Description
肺癌は、予後診断の不良と低い生存率によって特徴付けられる、工業国における死亡の主要な原因の1つである(Jettら、1983年; Pintoら、2011年)。新生物増殖の基礎となる細胞および分子メカニズムは、まだ十分に定義されていないが、肺癌の危険因子の1つは、発癌物質への暴露(吸入)である(Valavanidisら、2008年)。
著者らは、驚くべきことに、カスパーゼ、特にマウスカスパーゼ−11[NCBI受託番号CAA73531.1](配列番号4)およびヒト類似体カスパーゼ−4[NCBI受託番号NP_001216.1](配列番号1)、並びにマウスカスパーゼ−1[NCBI受託番号マウス:NP_033937.2 (NM_009807.2)](配列番号3)およびヒトカスパーゼ−1[CAA46153.1](配列番号2)が、肺における新生物増殖に関与していることを発見した。
したがって、著者らは、肺癌発生に関与する新規「経路」を同定しただけではなく、肺癌などの高死亡率を有する疾患の将来の治療戦略の開発のための新しい薬理学的標的を同定した。さらに、腫瘍組織で非常に高いレベルであることがすでに知られている、IL−1αおよびIL−1βなどの炎症性サイトカインの存在に関連する、ヒトにおけるカスパーゼ−4の活性化は、肺癌のための新しい診断の、および可能な予後診断のツールを示す。

IL−1βは、好ましくは、配列番号10または9により特徴付けられる。
IL−18は、好ましくは、配列番号14または13により特徴付けられる。
HMGB1は、好ましくは、配列番号12または11により特徴付けられる。
本発明による方法において、腫瘍は、好ましくは肺癌、より好ましくは、肺癌(lung carcinoma)である。
対象から単離されるサンプルは、好ましくは、生体液、細胞サンプルおよび/または組織サンプルである。
−上記で定義されるような、少なくとも1つのバイオマーカーの量を検出するおよび/または測定するための、および/またはそのバイオマーカーの量の変化を測定するための手段、および任意に、

本発明で記載されるバイオマーカーは、癌を予防するために使用されることができる。本発明を以下の図面を参照に、非限定的な実施例で説明する。
肺癌(Lung Carcinoma)のマウスモデル
C57Bl/6マウス(Harlan Laboratories、イタリア)および129Svマウス、およびカスパーゼ−1および11ノックアウトマウス(Charles River Laboratories、イタリア)(6〜8週間のメス)を、アルキル化および変異原性活性を有する、発癌物質、N−ニトロソ−N−メチル尿素(NMU)の気管内(i.t.)注入に供した(Damianiら、2008年)。NMUは、次の投与スケジュールおよび投与量に応じて、7日ごとに3回投与された:0日目、0.50μg/マウス:8日目、10μg/マウスおよび15日目、10μg/マウス(図1)。いくつかの実験において、抗IL−1α抗体(Ab)(2μg/ラット腹腔;eBioscience、米国)、またはカスパーゼ−1阻害剤(Ac−Y−VAD−cmk:10μg/マウス、腹腔内、Sigma Aldrich、米国)、または抗カスパーゼ−11抗体(10μg/マウス腹腔、Santa Cruz、米国)をNMUで処理されたC57Bl/6マウスに投与した。動物は、図1に示されるスキームにしたがって、異なる時点(最初のNMU投与から3−7−30日)で屠殺した。腫瘍病変は、腫瘍病変面積/総肺面積比として表された。
ヒトサンプルは、非小細胞肺癌タイプの上皮起源腺癌のステージIII癌の患者において、胸部手術および肺切除の後に得られた。Hで示された健康部分は、癌領域から巨視的に非常に遠い肺部分から得られた。ヒト組織は、イタリア、サレルノ(Salerno)、the Azienda Ospedaliera Universitaria San Giovanni di Dio e Ruggi d’Aragonaの胸部外科によって、提供された。
マウス肺およびヒトサンプルをコラゲナーゼ(1U/ml)およびDNAse I(20μg/ml)からなる消化溶液で消化した。タンパク質決定に従って、サンプルを12%ポリアクリルアミドゲル上に装填し(50μg/サンプル)、次に、ニトロセルロース膜上に移した。抗カスパーゼ−4(Santa Cruz、米国)、抗カスパーゼ−1(Santa Cruz、米国)、抗カスパーゼ−11(Santa Cruz、米国)、抗IL−1α(R&D Systems、イギリス)抗体を使用した。装填コントロールをGAPDH認識により行った。
ヒトおよびマウス肺ホモジネートを、キット製造業者によって提供される指示に従って、IL−1αおよびIL−1βの存在について試験した(eBioscience、米国)(インフォームドコンセントを得た)。
NMUで処置したマウスの左葉をOCT培地(TedPella Inc、ミラノ、イタリア)で固定し、次に7〜12μmの低温切開片に切り、ヘマトキシン&エオシン(H&E)で染色して組織の形態学的特徴を強調し、肺癌病変において、K−Rasの存在を確認するための免疫蛍光染色(Cell Signalling、イギリス)に供される低温切開片、および/または二次HRP抗体と腫瘍マーカーであるKi−67(Invitrogen、イタリア)からなる免疫複合体を検出するために、ジアミノベンジジン法(DAB)による免疫組織化学分析に供される低温切開片と、関連づけた。Ki−67の対照アイソタイプ(抗ラットIgG)を陰性対照として使用した。
結果は、平均±SEMとして表される。種々の群間の差は、適切な一元配置分散分析および/またはスチューデントのt検定を使用して統計的に分析された。0.05より低いp値は、統計的に有意であるとみなされた。
1.カスパーゼ−11は、マウスの肺癌増殖に関与する。
C57B1/6マウスにおいて、NMUでの処置は、Ki−67(図2B)、およびK−Ras(図2C)などの腫瘍増殖マーカーについて陽性であった肺の低温切開片(図2A)によって示されるように、腫瘍病変を生じた。NMUで処理されたマウスにおいて、腫瘍面積と総面積の間の比として計算される腫瘍質量増殖は、指数関数型である(図2D)。
本研究を橋渡しするために、カスパーゼ−11のヒトアナログ、すなわち、カスパーゼ−4の役割が分析された。カスパーゼ−4は、健康組織と比較して、7人の患者から分析された全ての腫瘍細胞で活性であった(p20 kDa)(図9A)。また、同じ組織において、カスパーゼ−1は、健康部分より腫瘍部分においてより活性化される(p20 kDa)ことがわかった(図9B)。したがって、ヒトでのこれらの酵素の活性の存在は、マウスで観察されたものと同様である。また、IL−1α(図10AおよびB、**p<0.005)およびIL−1β(図10B、*p<0.05)の存在は、正常組織よりも腫瘍組織において高かった。マウスで観察されたものと同様に、カスパーゼ4は、ヒト健康肺および腫瘍病変を有する肺のホモジネート組織で実施された免疫沈降実験、その後のウエスタンブロット分析によって証明されるように、AIM2に関連した(図11)。

Claims (9)
- 肺癌(Lung Cancer)の診断および/または予後診断のための、および/または肺癌(Lung Cancer)の発達および/または進行をモニタリングするためのインビトロまたはエクスビボの方法において、
対象から単離されたサンプルにおけるヒトカスパーゼ−4−タンパク質の活性型(配列番号1)を検出するおよび/または定量するステップを含む、方法。 - a)対象から単離されたサンプルにおけるヒトカスパーゼ−4−タンパク質の活性型(配列番号1)を検出するおよび/または定量するステップと、
b)それを所定の対照と比較するステップと
を含む、請求項1に記載の方法。 - 少なくとも1つの追加腫瘍マーカーの検出および/または定量、および適切な対照サンプルとの比較をさらに含み、
前記追加腫瘍マーカーは、IL−1α、IL−1β、IL−18またはHMGB1である、請求項1又は2に記載の方法。 - 対象から単離された前記サンプルは、生体液、細胞サンプルおよび/または組織サンプルである、請求項1〜3のいずれかに記載の方法。
- 前記肺癌(Lung Cancer)が、肺癌(Lung Carcinoma)である、請求項1から4の何れか1項に記載の方法。
- 肺癌(Lung Cancer)の診断および/または予後診断のための、および/または肺癌(Lung Cancer)の発達および/または進行のモニタリングのためのキットにおいて、
ヒトカスパーゼ−4−タンパク質の活性型(配列番号1)の量を検出するおよび/または測定するための試薬と、
対照試料と
を含む、キット。 - 肺癌(Lung Cancer)の予防および/または治療における使用のための、ヒトカスパーゼ−4−タンパク質の活性型(配列番号1)に対する特異的阻害剤であって、抗ヒトカスパーゼ−4抗体、合成ペプチド、アミノ酸および/またはヌクレオチド配列、ワクチン、siRNA、または低分子量薬物である、阻害剤。
- ヒトカスパーゼ−4−タンパク質の活性型(配列番号1)の肺癌(Lung Cancer)の診断のためのインビトロバイオマーカーとしての使用。
- 前記肺癌(Lung Cancer)が、肺癌(Lung Carcinoma)である、請求項8に記載の使用。
