Abstract
Description

i.タグを含む認識ドメインに特異的に結合することができるCARであって、応答エレメントに作用的に結合されている、CAR;
ii.応答エレメントの制御下のレポーター遺伝子
を含む、抗原結合分子をCAR−T細胞と接触させる工程;及び
d)レポーター遺伝子の発現を測定することによりT細胞活性化を決定し、標的抗原結合部分の特異性を確立する工程

「親和性」は、分子(例えば、抗体又はCAR)の単一結合部位とその結合パートナー(例えばリガンド)との間の非共有結合的相互作用の総和の強度を指す。本明細書で使用する場合、特に断らない限り、「結合親和性」は、結合対(例えば、抗原結合部分と抗原及び/又は受容体とそのリガンド)のメンバー間の1:1の相互作用を反映する固有の結合親和性を指す。分子Xの、そのパートナーYに対する親和性は、通常、解離定数(KD)によって表され、この解離定数(KD)は、解離速度定数と会合速度定数(それぞれ、koff及びkon)の比である。したがって、速度定数の比が同じである限り、同等の親和性が異なる速度定数を含むことがある。親和性は、ここに記載のものを含む当技術分野で既知の確立された方法により測定することができる。親和性を測定するための好ましい方法は、表面プラズモン共鳴(SPR)であり、測定に好ましい温度は25℃である。

Sambrook et al, Molecular cloning: A laboratory manual; Cold Spring Harbor Laboratory Press, Cold Spring Harbor, New York, 1989に記載されているように、標準的な方法を使用してDNAを操作した。分子生物学的試薬を、製造元の指示に従って使用した。ヒト免疫グロブリンの軽鎖及び重鎖のヌクレオチド配列に関する一般情報は以下に与えられている:Kabat, E.A. et al., (1991) Sequences of Proteins of Immunological Interest, Fifth Ed., NIH Publication No 91-3242。
DNA配列は、二本鎖シーケンシングにより決定された。
所望の遺伝子セグメントを、適切なテンプレートを用いてPCRによって生成するか、又は自動化された遺伝子合成による合成オリゴヌクレオチド及びPCR生成物からGeneart AG(Regensburg, Germany)により合成した。単一の制限エンドヌクレアーゼ切断部位に隣接する遺伝子セグメントを、標準的なクローニング/シーケンシングベクター中でクローニングした。形質転換した細菌からプラスミドDNAを精製し、UV分光法により濃度を決定した。サブクローニングされた遺伝子断片のDNA配列を、DNA配列決定によって確認した。遺伝子セグメントは、それぞれの発現ベクター中へのサブクローニングを可能にする適切な制限部位を用いて設計された。全てのコンストラクトは、真核細胞における分泌のためのタンパク質を標的とするリーダーペプチドをコードする、5’末端DNA配列を含むように設計された。
標準的なプロトコールを参照して、濾過された細胞培養上清からタンパク質を精製した。簡潔には、抗体を、プロテインA Sepharoseカラム(GE healthcare)に適用し、PBSで洗浄した。抗体の溶出はpH2.8で達成され、その直後に試料を中和した。凝集したタンパク質を、サイズ排除クロマトグラフィー(Superdex 200、GE Healthcare)により、PBS中又は20mMのヒスチジン(150mMのNaCl、pH6.0)中の単量体抗体から分離させた。単量体抗体画分をプールし、例えばMILLIPORE Amicon Ultra (30 MWCO)遠心濃縮機を使用して濃縮し、−20℃又は−80℃で冷凍し、保存した。試料の一部が、その後のタンパク質分析、及び例えばSDS−PAGE及びサイズ排除クロマトグラフィー(SEC)による分析的特徴付けのために提供された。
NuPAGE(登録商標)Pre−Castゲルシステム(Invitrogen)を製造者の指示書に従って使用した。特に、10%若しくは4〜12%のNuPAGE(登録商標)Novex(登録商標)Bis−TRIS Pre−Castゲル(pH6.4)及びNuPAGE(登録商標)MES(還元ゲル、NuPAGE(登録商標)抗酸化剤ランニングバッファー添加剤を含む)又はMOPS(非還元ゲル)ランニングバッファーを使用した。
抗体の凝集及びオリゴマー状態の決定のためのサイズ排除クロマトグラフィー(SEC)を、HPLCクロマトグラフィーによって実施した。簡潔には、プロテインA精製抗体を、Agilent HPLC 1100システムで300mM NaCl、50mM KH2PO4/K2HPO4(pH7.5)中のTosoh TSKgel G3000SWカラムに、又はDionex HPLC−システムで2倍のPBS中のSuperdex 200カラム(GEヘルスケア)に適用した。UV吸光度及びピーク面積の積分により、溶出したタンパク質を定量化した。BioRad Gel Filtration Standard 151−1901が標準として機能した。
それぞれの抗体は、ポリエチレンイミンを使用してHEK293−EBNA細胞に哺乳動物発現ベクターをコトランスフェクトすることにより産生された。細胞は、対応する発現ベクターを重鎖と軽鎖の1:1の比で用いてトランスフェクトされた。
レンチウイルスベクターを生成するために、構成的に活性なヒトサイトメガロウイルス前初期プロモーター(CMV)の存在下、レンチウイルスポリヌクレオチドベクター中のフレームでそれぞれのDNA配列をクローニングした。レトロウイルスベクターには、ウッドチャック肝炎ウイルスの転写後調節エレメント(WPRE)、中央ポリプリントラクト(cPPT)エレメント、pUC複製起点、及び細菌の増殖と選択を促進する抗生物質耐性をコードする遺伝子が含有されていた。
抗CD20抗体GA101をジゴキシゲニル化し、ジゴキシゲニン(DIG)分子の組み込みをウェスタンブロット分析によって実証した。抗体とジゴキシゲニンのカップリング反応について、20mM His、140mM NaCl、pH6に溶解した抗体を、初めに脱塩し、ZebaTMSpin Desalting Columns (ThermoFisherカタログ番号89889)を使用して、0.1M重炭酸ナトリウム(pH8)バッファーにバッファー交換した。等モル又はより多くの量(1:3の比)の抗体とジゴキシゲニン−3−O−メチルカルボニル−e−アミノカプロン酸−N−ヒドロキシスクシンイミドエステル(Sigma Aldrichカタログ番号11333054001)を、300rpm、振とう機で、室温で1時間インキュベートした。抗体−ジゴキシゲニンコンジュゲートを再度脱塩し、バッファーを20mM His 140mM NaCl pH6に交換した。非コンジュゲートDig−NHSを同じ工程で除去した(カットオフ7kDa)。
Jurkat NFATレポーターCAR−T細胞における抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSDの発現及びジゴキシゲニン−Cy5のCARへの結合を、FACSによって実証した。抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSD形質導入Jurkat NFATレポーターCAR−T細胞を室温で3分間300gでペレットし、適切な体積でフレッシュなRPMI− 1640+10% FCS+1% Glutamax(成長培地)に再懸濁させた。その後3x105細胞を96ウェルプレートの各ウェルに添加し、300gで5分間スピンダウンし、2% FCSを含むPBSの100μl中に再懸濁させた。Dig−Cy5を添加して20nMの最終濃度にし、氷で45分間インキュベートした。その後細胞をペレットし、氷冷PBSに再懸濁させた。洗浄工程をさらに2回繰り返した。その後細胞をフローサイトメトリーを介してCy5シグナル(APCチャネル)について分析した(図7)。ネガティブコントロールとして、非形質導入NFAT細胞を等しく処理及び分析した。
ここでは、CD20発現SUDHDL4腫瘍細胞を標的細胞として使用するレポーターCAR−T細胞アッセイ及びレポーター細胞としての抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSD発現Jurkat NFATレポーターCAR−T細胞のソート済プールが記載される(図9)。ジゴキシゲニル化GA101 IgG(抗体:Dig−NHS比1:10)をIgGとして使用した。これは一方では腫瘍抗原を認識し、他方では、形質導入Jurkat NFATレポーターCAR−T細胞により認識される。ポジティブコントロールとして、96ウェルプレート(Cellstar Greiner−bio−one、カタログ番号655185)をリン酸緩衝食塩水(PBS)中10μg/mlCD3抗体(Biolegend(登録商標))で、4℃で一晩又は37℃で少なくとも1時間コーティングした。CD3をコーティングしたウェルをPBSで2回洗浄し、最終の洗浄工程後にPBSを完全に除去した。レポーター細胞又はJurkat NFAT野生型細胞を数え、Cedex HiResを使用してそれらの生存率について調べた。細胞数を1x106生存細胞/mlに調整した。したがって、細胞懸濁液の適切な一定分量を室温(RT)で5分間、210gでペレットし、フレッシュなRPMI−1640+10% FCS+1% Glutamax(成長培地)に再懸濁させた。対象の抗原を発現する標的細胞を数え、それらの生存率も調べた。細胞数を、成長培地中1x106生存細胞/mlに調整した。標的細胞及びレポーター(エフェクター)細胞を5:1のE:T比(合計で1ウェル当たり110.000細胞)で96ウェル懸濁液培養プレート(Greiner−bio one)中に三重にプレートした。次の工程として、連続希釈したジゴキシゲニル化GA101抗体(対象の抗原を標的化)を、2mlの深さのウェルプレート(Axygen(登録商標))を使用して成長培地中に調製した。1ウェル当たり200μlの最終体積中1μg/mlから0.01pg/mlの最終濃度を得るために、50μlの一定分量の異なる希釈物をそれぞれのウェルにピペットした。96ウェルプレートを190g及びRTで2分間遠心分離した。Parafilm(登録商標)で密封したプレートを、湿度の高い雰囲気中37℃及び5% CO2でインキュベートした。インキュベーションの20時間後、多重チャンネルピペットを使用して10回ピペットアップ及びダウンすることにより、各ウェルの内容物を混合した。100μl細胞懸濁液を新たな白色透明平底96ウェルプレート(Greiner−bio−one)に移し、100μlのONE−GloTMLuciferase Assay(Promega)を添加した。300rpm及び室温で、回転振とう機で暗所における15分間のインキュベーション後、検出時間1秒/ウェルでTecan(登録商標)Spark10Mプレートリーダーを使用して発光を測定した。標的細胞及びレポーター細胞の5:1の比(灰色の点)での20時間の共培養において、グラフは、ジゴキシゲニル化GA101 IgGを抗体として使用したときの抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSD発現Jurkat NFATレポーターCAR−T細胞の用量依存的活性化を示す(図9)。ジゴキシゲニル化されていないGA101 IgG(図9、灰色の四角形で図示)を使用したとき、検出可能な形質導入Jurkat NFATレポーターCAR−T細胞の活性化はなかった。1μg/ml ジゴキシゲニル化GA101でインキュベートされたが標的細胞を含まないさらなるJurkat NFAT野生型細胞は、活性化を示さなかった(図9、黒色の四角形)。対照的に、1μg/ml ジゴキシゲニル化GA101 IgGでインキュベートしたが、標的細胞を含まない抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSD発現Jurkat NFATレポーターCAR−T細胞は、活性化を示した(図9、黒色の三角形)。
レポーター細胞としての抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSD発現Jurkat NFATレポーターCAR−T細胞(図10)、及び異なる量のジゴキシゲニン(DIG)分子に結合した抗CD20 IgG抗体(GA101)のソート済プールを使用したレポーターCAR−T細胞アッセイが本明細書で記載される。エフェクター細胞を数え、Cedex HiResを使用してそれらの生存率を調べた。細胞数を1x106生存細胞/mlに調整した。したがって、細胞懸濁液の適切な一定分量を室温(RT)で5分間、210gでペレットし、フレッシュなRPMI−1640+10% FCS+1% Glutamax(成長培地)に再懸濁させた。1x105レポーター細胞を、96ウェル懸濁液培養プレート(Greiner−bio one)に三重にプレートした。連続希釈抗GA101抗体を、2mlの深さのウェルプレート(Axygen(登録商標))を使用して成長培地中に調製した。抗GA101抗体は、平均して、1(1:1 GA101−Dig)、3(1:3 GA101−Dig)又は10(1:10 GA101−Dig)ジゴキシゲニン分子のいずれかの特徴を使用した。コントロールとして、非ジゴキシゲニル化抗体(GA101 wt)を使用した。

ここでは、LeY発現MCF7腫瘍細胞を標的細胞として使用するレポーターCAR−T細胞アッセイ及び抗ジゴキシゲニン−ds−scFv−CD28ATD−CD28CSDCD3zSSD発現Jurkat NFATレポーターCAR−T細胞のソート済プールが記載される。2つの抗LeY/ビオチン抗体(2+1、2+2)及び架橋ビオチン−ジゴキシゲニンアダプターを使用した。抗体は、一方では腫瘍関連抗原を認識し、他方ではビオチン化アダプター分子を認識する。アダプター結合ジゴキシゲニンは、本発明によるCARを発現するJurkat NFATレポーターCAR−T細胞により認識される(図13)。
Claims (15)
- 標的抗原に特異的に結合することができる標的抗原結合部分の特異性を評価するための方法であって、以下の工程:
a)抗原結合ドメイン及び認識ドメインを含む抗原結合分子を提供する工程であって、抗原結合ドメインが標的抗原結合部分を含み、認識ドメインがタグを含む、工程;
b)抗原結合分子を、表面上に標的抗原を含む標的細胞と接触させる工程であって、標的細胞ががん細胞である、工程;
c)抗原結合分子を、キメラ抗原受容体(CAR)発現レポーターT(CAR−T)細胞と接触させる工程であって、レポーターCAR−T細胞が以下:
i.タグを含む認識ドメインに特異的に結合することができるCARであって、応答エレメントに作用的に結合されている、CAR;
ii.応答エレメントの制御下のレポーター遺伝子;
を含む工程;及び
d)レポーター遺伝子の発現を測定することによりT細胞活性化を決定し、標的抗原結合部分の特異性を確立する工程
を含む方法。 - 抗原結合分子が、IgGクラス抗体、特にIgG1又はIgG4アイソタイプ抗体、又はその断片である、請求項1に記載の方法。
- 抗原結合ドメインがFab断片であり、認識ドメインがFcドメインである、請求項1又は2に記載の方法。
- 抗原結合ドメイン及び認識ドメインが、同じドメイン、特にFab断片である、請求項1又は2に記載の方法。
- タグがハプテン分子である、請求項1から4のいずれか一項に記載の方法。
- ハプテン分子がジゴキシゲニン(DIG)である、請求項1から5のいずれか一項に記載の方法。
- タグがポリペプチドタグである、請求項1から4のいずれか一項に記載の方法。
- ポリペプチドタグが、mycタグ、HAタグ、Aviタグ、FLAGタグ、Hisタグ、GCN4タグ、及びNEタグからなる群より選択される、請求項7に記載の方法。
- 標的抗原が細胞表面抗原又は受容体である、請求項1から8のいずれか一項に記載の方法。
- 標的抗原が、ヒト主要組織適合複合体(MHC)の分子に結合したペプチドであり、標的抗原結合部分が、T細胞受容体様(TCRL)抗原結合部分である、請求項1から9のいずれか一項に記載の方法。
- TCB抗体を生成するための方法であって、TCB抗体が、標的抗原に特異的な第1の抗原結合部分と、T細胞活性化受容体に特異的に結合することができる第2の抗原結合部分とを含み、第1の抗原結合部分が請求項1から10のいずれか一つに記載の方法に従って選択される、方法。
- T細胞活性化受容体がCD3である、請求項11に記載の方法。
- 固定膜貫通ドメインと、抗原結合部分を含む細胞外ドメインとを含むキメラ抗原受容体(CAR)であって、抗原結合部分が、タグを含む認識ドメインに特異的に結合することができるが、タグを含まない認識ドメインには特異的に結合することができない、キメラ抗原受容体(CAR)。
- タグがハプテン分子である、請求項13に記載のCAR。
- ハプテン分子がジゴキシゲニン(DIG)である、請求項14に記載のCAR。
