処置後48時間の胎児心臓の血管形成:図14に示すように、siRNA処置2日後のCep192 siRNA処置済み心臓は、明らかな血管を有していない。しかしながら、対照群では、移動性の心内膜細胞が心室壁を貫通して、細かい血管ネットワークを形成した。ゆえに、Cep192の枯渇は劇的に、心内膜細胞による血管形成プロセスを阻害する。
実験用のnp−si適用:これは、滅菌生理食塩水または水中にナノ粒子を混合して、10μl以下のアリコート中で標的濃度を達成することによって、実行することができる。溶液を直接、創傷または標的エリアに適用すると、溶液はそこで迅速に吸収される。対照群は、i)非特異的siRNAナノ粒子、およびii)水または生理食塩水のみを含む。2つの異なる処置レジメンをここに例示する:レジメン1において、np−siは、創傷を負わせた30分後から始めて8日目まで毎日投与する。レジメン2において、np−siは、創傷を負わせた30分後から始めて1日おき(0、2、4、6、および8日目)に投与する。

Claims (19)
- 対象において、線維症もしくは瘢痕化を処置する、または線維症もしくは瘢痕化を阻害するための薬剤を製造するための、CEP192の阻害剤の使用であって、CEP192の阻害剤は、CEP192を対象とするRNAi核酸である、前記使用。
- 創傷を有する対象において、創傷の治癒と関連する疼痛を処置するための薬剤を製造するための、CEP192の阻害剤の使用であって、CEP192の阻害剤は、CEP192を対象とするRNAi核酸である、前記使用。
- 前記CEP192は、ヒトCEP192である、請求項1又は2に記載の使用。
- 前記RNAi核酸は、CEP192を対象とするsiRNA、またはCEP192を対象とするshRNAである、請求項1又は2に記載の使用。
- 前記RNAi核酸はsiRNAである、請求項4に記載の使用。
- 前記RNAi核酸はshRNAである、請求項4に記載の使用。
- 前記薬剤は、ナノ粒子と結合したsiRNAを含む、請求項5に記載の使用。
- 前記線維症は、損傷に起因して生じたものである、請求項1に記載の使用。
- 前記線維症は、線維腫、肺線維症、嚢胞性線維症、肝硬変、過去の心筋梗塞に由来する心内膜心筋線維症、心房性線維症、縦隔線維症、骨髄線維症、後腹膜線維症、肺の進行性塊状線維症、塵肺の合併症、腎性全身性線維症、クローン病線維症、ケロイド線維症、皮膚もしくは肺の硬皮症/全身性硬化症、関節線維性癒着、または癒着性関節包炎線維症である、請求項1に記載の使用。
- 前記瘢痕化は、皮膚瘢痕化、心血管瘢痕化、心臓組織瘢痕化、またはニューロン瘢痕化である、請求項1に記載の使用。
- 前記創傷は、皮膚創傷、心血管創傷、心臓組織創傷、またはニューロン創傷である、請求項2に記載の使用。
- 前記皮膚創傷は熱創傷である、請求項11に記載の使用。
- 抗線維化剤を同定する方法であって、Cep192遺伝子産物をコードする核酸を候補剤と接触させること、またはCep192遺伝子産物を候補剤と接触させること、前記候補剤が、前記核酸にコードされる前記Cep192遺伝子産物の発現を阻害する、もしくは前記Cep192遺伝子産物の活性を阻害するかをそれぞれ判定すること、およびその後、前記候補剤が抗線維化剤であるか否かを同定し、Cep192発現またはCep192遺伝子産物を阻害する候補剤が、抗線維化剤として同定される、方法。
- 鎮痛剤を同定する方法であって、Cep192遺伝子産物をコードする核酸を候補剤と接触させること、またはCep192遺伝子産物を候補剤と接触させること、前記候補剤が、前記核酸にコードされる前記Cep192遺伝子産物の発現を阻害する、もしくは前記Cep192遺伝子産物の活性を阻害するかをそれぞれ判定すること、およびその後、前記候補剤が鎮痛剤であるか否かを同定し、Cep192発現またはCep192遺伝子産物を阻害する候補剤が鎮痛剤として同定されることを含む、方法。
- 前記候補剤は、小さな有機分子、ペプチド、核酸、オリゴヌクレオチド、抗体、抗体の抗原結合フラグメント、またはアプタマーである、請求項13又は14に記載の方法。
- CEP192の阻害剤、またはCep192遺伝子産物の阻害剤を含む、線維症を処置する、または瘢痕化を処置することを必要とする対象において、線維症を処置する、または瘢痕化を処置するための医薬であって、前記CEP192の阻害剤は、CEP192を対象とするRNAi核酸である、医薬。
- CEP192の阻害剤、またはCep192遺伝子産物の阻害剤を含む、対象において創傷の治癒と関連する疼痛を処置するための医薬であって、前記CEP192の阻害剤は、CEP192を対象とするRNAi核酸である、医薬。
- 前記RNAi核酸がsiRNAである、請求項16又は17に記載の医薬。
- 前記RNAi核酸がshRNAである、請求項16又は17に記載の医薬。
