Description
本出願は、2013年2月7日に出願された米国仮特許出願第61/762,013号の利益を主張するものであり、その仮特許出願は、その全体が参照により本明細書に援用される。

5つの生物医学的ポリマーの全てにドープされたSNAPは、何らの観察可能な相分離もなしに、緑色の均一かつ透明なフィルムを生じた。10wt%SNAPフィルムは、ポリマーフィルム1mg当たりおよそ0.42μmolのSNAP(または6μmol/cm2)を貯蔵し、一方、5wt%SNAPフィルムは、ポリマーフィルム1mg当たりおよそ0.21μmolのSNAP(または3μmol/cm2)を貯蔵した。
SNAP/E2Asフィルムを用いて、これらのフィルムからの長期NO放出およびSNAP滲出を調べるためにインビトロ調査を行った。経時的なSNAP/E2AsフィルムからのNO放出を、ポリマー相内で分解されたSNAPの量に基づき(すなわち、所与の時点においてフィルムを溶解した後に残存するSNAPを測定することにより)決定した。10wt%フィルム中のSNAPの初期濃度は、420nmolSNAP/mgフィルムであった。図5Aは、N,N−ジメチルアセトアミド(DMAc)に再溶解させた1.0mM SNAP溶液、E2As中10wt%SNAPフィルム、およびDMAcに溶解させたE2AsのUV−Visスペクトルを示している。
乾燥貯蔵の間のE2AsポリマーにドープされたSNAPの安定性も評価した。E2Asに組み込まれたSNAPは、貯蔵の間に熱分解または光化学分解を受ける可能性があり得、したがって使用時に利用可能なNO放出能力を制限し得る。したがって、10wt%SNAP/E2Asフィルムを、乾燥剤と共に冷凍庫中で暗闇の中で、乾燥剤と共に室温において暗闇の中または周囲光の中で乾燥した状態で、ならびに乾燥剤と共に37℃および50℃において暗闇の中で乾燥した状態で、貯蔵した。これらの安定性調査は、累積NO放出実験と類似のやり方で行われ、(本明細書に記載される)様々な時点でポリマー中に残存するSNAPの量を、フィルムをDMAcに溶解させて測定した。その結果は、SNAPが、4ヵ月後にE2Asポリマーマトリックス内で安定であることを示す。例えば2ヵ月後に、冷凍庫(−20℃)中で暗闇の中で貯蔵された10wt%SNAPフィルムは、室温で貯蔵されたフィルムについての89%および37℃で貯蔵されたフィルムについての82%と比較して、初期SNAP化学種の約96%を維持する(図8参照)。図8に示される結果は、貯蔵の間のSNAPの向上した安定性を説明している。第3級RSNO(例えば、SNAP)は、硫黄原子の周りの立体障害により、第1級RSNOよりも高い安定性を有している。E2Asポリマーの安定化効果と組み合わせたSNAPの増大した熱安定性が、SNAP/E2As物質の優れた貯蔵安定性をもたらす。

E2As中5wt%SNAPとE2Asの上塗りとで被覆された活性ECCループ(これの概略断面図が図10に示されている)およびE2Asのみで被覆されたコントロールループを調製した。一つにはECC実験の短い継続期間のため、5wt%SNAPをこれらの試験において使用した。上に記載したように、SNAP/E2Asコーティングは、浸漬の初日の間に溶液中へのSNAP拡散の初期噴出を有する。短期ECC実験の間のこの噴出の影響を低減するために、まず全てのループを生理食塩水中に一晩浸漬し、ECC実験の前にその浸漬溶液を捨てた。被覆ECCループの試料から放出される酸化窒素を、(生理食塩水中への一晩の浸漬後の)血液曝露前のNO放出について、NOAを用いて測定した。このSNAP/E2As被覆ループのNO放出は、(37℃において周囲光の場合に)4時間にわたり約2×10−10mol cm−2 分−1の平均フラックスを維持する(図11A参照)。流れる血液への4時間にわたる曝露後に、このECCループは、さらなる1時間の期間にわたり、少なくとも1.5×10−10mol cm−2 分−1のNOフラックスを依然として示す(図11B参照)。
4時間にわたるECCを通じての血小板の活性化および機能を、血小板数(例えば、消費量、図12A参照)および血漿フィブリノーゲンレベル(図12B参照)(これらの両方が、添加されたIV流体による血液希釈について補正された)ならびに%血小板凝集を記録することにより評価した。ベースライン血小板数(×108血小板/mL)は、5wt%SNAP/E2As回路およびE2Asコントロール回路について、それぞれ3.5±0.6および4.8±0.5であった。SNAP/E2As回路については、血小板数は最初に僅かに増加し、ECC上での4時間の終わりにベースラインレベルの100±7%で維持された。コントロール回路についての血小板数は、血小板の時間依存的喪失を示し、4時間後にはベースラインの60±6%まで減少した。血小板の機能的凝集のパーセントを、PRPのエキソビボコラーゲン刺激により決定し、光学的濁度により測定した。SNAP/E2As回路およびコントロール回路を通る循環から採取された血液からの血小板は、4時間にわたる血液曝露の間、コラーゲン刺激血小板凝集に対して同様の反応を示し、両方とも56±12%を維持した(ベースライン値は68±6%)。
本開示はさらに、市販のSRまたはPUチューブにSNAPを添加するための方法を含む。市販のSRおよびPUには、医用グレードのチューブ(US plastics、Cole Palmer、Professional Plastics、ICORally、およびThermedics,Inc.から入手可能なものを含む)が含まれる。図15は、1または2日間のいずれかにわたりSNAP/アセトン溶液中に浸漬されたポリウレタン(PU)チューブのNO放出プロファイルを示している。チューブは次いでアセトンですすがれ、NOA試験の前に乾燥された。NOA試験のために、チューブは37℃においてPBS中に浸漬された。
図16Aは、本開示の実施例に従って5wt%または10wt%SNAP/E2Asを用いて調製され、続いてE2Asの上塗りが適用された、E2Asカテーテルチューブの概略図である。図16Bは、このカテーテルチューブの断面図であり、各種の層を図示している。一般的に、3層カテーテルは、E2As溶液の5枚の下塗り、それぞれの活性溶液の25枚の被膜、およびE2As溶液の5枚の上塗りを浸漬被覆することにより調製された。このSNAP/E2Asカテーテルは、37℃で暗闇の中においてリン酸緩衝生理食塩水(PBS)中で維持された。
N−アセチル−DL−ペニシラミン(NAP)、塩化ナトリウム、塩化カリウム、リン酸ナトリウム(2塩基)、リン酸カリウム(1塩基)、エチレンジアミン四酢酸(EDTA)、テトラヒドロフラン(THF)、硫酸およびN,N−ジメチルアセトアミド(DMAc)は、Sigma−Aldrich(St.Louis,MO)から購入した。メタノール、塩酸および硫酸は、Fisher Scientific(Pittsburgh,PA)から入手した。Tecophilic(商標)SP−60D−60およびTecoflex(商標)Sg−80Aは、Lubrizol Advanced Materials Inc.(Cleveland,OH)の製品であった。Dow Corning RTV3140 シリコーンゴム(SR)は、Ellsworth Adhesives(Germantown,WI)から購入した。CarboSil(登録商標)20 90Aは、Polymer Technology Group(Berkeley,CA)からのものであった。Elast−Eon(商標)E2Asは、AorTech International,PLC(Scoresby,Victoria,Australia)から入手した。>90%凝固可能タンパク質を含有するヒト血漿フィブリノーゲンは、Calbiochem(La Jolla,CA)の製品であり、変性ヒトフィブリノーゲンに対するフルオロセイン標識ヤギIgG(ポリクローナル)は、MP Biomedicals,LLC(Solon,OH)から購入した。蛍光測定に使用した黒色ポリプロピレン96ウェルマイクロタイタープレートは、Nalge Nunc International(Rochester,NY)から入手した。全ての水溶液を、Milli−Qフィルター(Millipore Corp.,Billerica,MA)を用いて18.2MΩ脱イオン水を用いて調製した。138mM NaCl、2.7mM KCl、10mM リン酸ナトリウム、100μM EDTAを含有するリン酸緩衝生理食塩水(PBS)(pH7.4)を、全てのインビトロ実験のために使用した。
先に報告された方法(I.Chipinda,R.H.Simoyi,Journal of Physical Chemistry B 2006,110,5052)の修正版を用いて、SNAPを合成した。簡潔に述べると、等モル比のNAPおよび亜硝酸ナトリウムを、2M HClおよび2M H2SO4を含有する水とメタノールとの1:1混合物に添加した。30分間にわたる撹拌後、反応容器を氷浴中で冷却して、緑色SNAP結晶を沈澱させた。結晶を濾過により収集し、水で洗浄し、風乾させた。この反応および結晶は、常に光から保護された。
5wt%および10wt%のSNAPを含有するポリマーフィルムを、溶媒エバポレーションにより調製した。10wt%SNAPフィルムについては、THFに180mgのそれぞれのポリマーを溶解させることにより流延溶液を調製した。ポリウレタン(SP−60D−60、SG−80A、CarboSil(登録商標)およびElast−Eon(商標)E2As)は3mLのTHFに溶解させ、SRは1mLのTHFに溶解させた。次いで、SNAP(20mg)をこのポリマー溶液に添加し、この混合物を10分間撹拌した。5wt%SNAPフィルムは、SNAP(10mg)およびポリマー(190mg)を用いて同様に調製した。このフィルム溶液を、Teflon(登録商標)プレート上のTeflon(登録商標)リング(直径=2.5cm)中で流延し、周囲条件下で一晩乾燥させた。小円板(直径=0.7cm)を親フィルムから切り取り、上塗り溶液(4mLのTHF中の200mgのそれぞれのポリマー(SNAPは添加されていない))で2回浸漬被覆し、一晩乾燥させた。そういうものであるから、各試料のための上塗りは、親フィルムと同じポリマーでできていた。各小円板の重量を、上塗り被覆前に記録した。全てのフィルムおよびフィルム溶液を光から保護した。最終フィルムは、厚さ約150μmのSNAPドープ層および厚さ約50μmの上塗りを有していた。
インビトロウサギ調査において採用されたECC構成は、16ゲージおよび14ゲージのIVポリウレタン血管カテーテル(Kendall Monoject Tyco Healthcare,Mansfield,MA)、2つの長さ16cmの内径(ID)1/4インチTygon(登録商標)チューブ、およびより荒い血流のためより容易に血栓が形成し得る血栓形成チャンバを作る長さ8cmのID3/8インチTygon(登録商標)チューブからなった。
<UV−Visスペクトル>
室温で、UV−Vis分光光度計(Lambda 35,Perkin−Elmer,MA)を用いて、200nm〜700nmの波長範囲において全てのUV−Visスペクトルを記録した。SNAPのS−NO基の存在は、π→π*電子遷移およびnN→π*電子遷移に相当する特徴的な吸光度極大を340nmおよび590nmにおいてもたらす。
上塗り被覆フィルムを、個々のバイアルに入れ、微量金属イオンにより触媒されるあらゆるSNAP分解を最小限にするために100μM EDTAを含有する10mM PBS(pH7.4)中に浸漬した。フィルムを、室温(22℃)または37℃において暗闇の中でインキュベートした。様々な時点で、SNAP濃度の迅速な測定のために、PBSの1mLアリコートのUV−Visスペクトルを測定した。実験の継続期間の間、これらのアリコートは光から保護され、試料バイアルに直ちに戻された。フィルムは、毎日新しいPBS緩衝液に入れられた。340nmにおけるPBS中のSNAPについてのモル吸光係数は、εSNAP=1024M−1cm−1であると決定された。
E2As中10wt%SNAPフィルムを調製した後に、フィルム内のSNAPの初期濃度(nmolSNAP/mgフィルム)を決定するために、DMAcに溶解させた個々のフィルムのUV−Visスペクトルを記録した。次いで、同等のフィルムを、100μM EDTAを含有する3mLのPBS(pH7.4)を含んだ個々のバイアルに入れた。フィルムを、以下の様々な条件下でインキュベートした:周囲光下で室温、周囲光下で37℃、暗闇の中で37℃、および100W投光照明下で37℃。実験室中の蛍光灯を周囲光と呼ぶ。フィルムは、毎日新しいPBSに入れられた。様々な時点で、フィルム中に存在するSNAPの迅速な測定のために、フィルムをDMAcに溶解させた。放出されたNOの量を、様々な時点で、分解されたSNAPの量から間接的に測定した。経時的な累積放出NO([NO]t)を、フィルム中の初期SNAP量([SNAP]0)と時間tにおけるSNAP量([SNAP]t)との間の差異によって計算した:[NO]t=[SNAP]0−[SNAP]t(ここで、濃度は、nmol/mgフィルムの単位である)。この計算は、SNAPの340nm吸収帯の減衰が、S−NO結合の溶血開裂および付随するNO放出と直接関係しているという事実に基づいた。340nmにおけるDMAc中のSNAPについてのモル吸光係数は、εSNAP=1025M−1cm−1であると決定された。
フィルムから放出される酸化窒素を、Sievers化学ルミネセンス酸化窒素分析器(NOA)280(Boulder,CO)を用いて測定した。フィルムを、試料容器に入れ、100μM EDTAを含有するPBS(pH7.4)中に浸漬した。酸化窒素を、N2スイープガスおよびバブラーを用いて連続的に緩衝液から取り除き、上部空間から化学ルミネセンス検出チャンバ中に一掃した。周囲光および光開始NO放出実験のために、透明ガラス試料容器を使用した。光分解実験のために、100Wハロゲン投光照明(GE型式17986)を試料セルから約60cmの所に設置した。フィルムは、NOA測定と同じ条件(37℃で周囲光または100W投光照明照射)下においてPBS中でインキュベートされた。
SNAP/E2Asフィルム(10wt%SNAPからなる)を、乾燥剤を加えたバイアル中で以下の条件下に置いた:周囲光を用いて室温、暗闇の中で室温、暗闇の中で37℃、および冷凍庫(−20℃)中で暗闇の中。4ヵ月の期間の間の様々な時点で、フィルムをDMAcに溶解させ、UV−Visスペクトルを記録して、初期10wt%SNAPと比較した場合のフィルム中に残存するSNAPの%を決定した。
インビトロフィブリノーゲン吸着免疫蛍光アッセイを、96ウェルフォーマットにおいて行った。ECC回路を調製するために使用されたSNAP/E2Asポリマー溶液およびE2Asコントロールポリマー溶液を、96ウェルマイクロタイタープレートのマイクロウェルを被覆するためにも採用し、ECCループと同じ条件下で乾燥させた。簡潔に述べると、ヒトフィブリノーゲンを、CaCl2およびMgCl2を含まないダルベッコリン酸緩衝生理食塩水(dPBS)(Gibco Invitrogen,Grand Island,NY)を用いてヒト血漿濃度に相当する3mg/mLとなるように希釈し、次いで吸着実験のために使用した。100μLのこの溶液を各ウェルに加え、被覆ウェルをこの溶液と共に37℃で1.5時間インキュベートした。この後に、0.05%Tween 20(Calbiochem La Jolla,CA)を含有するAbD Serotec Block ACE緩衝液(Raleigh,NC)の10倍希釈物からなる洗浄緩衝液(100μL)を各回の洗浄のために用いる8回の洗浄工程が続いた。非特異的抗体結合を阻止するために、被覆ウェルを100μLの阻止緩衝液(Serotec Block ACE緩衝液の4倍希釈物)と共に37℃で30分間インキュベートした。洗浄緩衝液(1ウェル当たり100μL)での3回のすすぎの後に、プレートのバックグラウンド蛍光測定を、Synergy 2蛍光マイクロプレートリーダー(Biotek Winooski,VT)により485nm(励起)および528nm(発光)において行った。吸着されたフィブリノーゲンを検出するために、フルオレセイン標識ヤギ抗ヒトフィブリノーゲン抗体をSerotec Block ACE緩衝液の10倍希釈物で希釈し(1:10)、100μLのこの最終溶液を各ウェルに加えた。表面吸着されたフィブリノーゲンに、37℃で1.5時間にわたり抗体を結合させた。非被覆ポリプロピレンへのヒトフィブリノーゲン吸着を、異なるプレート内の蛍光シグナルを正規化するための内部コントロールとして使用した。全ての測定を三重反復試験で行った。
採用された全ての動物取扱手順および外科的手順は、大学および連邦政府の規則に従ってUniversity Committee on the Use and Care of Animalsにより承認されたものである。合計8匹のニュージーランド白ウサギ(Covance,Battle Creek,MI)をこの調査で使用した。全てのウサギ(2.5kg〜3.5kg)に、5mg/kgのキシラジン注射可能薬物(AnaSed(登録商標)Lloyd Laboratories Shenandoah,Iowa)および30mg/kgの塩酸ケタミン(Hospira,Inc.,Lake Forest,IL)の筋肉内注射で最初に麻酔をかけた。維持麻酔は、気管切開によってA.D.S.2000 Ventilator(Engler Engineering Corp.Hialeah,FL)を用いて行われた機械換気による1.5%〜3%の速度でのイソフルランガス吸入により投与した。最大吸気圧を15cmH2Oに設定し、人工呼吸器流量を8L/分に設定した。血圧安定性の維持を助けるために、乳酸加リンゲルのIV流体を、10mL/kg/時間の速度で投与した。血圧をモニタリングするためおよび血液試料を採取するために、ウサギの右頸動脈に、16ゲージのIV血管カテーテル(Jelco(登録商標),Johnson & Johnson,Cincinnati,OH)を用いてカニューレを挿入した。血圧および導出される心拍数を、Series 7000 Monitor(Marquette Electronics Milwaukee,WI)を用いてモニタリングした。体温を、直腸プローブを用いてモニタリングし、水ジャケット付加熱用ブランケットを用いて37℃に維持した。動静脈(AV)特注体外回路(ECC)の設置の前に、ウサギ左頸動脈および右外頸静脈を分離し、ベースライン血行動態ならびに動脈血pH、pCO2、pO2、総ヘモグロビンおよびメトヘモグロビンをABL 825血液−ガス分析器およびOSM3 Hemoximeter(Radiometer Copenhagen,DK)を用いて測定した。さらに、血小板数および総白血球(WBC)数のためにベースライン血液試料を採取し、これらをCoulter Counter Z1(Coulter Electronics Hialeah,FL)によって測定した。血漿フィブリノーゲンレベルをDade Behring BCS Coagulation Analyzer(Siemens,Deerfield,IL)を用いて測定し、活性凝固時間(ACT)をHemochron Blood Coagulation System Model 801(International Technidyne Corp.,Edison,NJ)を用いてモニタリングし、そして血小板機能をChrono−Log光学凝集測定器型式490(Havertown,PA)を用いて評価した。
ウサギ全血試料を、ACT用には血液凝固阻止されていない1cc注射器中に、細胞数および凝集測定用には3cc体積を有する3.2%クエン酸ナトリウムバキュテナー(Becton,Dickinson,Franklin Lakes,NJ)中に、血液−ガス分析用には40U/mLのヘパリンナトリウム(APP Pharmaceuticals,LLC,Schaumburg,IL)を含有する1cc注射器中に採取した。ECC血流の開始後に、これらのインビトロ測定のために、4時間の間1時間毎に血液試料を採取した。血小板、単球または血漿フィブリノーゲンのいかなる活性化も回避するために、採取から2時間以内に試料を使用した。
ウサギ血小板凝集を、Chrono−Log光学凝集測定器を用いてBorn比濁法に基づいてアッセイした。簡潔に述べると、クエン酸塩添加血(1:10の血液対3.2%クエン酸ナトリウム溶液)を採取し(6mL)、血小板を多量に含んだ血漿(PRP)を、15分にわたる110×gでの遠心分離により得た。血小板の乏しい血漿(PPP)を、PRPを除去した血液試料の15分にわたる2730×gでの別の遠心分離により得、凝集のためのブランクとして使用した。
長さ18cmの直径2mmのステンレス鋼マンドレル(McMaster Carrから購入)上にポリマー溶液を浸漬被覆することにより、カテーテルを調製した。E2Asコントロールカテーテルについては、ポリマー溶液は、THFに溶解させたE2As(150mg/mL)からなった。各被覆の間に2分間の間隔をおいて浸漬被覆することにより、E2As溶液の35枚の被膜をマンドレル上に適用した。SNAP/E2Asカテーテルについては、2つの異なる溶液、すなわち、上塗り/下塗り溶液および活性溶液を、3層カテーテル(図16参照)を作製するために調製した。上塗り/下塗り溶液は、THFに溶解させたE2As(150mg/mL)からなった。活性溶液は、10wt%のSNAPおよび90wt%のE2AsをTHFに溶解させて全体濃度を150mg/mLとしたものから構成された。3層カテーテルを、E2As溶液の5枚の下塗り、活性溶液の25枚の被膜、およびE2As溶液の5枚の上塗りを浸漬被覆することにより調製した。ヒツジ調査のために使用したカテーテルは、長さ15cmであった。
<ヒツジカテーテル植込み>
全ての動物は、National Society for Medical Researchにより策定された「Principles of Laboratory Animal Care」およびNational Academy of Sciencesにより作成されNIHにより公表された「Guideline for the Care of Use of Laboratory Animals」に準拠したケアを受けた。この調査は、University of Michigan Committee on Use and Care of Animalsによって承認された。
取り出し(explanting)後、カテーテルをPBS中ですすいだ。Nikon L24デジタルカメラを用いて、カテーテル全体の外部および長手方向に切った1cm片の内部の写真を撮影した。血栓の程度を、NIHからのImage J画像化ソフトウェアを用いて定量した。生菌を定量するために、1cm片を長手方向に切り、1mLのPBS緩衝液中で均質化した。最適均質化速度は、異なる均質化速度および時間を比較した別個の実験を用いて見出された。結果として生じたホモジェネートを、滅菌PBS中で段階的に希釈した。各希釈物の三重反復試験アリコート(10μL)を寒天プレート上に置いた。この寒天プレートを24時間にわたり37℃でインキュベートし、続いて、カテーテル表面積当たりのコロニー形成単位(CFU/cm2)を計算した。
本開示の全体にわたって、データは、平均±SEM(平均値の標準誤差)として表される。種々のSNAP/E2Asポリマー群とE2Asコントロールポリマー群との間の比較を、スチューデントt検定を用いた平均値の比較により分析した。全ての試験について、p<0.05の値を統計学的に有意であると見なした。
本開示の実施例は、疎水性ポリウレタン、例えば、シロキサンベースのポリウレタンエラストマー(その一例はElast−Eon(商標)E2Asポリマーである)が、SNAPのためのレザバとして機能する優れたマトリックスであり、結果として生じるフィルムが、NOおよびSNAPの制御放出のために使用され得ることを明らかにした。SNAPは、このポリマーフィルムからゆっくりと拡散し、このフィルム/コーティングからのNO放出は、血液がECCループを通って流れる時に光分解および/または熱分解によって開始され得る。安定性調査は、37℃での4ヵ月にわたる貯蔵の間でさえも、SNAPがE2Asマトリックス内で極めて安定であることを証明する。E2Asポリマーはそれだけで優れた固有の生体適合特性を有しているが、E2Asポリマーマトリックス中にSNAPを組み込むことで、NO/SNAPの制御送達がもたらされてポリマー血液適合性がさらに改善される。SNAP/E2As被覆ECCループは、4時間にわたるECC血流の間、血小板の数および機能を有意に維持したのであるが、凝塊面積もまた、対応するE2As被覆コントロールループに比べて減少した。さらに、SNAP/E2Asカテーテルから放出されるNOは、ヒツジへの7日間にわたる植込みの間、血栓および細菌の付着を有意に低減することができ、その結果、カテーテルの開存性を改善することができた。Elast−Eon(商標)E2Asポリマーフィルム/コーティング内にSNAPを組み込むことは、NO/SNAPを局所的に送達するための簡単な方法を提供し、多様な血液接触性医用デバイスの血液適合性を改善する可能性を有している。
Claims (12)
- ポリマーマトリックスと、5〜10wt%の範囲で前記ポリマーマトリックス中に導入されるS-ニトロソ-N−アセチルペニシラミン(SNAP)とからなるポリマーフィルムであって、
前記SNAPは、酸化窒素(NO)を放出することが可能であり、前記ポリマーマトリックスは、シリコーンゴムポリマーマトリックス、熱可塑性シリコーン−ポリカーボネート−ウレタン及びシロキサンベースのポリウレタンエラストマーからなる群より選択され、
前記ポリマーフィルムは、37℃の乾燥条件下において安定性を示し、かつ前記SNAPから水分またはS-ニトロソチオール結合を光分解することが可能な光に曝露されると、少なくとも2日間にわたって制御可能なNO放出速度を示す、ポリマーフィルム。 - 前記ポリマーマトリックスが、1.2wt%±0.3の水吸収を有する前記シリコーン
ゴムポリマーマトリックス、1.5wt%±0.3の水吸収を有する前記熱可塑性シリコーン−ポリカーボネート−ウレタン及び1.2wt%±0.1の水吸収を有する前記シロキサンベースのポリウレタンエラストマーからなる群より選択される疎水性ポリマーを含む、請求項1に記載のポリマーフィルム。 - 前記ポリマーマトリックスが、前記シロキサンベースのポリウレタンエラストマーであり、
前記SNAPが10wt%の量で存在し、かつ
前記NOが少なくとも20日間にわたって放出される、請求項1に記載のポリマーフィルム。 - 前記ポリマーマトリックスが前記シロキサンベースのポリウレタンエラストマーであり、前記SNAPが5wt%の量で存在し、かつ前記NOが少なくとも7日間にわたって放出される、請求項1に記載のポリマーフィルム。
- 前記SNAPが、前記ポリマーマトリックス内で分散しており、かつ前記SNAPが10wt%の量で存在する、請求項1に記載のポリマーフィルム。
- 下部ポリマー層と、
前記下部ポリマー層の上に配置された上部ポリマー層と、
前記下部ポリマー層と前記上部ポリマー層との間にある少なくとも1つの活性中間層と
を含むデバイスであって、前記少なくとも1つの活性中間層が、請求項3に記載のポリマーフィルムを含む、デバイス。 - 前記下部ポリマー層が、シロキサンベースのポリウレタンエラストマー、ポリ(塩化ビニル)、架橋ポリウレタン、架橋シリコーンゴム、およびポリテトラフルオロエチレンからなる群より選択され、かつ
前記上部ポリマー層が、シロキサンベースのポリウレタンエラストマー、ポリ(塩化ビニル)、架橋ポリウレタン、架橋シリコーンゴム、およびポリテトラフルオロエチレンからなる群より選択される、請求項6に記載のデバイス。 - 溶媒の蒸発によって、S-ニトロソ-N−アセチルペニシラミン(SNAP)を含む熱可塑性シリコーン−ポリカーボネート−ウレタン又はシロキサンベースのポリウレタンエラストマーのポリマーフィルムを形成する工程であって、
前記熱可塑性シリコーン−ポリカーボネート−ウレタン又は前記シロキサンベースのポリウレタンエラストマーを溶媒中に溶解して溶液を形成すること、 前記溶液中に前記SNAPを添加すること、
前記溶液を規定の時間攪拌すること、
前記溶液を乾燥させること、
を含むポリマーフィルムを形成する工程を備え、
前記SNAPを含むポリマーフィルムは少なくとも3日間にわたって酸化窒素(NO)を放出することが可能である、NO放出性ポリマー組成物を製造するための方法。 - 前記ポリマーフィルムが、SNAPを含む前記シロキサンベースのポリウレタンエラストマーであり、NOを少なくとも7日間にわたって放出することができる、請求項8に記載の方法。
- 前記溶液を下部ポリマー層上に流延することと、
前記流延された溶液を乾燥させて前記下部ポリマー層上に前記NO放出性ポリマー組成物の層を形成することと
をさらに含む、請求項9に記載の方法。 - 他のポリマーを前記NO放出性ポリマー組成物の前記層の上に被覆して上部ポリマー層を形成することをさらに含み、前記他のポリマーはSNAPを含まない、請求項10に記載の方法。
- 前記デバイスは体外チューブであり、
前記下部ポリマー層はポリ(塩化ビニル)からなり、
