Description
フィルムの製造
溶媒キャスティング法(solvent casting method)によりゼラチン系フィルムおよび低メトキシルペクチン系フィルムを調製した。
開発した表面の特性解析
本発明のホウ素ドープ抗微生物性ゼラチン系フィルムおよびホウ素ドープ抗微生物性ペクチン系フィルムの表面の表面特性解析を行った。開発したフィルム表面のレオロジー特性、機械的特性および形態学的特性を対照群と比較して検討した。
改良ディスク拡散法
試験する各微生物におけるホウ素化合物の抗微生物活性を測定するために、NCCLS標準ディスク拡散法[24]を改良して使用した。108cfu/mlの細菌、106cfu/mlの酵母および104spor/mlの真菌を含む100μlの溶液を、新しい培養物を用いて調製し、散布法(spreading method)により普通寒天培地(Nutrient Agar)(NA)、サブロー・デキストロース寒天培地(Sabouraud Dextrose Agar)(SDA)およびポテト・デキストロース寒天培地(Potato Dextrose Agar)(PDA)のそれぞれに接種した。グラム陽性細菌の中の黄色ブドウ球菌、グラム陰性細菌の中の大腸菌および緑膿菌、酵母の中のカンジダ・アルビカンス、ならびに真菌の中のクロコウジカビに対して抗微生物活性試験を行った。開発したフィルム表面および対照群を1×1cmのサイズに切断し、接種したペトリ皿の中に置いた。ペトリ皿におけるホウ素含有フィルム材料の配置を図1に示す。接種され且つ改良ディスク拡散法が適用されたペトリ皿を、細菌については24時間、酵母については48時間、36±1℃で培養し、真菌については25±1℃で72時間培養した。改良ディスク拡散法を用いて試験した微生物に対する抗微生物活性は、阻止域(inhibition zone)(微生物が増殖しない領域)を測定することにより評価した。試験したホウ素含有フィルム表面の抗微生物活性試験結果を表1にまとめ、全ての試験は少なくとも2回繰り返した。
抗微生物性試験結果
試験したホウ素化合物の抗微生物活性試験結果を表1にまとめている。全ての試験は少なくとも2回繰り返した。
SEM装置(EVO 40シリーズ、Carl Zeiss、独国)を用いることにより、開発したゼラチン系またはペクチン系ホウ素ドープ食品フィルム包装の形態学的特性を測定した。SEM画像の結果、ホウ素化合物がフィルムにおいて完全に分散していることが観察された。SEM画像サンプルを図9〜図15に示す。
フィルムの引張強度はそのフィルムの機械的強度を示す。生体高分子フィルムの機械的特性は、包装材料としての使用を可能にするのに非常に重要である。ホウ素誘導体をドープすることによりフィルムの機械的特性が変化し得るので、製造したフィルムを物理的特性解析試験にかけた。本発明のフィルム試料の引張強度を表2および表3にそれぞれ示す。得られた結果は、ホウ素誘導体をドープしていない陰性対照試料が他の試料と比較してより低い抵抗力(resistance)を有することを示している。
ゼラチンは熱可逆性ゲル化機構を有するので、ゲル化および溶融温度に対するホウ素化合物の影響はレオロジー試験によって調べられる。ホウ素化合物はゲル化および溶融温度を著しく変化させず、またはゲルの構造を変形させないことが観察される。ゼラチンゲルの溶融温度(Tm)およびゲル化温度(Tg)の変化を表6に示す。
本発明は、ゼラチン系またはペクチン系抗微生物性表面コーティング材料に関する。表面コーティング材料は、フィルム形状で得られ、産業的用途において包装材料として用いられる。コーティング材料として用いられるこの包装は、衛生を必要とする全ての分野において、特に食品産業において用いることができる。食品包装において、食品および外部環境または消費者に接触する表面は、人間の健康に有害でないように抗菌性、抗真菌性および抗カンジダ性の表面である。本発明により、食品包装の表面および内側の両方に存在する全ての種類の病原性因子(細菌、真菌およびウイルス)に対して有効であり、かつ人間の健康または食品の品質に害を及ぼさないコーティング材料が開発される。
Claims (9)
- ホウ酸、五ホウ酸ナトリウムまたは八ホウ酸二ナトリウムを含むことを特徴とする、ペクチン系抗微生物性表面コーティング材料であって、
前記ペクチン系抗微生物性表面コーティング材料がフィルム状であり、1070g〜2170gの平均引張強度を有し、
前記ホウ酸、五ホウ酸ナトリウムおよび八ホウ酸二ナトリウムが、SEM画像で前記ペクチン系抗微生物性表面コーティング材料において完全に分散している、前記ペクチン系抗微生物性表面コーティング材料。 - ホウ酸、五ホウ酸ナトリウムまたは八ホウ酸二ナトリウムを含むことを特徴とする、ゼラチン系抗微生物性表面コーティング材料であって、
前記ゼラチン系抗微生物性表面コーティング材料がフィルム状であり、11375g〜17172gの平均引張強度を有し、
前記ホウ酸、五ホウ酸ナトリウムおよび八ホウ酸二ナトリウムが、SEM画像で前記ゼラチン系抗微生物性表面コーティング材料において完全に分散している、前記ゼラチン系抗微生物性表面コーティング材料。 - 5質量%〜15質量%のホウ酸を含むことを特徴とする、請求項1または2に記載の抗微生物性表面コーティング材料。
- 5質量%〜15質量%の五ホウ酸ナトリウムを含むことを特徴とする、請求項1または2に記載の抗微生物性表面コーティング材料。
- 5質量%〜15質量%の八ホウ酸二ナトリウムを含むことを特徴とする、請求項1または2に記載の抗微生物性表面コーティング材料。
- 大腸菌、黄色ブドウ球菌、緑膿菌細菌に対する抗菌特性を有することを特徴とする、請求項1〜5のいずれか1項に記載の抗微生物性表面コーティング材料。
- カンジダ・アルビカンス酵母に対する抗カンジダ特性を有することを特徴とする、請求項1〜6のいずれか1項に記載の抗微生物性表面コーティング材料。
- クロコウジカビ真菌に対する抗真菌特性を有することを特徴とする、請求項1〜7のいずれか1項に記載の抗微生物性表面コーティング材料。
- 衛生を必要とする包装産業で用いられることを特徴とする、請求項1〜8のいずれか1項に記載の抗微生物性表面コーティング材料。
