本実施の形態に係る試料計測方法は、はじめに、マルチウエルプレート10の各ウエル12内で、接着依存性の細胞Cを培養する工程(ステップS10)を含む。具体的には、図5(A)に示すように、マルチウエルプレート10の各ウエル12に、細胞培養用に調整された培地Mを注入するとともに細胞を播種し、CO2インキュベーター内に静置する。これにより、図5(B)に示すように、マルチウエルプレート10のウエル12内(底面及び内側面)で、細胞Cを接着培養することができる。

以下、本実施の形態が奏する作用効果について説明する。
本実施の形態では、試料Sの状態を計測するための蛍光体30が、蓋20の凸部22の先端に設けられる。このことから、マルチウエルプレート10のウエル12に蛍光体を設ける必要がなくなるため、ウエル12の内面の組成を均一にすることができ、接着依存性の細胞などを培養することが容易になるとともに、マルチウエルプレート10の底面からの観察(顕微鏡観察など)が容易になる。また、本実施の形態では、蛍光体30への励起光の照射と、蛍光の受光を、マルチウエルプレート10の下側から(マルチウエルプレート10の底面を介して)行う。これによると、簡易な構造の蓋20を利用して、試料Sの状態を正確に計測することが可能になる。すなわち、この蓋20を利用して蓋20側から励起光の照射と蛍光の受光を行う場合、凸部22内での光の反射や、近接する凸部22を透過する光のクロストークが発生するおそれがあり、試料Sの状態を正確に計測することが難しくなる懸念がある。また、蓋20(凸部22)に光学素子や光ファイバーを設けるなどの工夫をすれば、試料Sの状態を正確に計測することが可能になるが、蓋20の構造を簡素化することが難しくなる。これに対して蛍光体30への励起光の照射と蛍光の受光をマルチウエルプレート10の下側から行うことで、簡易な構造の蓋20を利用して、試料Sの状態を正確に計測することが可能になる。
また、本実施の形態では、蓋20に、各ウエル12に対応した蛍光体30(凸部22)が備えられており、蓋20を取り付けるだけの作業で、すべてのウエル12内の試料Sの状態を計測することが可能になる。そのため、試料Sの状態を計測する工程を、効率よく実施することが可能になる。
なお、本実施の形態では、蛍光及び励起光は細胞Cを透過することになる。そのため、ウエル内で細胞が過密になる細胞などでは、計測に影響が出ることも懸念される。この場合には、細胞が過密にならない範囲で培養工程を終了させて計測工程を開始することで、試料Sの状態を精度よく計測することが可能になる。また、励起光及び蛍光に基づいて試料の状態を計測する方法には、蛍光の強度を利用する方法と、蛍光の位相差や消失時間に基づいて計測する方法とが存在する。本実施の形態では、蛍光の位相差や消失時間に基づいて試料の状態を計測する方法を適用することで、蛍光の強度変化の影響を受けにくくすることができ、細胞Cの培養が進んだ状態でも、試料Sの状態を正確に計測することが可能になる。(4) Operational Effects Hereinafter, the operational effects exhibited by the present embodiment will be described.

なお、個別蓋70は、ウエル12の底部が気密になるように構成することも可能である。例えば個別蓋70を、ウエル12の内形と同じ形とすることで、ウエル12の個別蓋70よりも下側の領域を気密に維持することができる。これにより、気相の影響をより小さくすることが可能になる。

本変形例では、培養と計測を同時に行う。言い換えると、本変形例では、試料Sの状態を計測しながら、ウエル12内で、細胞や微生物などの培養を行う。そして、本変形例に係る試料計測方法は、培養によって経時的に変化する試料Sの状態を計測する。
本変形例に係る試料計測方法は、ウエル12に、試料Sを注入する工程(ステップS50)を含む。試料Sは、培養対象物(例えば微生物、動物細胞、植物細胞など)と、培地との混合液である。なお、計測の目的によっては、培養対象物に対する試薬を混合することも可能である。
本変形例に係る試料計測方法は、マルチウエルプレート10に蓋20をした状態で、試料Sを経時変化させる工程を含む(ステップS60)。なお、本工程は、培養対象物に適した条件下で行われる。例えば本工程は、温調機能のついたCO2インキュベーター内に所定期間静置することによって実現することができる。
本実施の形態に係る試料計測方法は、試料Sの状態を計測する工程(ステップS70)を含む。本工程では、マルチウエルプレート10の下側から蛍光体30に励起光を照射するとともに、マルチウエルプレート10の下側から蛍光体30の蛍光を受光し、所定の演算を行うことによって、試料Sの状態を計測する。なお、本工程は、所定期間ごとに行ってもよく、連続的に行ってもよい。例えば、予め定められたタイミングでマルチウエルプレート10を試料計測装置2の計測台50に配置し、試料Sの状態を間欠的に計測することができる。あるいは、マルチウエルプレート10を計測台50に配置して経時変化させ、経時変化の様子を連続的に計測することも可能である。
なお、本工程では、試料Sの状態を計測する工程は、試料Sに含まれる特定成分の濃度を計測することによって実現することができる。具体的には、試料Sに含まれる溶存酸素濃度の情報として計測することも可能であるが、pHや溶存二酸化炭素濃度として計測することも可能である。あるいは、試料Sの状態を、濁度情報として計測することも可能である。
この試料計測方法によると、ウエル12ごとに、対象物の培養の成否を確認することができるため、培地や試薬の対象物への影響の有無を確認することが可能になる。
なお、本変形例では、試料Sの状態を計測する工程(ステップS70)で、蛍光の消失時間に基づいて試料の状態を計測する方法を適用してもよいが、蛍光の強度に基づいて試料の状態を計測する方法を適用することも可能である。例えば対象物の培養が進んでウエル12内で増殖が進めば、計測される蛍光の強度は小さくなる。そのため、蛍光の強度を利用することで、培養の進度を確認することが可能になる。FIG. 9 is a diagram for explaining a third modification of the embodiment to which the present invention is applied.
