Description
本発明は以下を含むものである。
[1]以下のアミノ酸配列:
(i)SLSHSPQ(配列番号1)からなる配列、または
(ii)上記配列番号1に記載のアミノ酸配列において、Pが、A,I,L,V,M,F,W,Y,S,T,N,Q,HおよびGからなる群より選ばれるアミノ酸(好ましくは、A,I,L,V,M,S,T,N,QおよびGからなる群より選ばれるアミノ酸、より好ましくは、A,I,L,V,S,TおよびGからなる群より選ばれるアミノ酸)に置換された配列、
を含む血液脳関門透過性ペプチド。
[2]少なくとも一つの非天然アミノ酸を含む上記[1]に記載の血液脳関門透過性ペプチド。
[3]前記ペプチドが環状ペプチドである上記[1]または[2]に記載の血液脳関門透過性ペプチド。
[4]前記ペプチドが下記の(a)~(f)のいずれかである、上記[1]に記載の血液脳関門透過性ペプチド、

(f)SLSHSPQ(配列番号1)のアミノ酸配列のC末端およびN末端に1~15個の任意のアミノ酸を有するペプチドであって、該配列の両端に存在する任意のアミノ酸同士が架橋されているペプチド。

本明細書中で、「X~Y」という表現を用いた場合は、下限としてXを上限としてYを含む意味で、或いは上限としてXを下限としてYを含む意味で用いる。
本明細書中でアミノ酸を示す場合は、1文字表記である、A(アラニン)、R(アルギニン)、N(アスパラギン)、D(アスパラギン酸)、C(システイン)、Q(グルタミン)、E(グルタミン酸)、G(グリシン)、H(ヒスチジン)、I(イソロイシン)、L(ロイシン)、K(リジン)、M(メチオニン)、F(フェニルアラニン)、P(プロリン)、S(セリン)、T(トレオニン)、W(トリプトファン)、Y(チロシン)、V(バリン)で表す。
本発明のBBB透過性ペプチド(以下、単に本発明のペプチドという場合がある)は、アミノ酸配列:SLSHSPQ(配列番号1)またはその類似する配列を有することを特徴とする。類似する配列とは、配列番号1に示される配列SLSHSPQにおいて、Pが、A,I,L,V,M,F,W,Y,S,T,N,Q,HおよびGからなる群より選ばれるアミノ酸、好ましくは、A,I,L,V,M,S,T,N,QおよびGからなる群より選ばれるアミノ酸、より好ましくは、A,I,L,V,S,TおよびGからなる群より選ばれるアミノ酸に置換された配列を意味し、本明細書では、単に、配列番号1の類似配列という場合がある。本発明のペプチドは、全てが天然のL体のアミノ酸から構成されるものに限定されず、L体またはD体のいずれのアミノ酸から構成されてもよく、また、L体およびD体のアミノ酸が混合したもので構成されてもよい。本発明のペプチドにおいては、天然に存在するアミノ酸である立体配置がL体であるアミノ酸の他に、天然のアミノ酸の構造を一部改変した誘導体など非天然のアミノ酸も使用されうる。非天然アミノ酸としては、特に制限されず、公知の任意のアミノ酸を用いることができる。例えば、立体配置がD体のアミノ酸やN-メチルアミノ酸は、蛋白分解酵素による分解を受けにくいことから本発明のペプチドに有効に使用されうる。よって、本発明のペプチドは、そのアミノ酸配列のうち、少なくとも一部がD体やN-メチルアミノ酸などの非天然型アミノ酸から構成されるのが好ましい。
(a)アミノ酸配列:SLSHSPQ(配列番号1)からなるペプチド、
(b)SLSHSPQ(配列番号1)のアミノ酸配列のC末端および/またはN末端に1~5個の任意のアミノ酸を有するペプチド、
(c)アミノ酸配列:CSLSHSPQC(配列番号2)からなるペプチドであって、前記配列中のシステイン残基同士がジスルフィド結合しているペプチド、
(d)CSLSHSPQC(配列番号2)で示されるアミノ酸配列のC末端および/またはN末端に1~10個の任意のアミノ酸を有するペプチドであって、前記配列中のシステイン残基同士がジスルフィド結合しているペプチド、または
(e)下記式(1):
(f)SLSHSPQ(配列番号1)のアミノ酸配列のC末端およびN末端に1~15個の任意のアミノ酸を有するペプチドであって、該配列の両端に存在する任意のアミノ酸同士が架橋されているペプチド。
(g)上記(a)~(f)のいずれかに記載のペプチドにおいて、アミノ酸配列SLSHSPQにおけるPが、A,I,L,V,M,F,W,Y,S,T,N,Q,HおよびGからなる群より選ばれるアミノ酸、好ましくは、A,I,L,V,M,S,T,N,QおよびGからなる群より選ばれるアミノ酸、より好ましくは、A,I,L,V,S,TおよびGからなる群より選ばれるアミノ酸に置換された配列であるペプチド。
ここで、AAaおよびAAdは、それぞれ独立に、1~10個、好ましくは1~7個、より好ましくは1~5個の任意のアミノ酸を表し、AAbおよびAAcは、それぞれ独立に、1~5個の、好ましくは1~3個の、より好ましくは1~2個の任意のアミノ酸を表す。上記式において、AAa~AAdで表されるそれぞれのアミノ酸は、天然アミノ酸、非天然アミノ酸のいずれでもよく、またそれらの任意の修飾アミノ酸でもよい。
架橋を形成するための環化は、アミノ酸合成技術の分野において用いられている公知の技術を制限なく用いることができる。代表的な環化の例としては、ジスルフィド結合(SS結合)、アミド結合、チオエーテル結合、オレフィン結合、ラクタム環を介した架橋をあげることができる。なお、ジスルフィド結合(SS結合)の場合は、上記(c)~(d)の態様を含む。これに限定されないが、具体的な例としては、二つのペニシラミン残基をジスルフィド架橋で連結することにより環化するもの(Mosberg ら、P.N.A.S. US, 80:5871, 1983)、リジンとアスパラギン酸との間にアミド結合を形成することにより環化するもの(Flora ら、Bioorg. Med. Chem. Lett. 15 (2005) 1065-1068)、または予めチオエーテル結合を導入した架橋部位を含むアミノ酸誘導体をペプチド結合に導入し、縮合反応で環化するもの(Melinら 米国特許第6,143,722号)、およびオレフィンメタセシス反応を用いて主鎖に導入した(S)-α-2'-ペンテニル)アラニン同士を架橋させて環化するもの(Schafmeisterら、J. Am. Chem. Soc., 122, 5891-5892, 2000)をあげることができる。また、架橋を構成するアミノ酸の間に任意のスペーサーを導入することもできる。スペーサーは、これに限定されないが、例えば、任意の長さのポリエチレングリコールをあげることができる。その他、上記のアミノ酸以外にも、架橋を形成するための特別な構造のアミノ酸を、アンカー部分を構成するアミノ酸の位置に導入し、架橋を形成する技術が報告されているが、それらも本発明のペプチドの合成において用いることができる。
本明細書において「環状ペプチド」という場合は、ペプチドのN末端とC末端が連結されて環状を形成している場合、ペプチドのN末端またはC末端の一方とペプチド鎖の任意の場所のアミノ酸残基が連結されて環状を形成している場合、または、ペプチド鎖の任意の2箇所のアミノ酸残基が連結されて環状を形成している場合のいずれも含む意味であり、また、環を形成する連結様式は、アミノ酸残基同士が直接結合する場合、架橋を構成するリンカーを間に含む場合のいずれも含まれる。
環状ペプチドを形成する方法は、様々な方法が報告されており、それらの方法を適宜使用し、環状を有する本発明のBBB透過性ペプチドを作製することができる。それらの報告は、修飾アミノ酸を含むアミノ酸残基同士を直接結合する方法、リンカーを介して2つのアミノ酸残基を架橋する方法を含む。本発明のペプチドにおいて、環を形成する場合の環の種類や環の形成様式は特に制限がなく、目的に応じて、任意の種類や様式を選択できる。例えば、ある態様においては、システイン残基同士のジスルフィド結合が簡便であり好ましいが、別の態様においは、血中での分解耐性を望む場合は、他の結合様式、例えば、オレフィン結合による環化を選択することもできる。
上記(a)で表される本発明のペプチドは、配列番号1で示されるアミノ酸配列を有し、そのC末端およびN末端が、直接、或いは任意のスペーサーを介して、結合し環を形成してもよい。
上記(b)で表される本発明のペプチドは、そのC末端およびN末端が、直接、或いは任意のスペーサーを介して、結合し環を形成してもよく、また、ペプチド鎖の任意の位置のアミノ酸残基同士が、好ましくは、配列番号1のアミノ酸配列を環内に含むようにして、架橋を形成して環を形成してもよい。
上記(c)~(e)で表される本発明のペプチドは、システイン残基間のジスルフィド結合により環を形成している。
本発明のBBB透過性ペプチドは、BBB透過性を有しており、他の分子(本明細書中では、「目的分子」という場合がある)とともに用いることにより、目的分子のBBB透過を助け、目的分子の脳内への移行を可能とする。目的分子としては、これに限定されないが、例えば、その分子そのものが脳内において生理活性を有する物質(以下、単に生理活性物質という場合がある)または脳内において機能を発揮する物質(以下、これらをあわせて脳内活性物質という場合がある)である分子、または、脳内活性物質を含むまたは保持できる薬物送達に用いることができる分子(本明細書では、薬物送達用キャリア分子または単にキャリア分子ともいう)をあげることができる。後者の場合、本発明のBBB透過ペプチドを用いることにより、キャリア分子とともにそれに含まれるまたは保持された脳内活性物質がBBBを透過でき、脳内へと移行される。また、本発明のBBB透過性ペプチドは、従来のBBB透過性ペプチドに比べ、より大きな目的分子を透過(脳内へ移行)させることができる。
公知の方法を用いて、これらの低分子化合物と本発明のペプチドからなる複合体を形成させることができる。例えば、これに限定されないが、低分子化合物および/または本発明のペプチドを化学的に修飾し、場合によりスペーサーを介して、両者を結合されればよい。目的分子である低分子化合物と本発明のペプチドからなる複合体も、本発明に含まれる。
公知の方法を用いて、これらのタンパク質と本発明のペプチドからなる複合体を形成させることができる。例えば、これに限定されないが、タンパク質および/または本発明のペプチドを化学的に修飾し、場合によりスペーサーを介して、両者を結合されればよい。或いは、非共有結合的に、例えば、静電的にタンパク質と本発明のペプチドを結合させてもよい。さらには、遺伝子工学的手法を用いて、目的分子であるタンパク質のアミノ酸配列と本発明のペプチドのアミノ酸配列をともに有する融合タンパク質(好ましくは、本発明のペプチドの配列をN末端またはC末端に有する融合タンパク質)として複合体を作製することもできる。また、目的分子であるタンパク質の配列と本発明のペプチドの配列の間に任意の配列を入れることも可能である。目的分子であるタンパク質と本発明のペプチドからなる複合体も、本発明に含まれる。
公知の方法を用いて、これらのペプチドと本発明のペプチドからなる複合体を形成させることができる。例えば、これに限定されないが、以下の方法をあげることができる。目的分子であるペプチドおよび本発明のペプチドの両者を含む(場合により間に任意のアミノ酸配列を含む)配列のペプチドを、ペプチド合成の技術を用いて合成することができる。目的分子であるペプチドおよび/または本発明のペプチドを化学的に修飾し、場合によりスペーサーを介して、両者を結合させればよい。或いは、非共有結合的に、両者を結合させてもよい。さらには、遺伝子工学的手法を用いて、目的分子であるペプチドのアミノ酸配列と本発明のペプチドのアミノ酸配列をともに有する融合ペプチド(好ましくは、本発明のペプチドの配列をN末端またはC末端に有する融合ペプチド)として複合体を作製することもでき、また、目的分子であるペプチドの配列と本発明のペプチドの配列の間に任意の配列を入れることも可能である。目的分子であるペプチドと本発明のペプチドからなる複合体も、本発明に含まれる。
公知の方法を用いて、これらの核酸と本発明のペプチドからなる複合体を形成させることができる。例えば、これに限定されないが、核酸および/または本発明のペプチドを化学的に修飾し、場合によりスペーサーを介して、両者を結合されればよい。或いは、非共有結合的に、例えば、静電的に核酸と本発明のペプチドを結合させてもよい。目的分子である核酸と本発明のペプチドからなる複合体も、本発明に含まれる。
公知の方法を用いて、これらの物質と本発明のペプチドからなる複合体を形成させることができる。目的分子であるこれらの物質と本発明のペプチドからなる複合体も、本発明に含まれる。
よって、本発明のBBB透過性ペプチドおよび本発明のBBB透過性ペプチドが付加された薬物送達用キャリア分子は、BBBを透過しないまたはBBB透過性が低い分子を脳内に送達させるための、脳内送達用キャリアでもある。
よって、本発明はまた、本発明のBBB透過性ペプチド、目的分子(脳内活性物質や薬物送達用キャリア分子)、および、トランスフェリン、フィブリノーゲンまたはRGD配列を含むペプチドから選ばれる物質、を含む組成物でもある。好ましくは、本発明のBBB透過性ペプチド、目的分子(脳内活性物質や薬物送達用キャリア分子)、およびトランスフェリンを含む組成物である。
さらに本発明はまた、本発明のBBB透過性ペプチド、目的分子(脳内活性物質や薬物送達用キャリア分子)、および、トランスフェリン、フィブリノーゲンまたはRGD配列を含むペプチドから選ばれる物質、を含む複合体でもある。好ましくは、本発明のBBB透過性ペプチド、目的分子(脳内活性物質や薬物送達用キャリア分子)、およびトランスフェリンを含む複合体である。
フィブリノーゲンは、肝臓で産生される凝固因子であり血栓の骨格となるフィブリンの前駆体である。本発明で用いることができるフィブリノーゲンは、その由来は特に制限がないが、好ましくはヒト由来のものである。
上記RGD配列を含むペプチドは、例えば、RGD配列を含む100アミノ酸配列以下の、好ましくは50アミノ酸配列以下の、さらに好ましくは20アミノ酸配列以下の、よりさらに好ましくは10アミノ酸配列以下のペプチドである。RGDモチーフは、多くの細胞接着性タンパク質に共通の細胞接着活性配列である。
本発明のBBB透過ペプチドと目的分子(脳内活性物質や薬物送達用キャリア分子)を含む複合体は医薬として利用が可能である。本発明の複合体を含む医薬品は、公知の方法に従って製剤化し、投与することができる。例えば、そのまま液剤としてまたは適当な剤型の医薬組成物として、ヒトを含む哺乳動物に対して非経口的または経口的に投与することができる。非経口的投与方法としては、例えば、注射剤又は貼付剤(経皮投与)が例示できる。本発明の複合体を含む医薬組成物は、好ましくは非経口的に投与される。
(実施例1)BBB透過ペプチドのスクリーニング
BBBを効率的に透過するペプチドの同定を目的としてペプチドのスクリーニングを行った。ペプチドのスクリーニングは、7アミノ酸がランダムに提示されるphageライブラリー(1x109種類)とヒトBBBモデル細胞であるhCMEC/D3細胞を用いた。
(1)phageライブラリーの作製
Ph.D.-C7C Phage Display Peptide Library Kit(New England BioLabs社)を購入し、phageライブラリーを取得した。phageにジスルフィド結合を形成したシステインに挟まれた7アミノ酸を持つ環状ペプチドがランダムに提示される。phageにディスプレイされる環状ペプチド構造を図1に示す。
hCMEC/D3細胞を用いた透過実験は、以下のようにして行った。概要を図2に示す。トランスウエルにhCMEC/D3細胞を1.0×105 cells/wellずつ播種し、37℃にてCO2インキュベーター内で4~6日間培養した。細胞のluminal側にphage library(1.0×1011pfu)を添加し、abluminal側へ透過したphageの力価をプラークカウント法によって測定した。abluminal側へ透過したphageを回収し、ER2738を用いて増殖させ、2回目のスクリーニングに用いた。hCMEC/D3細胞透過実験によるphage libraryスクリーニングを3回繰り返した。それぞれの回における、badal側へのphageの透過の結果を図3に示す。3回目にabluminal側へ透過したphageのプラークからphage DNAのシークエンス解析を行うことで、hCMEC/D3細胞を透過する環状ペプチドのアミノ酸配列を決定した。
その結果、3回目にabluminal側に透過したphage群から、178クローン(1分までにhCMEC/D3細胞を透過したphage群から89クローン、3分~5分までにhCMEC/D3細胞を透過したphage群から89クローン)を同定した。この内、同じ配列を示したものが、それぞれ2クローン同定された。それらのペプチドについて、hCMEC/D3細胞の透過性を確認したところ、アミノ酸配列:SLSHSPQ(配列番号1)をもつ環状ペプチドを提示したphageが顕著な透過性を示すことが確認できたので、そのペプチドを提示するphage(以下、SLS-phageともいう)を以下の実験に使用した。
インビトロのBBBモデルを用いて、SLS-phageの時間依存的な透過量を以下のようにして確認した。トランスウエルにhCMEC/D3細胞を1.0×105 cells/wellずつ播種し、37℃にてCO2インキュベーター内で4~6日間培養した。細胞のluminal側にphage(1.0×1011pfu)を添加し、abluminal側へ透過したphageの力価を定量PCR法によって測定した。コントロールとして、ペプチド非提示phageを用いた。結果を図4に示す。環状ペプチドSLSHSPQを提示するphage(SLS-phage)は1分後からhCMEC/D3細胞を透過し、30分後までの透過量はペプチド非提示phageと比較して3.5倍高かった。
さらに、サルおよびラット型のインビトロBBB透過モデルを用いて、透過量を測定した。サルおよびラットのインビトロBBBキットはファーマコセル(株)から購入したものを用い、透過実験はhCMEC/D3細胞の場合と同様にして行った。結果を図5に示す。SLS-phageは、アストロサイトおよびペリサイトと共培養したインビトロサル由来およびラット由来BBBもそれぞれ7.6倍および28倍で透過した。
このことより、本発明のBBB透過性ペプチドは、種を超えて脳毛細血管内皮細胞を経細胞的に透過することが示された。
本発明のBBB透過ペプチドのhCMEC/D3細胞への取り込みを以下のように確認した。
FAM標識した、SLSHSPQ配列を有する合成環状ペプチドを株式会社スクラムに依頼し、調製した。CSLSHSPQCを含む ACSLSHSPQCGGGSの配列のペプチドを化学合成し、その後、システイン残基間でジスルフィド結合を形成させることにより環状ペプチドを形成させた。次いで、C末端にリジン残基を付加しさらにFAMを結合させることにより、FAM標識環状ペプチドを調製した。
8well スライドグラスにhCMEC/D3細胞を2.5×104 cells/wellずつ播種し、37℃にてCO2インキュベーター内で48時間培養した。細胞にFAM標識した合成環状ペプチドSLSHSPQ(10μM)を添加し、37℃ホットプレート上で10,30,60分間インキュベートした。Cold PBS 500μLで3回洗浄し、4% PFAで10分間固定した。VECTASHIELD(登録商標) Mounting Medium with DAPIを滴下後に、カバーガラスをのせて封入し、共焦点顕微鏡でFAMおよびDAPIを観察した。結果を図6に示す。FAM標識ペプチドが細胞内に取り込まれていることが確認できた。これより、合成環状ペプチドSLSHSPQは、細胞内に内在化することが確認できた。
SLSHSPQ配列を有する本発明のBBB透過性ペプチドが密着結合に与える影響を以下のようにして検討した。トランスウエルにhCMEC/D3細胞を1.0×105 ccells/wellずつ播種し、37℃にてCO2インキュベーター内で4~6日間培養した。細胞に実施例3で調製した標識前の合成環状ペプチドSLSHSPQ(0~100μM)を添加し、37℃にてCO2インキュベーター内でインキュベートした。48時間後まで経時的にTEERを測定することで密着結合への影響を評価した。図7に示すように、環状ペプチドSLSHSPQは、細胞の密着結合を低下させなかった。
SLSHSPQ配列を有する本発明のBBB透過性ペプチドの細胞増殖に与える影響を以下のようにして検討した。96well plateにhCMEC/D3細胞を1×104 cells/wellずつ播種し、37℃にてCO2インキュベーター内で48時間培養した。細胞に合成環状ペプチドSLSHSPQ(0~100μM)を添加し、37℃にてCO2インキュベーター内でインキュベートした。24、48時間後の細胞生存率をcell counting kit-8を用いて算出した。図8に示ように、合成環状ペプチドSLSHSPQは細胞に対して細胞毒性を示さなかった。
SLSHSPQ配列を有する本発明のBBB透過性ペプチドの脳内移行を以下のようにして検討した。SLS-phageおよびペプチド非提示のcontrol Phage(それぞれ、1.0×1011pfu)をICRマウス(雄、7~10週齢)の経静脈へ投与し、1時間後に採血およびPBS灌流を行った。血液は4℃、8000rpmで5分間遠心し、血漿を分離した。摘出した脳をビーズ式ホモジナイザーで4000rpm, 30 秒×4回ホモジネートし、4℃、20000g、30分遠心後に上清を回収した。血漿および脳ホモジネート上清の力価をプラークカウント法によって算出した。結果を図9に示す。インビボマウス静脈内投与1時間後のSLS-phageの脳/血漿比をcontrol phageと比較すると12倍高かった。
以上の結果より、本発明のBBB透過性ペプチドは、脳毛細血管内皮細胞を経細胞的に透過することによってBBB透過能を示すことが示された。
実施例6と同様にしてphageをマウスに経静脈投与し、摘出した脳を切片にした。マウス脳切片を免疫染色し、SLS-phageの存在を確認したとこと、SLS-phageの脳実質移行が確認できた。結果を図10に示す。
hCMEC/D3細胞を用い、本発明のペプチドSLSHSPQを提示するphageのBBB透過性に対する各種リガンド(ビトロネクチン、フィブリノーゲン、RGDペプチド、トランスフェリン)の影響を確認した。
トランスウエルにhCMEC/D3細胞を1.0×105cell/wellずつ播種し、37℃にてCO2インキュベーター内で4~6日間培養した。細胞のluminal側にペプチドSLSHSPQを提示するphage(1.0×1011pfu)を添加し、abluminal側へ透過したphageの力価を定量PCR法によって測定した。コントロールとして、ペプチド非提示phageを用いた。+vitronectin群は20μg/mLとなるようにhuman vitronectin(R&D Systems, Inc.)を添加した。+fibrinogen群は50μg/mLとなるようにhuman fibrinogen(Wako)を添加した。+RGD群は100μMとなるようにRGDペプチド(SIGMA)を添加した。+holo-Tf(トランスフェリン)群は100μMとなるようにholo-Tf(SIGMA)を添加した。その後、abluminal側へ透過したphageの力価を定量PCRによって測定した。結果を図11に示す。
VitronectinによってSLS-phageの透過が阻害された。一方で、fibrinogenおよびRGDはSLS-phageの透過を阻害しなかった。従って、SLS-phageがhCMEC/D3細胞に発現するfibrinogenおよびRGDが認識しないintegrinを介して透過している可能性が示唆された。
Holo-transferrinによってSLS-phageの透過が促進された。Transferrinはエクソソームの分泌を促進する作用がある。また、integrinはエクソソーム分泌を介して細胞から細胞へと輸送されることが報告されている。従って、SLS-phageはエクソソームとしてhCMEC/D3細胞から分泌されることで透過している可能性がある。fibrinogenおよびRGDは、SLSHSPQが結合しないintegrinのサブタイプに結合することで、多胞体におけるエクソソーム合成を促進し、その結果、SLS-phageの細胞外への分泌が亢進し、透過量が増大した可能性がある
トランスウエルにhCMEC/D3細胞を1.0×105cell/wellずつ播種し、37℃にてCO2インキュベーター内で4~6日間培養した。luminal側およびabluminal側の培地をモネンシン含有培地に置換し、さらに18時間培養した。細胞のluminal側にプチドSLSHSPQを提示するphage(1.0×1011pfu)を添加し、abluminal側へ透過したphageの力価を定量PCR法によって測定した。結果を図12に示す。モネンシン濃度依存的にSLS-phageのhCMEC/D3細胞透過量が増大した。従って、SLS-phageはエクソソームを介してhCMEC/D3細胞内からabluminal側へと分泌されていることが示唆された。
hCMEC/D3細胞を用い、本発明のペプチドSLSHSPQを提示するphage(SLS-phage)の細胞内への取り込みを確認した。
8well スライドグラスにhCMEC/D3細胞を2.5×104 cells/wellずつ播種し、37℃にてCO2インキュベーター内で48時間培養した。培養した細胞にSLS-phage(1.0×104 pfu)を添加し、37℃ホットプレート上で10分間インキュベートした。Cold PBS 500μLで3回洗浄し、4% PFAで10分間固定した。その後、0.1% triton-X100/PBS 100 μLで10分間処理し、1% BSA/PBS 250 μL加え、室温で1時間ブロッキング後、1% Anti-m13+fd bacteriphage coat proteins antibody/Can Get Signal immunostain B 100 μLを添加し、4℃で一昼夜インキュベーションした。PBS-T 500 μLで3回洗浄し、0.1% Goat Anti-Rabbit IgG H and L(Alexa Fluor 568)(abcam、ab175695)/Can Get Signal immunostain B 100 μLを添加し、室温で1 時間インキュベーションした。再度、PBS-T 500 μLで3回洗浄し、VECTASHIELD(登録商標) Mounting Medium with DAPIを滴下後に、カバーガラスをのせて封入し、共焦点顕微鏡でSLS-phageおよびDAPIを観察した。結果を図13に示す。SLS-phageが細胞内に取り込まれていることが確認できた。これより、本発明のペプチドを持つ非常に大きな分子であるM13phage(SLS-phage)は、細胞内に内在化することが確認できた。
また、SLS-phageに加え、さらに、SLSペプチド(10 μM)、またはマクロピノサイトーシス阻害剤であるEIPA(100μM、CYAMANから購入)を添加し、37℃で培養して検討した。SLS-phageの取り込み阻害の結果を図14に示す。SLS-phageの細胞内内在化にはマクロピノサイトーシスが関与していることが判った。
薄膜水和法によって、組成:COATSOME NC-50/Cholesterol/DIO (210 nmol : 90 nmol : 5 nmol)からなる蛍光標識リポソームを合成し、ステアリル化SLSペプチド(ACSLSHSPQ-stearic acid、スクラムから購入)を混合することにより、SLS-ペプチドが結合したリポソームを調製した。
トランスウエルにhCMEC/D3細胞を1.0×105 cells/wellずつ播種し、37℃にてCO2インキュベーター内で4~6日間培養した。細胞のluminal側にSLS-リポソーム(50 nmol)を添加し、abluminal側へ透過したSLS-リポソームの量を蛍光プレートリーダーよって測定した。コントロールとして、SLSペプチドを結合していないリポソームを用いた。結果を図15Aに示す。SLSペプチドを結合させたリポソームはコントロールに比べて有意に透過が促進していた。なお、コントロールでも透過が観察されたのは、細胞の密着結合の程度が低いためその間隙から透過したと考えられる。
さらに、サルおよびラット型のインビトロBBB透過モデルを用いて、SLS-リポソームの透過量を測定した。サルおよびラットのインビトロBBBキットはファーマコセル(株)から購入したものを用い、透過実験はhCMEC/D3細胞の場合と同様にして行った。結果を図15B及びCに示す。SLS-リポソームは、アストロサイトおよびペリサイトと共培養したインビトロサル由来およびラット由来BBBもそれぞれ有意に透過が促進していた。
hCMEC/D3細胞を用い、実施例11で調製した蛍光標識SLS-リポソームの細胞内への取り込みを確認した。
24well プレートにhCMEC/D3細胞を5.0×104 cells/wellずつ播種し、37℃にてCO2インキュベーター内で48時間培養した。培養した細胞にSLS-リポソームまたはコントロールとしてSLSが結合していないリポソームを添加し(添加量:30 nmol)、37℃ホットプレート上で5、10、30分間インキュベートした。その後、hCMEC/D3細胞内に取り込まれたリポソームを1% triton-X100/PBSで抽出し、蛍光プレートリーダーで測定した。結果を図16に示す。SLSペプチドをリポソームに結合させることで、BBBを構成する細胞へのリポソームの取り込みが顕著に促進されることが判った。
SLS-リポソームの脳内移行を以下のようにして検討した。実施例11で調製したSLS-リポソームおよびSLSが結合されていないリポソーム(それぞれ、50 nmol)をICRマウス(雄、7~10週齢)の経静脈へ投与し、55分後に採血、60分後にPBS灌流を行い、脳を摘出した。血液は4℃、8000rpmで5分間遠心し、血漿を分離した。摘出した脳をビーズ式ホモジナイザーで4000rpm, 30 秒×4回ホモジネートし、4℃、20000g、30分遠心後に上清を回収した。血漿および脳ホモジネート上清中の蛍光量を測定した。結果を図17に示す。脳内からはSLSリポソームが検出されたが、対照としたリポソームは検出されなかった。また、血漿中のSLS-リポソームと対照リポソームの濃度は同程度であった。このことは、SLSペプチドが、リポソーム等のナノキャリの脳移行性を促進していることを示している。
Claims (23)
- 以下のアミノ酸配列:
(i)SLSHSPQ(配列番号1)からなる配列を含む血液脳関門透過性ペプチド。 - 少なくとも一つの非天然アミノ酸を含む請求項1に記載の血液脳関門透過性ペプチド。
- 前記ペプチドが環状ペプチドである請求項1または2に記載の血液脳関門透過性ペプチド。
- 前記ペプチドが下記の(a)~(f)のいずれかである、請求項1に記載の血液脳関門透過性ペプチド、
(a)アミノ酸配列:SLSHSPQ(配列番号1)からなるペプチド、
(b)SLSHSPQ(配列番号1)のアミノ酸配列のC末端および/またはN末端に1~5個の任意のアミノ酸を有するペプチド、
(c)アミノ酸配列:CSLSHSPQC(配列番号2)からなるペプチドであって、該配列中のシステイン残基同士がジスルフィド結合しているペプチド、
(d)CSLSHSPQC(配列番号2)で示されるアミノ酸配列のC末端および/またはN末端に1~10個の任意のアミノ酸を有するペプチドであって、該配列中のシステイン残基同士がジスルフィド結合しているペプチド、
(e)下記式(1):
(f)SLSHSPQ(配列番号1)のアミノ酸配列のC末端およびN末端に1~15個の任意のアミノ酸を有するペプチドであって、該配列の両端に存在する任意のアミノ酸同士が架橋されているペプチド。 - 少なくとも一つの非天然アミノ酸を含む請求項4に記載の血液脳関門透過性ペプチド。
- 請求項1~5のいずれかに記載のペプチドを含む脳内送達用キャリア。
- 前記脳内送達用キャリアが、請求項1~5のいずれかに記載のペプチドと、リポソーム、ナノキャリア、エキソソーム、ファージ、ポリロタキサン、シクロデキストリン、マイクロカプセル、およびミセルからなる群より選ばれる薬物送達用キャリア分子とからなる、請求項6に記載の脳内送達用キャリア。
- トランスフェリン、フィブリノーゲン、およびRGD配列を含むペプチドからなる群より選ばれる物質とともに使用されることを特徴とする請求項6または7に記載の脳内伝達用キャリア。
- さらに、トランスフェリン、フィブリノーゲン、およびRGD配列を含むペプチドからなる群より選ばれる物質を含む請求項6または7に記載の脳内伝達用キャリア。
- 請求項6~9のいずれか一つに記載の脳内送達用キャリアと脳内に送達される薬物とからなる組成物。
- 前記薬物が、脳内で薬理学的作用を示す分子または脳内イメージング分子である請求項10に記載の組成物。
- 前記脳内で薬理学的作用を示す分子が、低分子化合物、ポリペプチド、オリゴペプチド、タンパク質、または核酸である請求項11に記載の組成物。
- 脳疾患の予防および/または治療用の医薬組成物であって、請求項10~12のいずれか一つに記載の組成物および薬理学的に許容され得る添加剤とからなる医薬組成物。
- 請求項1~5のいずれか一つに記載のペプチド、および該ペプチドとともに血液脳関門を透過する分子からなる複合体。
- トランスフェリン、フィブリノーゲン、およびRGD配列を含むペプチドからなる群より選ばれる物質とともに使用されることを特徴とする請求項14に記載の複合体。
- さらに、トランスフェリン、フィブリノーゲン、およびRGD配列を含むペプチドからなる群より選ばれる物質を含む請求項14に記載の複合体。
- 前記分子が、脳内で薬理学的作用を示す分子または脳内イメージング分子である請求項14~16のいずれか一つに記載の複合体。
- 前記分子が、低分子化合物、ポリペプチド、オリゴペプチド、タンパク質、または核酸である請求項14~16のいずれか一つに記載の複合体。
- 脳疾患の予防および/または治療用の医薬組成物であって、請求項14~18のいずれか一つに記載の複合体および薬理学的に許容され得る添加剤とからなる医薬組成物。
- 請求項1~5のいずれかに記載のペプチドおよび該ペプチドの存在下で血液脳関門を透過する少なくとも一つの分子との混合物、および薬理学的に許容され得る添加剤とからなる組成物。
- 前記混合物がさらに、トランスフェリン、フィブリノーゲン、およびRGD配列を含むペプチドからなる群より選ばれる物質を含む請求項20に記載の組成物。
- 前記分子が、低分子化合物、ポリペプチド、オリゴペプチド、タンパク質、核酸、リポソーム、ナノキャリア、エキソソーム、ファージ、ポリロタキサンおよび脳内イメージング分子からなる群より選ばれる1またはそれ以上の分子である請求項20または21に記載の組成物。
- 請求項20~22のいずれか一つに記載の組成物からなる脳疾患の予防および/または治療用の医薬組成物。
