実験手順
動物
マウスは、イタリアのGenoaにあるIstituto Italiano di Tecnologia(IIT)の動物施設の標準的な実験室条件下で飼育した。すべての実験および手順はItalian Ministry of Health(許可番号056/2013および214/2015−PR)および地方の動物使用委員会(Animal Use Committee)によって承認されており、実験動物の使用および飼育に関する欧州の法律に従って行った。Dicerflox/floxマウス(Murchison et al., 2005)を、Td−Tomato flox/wtノックインレポーターマウス(Jacksonラボストック番号007908;(Madisen et al., 2010))と交配させた。Dicerwt/wt Td−Tomato flox/wt(DicerWT)、Dicerflox/wt Td−Tomato flox/wt(DicerHT)およびDicerflox/flox Td−Tomato flox/wt(DicercKO)マウスを、全ての実験に用いた。Split−Cre−リコンビナーゼを発現するウイルス(Beckervordersandforth et al., 2010)で感染させた時点で、全てのマウスは8週齢であった。1ヵ月後、マウスに5日間、BrdU(50mg/kg)を毎日腹腔内注射した。マウスを屠殺し、BrdU注射の10日後または1ヶ月後に解析した。
6〜8週齢のDicerwt/wt Td−Tomato flox/wt(DicerWT)、Dicerflox/wt Td−Tomato flox/wt(DicerHT)およびDicerflox/flox Td−Tomato flox/wt(DicercKO)マウスの海馬から、成体の神経幹細胞(aNSC)を得て、記載されているように(Babu et al., 2011; Walker and Kempermann, 2014)増殖培地中で増殖させた。CMVプロモーターエンハンサーと融合した構成的チキンβ−アクチンプロモーター(CAG)の制御下の5μgのCre−リコンビナーゼ発現ベクター(pCAGGS−CRE)のヌクレオフェクション(nucleofection)(Amaxa,Lonza)によって、増殖中のaNSCからDicer遺伝子を取り除いた。固定の2時間前に、BrdU(Sigma−Aldrich)を増殖培地中のaNSCに、最終濃度10μMとなるように添加した。分化:aNSCを、FGFを添加(20ng/ml)した培養培地中に、1.2×104細胞/cm2でプレーティングし、24時間培養した。次いで、培地を、レチノイン酸(RA)およびFGF(5ng/ml)と共に、B27を含む培地と交換して24時間培養し、次の4日間にわたってFGF(1ng/ml)と交換して培養した。細胞を、培養中、6日間インビトロで(6 days in vitro(DIV))、分化させた。星状細胞分化培地:aNSCを、増殖因子を含まない10%FBSを含む増殖培地中に、1.2×104細胞/cm2の密度で、プレーティングし、6DIVにわたって培養した。レトロウイルス媒介誘導性ニューロン分化:Ascl1−ERT2を発現するウイルス構築物および感染条件は、以前に記載された通りであった(Braun et al., 2013)。ニューロン分化は、増殖因子の除去および0.5mM OH−TAM(Sigma)の添加によって、2日間誘導した。培地は2〜3時間ごとに交換した。細胞を、OH−TAMへの曝露の、7、14または21日後に固定した。miRNA投与:増殖aNSCを、250nMの「コントロール模倣物」(ネガティブコントロール、CN−001000−01−05; Dharmacon)、または個々の「miRNA模倣物」(Dharmacon)の混合物で、ヌクレオフェクトし(Amaxa、Lonza)、「miRNA模倣物」はそれぞれ25nMであり、ネガティブコントロールと合わせて最終濃度250nMであった。「miRNA模倣物」がプールとしてトランスフェクトされた場合は、等モル濃度を用い、最終濃度は250nMであった。ヌクレオフェクションの24時間後に、細胞を分化培地にプレーティングし、6日後に分析用に回収した。
脳切片の免疫蛍光染色は、背側海馬全体をカバーする6つの切片のうちの1つで行った。一次抗体:ラット抗BrdU(1:200;ab−6326;Abcam)、ウサギ抗ダブルコルチン(1:1000;ab18723:Abcam)、ウサギGFAP(1:1000;Z−0334;Dako)、マウス抗S100b(1:250;Sigma),マウス抗NeuN(1:250;Millipore)。二次蛍光抗体(1/1000;ヤギAlexa488、568、および647nm、Invitrogen)。40倍の対物レンズを有する共焦点A1ニコン(Nikon)倒立顕微鏡SFCで、画像を得た。DG(歯状回)における定量および解析は、NIS−Elementsソフトウェア(Nikon)を用いて行った。細胞培養物についての免疫蛍光法は、以前に記載された通りに実施した(Babu et al., 2011)。一次抗体:ラット抗BrdU(1:200;ab−6326;Abcam)、ヤギ抗ダブルコルチン(1:200;Santacruz)、ウサギ抗ダブルコルチン(1:500;ab18723;Abcam)、ウサギGFAP(1:1000;Z−0334;Dako)、Abcam)、ラット抗Nestin(1:200;BD−Pharmigen)、ラット抗pH3(1:500;Abcam)、ウサギ抗Sox2(1:500;Millipore)、マウス抗S100b(1:500;Sigma)。AlexaFluor488−標識二次抗体(Invitrogen)は、1:1000(ヤギで得られた)または1:500(ロバ抗ヤギ)で用いた。画像は、20倍または40倍の倍率でNikon Eclipse顕微鏡を用いて得た。Image JソフトウェアのCell Counter Plugin(Macbiophotonics)を用いて、計数された細胞を追跡した。
RNA抽出およびcDNA調製:68匹のマウス(それぞれDicer遺伝子型)を、示した時点で屠殺した。DG aNSCを、Neural Tissue Dissociation Kit P(Miltenyi Biotec)を用いて分離し、FACS選別細胞を直ちにRNA抽出のために処理した。増殖培地中のCreをヌクレオフェクトしたaNSC、または分化したaNSCを、示した時点で回収した。全RNAをQIAzolプロトコール(Qiagen)で抽出し、RNAをRNeasy Mini kitまたはmiRNeasy Mini Kit(Qiagen)を用いて製造元の説明書に従い精製した。cDNA合成(mRNA由来)は、ImProm−II(商標)逆転写酵素(Promega)によって得た;cDNA(miRNA由来)は、製造元の説明書に従って、HiSpecバッファー(Qiagen)を用いてmiScript II RTキットで調製した。mRNAは、ABI−7500 Real−Time PCRシステム(Applied Biosystems)において、QuantiFast SYBR Green PCR Kit(Qiagen)を用いて定量した。各サンプルをGAPDHまたはアクチンのレベルに対して正規化した。miRNAは、ABI−7500 Real−Time PCRシステム(Applied Biosystems)において、製造元の推奨事項に従い、Mouse Cell Differentiation&Development miScript miRNA PCR Array(Qiagen)およびmiScript SYBR Green PCR kit(Qiagen)を用いて、あるいは、ViiA 7 Real−Time PCRシステム(Thermo Fisher)で、製造元の推奨事項に従い、TaqMan(登録商標) Array Rodent MicroRNA A Cards Set v3.0(Thermo Fisher)を用いて定量した。
データは平均±平均の標準誤差として表し、Prism 6(GraphPad, San Jose, CA, USA)で分析した。統計的有意性は、2つの実験群については、両側の独立t検定で、評価した。3つ以上の群を有する実験については、ボンフェローニ事後多重比較検定を用いた一元配置分散分析を使用した。P<0.05の場合に、結果は有意であると考えた。
インビボでのSplit Creウイルス媒介Dicer切断は、成体の海馬において、ニューロン新生、ニューロンの成熟および生存を損なうが、星状グリア形成は損なわない。
インビボで成体海馬ニューロン新生におけるDicerの役割を研究するために、本発明者らは、Dicerについての条件付き(conditional)対立遺伝子を有するマウス系統(Dicerflox、(Murchison et al., 2005))と、Cre誘導性レポーターマウス系統 (Td−Tomatoflox、(Madisen et al., 2010))とを交配した。本物の(bona fide)タイプ1aNSCにおいて、Dicerの条件付き切断を得るために、Split−Creウイルス(図1A)を、8週齢のDicerwt/wt Td−Tomato flox/wt(WT)、Dicerflox/wt Td−Tomato flox/wtDicer(HT)、およびDicerflox/flox Td−Tomato flox/wt(DicercKO)マウスのDGに、注射した。このアプローチにより、hGFAPプロモーターとProminin1プロモーターの同時活性に基づいて(Beckervordersandforth et al., 2014)、Split−Creリコンビナーゼをタイプ1aNSCで発現させることが可能になったことが注目されるべきである(図2A)。このようにして、インビボでの成体海馬の顆粒細胞下層(SGZ)および顆粒細胞層(GCL)における細胞運命を、以前に発表されたように分析した(Beckervordersandforth et al., 2014)。
インビトロでDicer切断の効果を評価するために、WT、HTおよびDicerflox対立遺伝子についてホモ接合性(Cre誘導性Td−Tomato対立遺伝子についてはヘテロ接合性)のマウスのDGから、以前に記載された通り (Babu et al., 2011; Walker and Kempermann, 2014)に、初代aNSCを生成し、これらの細胞を単層として培養した(図2B)。Dicerの枯渇は、構成的プロモーターの制御下でCreリコンビナーゼを発現するプラスミド(pCAGGS−Cre)の、ヌクレオフェクションによって得た。これらの細胞におけるCreヌクレオフェクションは、Td−Tomatoタンパク質発現を活性化し(図3A)、効果的にDicer遺伝子座の組換えをもたらした(図3B)。一貫して、Dicerをコードする転写物は、WTaNSCと比較して、DicerHTaNSCにおいて50%まで減少し(図3C、p=0.0082)、DicercKOaNSCにおいてほとんど完全に存在しなかった(図3C、p=0.0003)。その結果、成熟mRNAレベルは、WTaNSCと比較して、DicerHTaNSCにおいて50%まで減少し(図3D、p=0.01)、DicercKOaNSCにおいてほとんど完全に存在しなかった(図3D、残存miRNAレベルはおよそ7%、p<0.0001)。これらの結果は、インビトロで、Dicerflox対立遺伝子の組み換えが、海馬のaNSCからのDicer転写物および成熟mRNAの両方の効率的な枯渇をもたらすことを実証した。
aNSCにおけるDicerRNAおよびmiRNAの効率的な枯渇にもかかわらず、細胞は増殖条件下で培養すると、少なくとも18日間インビトロで(18DIV)維持することができた。DicerHTおよびWTaNSCと比較して、これらの細胞の細胞形態、細胞の培地交換についての必要性、および比率に、大きな違いは見られなかった。さらに、増殖条件下での、2時間のパルス後にBrdUを組み込んだTd−Tomato+細胞の割合(図4A−C)、またはホスホヒストンH3に対する免疫陽性のTd−Tomato+細胞の割合(PH3 図4D−E)、または増殖因子の滴下の際の2時間のパルス後にBrdUを組み込んだTd−Tomato+細胞の割合(図4G−I)に関して、遺伝子型間の差異は観察されなかった。一貫して、培養中に数日間成長曲線を描いた場合に、遺伝子型の間に違いは見られなかった(図4F)。従って、本発明者らは、Dicer/miRNA枯渇はaNSC増殖に影響を及ぼさないと結論付けた。
本発明者らによって得られたインビボデータは、aNSCにおけるDicerの枯渇は、成体マウスの海馬において、ニューロン新生を損なうが、星状グリア形成は損なわないことを示唆している(図1)。そこで、本発明者らは、組換え(Td−Tomato+)DicerWT、HTおよびcKOのaNCSを単離し、インビトロで分化した際のニューロン新生およびニューロンの成熟について調べた(図7A)。本発明者らは、DicerHTおよびcKOのaNSCは、WTaNSCと比較して、有意に少ないDCX+細胞を生成することを見出した(図7B−C、DicerHTでは約12%のDCX+、p=0.04:DicercKOでは約9%のDCX+、p=0.0013)。さらに、本発明者らは、DicerWTおよびHT細胞と比較して、DCX+DicercKO細胞において、神経突起の数、分枝、およびスパインの減少を観察した(図7D)。
本発明者らは、Dicer依存性miRNAまたは他のRNA干渉(RNAi)関連のDicerの機能が、成体海馬ニューロン新生の制御に関与するかどうかを明らかにしようと試みた。最初に、本発明者らは、増殖条件下での、およびニューロン分化の7、14、および21DIV後での、WTaNSCにおけるmiRNA発現の動態を、ウイルス誘導性Ascl1発現によって解析した(Braun et al., 2013)(図9A)。このアプローチにより、aNSCのウイルス感染後に95%のMAP2+ニューロンを得ることができた(図10)。これらの細胞から335個の成熟mRNAをqRT−PCRによって検出し、それらの増殖下の発現レベルおよび動態に従って、それらを、初期ニューロン分化(7DIV)、および後期ニューロン分化(14および21DIV)の3つのグループに分類した(図9B)。増殖またはニューロン分化に関与することが知られているmiRNAは、動的に制御され(図11A−B、(Schouten et al., 2012))、したがって本発明者らのアプローチの妥当性を支持している。
本発明者らが行った研究は、成体の海馬において11種のmiRNAのセットが、星状グリア形成を犠牲にした、aNSCのニューロンへの分化に重要であることを初めて示した。驚くべきことに、これらのmiRNAは、プールとして投与された場合にのみ、既に損なわれたニューロン新生を正常レベルまで救済したが、個々のmiRNAの投与は効果を示さなかった。それゆえ、本発明者らが行った研究は、相乗的に遺伝子調節メカニズムを活性化することによって、aNSCの分化プログラムに影響を及ぼすという、miRNAの協同性の新たな概念に対する実験的証拠を提供した。
U87MG(ATCC HTB−14)は、一般に研究されているグレードIVの神経膠腫細胞株であり、40年以上にわたり少なくとも1,700の文献で解析されており、ヌードマウスの脳内注射で悪性神経膠腫を誘発することが示されている。この細胞株を、J.Pontenおよび准教授が1966年から1969年までの間に、ステージIVの44歳の男性がん患者(ATCC、HTB−15およびATCC HTB−16も参照)から入手した。U87MGのゲノム配列の解析により、他のどの細胞株と比較しても比類のないレベルの突然変異の結果が得られ、ホモ接合性変異を持つ512個の遺伝子が明らかになり、154個のSNV(単一ヌクレオチド変異)、178個の小さなインデル、145個の大きな微小欠失および35個の染色体間転座が含まれていた。
ヒト初代神経膠芽腫細胞株U87MGを、1.2×104細胞/cmでプレーティングして、培養培地で24時間培養した。3つの培地条件を試験した:i)10%FBSを添加したEMEM培地;ii)FBSなしのEMEM培地;および(iii)レチノイン酸を添加したNB。翌日、細胞を、250nMの模倣物(ネガティブコントロール、CN−001000−01−05;Dharmacon)、または、本発明の11種のmRNAのプールで、Lipofectamine2000(Thermofisher)を用いて、製造元の説明書に従いトランスフェクトした。トランスフェクションの24時間後に、細胞をトリプシンで剥がし、計測して、スフェロイドに好適な適切な密度でプレーティングした。およそ500個の細胞を含むスフェロイドを、3Dコラーゲンゲルに移し、加湿チャンバー内で37℃、5%CO2でインキュベートした。ゲル中にスフェロイドを含有させた1、4および6日後に、画像を得た。
神経膠芽腫U87MGスフェロイドをコラーゲンゲル中で6日間培養し、本発明のmiNプールがその浸潤性および増殖特性に影響を及ぼし得るかどうかを試験した。図14に示したように、6日間インキュベーションした後に、本発明のmiRNAプールをトランスフェクトしたU87MGスフェロイドは、全ての試験条件で、コントロールスフェロイドと比較した場合に、中心核の半径の顕著な減少を示し(二元配置分散分析、p<0.05 EMEM−FBS;p<0.01 NB)、10%FBSを添加したEMEMにおいて浸潤性を測定した場合にも、中心核の半径の顕著な減少を示した(二元配置分散分析、p<0.001))。これらの結果は、本発明の選択されたmiRNAプールがヒト神経膠芽腫U87MG細胞株の増殖および転移特性をインビトロで阻害することを示し、グレードIVのヒト神経膠芽腫の浸潤性を阻害するための新しい治療アプローチの始まりを示すものである。
Claims (14)
- 複数のマイクロRNA、並びに医薬的に許容される担体および/または希釈剤および/または賦形剤を含む、医薬組成物であって、複数のマイクロmRNAが、配列番号1のヌクレオチド1〜8を含むマイクロRNA、配列番号2のヌクレオチド2〜11を含むマイクロRNA、配列番号3のヌクレオチド2〜11を含むマイクロRNA、配列番号4のヌクレオチド2〜8を含むマイクロRNA、配列番号5のヌクレオチド2〜11を含むマイクロRNA、配列番号6のヌクレオチド2〜8を含むマイクロRNA、配列番号7のヌクレオチド2〜11を含むマイクロRNA、配列番号8のヌクレオチド2〜11を含むマイクロRNA、配列番号9のヌクレオチド2〜11を含むマイクロRNA、配列番号10のヌクレオチド2〜11を含むマイクロRNA、および配列番号11のヌクレオチド2〜11を含むマイクロRNAを含み、ここで、ヌクレオチド位置は、配列の5’末端を基準に示されている、医薬組成物。
- 以下のマイクロRNAを含む、請求項1に記載の医薬組成物:
hsa−miR−124−3p MIMAT0000422(配列番号1);
hsa−miR−127−3p MIMAT0000446(配列番号2);
hsa−miR−134−5p MIMAT0000447(配列番号3);
hsa−miR−135a−5p MIMAT0000428(配列番号4);
hsa−miR−139−5p MIMAT0000250(配列番号5);
hsa−miR−218−5p MIMAT0000275(配列番号6);
hsa−miR−370−3p MIMAT0000722(配列番号7);
hsa−miR−376b−3p MIMAT0002172(配列番号8);
hsa−miR−382−5p MIMAT0000737(配列番号9);
hsa−miR−411−5p MIMAT0003329(配列番号10);および
hsa−miR−708−5p MIMAT0004926(配列番号11)。 - 医薬として使用するための、請求項1または2に記載の医薬組成物。
- ニューロン分化を、促進、刺激、または増大させることができる医薬として使用するための、請求項1または2に記載の医薬組成物。
- 神経変性疾患の治療的処置もしくは予防、または加齢性神経変性症状の治療的処置もしくは予防、または神経変性関連の認知機能低下の治療的処置もしくは予防、または神経組織の腫瘍性疾患の治療的処置もしくは予防、またはてんかんおよび/または脳卒中により引き起こされた神経組織への損傷の治療的処置もしくは予防において使用するための、請求項1または2に記載の医薬組成物。
- 神経組織の腫瘍性疾患が、神経膠芽腫である、請求項5に記載の医薬組成物。
- 抗うつ薬として使用するための、請求項1または2に記載の医薬組成物。
- ヒトに投与するための、請求項1〜7のいずれか一項に記載の医薬組成物。
- ニューロン分化を、促進、刺激、または増大させる上で、同時に、別々に、または連続的に使用するための組み合わせ剤としての、
複数のマイクロRNA、並びに医薬的に許容される担体および/または希釈剤および/または賦形剤を含む部品キットであって、複数のマイクロmRNAが、配列番号12のシード配列を含むマイクロRNA、配列番号13のシード配列を含むマイクロRNA、配列番号14のシード配列を含むマイクロRNA、配列番号15のシード配列を含むマイクロRNA、配列番号16のシード配列を含むマイクロRNA、配列番号17のシード配列を含むマイクロRNA、配列番号18のシード配列を含むマイクロRNA、配列番号19のシード配列を含むマイクロRNA、配列番号20のシード配列を含むマイクロRNA、配列番号21のシード配列を含むマイクロRNA、および配列番号22のシード配列を含むマイクロRNAを含む、部品キット。 - 前記使用が、神経変性疾患の治療的処置もしくは予防、または加齢性神経変性症状の治療的処置もしくは予防、または神経変性関連の認知機能低下の治療的処置もしくは予防、または神経組織の腫瘍性疾患の治療的処置もしくは予防、またはてんかんおよび/または脳卒中により引き起こされた神経組織への損傷の治療的処置もしくは予防を含む、請求項9に記載の部品キット。
- 神経組織の腫瘍性疾患が神経膠芽腫である、請求項10に記載の部品キット。
- 抗うつ薬として使用するための、請求項9に記載の部品キット。
