以下及び実施例においてより詳細に説明されるとおり、種々の組成物及び成分は、標的タンパク質を分泌する特定の細胞を調製する際に有用であり、多成分複合体(multi-part complex)の一部として細胞の形質膜外側上に標的タンパク質を捕捉するための方法において有用である。多成分複合体は、細胞結合アンカーとリンカーとの間の「第1の」複合体、及び、結合リンカーと標的タンパク質との間に形成される「第2の」複合体を含む。更なる他の複合体は、細胞に結合した多成分複合体の一部の場合がある。例えば、以下に説明するとおり、標的が抗体である場合、「第3の」複合体は、結合した標的タンパク質とその抗原との間で形成され得る。また、「第4の」複合体は、結合した抗原と更なる標的との間に形成され得る。本明細書中の教示を利用することによって、細胞を、それが分泌する標的タンパク質(例えば、mAb、IgG、IgA、又は、ヒトIg)に物理的に付着させることが可能である。その標的は、適切な特異性のリンカーが第1及び第2の複合体を形成することによってアンカーを介して細胞に結合される場合にのみ、標的を分泌する細胞に付着するであろう。
上記のこれらの核酸分子、ベクター及び細胞型のいずれか又は全ては、選択された分泌標的タンパク質の分析及び/又は生成又は産生のため、所望の特徴を有する細胞(例えば、標的タンパク質高産生体)の生成及び/又は同定のため、並びに、ハイブリドーマのハイスループットスクリーニング及び評価のためのいずれかの数の方法において使用される場合がある。これらの核酸によってコードされる遺伝子の安定な発現又は一過性の発現を生じる条件下で核酸を細胞に導入するための多様な方法は、一般的に使用されており、当業者に周知である。本明細書に説明する組成物は、以下の方法その他の類似の方法において使用される場合がある。
(i)B細胞の細胞系譜の細胞を、選択されたリンカーとの第1の複合体を形成するために設計されたアンカーをその形質膜外側表面上に発現した不死化細胞と融合させることによって調製された、ハイブリドーマ細胞の集団を、
(ii)アンカーとの第1の複合体、及び、単一のハイブリドーマ細胞によって分泌される単一のモノクローナル抗体との第2の複合体を形成するために設計されたリンカーであって、モノクローナル抗体がアンカーによって認識されない、リンカーと接触させる工程であって、
前記接触は、第1の複合体においてアンカーと結合することによって、リンカーがハイブリドーマ細胞の形質膜外側に付着することを可能にする、工程、
(b)(a)の細胞を、リンカーがハイブリドーマ細胞によって分泌されるモノクローナル抗体と第2の複合体を形成し、モノクローナル抗体が、付着した第1及び第2の複合体を介してそれを分泌するハイブリドーマ細胞に結合する条件下でリンカーの存在下で維持する工程、
(c)ハイブリドーマ細胞によって分泌され、アンカー-リンカー-モノクローナル抗体によって形成された細胞結合多重免疫複合体によって結合されたモノクローナル抗体の存在、抗原特異性、抗原結合親和性、力価、量、又は、生物学的活性を同定する工程と、
(d)所望の抗原特異性、抗原結合親和性、力価、量、又は、生物学的活性を有するモノクローナル抗体を分泌するハイブリドーマ細胞の集団を単離する工程、及び、

本明細書に提供される教示を使用して構築物において生成できる変形の数を考慮すると、これらの組成物を使用する多くの他の方法は、迅速かつ複雑な標的同定又は他の使用のために使用できる。例えば、組成物及び方法は、例えば、品質管理のために、細胞によって分泌されるいずれかの分子(例えば、タンパク質)を追跡するための方法において使用される場合がある。別の実施形態では、そのハイブリドーマ細胞に結合したモノクローナル抗体は、その抗原への結合について評価される場合があるか、又は、T細胞活性化のような免疫複合体に組み込まれた場合にのみ発現する場合がある活性について評価できる。
以下の実施例は、リンカー分子と接触した場合に、細胞によって分泌される標的分子をそれらの形質膜外側上に捕捉する哺乳動物細胞の生成の証拠を提供する。また、これらの教示による特定の融合パートナー細胞及びハイブリドーマも開示される。以下の実施例は、本明細書に説明する方法及び組成物の特定の実施形態を開示する。これらの実施例は、本明細書に提供される教示の結果として明らかになるいずれかの及び全ての変形を包含すると解釈されるべきである。
アンカーをコードする核酸分子は、下記の表中の以下の要素を用いて構築される。簡潔には、マウス抗ウサギ抗体重鎖(VH)の可変ドメインをコードする核酸配列を、(Gly4Ser)3をコードするスペーサーを介して抗体軽鎖(VL)の可変ドメインに連結し、単鎖可変断片(scFv)をコードする配列を生じた。更に、scFvをコードする2個の核酸配列を、上記のスペーサーによって一緒に連結し、タンデムscFvコード配列を生じた。核酸分子中の更なる配列は、細胞表面上でのscFvの発現を確実にするためのκ軽鎖免疫グロブリンタンパク質由来のN末端リーダー配列(AKA、シグナルペプチド)をコードする配列を含んでいた。配列は、NCBI参照配列:XP_003753129.1(Igκ鎖V-III領域MOPC 63アイソフォームX2;種ドブネズミ(Rattus norvegicus))の下でNCBIデータベースに提供される。核酸分子又は構築物中の更なる配列は、エピトープタグ又はプロテアーゼタグ配列(即ち、c-Myc)、及び、血小板由来成長因子受容体(PDGF受容体、PDGFR)膜貫通(TM)ドメインをコードする配列である。58.92%のシトシン及びグアニンを含むヌクレオチド1781個を有する核酸分子/構築物を、マウス細胞発現について更に最適化した。アンカーの実施形態をコードする核酸分子/構築物の配列は、配列番号1として提供される。対応するタンパク質配列は、配列番号2である。核酸分子を、説明したプロトコルに従って、レトロウイルス導入プラスミドpMSCV及びレンチウイルス導入プラスミドpLION IIに連結した。13、14、15
IL-6の異所性発現によるシグナル伝達は、hTERTも発現するハイブリドーマによるヒト免疫グロブリンの安定な分泌に不可欠である。従って、ハイブリドーマプロトコルの最適化及び標準化は、バルク細胞集団における個々の細胞のIL-6刺激活性がリアルタイムで効率的に評価可能であることを必要とする。しかし、ポリクローン集団における個々の細胞によるIL-6の分泌速度を測定することは困難である。LCX細胞株は、異所性hTERTを発現し、IL-6様シグナルを形質導入する分子の表面発現を評価することによって監視することができるIL-6シグナル伝達活性を提供するために作製された。親K6H6/B5細胞を、レトロウイルス形質導入によってgp130 ΔYYタンパク質(IL-6受容体ファミリーの構成的に活性なメンバー)に加えてhTERTを発現するために改変し、RT-PCRを行って、説明されるとおりLCX細胞における異所性hTERT遺伝子の発現を確認した(図14を参照されたい)。13
LCX細胞株を使用して、初代ヒトB細胞との融合後にヒトIgを安定的に分泌するハイブリドーマを作製した。B5-6T融合パートナー細胞株の使用で標準化されたプロトコルに従って、本発明者らは、ポリオウイルス(PV)(1~3型)に対して免疫性のある個体由来のB細胞にLCX細胞株を説明したプロトコルに従って融合させた。20これは、ポリオウイルスと免疫反応性のヒトIgGの発現について評価された、ヒトを分泌するハイブリドーマの集団を作製した。ハイブリドーマ上清をELISAによって評価し、PV結合可能なヒトmAbを安定的に分泌するハイブリドーマの同定及びサブクローニングを可能にした。PV反応性mAbを、表2に列挙されるマイクロ中和試験を用いて、インビトロでの中和について試験した。多くのmAbsは、高い中性化能(LX-2D6、LX-5G5、LX-4A4、LX-3E9、LX-1A7、LX-10C6及びLX-2E1など)を有することが確認され、4個のmAbsは、2株以上を中性化することが確認された。
Igリンカータンパク質と相互作用することができるアンカータンパク質を細胞の形質膜外側上に発現する哺乳動物細胞株を作製し、確立するために、以下の実験を行った。現在の実験環境では、アンカーは、実施例1に説明されるとおりIgリンカーとしてのウサギ抗ヒトIgG(RAH)に結合し、第1の免疫複合体を形成するために設計された、タンデムマウス抗ウサギscFv分子部分を含んでいた。LCX及びB5-6T細胞を培養し、説明のとおり維持した。21LCX細胞を培養し、10%ウシ胎仔血清を含むATCC処方RPMI-1640培地中で、37℃で5% CO2を含む大気中で維持した。
アンカーは、2つの方法のうちの1つでリンカーと相互作用することができる。一実施形態では、アンカーは、リンカーによって認識又は結合できる。例えば、アンカーは、抗原(例えば、c-mycタグ又はscFv配列自体(本実施形態におけるとおり))を含む場合があり、これは、リンカータンパク質(例えば、及び、mycタグ又はscFv分子部分に特異的な抗体)によって認識される。例えば、細胞を、c-mycタグ配列に特異的なヤギ抗体、又は、マウスscFv抗原に特異的なヤギ抗体と接触させ、洗浄し、更にAPC標識抗ヤギ二次抗体とインキュベートし、次いでフローサイトメトリーによって分析し、リンカーとして使用してもよい抗体によって認識されるc-mycタグ又はマウスIgエピトープの能力を実証する。あるいは、アンカーがそれ自体、リンカータンパク質上のエピトープに結合することができる抗原結合ドメインを含む場合がある。
本明細書に説明する組成物及び方法の新規かつ有用な能力は、それらが、細胞によって分泌されるタンパク質をその細胞の表面に誘導的に付着可能にすることである。細胞に付着されている間、タンパク質の構造的及び/又は機能的特徴は、タンパク質が表面に固定化されることを必要とするアッセイを使用して評価される。分泌タンパク質がmAbである本実施例において、このようなアッセイは、抗原結合アッセイ、mAbグリコシル化又はアイソタイプについての試験、リンパ球刺激又は補体依存性細胞傷害性などの免疫複合体媒介機能、及び、触媒活性を含むが、これらに限定されない。
ヒトIgG及び抗原によって形成される第3の免疫複合体の存在を確立するために、更なる実験を行った。上記のとおり、8C5 mAbは、RABV GPに結合する。本発明者らは、B5-6T、8C5及び8C5 BGS細胞を過剰のRAHの存在下で一晩培養し、次いで細胞を洗浄し、追加のRAH、ビオチン化RABV GP抗原、及び、検出分子としてのAPC標識ストレプトアビジンを添加した。図13に示される結果は、B5-6T融合パートナー細胞及び8C5ハイブリドーマに比べて、8C5 BGSハイブリドーマのサンプルにおいて、抗原陽性細胞のより高いパーセンテージ及び抗原のより強い結合を明らかにし、これは、第1、第2及び第3の免疫複合体の特異的な形成に成功したことを示す。この実験は、細胞によって分泌されるmAbを含み、膜結合アンカータンパク質及びIgリンカータンパク質からなる架橋を介して細胞の形質膜外側に付着される有用かつ新規な細胞を含む免疫複合体の存在を確立し、mAbは、その抗原に結合し、抗原は、更に検出分子に結合する。本明細書に説明する方法及び組成物の使用によって、抗原に結合できるmAbを分泌する細胞は、容易に同定され、単離される。
本明細書に説明する方法及び組成物によって可能にされるアンカータンパク質を発現するハイブリドーマは、異なる方法によって確立できる。一実施例では、アンカータンパク質の発現は、既存のmAb分泌ハイブリドーマのレトロウイルス遺伝子導入によって誘導される。あるいは、アンカーを発現する融合パートナー細胞をB細胞に融合させて、アンカータンパク質及びB細胞に由来するIg遺伝子によってコードされるmAbの両方を発現するハイブリドーマを産生する。
本発明の融合パートナー細胞株は、モノクローナル抗体を発現するハイブリドーマのライブラリーの作製及びスクリーニングのために使用できる。原則として、この方法は、いずれかの哺乳動物種由来のB細胞の細胞系譜の細胞と共に使用される場合があるが、B5-6T BGS及びLCX BGS細胞株は、ヒトmAbを安定的に分泌するハイブリドーマを作製するために適する。B5-6T細胞株のような細胞を用いてハイブリドーマを作製する方法は、ヒトB細胞22、23、野生型マウス由来のマウスB細胞24、ヒト免疫グロブリン遺伝子配列に遺伝子導入されたマウス25、及び、ウサギ26を含む多くの様々な種由来のB細胞と共に使用するための最新技術として周知である。
ハイブリドーマ法によるmAbクローニングの現在の技術水準における重要な弱点は、mAbを分泌するハイブリドーマがより速く増殖する非分泌ハイブリドーマ細胞によって圧倒される傾向があるため、ハイブリドーマライブラリーをポリクローナル培養物中で維持することができないことである。その結果、ハイブリドーマライブラリーは、典型的には、スクリーニング後に廃棄される。これは、各ハイブリドーマについて実施可能な試験の数に実際的な制限を課し、潜在的に価値のあるB細胞サンプルを失う。
抗体によって発揮される多くの機能が免疫複合体の形成に依存することは、周知である。例えば、複数のIgG抗体からなる免疫複合体は、古典的補体経路を活性化し、T細胞を活性化し、Fc受容体を介して、マクロファージ、NK細胞、及び、好中球におけるエフェクター応答を活性化することができる。28、16、29
本明細書に説明のとおり調製されたmAbを分泌するハイブリドーマは、連結免疫複合体において、mAbをそれらの形質膜外側上に捕捉するが、ここで、前記連結免疫複合体は、アンカーが細胞表面から切断された後に残存するアビディティー効果によって安定化され、該アビディティー効果は、結合した抗原の特徴付けに使用することができる。以前に説明したMycタグの代わりにtevプロテアーゼタグを含むアンカー分子は、あまり特徴付けられていない抗原で、複数の抗原を含む抗原混合物内に存在することが知られている抗原に特異的なmAbを分泌するハイブリドーマ細胞において発現される。抗原混合物を最初にビオチン化する。次いで、アンカー発現細胞株を、過剰のIgリンカーと共にインキュベートし、これは、分泌されたmAbをハイブリドーマ細胞表面、ビオチン化抗原混合物、及び、ストレプトアビジンAlexa 488に付着させる。抗原が細胞表面に結合したことを視覚的に確認した後、細胞を培養培地から取り出し、AcTEVプロテアーゼ(Thermo Fisher)による切断に好ましい条件下で処理する。切断反応に続いて、細胞を遠心分離により上清から除去し、所望により、AcTEVプロテアーゼをサイズ排除クロマトグラフィーにより除去する。得られたサンプルを分析して、質量分析又は結合した抗原の特徴付けに適切な他の方法によって、mAbに結合した抗原の組成を同定する。
293T又は293T OCMS細胞を、24ウェルプレート中の丸いCorning(商標)BioCoat(商標)12mm #1German Glass Coverslips(Corning、カタログ番号354087)上に細胞5×104個/ウェルで蒔き、5%C02インキュベーター中、37℃でインキュベートした。一晩インキュベートした後、X-treme Gene 9 DNA遺伝子トランスフェクション試薬を使用して、A12重鎖及び軽鎖DNAの各々0.5μg又は1μgで一過性トランスフェクションを行った。24時間後、各ウェルから培地を除去し、1μg/mlのウサギ抗ヒトIgG Fc特異的mAb(Abcam H169-1-5)を有する10% FBSを有する500μlのDMEMを加え、一晩インキュベートした。翌日(トランスフェクションの48時間後)、細胞を、1% BSAを含むPBS(PBS-BSA)で3回洗浄し、PBS-BSA中に1:200希釈した、抗ヒトIgG F(ab)2断片特異的(Jackson Immuno Research、カタログ番号109-546-097)のAlexa Fluor 488結合ヤギF(ab)2断片とともにCO2インキュベーター中、37℃で1時間インキュベートした。1時間のインキュベーション後、細胞をPBS-BSAで3回洗浄し、室温で15分間PBS中の4%パラホルムアルデヒドで固定した。固定後、細胞をPBSで1回、dH2Oで1回洗浄し、次いでカバーガラスにDAPIを含むProLong(登録商標)Gold Antifade試薬(P36935、Thermo Fisher Scientific)をマウントし、63x/1.3 NAオイル対物レンズを有するC2+ニコン共焦点顕微鏡で画像化し、画像をImageJソフトウェア(https://imagej.nih.gov/ij/)で分析した。図19を参照されたい。
LCX BGS細胞株を用いて、初代ヒトB細胞との融合後にヒトIgGを安定的に分泌するハイブリドーマを作製した。B5-6T融合パートナー細胞株の使用により標準化されたプロトコルに従って、本発明者らは、LCX BGS細胞株を、ポリオウイルス(PV)(1~3型)に対して免疫性の2人の個体由来のB細胞に、説明されたプロトコルに従って融合させた。20実験は、全PVビリオン、非標識PVビリオン、生PVビリオン又はUV-C不活化PVビリオン(セービンワクチン株)を用いて行われる41。これは、3型ポリオウイルスと免疫反応性のIgGの発現について評価されたヒトIgGを分泌するハイブリドーマの集団を生成した。ハイブリドーマ上清をELISAによって評価し、PV結合可能なヒトmAbを安定的に分泌するハイブリドーマの同定及びサブクローニングを可能にした。本発明者らは、マイクロ中和アッセイにおいて7種のヒトmAbの中和力価(以下の表3)を試験し、100 TCID50のセービン3及びサウケット3 PV株でのチャレンジに対して細胞培養物の50%を保護する希釈の逆数として結果を表にした。全てのmAbは、両方の株に対して3型PV中和活性を有していた。
上記のとおり、以下は、普遍的な抗ウイルスmAbスクリーニング方法の例を提供する。本明細書に提供される特定の実施例において、狂犬病糖タンパク質GPに特異的なヒトmAbを分泌する8C5、及び、3型ポリオウイルス(PV)に結合するヒトmAbを分泌する4G4の2種のハイブリドーマが示される。ハイブリドーマを洗浄し、ヒトIgG(H+L)(Rockland、カタログ番号809-4102)及び3型PV又は狂犬病GPに特異的な過剰のウサギFAb断片を含有する細胞培養培地中で一晩インキュベートした。それらをPBS+BSAで洗浄し、PBS-BSA中で1:200で、ヤギ抗ヒトIgG Fc特異的(Jackson Immuno Research、カタログ番号109-546-170)Alexa Fluor 488結合F(ab)2断片と共に1時間インキュベートした。1時間後、細胞をPBS-BSAで3回洗浄し、PBS中4%パラホルムアルデヒドで15分間室温にて固定した。固定後、細胞をPBSで1回、dH2Oで1回洗浄し、次いで、カバーガラスに、DAPI(P36935、Thermo Fisher Scientific)を含むProLong(登録商標)Gold Antifade試薬(P36935、Thermo Fisher Scientific)をマウントし、63x/1.3 NAオイル対物レンズを有するC2+ニコン共焦点顕微鏡で画像化し、画像をImageJソフトウェア(https://imagej.nih.gov/ij/)で分析した。
治療用mAbは、それらが臨床開発のために指定できる前に、インビトロでウイルス中和活性を示す必要がある。第1のこのような試験は、典型的には、培養細胞株をウイルスに感染させ、細胞変性作用を予防するmAbの能力を測定するマイクロ中和試験である。20、43OCMS mAbクローニング方法の利点の1つは、組換えmAb発現を必要とせずに、マイクロ中和アッセイで試験することができる全長IgGを安定的に産生することである。普遍的な抗ウイルスmAbクローニングアッセイの必須のコンパニオンは、ウイルス中和を試験するために使用できる細胞株のパネルである。OCMSハイブリドーマ自体がウイルス中和を試験するために使用されるならば、有利であろう。
ウイルスの細胞への感染能は、多数の因子、とりわけ、細胞表面上のウイルスに対する受容体の存在、及び、感染した細胞のサイトカイン応答に起因する44。EV71誘導殺傷に対するLCX OCMS細胞株の感受性は、それらがヒトPV受容体(hPVR、CD155)を発現した場合、それらも殺傷されることを示唆する45。本発明者らは、レトロウイルス遺伝子形質導入を用いてLCX OCMS細胞中でCD155を発現させ、2logの範囲内のCD155発現の6種の細胞クローンのパネルを単離する。本発明者らは、上記のOperettaアッセイを用いてPV感染についてこれらを試験し、CD155発現及びPV用量(50%組織培養感染用量、TCID50)の最適レベルを同定する。次いで本発明者らは、既知の中和能力を有するヒトPV mAbのパネル(9H2、8H4、7E2、及び、対照mAb、6A)22、20、46を、先に説明したとおり、1mg/mlストックからの希釈系列において試験し、Karber式を使用して中和力価を計算する46、47。
本明細書で同定されるヒトmAbは、優れた抗ウイルス治療薬候補である可能性が高い。にもかかわらず、mAbの安定性、発現レベル、結合動力学、又は、結合特異性を改善するために、ヒトmAbを改変することが重要な場合がある。OCMSプラットフォームのハイスループットスクリーニング能力を使用して、本発明者らは、クローニングされたmAbの指向性進化、即ち、突然変異誘発、続いて改善されたmAb変異体の選択を行う。

Claims (14)
- ハイブリドーマ細胞の不均一な集団の中のモノクローナル抗体を分泌するハイブリドーマ細胞を同定する方法であって、
(a)アンカーをその形質膜外側表面上に発現した不死化細胞をB細胞の細胞系譜の細胞と融合させることによって調製された、ハイブリドーマ細胞の不均一な集団を提供する工程であって、前記アンカーは、i)選択されたリンカーに結合して第1の複合体を形成することができる抗体又はその断片を含む細胞外領域、及びii)アンカーを細胞表面に固定する膜貫通アミノ酸配列、を含み、前記アンカーは、前記モノクローナル抗体に結合できない、工程と、
(b)(a)の細胞を十分な量の前記リンカーと接触する工程であって、前記リンカーは、前記モノクローナル抗体の保存ドメインと結合して第2の複合体を形成する可変領域を含む可溶性の抗体又は抗体断片であり、前記接触は、前記第1及び第2の複合体を形成すると前記モノクローナル抗体がそれを分泌するハイブリドーマ細胞の表面に結合することを可能にする、工程と、
(c)前記ハイブリドーマ細胞によって分泌され且つアンカー-リンカー-モノクローナル抗体によって形成される前記第1の及び第2の免疫複合体によってそれに結合する、前記モノクローナル抗体の、存在、抗原特異性、抗原結合親和性、力価、量、又は、生物学的活性を同定する工程と、
を含む、方法。 - (b)は、前記細胞を過剰濃度の前記リンカーと接触させることを更に含み、前記過剰は、過剰な非結合リンカーを結合することによって、前記モノクローナル抗体とこれを分泌しない他の細胞との結合に比べて、ハイブリドーマの細胞表面上の第1の複合体のリンカーとそのハイブリドーマ細胞によって分泌されるモノクローナル抗体との間の結合の特異性を増加させる、請求項1に記載の方法。
- 第3の複合体において前記モノクローナル抗体が特異的に結合する、検出可能に標識された抗原に前記ハイブリドーマ細胞を接触させることと、第1の複合体アンカー-リンカーによって形成される多成分複合体に結合する細胞を形成することとを更に含み、前記リンカーは、前記抗原に結合された前記分泌されたモノクローナル抗体にも結合し、そして及び工程(c)は、前記細胞結合多成分複合体に会合する前記検出可能な標識を同定又は
定量することによって、選択された抗原に特異的なモノクローナル抗体を分泌するハイブリドーマ細胞を同定することを含む、請求項1~2のいずれか1項に記載の方法。 - 前記モノクローナル抗体はヒト抗体であり、前記アンカー分子は選択された非ヒト由来リンカーに特異的なscFvを含み、前記リンカーは抗ヒトIg抗体又は抗ヒトIg抗体の抗体結合断片である、請求項1~3のいずれか1項に記載の方法。
- 抗体又は抗体断片を含む選択された可溶性のリンカーと第1の複合体を形成するために設計されたアンカーをその形質膜外側表面上に発現するハイブリドーマ細胞であって、前記リンカーは、前記アンカーによって認識されない前記モノクローナル抗体と第2の複合体を形成するために設計されている、細胞であって、
前記アンカーが、
i.前記リンカー上の配列に特異的に結合する単鎖抗体可変領域(scFv)、各々が前記リンカー上の配列に結合するために設計されている2~6個のscFvから形成されるタンデムscFv、細胞表面上で発現されるときに1個又は2個の遊離Fv領域を有するFabであって前記リンカー上の配列に特異的に結合するFab、細胞表面上で発現されるときに1個又は2個の遊離Fv領域を有する抗体であって前記リンカー上の配列に特異的に結合する抗体、前記リンカー上の配列に特異的に結合する単一ドメイン抗体、又は前記リンカー上の配列に特異的に結合するラクダ抗体を含む細胞外領域ポリペプチドであって、前記リンカーに結合する細胞外領域ポリペプチド、及び
ii.内在性膜タンパク質に由来する膜貫通アミノ酸配列
を含む、ハイブリドーマ細胞。 - 前記ハイブリドーマ細胞の形質膜外側表面上が、前記アンカー及び前記リンカーを含む第1の免疫複合体である、請求項5に記載のハイブリドーマ細胞。
- 前記リンカーの抗体断片は、Fv断片、Fab断片、Fv若しくは単鎖Fv、又は、単一ドメイン抗体、およびそれらの組換えバージョンである、請求項5に記載のハイブリドーマ細胞。
- 前記ハイブリドーマ細胞の形質膜外側表面上が、前記リンカーに結合した前記アンカーを含む多重免疫複合体であり、前記リンカーは前記モノクローナル抗体に結合する、請求項5に記載のハイブリドーマ細胞。
- アンカーをその形質膜外側表面上に発現している不死化細胞とB細胞の細胞系譜の細胞を融合させることを含む、請求項5に記載のハイブリドーマ細胞を作製する方法であって、前記アンカーは、i)選択されたリンカーに結合して第1の複合体を形成することができる抗体又はその断片を含む細胞外領域、及びii)内在性膜タンパク質に由来する膜貫通アミノ酸配列、を含み、前記アンカーは、前記ハイブリドーマ細胞から分泌される前記モノクローナル抗体に結合せず、前記リンカーは、前記ハイブリドーマ細胞によって分泌される前記モノクローナル抗体に結合する抗体又は抗体断片を含む、方法。
- 請求項9に記載の方法であって、
(a)前記アンカーが、ポリペプチドタグを含むか;または、
(b)前記抗体断片は、Fv断片、Fab断片、Fv若しくは単鎖Fv、又は単一ドメイン抗体、およびそれらの組換えバージョンであるか;または、
(c)作製された前記ハイブリドーマ細胞が、前記B細胞の細胞系譜の細胞内に由来する核酸配列から発現されるモノクローナル抗体を分泌し、前記不死化細胞によって提供される核酸配列からの前記アンカータンパク質を発現する、
前記方法。 - ATCC受託番号PTA-124062によって指定される、細胞株LCX-BGS株03.06.17。
- B細胞の細胞系譜の細胞を請求項11に記載の細胞株と融合させることによって製造されたハイブリドーマ細胞株。
- ウイルス抗原に結合する、ハイブリドーマ細胞によって分泌されたモノクローナル抗体を同定するための方法であって、
(a)ハイブリドーマ細胞の不均一な集団を提供する工程であって、各ハイブリドーマ細胞は、選択された可溶性のリンカーと第1の複合体を形成するために設計されたアンカーをその形質膜外側上に含み、ここで、前記アンカーは、前記ハイブリドーマ細胞から分泌された前記モノクローナル抗体に結合することができず、且つ、前記リンカーは、前記ハイブリドーマ細胞によって分泌される前記モノクローナル抗体と第2の複合体を形成することができる抗体又はその断片を含み、
ここで、前記アンカーは、
i.前記リンカー上の配列に特異的に結合する単鎖抗体可変領域(scFv)、各々が前記リンカー上の配列に結合するために設計されている2~6個のscFvから形成されるタンデムscFv、細胞表面上で発現されるときに1個又は2個の遊離Fv領域を有するFabであって前記リンカー上の配列に特異的に結合するFab、細胞表面上で発現されるときに1個又は2個の遊離Fv領域を有する抗体であって前記リンカー上の配列に特異的に結合する抗体、前記リンカー上の配列に特異的に結合する単一ドメイン抗体、又は前記リンカー上の配列に特異的に結合するラクダ抗体を含む細胞外領域ポリペプチドであって、前記リンカーに結合する細胞外領域ポリペプチド、及び
ii.内在性膜タンパク質に由来する膜貫通アミノ酸配列
を含む、工程と、
(b)(a)の細胞を十分な量の前記リンカーと接触する工程であって、前記リンカーは、前記モノクローナル抗体の保存ドメインと結合して第2の複合体を形成する可変領域を含む可溶性の抗体又は抗体断片であり、前記接触は、前記第1及び第2の複合体を形成すると前記モノクローナル抗体がそれを分泌するハイブリドーマ細胞の表面に結合することを可能にする、工程と、
(c)前記ウイルス抗原が前記細胞結合アンカー-リンカー-mAb複合体に結合するために十分な時間、前記集団を多価ウイルス抗原と接触させ、細胞結合アンカー-リンカー-mAb-ウイルス抗原複合体を生じる工程と、
(d)前記細胞集団によって分泌される更なる免疫グロブリン(Ig)が、前記結合した多価ウイルス抗原上の他の結合部位に結合し、それによってアンカー-リンカー-mAb-ウイルス抗原-Igの検出可能な免疫複合体を形成することを可能にする工程と、
(e)細胞表面上の前記複合体のサイズ及び位置又は場所に基づいて、前記アンカー-リンカーmAbウイルス抗原-Ig複合体が結合した細胞を同定することを含む、工程とを含む、方法。 - 前記多価ウイルス抗原は、1個以上の精製ウイルス抗原、全ビリオン、又は、ウイルス様粒子(VLP)であるか;または、前記同定する工程(e)は、接触により得られる(a)の細胞と、一価ウイルス抗原又は非ウイルス抗原とを比較することを含むか;または、前記一価ウイルス抗原はFab断片である、請求項13に記載の方法。
