Description

(c)配列番号15に規定のアミノ酸配列を有する、DENV1のNS5タンパク質の断片
を含むか又はそれらからなり、(a)、(b)及び(c)の断片がこの順番で直接的又は間接的に融合している、600未満のアミノ酸残基を有するキメラポリエピトープ:
又はそのポリエピトープ変異体であって、該キメラポリエピトープは、(i)DENV NS断片のアセンブリを融合ポリペプチドの形態で含むか又はそれからなり、ここでそれら断片の配列が、DENV2、DENV3若しくはDENV4血清型であるウイルスのNSタンパク質の相応するNS3、NS4b及びNS5配列と、(a)、(b)及び(c)に記載のNS3 DENV1、NS4b DENV1、NS5 DENV1断片をアラインすることにより得られるか、又は(ii)融合断片(a)、(b)及び(c)からなる融合ポリペプチド(これからアミノ酸残基の変異により該キメラポリエピトープが誘導される)の配列とその全長にわたり、75%超の同一性、特に80%超又は85%超又は90%超の同一性を有するアミノ酸配列を有するキメラポリエピトープからなる、キメラポリエピトープに関する。

プラスミドpTM-MVSchwにクローンした、MVシュワルツ株のcDNAを使用して産生されるが、このプラスミドは2002年6月12日に番号1-2889でCNCMのInstitut Pasteur(パリ、フランス)により寄託され、その配列はCombredet[Combredet、C.ら、2003、J Virol、77(21): 11546〜11554頁]により記載され、且つWO2004/000876に開示もされている。プラスミドpTM-MVSchwは、Bluescriptプラスミドから得られたが、T7 RNAポリメラーゼのプロモーターの制御下に置かれた、シュワルツ株の全長MV (+) RNA鎖をコードするポリヌクレオチドを含むとともに、18967個のヌクレオチドを有する。他のMV株からのcDNAは、弱毒化生MVのウイルス粒子から精製した核酸から開始して同様に得ることができる。したがって、本発明の組換えMVゲノムベクターは、配列番号139により説明されているpTM-MVDVax6、配列番号140により説明されているpTM-MVDVax7、配列番号141により説明されているpTM-MVDVax8、配列番号142により説明されているpTM-MVDVax9、配列番号143により説明されているpTM-MVDVax10又は配列番号149により説明されているpTM-MVDVax11である。
a)T7 RNAポリメラーゼ並びにMVのN、P及びLタンパク質を発現するヘルパー細胞株中に、本発明に記載の組換えMVゲノムベクターをトランスフェクトする工程、
b)前記ヘルパー細胞株を、本発明に記載のキメラポリエピトープと、任意選択で、DENV1のectoMに融合した、4種の血清型のEDIIIポリペプチドの融合体から構成される4価DENV抗原、特に配列番号145の配列を有するDENV抗原を含むポリペプチドとを発現する組換えMV粒子の複製及び産生を維持する能力がある細胞上に移す工程、及び
c)工程b)で産生された組換えMV粒子を回収する工程
を含むか又はそれらの工程からなる、方法にも関する。
ベロ細胞は熱失活させたウシ胎児血清5%(FCS、Invitrogen社 Frederick、MD)で補ったDMEM Glutamax(Gibco-BRL社)中で維持した。ウイルスレスキューに使用した安定なHEK293-T7-MVヘルパー細胞(WO2004/000876)は10%FCSで補ったDEME中で成長させた。
プラスミドpTM-MVSchwについては、シュワルツMVワクチン株の抗ゲノムに相応する感染性MV cDNAを含有するが、先に記載した[Combredetら、J Virol.、2003, 77(21): 11546〜11554頁]。DENV1からの非構造タンパク質のポリエピトープ構築物をコードする合成cDNAを、先に記載の4価EDIII抗原と組み合わせて又は組み合わせずにBsiWI/BssHII-消化pTM-MVSchwベクターに様々な位置で挿入した[Brandlerら、PLoS 2007、1(3)、e96; Brandlerら、Vaccine, 2010、28、6730〜6739頁](図7)。組換えウイルスのレスキューは、先に記載のように[Combredetら、J Virol.、2003、77(21): 11546〜11554頁]安定なHEK293-T7-MVヘルパー細胞を使用して行った(WO2004/000876)。組換えウイルスはベロ細胞上で成長させ、ウイルス力価はベロ細胞上で終点希釈アッセイにより決定した。
MVベクター又はpcDNA3発現プラスミドによるDENV抗原の発現を明示するため、MV-DENVで感染させたベロ細胞からの溶解物又はpcDNA3-NSプラスミドでトランスフェクトしたHEK293細胞からの溶解物をSDS-PAGEで分画し、セルロース膜(Amersham Pharmacia Biotech社)に転写し、抗EDIII DENV1 mAb 4E11又は抗NS3抗体でプローブした。ヤギ抗マウスIgG-西洋ワサビペルオキシダーゼ(HRP)コンジュゲート(Amersham社)を二次抗体として使用した。ペルオキシダーゼ活性は、増強化学蛍光検出キット(Pierce社)で可視化した。この分析により、MVベクターがEDIII 4価抗原及びNSポリエピトープ抗原の双方を発現していることが示された。同様に、pcDNA3-NSプラスミドは、高レベルのNSポリエピトープ抗原を発現した(図8)。
IFNα/β受容体欠失及びヒトMV受容体(hCD46+/-IFNα/β-/-又はCD46-IFNAR)を発現するマウスを先に記載のように作出し[Combredetら、J Virol.、2003、77(21): 11546〜11554頁]、Institut Pasteur動物施設にて病原体フリーの条件下で収容した。実験はInstitut PasteurにてOffice of Laboratory Animal Careの指針に従って実施した。6週齢のCD46-IFNARマウス6匹の群にMV-DENVベクター105TCID50又は空のMVの単回投与を腹腔内(ip)経路で接種した。マウスは免疫化後7日目に安楽死させ、脾臓を採取した。抽出直後の脾臓細胞をDENVペプチド(2μg/ml)又はMV(MOI 1)で18時の間特異的に刺激した。細胞はまた、陽性対照としてコンカナバリンA(5μg/ml、Sigma社)及び陰性対照としてRPMI-IL-2(10U/ml)でも刺激した。刺激するとIFN-γを分泌するそれらの細胞の能力は、先に記載のELISPOTアッセイにより試験した[Guerboisら、Virology、2009、388(1): 191〜203頁]。
NSポリエピトープ配列全体をカバーする36の重複ペプチド(5個のアミノ酸が重複する9〜15個のアミノ酸)の系列を合成した(図9)。CD46-IFNARマウスにおいて発現したH2-Db T細胞受容体及びH2-Kb T細胞受容体に結合可能な5つの特異的ペプチドも、予測アルゴリズムを使用して同定された(図9)。これらのペプチドを、プールするか又は個々のペプチドとしてのいずれかでELISPOT実験で使用して、免疫CD46-IFNARマウスからの脾臓細胞におけるT細胞応答を再刺激した。
MV-DENVでの免疫化により誘発される細胞性免疫応答(CMI)は、単回免疫化を行って7日後に採取した脾臓細胞に関するIFN-γELISPOTアッセイを使用して評価した。DENV NSペプチド又はMVによるex vivo刺激の後、有意な数のDENV-特異的IFN-γ分泌細胞(400〜800のスポット形成細胞/106脾臓細胞)がMV-DENV免疫マウスに検出され(図10、図11、図12)、これは類似の刺激条件におけるMV特異的応答の1/3であった(1500のスポット形成細胞/106脾臓細胞)。MV-DENVで免疫したほとんどのマウス(5/6)はDENVに対して有意なCMI応答を有し、本実験モデルにおける単回MV-DENV免疫化により、1週間以内に抗DENV細胞性免疫をプライミング可能であることが実証された。
CMVプロモーター下でDENV1 NSポリエピトープ配列を発現するpcDNA3プラスミドを、HLA-A*02-01、HLA-A*24: 02、HLA-B*07: 02及びHLA-B*35: 01単鎖形質転移/H-2 Class Iヌルマウスにおいてエレクトロポレーションにより注入し、脾臓細胞のINF-γ応答を、個々のペプチドに対するELISPOTアッセイにより定量した(図13)。
A.HLAクラスIトランスジェニックマウスにおけるDENV-特異的細胞障害性T細胞の誘導
防御免疫応答におけるCD8 T細胞の役割を調査するために、4群のH-2クラスIヌル/HLAクラスIトランスジェニックマウス(HLA-A*02: 01、HLA-A*24: 02、HLA-B*07: 02及びHLA-B*35: 01)にDNA免疫法によりワクチン接種をした。7匹のトランスジェニック動物から構成されている各群において、5匹及び2匹のマウスが、それぞれ、DENV1-NSポリエピトープ構築物及び対照プラスミドを受けた。全ての動物を皮内注射(100μgのDENV1-NSポリエピトーププラスミド又は対照プラスミド)、続いてin vivoエレクトロポレーションにより免疫した。2回の免疫化を3週間隔で実施し、脾臓細胞を第2の注射後10日目にELISPOTアッセイによりin vitro及びin vivo細胞障害性活性並びにIFN-γ分泌について試験した。
HLAクラスIトランスジェニック動物の各群において、DENV1-NSポリエピトープ構築物及び対照プラスミドをそれぞれ受けた3匹及び1匹のマウスを、それらの抗原特異的細胞障害性T細胞応答について試験した。T細胞応答の定量化は、グランザイムB(GrB)ELISPOTアッセイを使用して得た。このアッセイにより単一細胞レベルで細胞障害性メディエーターグランザイムBの放出を測定するが、この放出は標的細胞を溶解するT細胞の能力と強く相関すること[Ewen CLら、2006、J Immunol Methods、308(1〜2): 156〜166頁; Kalia Vら、2010、Adv Exp Med Biol、684: 79〜95頁; Zanetti Mら2010、Adv Exp Med Biol、684: 108〜125頁]及びウイルス産生を阻害すること[Marcet-Palacios Mら、2011、PLoS Pathog、7(12): e1002447]が示された。このアッセイにおいて、同族ペプチド又は無関係ペプチドによるin vitro刺激に対してグランザイムBを放出する脾臓細胞の頻度を、IFN-γを分泌する細胞の頻度と並行して測定した。結果は、対照pcDNA3.1プラスミドで免疫した動物と比較して、DENV1-NSポリエピトープ構築物で免疫したマウスにおいて有意なINF-γ及びGrB応答を示した(図16A及び図16B)。INF-γ応答とGrB放出との間に相関関係もあり、INF-γ及びGrB双方について最も大きい応答は、HLA-A24マウスについてはペプチドP32に対して、HLA-A2マウスについてはP56に対して、HLA-B7マウスについてはP15に対して、及びHLA-B35マウスについてはP49、P50及びP51に対して得られた。
in vivo細胞障害性のアッセイを先に記載のように実施した[Clemente T.ら、2013、Methods、61(2): 105〜109頁]。要約すると、DENV1-NSポリエピトープ構築物又は対照プラスミドで免疫したマウスに、ペプチドを負荷させ且つ蛍光染料(Cell trace violet)で標識した同系細胞で養子移入を行った。それぞれ、特異的ペプチド及び対照ペプチドでパルスされた、107の高蛍光染色細胞及び107の低蛍光染色細胞の混合物を、レシピエント動物に静脈内注射した。注射して18時間後に、レシピエントマウスの脾臓細胞をフローサイトメトリーで分析し、Cell trace高ドナー細胞(特異的ペプチド混合物でパルスした細胞)とCell trace低ドナー細胞(パルスされなかったか又は対照ペプチドでパルスされた)との間の比を決定した。同数の高染色細胞及び低染色細胞をレシピエントマウスに注射した場合、免疫マウスにおける特異的死滅活性は特異的ペプチドの混合物で負荷させた高染色細胞のより低い画分にもたらされた(図17)。特異的溶解のパーセンテージは式:
タイプI IFNが十分なマウスはDENV感染症に耐性があるので、DENVでの攻撃に対するワクチン誘導の防御免疫応答は、タイプI欠損マウスおいて、又はヒト造血幹細胞が生着した無リンパ球マウスにおいてのみ考慮することができる。HLA制限応答によりヒト骨髄コンパートメント及びリンパ球コンパートメントで再構成したヒト化マウスの新世代に関する近年の発展に即して[Legrand Nら、2011、Proc Natl Acad Sci U S A、108(32): 13224〜13229頁; Garcia Sら、2012、Immunol Lett、146(1〜2): 1〜7頁; Serra-Hassoun Mら、2014、J Immunol、193(3): 1504〜1511頁]、
本発明者らは、ヒトSIRPアルファに対してトランスジェニックしたRAG-/-γc-/-マウスを使用するが、このマウスでは、マウスMHCクラスI分子及びクラスII分子が、それぞれ、ヒトHLA-A2分子及びDR1分子で置き換えられた(CH1-2hSa)。
本発明者らは、DENV1の膜タンパク質のエクトドメイン(ectoM)に融合した、4種のDENV血清型のエンベロープドメインIII(EDIII)の融合体から構成される4価DENV抗原を発現し、並びに本発明のDENV1 NS抗原を同時に発現する組換えMVDVaxベクターの、CD46-IFNARマウスにおける防御有効性を評価した(図7に記載)。先に記載のように、CD46-IFNARマウスを作出し[Combredet、C.ら、2003、J Virol、77(21): 11546〜11554頁]、Institut Pasteur動物施設にて病原体フリーの条件下で収容した。実験はInstitut PasteurにてOffice of Laboratory Animal Careの指針に従って実施した。6週齢のCD46-IFNARマウス6匹の群をMV-DENVベクター105TCID50又は空のMVを腹腔内(ip)経路で接種した。同一用量の第2の投与をプライミングを行って1月後に実施した。抗MV抗体及び抗DENV抗体の存在を分析するため、血液を規則的に眼窩周囲経路により採取した。最後の免疫後2か月後に、全ての動物を、ベロ細胞上で成長させたマウス神経適応DENV1ハワイ株の105TCID50の腹腔内注射による実験的攻撃に供した[Despresら、J. Virol.、1998、72(1)、823〜829頁]。攻撃後、1、2、3、5、7日目に血液を眼窩周辺経路にて採取し、動物は続く15日間、臨床徴候について及び体重について追跡調査した。DENV1ゲノムウイルス負荷を、下に記載のワンステップqRT-PCRによりマウス血清において決定した。血清は56℃で30分間熱不活性化し、抗MV抗体の存在は、ELISA(Trinity Biotech社)を使用して検出した。HRPコンジュゲート抗マウス免疫グロブリン(Jackson Immuno Research社)を二次抗体として使用した。抗DENV抗体は先に記載のように[Brandlerら、Vaccine、2010、28、6730〜6739頁]、大腸菌(E.coli)中で産生したか又はin vitroで合成したDENV1、DENV2、DENV3、及びDENV4からの組換えEDIIIタンパク質で被覆した96-ウエルプレートを使用するELIZAにて検出した。HRPコンジュゲート抗マウス免疫グロブリンを二次抗体として使用した。プールされた血清の終点力価を、陰性対照として利用するMVを接種されたマウスからの血清の吸光度の2倍を与える最終希釈の逆数として算出した。
本発明者らは、非ヒト霊長類(NHP)において、組換えMVDVaxベクターの防御有効性を評価する。
ウイルス。攻撃ウイルス: DENV1ジャマイカ株CVI 1636 3Pを、1977年に単離、2/3 MP/3P C6/36HT 2Pベロ細胞の培養上清で継代培養する。DENV1株KDH0026Aを、C6/36細胞培養上清で継代培養する。DENV2 DJ.M.O.1.7.12を、C6/36細胞培養上清で継代培養する。DENV4ドミニカ814669株を、1981年に単離、4P C6/36 HT 2Pベロ細胞の培養上清で継代培養する。これらのウイルスを中和化試験に使用する。組換えMVDVaxワクチンウイルスを先に記載のように(Brandlerら、Vaccine、2010、28、6730〜6739頁)調製し、ウイルス力価をベロ細胞上で終点限界希釈アッセイにて決定する。
Claims (21)
- 融合ポリペプチドに組み立てられた以下の(a)、(b)及び(c)の断片:
(a)2つの領域を含むか又はそれらからなるデングウイルス(DENV)血清型1(DENV1)の非構造(NS)NS3タンパク質の2つの断片であって、第1の領域が配列番号6に規定のアミノ酸配列を有し、第2の領域が配列番号9に規定のアミノ酸配列を有する、2つの断片、
(b)配列番号12に規定のアミノ酸配列を有する、DENV1のNS4bタンパク質の断片、
(c)配列番号15に規定のアミノ酸配列を有する、DENV1のNS5タンパク質の断片
を含むか又はそれらからなり、(a)、(b)及び(c)の断片がこの順番で直接的又は間接的に融合している、600未満のアミノ酸残基を有するキメラポリエピトープ:
又はそのポリエピトープ変異体であって、前記ポリエピトープ変異体が、融合断片(a)、(b)及び(c)からなる融合ポリペプチドの配列とその全長にわたり90%超の同一性を有するアミノ酸配列を有するキメラポリエピトープからなり、前記融合ポリペプチドからアミノ酸残基の変異により該キメラポリエピトープが誘導される、キメラポリエピトープ。 - 配列番号3、配列番号146、配列番号147及び配列番号148からなる群から選択されるアミノ酸配列を有する、請求項1に記載のキメラポリエピトープ。
- DENV1、DENV2、DENV3及びDENV4に対するヒト白血球抗原(HLA)制限CD8+及び/又はCD4+T細胞応答を誘発する、請求項1又は2に記載のキメラポリエピトープ。
- 請求項1から3のいずれか一項に記載のキメラポリエピトープをコードする単離又は精製されたポリヌクレオチド。
- 配列番号1又は配列番号2からなる群から選択されるヌクレオチド配列を有する、請求項4に記載のポリヌクレオチド。
- 請求項4又は5に記載のポリヌクレオチドを含むプラスミドであるベクター。

- T7 RNAポリメラーゼ並びにMVのN、P及びLタンパク質を発現し、請求項6に記載のベクターをコードする核酸又は請求項7から9のいずれか一項に記載の組換えMVゲノムベクターをコードする核酸を更に発現するヘルパー細胞株からレスキューされる、組換え麻疹ウイルス(MV)粒子。
- (a)DENV1のectoMに融合した、4種のDENV血清型のEDIIIポリペプチドの融合体から構成されるDENV抗原、及び(b)請求項1から3のいずれか一項に記載のキメラポリエピトープを含み、任意選択でアクセサリーアジュバントを含まない、免疫原性組成物。
- 請求項12に記載の組換えMV粒子を含み、任意選択でアクセサリーアジュバントを含まない、免疫原性組成物。
- 皮下(s.c.)注射、皮内(i.d.)注射、筋肉内(i.m.)注射、腹腔内(i.p.)注射又は静脈内(i.v.)注射などの非経口経路による投与のために製剤化されている、請求項13又は14に記載の免疫原性組成物。
- 単回又は多回投与で投与される、請求項13から15のいずれか一項に記載の免疫原性組成物。
- ヒト対象においてデングウイルス感染症を予防及び/又は処置するための、請求項13から16のいずれか一項に記載の免疫原性組成物であって、薬学的に許容される媒体及び/又はアジュバントと混合されている、免疫原性組成物。
- 抗デングウイルスワクチンを調製するための組換えMV粒子を産生するための方法であって、
