Description
本発明は、一般に、細胞培養を使用する特異性アッセイ、特に、異なるフォーマットで新規抗原結合部分を試験するためのキメラ抗原受容体(CAR)発現レポーターT(CAR-T)細胞アッセイに関する。さらに、本発明は、抗原結合分子の認識ドメインに特異的に結合することができる標的抗原結合部分を含む操作されたキメラ抗原受容体(CAR)でトランスフェクト/形質導入されたCAR-T細胞の使用に関する。本発明はまた、候補抗原結合部分及び/又は操作されたCARを発現する核酸分子及びベクターの特異性試験のための方法及びキットに関する。
本発明は、一般に、特に薬物開発プロセスにおいて、新規抗原結合部分を選択する方法に関し、例えば腫瘍細胞上の標的抗原への結合の評価と、抗体-標的結合に応答したT細胞の活性化とを組み合わせる。提供されるのは、以下の工程:
a)抗原結合ドメイン及び認識ドメインを含む抗原結合分子を提供する工程、ここで、抗原結合ドメインは抗原結合部分を含み、抗原結合部分は標的抗原に特異的である;
b)抗原結合分子を、表面上に標的抗原を含む標的細胞、特に標的細胞ががん細胞である標的細胞と接触させる工程;
c)抗原結合分子を、キメラ抗原受容体(CAR)発現レポーターT(CAR-T)細胞と接触させる工程、ここで、レポーターCAR-T細胞は、
i.認識ドメインに特異的に結合することができるCARであって、抗原結合部分が応答エレメントに作動可能に連結されているCAR;
ii.応答エレメントの制御下にあるレポーター遺伝子
を含む;及び
d)抗原結合部分の特異性を確立するためにレポーター遺伝子の発現を決定することによってT細胞活性化を決定する工程
を含む、抗原結合部分の特異性を評価するための方法である。
e)レポーター遺伝子の発現を参照と比較する工程
を含む。
f)標的細胞の非存在下でのレポーター遺伝子の発現と比較して、標的細胞の存在下でのレポーター遺伝子の発現が所定の閾値よりも高い場合、新規抗原結合部分を選択する工程
を含む。
定義
用語は、以下で特に定義されない限り、当該技術分野で一般的に使用されるように本明細書で使用される。
「活性化Fc受容体」は、抗体のFcドメインの結合に続いて、エフェクター機能を実行するように受容体保有細胞を刺激するシグナル伝達現象を生じさせるFc受容体である。ヒト活性化Fc受容体には、FcγRIIIa(CD16a)、FcγRI(CD64)、FcγRIIa(CD32)、及びFcαRI(CD89)が含まれる。
用語「可変領域」又は「可変ドメイン」は、抗原との結合に関与する免疫グロブリン重鎖又は軽鎖のドメインを指す。天然抗体の重鎖及び軽鎖の可変ドメイン(それぞれ、VH及びVL)は、一般的に類似の構造を有し、各ドメインは4つの保存されたフレームワーク領域(FR)及び3つの超可変領域(HVR)を含む。例えば、Kindt et al., Kuby Immunology, 6th ed., W.H. Freeman and Co, page 91 (2007)を参照のこと。抗原結合特異性を付与するには通常、単一のVH又はVLドメインで十分である。
2表Aにおいて使用されている小文字の「b」を含む「AbM」は、Oxford Molecularの「AbM」抗体モデル化ソフトウエアによって定義されるCDRを指す。
100×分率X/Y
ここで、Xは、配列アラインメントプログラムALIGN-2により、そのプログラムのAとBとの配列比較において、完全一致を記録したアミノ酸残基の数であり、YはBの全アミノ酸残基数である。アミノ酸配列Aの長さがアミノ酸配列Bの長さと異なる場合、AのBに対する%アミノ酸配列同一性は、BのAに対する%アミノ酸配列同一性とは異なることは理解されるであろう。特に断らない限り、本明細書で使用される全ての%アミノ酸配列同一性値は、ALIGN-2コンピュータプログラムを使用し、直前の段落で説明したように得られる。

例えば、本明細書に記載のCARにおいて、認識ドメインに特異的に結合することができる少なくとも1つの抗原結合部分を含む細胞外ドメインとアンカー膜貫通ドメインとの間のリンカーは、配列番号17に示されるアミノ酸及びアミノ酸配列を含むか、又はそれらからなり得る。したがって、アンカー膜貫通ドメイン、共刺激シグナル伝達ドメイン及び/又は刺激ドメインは、ペプチドリンカーによって、あるいはドメインの直接融合によって、互いに連結され得る。

DNA配列は、二本鎖配列決定により決定された。
所望の遺伝子セグメントは、適切な鋳型を用いたPCRによって生成されるか、又は自動遺伝子合成による合成オリゴヌクレオチド及びPCR産物からGeneart AG(Regensburg,Germanyによって合成された。特異的制限エンドヌクレアーゼ切断部位に挟まれた遺伝子セグメントを、標準的なクローニング/配列決定ベクターにクローニングした。形質転換した細菌からプラスミドDNAを精製し、UV分光法により濃度を決定した。サブクローニングされた遺伝子断片のDNA配列を、DNA配列決定によって確認した。遺伝子セグメントは、それぞれの発現ベクター中へのサブクローニングを可能にするために適切な制限部位を用いて設計された。全ての構築物は、真核細胞内の分泌のためにタンパク質を標的とするリーダーペプチドをコードする5’末端DNA配列を含むように設計された。
標準プロトコールを参照して、濾過した細胞培養上清からタンパク質を精製した。簡潔には、抗体を、プロテインA Sepharoseカラム(GE healthcare)に適用し、PBSで洗浄した。抗体の溶出は、pH2.8で達成され、続いて試料を直ちに中和した。凝集したタンパク質を、PBSにおける、又は20mMのヒスチジン、150mMのNaCl、pH6.0における、サイズ排除クロマトグラフィー(Superdex 200、GE Healthcare)によって、単量体抗体から分離した。単量体抗体画分をプールし、(必要に応じて)例えば、MILLIPORE Amiconultra(30 MWCO)遠心濃縮機を使用して濃縮し、凍結して-20℃又は-80℃で保存した。試料の一部は、例えば、SDS-PAGE及びサイズ排除クロマトグラフィー(SEC)による後続のタンパク質分析及び分析特性評価のために提供された。
NuPAGE(登録商標)Pre-Castゲルシステム(Invitrogen)を製造者の指示に従って使用した。特に、10%又は4~12%のNuPAGE(登録商標)Novex(登録商標)ビス-TRIS Pre-Castゲル(pH6.4)及びNuPAGE(登録商標)MES(還元ゲル、NuPAGE(登録商標)抗酸化剤ランニングバッファー添加剤を含む)又はMOPS(非還元ゲル)ランニングバッファーを使用した。
抗体の凝集及びオリゴマー状態の測定のためのサイズ排除クロマトグラフィー(SEC)は、HPLCクロマトグラフィーによって行った。簡潔には、プロテインA精製抗体を、Agilent HPLC 1100システムで、300mMのNaCl、50mMのKH2PO4/K2HPO4、pH7.5の中のTosoh TSKgel G3000SWカラムに適用するか、又はDionex HPLCシステムで、2xPBSの中のSuperdex 200カラム(GE Healthcare)に適用した。溶出したタンパク質を、UV吸光度及びピーク面積の積分によって定量した。BioRadゲル濾過標準151-1901を標準とした。
国際特許出願公開番号WO2012/130831A1に記載の方法に従って、Fcガンマ受容体への結合を抑止するために、定常領域にPro329Gly、Leu234Ala及びLeu235Ala変異が導入された。したがって、I253A、H310A及びH435A(「AAA」)変異が、FcRnへの結合を抑止するために定常領域に導入された。それぞれの抗体は、ポリエチレンイミンを使用してHEK293-EBNA細胞に哺乳動物発現ベクターをコトランスフェクトすることにより産生された。細胞に、重鎖用及び軽鎖用の対応する発現ベクターを1:1の比でトランスフェクトした。
レンチウイルスベクターを作製するために、抗原結合受容体の正しい組み立てのためのそれぞれのDNA配列を、恒常的に活性なヒトサイトメガロウイルス前初期プロモーター(CMV)の下でレンチウイルスポリヌクレオチドベクターにインフレームでクローニングした。レトロウイルスベクターは、ウッドチャック肝炎ウイルス転写後調節エレメント(WPRE)、セントラルポリプリントラクト(cPPT)エレメント、pUC複製起点、及び細菌における増殖と選択を促進する抗生物質耐性をコードする遺伝子を含んでいた。
本明細書に記載されるのは、標的細胞として、CD20発現SUDHDL4腫瘍細胞を使用し、レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞の選別されたプール(図6A)又は抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞のプール(図6B)を使用するJurkat NFATレポーターCAR-T細胞アッセイである。P329G LALA変異を有するGA101 IgGをIgGとして使用し、これは、一方では腫瘍抗原を認識し、他方では形質導入されたJurkat NFATレポーターCAR-T細胞によって認識される。陽性対照として、96ウェルプレート(Cellstar Greiner-bio-one、カタログ番号655185)を、リン酸緩衝生理食塩水(PBS)中で10μg/mlのCD3抗体(Biolegend(登録商標)から)で、4℃で一晩又は37℃で少なくとも1時間コーティングした。CD3でコーティングされたウェルをPBSで2回洗浄し、最終洗浄工程の後、PBSを完全に除去した。レポーター細胞又はJurkat NFAT野生型細胞を計数し、Cedex HiResを使用してそれらの生存率をチェックした。細胞数を1×106個の生細胞/mlに調整した。したがって、細胞懸濁液の適切なアリコートを室温(RT)で210gで5分間ペレット化し、新鮮なRPMI-160+10%のFCS+1%のGlutamax(増殖培地)に再懸濁した。目的の抗原を発現する標的細胞を計数し、その生存率についてもチェックした。細胞数を、レポーター細胞について記載されているように、増殖培地中で1×106生存細胞/mlに調整した。標的細胞及びレポーター細胞を5:1又は1:1のE:T比(合計でウェルあたり110.000個の細胞)で、96ウェルの懸濁培養プレート(Greiner-bio one)に3重に播種した。次の工程として、目的の抗原を標的とする、P329G LALA変異を有するGA101の段階希釈物を、2mlのディープウェルプレート(Axygen(登録商標))を使用して増殖培地中で調製した。ウェルあたり200ulの最終容量で1μg/mlから0.0001μg/mlの範囲の最終濃度を得るために、異なる希釈の50μlのアリコートをそれぞれのウェルにピペットで移した。96ウェルプレートを190g及びRTで2分間遠心分離した。Parafilm(登録商標)で密封し、プレートを37℃、5%CO2の湿度雰囲気中でインキュベートした。20時間のインキュベーション後、各ウェルの内容物を、マルチチャンネルピペットを使用して10回上下にピペッティングすることにより混合した。100μlの細胞懸濁液を新しい白色平坦透明底の96ウェルプレート(Greiner-bio-one)に移し、100ulのONE-GloTMLuciferase Assay(Promega)を加えた。暗所で、ロータリーシェーカー上で、300rpm、室温で15分間インキュベートした後、Tecan(登録商標)Spark10Mプレートリーダーを使用して、検出時間を1秒/ウェルとして発光を測定した。

本明細書に記載されるのは、標的細胞として付着性CEA発現MKN45腫瘍細胞を使用するJurkat NFATレポーターCAR-T細胞レポーターである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSD発現Jukat NFAT T細胞(図10C)又は抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞(図10A)の選別されたプールを使用した。両方ともP329G LALA変異を有するCH1A1A 98 99又はCEA hMN14 IgGのいずれかを使用した。さらに、アイソタイプ対照として、P329G LALA変異を有するIgG DP47/vk3を含めた。
本明細書に記載されるのは、標的細胞として付着性TNC発現CT26TNC c119腫瘍細胞を使用する Jurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞(図11C)又は抗P329G-ds-scFv-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞(図11A)の選別されたプールを使用した。P329G LALA変異を有するTNCA2B10をIgGとして使用した。さらに、アイソタイプ対照として、P329G LALA変異を有するIgG DP47/vk3を含めた。
本明細書に記載されるのは、標的細胞として付着性TNC発現CT26TNC c119腫瘍細胞を使用するJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、Jurkat NFATレポーターCAR-T細胞を発現する抗P329G-Fab-CD28ATD-CD28CSD-CD3zSSDの選別されたプールを使用した(図12A)。P329G LALA変異を有するTNCA2B10をIgGとして使用した。さらに、アイソタイプ対照として、P329G LALA変異を有するIgG DP47/vk3を含めた。
本明細書に記載されるのは、RMFペプチド又はVLDペプチドでパルスされたT2細胞を用いたフローサイトメトリーによる、HLA-A2/WT1ペプチドバインダー5E11(配列番号102及び103)並びに33H09(配列番号100及び101)の特異性の評価である。HLA-A2/WT1ペプチド結合抗体とのインキュベーションの前に、T2細胞にそれぞれのペプチドを10-5Mで37℃で2時間パルスするか、又はパルスせずに放置した。100000細胞の細胞アリコートへのそれぞれのIgGの結合を、それぞれ、問題の抗体の異なる濃度で氷上で1時間許容し、続いてPBSで2回洗浄し、Fortessaアナライザー(BD Biosciences)のフローサイトメトリーにおいて90nMの濃度で、抗huFc検出(Jackson ImmunoResearchからの抗ヒトF(ab)2_AF647)を介して評価した。バインダー5E11と33H09は両方とも、RMFペプチドパルスでは明確な濃度依存性結合シグナルを与えるが、VLDペプチドパルスT2細胞では示さない(図13A及びB)。このフローサイトメトリーに基づく評価によれば、両抗体候補はRMFペプチドパルスT2細胞に特異的に結合するが、VLDペプチドパルスT2細胞には結合しないようである。
本明細書に記載されるのは、HLA-A2/WT1ペプチドバインダー33F05(配列番号96及び97)、11D06(配列番号98及び99)、33H09(配列番号100及び101)及び5E11(配列番号102及び103)の特異性を評価するためにペプチドパルスT2細胞を標的細胞として使用するJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞の選別されたプールを使用した。P329G LALA変異を有するHLA-A2/WT1ペプチドバインダーをIgGとして使用した。HLA-A2/WT1ペプチド結合抗体及びレポーター細胞とのインキュベーションの前に、T2細胞にそれぞれのペプチドを10-5Mで37℃で2時間パルスするか、又はパルスせずに放置した。標的細胞及びレポーター細胞を5:1のE:T比(1ウェルあたり2000個の標的細胞あたり10000個のエフェクター細胞)で、384ウェルの白色平坦透明底プレート(Greiner-bio-one)に3重に播種した。次の段階として、当該IgGの段階希釈物を増殖培地中で調製した。レポーター細胞、T2細胞及びIgGを37℃で16時間インキュベートし、続いて1ウェルあたり6μlのONE-Glo(商標)ルシフェラーゼ基質(Promega)を添加し、TECANインフィニットM1000Proプレートリーダーを使用して発光を直接測定した。
本明細書に記載されるのは、HLA-A2/WT1ペプチドバインダー11D06/D43の変異体の特異性を評価するために、標的細胞としてペプチドパルスT2細胞を使用するJurkat NFATレポーターCAR-T細胞アッセイである。レポーター細胞として、抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFATレポーターCAR-T細胞の選別されたプールを使用した。P329G LALA変異を有するHLA-A2/WT1ペプチドバインダーをIgGとして使用した。HLA-A2/WT1ペプチド結合抗体及びレポーター細胞とのインキュベーションの前に、T2細胞にRMFペプチドを10-5Mで37℃で2時間パルスするか、又はパルスせずに放置した。標的細胞及びレポーター細胞を5:1のE:T比(1ウェルあたり2000個の標的細胞あたり10000個のエフェクター細胞)で、384ウェルの白色平坦透明底プレート(Greiner-bio-one)に3重に播種した。次の段階として、問題のIgGの段階希釈物を増殖培地で調製した。レポーター細胞、T2細胞及びIgGを37℃で16時間インキュベートし、続いて1ウェルあたり6μlのONE-Glo(商標)ルシフェラーゼ基質(Promega)を添加し、TECANインフィニットM1000Proプレートリーダーを使用して発光を直接測定した。
本明細書に記載されるのは、レポーター細胞として抗P329G-ds-Fab-CD28ATD-CD28CSD-CD3zSSD発現Jurkat NFAT CAR-T細胞の選別されたプールを用いたJurkat NFAT レポーターCAR-T細胞アッセイである。レポーター細胞は、P329G LALA変異を有するIgG形式のHLA-A2/WT1ペプチドバインダーに結合し、次いで、試験されたHLA-A2/WT1ペプチド(それぞれ、RMF又はVLD)を異なる程度で認識する。
Claims (16)
- 以下の工程;
a)抗原結合ドメイン及び認識ドメインを含む抗原結合分子を提供する工程、ここで、抗原結合ドメインは抗原結合部分を含み、抗原結合部分は標的抗原に特異的である;
b)抗原結合分子を、表面上に標的抗原を含む標的細胞と接触させる工程;
c)抗原結合分子を、キメラ抗原受容体(CAR)発現レポーターT(CAR-T)細胞と接触させる工程、ここで、レポーターCAR-T細胞は、
i.認識ドメインに特異的に結合することができるCARであって、ここでCARが応答エレメントに作動可能に連結されているCAR;
ii.応答エレメントの制御下にあるレポーター遺伝子
を含む;及び
d)抗原結合部分の特異性を確立するためにレポーター遺伝子の発現を決定することによってT細胞活性化を決定する工程
を含む、抗原結合部分の特異性を評価するための方法。 - 標的細胞ががん細胞である、請求項1に記載の方法。
- 認識ドメインが免疫グロブリンドメインである、請求項1又は2に記載の方法。
- 認識ドメインがFcドメインである、請求項1から3のいずれか一項に記載の方法。
- Fcドメインが変異Fcドメインであり、ここで、変異Fcドメインが、非変異親Fcドメインと比較して少なくとも1つのアミノ酸置換を含み、ここで、CARは、変異Fcドメインに特異的に結合することができるが、非変異親Fcドメインに特異的に結合することができない、請求項4に記載の方法。
- 変異Fcドメインが、EU番号付けによるL234、L235、I253、H310、P331、P329及びH435からなる群から選択される位置に少なくとも1つのアミノ酸変異を含み、特に、アミノ酸変異がL234A、L235A、I253A、N297A、H310A、P329G及び/又はH435Aである、請求項5に記載の方法。
- 変異Fcドメインが、EU番号付けによるアミノ酸変異P329Gを含む、請求項5又は6のいずれか一項に記載の方法。
- 抗原結合部分が、Fab断片、特にファージディスプレイライブラリースクリーニングに由来するFab断片である、請求項1から7のいずれか一項に記載の方法。
- 応答エレメントの活性化が、レポーター遺伝子の発現をもたらす、請求項1から8のいずれか一項に記載の方法。
- レポーター遺伝子が、緑色蛍光タンパク質(GFP)又はルシフェラーゼをコードしている、請求項1から9のいずれか一項に記載の方法。
- 標的抗原が、CD20、CEA、HER2、TYRP、EGFR、MCSP、STEAP1、WT1、及びFolR1、又はそれらの断片からなる群から選択される、請求項1から10のいずれか一項に記載の方法。
- 標的抗原が、ヒト主要組織適合性遺伝子複合体(MHC)の分子に結合したペプチドである、請求項1から11のいずれか一項に記載の方法。

- 抗原結合部分が、T細胞二重特異性(TCB)抗体に含まれ、標的細胞の存在下でのレポーター遺伝子の高レベルの発現及び標的細胞の非存在下でのレポーター遺伝子の低レベルの発現が、TCB抗体の高特異性を示す、請求項1から14のいずれか一項に記載の方法。
- TCB抗体を生成するための方法であって、TCB抗体が、標的抗原に特異的な第1の抗原結合部分と、T細胞活性化受容体に特異的に結合することができる第2の抗原結合部分とを含み、ここで、第1の抗原結合部分が、請求項1から15のいずれか一項に記載の方法に従って選択される、方法。
