Description
アミノ酸配列はアミノ末端からカルボキシル末端へ向かって与えられる。配列位置の大文字は1文字コードのL−アミノ酸を指す(Stryer、Biochemistry、3rd ed.p.21)。
項目1:癌の治療または予防において使用するための、関連するβ2ミクログロブリンを本質的に含まない単離された単一のHLA−B27重鎖ポリペプチドモノマー。
項目2:前記モノマーがペプチドエピトープ断片をさらに含む、項目1に記載の癌の治療または予防において使用するための単離された単一のHLA−B27重鎖ポリペプチドモノマー。
項目3:前記HLA−B27鎖がHLA−B27α1、2および3ドメインのみからなる、項目1または2に記載の癌の治療または予防において使用するための単離された単一HLA−B27重鎖ポリペプチドモノマー。
項目4:HLA−B27鎖が膜貫通ドメインを含み、細胞内ドメイン(細胞質尾部)を含まない、前記項目のいずれか1項に記載の癌の治療または予防において使用するための単離された単一のHLA−B27重鎖ポリペプチドモノマー。
項目5:HLA−B27鎖が、連続して番号を付した配列番号006から配列番号172によって同定される配列のいずれか1つに対して、70%以上、80%以上、85%以上、90%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上または98%以上、または100%の配列同一性を有する、上記項目のいずれか1項に記載の癌の治療または予防において使用するための単離された単一HLA−B27重鎖ポリペプチドモノマー。
項目6:
a.項目1〜5のいずれか1項に記載の単離された単一のHLA−B27重鎖ポリペプチドモノマー、および
b.チェックポイント阻害剤、
を含む併用薬剤。
項目7:前記チェックポイント阻害剤が、CTLA4とCD80またはCD86との相互作用の阻害剤、PD−1とそのリガンドPD−L1との相互作用の阻害剤、およびリガンドTIM−3から、特にCTLA4、CD80、CD86、PD−1、PD−L1またはTIM−3のいずれか1つに対する抗体、より特にヒトCTLA4またはPD−1に対するモノクローナル抗体から選択される、項目6に記載の併用薬剤。

を哺乳類細胞(「産生細胞株」)において共発現する工程;
b.精製工程:得られたHLA−重鎖/β2ミクログロブリン複合体を哺乳動物細胞(産生細胞株)から精製する工程;
c. 解離工程:精製HLA−重鎖/β2−ミクログロブリン複合体を適切な条件下で解離させ、HLA重鎖ポリペプチドをβ2ミクログロブリンポリペプチドから分離する工程;
d.リフォールディング工程:分離されたHLA重鎖ポリペプチドを、(生理的に活性なHLAオープン配座異性体分子において見られるそれらの天然のタンパク質三次構造の)リフォールディングに至る条件下でインキュベートする工程。
項目B:前記HLAをコードする核酸配列が、ポリペプチドのN末端からC末端にかけてα1鎖、α2鎖、α3鎖および安定化配列をコードする配列を含む、項目Aに記載のヒトHLA重鎖ポリペプチドの産生方法。
項目C:前記安定化配列が、ウシ血清アルブミンおよび免疫グロブリン定常フラグメント(Fc)、特に免疫グロブリンG定常フラグメント、より特にIgG4Fcから選択される、項目AまたはBに記載のヒトHLA重鎖ポリペプチドの産生方法。

項目G:前記HLAをコードする核酸配列が安定化配列として免疫グロブリンFc断片を含み、および前記精製工程は、組換えHLA重鎖ポリペプチドをプロテインAに結合された表面に吸着させることによって行われる、前記項目のいずれかに記載の方法。
項目H:前記解離工程は、酸性条件下、特に約pH2での処理、および還元条件下での透析によって行われる、前記項目のいずれかに記載の方法。
項目I:前記リフォールディング工程は、中性条件下の処理で行われる、前記項目のいずれかに記載の方法。
B272−Tet分子は、インビトロにおいて、Treg抑制への作用およびT細胞活性化を通して、ヒトとげっし類の両方において免疫応答を調節する(図1〜3)。
iTregの変換について、HLA分子とナイーブCD4+T細胞との作用を解析した。0.5 μg/mLのB272四量体、HLA−B27四量体、HLA−B8四量体を、iTreg変換について最適な培養条件でナイーブCD4+T細胞とインキュベートした。B272−TetはFoxP3の誘導を下方調節することが示された(図1)。
応答細胞として、紫色にラベルしたナイーブCD8+T細胞を用いて、マウスおよびヒトのTreg細胞の抑制機能を決定した(図2)。TregはHLA四量体およびBSA四量体と共培養し、96時間後のCD8+T細胞の増殖を測定した。CD8+T細胞単独では、期待通り強い増殖を示し、そしてTreg細胞は、コントロール四量体(HLA−B27、HLA−B8およびBSA)とインキュベートしたときはCD8+T細胞の増殖を抑制した。驚くべきことに、B272四量体の存在によるTregの抑制機能の大幅な減少はマウス(図2A)およびヒト(図2B)の両方の抑制アッセイにおけるCD8+T細胞の強力な増殖によって示された。B272−Tetの効果は、マウス(図2C)およびヒト(図2D)の両方で量依存的であった。
インビボ疾患におけるB27オープン配座異性体ホモ二量体の関与を実証するために、B27オープン配座異性体ホモ二量体に特異的な抗体(HD5)を作製した。インビボモデルとして、発明者らは、炎症誘発性リンパ球(Th1、Th17、TNF+CD4+T細胞)の数の増加で、ヒトSpA症状に似た疾患へ進行するSpAのHLA−B27ラットモデルを用いた。HLA−B27ラットリンパ球からのエクスビボデータは、B272−Tetの唯一の存在がCD4+T細胞由来の炎症誘発性サイトカインの発現を誘導できることを示した(図3)。さらに、抗B27オープン配座異性体ホモ二量体抗体(HD5)(図4A〜C)を注入することによって得られたインビボの結果は、インビボでB27オープン配座異性体ホモ二量体分子を遮断することによって免疫応答が調節され、そして異なる器官においてTNF CD4+T細胞(図4B)およびTh17細胞(図4C)の増殖が有意に減少することを示した。
有効な戦略は、治療の観点でいうと、安定した状態でHLA−B27オープン配座異性体タンパク質を産生し、溶解性、安定性、結合活性、半減期を向上させることであり、技術的観点でいうと、哺乳類システムにおけるコスト効率の良い産生および精製である。B272−β2m−Fc複合体の産生は、HLA−B27のα1、2および3ドメインを、B272−β2m−Fcタンパク質の細胞外産生に必要なヒトβ2mベクターと一緒に、ヒトIgG4−Fcベクターカセット(図5A)へ挿入することによって成功した(図5A、B)。チャイニーズハムスター卵巣(CHO)細胞におけるトランスフェクションは、B27−Fcベクターとβ2mベクターの両方を1:3の比率で用いて行った。標準的な抗体精製プロトコール(組換えタンパク質精製ハンドブック、原理および方法、2009.GE Healthcare、18−1142−75)を用いて上清を回収し、B272−β2m−Fcを精製した(図5B)。B27−Fc遊離重鎖からのβ2mの分離は、変性条件でSECまたは透析法によって行った(図6)。B272−Fcのリフォールディングは、リフォールディング緩衝液中の希釈法を用いて行い、ウエスタンブロットにより分析した(図6D)。
毒性試験は6週齢のC57BL/6ワイルドタイプマウスにおいて行った。B272−Fcを0、0.1 mg/kg、1 mg/kg、10 mg/kg の用量で3週間にわたって週2回、腹腔内投与した。PBS群と処置群とを比較すると、健常群に変化はなかった(麻痺、ストレス、不安、無力感、感染の徴候、腹部呼吸、腰掛けまたは毛皮のフリル、体重および便)。リンパ球集合は群間で一定であり、ナイーブおよびエフェクターCD4+T細胞、CD8+T細胞、NK、B細胞ならびに単球は群間の数に変化を示さなかった。このデータに基づいて、本発明者らは同系癌マウスモデルにおけるインビボ試験について、B272−Fcを10 mg/kgで試験を進めることとした。
B272−Fc分子(10mg/kg)の注射を治療計画(図7A)において行った。確立されたプロトコールに従って、MC38−OVAdim断片腫瘍を同系マウスの脇腹に皮下注射した。腫瘍が100 mm3(腫瘍の注射後1〜2週間)に達すると、B272−Fcを腹腔内注射した。3週間にわたって週に2回(図7A)、腫瘍のサイズを測り、リンパ球の浸潤について分析した(図7B〜E)。
確立されたプロトコールに従って、Pan02を1×106細胞、EMT6を1×106細胞およびEG.7を1×106細胞、同系マウス群の脇腹にそれぞれ皮下注射した。マウスは腫瘍体積(Pan02およびEMT6)または体重(EG.7)に従ってグループ分けした。B272−Fcを3日間ごとに7回腹腔内注射し(Q3Dx7)、CTLA4 を選択した間隔で3回注射し(Q3Dx7)(図8)、PD−1を週2回計6回(biwk×3)、注射した(図8)。
癌と戦うB272−Fc分子の使用についての原理は、単独療法またはCTLA4もしくはPD−1抗体との併用療法のいずれかとして、異なる同系癌マウスモデルにおける前臨床実験で証明された。
動物と細胞株
iTregアッセイのためのFoxP3(eGFP)レポーターマウス(B6.Cg−Foxp3tm2Tch/J)はジャクソン研究所から購入し、スイス医科大学動物施設で飼育した。HLA−B27/hβ2mトランスジェニック(33−3系統)(Tg)および野生型(WT)Fischer F344雄ラットをTaconic(Germantown、NY)から入手し、スイス医科大学動物施設で飼育した。野生型C57Bl/6マウスを繁殖させ、バーゼル大学病院動物施設で維持した。MC38−OVAdimは、マウス由来の結腸癌腫瘍細胞株である。
HLA−B27/hβ2mトランスジェニック(33−3系統)(Tg)および野生型(WT)Fischer F344雄ラットを3群に分け、それに応じてランダムにHD5 mAb(抗B272)または抗Her2neuで処置した:a.WT−同腹子(n=8)、b. Tg−HD5(n=10)、c.Tg−コントロール(抗Her2neu)コントロール群(n=10)。各Tgラットに、15週齢または23週齢までHD5または抗Her2neu(コントロール)(10 mg/kg)を毎週腹腔内(i.p.)注入した。WT−同腹仔は治療を受けなかった。
マウスのリンパ球集合を、CD4(FITC−BD Bioscience)、FoxP3 +(efluor 450−eBioscience)、CD3(PE−Cy7−eBioscience)、CD45(PerCP−eBioscience)、CD3(PE−eBioscience)、Granzyme B CD11b(FITC−eBioscience)、CD11c(FITC−eBioscience)、およびTer119(FITC−eBioscience)で染色した。
FoxP3+Treg細胞の検出のための細胞内サイトカイン染色(ICS)を、1%パラホルムアルデヒドを用いて行い、FACS緩衝液中0.1%サポニンで浸透させ、一次抗体(FoxP3、CD4およびCD3)で染色した。
インフルエンザ核タンパク質NP383−391ペプチドエピトープSRYWAIRTR(配列番号174)またはEBV EBNA3CエピトープRRIYDLIEL(配列番号175)の存在下で、β2m有りまたは無しでの制限希釈により、HLA−B*2705オープン配座異性体ホモ二量体およびヘテロ三量体複合体をリフォールディングした。コントロールHLA−B8ヘテロ三量体複合体をhCMVpp65(TPRTGGGAM;配列番号176)でリフォールディングした。ストレプトアビジンまたはストレプトアビジン−PE(Life technologies)を用いてHLA−テトラマーを構築した。ビオチン化BSA(Sigma)をコントロール四量体およびHLA複合体と組み合わせて使用した。
マウスCD4+T細胞におけるFoxp3の発現を誘導するために、F344FoxP3EGFPマウスからの脾臓細胞を回収した。脾細胞を選別してCD4+Tナイーブ細胞を得た。 次いで、5μg/mLの抗CD3mAb(eBioscience)、2μg/mLの可溶性抗CD28 mAb(Biolegend)、10μg/mL TGF−β1(R&Dsystems)および100 IU/mLのIL−2(R&Dsystems)でコーティングした96ウェルプレート内で、細胞を105細胞/200μL/ウェルで96時間培養した。
iTreg変換の最適培養条件におけるマウスナイーブCD4+T細胞を、0.5μgのB272−Tet、HLA−B27−TetおよびHLA−B8−Tetの存在下で72時間インキュベートした。iTreg変換をフローサイトメトリーにより測定した。
CD4+またはCD8+T−エフェクター細胞を、マウスまたはヒトのいずれか(マウスナイーブCD4+T細胞単離キット−、Easy Sep;Dynabeads(登録商標)FlowCompTMマウスCD8−、Life Technologies; Dynabeads(登録商標)CD8 ヒト−、Life Technologies)から精製し、10μM 細胞トレースバイオレット増殖染色(Molecular Probes)。CD3(eBioscience)(3μg/mL)および可溶性CD28(eBioscience)(1μg/mL)抗体が被覆された96ウェルU底プレートにおいて、Tregs(2.5×104)細胞およびT−エフェクター細胞(2.5×104)を96時間インキュベートした。FACS canto IIを用いてT−エフェクター細胞の増殖を測定し、FlowJoバージョン7.6.4の増殖分析ソフトウェアを用いてデータを分析した。
ヒトIgG4−Fcベクター(InvivoGen)およびヒトβ2ミクログロブリン(β2m)にHLA−B27のα1、2および3ドメインを別々のベクターに挿入することにより、B272−β2m−Fcの組換え産物の産生を行った。組換えB272−β2m−Fcの産生は、B27−Fc−ベクターおよびβ2m−ベクターのチャイニーズハムスター卵巣(CHO)細胞への同時トランスフェクションによって行った。B272−β2m−Fcの産生は、Evitria AGに委託した。
N末端からC末端までの完全長HLA−B27α鎖の機能的ドメインは:シグナルペプチド、α1、α2、α3(αドメイン1〜3は下線、α2は太字)、膜貫通ドメインおよび細胞質尾部である。
配列番号001から配列番号172の完全配列および部分配列は付属の配列プロトコルにおいて提供される。
Claims (16)
- 癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体であって、ここで前記ホモ二量体は、第1および第2のモノマーで構成され、または含み、各モノマーは他のモノマーとは独立して、
i.HLA−B27重鎖、および
ii.Fcフラグメント、および
iii.任意に、該HLA−B27重鎖と該Fcフラグメントを繋ぐアミノ酸リンカー
を含む、癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。 - 前記第1および第2のモノマーが同一である、請求項1に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- さらにペプチドエピトープフラグメントを含む、請求項1又は2に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 前記モノマーがさらにペプチドエピトープフラグメントを含む、請求項1または2に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 前記HLA−B27重鎖が、HLA−B27α1、α2およびα3ドメインのみで構成される、請求項1〜4のいずれか1項に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 前記HLA−B27重鎖が膜貫通ドメインを含み、および細胞内ドメイン(細胞内尾部)を含まない、請求項1〜4のいずれか1項に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 前記HLA−B27重鎖が、連続する配列番号6から配列番号172で番号付けられた配列番号によって同定される配列のいずれか1つに対して、90%以上の配列同一性を有する、請求項1〜6のいずれか1項に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 2つのモノマーをジスルフィド結合で連結する、請求項1〜7のいずれか1項に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 前記Fcフラグメントが、免疫グロブリンG型(IgG)、A型(IgA)、D型(IgD)、E型(IgE)、またはM型(IgM)から選択される重鎖定常領域C H 2およびC H 3を含む、請求項1〜8のいずれか1項に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- 前記アミノ酸リンカーが1から50アミノ酸を含み、HLA−B27重鎖とFcフラグメントとを1つの単一ペプチド鎖として連結する、請求項1〜9のいずれか1項に記載の癌の治療または予防において使用するための、HLA−B27融合タンパク質ホモ二量体。
- a.請求項1〜10のいずれか1項に記載のHLA−B27融合タンパク質ホモ二量体、および
b.チェックポイント阻害試薬、
を含む、併用医薬。 - 前記チェックポイント阻害剤が、CTLA4とCD80またはCD86との相互作用の阻害剤、PD−1とそのリガンドPD−L1との相互作用の阻害剤、およびリガンドTIM−3から選択される、請求項11に記載の併用医薬。
- 癌の治療または予防において使用するための核酸分子であって、前記核酸分子が、請求項1〜10のいずれか1つに記載のHLA−B27融合タンパク質ホモ二量体またはHLA−B27融合タンパク質モノマーをコードする、癌の治療または予防において使用するための核酸分子。
- 癌の治療または予防において使用するための、請求項13に記載の核酸分子を含む組換え発現ベクター。
- 癌の治療または予防において使用するための、哺乳動物細胞において作動可能なプロモーター配列の制御下で請求項13に記載の核酸分子を含むウイルス。
- 請求項13に記載の核酸分子を含むインビトロ遺伝子改変宿主細胞。
