Description

The present invention relates to a cerebral blood flow improving agent.

In Japan, as described in the phrase "100 million people who are sick and half sick," many people have mild physical problems such as insomnia, headache, stiff shoulders, low back pain, stomach pain, nausea, tiredness, lack of motivation, and irritation. I have a problem. Furthermore, with the aging society, the number of sickly sick people is expected to increase in the future, and is regarded as a major social problem leading to an increase in medical expenses. Many of these physical disorders are related to brain function. Among them, the cerebral cortex is a site that has functions that control memory, judgment, sensation, movement, language, etc., which are important in human life, and has a great relationship with everyday life, so the activity of the cerebral cortex is It is considered important for health.

For example, Patent Document 1 proposes a cerebral blood flow improving agent containing cells of Lactobacillus gasseri CP2305 strain, and the vagus nerve of the stomach and intestine is ingested by ingesting the cells of Lactobacillus gasseri CP2305 strain. It has been reported that the activity of the afferent branch is increased, the cerebral blood flow (increased blood flow in the cerebral cortex, decreased blood flow in the basal ganglia, decreased blood flow in the occipital lobe 8 region) is improved, and brain function is improved. ing. These changes in cerebral blood flow are thought to be effective in avoiding epileptic seizure risk, reducing stroke risk, avoiding cerebral aneurysm risk, calming behavior, and suppressing frustrating behavior. In addition, it is thought to partially reduce cerebral blood flow and contribute to insomnia and sleep quality improvement. In fact, in human intake tests, sleep quality, falling asleep, and alleviating sleep disorders have been shown to improve sleep Is shown.

On the other hand, the technique of "healing" using scents and music has been generalized. Above all, aromatherapy using scent is a mind and body healing therapy that is taken in daily by many lovers. In addition, some research results have been reported on the effects of aromatherapy and other fragrances on diseases, but most are reports on essential oils containing multiple fragrance components, and reports on single fragrance components Almost no.

The present invention relates to a cerebral blood flow improving agent containing camphor.
The cerebral cortex is a site that has functions that control memory, judgment, sensation, movement, language, etc., which are important in human life, and has a great relationship with daily life, so the activity of this site is important for physical and mental health , Considered important.
The cerebral cortex is roughly divided into frontal lobe, parietal lobe, temporal lobe, and occipital lobe.

(Penfield W and Evans J, The frontal lobe in man: A clinical study of maximum removals., Brain 1935 58:115-133)。
The motor cortex, premotor cortex, prefrontal cortex, frontal eye cortex, motor language cortex, etc. are known as functional localization of the frontal lobe.
The prefrontal cortex of the frontal lobe has both cognitive / executive functions and emotion / motivation functions. The prefrontal cortex plays a role as a whole `` understand cognitive and motivating situations, make appropriate judgments, and organize behaviors adaptively in situations that can not be dealt with with a typical response style '' . The prefrontal cortex of the frontal lobe supports working memory, response suppression, behavior switching, cognitive and executive functions such as planning and inference, emotional and motivational functions that play an important role in the decision-making process, and social behavior. It is responsible for functions such as solution and selection based on rewards.
The prefrontal cortex has been shown to balance intelligence and impulses and to be involved in future planning (Harlow JM, Boston Medical and surgical Journal 1848 39: 389-393).
If the prefrontal cortex is damaged, it becomes impossible to assemble the sequenced actions, that is, to make arrangements (Penfield W and Evans J, The frontal lobe in man: A clinical study of maximum removals. , Brain 1935 58: 115-133).

(Boller F, Grafman J(Eds.) Handbook of Neuropsychology, 2ndEdition: The Frontal Lobes Elsevier 2002)。
Patients with prefrontal cortex injuries have difficulty in planning and executing plans, including "planning in order,""finishingplans,""distinguishing important and trivial items," or cause a failure to "or so as not to bring the goals and unrelated matters" (Boller F, Grafman J (Eds ) Handbook of Neuropsychology, 2 nd Edition:. the Frontal Lobes Elsevier 2002).
The frontal pole in the prefrontal cortex of the frontal lobe (Broadman's brain map area 10) is involved in thinking about the future (Jiro O, et. Al., NeuroImage 2003 19: 1369-1380), and is responsible for making decisions made in the past. Involved in the assessment (Satoshi T, et. Al., Nature Neuroscience 2010 13: 120-126).

(Kringelbach ML and Rolls ET, Progress in Neurobiology, 2004 72:341-372)。
Human Brain Mapping Aug 2007)。
The orbital frontal cortex in the prefrontal cortex of the frontal lobe (Broadman's brain map area 11) is involved in the expression of emotional values, decision making and expectations, and plays a role in mediating subjective pleasure experiences ( Kringelbach ML, Nature Reviews Neuroscience, 2005 6: 691-702). It also controls action plans related to sensitivity to reward and punishment (Kringelbach ML and Rolls ET, Progress in Neurobiology, 2004 72: 341-372).
In the lower frontal gyrus of the frontal lobe, there is a Broca area (Broadman brain areas 44 and 45), also called the motor language center, which is involved in language processing, speech language, and the production and understanding of sign language (Horwits B, Amunts K, Bhattacharyya R, Patkin D, Jeffries K, Zilles K, Braun AR, Neuropsychologia 2003 41: 1868-76).
In the lower frontal gyrus of the frontal lobe, there is a triangular part (Broadman's brain area 45), which is strongly active when reading exceptional words where the relationship between sound and spelling is irregular (Andrea M, Jennifer TC, Steven L, Karl JF, Matthew ALR, Karalyn P, James LM, and Cathy JP, Journal of Cognitive Neuroscience 2005 17: 11: 1753-1765). It also has the role of deciphering the meaning of words (Cortical dynamics of word recognition. Human Brain Mapping Aug 2007).

Opin.
In the parietal lobe, the sensory area, the taste area, the sensory association area, the parietal association area, and the like are known as functional localization.
The parietal association area of the inferior parietal lobule of the parietal lobe (Broadman's brain area 39, 40) is a function such as expression and regulation of movement associated with the premotor area (Leonardo F, Giuseppe L, Curr. 2005 15: 626-31), which is responsible for controlling attention associated with prefrontal cortex function (JB Hopfinger, MH Buonocore, GR Mangun, Nat. Neurosci., 2000 3: 284-91).

In the temporal lobe, the auditory cortex, auditory cortex cortex, auditory speech cortex, and olfactory cortex are known as functional localization.
The auditory cortex of the temporal lobe of the temporal lobe (Broadman's brain map 41) and the auditory association area (Broadman's brain map 42) are called primary auditory cortices and function to discriminate pitch ( Bendor D, Wang X, Nature 2005 436: 1161-5). This area can identify basic parts of music, such as pitch and volume.

Focusing on the prefrontal cortex, which is said to be the control tower of the brain, the odor components that activate the frontal lobe, parietal lobe, and temporal lobe (hereinafter also referred to as the "region") have been studied intensively. Found that most of this region was activated. The activity was measured using oxyhemoglobin concentration change as an index.

Figure 0006660783
Camphor is one of the monoterpene ketones, also called camphor. Formula 1 shows the structural formula of camphor. Compound names are 1,7,7-trimethyl bicyclo [2.2.1] - heptan-2-one, the composition formula is C 10 H 16 O. Camphor has a melting point of 180 ° C. and a boiling point of 208 ° C., is a waxy white translucent solid at room temperature, and has sublimability.
Figure 0006660783

Camphor was originally produced by steam distillation of camphor wood pieces, but is now mostly produced by chemical synthesis using pine or other essential oil as a starting material.
Camphor is used as an external preparation for muscle pain, bruises, bruises, sprains, frostbite (first degree), frostbite, and pruritus cutaneous for the purpose of improving blood circulation, inflammation, analgesia, and itching. It is also used as an insect repellent for clothes.

The cerebral blood flow improving agent of the present invention may be any as long as it contains camphor. The camphor may be used alone as it is, or an essential oil containing camphor may be used. Further, camphor can be diluted with alcohol or oil, or blended in a formulation such as an ointment. A solution in which camphor is dissolved in an organic solvent or the like can be used by impregnating it with silica gel, or can be kneaded into a candle and used as an aroma candle.

The method of using the cerebral blood flow improving agent of the present invention includes, for example, wrapping camphor crystals in a paper bag or cloth bag and inhaling volatilized camphor therefrom. The camphor solution impregnated in silica gel may be similarly wrapped in a paper bag or cloth bag to volatilize the camphor. Further, by externally using an ointment containing camphor, camphor that volatilizes from the application part may be inhaled. Oral administration of a beverage containing camphor may inhale the camphor evaporating from the beverage. The camphor may be inhaled by spraying a camphor solution to inhale the volatilizing camphor, or by burning an aroma candle containing camphor.

By inhaling camphor contained in the cerebral blood flow improving agent of the present invention, blood flow in the brain, particularly in the frontal lobe, parietal lobe, and temporal lobe of the cerebral cortex increases. By increasing the blood flow, activation of the brain function controlled by the frontal lobe, parietal lobe, and temporal lobe can be expected, and improvement of the function localized in this region can be expected.

[result]
Camphors of "whole" and "above right head", "below right head", "above frontal", "below frontal head", "left above head", "below left head" of cerebral cortex, Comparison of changes in oxyhemoglobin concentration between α-pinene and control is shown in FIGS. The vertical axis of the figure is the unit of the NIRS measurement value, which is the oxyhemoglobin concentration length represented by the product of the oxyhemoglobin concentration and the optical path length. Further, as a result of the statistical analysis, a point having a statistically significant difference (p <0.05) is indicated by *.

By camphor, it was confirmed that the cerebral blood flow increased in all six areas obtained by dividing this region. On the other hand, α-pinene hardly increased cerebral blood flow in all of the six areas obtained by dividing this region, and was inferior to dipropylene glycol as a control.
Although camphor and α-pinene are both monoterpenes and have similar molecular skeletal structures, it was confirmed that camphor significantly increased cerebral blood flow.
