Each of the obtained liquid compositions was evaluated for sterilization or bacteriostatic properties by the following procedure.

〔殺菌又は静菌特性評価〕
(1)サンプル菌液の調製
対象菌には、4種の乳酸菌Weissella viridescens、Leuconostoc mesenteroides、Leuconostoc citreum、及びLeuconostoc pseudomesenteroidesを用いた。対象菌のシングルコロニーを白金耳を用いて数コロニー釣菌し、TSB培地に植菌して30℃で20時間種培養したものを乳酸菌液(107cfu/mL)とした。得られた乳酸菌液を、滅菌済み精製水10mLに0.1mL混合し、さらにこれを10倍希釈した。4種の乳酸菌の10倍希釈液を等量混合してサンプル菌液を調製した。

(2)液状組成物の適用及び菌数測定
塗布直後及び18時間後の液状組成物による静菌特性を調べた。滅菌プラスチックシャーレを二群に分けた。一方の群では、各液状組成物を1.0mLずつ空のシャーレに入れ、コンラージ棒で均一に塗布し、次いでこれにサンプル菌液を1.0mL接種し、20秒間コンラージ棒で混合しながら塗り広げた(0h群)。もう一方の群では、各液状組成物を1.0mLずつ空のシャーレに入れ、コンラージ棒で均一に塗布した後、安全キャビネット内で18時間風乾し、水分とエタノールを完全に蒸発させた。これにサンプル菌液を1.0mL接種し、20秒間コンラージ棒で混合しながら塗り広げた(18h群)。初発菌数は105~106cfu/mLに調整した。サンプル菌液を塗布したシャーレを、5分間静置後、シャーレ内の液体を採取し、MRS寒天培地を用いて菌数を測定した。

B:初発の1/100以下~1/1000超に菌数が減少
C:初発の1/10以下~1/100超に菌数が減少
D:菌数が初発の1/10よりも多い

試験例3
試験例2と同様の手順で、ただしカプリル酸モノグリセリド又はカプリン酸モノグリセリドの濃度を変更して、表6~7記載の液状組成物を調製した。得られた組成物について、参考例と同様の手順で呈味性(苦味)を評価し、また試験例2と同様の手順で殺菌又は静菌特性を評価した。結果を表6~7に示す。カプリル酸モノグリセリド又はカプリン酸モノグリセリドを0.05質量%以上含有する組成物は、苦味がなく、かつ持続的な殺菌又は静菌作用を有していた。

試験例4
試験例2と同様の手順で、ただしウルトラリン酸塩の濃度を変更して、表8~9記載の液状組成物を調製し、試験例3と同様の手順で、呈味性、及び殺菌又は静菌特性を評価した。結果を表8~9に示す。ウルトラリン酸塩の濃度が低い組成物は苦味を有し、また持続的な殺菌又は静菌作用を示さなかった。

室温下で、C8~C12脂肪酸モノグリセリドと精製水を混合し、これにウルトラリン酸Naを加えてpHを調整し、表10に示す組成の液状組成物を調製した。得られた組成物の呈味性(苦味)及び液の均質さ(成分の溶解性)を評価した。呈味性については、参考例と同様の基準で評価した。液の均質さについては、調製直後又は冷蔵庫で一晩放置後の組成物について、成分の析出(白濁又は沈殿)の有無を目視で確認した。

試験例6
様々な素材上における液状組成物の殺菌又は静菌作用を調べた。
試験例2と同様の手順で、液状組成物を調製した(カプリル酸モノグリセリド 0.15%、ウルトラリン酸Na 0.40%、エタノール 40.00%、水 残余;単位は質量%;pH2.4)。サンプル菌液は、試験例2と同様の手順で4種の菌を用いて調製した。
対象素材として、金属(ステンレス板)、ゴム(ゴム手袋)、プラスチック(番重ふた)を準備した。ステンレス板及び番重ふたは、10cm四方に切断して使用した。ゴム手袋は、10cm四方のプラスチック板に皺のないように伸ばして両面テープで貼り、余った部分はカットして使用した。

The rubber gloves were stretched on a 10 cm square plastic plate without wrinkles and attached with double-sided tape, and the excess part was cut and used.

菌数の測定は、各素材につき以下の手順で行った。各対象素材を3群に分けた。第一の群では、液状組成物0.5mLを、コンラージ棒で対象素材の上に均一に塗布し、次いでサンプル菌液0.1mLを接種し、40秒間コンラージ棒で混合しながら塗り広げた(0h群)。残りの2群では、液状組成物0.5mL又は1.0mLを、コンラージ棒で対象素材の上に均一に塗布した後、安全キャビネット内で18時間風乾し、水分とエタノールを完全に蒸発させた。これにサンプル菌液1.0mLを接種し、40秒間コンラージ棒で混合しながら塗り広げた(18h群)。サンプル菌液の初発菌数は105~106cfu/mLに調整した。
サンプル菌液を塗布した対象素材を5分間静置後、無菌パウチに入れ、上から滅菌水10mLを加え、熱シールして上下に5回程度軽く振った。パウチを開封して中の液体を採取し、MRS寒天培地で菌数を測定した。
