「ポリペプチド」、「オリゴペプチド」、「ペプチド」、および「タンパク質」という用語は、任意の長さのアミノ酸鎖を指し、本明細書では同義に用いられる。例えば、鎖長が比較的短い場合もあれば(例えば、10以上100以下のアミノ酸)それより長い場合もある。鎖は、線状または分岐状であることがあり、改変アミノ酸を含むことがあり、非アミノ酸が割り込むこともある。これらの用語は更に、自然または介入により改変されたアミノ酸鎖も網羅する。例えば、ジスルフィド結合の形成、グリコシル化、脂質化、アセチル化、リン酸化、または標識成分との結合など、その他のあらゆる操作または改変などである。本定義には更に、例えば1以上のアミノ酸アナログを含むポリペプチド(例えば、非天然アミノ酸などを含む)、および当業者に公知のその他の改変も含まれる。ポリペプチドは、単鎖または付随鎖として発生し得ることが知られている。
本明細書で提供する開示は、改変カスパーゼ-9ポリペプチドに関する。本明細書で使用される場合、「カスパーゼ-9ポリペプチド」とは、野生型ヒトカスパーゼ-9のアミノ酸配列を含むポリペプチド(すなわち、配列番号1)か、野生型ヒトカスパーゼ-9のアミノ酸配列の一部を含むポリペプチドであって、その一部が、カスパーゼ9媒介のシグナル伝達を誘導できる(例えば、配列番号2に示すアミノ酸配列を有する、カスパーゼリクルートメントドメイン(「CARD」)のないヒトカスパーゼ-9)ポリペプチドか、または配列番号1もしくは2に示すアミノ酸配列に対して、60%、65%、70%、75%、80%、85%、90%、95%、96%、97%、98%、もしくは99%以上の配列同一性を有するポリペプチドを指す。本明細書で使用される場合、「改変カスパーゼ-9ポリペプチド」とは、ポリペプチドが、対応する野生型ヒトカスパーゼ-9ポリペプチドに対して1以上のアミノ酸変異を有する、上述の通りのカスパーゼ-9ポリペプチドを指す。
いくつかの実施形態によれば、本明細書は、少なくとも2つの成分1)改変カスパーゼ-9ポリペプチド、および2)二量体化ドメイン、を含む操作されたキメラタンパク質を提供する。これら2つの成分を含むキメラタンパク質は、本明細書では「キメラカスパーゼ-9タンパク質」とも呼称される。
キメラカスパーゼ-9タンパク質のカスパーゼ-9ポリペプチド部は、改変カスパーゼ-9ポリペプチドが上記で提供される特性のいずれかを有してよい。
キメラカスパーゼ-9タンパク質の「二量体化ドメイン」は、二量体化ドメインに結合できる二量体化リガンドなどによって二量体化を誘導できる任意のアミノ酸配列であってよい。例えば、二量体化ドメインは、FK506結合タンパク質(「FKBP」)のアミノ酸配列を含んでよい。FKBPタンパク質は、薬剤FK506に特異的に結合する。FK506の多量体アナログ(すなわち、FK506のコピーを2以上含むリガンド、またはその誘導体)であるリガンドは、第1タンパク質および第2タンパク質双方に結合することにより、FKBPのアミノ酸配列をそれぞれ含む第1タンパク質および第2タンパク質の二量体化を誘導し、それにより両者を結合する。そのため、本明細書で提供するキメラカスパーゼ-9タンパク質は、キメラカスパーゼ-9タンパク質の二量体化ドメインに結合する好適な二量体化リガンドに曝露することで、二量体化を誘導できる。

いくつかの実施形態によれば、本明細書で提供するキメラカスパーゼ-9タンパク質は、好適な二量体化リガンドを介して二量体化するように誘導されてよい。二量体化リガンドにより(両タンパク質のコピーの二量体化ドメインを介して)キメラカスパーゼ-9タンパク質の2つのコピーが結合され、それによりキメラカスパーゼ-9タンパク質の二量体化が誘導される。通常、本明細書で提供する実施形態に用いられる二量体化リガンドは、被験体に容易に投与できる3kDa未満の小分子である。
いくつかの実施形態によれば、本明細書は、本明細書で説明する改変カスパーゼ-9ポリペプチドまたはキメラカスパーゼ-9タンパク質をコードする核酸配列を含むポリヌクレオチドを更に提供する。
いくつかの実施形態によれば、本明細書は、宿主細胞を調製する方法を提供する。
本開示は更に、本明細書で提供する改変カスパーゼ-9ポリペプチドまたはキメラカスパーゼ-9タンパク質のいずれかをコードするポリヌクレオチドを含む操作された宿主細胞も提供する。いくつかの実施形態によれば、本明細書で提供するカスパーゼ-9タンパク質をコードするポリヌクレオチドは、導入遺伝子としてプラスミドベクターを介して宿主細胞に導入できる。いくつかの実施形態によれば、プラスミドベクターは更に、例えばベクターを受け取った細胞の識別および/または選択を行う選択マーカーを含んでもよい。通常、本明細書で提供する宿主細胞は免疫細胞であり、例えばプライマリT細胞である(例えば、図4から図9までを参照)が、他の種類の宿主細胞も可能である。
いくつかの実施形態によれば、本明細書で提供するキメラカスパーゼ-9タンパク質、または改変カスパーゼ-9ポリペプチドをコードするポリヌクレオチドを含む宿主細胞は、治療目的で被験体に導入するために用いることができる。例えば、宿主細胞は免疫細胞であってもよく、被験体内の癌を治療するために免疫細胞(例えばCAR-T細胞)を被験体に導入してもよい。宿主細胞を被験体に投与する前に、本明細書で提供するキメラカスパーゼ-9タンパク質をコードするポリヌクレオチドを宿主細胞に組み込むことにより、本明細書で提供するシステムおよび方法に従い宿主細胞を被験体内で(必要に応じて)改変できる。例えば、宿主細胞を被験体に導入した後、本明細書で提供するキメラカスパーゼ-9タンパク質を含む宿主細胞でアポトーシスを誘導することが必要であるか望ましい場合、宿主細胞のキメラカスパーゼ-9タンパク質の二量体化ドメインに結合する好適な二量体化リガンドを被験体に投与できる。導入した細胞を排除するかどうかの判断は、例えば、許容できないレベルの毒性が検出された場合、または細胞が望ましくない状態で増殖している場合など、導入された細胞に起因して患者に望ましくない影響が検出された際に必要となることがある。例えば、本明細書で提供するキメラカスパーゼ-9タンパク質を含む宿主細胞を被験体に投与した後、いくつかの実施形態によれば、被験体に宿主細胞を投与した後、2時間、4時間、8時間、12時間、16時間、24時間、36時間、48時間、72時間、96時間、120時間、または144時間以内に、対応する二量体化リガンドを被験体に投与することができる。
本開示は更に、本方法で用いるためのキットも提供する。本開示のキットには、i)本明細書で説明されるように改変カスパーゼ-9ポリペプチドまたはキメラカスパーゼ-9タンパク質をコードするポリヌクレオチド、または改変カスパーゼ-9ポリペプチドもしくはキメラカスパーゼ-9タンパク質をコードするポリヌクレオチドを含む操作された免疫細胞と、ii)本明細書で説明される開示の方法のいずれかに従って使用するための説明書とを含む。一般的に説明書には、上記の治療処置のために操作された免疫細胞を投与するための説明が含まれる。任意にて、キットには更に、キメラカスパーゼ-9タンパク質の二量体化ドメインに結合する好適な(例えば、AP1903などの)二量体化リガンドと、例えば必要とする被験体に二量体化リガンドを投与する必要性と時期を含めた説明書とが含まれてよい。

次のように、4つの異なるヒト血液ドナー(「ドナー1」、「ドナー2」、「ドナー3」、および「ドナー8」)から、それぞれプライマリT細胞を調製した。

前記二量体化リガンドが、キメラタンパク質の二量体化ドメインに結合し、前記リガンドが前記二量体化ドメインに結合されると、カスパーゼ-9活性が誘導される方法。
〔26〕前記リガンドがAP1903である、前記〔25〕に記載の方法。
〔27〕操作された免疫細胞を調製する方法であって、
前記方法が、前記〔6〕、〔7〕、〔19〕もしくは〔20〕に記載のポリヌクレオチド、または前記〔8〕もしくは〔21〕に記載の発現ベクターを、前記免疫細胞に導入することを含む方法。
Claims (23)
- 配列番号2に対して90%以上の配列同一性を有するアミノ酸配列と、1以上のアミノ酸置換とを含む改変カスパーゼ-9ポリペプチドであって、
前記アミノ酸置換が、S271Y、S274E、F342H、I370E、D379E、V387A、A390N、およびK409Nからなる群から選択され、各アミノ酸位置は配列番号1の各アミノ酸の位置を示す、改変カスパーゼ-9ポリペプチド。 - 前記改変カスパーゼ-9ポリペプチドが、Q221R-V387A、D330L-I370E、D330L-D379E、D330M-S271Y、D330M-S274E、D330M-F342H、D330M-I370E、D330M-D379E、D330M-V387A、D330M-A390N、およびD330M-K409Nからなる群から選択される2以上のアミノ酸置換を含む、請求項1に記載の改変カスパーゼ-9ポリペプチド。
- 前記改変カスパーゼ-9ポリペプチドが、配列番号3に示すアミノ酸配列を含む、請求項1または2に記載の改変カスパーゼ-9ポリペプチド。
- 請求項1から3のいずれか1項に記載の改変カスパーゼ-9ポリペプチドをコードする核酸配列を含む、ポリヌクレオチド。
- 前記ポリヌクレオチドが、配列番号11に示す核酸配列を含む、請求項4に記載のポリヌクレオチド。
- 請求項4または5に記載のポリヌクレオチドを含む、発現ベクター。
- 請求項1から3のいずれか1項に記載の改変カスパーゼ-9ポリペプチド、または請求項4もしくは5に記載のポリヌクレオチドを含む、操作された免疫細胞。
- 二量体化ドメインと改変カスパーゼ-9ポリペプチドとを含むキメラタンパク質であって、
前記改変カスパーゼ-9ポリペプチドが、配列番号2に対して90%以上の配列同一性を有するアミノ酸配列と、および1以上のアミノ酸置換とを含み、
前記アミノ酸置換が、S271Y、S274E、F342H、I370E、D379E、V387A、A390N、およびK409Nからなる群から選択され、各アミノ酸位置は配列番号1の各アミノ酸の位置を示す、キメラタンパク質。 - 前記改変カスパーゼ-9ポリペプチドが、Q221R-V387A、D330L-I370E、D330L-D379E、D330M-S271Y、D330M-S274E、D330M-F342H、D330M-I370E、D330M-D379E、D330M-V387A、D330M-A390N、およびD330M-K409Nからなる群から選択される2以上のアミノ酸置換を含む、請求項8に記載のキメラタンパク質。
- 前記二量体化ドメインが、FKBPポリペプチドおよびシクロフィリンポリペプチドからなる群から選択されるポリペプチドを含む、請求項8または9に記載のキメラタンパク質。
- 前記二量体化ドメインが、FKBP12ポリペプチドを含む、請求項8から10のいずれか1項に記載のキメラタンパク質。
- 前記FKBP12ポリペプチドが、アミノ酸置換F36Vを含む、請求項11に記載のキメラタンパク質。
- 前記キメラタンパク質が、配列番号6に示すアミノ酸配列を含む、請求項8から12のいずれか1項に記載のキメラタンパク質。
- 前記二量体化ドメインが、二量体化リガンドAP1903、AP20187、二量体化FK506、または二量体化FK506様アナログに結合する、請求項8から13のいずれか1項に記載のキメラタンパク質。
- 請求項8から14のいずれか1項に記載のキメラタンパク質をコードする核酸配列を含む、ポリヌクレオチド。
- 前記ポリヌクレオチドが、配列番号8に示す核酸配列を含む、請求項15に記載のポリヌクレオチド。
- 請求項15または16に記載のポリヌクレオチドを含む、発現ベクター。
- 請求項8から14のいずれか1項に記載のキメラタンパク質、または請求項15もしくは16に記載のポリヌクレオチドを含む、操作された免疫細胞。
- キメラ抗原受容体(CAR)、またはCARをコードするポリヌクレオチドをさらに含む、請求項18に記載の操作された免疫細胞。
- 前記免疫細胞がT細胞である、請求項18または19に記載の操作された免疫細胞。
- 請求項18から20のいずれか1項に記載の操作された免疫細胞を含む、前記操作された免疫細胞を被験体内で改変する方法に使用するための、免疫療法用医薬組成物であって、、
前記方法が、前記操作された免疫細胞を先に投与された被験体に、二量体化リガンドを投与することを含み、
前記二量体化リガンドが、キメラタンパク質の二量体化ドメインに結合し、前記リガンドが前記二量体化ドメインに結合されると、カスパーゼ-9活性が誘導される方法である、上記医薬組成物。 - 前記リガンドがAP1903である、請求項21に記載の医薬組成物。
- 操作された免疫細胞を調製する方法であって、
前記方法が、請求項4、5、15もしくは16に記載のポリヌクレオチド、または請求項6もしくは17に記載の発現ベクターを、前記免疫細胞に導入することを含む方法。
