本明細書において使用される場合、用語「ヌクレオチド」は、リボヌクレオチド若しくはデオキシリボヌクレオチド又はこれらの修飾形態及びこれらの類似物を指す。ヌクレオチドとして、プリン(例えば、アデニン、ヒポキサンチン、グアニン並びにこれらの誘導体及び類似物)並びにピリミジン(例えば、シトシン、ウラシル、チミン並びにこれらの誘導体及び類似物)を含む化学種が挙げられる。
タンパク質-セレックス技術に基づいて、本発明者らは、陽性スクリーニングのための標的タンパク質としてスクレロスチン、及び陰性スクリーニングのために無関係のタンパク質を使用し、高親和性を有してスクレロスチンに特異的に結合するアプタマーを最終的に選択した。本明細書に記載のスクレロスチンは、好ましくはヒトスクレロスチン、例えばそのアミノ酸配列が配列番号18に示されるスクレロスチンである。

別の態様では、本発明は、本発明のスクレロスチンに対するアプタマーによって疾患を処置する方法であって、本発明のスクレロスチンに対するアプタマーの治療有効量をそれを必要とする対象に投与することを含む方法を提供する。
別の態様では、本発明は、本発明の少なくとも1つのスクレロスチンに対するアプタマー及び薬学的に許容できる担体又は賦形剤を含む医薬組成物も提供する。前記医薬組成物は、例えば、スクレロスチン関連疾患を処置するために使用される。
ssDNAライブラリーは、各末端の18nt保存領域及び中央の無作為領域からなる。異なる長さの無作為化配列を含む2つのssDNAライブラリーを本プロジェクトにおいて使用した。長いssDNAライブラリーは、40ntランダム領域(5’-CGTACGGTCGACGCTAGC-(N)40-CACGTGGAGCTCGGATCC-3’)を含有し、短いssDNAは、25ntランダム領域(5’-CGTACGGTCGACGCTAGC-(N)25-CACGTGGAGCTCGGATCC-3’)を含有する。フォワードプライマー(FP:5’-CGTACGGTCGACGCTAGC-3’)及びビオチン化リバースプライマー(Bio-RP:5’-ビオチン-GGATCCGAGCTCCACGTG-3’)を選択の際のssDNAの増幅のために合成した。すべてのオリゴは、合成後にHPLCによって精製した。
NGS結果により、高出現率を有する代表的なアプタマーを特異性アッセイのために合成した。これらのアプタマー候補の詳細な配列を表1に列挙した。
酵素結合オリゴヌクレオチドアッセイ(ELONA)をスクレロスチンへのアプタマー候補の結合親和性を決定するために実施した(Drolet,Moon-McDermottら、1996)。同様に、酵素結合免疫吸着アッセイ(ELISA)を、ヒトスクレロスチンへの抗スクレロスチン抗体の結合親和性を決定するために実施した(Engvall及びPerlmann 1971)。精製組換えヒトスクレロスチン160ngを、96-ウエルマイクロタータープレートに100μl PBS中、4℃で一晩、インキュベートすることによってコートした。次にプレートをブロッキング緩衝液(PBS、0.1%ツイーン20及び1%BSA)を用いて1時間、室温でブロックし、セレックスB&W緩衝液(PBS、1mM MgCl2、0.1%ツイーン20及び0.1%BSA)を用いて4回洗浄した。アプタマー候補を95℃で10分間変性させ、使用前に氷上で10分間急速に冷却した。適切な濃度のビオチン化アプタマー/抗体を各ウエルに加え、次にセレックスB&W緩衝液を100μlまで加え、45分間、室温で継続して穏やかに振とうしながらインキュベートした。結合後、プレートを非特異的な及び非常に弱い結合を除去するためにセレックスB&W緩衝液を用いて4回洗浄し、その後PBST+0.1%BSAを用いて4回洗浄した。100μlストレプトアビジン-HRP/ヤギ抗ヒトIgG Fc-HRP(PBST+0.1%BSAに1:10000希釈)を各ウエルに加え、30/60分間インキュベートし、PBST+0.1%BSAを用いて4回洗浄した。50μl TMBを各ウエルに加え、20分間インキュベートした。反応を、50μl 2M H2SO4を加えることによって停止させた。450nmでの吸光度を、マイクロプレートリーダーを用いて測定した(Stoltenburg,Krafcikovaら、2016)。データをオリジンソフトウェア(Origin software)(OriginLab、Northampton、MA)を用いて分析した。非線形カーブフィッティングモデルHyperblを、結合曲線をプロットするために使用した。Hyperblモデルの式は、y=P1×x/(P2+x)であり、P2はKd値である。
Wntシグナル伝達でのスクレロスチンに対するアプタマーの拮抗作用の阻害強度を研究するために、TOP-Wnt誘導ルシフェラーゼレポーターアッセイを骨芽細胞MC3T3-E1細胞において使用した(van Bezooijen,Svenssonら、2007、Shum,Chanら、2011)。
スクレロスチンへの高い親和性及び阻害強度を示したAptscl3を、切断した(表2)。結合親和性及びin vitro阻害強度は、先の研究と同じプロトコールを使用して実施した。Aptscl 3-1、-2、-3、-4及び-5は、それぞれ0.86、0.52、0.2及び0.22nMのKd値を有してスクレロスチンへの高い結合親和性を残していた。一方aptscl 3-6は、この濃度範囲で結合曲線にフィットできず、スクレロスチンへの低い結合能力を示している(図6-1)。さらに、aptscl3-5は、Wntシグナル伝達でのスクレロスチンの拮抗作用への高い阻害強度を保持していた(EC50=28.4μg/ml)(図6-2)。
本発明者らは、スクレロスチンに対するDNAアプタマーを選択し、aptscl 56及びaptscl 3-5と呼ばれ、低ナノモル範囲の解離定数を有してスクレロスチンに特異的且つ堅く結合する2つの切断アプタマーを最終的に開発した。核酸アプタマーのかさ高い2’-O-メチル(2’-OMe)修飾は、臨床例において見られるそれらのヌクレアーゼ耐性の増強及び2本鎖融解温度の上昇のために選択後修飾として以前から使用されている(Fine,Martinら、2005;Gupta,Hirotaら、2014)。逆位dTを用いた3’末端キャッピングも進行中又は完了した臨床試験での疾患治療のためのアプタマーにおける一般的戦略である(Padilla,Sousaら、1999;Ruckman,Greenら、1998)。合わせて、本実施例は、aptscl 56及びaptscl 3-5の血清安定性が2’-OMe及び3’末端逆位dT(3’-idT)修飾によって改善され得るかどうかを評価するものである。
修飾ヌクレオチドは、合成中に導入された。修飾及び未修飾アプタマーの血清代謝安定性を新たに調製されたマウス血清において評価した。すべてのアプタマー試料を10%及び100%マウス血清を用いてそれぞれ37℃で、0、2、4、8、12、24、36、48及び72時間インキュベートした。指定時間に、アプタマー試料をドライアイス浴中で急速凍結し、次いで評価のためにすべての試料を採取するまで-80℃で保存した。すべてのアプタマー試料の安定性は、アガロースゲル電気泳動によって決定できる、インキュベーション後に残っている未処置アプタマーのバンド密度で表された。
修飾及び未修飾DNA配列を、商業的に入手可能な5’-O-DMT-2’-デオキシヌクレオシド(ABz、CAc、GiBu及びT)ホスホラミダイトモノマー、5’-O-DMT-2’-O-メチルヌクレオシド(ABz、CAc、GiBu及びT)ホスホラミダイトモノマー及び/又は5’-O-DMT-2’-F-ヌクレオシド(ABz、CAc、GiBu及びT)ホスホラミダイトモノマーを使用して、K&A H8スダンダードDNA/RNA合成機で、1μmolスケールで合成した(Beaucage及びCaruthers 2001)。aptscl56の修飾配列は、CGGGG TGTGG GTTCG TCGTT AGCTT GATTT GGCAG CTGCCC-idTであり、下線を付けたヌクレオチドは、2’-OMe修飾された。aptscl 3-5の修飾配列は、GCTAG CTGTT GTACA TCGCC TTACG CACGT G-idTであり、下線を付けたヌクレオチドは、2’-OMe修飾された。
すべてのアプタマー試料のバンド密度をモレキュラーイメイジャー(molecular imager)(Bio-Rad)によってアッセイした(Klussmann,Nolteら、1996、Siller-Matula,Merhiら、2012)。結合親和性及びin vitro阻害強度を先の研究と同じプロトコールを使用して実施した。
aptscl 56について、未修飾アプタマーは、10%血清中48時間後では完全に分解され、100%血清中わずか8時間では残存した。2’-OMe及び3’idT修飾aptscl 56は、10%マウス血清中72時間では残存し、12時間後に分解された。72時間では、少量の修飾アプタマーがまだ残っていた(図7)。
2’-OMe及び3’idTを用いた修飾は、治療用ヌクレアーゼ耐性アプタマーに向けたaptscl 56及びaptscl 3-5の開発をさらに促進できる。
これは、ラットでの皮下投与後のスクレロスチンに対するPEG修飾を有する及び有さないアプタマー(PEG40K-aptscl 56及びaptscl 56)の血漿薬物動態を決定するためである。aptscl 56の配列は、CGGGG TGTGG GTTCG TCGTT AGCTT GATTT GGCAG CTGCCC-idTであり、開始CGGG及び末端GCCCの各ヌクレオチドは、2’-OMeを用いて修飾される。これに基づいて、PEG40K(40,000の分子量を有するPEG)が、PEG40K-aptscl 56を得るように5’末端にさらに付着される。
アプタマーaptscl56及びPEG40K-aptscl 56の薬物動態研究を、標準的実験室餌を自由摂取させ、管理された条件(12時間光サイクル、20℃)で飼育された6ヵ月齢メスバージンスプラーグドーリーラットで実施した。ラットをそれぞれ、6.1mg/kg aptscl56及び25mg/kg PEG40K-aptscl 56を用いて単回皮下注射によって処置した。Aptscl56及びPEG40K-aptscl 56は、投与のためにそれぞれ生理食塩水に1.6mg/ml及び6.2mg/mlの濃度で溶解した(Judith M.Healy,Ryan M.Boomerら、2004)。血液試料を各処置群のラット複製(n=6)からさまざまな時点(aptscl56:5分間、15分間、30分間、1時間、2時間、4時間、8時間、12時間、24時間;PEG40K-aptscl 56:30分間、1時間、2時間、4時間、8時間、12時間、24時間、30時間、36時間、48時間、54時間、62時間、70時間、76時間、84時間、96時間、107時間)で回収し、血漿を単離した。プロテイナーゼKによって処置した後、血漿中の残りのaptscl56及びPEG40K-aptscl 56をHPLCによって定量した。
試料調製:およそ800μlの血液を各ラットから眼窩静脈を介して取り、抗凝固剤としてヘパリンナトリウムを含有するチューブ(1.8mlバキュテナー(vacutainer)、BD Biosciences)に回収し、次いで直ちに湿らせた氷の上に置いた(Healy,Lewisら、2004、Perschbacher,Smestadら、2015)。血漿を回収後1時間以内で可能な限り速やかに、6000g、10分間、4℃での遠心分離によって単離し、分析まで-80℃で保存した(Healy,Lewisら、2004、Siller-Matula,Merhiら、2012、Gao,Shenら、2016)。分析の前に、25μl消化緩衝液(60mM Tris-HCl、pH8.0、100mM EDTA及び0.5%SDS)並びに75μlのプロテイナーゼ溶液(10mM Tris HCl、pH7.5、20mM CaCl2、10%グリセロールv/v中1mg/mLプロテイナーゼK)を50μl血漿試料に加えた。次に試料を55℃で一晩、振とうしながらインキュベートした。インキュベーション後、試料を遠心分離(14000rpm;4℃;15分間)し、100μlの上清を取り、HPLCインジェクションバイアルに移した(Siller-Matula,Merhiら、2012)。
aptscl 56についてのHPLC定量の下限は、10ug/mL~360ug/mLの線形濃度範囲を有して10ul/mLであった。皮下投与後のスプラーグドーリーラットにおけるaptscl56アプタマーの排出半減期(Elim.T1/2)の平均値は、1.8時間であった。Tmaxは0.5時間で、Cmaxの平均値は265.5ug/mlであった(図10、表3)。
本研究は、S.D.ラットに皮下投与されたaptscl56-PEG及びaptscl56の薬物動態特徴を、それぞれ調査した。aptscl 56の1.8時間の排出半減期と比較して、PEG40K-aptscl 56の半減期は、65.1時間まで劇的に延長された。これは、PEGがaptscl56のin vivo滞留時間の延長に顕著な効果を有したことを示している。しかし、血漿中のPEG40K-aptscl 56の濃度は、aptscl56の濃度より完全に低かった(図12)。多数の研究は、ペグ化のサイズ拡大が、血液への吸収におけるシステマティックな減少をもたらしたことを実証した(Caliceti 2003、Kaminskas,Kotaら、2009)。血液へのaptscl 56の吸収は、ペグ化により妨げられる可能性がある。
骨粗鬆症ラットにおけるPEG40Kコンジュゲートアプタマーの処置プロトコール。卵巣切除誘発骨粗鬆症ラットでの骨同化における化学修飾aptscl56/aptscl3とPEG40Kとの組合せの有効性を評価するために、3ヵ月齢のメススプラーグドーリーラット70匹を卵巣摘出(OVX、n=50)又はシャム手術(シャム、n=20)に供し、2ヵ月間処置しなかった。処置前に10匹のOVXラット及び10匹のシャム手術ラットをベースライン(OVX-BS及びシャム-BS)として安楽死させた。残りのシャム又はOVXラットにビヒクル(Veh)、PEG40K-aptscl56/aptscl3(25mg/kg)又はPEG40K-無作為配列(RS、25mg/kg)を6週間、1週間に1回、皮下注射した(1群あたりn=10)。すべての動物を最初の注射の6週間後に安楽死させた。安楽死の前に、すべての動物にカルセイン(20mg/kg)を13日目及び3日目に、それぞれ腹腔内注射した。安楽死後に、右大腿骨を回収し、骨微細構造をマイクロCTによって検査した。左大腿骨を、未脱灰切片及びさらなる組織形態学的分析のために回収した。すべての実験を関連する指針及び規制に従って実施し、すべての実験手順はAnimal Ethics Committee and Experimental Safety Committee of Hong Kong Baptist Universityによって承認された。
骨梁について、マイクロコンピュータ断層撮影(マイクロCT)分析は、卵巣切除された(OVX)ベースライン(OVX-BS)ラットと比較して、シャムベースライン(シャム-BS)ラットの遠位大腿及び近位脛骨、遠位及び第5腰椎の海綿骨質は、顕著に高く、海綿骨構造性能は良好である(図13-1~13-4)。PEG40K-aptscl56/aptscl3、無作為配列又はベクターの6回の定期注射を、OVXラットを処置するために1週間間隔で使用した(それぞれ、OVX+aptscl56、OVX+aptscl3、OVX+RS及びOVX+Vehと名付けた)。シャムラットを、ビヒクルを用いて6週間、1週間に1回処置した(シャム+Veh)(図13-1)。遠位大腿、近位脛骨及び第5腰椎の骨梁領域の定量的分析は、OVX+Vehの骨量がシャム+Veh対照と比較して顕著に低減したことを示した(図13-2、13-3及び13-4)。これらのデータは、骨粗鬆症がOVXラットにおいて良好に誘発されたことを示している。

Claims (17)
- スクレロスチンに対するアプタマーであって、配列番号1、3、10又は19~23のいずれか1つの配列を含み、スクレロスチンに特異的に結合する、アプタマー。
- 100nM未満のスクレロスチンに対するKdを有する、請求項1に記載のアプタマー。
- Wntシグナル伝達経路でのスクレロスチンの拮抗作用を阻害することができる、請求項1に記載のアプタマー。
- 細胞ベースWntシグナル伝達アッセイにおいてスクレロスチンの拮抗作用を遮断できる、請求項1に記載のアプタマー。
- Wntシグナル伝達経路での前記スクレロスチンの拮抗作用を阻害することについて、100μg/ml未満のEC50値を有する、請求項1に記載のアプタマー。
- アプタマーにヌクレアーゼ耐性の増強を付与する、及び/又はアプタマーのin vivo半減期を増強する1つ又は複数の修飾をさらに含む、請求項1に記載のアプタマー。
- 前記修飾が3’逆位デオキシチミジン(3’idT)修飾を含む、請求項6に記載のアプタマー。
- 前記修飾が、1つ又は複数の天然に存在するヌクレオチドを、2’-フルオロ、2’-メトキシエチル、2’-メトキシ又は2’-アリルオキシ修飾ヌクレオチドからなる群から選択される修飾ヌクレオチドを用いて置換することを含む、請求項6に記載のアプタマー。
- 前記修飾が、ヌクレオチド間修飾を含む、請求項6に記載のアプタマー。
- 前記修飾が、ヌクレオチド間ホスホロチオエート結合修飾を含む、請求項6に記載のアプタマー。
- 前記修飾が、PEG修飾を含む、請求項6に記載のアプタマー。
- 前記PEGの分子量が1kDa~100kDaである、請求項11に記載のアプタマー。
- 前記PEGの分子量が40kDaである、請求項11に記載のアプタマー。
- 2’-メトキシ(2’-OMe)修飾、3’逆位デオキシチミジン(3’idT)修飾及び/又はPEG修飾を含む、請求項6に記載のアプタマー。
- 請求項1~14のいずれか一項に記載の少なくとも1つのスクレロスチンに対するアプタマー及び薬学的に許容できる担体又は賦形剤を含む医薬組成物。
- スクレロスチン関連疾患を処置するための医薬の調製における、請求項1~14のいずれか一項に記載のスクレロスチンに対するアプタマー又は請求項15に記載の医薬組成物の使用。
- 前記スクレロスチン関連疾患が、骨粗鬆症、骨減少症、骨軟化症、骨形成不全症(OI)、虚血性骨壊死、関節リウマチ、骨折、変形性関節症及びミエローマから選択される、請求項16に記載の使用。
