Description
から、有利な薬学的性質、特に、保存中の物理的な安定性及び考えられる投与経路、特に小さい液滴の使用を要する静脈内投与の点で有利な薬学的性質をもたらす。しかし、これらの系はそのエマルションを構成する油(大豆油、オリーブ油)に可溶である非常に親油性の高い有効成分しか組み入れることができず、それらの潜在用途を制限する。

殿及び再水和を要するので工業規模で適用するには困難が伴う。実際に、これらの粒子は主に、水相が予め形成される脂質ナノ粒子を含有することを除いてはリポソームに用いられるものと非常に類似したリン脂質膜水和技法によって得られる。最終的な粒子は、リン脂質二重層中の油滴の「統計的な閉じ込め(statistical confinement)」に由来する。結果として、該方法は先験的に(a priori)、脂質とリン脂質部分を結びつけること無く物体(エマルソーム、リポソーム、ナノエマルション、又は固体ナノ粒子)のさまざまな集団を生成する。これは先験的に(a priori)、ある特定の精製操作(例えば、遠心分離)又は保存中に系の安定性の問題を引き起こしうる。
てこれらの賦形剤を混合及び処理することにより、典型的なナノエマルション(NE)のイメージと明らかに異なる本発明のセリソームが作られる。
くとも1つ含有する親水性非イオン性界面活性剤混合物の使用に基づいているようである。疎水性セグメントは、脂肪族アルコール、脂肪酸、グリセリド、コレステロール、又はリン脂質膜に親和性をもつ他の基から成ることができる。親水部は、ポリオキシエチレン、ポリプロピレン、多糖類型のポリマー、又は疎水性セグメントと併せて、HLB値が11を超える界面活性剤を生成できる任意の型のポリマーから成ることになる。HLB値はグリフィンによって記載された尺度に従って本明細書で定義される。例えば、これらの界面活性剤は、ポリオキシルグリセリド若しくはマクロゴールグリセリド、ポリオキシエチレン脂肪酸エステル、又はポリオキシエチレンアルキルエーテルのファミリーに属することがあり、好ましくはゲルシア(登録商標)50/13又はポリオキシエチレン(40)ステアレートであることになる。ポリオキシエチレン脂肪酸エステルは、親水性ポリ(オキシエチレングリコール)鎖でエステル化された脂肪酸の混合物であり、脂肪酸−PEG型(モノエステル)の2つのブロックが並んだ誘導体及び脂肪酸−PEG−脂肪酸型(ジエステル)の3つのブロックが並んだ誘導体のさまざまな割合での混合物を含有する。また、マクロゴールグリセリドはグリセリドを含有する。特に、ゲルシア(登録商標)50/13は、グリセリドと1500g/molの親水性ポリ(オキシエチレングリコール)鎖でエステル化された脂肪酸の混合物であり、脂肪酸−PEG−脂肪酸型(図4A)の3つのブロックが並んだ誘導体を約40%含む。提案された系では、ジエステル部分は、間違いなく二重層と脂質部の間の固定位置(anchor point)である(図4B)。実際に、リン脂質の脂質粒子型製剤への組み込みが、一般に2つのタイプの物体、ナノ粒子及びリポソームを含有する不均一な製剤を生じることは当該技術分野において公知である。グリフィンHLB値が11を超える「疎水性−親水性−疎水性」型の非イオン性界面活性剤が非存在の場合に本発明の実験プロトコールを適用することによって同じ結果が得られた(データ示さず)。提案された系では、これらの界面活性剤部分は、間違いなく二重層と脂質部の間の固定位置である(図4B)。
本発明はまた、以下の工程を含む本発明のマルチコンパートメント脂質ナノ粒子(セリソーム)を調製する方法を目的として有する:
− 少なくとも1つの脂質、1つのリン脂質、1つの親水性非イオン性界面活性剤を含み、少なくとも1つの対象となる分子を含んでいてもよい、油/水エマルションを調製する工程;
− 高圧下で前記エマルションをホモジナイゼーションする工程。

好ましくは、本発明の方法の油/水エマルションは以下を含む:
− 5〜30%の脂質、好ましくは、ラブラフィル(登録商標)M2125CS又はM1944CS;
− 0.5〜5%のリン脂質、好ましくはホスフォリポン90G(登録商標);
− 0.5〜5%のマクロゴールグリセリド又はポリオキシエチレン脂肪酸エステル、好ましくはゲルシア(登録商標)50/13又はポリオキシエチレン(40)ステアレート;
− 水、適量 100g。
調製
マルチコンパートメント脂質ナノ粒子の調製を図5に模式的に示す。
各バッチについて、以下の3つの工程を含むプロトコールに従って50gのナノ分散体を調製した。
上記調製方法によって得られたマルチコンパートメント脂質ナノ粒子は例えば以下を含む。
脂質(ラブラフィル(登録商標)) 5〜20%
リン脂質(ホスフォリポン90G) 1%
マクロゴールグリセリド(ゲルシア(登録商標)50/13) 2%
水(適量) 100g
粒子のサイズ及び多分散性について、水に懸濁したマルチコンパートメント脂質ナノ粒子の室温での保存を数箇月にわたってモニターした。ラブラフィル(登録商標)M2125CSについて、このモリタリングの例を示した。平均直径(n=3)の標準偏差は概し
て記号で隠され、方法の良好な再現性を裏付けた。200nm未満のナノ粒子直径は静脈内投与に適する。0.2未満の多分散性は平均値前後の狭いサイズ分布を表わす。
有効成分用担体としてのマルチコンパートメント脂質ナノ粒子の調製
上記プロトコールによって得られたマルチコンパートメント脂質ナノ粒子を、医薬的有効成分を封入するその能力について試験した。
水性媒体中で加水分解され、本発明のマルチコンパートメント脂質ナノ粒子に封入されたクェルセチンの安定性を調べた。
さまざまな賦形剤から上記プロトコールに従って、他のセリソームを得た。
調製
マルチコンパートメント脂質ナノ粒子の調製を図5に模式的に示す。
以下の3つの工程を含むプロトコールに従って10グラムのナノ分散体を調製した。
上記調製方法によって得られたマルチコンパートメント脂質ナノ粒子は以下を含む(質量%):
ラブラフィル(登録商標)M2125CS 20%
ホスフォリポン90G 1%
ゲルシア(登録商標)50/13 2%
水(適量) 10g
この調製法の後、ナノ粒子は流体力学的直径が228±2nmである。さらに、クライオ−TEM観察により、ナノ粒子がコンパートメント化され、図3に示されるものと同様の形態であることを確認した。
実施例5:医薬及び/又は化粧品目的の分子のための担体としての10gバッチのマルチコンパートメント脂質ナノ粒子(セリソーム)
この調製法の後、カフェインを含有するナノ粒子は流体力学的直径が223±3nmであり、25℃で少なくとも12日間安定した状態を保ち、イルガサンを含有するナノ粒子は流体力学的直径が224±4nmであり、25℃で少なくとも12日間安定した状態を保ち、クロロキシレノールを含有するナノ粒子は流体力学的直径が245±2nmであり、25℃で少なくとも1箇月間安定した状態を保った。
実施例4に記載のプロトコールに従って、マルチコンパートメント脂質ナノ粒子を調製した。
標)974NF中に0.2%及び0.4%に濃縮し、それぞれ脂質賦形剤(質量による)を20.7%及び18.4%含有し、最終的なゲルはナノ粒子を含まないカーボポール(登録商標)974NFゲルのずり減粘性を保持する。
調製
マルチコンパートメント脂質ナノ粒子の調製を図5に模式的に示す。
上記調製方法によって得られたマルチコンパートメント脂質ナノ粒子は以下を含む(質量%):
ラブラフィル(登録商標)M2125CS 20%
ホスフォリポン90G 1%
ゲルシア(登録商標)50/13 2%
水(適量) 1kg
この調製法の後、クライオ−TEM観察により、ナノ粒子がコンパートメント化され、図3に示されるものと同様の形態であることを確認した。

Claims (25)
- 以下を含むことを特徴とする、平均直径が10〜500nmであるマルチコンパートメント脂質ナノ粒子:
− リン脂質二重層によって区切られた第2の親水性コンパートメントによって部分的に囲まれた、脂質を含有する第1の親油性コンパートメントであり、前記第2のコンパートメントに部分的に固定された前記第1のコンパートメント;
− (a)リン脂質、及び、(b)グリフィンHLB値が11を超える「疎水性−親水性−疎水性」型の3連配列界面活性剤を含む非イオン性界面活性剤混合物。 - 多分散指数が5〜15%である請求項1に記載のナノ粒子。
- 前記平均直径が100〜250nmである請求項1又は2に記載のナノ粒子。
- 前記親油性コンパートメントが、25℃で液体状態又は半固体状態である脂質から成る請求項1〜3のいずれか一項に記載のナノ粒子。
- 前記親油性コンパートメントが、グリセリドが混合されていてもよい脂肪酸エステルを含む請求項1〜4のいずれか一項に記載のナノ粒子。
- 前記脂肪酸エステルが、親水部のモル質量が100〜700g/molの範囲であるポリエチレングリコールモノエステル及びポリエチレングリコールジエステルの混合物である請求項5に記載のナノ粒子。
- 前記グリセリドが、25℃で液体又は固体であるグリセロールモノエステル、グリセロールジエステル、及びグリセロールトリエステルの混合物からなる群から選択される請求項5又は6に記載のナノ粒子。
- 前記リン脂質二重層が、少なくとも1つのリン脂質及び1つの親水性非イオン性界面活性剤を含む請求項1〜7のいずれか一項に記載のナノ粒子。
- 前記リン脂質が、双性イオン性のリン脂質の混合物からなる群から選択される請求項8に記載のナノ粒子。
- 前記親水性非イオン性界面活性剤が、ポリオキシルグリセリド、ポリオキシエチレン脂肪酸エステル、及びポリオキシエチレンアルキルエーテルからなる群から選択される請求項8又は9に記載のナノ粒子。
- 少なくとも1つの対象となる分子をさらに含む請求項1〜10のいずれか一項に記載のナノ粒子。
- 前記対象となる分子が、核酸、タンパク質、多糖類、及び40〜2000Daの分子から選択される請求項11に記載のナノ粒子。
- 薬剤的に許容可能な賦形剤中に請求項1〜12のいずれか一項に記載のナノ粒子の懸濁液を含む医薬組成物。
- 以下の工程を含む、請求項1〜12のいずれか一項に記載のマルチコンパートメント脂質ナノ粒子を調製する方法:
− 少なくとも1つの脂質、リン脂質、及び、1つの親水性非イオン性界面活性剤を含み、少なくとも1つの対象となる分子を含んでいてもよく、前記親水性非イオン性界面活性剤は、グリフィンHLB値が11を超える「疎水性−親水性−疎水性」型の3連配列界面活性剤を含む非イオン性界面活性剤混合物を含む、油/水エマルションを調製する工程;
− 200〜600barの圧力下で前記エマルションをホモジナイゼーションする工程。 - 前記エマルションをホモジナイゼーションする前に30〜80℃の温度に加熱する工程をさらに含む請求項14に記載の方法。
- 前記脂質が、親水部のモル質量が100〜700g/molの範囲であるポリエチレングリコールモノエステル及びポリエチレングリコールジエステルの混合物である請求項14又は15に記載の方法。
- 前記リン脂質が双性イオン性のリン脂質の混合物からなる群から選択される請求項14〜16のいずれか一項に記載の方法。
- 前記親水性非イオン性界面活性剤が、ポリオキシルグリセリド、ポリオキシエチレン脂肪酸エステル、及びポリオキシエチレンアルキルエーテルからなる群から選択される界面活性剤を含む請求項14〜17のいずれか一項に記載の方法。
- 前記油/水エマルションが
− 5〜30質量%の脂質;
− 0.5〜5質量%のリン脂質;
− 0.5〜5質量%のポリオキシルグリセリド、ポリオキシエチレン脂肪酸エステル又はポリオキシエチレンアルキルエーテルを含む請求項14〜18のいずれか一項に記載の方法。 - 前記脂質が、ラブラフィル(登録商標)M2125CS又はM1944CSである請求項19に記載の方法。
- 前記リン脂質が、ホスフォリポン90G(登録商標)である請求項19または20に記載の方法。
- 前記ポリオキシルグリセリド又は前記ポリオキシエチレン脂肪酸エステルが、ゲルシア(登録商標)50/13又はポリオキシエチレン(40)ステアレートである請求項18〜21のいずれか一項に記載の方法。
- 請求項1〜10のいずれか一項に記載のナノ粒子を含む、対象となる分子を投与するための担体。
- 請求項11又は12に記載のナノ粒子を含む医薬品。
- 前記医薬品は、注射、経口、経皮又は経鼻で投与される医薬品である請求項24に記載の医薬品。
