Description
a)PUFA含有脂質を有する細胞を含むバイオマスの懸濁液を提供するステップ;
b)(a)の懸濁液を、50℃から70℃の温度、好ましくは55℃から65℃の温度まで加熱し、懸濁液に細胞壁分解酵素を添加し、さらに、必要に応じて酵素が適切に作動するのに適したpH値に調整するステップ;
c)少なくとも1時間、好ましくは少なくとも2時間、より好ましくは2から4時間、温度及びpHを(b)に示される範囲内で維持するステップ;
d)全乾物含量が30から60重量%、より好ましくは35から55重量%、特に40から50重量%に達するまで、100℃以下、好ましくは70℃から100℃、より好ましくは80℃から90℃の温度で水を蒸発させることによって、ステップ(c)で得られた懸濁液を濃縮するステップ;
e)ステップ(d)で得られた懸濁液において、温度を80℃から100℃、好ましくは85℃から95℃、より好ましくは約90℃に調整し、さらに、アルカリ化剤、好ましくは苛性ソーダを添加して、pH値を9.5から11.5、好ましくは10.0から11.0、より好ましくは10.3から10.7に調整するステップ;
f)少なくとも10時間、好ましくは15から40時間、より好ましくは20から36時間、温度を(e)に示される範囲内で維持し、必要に応じてさらなるアルカリ化剤、好ましくは苛性ソーダを添加することによってpH値を9.0から11.5、好ましくは9.0から11.0、より好ましくは9.0から10.5の範囲内で維持し、従って、懸濁液の解乳化を可能にするステップ;
を含む。
Claims (6)
- 多価不飽和脂肪酸(PUFA)含有脂質を得る方法であって、
a)トラウストキトリド科のPUFA含有脂質を有する細胞を含むバイオマスの懸濁液を提供するステップと、
b)(a)の懸濁液を、50℃~70℃の温度まで、又は55℃~65℃の温度まで加熱し、前記懸濁液に細胞壁分解酵素を添加し、さらに、必要に応じて前記酵素が適切に作動するのに適したpH値に調整するステップと、
c)少なくとも1時間、好ましくは少なくとも2時間、又は2~4時間、前記温度及び前記pHを(b)に示される範囲内で維持するステップと、
d)全乾物含量が30~60重量%、又は35~55重量%に達するまで、100℃以下、70℃~100℃、又は80℃~90℃の温度で水を蒸発させることによって、ステップ(c)で得られた前記懸濁液を濃縮するステップと、
e)ステップ(d)で得られた懸濁液において、温度を80℃~100℃、85℃~95℃、又は約90℃に調整し、さらに、アルカリ化剤、又は苛性ソーダを添加して、pH値を9.5~11.5、10.0~11.0、又は10.3~10.7に調整するステップと、
f)少なくとも10時間、15~40時間、又は20~36時間、前記温度を(e)に示される範囲内で維持し、必要に応じてさらなるアルカリ化剤、又は苛性ソーダを添加することによって前記pH値を9.0~11.5、9.0~11.0、又は9.0~10.5の範囲内で維持し、従って、前記懸濁液の解乳化を可能にするステップと、
さらに、PUFA含有油を収集するステップであって、解乳化された前記懸濁液を中和し、続いてそのようにして得られた油含有軽質相を、水、塩及び細胞片含有重質相から分離することを含むステップと、を含み、
ここで、1重量%未満、0.5重量%未満、又は0.1重量%未満の塩化物が使用される、
む方法。 - 前記PUFA含有油を収集するステップにおいて、中和は、酸、又は硫酸を添加して、pH値を5.5~8.5、6.5~8.5、又は7.0~8.0に調整することによって実現され、水、塩及び細胞片含有重質相からの油含有軽質相の分離は、機械的手段によって、60~90℃、又は70~80℃の温度、及び、6~9、又は7~8.5のpH値で実現される、請求項1に記載の方法。
- 1重量%未満、0.5重量%未満、又は0.1重量%未満の有機溶媒が当該方法において使用される、請求項1又は2に記載の方法。
- 90重量%を超える全乾物含量まで前記バイオマスを乾燥させることによって、水、塩、残油及び細胞片含有重質相を乾燥バイオマスに変換するステップをさらなるステップとして含み、乾燥バイオマスへの変換は、30~50重量%の乾物含量まで前記重質相を濃縮し、続いて流動層造粒装置を使用することにより前記バイオマスを噴霧造粒することによって行われる、請求項2又は3に記載の方法。
- 前記懸濁液は、少なくとも80、100、120又は140g/lのバイオマス密度を有する発酵ブロスとして提供される、請求項1~4のいずれか一項に記載の方法。
- 前記細胞壁分解酵素は、プロテアーゼ、セルラーゼ、ヘミセルラーゼ、キチナーゼ、ペクチナーゼ、スクラーゼ、マルターゼ、ラクターゼ、アルファ-グルコシダーゼ、ベータ-グルコシダーゼ、アミラーゼ、リゾチーム、ノイラミニダーゼ、ガラクトシダーゼ、アルファ-マンノシダーゼ、グルクロニダーゼ、ヒアルロニダーゼ、プルラナーゼ、グルコセレブロシダーゼ、ガラクトシルセラミダーゼ、アセチルガラクトサミニダーゼ、フコシダーゼ、ヘキソサミニダーゼ、イズロニダーゼ、マルターゼ-グルコアミラーゼ、ベータ-グルカナーゼ、マンナナーゼ、及びその組み合わせから選択される、請求項1~5のいずれか一項に記載の方法。
