Description
(1) αシヌクレイン遺伝子を導入したトランスジェニック非ヒト動物であって、導入したαシヌクレイン遺伝子を、前記動物の中年期以降の時期にオリゴデンドロサイトにおいて発現させた、非ヒト動物。
(2) オリゴデンドロサイトにおいて異常リン酸化αシヌクレインの凝集が認められる、(1)に記載の非ヒト動物。
(3) 不随意運動が認められる、(1)または(2)に記載の非ヒト動物。
(4) 導入したαシヌクレイン遺伝子を、前記動物の中年期以降の時期にオリゴデンドロサイトにおいて発現させた後10日間の死亡率が20%以上である、(1)から(3)の何れか一に記載の非ヒト動物。
(5) 薬物の投与によりαシヌクレインの発現が誘導される、(1)から(4)の何れか一に記載の非ヒト動物。
(6) 薬物が、タモキシフェンである、(5)に記載の非ヒト動物。
(7) loxp配列に挟まれたストップコドン配列を上流域に有するαシヌクレイン遺伝子と、オリゴデンドロサイト特異的プロモーターおよび刺激応答性配列に連結したCre遺伝子とを有する、(1)から(6)の何れか一に記載の非ヒト動物。
(8) loxp配列に挟まれたストップコドン配列を上流域に有するαシヌクレイン遺伝子を有する非ヒト動物と、オリゴデンドロサイト特異的プロモーターおよび刺激応答性配列に連結したCre遺伝子を有する非ヒト動物とを交配することにより得られる、(7)に記載の非ヒト動物。
MSAモデル動物は、MSAの治療法や治療薬開発において有用である。MSA患者ではαシヌクレインタンパク質がオリゴデンドロサイト特異的に凝集していることから、αシヌクレインを過剰発現するように設計されたMSAモデルマウスが報告されている(非特許文献1から3)。しかし、従来のMSAモデルマウスでは、αシヌクレインの過剰発現が胎生もしくは幼若期から始まっており、これはヒトのMSA病態と大きく異なる。本発明においては、ヒトのMSA病態により近いモデル動物を作製することを意図して、任意の時期に薬物などの刺激を投与することでαシヌクレインの発現を誘導することができるように設計した。

ヒトのcDNA遺伝子ライブラリーからPCR等の方法によりαシヌクレイン遺伝子断片を得ることができ、これを用いてαシヌクレイン遺伝子をスクリーニングし、全長のαシヌクレイン遺伝子を取得することができる。予めαシヌクレイン遺伝子が挿入された市販のプラスミドベクターを使用してもよい。なお、αシヌクレイン遺伝子の塩基配列情報は、NCBIのウェブサイトにおいてアクセッション番号NP_000336として登録されている。

ES細胞については、ES細胞を適当な培地、例えば胎児ウシ血清を補充したDulbecco’s Modified Eagle’s medium (DMEM)中でフィーダー層に植え付けることができる。ターゲッティングベクターを含む細胞は選択培地を用いて検出することができ、コロニーが成長するまで十分な時間後にコロニーを拾い、相同組換えの発生を分析することができる。ターゲッティングベクターが相同組み換えにより導入されたES細胞をマウス胚盤胞にマイクロインジェクションする。少なくとも1個、通常は約10~15個のES細胞を胚盤胞の胞胚腔にインジェクトすることができる。胚盤胞は4から6週齢の過排卵した雌から排卵3.5日後に子宮を洗い流すことによって得ることができる。マイクロインジェクション後の胚を、仮親となる偽妊娠雌マウスの子宮に移植する。移植後の雌マウスを通常の飼育条件下で飼育し、仔マウスを出産させる。仔マウスの一部(尾の先端や耳介片など)からゲノムDNAを抽出し、PCR法又はサザンブロット法等により、αシヌクレイン遺伝子の導入の成否を確認することができる。
本発明の非ヒト動物は、好ましくは、導入したαシヌクレイン遺伝子を、当該動物の中年期以降の時期にオリゴデンドロサイト特異的に発現させた後10日間の死亡率が20%以上である。
不随意運動については、カメラにてビデオを撮影することにより評価することができる。
また生存観察は目視により行なうことができる。
本発明の非ヒト動物に薬物等を投与することにより、MSAの治療のための薬物のスクリーニングや遺伝子治療の条件検討を行うことが可能である。例えば、本発明の非ヒト動物に候補物質を投与し、化合物の薬効を評価することにより、MSAの治療薬をスクリーニングすることができる。更には、脳室、くも膜下腔からのウイルスベクターなどを用いた遺伝子導入によって、症状の回復を図ることもできると考えられるため、遺伝子治療のモデルとして用いることも可能である。

(1)中程度の発現能が知られているROSA26領域に遺伝子をノックインし、さらに強力な発現誘導を可能にするCAGプロモーターを用いた。
(2)タンパク質の分解を抑制する分解抑制ペプチド(WPRE)を付加した。
マウス胚盤胞株129由来のE14細胞をES細胞系として用いた(Hooper et al.,Nature 326, 292-295 1987)。細胞培養とターゲティング実験は、既報に従って行った(Itohara et al.,Cell 72, 337-348 1993)。作成したターゲティングベクターは複数の制限酵素処理により確認した(図2)。図2に示す通り各種の制限酵素処理後に予想される大きさのバンドが認められた。
150 μg/mlのG418で選択したクローン由来ゲノムDNAをAvrII消化後、5'外部プローブによるサザンハイブリダイゼーション法によってスクリーニングを行った(図3)。図3に示す結果の通り、6個のES細胞で予想される大きさに陽性のシグナルが確認された。
マウスの遺伝子型は、尾から調製したゲノムDNAのPCR解析によって決定した。オリゴデンドロサイト特異的にαシヌクレインを発現するために、Plp1-Cre/ERTマウス(Jacksonラボラトリーより購入)と、上記により選別されたマウス(αSyn flox/flox)とを交配した(図7)。PCR法によりCre陽性およびαSyn flox/flox陽性のダブル陽性のマウス(Plp1-Cre/ERT; αSyn flox/flox)をスクリーニングした。用いたプライマーと予想されるPCR産物の大きさは以下である。

結果はImage J 1.44p software(NIH)を用いて、数値化した。

上述のようにマウスを還流後、脳を摘出した。大脳半球を 4%パラホルムアルデヒド/リン酸緩衝液に漬け、4℃で 2 日間、浸漬固定した。パラフィン包埋した後、4 μmの厚さの切片を作製した。100%キシレンに10分間浸漬することを3回繰り返すことにより脱パラフィン処理を行った。その後、更に100%アルコール2分、90%アルコール2分、80%アルコール2分、70%アルコール2分に浸漬し、さらに蒸留水に浸漬することを2回繰り返して水和処理を行った。このような処理に供した各試料を、以下A、B、Cに記載される抗体を用いて免疫染色を行い、MSAの病理学的特徴について評価した。
MSAの患者では、脳のオリゴデンドロサイト内にαシヌクレインタンパク質が異常蓄積し、それがグリア封入体を形成することが知られている。そこで、MSAモデルマウスおよびコントロールマウスの脳におけるグリア封入体の形成、αシヌクレインタンパク質の蓄積、およびαシヌクレインの分布について観察した(図8)。

前述のように脱パラフィン、水和処理に供された脳切片に対し、抗体反応の前に、内因性のペルオキシダーゼ活性を抑制するため、0.3% H2O2/メタノールによる処理を30分間おこなった。その後PBSで20%に希釈した仔ウシもしくはヤギ血清で1時間インキュベートすることによりブロッキング処理を行った。一次抗体を用いて、室温(約25℃)で12時間反応させた。用いた一時抗体は以下である。一次抗体反応後、脳切片を100 mM Tris-HCl(pH 7.6)、150 mM NaCl(Tris buffered saline (TBS))を使用して洗浄し、その後、二次抗体を一次抗体の時と同様に反応させ、洗浄した。用いた二次抗体は以下である。C2siデジタルカメラ(ニコン)を搭載したECLIPSE Ti共焦点レーザー顕微鏡(ニコン)を用いて染色像を撮影した。Image J 1.44p software(NIH)を用いて、数値化した。

MSA患者における病変の1つに、αシヌクレインタンパク質が異常なリン酸化を受けて細胞質中で凝集する現象が知られている。αシヌクレインタンパク質は、中枢神経細胞に多量に存在し、脳の神経ネットワークを構成する前シナプス末端の機能に重要なタンパク質であるが、異常リン酸化により蓄積して神経ネットワークの障害を生じ、脳機能の低下を招くとされている。
カメラ(COOLPIX P80、ニコン)にてビデオを撮影した。生存観察は目視によりおこなった(図10)。
図10はタモキシフェン投与後のMSAモデルマウス(オス)における生存割合を示すグラフである。カプラン・マイヤー曲線では、MSAモデルマウス(破線25例)もしくはコントロールマウス(実線11例)に対してタモキシフェン投与後の日数(横軸)ごとの生存率(縦軸)を示している。MSAモデルマウスでは投与開始時(100%)から3日後に1匹が死亡した。以降、4日後に1匹、5日後に1匹、6日後に2匹、8日後に1匹、10日後に1匹が死亡した。一方、残りのMSAモデルマウスはタモキシフェン投与後140日の時点まで生存している。コントロールマウスで死亡したマウスはおらず、タモキシフェン投与後140日の時点で全例が生存している。
データは平均±標準偏差で表し、SAS software(SAS Institute)を用いて統計解析した。2群間の解析には Student t 検定、3群間以上の解析には ANOVA検定を用い、有意水準は p< 0.05 とした。
MSA患者に認められるαシヌクレインの異常凝集は従来のモデルA、B、Cに加え、本モデルでも認められる。
MSA患者に認められるαシヌクレインの異常リン酸化がモデルAおよびCで報告されており、本モデルで認められた。
MSA患者では運動失調(不随意運動を含む)が臨床症状として認められる。モデルマウスBおよびCで運動失調の記載が認められた。本モデルマウスでは25匹中4匹に不随意運動が認められた(ビデオあり)。
MSA患者では主に中高年期以降に症状が発症する。従来のモデルでは出生直後からαシヌクレインの過剰産生がおこっている。一方、本モデルマウスではタモキシフェン投与の時期を任意に選べるために、マウスの中高年期(成熟年齢を元にマウスでは50週以降)にαシヌクレインの発現を誘導した。
<オープンフィールドテスト>
マウス行動実験には、MSA マウスおよび対照例マウスを少なくとも6匹用いた。ビデオ追跡システム(CaptureStar、CleverSys Inc.、アメリカバージニア州)を用いて、オープンフィールド試験を行なった。マウスをオープンフィールドアリーナ(30cm×30cm×30cm、明るさ30ルクス)の中央に配置し、10分間自由行動を撮影した。 全移動距離(mm)は、TopScan行動分析ソフトウェア(CleverSys Inc.)によって分析した。各マウスの平均移動速度は、平均速度(mm / s)=移動距離(mm)/ 600秒で算出した。結果を図11に示す。対照例マウスと比べてMSA マウスでは、移動距離及び平均速度とも低下していた。
回転ロッド装置(Accelerating Model、Ugo Basile、Biological Research Apparatus、Varese、イタリア)を使用した。マウスの前肢及び後肢の運動協調、バランスを測定した。訓練期間として、各マウスをロタロッド上に一定速度で5rpmで5分間置いた後、マウスを1日5回を3日間行った。訓練期間にMSAマウス、対照例マウスともにロタロッドの操作を学習した。その後、3週間連続して5分/日/週で4~40rpmの速度レベルで4回テストを行った。ロタロッドから落下する平均時間を記録し、その後の分析に使用した。結果を図12に示す。対照例マウスと比べてMSA マウスでは、ロタロッドから落下する平均時間が短かった。
すべてのデータは平均+標準偏差(SD)として表した。 2群を分析するためにスチューデントのt検定を用いて一元配置分散分析を用いて統計的有意性を評価した。 0.05未満の確率値(p <0.05)を有意とした。0.01以下を顕著に有意とした。

- αシヌクレイン遺伝子を導入したトランスジェニックマウスであって、
導入したαシヌクレイン遺伝子を、中年期より前は発現させずに、前記動物の中年期以降の時期にオリゴデンドロサイトにおいて発現させ、
導入したαシヌクレイン遺伝子を、前記動物の中年期以降の時期にオリゴデンドロサイトにおいて発現させた後10日間の死亡率が20%以上である、
トランスジェニックマウス。 - オリゴデンドロサイトにおいて異常リン酸化αシヌクレインの凝集が認められる、請求項1に記載のトランスジェニックマウス。
- 不随意運動が認められる、請求項1または2に記載のトランスジェニックマウス。
- タモキシフェンの投与によりαシヌクレインの発現が誘導される、請求項1から3の何れか一項に記載のトランスジェニックマウス。
- loxp配列に挟まれたストップコドン配列を上流域に有するαシヌクレイン遺伝子と、オリゴデンドロサイト特異的プロモーターおよびタモキシフェン応答性配列に連結したCre遺伝子とを有する、請求項1から4の何れか一項に記載のトランスジェニックマウス。
- loxp配列に挟まれたストップコドン配列を上流域に有するαシヌクレイン遺伝子を有するマウスと、オリゴデンドロサイト特異的プロモーターおよびタモキシフェン応答性配列に連結したCre遺伝子を有するマウスとを交配することにより得られる、請求項5に記載のトランスジェニックマウス。
