細胞培養法
用語「培養物」および「細胞培養物」とは、本明細書で使用する場合、細胞集団の生存および/または増殖に適切な条件下で培地中に懸濁される細胞集団を指す。当業者に明らかなように、一部の実施形態では、これらの用語は、本明細書で使用する場合、細胞集団、および集団が懸濁される培地を含む組み合わせを指す。一部の実施形態では、細胞培養物の細胞は、哺乳動物細胞を含む。

アミノ酸の濃度は、当業者に公知の任意の方法によって測定され得る。オンラインまたはオフラインの方法でアミノ酸の濃度を測定するための好ましい方法には、例えば、液体クロマトグラフィー、例えば、HPLC、UPLCもしくはLCMS、NMRまたはGCMSが含まれる。
薬物対抗体比(DAR)は、当業者に公知の任意の方法によって測定され得る。DARを測定するための好ましい方法には、例えば、液体クロマトグラフィー、例えば、HPLC、UPLCまたはLCMS、質量分析、およびNMRが含まれる。

通常のCD CHO培地における高い細胞密度の安定なCHO発現による、完全にキャップされていないシステイン変異体抗体の生成
システイン変異体抗体トラスツズマブK290C-K334CまたはK392C-K334Cを安定に発現するCHO-K1細胞を、3E6細胞/mLまたは6E6細胞/mLの密度で通常のCD CHO培地(Thermo Fisher、Waltham、MA)中に播種し、増殖させ、5%CO2を有する加湿インキュベーター中で37セルシウス度に維持した。1つの条件では、5mMのシスチンを、CD CHO培地に添加した。細胞を、37セルシウス度で72時間培養した。細胞生存度を測定し、順化培地を収集した。細胞は、98%よりも高くが生存していた。
通常のCHO培地における高い細胞密度の安定なCHOによる、完全にTNBキャップされたシステイン変異体抗体の生成
実施例1に示されるように、高い密度での細胞増殖は、培地中の顕著な量のシステインまたはシスチンを消費し得る。結果的に、完全にキャップされていないシステイン変異体抗体が生成された。実施例1に基づくと、これは、異なる時点において種々の濃度のDTNBを細胞培養物に添加することによって、完全にTNBキャップされたシステイン変異体抗体の大量の生成を可能にした。DTNBは、キャップされていない変異体抗体上の遊離チオールを効率的にアルキル化し、TNBキャップされた抗体の生成を生じた。
制限されたシステイン/シスチン曝露を標的化するプロセスにおける、完全にTNBキャップされたシステイン変異体抗体の生成
以下の実施例では、システイン変異体抗体についてのキャッピング効率の読み出しは、最終精製の前の抗体へのリンカー-ペイロードのコンジュゲーション後に得られる粗製DAR4(薬物対抗体比)百分率によって示される。粗製DAR4値は、これらの実施例において、キャッピング効率の代用マーカーとして使用されている;しかし、抗体上の各変異したシステイン残基のキャッピング状態を決定することは、コンジュゲーションプロセスの前に長期LC/MS解析(実施例1に記載される)によって決定され得る。LC/MS解析の一例は、図7に見出すことができ、図中、抗体1つ当たりの異なるキャッピング種が特定および定量化されている。この解析に使用した試料は、TNBキャップされた抗体を産生するためのDTNB供給を伴う、低いシステイン/シスチンプロセスであった。所望の標的種は、抗体1つ当たり4つのTNBであり、これは、低レベルの還元不能なキャップおよびいくらかのサブ-4抗体種を伴って、この特定の実施例のLC/MS結果において>95%を占める。
供給培地中に処方された濃度を減少させることによって、培養物中のシステイン/シスチンを計画的に制限する
アプローチ1:この実施例は、補足的供給培地中に処方されたいずれかの成分の量を減少させることによって、培養物中のシステインおよび/またはシスチン濃度を意図的に制限することを示す。この実施例で使用した2つの条件は、基本培地中に類似の出発濃度のシステインおよび/またはシスチンを有した;しかし、1つの条件は、栄養素供給培地中にシステイン成分もシスチン成分も有さず、第2の条件は、シスチンのみを含有する供給培地を使用した(表1);全ての他のプロセスパラメーターは、これら2つの条件について同一であった。標準的な哺乳動物細胞培養プロセスは、細胞培養培地中でのこれらの成分の溶解度の課題に起因して、さらなる補足的供給中にシステインおよび/またはシスチンを取り込む。
化学量論的アプローチおよび標的化された分数システイン制限比を使用して、培養物中のシステイン/シスチンを制限する:以前に記載された合理的培地設計および化学量論的アプローチを使用して、特定のピーク細胞密度および産生される産物の量のために必要とされるシステイン/シスチンの必要量を計算でき、その特定のプロセスに最適な培地を設計するためにそれを使用できる(方程式1)。この実施例では、システイン/シスチンの必要量を、プロセスの推定ピーク生存細胞密度および収集力価を使用して、合理的培地設計に基づいて計算した。システイン/シスチンの計算された必要量を(例えば、米国特許第8,232,075号で教示される比と比較して)下回る異なる分数システイン制限比の範囲を評価して、許容可能なピーク生存細胞密度および収集力価になおも達しつつ、抗体のTNBキャッピングを促進するために、システイン/シスチンを制限または枯渇させる理想的な標的比を見出した(方程式2を参照のこと)。
ピーク細胞密度を増加させることによって、培養物中のシステイン/シスチンを制限する:実施例3.1で2つの細胞株によって以前に実証されたように、バッチの間に、システイン/シスチンが低い濃度に制限される場合または完全に枯渇される場合、TNBキャッピング効率は増加する。培養物中のシステイン/シスチン濃度のレベルを意図的に減少または枯渇させる第2の例は、ピーク生存細胞密度を使用することによって実施され得る。所与の培養物中の生存細胞密度が増加するにつれて、アミノ酸の消費およびアミノ酸に対する需要も、典型的には同様に増加する(米国特許第8,232,075号)。この実施例では、異なるピーク生存細胞密度が達成された2つの別々の条件を評価した。
細胞の非存在下でのTNBキャップされたCys変異体抗体の生成。所望の抗体薬物コンジュゲートを生成するためのコンジュゲーションステップにおいて引き続いて使用され得るような形態での抗体または融合タンパク質の生成は、慣用的には、製造細胞株の開発の一部として達成される、および/または製造プロセスの上流/細胞培養プロセス部分の一部として達成される。TNBキャップされた抗体の生成の場合、これまでに記載されたアプローチは、以前に記載された細胞培養プロセスの種々のさらなる改変(例えば、システイン/シスチン供給濃度の調整、バッチ長さの改変であるがこれらに限定されない)と共にまたはそれなしで、細胞培養プロセスの一部としてDTNB供給を含めることを介したものであった。これは、所望のキャップされた抗体の産生を可能にするが、これらのアプローチは、製造プロセスの細胞培養物部分に依存し、製造設備の全体的生産性を低減させ得る。したがって、TNBキャップされた抗体の処理量は、設備、特に、細胞培養に使用される産生バイオリアクターまたは発酵槽の最適未満の利用に起因して制限される。
TNBコンジュゲーションプロセス。TNBでキャップされたシステイン変異体抗体を、TSPPで選択的に還元する。生成された遊離チオール基は、限外濾過/ダイアフィルトレーション(UF/DF)および再酸化ステップなしの直接的薬物コンジュゲーションを可能にし、プロセスを単純化する。抗体1つ当たり4つのキャッピング(DAR4)の形態の、TNBで完全にキャップされたシステイン変異体抗体トラスツズマブK183C-K290Cが生成され、TSPP処理後に、70%の効率で直接コンジュゲートされた。
還元後ダイアフィルトレーションを用いるTNBコンジュゲーションプロセス。実施例5に記載されるプロセスにおいて使用される過剰量のリンカー-ペイロードは、TSPP還元後に、リンカー-ペイロードと反応する種を除去するためのダイアフィルトレーション(緩衝液交換)ステップを追加することによって低減させることができる。図14に示されるように、70%のDAR4が、緩衝液交換後に6当量のリンカーペイロードを用いて達成されるが、緩衝液交換が実施されない場合、有意により高い当量が必要とされる。また、TSPPの量は、還元温度および/または時間を増加させることによって、わずかに低減され得る。
増加したTSPP化学量論、還元後ダイアフィルトレーションおよび再酸化を用いる、TNBコンジュゲーションプロセス。実施例6に記載されるプロセスによって産生される凝集物の量は、TSPP化学量論を増加させることによって低減させることができる。増加したTSPP化学量論は、変異したシステインからTNBをより迅速に除去し、抗体間でのジスルフィド結合の形成を防止する。しかし、増加したTSPP化学量論は、鎖間ジスルフィド結合のごく一部の還元もまた生じる。塩化ナトリウムおよび他の塩の存在(実施例8を参照のこと)は、より高いTSPP化学量論における鎖間ジスルフィド還元の程度を減少させる。ダイアフィルトレーション後の再酸化ステップの追加は、還元された鎖間ジスルフィド結合の一部を修復する。図15に示されるように、20当量のTSPPによる還元および緩衝液交換の後のコンジュゲーションは、再酸化ステップなしの場合、鎖間ジスルフィド結合還元に起因する低いレベルの過剰コンジュゲートした種を生じる。再酸化をプロセスに追加すると、過剰コンジュゲートした種は形成されない。
増加したTSPP化学量論、還元後ダイアフィルトレーション、再酸化およびリンカー-ペイロードクエンチを用いる、TNBコンジュゲーションプロセス。還元混合物中の、実施例7中の塩化ナトリウムなどの塩の存在は、より高いTSPP化学量論において鎖間ジスルフィド還元の程度を減少させる。
増加したTSPP化学量論、還元後ダイアフィルトレーション、再酸化およびリンカー-ペイロードクエンチを用いる、TNBコンジュゲーションプロセス。この実施例は、実施例7と類似であるが、以下を特徴とする:(1)再酸化時間を顕著に低減させ、ダイアフィルトレーション後の時間のかかる冷却およびコンジュゲーション前の加熱を排除する、25℃における再酸化実行、および(2)特定の条件下での下流の精製を改善する、システインとの反応によるリンカー-ペイロードのクエンチ(以下の実施例14を参照のこと)。
TNBコンジュゲーションプロセス-再酸化による抗体断片修復。実施例7および9のような再酸化ステップの追加は、入ってくる抗体中に存在する断片を修復し、そうして、最終コンジュゲートの質および収量を改善した。断片は、鎖間ジスルフィド結合のうち1つまたは複数が完全なままではない抗体に起源する。したがって、実施例9に記載されるプロセスを使用する、15~35%の断片レベルを含む抗体ロットのコンジュゲーションは、低いおよび同等のレベルの断片とのコンジュゲートを産生した。
抗EDB K183C-K290C抗体を用いたTNBコンジュゲーションプロセス。TNBコンジュゲーションプロセスは、トラスツズマブ以外のTNBキャップされた抗体に適切である。
TNBコンジュゲーションプロセスによって産生されたコンジュゲートの精製。このプロセスによって産生された粗製コンジュゲートは、疎水性相互作用クロマトグラフィー(HIC)によって精製することができる。HIC精製は、遊離リンカー-ペイロード、凝集物、断片およびより低いDARのコンジュゲートを除去するまたは顕著に低減させる。
増加した回収を有するTNBコンジュゲーションプロセスによって産生されたコンジュゲートの精製。HICによって精製する場合、DAR4 ADCの回収は、移動相緩衝液Bへのイソプロパノールの添加によって改善され、したがって、プロセス収量を増加させることができる。
リンカー-ペイロードクエンチを用いたTNBコンジュゲーションプロセスによって産生されたコンジュゲートの精製。mcvcPABC0101リンカー-ペイロードを、実施例9のように、クエンチステップにおいてシステインでキャップする場合、HIC精製は、Capto(商標)Butyl ImpRes樹脂(GE)カラム単独を使用して達成することができる。システインキャップされたmcvcPABC0101は、DAR4コンジュゲートよりも早く溶出し、Capto(商標)Butyl ImpResカラムで十分に分離される。図17に示されるように、DAR4コンジュゲートを、この実施例に記載される条件を使用して、システインキャップされたmcvcPABC0101、より低いDARのコンジュゲート、凝集物および断片から分離する。システインキャップされたmcvcPABC0101は、ダイアフィルトレーションによっても容易に清澄化される。
抗体のヒンジ領域におけるジスルフィドスクランブリングを解析するためのアッセイ。TNBコンジュゲーションプロセスの利点は、抗体上のヒンジジスルフィド還元の顕著な減少または完全な排除であり、したがって、ネイティブの鎖間ジスルフィド結合を有するコンジュゲートの産生である。抗体およびコンジュゲートの両方においてヒンジスクランブリングのレベルを測定するためのアッセイを開発した。このアッセイでは、抗体またはコンジュゲートを、以下に示されるように、ヒンジの下で切断する酵素、およびヒンジの上で切断する第2の酵素で処理する:
システインキャップされた抗体へのコンジュゲーションとの比較。システインキャップされた抗体から産生されたADCは、ヒンジ-スクランブルされた産物を含有する。システインキャップされた抗体をコンジュゲートするために、抗体は、過剰量のトリス(2-カルボキシエチル)ホスフィン塩酸塩(TCEP)で完全に還元されなければならず、還元された鎖間ジスルフィド結合は、デヒドロアスコルビン酸で再酸化されて、変異したシステイン残基のみを、コンジュゲーションのための遊離チオールとして利用可能なままにする。次いで、変異したシステイン残基をリンカー-ペイロードにコンジュゲートして、粗製ADCを得る。再酸化ステップは、抗体のヒンジ領域において、ネイティブの鎖間ジスルフィド結合およびスクランブルされたジスルフィド結合の両方を産生する。この完全還元/再酸化プロセスによって産生されたADCについて、実施例15に記載されるアッセイによって測定されたヒンジスクランブリングのレベルは、典型的には5~20%である。ヒンジスクランブリングは出発抗体中では検出されていないので、全てのヒンジスクランブリングがADCコンジュゲーションプロセスに起源することに留意されたい。
Claims (7)
- 化学物質ペイロードにコンジュゲートされることが可能なシステイン含有抗体を生成するためのプロセスであって、
(a)システイン、シスチンおよびグルタチオンから選択される1種または複数の増殖成分を含む細胞増殖培地に、システイン含有抗体を発現することが可能な細胞を播種するステップ;
(b)前記細胞をインキュベートして、少なくとも5E6細胞/mLの細胞密度を達成するステップ;ならびに
(c)前記細胞をさらにインキュベートして、遊離チオールを含む1つまたは複数のキャップされていないシステイン残基を有するシステイン含有抗体を発現させるステップ
を含むプロセスであって、システイン含有抗体を発現することが可能な細胞がCHO細胞であり、かつ細胞増殖培地中の分数システイン制限比が0.8×未満であるプロセス。 - (d)前記発現されたシステイン含有抗体に、所定のキャッピング部分またはその前駆体を導入するステップ
をさらに含み、それにより、前記抗体上の1つまたは複数のシステインが、前記所定のキャッピング部分でキャップされ、かつ前記所定のキャッピング部分が、5-チオ-2-ニトロ安息香酸(TNB)、またはその前駆体である5、5‘―ジチオビズ(2-ニトロベンゾエート(DTNB)である、請求項1に記載のプロセス。 - 前記キャップされた抗体が、単離、精製、および濃縮のうち1つまたは複数からなる更なるプロセスに供される、請求項2に記載のプロセス。
- 前記細胞増殖培地中の分数システイン制限比が、0.75×、0.70×、0.65×、0.60×、0.55×、0.50×、0.45×、0.40×、0.35×、0.30×、0.25×、0.20×、0.15×、0.10×または0.05×未満である、、請求項1から3のいずれか一項に記載のプロセス。
- 前記細胞密度が、少なくとも10E6細胞/mL、50E6細胞/mL、100E6細胞/mLまたは500E6細胞/mLである、請求項1から4のいずれか一項に記載のプロセス。
- 前記システイン含有抗体が、抗EDB抗体および抗HER2抗体から選択される抗体である、請求項1から5のいずれか一項に記載のプロセス。
- 前記抗HER2抗体がトラスツズマブである、請求項6に記載のプロセス。
