図2はIncucyte(登録商標)S3装置を用いて撮影された、DIV14における成熟ニューロンの白黒ネガティブ位相差顕微鏡画像であり、研究で使用されたものを代表する。
図3A~3Cは、LIPID MIX A(3B)を用いて調製したプラスミドGFP-LNP、またはLIPID MIX L(3C)を用いて調製したプラスミドGFP-LNPでの処理後、DIV14、未処理(3A)、および48時間でIncucyte(登録商標)機器を用いて撮影したiPSC由来ニューロンの代表的な位相コントラスト黒および白ネガティブ画像である。
図4A~4Dは4日目にIncucyte(登録商標)S3機器を用い、未処理(4A)、およびN/P6(4B)のLIPID MIX A、N/P6(4C)のLIPID MIX L、またはGFPをコードするプラスミドを含むLipofectamine(登録商標)(4D)からなる脂質粒子での処理の48時間後に撮影した、iPSC由来神経前駆細胞の黒色および白色のネガティブ位相コントラスト画像である。
図5は、LIPID MIX A、LIPID MIX T20、LIPID MIX T80、LIPID MIX K、またはLIPID MIX Lを用いて調製したプラスミドGFP-LNP(N/P4)での処置後0~144時間で捕捉した画像からの、DIV14でGFPを発現するiPSC由来ニューロンのパーセンテージのグラフである。

代表的なアミノ脂質としては、1,2-ジリノレオキシ-3-(ジメチルアミノ)アセトキシプロパン(DLin-DAC)、1,2-ジリノレオキシ-3-モルホリノプロパン(DLin-DAP)、1,2-ジリノレオイル-3-ジメチルアミノプロパン(DLin-S-DMA)、1,2-ジリノレオイル-3-トリメチルアミノプロパンクロリド(DLin-TAPOOACI)、1,2-ジリノレオイルオキシ-3-(N-メチルピペラジノ)プロパン(DLin-MPZ)3-(N,N-ジリノールアミノ)-l,2-プロパンジオール(DLinAP)、3-(N,N-ジオレイルアミノ)-l,2-プロパンジオ(DOAP)、1,2-ジリノレイル-3-(2-N,N-ジメチルアミノ)エトキシプロパン(DLin-EG-DMA)、2,2-ジリノレイル-4-ジメチルアミノメチル-[l,3]-ジオキソラン(DLin-K-DMA)1,2-ジリノレイルオキシ-N,N-ジメチル-3-アミノプロパン(DLin-DMA)、ヘプタトリアコンタ-6,9,28,31-テトラエン-19-イル4-(ジメチルアミノ)ブタノエート(DLin-MC3-DMA)、2,2-ジリノレイル-4-(2-ジメチルアミノエチル)-[1,3]-ジオキソラン(DLin-KC2-DMA)、および1,2-ジオレイルオキシ-3-ジメチルアミノプロパンが挙げられる。
本明細書中で使用される場合、用語「約」は、列挙された数の10%±を意味すると定義される。これは所望の目標濃度が例えば、40モル%であってもよいが、混合の不一致によって、実際のパーセンテージが+/5モル%だけ異なってもよいことを示すために使用される。
材料及び方法
脂質混合物のいくつかは、Avanti Polar Lipids(Alabaster、AL、米国)によって合成された1,17-ビス(2-オクチルシクロプロピル)ヘプタデカン-9-イル4-(ジメチルアミノ)ブタノエート(BOCHD-C3-DMA)で形成された。他の脂質混合物を、DODMA(Avanti Polar Lipids)またはCOATSOME(登録商標)SS-E-P4C2、カタログ番号SS-33/4PE-15(NOF America Corporation、アービン、カナダ)を用いて形成した。
非組み込みoriP/EBNAlプラスミドを用いて、再プログラムドナー由来線維芽細胞から人工多能性幹細胞(iPSC)を作製した。細胞をCorning(登録商標)増殖因子減少(GFR)基本膜マトリックス(#354230)上で維持し、完全mTeSR培地(補充物を含む基本培地)(StemCell Technologies、#05850)中で維持した。3人の個々のドナーからの3つのiPSCクローンを、250ng/mLのGFP mRNA脂質粒子および100ng/mLのApoEで48時間処理した。
NPCは誘導された多能性幹細胞に由来する未熟なニューロン細胞型であり、特定の培養条件、試薬および分化を促進する細胞培養培地を用いて成熟皮質ニューロンに分化させることができる。iPSCクローンから機能的および生理学的に定義された成熟皮質ニューロンへの分化過程は、時間がかかり技術的に困難な過程である。市販のNPCは、これらの細胞型が分化経路にさらに沿っているので、ニューロン培養のプロセスを加速する。これらの研究において、十分に特徴付けられたNPCを、StemCell Technologies(Vancouver、Canada、Female cell line Cat #70902、Male line: StemCell Technologies Cat #70901)から購入し、培養して成熟皮質ニューロンを誘導した。本発明の脂質粒子製剤は、この分化経路に沿って、前駆段階からニューロン成熟の時点までの様々な時点に適用することができる。これはまた、脂質粒子ベースの核酸ベクターの、iPSC自体、またはNPC段階の上流のニューロン細胞型の遺伝子操作への可能な適用を示唆する。
ニューロンが必要な日(1日目、7日目、10日目、14日目など)まで成熟したとき、1 ug/mLのApoEを各ウェルに添加し、続いて600ng/mLの核酸治療薬/LのLipid Mix製剤を添加した。次に、処理したプレートを、細胞培養インキュベーター内に置いたIncucyte(登録商標)S3 Live-Cell Analysis System(Essen Biosciences、Ann Arbor、Michigan、USA)を用いてリアルタイムで分析した。Incucyte(登録商標)S3は実験の間、6時間毎に、各ウェル内の6つの視野位置で10倍の倍率で画像を撮影するようにプログラムされた。データ取得に続いて、Incucyte(登録商標)S3ソフトウェアを用いてアルゴリズムを作成し、位相差画像および緑色蛍光タンパク質(「GFP」)画像の両方について細胞体および%コンフルエンスを同定した。
核酸治療薬(NAT)の脂質ナノ粒子(LNP)へのマイクロ流体混合
脂質混合物溶液は、エタノール中の個々の脂質ストックからの規定量の脂質(表1を参照のこと)を合わせることによって、エタノール中で調製した。NanoAssemblr(登録商標)SPARK法では50mMの脂質混合溶液濃度を使用し、NanoAssemblr(登録商標)ベンチトップでは典型的には12.5mMの脂質混合溶液を使用した。
リピドミックスAの成分は1,17-ビス(2-オクチルシクロプロピル)ヘプタデカン-9-イル-4-(ジメチルアミノ)ブタノエート塩酸塩、中性脂質DOPE、安定剤Myrj52(ポリオキシエチレン(40)ステアレート)であり、リピドミックスA DODMAには、1,17-ビス(2オクチルシクロプロピル)ヘプタデカン-9-イル-4-(ジメチルアミノ)ブタノエート塩酸塩の代わりにDODMAを用い、リピドミックスA-COATSOMEにはCOATSOME(登録商標)を用いた。安定化剤Myrjを、最初に単一の界面活性剤、次に2つの界面活性剤、次に3つの界面活性剤で置き換えた。
*ILはイオン化可能な脂質。BOCHD-C3-DMA、DODMA、およびCoatsornet(登録商標)は種々の実施例のILであった。
LipidMixAまたはLipidMixKの後の接尾辞DODMAまたはCOATSOMEは、これらの脂質が使用された場所を示すために使用される。他のすべての状況では、ILはBOCHD-C3-DMAである。

脂質粒子または「LNP」キャラクタリゼーションおよびカプセル化
粒子径(粒子の流体力学的直径)は、ZetaSizer Nano ZS(登録商標)、英国Malvern Instruments社を用いてDynamic Light Scattering(DLS)によって決定した。光源には波長633nmのHe/Neレーザを用いた。後方散乱検出モード(測定角度173)で行った散乱強度データからデータを測定した。測定は、各サンプルあたり2サイクルの平均10回の実験であった。Z-平均サイズは、脂質粒子サイズとして報告され、高調波強度平均粒子直径として定義される。
プラスミドLNPによるニューロンの処置およびIncucyte(登録商標)S3-GFP発現を使用する評価
ddH2O中のアンピシリン耐性、制限酵素HINDIIIを含む、GenScript USA Inc., Piscataway、NJによってカスタムメードされたプラスミド調製物pCX-EGFPプラスミドサイズ5514 nt、配列番号1を、この評価のために使用した。プラスミドは、プラスミドが細胞内で発現される場合にのみ標的タンパク質を産生するGFP発現成分を含んでいた。
細胞の健康に及ぼす製剤の影響
参考のポイントとして、図2はニューロン成熟14日目(DIV14)の未処理成熟ニューロンの位相差画像である。図3A、3B、および3Cは、アミノ脂質対ヌクレオチドリン酸モル比N/P6でGFPをコードするプラスミドを含有するLIPID MIX AまたはLIPID MIX Lからなる脂質粒子で処理した後、未処理および48時間後の、DIV14におけるiPSC由来ニューロンの位相差画像である。示されているデータは、Incucyte(登録商標)のライブイメージング機器からのものである。観察:位相コントラスト画像中の白色丸細胞は死細胞/死細胞を示し;脂質混合物Aの画像が製剤がヒトiPSC由来ニューロンに対して毒性であることを示し;および脂質混合物Lが未処理細胞に非常に類似している(すなわち、毒性がない)。
フローサイトメトリーでそのようにして確認されたiPSC由来の成熟ニューロン
ニューロンの処理およびインキュベーション後、各ウェルから培地を回収し、PBS中の3% FBSで不活性化した0.25%トリプシンを用いて細胞を回収し、それらの対応する培地中でペレット化した。遠心分離後、細胞をPBSで1回洗浄し、再びペレット化した。次いで、細胞をBinding Buffer(登録商標)(BD Biosciences)に再懸濁し、ヨウ化プロピジウムを添加して、完全に破裂した膜(死細胞)を有する細胞について染色した。次いで、BD Biosciences Canto(登録商標)IIフローサイトメーターを用いて細胞を評価した。
ニューロンの治療とニューロン生存能の評価
細胞生存率および神経突起伸長は、神経細胞の健康および機能の最も一般的に測定される指標である。細胞傷害性処理を受けた神経細胞は、しばしばその神経突起を短縮することによって反応する。また、神経突起伸長は、生存可能なニューロン培養のためのゴールドスタンダードマーカーである。神経突起長の測定単位はmm/mm2である。「平均神経突起長」は、mm2当たりの神経突起の長さを見て、ウェルの全ての画像からのデータから導出される。
mRNAトランスフェクション
非組み込みoriP/EBNAl{Yu、2009、Human induced pluripotent stem cells free vector and transgene sequences}プラスミドを用いて、再プログラムドナー由来線維芽細胞から人工多能性幹細胞(iPSC)を作製した。細胞をCorning(登録商標)増殖因子減少(GFR)基本膜マトリックス(#354230)上で維持し、完全mTeSR培地(補充物を含む基本培地)(StemCell Technologies、#05850)中で維持した。3人の個々のドナーからの3つのiPSCクローンを、250ng/mLのGFP mRNA(CleanCap(登録商標)EGFP mRNA CleanCap(登録商標)Enhanced Green Fluorescent Protein mRNA(Trilink Biotechnologies、San Diego、CA))脂質粒子および100ng/mLのApoEで48時間処理した。996bpのmRNAは、配列番号2として表される。DAPI核標識を用いて細胞の総数を決定し、次いで、ウェル当たりの細胞の総数に対するGFP発現細胞のパーセンテージを計算した。図7に示されるように、Lipid Mix Lから作製された脂質粒子は、未処理およびLipid Mix A群と比較して、GFP発現iPSCの最も高い比率を示した(40.4%対8.36%)。グラフは、処置群あたりのGFP陽性細胞の平均パーセンテージを表す。この実験は、mRNAならびにプラスミドトランスフェクションが本発明の脂質混合物によって好都合に達成され、そしてiPSCが首尾よくトランスフェクトされ得ることを示す。
インビトロでのプラスミドトランスフェクション、GFP発現
プラスミドおよび脂質粒子の調製は、上記で特定した通りであった。この実験では、イオン化可能な脂質、中性脂質、コレステロールおよび界面活性剤の異なる比率を、プラスミドカプセル化と共に使用した。記載された製剤の詳細については表1を参照されたい。LNP Tween 20およびLNP Tween 80は、mRNA Lipid Mix T20およびT80に使用したものとはわずかに異なる比率のコレステロールおよび安定化剤を有していた。実験の目的は個々の界面活性剤と組み合わせた脂質粒子組成物を使用して、インビトロで神経前駆細胞(NPC)におけるGFPのプラスミド媒介発現の効果を研究することであり、第2の部分では、標準的な界面活性剤Myrj52に対して、単一の薬剤として異なる界面活性剤を使用した。脂質粒子中に1モル%のtween 20、1モル%のTween80、および1モル%のtween 20およびTween80の1対1の組み合わせを含む製剤によって媒介されるトランスフェクション活性を図8Aに示す。2つのTweensの組み合わせは、細胞中にGFPタンパク質発現を点滴注入するために、各Tweensよりも個々に良好に機能した。
ロニザブル脂質同一性に関わらず存在する界面活性剤併用の効果
NPCを、交互のイオン化可能な脂質含有形態のLipid Mix AおよびLipid Mix Kカプセル化mRNAに曝露して、任意のイオン化可能な脂質特異的効果を同定した。図9Aは、Lipid Mix KおよびLipid Mix K DODMAと比較した、Lipid Mix AおよびLipid Mix A DODMAについてのGFPのパーセントとしてのmRNA発現を示す。同様の結果がDODMAとは対照的に、1,17-ビス(2-オクチルシクロプロピル)ヘプタデカン-9-イル4-(ジメチルアミノ)ブタノエートHClを用いて作製された脂質混合物について見られる。
トランスフェクタミン(登録商標)トランスフェクション剤(Life Technologies Inc.)を、NPC中のLipid Mix KおよびLipid Mix L(1,17-ビス(2-オクチルシクロプロピル)ヘプタデカン-9-イル4-(ジメチルアミノ)ブタノエートHCI)と比較して、細胞の健康に対するその効果を調べた。結果を図10に示す。継代培養を含む正常細胞培養144時間後、Lipofectamine(登録商標)で処理した細胞は生存不能であったが、Lipid Mix KおよびLで処理した細胞はほぼ100%コンフルエントであった。
一般的に使用される2つのイオン化可能な脂質、KC2およびDLin-MC3-DMAを、脂質混合物AおよびKのイオン化可能な脂質成分の代わりに用いた。NPCを、上記のようにトランスフェクトし、フローサイトメトリーによるGFP中央値蛍光強度についての48Hでの結果を図11に示す。参考のために、脂質粒子サイズ、PDI、およびカプセル化効率を表4に列挙する。KC2およびDLin-MC3-DMA(グラフ中のMC3)はともに、Lipid Mix Aに対するGFP表現を強化するためにLipid Mix Kで有効であり、PNI-IL-1はl,17-bis(2-オクタイシクロピロピル)ヘプタデカン-9-yl 4-(ジエチルアミノブタノベートHCI)である。

Claims (14)
- 35~50モル%のイオン化可能な脂質またはその薬学的に許容される塩と、
15~25モル%の構造脂質と、
35~41モル%のステロールと、
0.5~約10モル%の安定剤を含み、
安定化剤がポリソルベート80、ポリソルベート20、およびトリデシル-D-マルトシドの混合物であり、前記混合物におけるポリソルベート80、ポリソルベート20、およびトリデシル-D-マルトシドの比率が等しい、トランスフェクション試薬組成物。 - 前記イオン化可能な脂質が、アミノ脂質またはその薬学的に許容される塩である、請求項1に記載の組成物。
- イオン化可能な脂質が組成物の約40モル%を構成する、請求項1または2に記載の組成物。
- 安定化剤が組成物の約0.5~5モル%を構成する、請求項1~または2に記載の組成物。
- ステロールがコレステロールである、請求項1~4のいずれか1項に記載の組成物。
- 核酸またはペプチドをさらに含む、請求項1~5のいずれか1項に記載の組成物。
- 核酸が、DNA、RNA、またはRNAを発現することができるプラスミドである、請求項6に記載の組成物。
- 前記イオン化可能な脂質が、BOCHD-C3-DMA、KC2、MC3、a-D-トコフェロルスクシノイルおよびDODMA、またはその薬学的に許容される塩から選択される、請求項1~7のいずれか1項に記載の組成物。
- 構造脂質が、1,2ジオレオイル-sn-グリセロ-3-ホスホエタノールアミン(DOPE)またはDSPEまたはDSPCである、請求項1~8のいずれか1項に記載の組成物。
- 前記組成物が、約15nm~約300nmの直径を有する脂質ナノ粒子の形態で存在する、請求項1~9のいずれか1項に記載の組成物。
- iPSCまたはiPSC様細胞を、核酸の活性および細胞の生存率を維持しながら、核酸治療剤でトランスフェクトするためのインビトロの方法であって、細胞を請求項1~10のいずれか1項に記載の組成物と接触させるステップを含む、方法。
- 核酸の活性および細胞の生存率を維持しながら、ニューロン細胞を核酸治療剤でトランスフェクトするためのインビトロの方法であって、細胞を請求項1~10のいずれか1項に記載の組成物と接触させるステップを含む、方法。
- 細胞が哺乳動物細胞である、請求項11または12に記載の方法。
