Description
治療への応用のための生体外血小板産生は、魅力的な選択肢であるが、依然として大きな技術的課題がある。
a)巨核球または巨核球断片を含む細胞懸濁液を提供し、
b)流体装置のチャネルに前記細胞懸濁液を導入し、
c)巨核球の伸長および断片化並びに血小板放出に適したずり速度(shear rate)を有するフローにチャネル中の前記細胞懸濁液をさらし、
少なくとも1つの巨核球調節因子化合物を前記細胞懸濁液に添加され、前記巨核球調節因子化合物は、巨核球倍数性を増加させる際に直接的または間接的に関与する化合物および/または細胞骨格構築(cytoskeleton organization)に関与する化合物である。
(i)Rho/ROCK経路の阻害剤、
(ii)MLCK経路の阻害剤、
(iii)サーチュイン経路の阻害剤、
(iv)NMM II ATPaseの阻害剤、および
(v)微小管組織化調節剤、
(vi)細胞骨格シグナル伝達阻害剤、
(vii)マトリックスメタロプロテナーゼ阻害剤、
を含むグループから選択される。
a)巨核球前駆体および/または幹細胞、例えば、ヒト臍帯血からのCD34+細胞を提供し、
b)前記巨核球前駆体をおよび/または幹細胞を培養して細胞を膨張させ、膨張した細胞を巨核球に分化させ、
c)細胞を洗浄して培地を除去し、
d)かん流培地中で当該細胞を再懸濁する、
ことにより得られる。
(a)(i)非多孔質壁、(ii)一端に少なくとも1つの入口開口部、および(iii)他端に少なくとも1つの出口開口部、により画定された少なくとも1つのチャネルを含む産生チャンバを有する流体装置を準備し、前記チャネルの前記壁の内壁の少なくとも一部は、巨核球に対して親和性を有するリガンド、例えば、(i)フォン・ヴィレブランド因子(VWF)またはその機能的変異体、(ii)フォン・ヴィレブランド因子の断片を含むポリペプチド、フィブリノゲン、または(iv)フィブロネクチン、でコーティングされており、

(d)チャネルの出口で血小板を収集し、
前記細胞懸濁液に前記巨核球調節因子化合物を添加する方法である。
a)(i)非多孔質壁、(ii)一端に少なくとも1つの入口開口部、および(iii)他端に少なくとも1つの出口開口部、により画定された少なくとも1つのチャネルを含む産生チャンバを有する流体装置を準備し、前記チャネルの壁の内面の少なくとも一部の複数の障害物がテクスチャ加工(textured)されており、

c)前記チャネルの入口から出口へのフローに前記懸濁液をさらすことにより、前記チャネルにおける巨核球の伸長および断片化並びに血小板放出に適したずり速度を生成し、
d)チャネルの出口で血小板を収集し、
前記細胞懸濁液に前記巨核球調節因子化合物を添加する方法である。
(i)ポストの半径rは、好ましくは、50nmから15mmの間、より好ましくは、500nmから1.5mmの間の値であり、
(ii)2つのポストの中心間の最も近い距離pは、少なくとも100nm、好ましくは、100nmから50mmの間の値、より好ましくは、500nmから10mmの間の値、さらに好ましくは、5μmから1mmの間の値、に等しく、
(iii)チャネルの縦方向により規定される最小角度である角度α、および、六方晶系のブラベー格子の格子ベクトルのうちの1つ、例えば、αは、0°から90°の間、好ましくは、0°から30°の間であり、
(iv)任意に、前記ポストは、0<h≦Hのような高さhを有し、Hは、チャネルの断面における2つの対向する壁の間で計測される最小距離である。
a)巨核球または巨核球断片を含む細胞懸濁液を提供し、
b)流体装置のチャネルに前記細胞懸濁液を導入し、
c)チャネル中の前記細胞懸濁液を巨核球の伸長および断片化並びに血小板放出に適したずり速度を有するフローにさらし、前記細胞をチャネルの出口で収集し、血小板産生用のチャネルの入口に再導入する、
ことを含む。
巨核球または巨核球断片を含む細胞懸濁液を提供し、
流体装置のチャネルに前記細胞懸濁液を導入し、
巨核球の伸長および断片化並びに血小板放出に適したずり速度下のフローに前記懸濁液をさらし、
少なくとも1つの巨核球調節因子化合物が前記細胞懸濁液に添加され、前記巨核球調節因子化合物は、巨核球倍数性を増加させる際に直接的または間接的に、および/または細胞骨格構築に関与する化合物である。
血小板産生方法に用いる巨核球を含む細胞懸濁液は、好ましくは、以下により得られる。
(i)巨核球前駆細胞および/または幹細胞、例えば、臍帯血からのCD34+細胞を提供し、
(ii)細胞を増殖させ、および増殖した細胞を巨核球へ分化させるために前記巨核球前駆細胞および/または幹細胞を培養し、
(iii)必要に応じて、細胞を洗浄して培地を除去し、かん流培地に再懸濁させること、
を含む。
本発明者らは、巨核球の倍数性に直接的または間接的に関与することが知られている化合物および/または培養条件下の細胞骨格構築に関与する化合物が、かん流断段階で補足されると、血小板産生を増加させる能力も有することを見出した。
i. Rho/ROCK経路の阻害剤、
ii. MLCK経路の阻害剤、
iii. サーチュイン経路の阻害剤、
iv. NMM II ATPアーゼの阻害剤、
v. 微小管構築調節因子、
vi. 細胞骨格シグナル伝達阻害剤、および
vii. マトリックスメタロプロテイナーゼ阻害剤
巨核球倍数性を増加させることが知られ、巨核球調節因子化合物として用いられる化合物には、Rho/ROCK阻害剤が含まれる。
MLC2リン酸化反応は、主に2つの異なるキナーゼ:ROCKおよびMLCK(ミオシン軽鎖キナーゼ(Myosin Light Chain Kinase))によって調整される。
巨核球倍数性を増加させることが知られ、巨核球調節因子化合物として用いられる化合物は、巨核球倍数性に影響を与えるニコチンアミドのようなサーチュイン阻害剤を含む(Avanziら, British Journal of Haematology, Vol. 164, 867-876, 2014)。
巨核球倍数性の増大を示し、巨核球調節因子化合物として用いられる他の化合物は、巨核球倍数性に影響を及ぼすことが示されているブレビスタンチン(blebbistatin)などのNMMII ATPアーゼ阻害剤を含む(Avanziら, British Journal of Haematology, Vol. 164, 867-876, 2014)。
例えば、Shinら(PNAS, Vol. 108, n°28, 11458-11463, 2011)により実施されたリン酸化NMM IIAを検出する免疫ブロット法、またはLimouzeら(J Muscle Res Cell Motil, Vol.25(4-5), 337-341, 2004)が測定したアクチン活性化MgATPアーゼ活性として測定されるようなNMMII ATPアーゼ活性を削減し、減少させおよび/または阻害する物質を意味する。
巨核球調節因子化合物として用いられる微小管タンパク質は、微小管構築およびその機能的変異体および/またはアイソフォーム(isoforms)に直接的または間接的に関連するタンパク質を含む。
細胞骨格シグナル伝達を阻害することが知られており、巨核球調節因子化合物として用いられる化合物は、アクチン重合阻害剤を含む。
プロテアーゼを阻害することが知られている化合物は、マトリックスメタロプロテイナーゼ(MMP)阻害剤を含む。
チャネルの入口および出口としてさらに開示される少なくとも2つの開口部を含み少なくとも1つのチャネルを有する産生チャンバを備える流体装置が開示されている。チャネルは、巨核球懸濁液または巨核球断片懸濁液が、チャネルの入口から出口へと流れるものである。好ましくは、チャネルは、1つの入口と1つの出口を有し、前記入口から前記出口に巨核球懸濁液を流すためにメインフローを適用することができる。このような、流体装置は、血小板産生のための本発明の方法に用いることができる。
(i)縦軸および横軸に沿った2つのポスト中心間の最も近い距離pとgとは少なくとも2rに等しく、好ましくは、(2r+Dcell/10)から(2r+100×Dcell)の間、より好ましくは、(2R+Dcell)から(2r+10×Dcell)の間の値であり、
(ii)横軸に沿った2つのポスト中心間の距離pは、チャネルに沿って変化し、および/または、縦軸に沿った2つのポスト中心間の距離gは、フロー方向、すなわち、チャネルの入口から出口までの障害物密度を増加させるために、チャネルに沿って変化する。
(iii)チャネルの長手方向と六角形周期構造の単純格子の格子ベクトルの1つとによって定義される最小角度αは、
r≧Dcell/2の場合、αは、arcsin(r/10g)からarcsin(2r/g)の間であり、
r<Dcell/2の場合、αは、arcsin(Dcell/20g)からarcsin(Dcell/g)の間である。
(iv)必要に応じて、前記ポストは、0<h≦Hの高さhを有し、Hは、チャネルの高さであり、例えば、h=Hである。
(i)ポストの半径rは、好ましくは、50nmから15mmの間、より好ましくは、500nmから1.5mmの間の値である。
(ii)横軸に沿った2つのポスト中心間の最近接距離pおよび縦軸に沿った2つのポスト中心間の最近接距離gは、少なくとも100nmに等しく、好ましくは、100nmから50mmの間、好ましくは、500nmから10mmの間、より好ましくは、5μmから1mmの間の値である。
(iii)チャンネルの長手方向と六角形ブラベー格子の格子ベクトルの一つとで規定される最小角度αは、0°から90°の間、好ましくは、0°から30°の間の値である。
(iv)必要に応じて、前記ポストは、0<h≦Hの高さhを有し、Hは、チャンネルの断面の2つの対向する壁の間で測定される最小距離であり、例えば、h=Hである。
チャネル中の前記懸濁液のフローを制御する流速コントローラと、
必要に応じて、産生チャンバの上流で、巨核球懸濁液を濃縮し、巨核球懸濁液を均質化する巨核球ソータ(sorter)および/または巨核球ミキサ(mixer)と、
必要に応じて、巨核球または他の細胞残渣から血小板を選別することにより、流出懸濁液を精製する産生チャンバの下流にある血小板ソータと、
を加えることができる。
本発明はさらに、巨核球または巨核球断片を含む細胞懸濁液から血小板を産生する生体外方法に関する。前記方法は、上述した本発明の流体装置に細胞懸濁液をかん流することにより行われ、必要に応じて、少なくとも1つの巨核球調節因子化合物を補充した細胞懸濁液をかん流することにより行われる。したがって、流体装置のために上述した全ての特定の特徴および実施形態は、当該方法の特徴および実施形態である。
有利には、本発明の流体装置は、新たに産生された血小板の機能的性質を維持しながら、高収率産生で巨核球懸濁液から血小板を産生することができる。

CD34+細胞は、以前に報告されたように(Poirault-Chassacら, “Notch/Delta4 signaling inhibits human megakaryocytic terminal differentiation”, Blood, vol.116, n°25, p.5670-5678, 2010参照)、免疫磁性技術(Miltenyi Biotec, Paris, France)によりヒト臍帯血(UCB)から単離される。これらの血液サンプルは、インフォームド・コンセントおよび我々の学会倫理委員会(our Institute Ethics Committee)の承認を得た後に、ヘルシンキ宣言に従って採取された。CD43+細胞は、15%BIT9500血清代替物(Stem Cells Technologies, Grenoble, France)を添加したイスコフ改変ダルベッコ培地(Iscove modified Dulbecco medium)(IMDM;Gibco Life Technologies, Saint-Aubin, France)、α−モノチオグリセロール(α-monothioglycerol)(Sigma-Aldrich, Saint-Quentin Fallavier, France)およびリポソーム(ホスファチジル−コリン(phosphatidyl-choline)、コレステロール(cholesterol)およびオレイン酸(oleic acid);SigmaAldrich)を含む完全培地中で37℃、5%CO2の湿った環境で培養された。ヒト組み換え幹細胞因子(SCF、20ng/mL;Miltenyi Biotec)およびトロンボポエチン・ペプチド作動薬(thrombopoietin peptide agonist)AF13948(TPO、50nM)(Dunois-Lardeら, “Exposure of human megakaryocytes to high shear rates accelerates platelet production”, Blood, vol.114, n°9, p.1875-1883, 2009参照)が、0日目に培地に1回添加され、続いて、5日目にSCFなしで20nMのTPOが添加された。成熟巨核球は、培養の12から14日後に得られる。培養中およびせん断にさらす直前に形成された血小板の除去は、(Robert A, Cortin V, Garnier A, Pineault N. Megakaryocyte and platelet production from human cord blood stem cells. Methods Mol Biol. 2012; 788: 219-47)で報告された方法に従ったBSA勾配により行った。そして、濃度は、200,000巨核球/mLに調整した。10μMROCK阻害剤(Y27632, Sigma Aldrich, Saint Quentin Fallavier, France)が、培養の8日目に巨核球の培養物に添加される。10μMROCK阻害剤の添加は、マイクロ流体装置の入口チャンバに巨核球懸濁液を導入する前に行われる。6.25μMニコチンアミド(N0636, Sigma Aldrich, Saint Quentin Fallavier, France)が、培養の8日目に巨核球の培養物に添加される。6.25μMニコチンアミドの添加は、マイクロ流体装置の地理口チャンバに巨核球懸濁液を導入する前に行われる。20μMブレビスタンチン(203390, Calbiochem)が、培養の8日目に巨核球の培養物に添加される。20μMブレビスタンチンの添加は、マイクロ流体装置の入口チャンバに巨核球懸濁液を導入する前に行われる。10μMラトルンキュリン−A(sc202691, Santa Cruz Biotechnology)が、培養の8日目に巨核球の培養物に添加される。10μMラトルンキュリン−Aの添加は、マイクロ流体装置の入口チャンバに巨核球懸濁液を導入する前に行われる。25μMGM6001(ab120845, Abcam)が、培養の8日目に巨核球の培養物に添加される。25μMGM6001の添加は、マイクロ流体装置の入口チャンバに巨核球懸濁液を導入する前に行われる。
成熟巨核球懸濁液は、少なくとも300rpmで回転する、オービタルミキサ(orbital mixer)に固定された50mLチューブ(Falcon tube, Dutscher, France)に導入される。オービタルミキサは、懸濁液中の細胞濃度の均質性を維持するために用いられる。チューブ内の巨核球濃度範囲は、少なくとも100mL-1であり、1012mL-1を超えない。
マイクロ流体コンポーネントは、ソフトリソグラフィー・ラピッドプロトタイピング(Xiaら 1998. “Soft lithography”. Annual Review of Materials Science. vol.28, n°1, p.153-184)により製造される。まず。マイクロチャネルのデザインを含むコンピュータ支援設計ファイル(CADファイル)からトランスペアレンシー(transparencies)を作成する。これらのトランスペアレンシーは、典型的なフォトリソグラフィーにより、ネガティブ・フォトレジスト(SU-8 2000 and 3000 series, Microchem, US)にパターンを転写する際のマスクとして用いられ、マイクロチャネルの凸状の起伏(positive relief)を有するマスタが得られる。チャネルは、成形ポリジメチルシロキサン(PDMS, Sylgard, Dow Corning, USA)から作製され、ガラススライド上に密封される。PDMSプレポリマーおよび硬化剤が、混合され、脱気された。混合物がマスターの上に注がれ、70℃で2時間硬化させ、個々のチップにカットし、入口孔および出口孔が打ち抜かれた。ガラススライドをイソプロパノールで洗浄し、乾燥させた。PDMSの個々の構造およびガラススライドの両方を酸素プラズマオーブン(oxygen plasma oven)で処理し、その後密封した。
テクスチャ加工された表面は、チャネル壁(ガラスまたはPDMS)上の3Dパターンで定義される。これらのパターンは、ポスト密度が変化する(Oxy)平面内のディスクアレイを含む。ここでは、これらの可能なバリエーションの2つを提示する。
最初のものは、「勾配配置1」であり、血小板産生を増大させるために、巨核球捕捉を増やすために用いた。距離gは85μmに固定され、距離pはチャネルに沿って85μmから55μmまで変化する。したがって、ポスト密度は、チャネルに沿って徐々に増加する。
2つ目のものは、「勾配配置2」であり、開始距離p=120μmの結果、装置に沿って細胞密度を均質化するために用いた。さらに、この距離pは、チャネルに沿って120μmから70μmまで変化する。これらのバリエーションは、ポスト密度の急激な増加を誘発し、それにより、巨核球捕捉の急激な増加を誘発する。巨核球伸長のための十分な空間を確保するために、距離gを以下のように定義した。

我々は、ガラスまたは(PDMS障害物を含む)PDMS上のどのようなものであれ、チャネル壁上に表面要素を定義する。この表面要素では、平面の単位ベクトルそれに垂直に作用するベクトルnで定義する。(ベクトルn,ベクトルm)は平面デカルト座標系であるように、流体速度vの法線方向および局所方向に接する単位ベクトルmが決定される。壁ずり速度ベクトルγ(s-1)は、ベクトルγ=∂/∂n(ベクトルv・ベクトルm)で定義される。壁ずり速度は、装置内の流速と装置の幾何学的形状との両方によって制御される。
マイクロ流体チップは、倒立顕微鏡(DMI6000 B, Leica Microsystems GmbH, Germany)のステージ上に配置された。コンピュータ支援伝導ステージ制御を用いて、チャネル長に沿った位置を記録した。我々は、実験時間に沿って、観測フィールド位置とそれらの間の交互記録画像とを記録した。微分干渉対物レンズ(Differential interference contrast objective)を用いて、10倍から40倍の間の動画および画像を記録した。CMOS高速カメラ(Fastcam SA3, Photron, USA)を用いて、0.5から1500Hzの周波数で画像を記録した。
ヒト・フォン・ヴィレブランド因子(VWF)は、Laboratoire Francais du fractionnement et des Biotechnologiesの贈り物であった。それは、カルシウムイオンおよびマグネシウムイオンを含まないリン酸緩衝生理食塩水(PBS)(Lonza, Belgium)中で40μg・mL−1に希釈され、密封マイクロチャネルでかん流した。我々は、この表面コーティングを血小板産生チャネルにおいてのみ使用した。
血小板産生数を増加させるには、装置における巨核球の流速が高いことが望ましい。障害物のパターンおよびチャネルの高さについて、チャネル幅を大きくすることにより流速を増加させることができる。PDMSチャネルの機械的制約は、チャネル崩壊を回避する高さに対する最大幅の比を課すので、有効幅を広げるためにチャネルを並列化した。
裸の核および無傷の巨核球は、血小板産生チャネルからの流出懸濁液を連続的にソーティングすることにより血小板から除去することができる。これはマイクロ流体技術により行うことができる。アフェレーシス技術もたくさん考えられる。我々は、ピンチフロー分画法(Takagiら, Lab Chip, vol.5, n°7, p.778, 2005)および決定論的横置換法(Deterministic Lateral Displacement)((L. R. Huangら Science, 304, 987, 2004)を用いた血小板ソーティングの2つの例を与えた。決定論的横置換装置は、1つの入口、球形のポストアレイおよび2つの出口(中央および側方)を含む。この装置は、長さ5cm、幅3mmおよび高さ40μmである。ポストの直径は、85μmであり、ポスト間の間隔は15μmである。ポストのローシフト(row shifting)は、0.05ラジアンの角度を形成する。表面はBSAコーティングで覆われている。細胞は、中央出口で軌道を変えられ、ソートされるが、血小板は、側方出口で軌道を変えられず、ソートされない。細胞懸濁液から血小板を分離する他の技術は、超音波定常波力(ultrasonic standing wave forces)(Antfolkら, Lab Chip, vol 14, 2791-2799, 2014)を用いる技術である。連続的に流れる細胞懸濁液に音響定常波場(acoustic standing wave field)を適用することにより、細胞は、物理的性質に依存する周囲の培地から分離される。その後、血小板をソートすることができる。
勾配配置2のマイクロ流体装置における血小板産生が、マイクロ流体チップなしの24時間静的条件からの血小板産生を含む対照群サンプルと比較された。CD41抗原およびCD42抗原の発現は、フローサイトメータBD蛍光Accuri C6(登録商標)(flow cytometer BD Fluorescence Accuri C6)(BD Biosciences, Le Pont de Claix, France)と用いて特徴付けた。血小板をフルオレセインイソチオシアネート(FITC)共役抗ヒトCD41(αIIb)、R−フィコエリトリン(PE)共役抗ヒトCD42b(GPIbα)(いずれも、Beckman Coulter, Villepinte, France)およびFITC共役抗ヒト活性化αIIbb3(BD Biosciences)により、15分間、22℃で培養し、対照群は、FITCマウスIgG1(Beckman Coulter)、PEマウスIgG1(G1)を用いて行われた。血小板受容体の単色フローサイトメトリー分析は、GPスクリーンアッセイ(GP screen assay)(Biocytex, Marseille, France)を用いて行われた。抗原部位の数は、較正されたビーズ標準曲線(bead standard curve)に基づいて、蛍光強度を血小板に結合した対応するモノクロナール抗体の数に変換することにより決定される。活性化が、PAR−1トロンビン受容体(TRAP6)のペプチド作動薬の存在下で得られたことを除いて、上記のDunois-Larde,の刊行物に以前に報告されたように、フィブリノゲン接着分析(adhesion assay)および落射蛍光法(epifluorescence)を行った。落射蛍光法が、高解像度生体イメージングプラットフォーム(QIClick-F-CLR-12 Digital CCD camera, Q-imaging, Microvisions Instruments, Evry, France)を用いて、494nmおよび522nm(それぞれ、吸収および発光)で解析された。
我々は、(非移動細胞を含む)移動速度(translocation velocity)および伸長プロセスとは無関係に、表面付着巨核球により捕捉巨核球を定義する。我々は、特定のネックレス構造を既に示している巨核球と同様に、糸に通されたビーズ様構造(bead-on-a-thread-like structure)を形成するために巨核球が再組織化を開始する表面付着および伸長巨核球により伸長巨核球を定義する。我々は、図15および図16に示した勾配配置1または勾配配置2を示すマイクロ流体装置に導入された巨核球懸濁液の巨核球捕捉を評価した。結果は、図17および図18に示されている。捕捉巨核球の密度は、テクスチャ加工された領域(σ〜300から500mm−2)の入口で非常に高いが、この密度は、固定されたpで装置に沿って急激に減少する(テクスチャ加工された領域の端部で50mm−2)。対照的に、細胞がポストの勾配を経験する場合、付着巨核球の密度は、各遷移で増加する(距離pの減少に対応する)。細胞の密度が、維持され、テクスチャ加工された領域の端部で高くなる(「勾配配置1」で〜100mm−2)。
血管内皮細胞では、VWFは、GPIb受容体の結合を介して高いずり速度(>1000s-1)にさらされると、循環血小板の移動を可能とする(Huizingaら, Science, vol.297, n°5584, p.1176-1179, 2002)。試験管内において、VWFの吸着により、巨核球、血小板および血小板前駆体がPDMSおよびガラス表面へ移動することが可能となる(Dunois-Lardeら, Blood, vol.114, n°9, p.1875-1883, 2009)。我々は、培養の8日目およびマイクロ流体かん流の2時間の間に添加されたROCK阻害剤を有する巨核球が付着するチャネル表面をコーティングするVWFおよびBSAの効果を比較した。
我々は、表面付着巨核球からの血小板脱落を観察した。巨核球がチャネル壁上を移動すると、VWFとの一時的な相互作用が壁表面で起こり、巨核球からの血小板脱落まで、次第に形態変化が起こる。脱落は、伸長および細胞体の両方が移動している場合に起こる。このプロセスは、上記Dunois-Lardeの刊行物および特許文献8に記載されている。破裂の後、両者は壁面上を移動し続ける。
2時間のかん流の後、ROCK阻害剤を含む巨核球懸濁液を受け取った流体装置中の産生血小板は、ROCK阻害剤を含まない巨核球懸濁液を受け取った流体装置で産生された血小板と比較して優れたいくつかの特性を示した。図5に示したように、CD42b受容体を発現する血小板の割合は、ROCK阻害剤を含む巨核球懸濁液を受け取った流体装置により生成された場合、より高くなった。それらはまた、天然の血小板、すなわち血液から単離された血小板と同等の類似の特性を示した(図6)。興味深いことに、図7において、TRAP−6刺激時に、ROCK阻害剤を含む巨核球懸濁液からの血小板は、一時的活性化に特有であるチューブリン構造を示すのに対し、アクチンフィラメントは、血液から単離されたトロンビン活性化血小板として再構成される。それらは、ROCK阻害剤を添加しない場合に、流動対照群からのサンプルでは観察されない安静構造に戻ることができる。刺激がなければ、図7は、血小板のチューブリンリング特性が、流体装置から収集されたサンプル中に存在するが、静的サンプルからでは存在しないことを示している。事実、静的サンプルは、TRAP−6の存在下および不存在下での糸状仮足(filopodia)および葉状仮足(lamellipodia)の存在を示す。TRAP−6刺激時に、マイクロ流体装置で産生された血小板は、血小板を特徴付けることが知られているものと同様の活性化特徴を受けることができ、例えば、αIIbβ3受容体の活性化構造に特異的なPAC1モノクロナール抗体の結合レベルを増加させる。図8および図9によれば、ROCK阻害剤を含むまたは含まない巨核球懸濁液の両方から産生された血小板は、TRAP−6刺激時にαIIbβ3受容体の活性化構造を示す。
Claims (9)
- 巨核球から血小板を産生する生体外血小板産生方法であって、
(a)非多孔質壁を有し、一端に入口開口部を有し、他端に出口開口部を有する、少なくとも1つのチャネルを含む産生チャンバを有する流体装置を準備するステップと、
(b)巨核球を含む細胞懸濁液を前記チャネルの前記入口開口部に導入するステップと、
(c)血小板放出を目的とする巨核球伸長および巨核球断片化に適した、少なくとも300s-1のずり速度で、前記細胞懸濁液を前記入口開口部から前記出口開口部へ流して、前記出口開口部において、血小板を含み、裸の核および/または巨核球をさらに含み得る懸濁液を取得するステップと
(d)前記チャネルの前記出口開口部で前記懸濁液から血小板を収集するステップと、
(e)前記出口開口部で、未伸長の前記巨核球または前記懸濁液を収集する収集ステップと、
(f)前記収集ステップで収集された未伸長の前記巨核球または前記懸濁液を、再かん流して血小板を産生するために前記入口開口部に再導入する再導入ステップと、
を含み、
前記細胞懸濁液には少なくとも1つの巨核球調節因子の化合物が添加されており、
前記巨核球調節因子は、
i.Rho/ROCK経路阻害剤と、
ii.MLCK経路阻害剤と、
iii.サーチュイン経路阻害剤と、
iv.NMM II ATPase阻害剤と、
v.微小管組織化レギュレータと、
を含むグループから選択される、血小板産生方法。 - 前記流体装置の前記チャネルは、巨核球への結合親和性を有するリガンドでコーティングされ、前記リガンドは、
(i)フォン・ヴィレブランド因子(VWF:von Willebrand factor)またはその機能的変異体、
(ii)フォン・ヴィレブランド因子の断片を含むポリペプチド、
(iii)フィブリノゲン、または
(iv)フィブロネクチン、
を含む、請求項1に記載の血小板産生方法。 - (i)巨核球前駆細胞および/または巨核球幹細胞として、ヒト臍帯血由来のCD34+細胞を準備するステップと、
(ii)準備された前記CD34+細胞を培養して、増殖させ、巨核球へ分化させる分化ステップと、
(iii)培地を除去するために、前記分化ステップ後の前記CD34+細胞を洗浄する洗浄ステップと、
前記洗浄ステップ後に前記CD34+細胞に前記巨核球調節因子の化合物を添加する添加ステップと、
(iv)前記洗浄ステップ後の前記CD34+細胞をかん流培地において再懸濁させるステップと、
により前記細胞懸濁液を得る請求項1または2に記載の血小板産生方法。 - 前記再導入ステップの前に、前記収集ステップで収集された前記未伸長の巨核球を成熟させるために培養する培養ステップを更に含む請求項1に記載の血小板産生方法。
- 前記流体装置の前記チャネルは、正方形または長方形の断面を有する請求項1乃至4のいずれか1項に記載の血小板産生方法。
- 前記チャネルは、5μmから1mmの間、好ましくは、25μmから100μmの間の高さ、および、100μmから5mmの間、好ましくは、300μmから800μmの間の幅を有する請求項5に記載の血小板産生方法。
- 前記産生チャンバは、2〜106個の複数の並列チャネルを有する請求項1乃至6のいずれか1項に記載の血小板産生方法。
- 前記細胞懸濁液は、少なくとも600s-1、好ましくは、少なくとも1000s-1のずり速度下、300s-1から5000s-1の間、または、600s-1から2400s-1の間のずり速度下で、前記チャネルのフローにさらされる請求項1乃至7のいずれか1項に記載の血小板産生方法。
- 請求項1乃至8のいずれか1項に記載の血小板産生方法により得られた、または、得ることができる血小板。
