Description
本発明は、本明細書に記載される自然リンパ球前駆細胞(ILCP)を含む組成物を包含する。本明細書で(例えば、実施例で)使用されるマーカーの全ては、ILCPのマーカーとして使用するための任意の及び全ての組み合わせで明確に熟慮され、本発明の種々の実施形態で使用され得る。
本発明は、本発明の自然リンパ球前駆細胞(ILCP)の精製集団を作製する方法を包含する。一実施形態では、本方法は、ヒト細胞試料を用意する工程と、試料中でILCPについて選択する工程とを含む。ILCPは本明細書(例えば、実施例)に示されるマーカーの任意の組み合わせを用いて選択することができる。一実施形態では、細胞が、以下のマーカーLin-、CD34-CD7+CD127+CD117+CD45RA+、NKp44-、RORγt-、IL-1R1+、CD69-、CD62L+、CD26+;特に、Lin-、CD34-CD7+CD127+CD117+CD45RA+、NKp44-、IL-1R1+、CD69-、CD62L+、CD26+の任意の組み合わせについて選択される。Lin-は系統陰性細胞を指す;Lin-は、CD3-、CD4-、CD5-、TCRαβ-、TCRαβ-、CD14-及びCD19-を含む及び指す。選択は、例えば、実施例に記載されるように、FACS分析及び細胞選別等の当技術分野の日常的な技術によって行うことができる。
本発明は、添加されるILCP増殖の方法を包含する。細胞をIL-1βを用いて培養すると、細胞の堅牢な増殖が観察される。ILCPを増殖するために、培養培地はIL-1β、好ましくはIL-1β及びIl-2を含む。更に、培地は場合によりIL-7等の他のサイトカインを含むことができる。
本発明は、ILC1、ILC2、ILC3及びNK細胞を作製する方法を包含する。ILC1、ILC2、ILC3及びNK細胞を、当技術分野で日常的な技術によって、本発明のILCPから作製することができる。例えば、ILC1、ILC2、ILC3及びNK細胞を、実施例に開示される特定の技術を使用して作製することができる。一実施形態では、細胞系(例えば、Mohtashamiら(2010)に開示されるOP9間質細胞系)を使用して、本発明のILCPからILC1、ILC2、ILC3及びNK細胞を作製することができる。最も好ましくは、細胞が「可塑性」とも呼ばれる表現型の変化も機能の変化もなく増殖する。
本発明は、自然免疫系調節を必要とする患者を治療するために使用するための自然リンパ球前駆細胞(ILCP)を含む組成物を包含する。よって、本発明は、患者を治療するためのこれらの化合物の使用及び患者を治療する方法を包含する。
本発明は、ILCPのILC1、ILC2、ILC3及び/又はNK細胞への分化を調節(すなわち、阻害又は増強)する化合物をスクリーニングする方法を包含する。種々の実施形態では、本発明のILCPの集団を、インビボ又はインビトロで、試験化合物と接触させ、化合物の分化に対する効果を評価する。効果は、細胞集団中の細胞の表現型の変化を検出することによって観察することができる。
ヒト血液及び組織試料
健康なドナーの血液試料を、パスツール研究所と署名で合意してEstablishment Francais du Sang(EFS、パリ)から入手した。RORC突然変異(RORC-/--P1;RORC-/--P2、p.A421X/Q421X)を有する患者の血液試料は以前に報告されている(26160376)。臍帯血を正常分娩から得た。扁桃を、扁桃摘出術を受けた小児患者から入手した。胎児肝臓を14~20週齢に及ぶ妊娠期間での人工妊娠中絶から入手した。ヒト胎児肝臓を用いる実験は、パスツール研究所の医学倫理委員会によって承認され、フランスの法律を完全に順守して行った。肺は外科手術を受けている患者から入手し、試料はDr.JM Sallennave(ビシャ病院)によって提供された。腸は外科手術を受けた結腸がん患者から入手し、Dr.M Allez(セントルイス病院)によって提供された。インフォームドコンセントを、ネッカー医学学校、パリデカルト大学、ビシャ病院、セントルイス病院、パリ公立病院支援機構の治験審査委員会によって要求及び承認されるように各患者から入手した。
BALB/c Rag2-/-Il2rg-/- SirpaNOD(BRGD)マウスを記載し、パスツール研究所でアイソレーター中で維持した。CD34+HSC又はCD117+ILCを、FACS Ariaを使用して健康なドナーの末梢血から選別した。胎児肝臓CD34+ HSCを、CD34マイクロビーズキット(Miltenyi)を使用して単離した。インビボ移植実験については、1~3×105個のCD117+ ILC又はCD34+ HSCを、0.3μgのIL-2及びIL-7(Miltenyi)と合わせて、致死量以下照射(3Gy)新生(3~7日齢)BRGSマウスに肝内注射した。マウスに毎週腹腔内注射によってIL-2、IL-7、IL-1β、IL-23、IL-25及びIL-33(それぞれ0.3μg)を与え、移植4週間後に分析した。HISマウスの作製については、胎児肝臓由来CD34+ HSCを、致死量以下照射(3Gy)新生(3~7日齢)BRGSマウスに肝内注射した。マウスを注射8~9週後に屠殺した。実験は、パスツール研究所の倫理委員会によって承認され、フランス教育研究省によって検証された。
CB及びPBのヒト末梢血単核細胞(PBMC)を、Ficoll-Paque(GE Healthcare)密度勾配遠心分離によって単離した。胎児肝臓及び扁桃からの単一細胞懸濁液を、70μmフィルタを通した機械的破壊によって達成した。肺及び腸試料を切り刻み、37℃振盪インキュベーター中で45分間、Liberase TL(25μg/ml;Roche)及びDNアーゼI(50μg/ml;Sigma-Aldrich)で消化した。消化した組織を70μmフィルタに通過させた。肝臓、肺及び腸からのリンパ球を、Ficoll-Paque密度勾配遠心分離によって単離した。
FACS分析のために、最初に、細胞をFlexible Viability Dye eFluor 506(eBioscience)で10分間染色し、引き続いて氷上でBrilliant Stained Buffer(BD)で20分の表面抗体染色をした。細胞内TF染色を伴う実験については、細胞を固定し、透過処理し、Foxp3/転写因子染色緩衝液キット(eBioscience)を使用して染色した。細胞内サイトカイン染色については、細胞を、Golgi Plug(BD)の存在下で、PMA(10ng/ml;Sigma)+イオノマイシン(1μg/ml;Sigma)で3時間刺激した。細胞を固定し、透過処理し、Cytofix/Cytopermキット(BD)によって染色した。試料をLSRFortessa(BD)で獲得し、FlowJ10(Tree Star)によって分析した。
各集団の103個の細胞を、溶解/結合緩衝液(Life Technologies)50μl中に直接FACS選別した。mRNAをDynabeadsオリゴ(dT)(Life Technologies)で捕捉し、洗浄し、10mM Tris-HCl(pH7.5)10μlで70℃で溶出した。単細胞RNA-seq用に開発された記載される(24531970)MARS-seqの派生物を使用して、1集団当たり最小2つの複製を含む発現ライブラリーを作製することができる。1ライブラリー当たり平均400万の読み取りを配列決定し、デフォルトパラメータ(10289445)と共にTopHat v2.0.10を使用してヒト参照ゲノム(NCBI)に整列した。発現レベルを計算し、各試料について、HOMERソフトウェア(homer.salk.edu)を使用して読み取り総数に正規化した。任意の2つのサブタイプの平均間のノイズに対して2倍の差を有する高度に発現された遺伝子に焦点を当てた(8つの読み取り)。DAVID(12734009)を使用して、KEGG分析を行った。
FACS選別細胞(20~50K)を直ちに、ChIP-Seq分析のために室温で10分間、1%ホルムアルデヒド(Sigma)を含むPBS中で架橋させた。グリシン(0.125M最終濃度)を添加し、引き続いて室温で5分間インキュベートすることによって、架橋をクエンチした。細胞を氷上に置き、PBSで洗浄し、-80℃で貯蔵するために瞬間凍結した。技術的変動を最小化するためにペレットを同時に処理した。細胞を100μl超音波処理緩衝液(1%SDS、10mM EDTA、50mM Tris-HCl pH8及び1×EDTAフリー完全プロテアーゼ阻害剤;Roche)に再懸濁し、0.65ml Bioruptor超音波処理管(Diagenode)に移した。15分間の氷上でのインキュベーション後、Bioruptor Pico超音波処理器(Diagenode)を用いて、細胞を30サイクル(30秒間ON-30秒間OFF)の間超音波処理して、クロマチンを±250bpフラグメントまでせん断した。900μl 10×ChIP希釈緩衝液(0.01%SDS、1.1%Triton X-100、1.2mM EDTA、16.7mM Tris-HCl pH8、167mM NaCl)を添加することによって、クロマチンを平衡化し、1μlのH3K4Me2特異的抗体(ab32356、Abcam)又は陰性対照としての正常ウサギIgG(sc-2027、Santa Cruz)と共に4℃で一晩インキュベートした。更に、1IP当たり20μlのプロテインA Dynabeads(Thermo Fisher Scientific)を、4℃で一晩のインキュベーションによって0.1%BSA(Sigma)を含むPBS中でブロッキングした。翌日、ビーズをChIP希釈緩衝液によって元の体積に再懸濁し、クロマチン抽出物に添加した。4℃で2時間のインキュベーション後、ビーズを回収し、低塩緩衝液(0.1%SDS、1%Triton X-100、2mM EDTA、20mM Tris-HCl pH8、150mM NaCl)、高塩緩衝液(0.1%SDS、1%Triton X-100、2mM EDTA、20mM Tris-HCl pH8、500mM NaCl)及びLiCl緩衝液(10mM Tris-HCl pH8、1mM EDTA、250mM LiCl、0.5% NP-40、0.5%デオキシコール酸)で洗浄した。次いで、磁気ビーズ上に固定化されたクロマチン抗体を、近年記載されている(Schmidlら、2015)、タグメンテーション(tagmentation)に供した。溶出したDNAを、MinEluteスピンカラム(Qiagen)を使用して精製し、Nextera PCRプライマーを使用して8~12サイクル増幅した。二重(0.5×~2.0×)SPRI Ampure XPビーズ(Beckman Coulter)を使用してライブラリーを精製し、プールし(1配列決定実行当たり最大10)、単一読み取り75bpプロトコルを実行するNextSeq500(Illumina)で配列決定した。
読み取りを、BaseSpace(Illumina)を使用して多重分離し、標準的な設定で、唯一にマッピングされ得なかった読み取りを除去して、Bowtie(Langmead,B,&Salzberg,S.L. Fast gapped-read alignment with Bowtie 2. Nat Methods 9、357~359、doi:10.1038/nmeth.1923(2012))を使用してマウスゲノム(mm10構築)に割り当てた。インデックスされ、選別されたbamファイルを、更なる分析のためにHOMER(Heinz, S.ら Simple combinations of lineage-determining transcription factors prime cis-regulatory elements required for macrophage and B cell identities. Java TreeView(Saldanha, A.J. Java Treeview-extensible visualization of microarray data. Bioinformatics 20、3246~3248、doi:10.1093/bioinformatics/bth349(2004))を用いてヒートマップとして可視化した。領域/ピークを、推定上の標的遺伝子GREATに割り当てた(McLean,C.Y.ら GREAT improves functional interpretation of cis-regulatory regions. Nat Biotechnol 28、495~501、doi:10.1038/nbt.l630(2010))。その後、GREATを使用して、バックグラウンドとして全ゲノムを使用して、公知の経路サインについてのこれらの遺伝子の濃縮を計算した。
全てのインビトロ培養実験を、2%ヒトAB血清(EFS)を補足したYssel培地(18432890)で行った。2~3×103個の間質細胞を、培養の一晩前に96ウェル丸底プレートに事前に蒔いた。Yssel培地を、IMDM(Invitrogen)+0.25%(w/v)ウシ血清アルブミン(Sigma)、1.8μg/L 2-アミノエタノール、40μg/Lアポトランスフェリン、5μg/Lインスリン及びペニシリン/ストレプトマイシンを使用することによってインハウスで調製する。バルク培養については、100~300個のFACS選別細胞を間質細胞上に蒔いた。クローニング実験については、細胞を、間質細胞を事前に蒔いた96ウェルプレートに直接インデックス選別した。サイトカインIL-2、IL-7(それぞれ20ng/ml、Miltenyi)、IL-12、IL-18、IL-25、IL-33、IL-1β、IL-23(それぞれ20ng/ml、R&D)を指示されるように種々の組み合わせで用意した。バルク培養については、新鮮なサイトカイン及び培地を5日毎に補充し、10日間の増殖後に分析した。クローニング実験のために、サイトカイン及び培地を7日毎に補充し、14~18日間の培養後に分析した。
ヒト末梢血CD117+ ILCの特性評価
指定しない限り、データを中央値として表す。各実験についての標本サイズ及び実験の複製数を図の凡例に含める。循環ILCを、健康な個体並びに多様な臨床症候群を患っている患者の系統-CD7+CD56-CD127+末梢血(PB)細胞内の低頻度集団(全CD45+細胞の0.2%未満)として同定することができる((Hazenberg及びSpits、2014;Munnekeら、2014;図1A及び図IB)。PB ICLのILC1、ILC2及びILC3への更なる分画が、CD161、CRTh2、CD117及びNKp44を含む胎児組織及び扁桃中のILCサブセットを同定する表現型マーカーを使用して達成されている(Spitsら、2013)。よって、以前の報告は、ヒト血液中の循環ILC2(CD161+CRTh2+GATA-3+細胞)並びにILCl(CD161+CRTh2-CD117-T-BET+細胞)を同定している(Mjosbergら、2011)。循環ILCはまた、CRTh2発現を欠く優勢CD117+サブセットも含む((Munnekeら、2014;Velyら2016);図1A及び図IB)。組織常在性ILC3がCD117を強力に発現するので、以前の研究は、これらの細胞を循環ILC3とみなしていた(Cellaら、2009;Cupedoら、2009)。しかしながら、PB CD117+ ILCは、ILC3を同定するNKp44及び転写因子(TF)RORγtの発現を欠くという点で、腸CD117+ ILCとは劇的に異なることが分かった(図1C)。したがって、PB CD117+ ILCは刺激後にIL-17AもIL-22も産生しないが、腸CD117+細胞はこれらのILC3関連サイトカインを豊富に産生する(図1D)。興味深いことに、循環CD117+ ILCは高レベルのIL-1R1、CD45RAを発現し、CD69-である一方、腸常在性ILC3はCD69+であるが、IL-1R1-及びCD45RA-である(図1C)。これらの知見は、PC CD117+ ILCが正真正銘のILC3ではないことを示唆している。
CD117+ ILCの転写及びクロマチンランドスケープはILC前駆細胞プロファイルを明らかにする
CD117は血液リンパ系前駆細胞上で高度に発現するので(Ikuta及びWeissman、1992;Kikushigeら、2008)、PB CD117+ ILCが確約されていないリンパ球前駆細胞を含み得ると仮定された。PB CD117+ ILCの正体を更に理解するために、高度に精製された循環CD117+ ILCのトランスクリプトーム及びエピジェネティックランドスケープを明らかにし、CD34+ HSCと比較した(図2A);後者は多系統能力を有する未成熟造血前駆細胞を表す(Baumら、1992;Mohtashamiら、2010)。
末梢血CD117+ ILCは多能性ILC前駆細胞(ILCP)を含む
循環CD117+ ILCの造血能力を評価するために、これらの細胞を種々のサイトカインの存在下でバルク培養した。CD117+ ILCはCD25、CD127及びCD121a(IL-1R1)を発現するので(図1A、図1C)、IL-2、IL-7及び/又はIL-1βをこれらの培養物に添加した。バルク培養物はIL-2及びIL-7の存在下で最小限増殖したが、細胞を1L-1βで培養した場合、堅牢な増殖が観察された(図3A)。ILC1/NK(IL-12、IL-18)、ILC2(IL-25、IL-33)又はILC3(IL-23)発達を駆動することができるサイトカインの更なる存在は、IL-1βで得られたものよりは細胞収率を更には増加させなかった(図3A)。バルク培養細胞はB(CD19+)細胞もT(CD3+CD5+)細胞も持たなかったが、CD161+であるCD7+細胞の純粋な集団を含んでおり、可変レベルのCD117及びCD25を発現していた(図3B)。
循環CD117+ ILCPはヒト化マウスにおいてインビボで多ILC能力を有する
次に、PB CD117+ ILCPのインビボ能力を評価した。以前の研究により、ヒトCD34+造血幹細胞(HSC)前駆細胞を移植した重度免疫不全マウス株がヒトリンパ球(B、T、NK)及び骨髄(DC、マクロファージ、好中球)細胞サブセットを産生する能力が実証された((Shultzら、2012)に概説)。堅牢な多系統ヒト造血細胞移植を許容するBALB/c Rag2-/-Il2rg-/-SirpaNOD(BRGS)マウスを使用した(Legrandら、2011)。同じドナーからのヒトPB CD34+ HSC及びCD117+ ILCPを新生BRGSマウスに養子的に移植した;サイトカイン補充(IL-2、IL-7、IL-1β、IL-23)を提供し、4週間後にマウスを分析した(図4A)。ヒトCD34+ HSCを移植したBRGSマウスは骨髄でCD19+ B細胞及びCD14/CD33+骨髄細胞を発達した一方、CD3/CD5+ T細胞及びLin-CD7+ NK/ILCが腸で検出された(図4B)。対照的に、PB CD117+ ILCPを受けたBRGSマウスは、Lin-CD7+細胞を発達させたが、骨髄細胞も、B細胞も、T細胞も発達させなかった。移植CD117+ ILCPからのヒトCD45+子孫を、脾臓、肺、腸及び肝臓を含む複数の器官で検出した(図4C)。これらの組織部位の各々で、刺激時にエキソビボでIFN-γ、IL-13、IL-17A及び/又はIL-22を産生するEOMES+ NK細胞並びに多様なCD127+ ILCサブセットを同定することができた(図4C)。これらの結果は、PB CD117+ ILCPがインビボで全ての公知のILCサブセット及びNK細胞を産生する能力を有することを実証している。PB CD117+ ILCPは骨髄、B及びT細胞能力を欠いているので、これらの細胞が確約されたILC前駆細胞を構成すると結論付けられた。
ヒトCD117+ ILCPはインビボでCD34+ HSCから発達する
次に、CD34+ HSCとCD117+ ILCPの間の発達関係を調べた。免疫不全新生仔BRGSマウスに、精製CD34+ HSCを移植し、8~9週間後に屠殺した(図5A)。ヒトCD45+細胞を骨髄、肺、肝臓及び脾臓で分析した。予想されるように(Legrandら、2011)、これらの様々な組織部位は、種々の系統+ T、B及び骨髄細胞を含むヒトCD45+細胞を持っていた(図5B、データは示さず)。更に、Lin-CD7+細胞のサブセット内で、T-BET及びEOMES発現を欠くCD127+CD117+細胞の明確に規定される亜集団が複数の組織で識別された(図5B)。これらには、低レベルのGATA-3及びRORγtを発現し、NKp44-であり(図5D)、そのためPB CD117+ ILCPに似ているCD127+CD117+細胞が含まれた。エキソビボ刺激はCD127+CD117+細胞からサイトカイン産生を誘発できなかった(図5E)。これらの細胞を選別し、IL-2、IL-7、IL-1β及びIL-23の存在下でバルク培養した。増殖細胞はIFN-γ、IL-13、IL-17A及びIL-22を産生することができるサブセットを含んでおり(図5E)、それによってヒトILCPの存在が確認された。これらの結果は、CD34+ HSCがインビボでCD117+ ILCPをもたらすことができることを実証している。
ヒトCD117+ ILCPは胎児肝臓、臍帯血及び成人肺中に存在する
次に、ヒトCD117+ ILCPが生じる発達段階を評価した。最初に、胎児肝臓(FL)がいくつかの未成熟造血前駆細胞集団を持つことが示されており(Rolliniら、2007)、マウスにおけるリンパ組織誘導細胞の発達の見解として提案されている(Cherrierら、2012)ので、FL中のヒトILCサブセットを試験した。FL内のLin-CD127+ ILCは、優勢なCD117+サブセットを含んでいる。興味深いことに、これらの細胞は、その末梢血対応物を超えるレベルでRORγtを発現し(図1C)、更に、CCR6、ニューロピリン-1(NRP-1)を発現するが、NKp44は発現しない。これらの差異にもかかわらず、FL CD117+ ILCは、刺激後に有意な量のIL-17AもIL-22も産生せず、これらが完全成熟ILC3ではなかったことを示唆している。それにもかかわらず、FL CD117+ ILCをインビトロで増殖させると、IL-17A産生ILC3が豊富に産生された。更に、IL17A+ ILC3はDL4を欠く間質細胞で発達し、この過程に追加のNotch関与は必要でなかったことを示唆している(図6A)。興味深いことに、FL CD117+ ILCのバルク培養物はまた、頻度は低いが、検出可能なIL-13及びIFN-γ産生細胞を含んでいる。クローン分析によって、FL CD117+ ILCが、予想通り、高い割合のILC3確約前駆細胞を持つことが明らかになった。更に、この集団の相当な割合が、Notchリガンドの存在下でより明確に明らかにされる多能性ILCPを含んでいる(図6B、図6C)。これらの結果は、ヒトFLが、全ての公知のILCサブセットを産生することができるCD34-CD127+CD117+多能性ILCPを持つことを実証している。この組織部位のILC3確約前駆細胞の濃縮は、環境シグナルが、この期間中に多能性ILCPのILC3の運命への更なる指定を指示し得ることを示唆している。
ILC前駆細胞は二次リンパ組織内に存在する
ヒト二次リンパ組織(リンパ節、扁桃)は、多様なILCサブセット及びその前駆細胞を持つ(Berninkら、2013;Cellaら、2009;Fehnigerら、2003;Mjosbergら、2011;Renouxら、2015;Montaldoら、2015;Scovilleら、2016)。そのため、扁桃CD117+ ILCPを更に特徴付け、その細胞の運命の能力を評価することは興味深かった。小児扁桃のCD117+ ILCは、IL-17A及びIL-22を産生するように刺激することができる優勢なNKp44+ ILC3サブセットを持つ(Hoorwegら、2012)。この集団はまた、刺激(IL-1β、IL-12、IL-23を使用)によってこれらの細胞のサイトカイン出力を改変することができる(Berninkら、2015;Berninkら、2013;Cellaら、2010)ので、広範な機能的可塑性を有するように思われる。扁桃CD117+ ILC内では、NKp44-細胞がCD45RA+及びNRP-1-である一方、NKp44+細胞がCD45RA-及びNRP-1+であることが分かった。これらは、NLp44+ ILC3がより成熟しており、NLp44-細胞から分化することを示唆している(Berninkら、2015)。しかしながら、サイトカイン産生プロファイルは、扁桃NKp44-対NKp44+ CD117+ ILCからのバルク培養で異なっていた(図6J及び図6M)。特に、IfN-γ+細胞及びIL-13+細胞は、特にOP9間質でのNKp44-細胞由来の培養物でより自明であった(図6J)。
RORC欠損患者はILCPを持つが、IL-17A+ ILC3を産生できない
CD34+CD45RA+CD117+表現型を有するヒト二次リンパ組織に確約されたILCPは、TF RORCを高度に発現することが示された(Scovilleら、2016)。CD117+ ILCPはCD34+ HSCから発達的に下流にあるので(図5)、以前記載されたCD34+CD45RA+CD117+ ILCPサブセットはこの経路の中間体となることが可能である。RORCがヒトCD117+ ILCPの産生に必要であったかどうかを扱うために、RORC欠損患者を研究した。ヒトにおけるRORC欠損は粘膜皮膚カンジダ症に関連し、以前の研究によって、このTFが真菌病原体から防御するTh17細胞の分化に必須であることが実証された(Okadaら、2015)。RORC欠損を有する2人の患者の末梢血細胞のILCサブセットを試験した(図7A)。Lin-CD7+細胞は、対照とRORC欠損患者の両方でCD56+ NK細胞の優勢な集団を含んでおり(CD56brightとCD56dimの両方)、CD127+ ILCの控えめな亜集団も明らかに検出された。以前記載されたように(Okadaら、2015)、CD117発現ILCの頻度減少がRORC欠損を有する患者で認められたが、この集団はまだ明らかに存在していた(図7A及び図7B)。対照的に、RORCの非存在下で、ILC1は存在し、全ILCからのILC2の割合は有意に増加した(図7B)。対照及びRORC欠損患者からの選別したCD117+ ILCを、上記のようにバルク培養した。Lin-CD7+細胞の堅牢な増殖が観察され、WTとRORC欠損細胞の間に有意な差はなかった。IFN-γ、IL-13又はIL-22を産生するものを含む多様なサイトカイン産生細胞がこれらの培養物で同定されたが、IL-17A産生細胞は存在しなかった(図7C)。EOMES+IFN-γ+ NK細胞の発達は、RORCの非存在によって影響を受けなかった。これらの結果は、RORCがヒトにおいてNK細胞、ILC1、ILC2及びIL-22+ ILC3の発達に必要とされないが、ILCPからのIL-17+ ILC3の産生には必須であることを実証している。
血液中のILCPマーカーの分析
CD127及びCD117はILCPによって発現されるが、これらは特異的ではない。最小限の必須マーカーの洞察を得るために、ILCPについて高度に濃縮するために使用することができたマーカーの組み合わせを分析した。そのため、様々な細胞型の割合を、成人末梢血から細胞を単離するための様々なマーカーを使用してFACSによって決定した。結果は、マルチパラメータFACS分析を用いた、末梢血中のヒトILCPについての濃縮の分析を表す。異なるゲーティングスキームを使用して、ILCP並びに存在し得る他の細胞型(T細胞、NK細胞、ILC2)の濃縮を推定することができる。結果を図8A~図8F及び図9A~図9Gに提示する。
追加のILCPマーカー
ヒトILCP(Lin-CD127+CD117+CRTh2-として定義される)を、これらの細胞を単離するための有用な代用となり得る追加の細胞表面マーカーの発現について更にスクリーニングした。可変的CD62L及びCD26発現がヒトILCPで同定され、ほとんどの細胞がCD62L+であり、細胞の大部分がCD26を発現していた(図10)。
Claims (16)
- 自然リンパ球前駆細胞(ILCP)の精製集団であって、前記集団中の前記細胞の少なくとも75%が、表現型CD127+CD117+Lin-CRTh2-を有する、ILCPの精製集団。
- 前記集団中の前記細胞の少なくとも90%が、前記表現型CD127+CD117+Lin-CRTh2-を有する、請求項1に記載のILCPの精製集団。
- 自然リンパ球前駆細胞(ILCP)の精製集団を作製する方法であって、
ヒト細胞試料を用意する工程と、
表現型CD127+CD117+Lin-CRTh2-を有する、前記細胞試料中の細胞を選択して、細胞の集団を得る工程と
を含み、
前記集団中の前記細胞の少なくとも75%が、前記表現型CD127+CD117+Lin-CRTh2-を有する、方法。 - 免疫病理、感染性疾患又はがんの治療に使用するための、ILCPの精製集団を含む組成物であって、前記集団中の細胞の少なくとも90%が、表現型CD127+CD117+Lin-CRTh2-を有する、組成物。
