラグメント(ポリペプチド25-20)(図18)が、全長XPO1との安定な相互作用を形成し得る(図19)ことが発見された。ポリペプチド25-20と相互作用し得る低分子を同定するために、化合物ライブラリーを、Georgetown UniversityのBiacore Molecular Interaction Shared Resourceとの契約を通じて、表面プラズモン共鳴(SPR)ベースのBiacoreシステムを使用してスクリーニングした。このスクリーニングのために、Hisタグ化ポリペプチド25-20を細菌において合成し、精製し、抗Hisタグ抗体を使用してCM5チップ表面に固定化し、ポリペプチド25-20に対する化合物の親和性を、化合物をチップ表面を通して流しながら測定した。コントロールとして、これらの化合物とHisタグ化ヒト血清アルブミン(HSA)との相互作用もまた同様に評価した。2つの化合物ライブラリーを最初にスクリーニングした(1,200を超える大部分が承認された薬物を含むPrestwickライブラリーおよび2,500を超える化合物を有するNCI Developmental Therapeutics Program(DTP)ライブラリー)。合計で77の化合物を、このスクリーニングにおいてHSAに対するより少なくとも50倍高い親和性でポリペプチド25-20に結合すると見出した。いくつかの化合物は、1μM濃度もしくはこれより低い濃度でSetbp1によって不死化したマウス骨髄系前駆細胞の増殖を有意に阻害することが見出された。
化合物(1AA)のインビトロおよびインビボ試験. 骨髄系新生物に対する化合物1AAの活性を、20% ウマ血清+マウスScf(50ng/ml)およびIl-3(10ng/ml)を有するIMDMメチルセルロース培地上でSetbp1/Setbp1(D/N)不死化マウス骨髄系前駆細胞によるコロニー形成に対するその阻害効果を評価することによって、インビトロで先ず決定した。化合物1AAのインビボ活性を、Setbp1(D/N)誘導性白血病細胞を移植したマウスの脾臓サイズを低減し、生存を延ばすその能力を評価することによって評価した。白血病マウスの作製のために、Setbp1(D/N)誘導性白血病を有する原発性レシピエントマウスの5×105個の脾細胞を、各々致死量未満で照射した(137Cs源から550ラド)B6-Ly5.2雌性マウスの尾静脈へと注射した。移植後10日目から開始して、上記レシピエントマウスを、化合物(1AA)(33mg/kg、生理食塩水中に溶解)または生理食塩水で、5回の腹腔内注射によって処置した。固形腫瘍に対する化合物1AAの活性を、培養において種々のヒト固形腫瘍細胞株に対するその増殖阻害性効果を評価することによって決定する。具体的には、これらの細胞株の液体培養物の細胞ATPレベルを、系列希釈した濃度の化合物1AAでの処理後72時間で測定し、処理されていない細胞と比較する。
式1の代表的化合物としての化合物(1AA)は、Setbp1不死化細胞(図10)およびSetbp1(D/N)不死化細胞(図11)において、コロニー形成を阻害し、分化/細胞傷害性を誘導することが観察された。さらに、化合物(1AA)は、それぞれ、Setbp1不死化細胞およびSetbp1(D/N)不死化細胞において処置後12時間で、Hoxa9、Hoxa10およびMybのSetbp1(図12)およびSetbp1(D/N)(図13)標的転写レベルを低減した。
