Description
細胞嚢および細胞嚢管の発生
細胞境界および3Dの細胞の移動運動の力学を調べるため、正常初代ヒト乳腺上皮細胞(HMEC)を、本開示の特定の実施形態に従って、再構成された細胞外マトリックス(ECM)の代替である3Dマトリゲルマトリックスに埋め込んだ。3Dマトリゲル中の単一の球状のHMECは、細胞を封入する様々なサイズの円形/不整な形状で細胞外気泡様の嚢を発生させているが、2D環境における細胞は、それらを発生させなかった(図1A)。およそ96%の単一HMEC(6ウェルプレート中で1×103個/ウェル、n=3)が、12時間において細胞外嚢を発生させた。細胞外球状/卵形嚢の直径/長軸は有意に増加し、最大250μmに達した。嚢表面が膜性であるかどうかを確認するために、我々は、HMECにおいて、改良緑色蛍光タンパク質(EGFP)と形質膜タンパク質Ca2+ATPアーゼ2との融合物(EGFP−PMCA2)を一過性に過剰発現した。実際に、EGFP−PMCA2は、細胞の形質膜および細胞外嚢膜の双方に分布しており、このことは、細胞外嚢は膜により封入されていることを立証した(図1A)。これらの細胞外細胞嚢は、伸長して、様々な長さの長い管を形成し得る(図1B)。経時的に、単一の嚢は、自動的に脱細胞し(ecellularized)、緊張した膜を有する複数の形態(球状、卵形、または不整)の無細胞の閉鎖した嚢を残す(図.1C〜1D)。これらの細胞嚢は、様々なサイズであり、最大直径100μmまで大きくなり得る(図1E)。これらの細胞嚢は、膜性である(図1F)。これらの観察所見は、嚢膜は形質膜の外側に独立して位置すること、および無細胞の嚢は細胞なしで存在し得ることを証明した。このこれまで価値を認められなかった、単一の哺乳類細胞により発生する細胞外の膜性の嚢は、細胞嚢と命名した。
細胞嚢の脱細胞(ecellularization)
ほとんどの場合において、細胞嚢の脱細胞(ecellularization)後、細胞は、その無細胞の細胞嚢から完全に分離する。上記無細胞の細胞嚢は、崩壊して、収縮した凹形の円板を形成する(図1C)。時折、細胞嚢の脱細胞(ecellularization)は、排出された細胞のその無細胞の細胞嚢からの不完全な分離をもたらす。
細胞嚢および細胞嚢管は形質膜外の細胞小器官である
本発明者らは、細胞嚢および細胞嚢管は、形質膜の伸長でも、接続された細胞表面でもないことを発見した。それらは、単一細胞により発生され、形質膜とは異なるそれら自身が発生させた膜に囲まれており、それらを発生させた細胞を封入している。細胞嚢および細胞嚢管は、多面的な生体機能に加え、細胞を包む細胞外の膜性の嚢(または管)、ならびに脱細胞(ecellularization)および細胞の侵入の許可を含む独特な特徴を有する。他方、細胞嚢管は、複数の細胞を定方向に輸送するための長い管状の道を提供するが、細胞嚢はそれらを提供しない。細胞嚢および細胞嚢管は、形質膜の伸長ではなく、細胞を包む細胞外の膜性の細胞小器官である。細胞嚢および細胞嚢管は、2D細胞培養においては存在しない。細胞嚢および細胞嚢管は、支持足場および封入された細胞のための被覆の提供、ならびに脱細胞(ecellularization)および細胞の侵入の許可を含む多面的な生体機能を有する。細胞は、細胞嚢、細胞嚢管、および細胞嚢管ネットワークにおいて遊走することができる。細胞嚢および細胞嚢管の自己分解は、細胞の生存、増殖および成長に影響を及ぼさない。脱細胞された(ecellularized)細胞嚢および細胞嚢管は、最大98時間存在することができる。細胞嚢および細胞嚢管は、数十のなどの複数の細胞を収容することができる。細胞嚢管は相互接続し、複数の方向の定方向の細胞トランスロケーションのための開いた管状ネットワークを形成する。上記細胞嚢および細胞嚢管のこれらの特徴は、それらは、総てのこれまでに述べられた細胞小器官とは異なる2種の新規な通性細胞小器官であることを実証した。
細胞嚢管ネットワークの形成
6時間において、単一初代正常HMECは、示したマトリゲルマトリックスにおいて巨大な膜性の短い細胞嚢管を発生させた(図2)。10時間において、HMECは、短い細胞嚢管において遊走し、伸長した細胞嚢管を発生させた(図2)。36時間において、複数のHMECの長い細胞嚢管は、相互接続し、結合節を介してネットワークを形成した(図2)。68時間において、HMECは凝集し、細胞集団を形成し、総てのHMECの細胞嚢管は分解し、残存した細胞嚢管はなかった(図2)。
細胞輸送のための細胞嚢管の幹線道路
細胞は、管状細胞嚢管が細胞トランスロケーションのための幹線道路を提供する膜性の細胞嚢管ネットワークにおいて遊走する(図2〜3)。我々は、急速な細胞溶解および即時画像技術を用いて、細胞嚢管ネットワーク構造を調べた。BCSCの細胞嚢管は、広範囲かつ積極的に相互接続し、交差、開環、閉環、および多くの他の不整な形態を有意に形成し、複数の方向の離れた目的地までの癌細胞の定方向の細胞再配置のための超巨大な管状ネットワークを提供する(図4)。単一細胞および複数の細胞の双方とも、3D環境におけるものよりも、細胞嚢管において速く遊走する。細胞嚢管は、定方向の3D細胞輸送のための幹線道路を形成する。
シンシチン1は細胞嚢管の合併および細胞の侵入を調節する
異なる細胞の細胞嚢は、合併して、巨大な細胞嚢を形成し得、細胞は、他の細胞の細胞嚢に侵入し得る(図5A)。細胞は、他の細胞の細胞嚢に侵入し得、その結果、複数の細胞または細胞集団を内部に有する単一の巨大細胞嚢が生じる(図5B〜5C)。次に、我々は、細胞嚢管の融合、合併および細胞の侵入の根底にある分子機序を探索した。内在性の膜融合タンパク質シンシチン1が、HMEC、細胞嚢および細胞嚢管において発現した。HMECにおけるシンシチン1の一過性のノックダウンは、細胞嚢管の伸長、接続を有意に減少させたが、細胞嚢のイニシエーションを有意に減少させなかった(図5D〜5F)。さらに、シンシチン1のタンパク質レベルは、細胞嚢管の伸長中に増加した(図5G)。これらのデータは、シンシチン1は、細胞嚢管の合併および細胞の侵入を調節することを実証した。
インテグリンサブユニットβ−2(ITGB−2)は細胞嚢管の発生を促進する
インテグリンは、細胞の接着、付着、遊走および浸潤に必須の細胞表面支持分子である。我々は、細胞嚢管の発生中におけるBM−MSC、HMECおよびBCSCにおける26個のインテグリン遺伝子転写物を調べた。ITGB−2は、細胞嚢管の伸長の過程で有意に増加する(図6A)。さらに、遺伝子特異的shRNAを用いたITGB−2の一過性のノックダウンは、細胞嚢管の伸長の有意な減少をもたらしたが、ITGA−8のノックダウンではみられなかった(図6B)。これらのデータは、ITGB−2は、細胞嚢管における細胞遊走を媒介し、かつ、細胞嚢管の伸長を調節することを実証した。
材料と方法
細胞および試薬。初代正常ヒト乳腺上皮細胞(HMEC)を、ATCC(PCS−600−010(商標))から注文した。HMLER(CD44高/CD24低)FA部分集団細胞のBCSCを、以前の記載のとおりに調製した(27)。FITC抱合抗CD44(BD Biosciences;G44−26)抗体およびフィコエリトリン抱合抗CD24(BD Biosciences,ML15)抗体を、フローサイトメトリーを用いたセルソーティングに使用した。MEGM(商標)乳腺上皮細胞成長培地BulletKit(商標)(Clonetics(商標)MEGM(商標)乳腺上皮細胞成長培地およびSingleQuots(商標)キットパッケージ)を、Lonza(CC−3150)から注文した。マトリゲル(商標)基底膜マトリックス(CB−40234)を、Corningから購入した。BD Matrigel Matrix Growth Factor Reduced(GFR、カタログ番号356230)をBD Bioscienceから注文した。
総ての図において:有意差なし、ns、P>0.05;* P<0.05;** P<0.01;*** P<0.001。
Claims (12)
- 3Dマトリックスと、細胞嚢と、細胞嚢管とを含んでなる組成物であって、細胞嚢および細胞嚢管が、前記3Dマトリックスの上部に単一細胞懸濁液を埋め込むことにより発生するものである、組成物。
- 前記細胞嚢および細胞嚢管が、前記細胞嚢および細胞嚢管を発生させる前記細胞を封入する、請求項1に記載の組成物。
- 前記細胞嚢および細胞嚢管が、周囲環境から前記細胞嚢および細胞嚢管に侵入する複数の細胞をさらに含む、請求項1に記載の組成物。
- 前記細胞嚢および細胞嚢管が細胞を欠く、請求項1に記載の組成物。
- 前記3Dマトリックスが生分解性である、請求項1に記載の組成物。
- 前記3Dマトリックスが、エラスチン、ラミニン、コラーゲン、プロテオグリカン、または非プロテオグリカン多糖を含んでなる、請求項1に記載の組成物。
- 前記プロテオグリカンが、ヘパラン硫酸、コンドロイチン硫酸、およびケラチン硫酸を含んでなる、請求項6に記載の組成物。
- 前記非プロテオグリカン多糖がヒアルロン酸を含んでなる、請求項6に記載の組成物。
- 前記3Dマトリックスが、線維性タンパク質、FACITタンパク質、短鎖タンパク質および基底膜タンパク質をさらに含んでなる、請求項1に記載の組成物。
