Description
PUFAは多価不飽和脂肪酸を指す。
LC−PUFA(長鎖多価不飽和脂肪酸)は少なくとも20個の炭素原子を有するPUFAを指す。
HUFA(高度不飽和脂肪酸)は、少なくとも3つの二重結合を有するPUFAを指す。
LC−HUFA(長鎖高度不飽和脂肪酸)は、少なくとも20個の炭素原子と少なくとも3つの二重結合を有する多価不飽和脂肪酸を指す。
LC−PUFAとしては、例えば、
ジホモ−γ−リノレン酸(DGLA、20:3 ω−6)
アラキドン酸(ARA、20:4 ω−6)
エイコサペンタエン酸(EPA、20:5 ω−3)
ドコサペンタエン酸(DPA、22:5 ω−3、またはDPA22:5、ω−6)、
ドコサヘキサエン酸(DHA:22:6 ω−3)
が挙げられる。
[実施例1]
8日間の発酵後に得られたモルティエラ・アルピナ(Mortierella alpina)の発酵ブロスを70℃で1時間低温殺菌した。低温殺菌ブロスを濾過して、水分含量50重量%の濾過ケークを得た。濾過ケークを崩し、15℃未満の温度で押し出した。押し出し物(直径3mm)を3つのゾーンがある連続流動床乾燥機内で、水分含量7%に乾燥させた。第1のゾーンでは床温度は32℃であり、空気温度は50℃であった(Tdew point=15℃)。
第1のゾーン:床温度32℃、空気温度50℃(Tdew point=15℃):45分間
第2のゾーン:床温度32℃、空気温度35℃(Tdew point=1℃):45分間
第3のゾーン:床温度15℃、空気温度15℃(Tdew point=1℃):30分間
乾燥バイオマスの油含量は39%であった。ARA含量は、油中の総脂肪酸を基準にして46%であった。
発酵および低温殺菌を繰り返す。連続脱水機を使用して湿潤細胞を回収し、破壊して、次に振動流動床を用いて熱風乾燥(熱風温度120℃)により乾燥を実施し、水分含量1重量%に乾燥させる。室温の空気を流動床に供給して、乾燥細胞を冷却する。ARAおよび油含量は、実施例1と同様である。
国際公開第2008009600号パンフレットに記載される手順を使用して、形質転換された遺伝子導入アブラナ属(Brassica)植物から、19%アラキドン酸(総脂肪酸基準)を含有する種子を得る。
Claims (34)
- (i)少なくとも20個の炭素原子を有する多価不飽和脂肪酸(LC−PUFA)および(ii)細胞を含んでなる組成物であって、
1〜20重量%の水分含量を有し、
少なくとも30重量%の油含量を有し、
油中の総脂肪酸を基準にして、少なくとも40重量%の少なくとも3つの二重結合があるPUFAを含んでなる油を含んでなり、
前記細胞が微生物細胞であり、
40℃で>24時間の熱誘導時間(T.I.T.)を有する、組成物。 - 2〜15重量%の水分含量を有する、請求項1に記載の組成物。
- 3〜12重量%の水分含量を有する、請求項1に記載の組成物。
- 3.5〜10重量%の水分含量を有する、請求項1に記載の組成物。
- 4〜9重量%の水分含量を有する、請求項1に記載の組成物。
- 70重量%未満の油含量を有する、請求項1〜5の何れか一項に記載の組成物。
- 少なくとも40重量%の油含量を有する、請求項6に記載の組成物。
- 60重量%未満の油含量を有する、請求項6に記載の組成物。
- 油中の総脂肪酸を基準にして、80重量%未満の少なくとも3つの二重結合があるPUFAを含んでなる油を含んでなる、請求項1〜8の何れか一項に記載の組成物。
- 油中の総脂肪酸を基準にして、70重量%未満の少なくとも3つの二重結合があるPUFAを含んでなる油を含んでなる、請求項9に記載の組成物。
- 油中の総脂肪酸を基準にして、60重量%未満の少なくとも3つの二重結合があるPUFAを含んでなる油を含んでなる、請求項9に記載の組成物。
- 前記LC−PUFAが少なくとも3つの二重結合を有する、請求項1〜11の何れか一項に記載の組成物。
- 前記LC−PUFAがω−3またはω−6 PUFAである、請求項1〜12の何れか一項に記載の組成物。
- 前記LC−PUFAが、ジホモ−y−リノレン酸(DGLA、20:3 ω−6)、アラキドン酸(ARA、20:4 ω−6)、エイコサペンタエン酸(EPA、20:5 ω−3)、ドコサヘキサエン酸(DHA:22:6 ω−3)、ドコサペンタエン酸(DPA 22:5 ω−3、またはDPA 22:5、ω−6)から選択される、請求項1〜13の何れか一項に記載の組成物。
- 前記LC−PUFAがARAまたはDHAである、請求項1〜14の何れか一項に記載の組成物。
- 前記微生物細胞が、酵母細胞、細菌細胞、真菌細胞、または藻類細胞である、請求項1〜15の何れか一項に記載の組成物。
- 前記組成物が、モルティエラ(Mortierella)属の微生物を含んでなり、前記LC−PUFAがARAである、請求項1〜16の何れか一項に記載の組成物。
- 前記組成物が、モルティエラ・アルピナ(Mortierella alpina)種の微生物を含んでなる、請求項17に記載の組成物。
- 前記組成物が、ヤブレツボカビ(Thraustochytriales)目の微生物を含んでなり、前記LC−PUFAがDHAまたはEPAである、請求項1〜16の何れか一項に記載の組成物。
- 前記組成物が、スラウストキトリウム(Thraustochytrium)属またはシゾキトリウム(Schizochytrium)属の微生物を含んでなる、請求項19に記載の組成物。
- 前記組成物が、クリプテコジニウム(Crypthecodinium)属の微生物を含んでなり、前記LC−PUFAがDHAである、請求項1〜16の何れか一項に記載の組成物。
- 前記組成物が、クリプテコジニウム・コーニイ(Crypthecodinium cohnii)種の微生物を含んでなる、請求項21に記載の組成物。
- 請求項1〜22の何れか一項に記載の組成物を製造する方法であって、
細胞およびLC−PUFAを含んでなる組成物を、40℃未満の温度で乾燥させるステップを含んでなる、方法。 - 乾燥温度が35℃未満である、請求項23に記載の方法。
- 乾燥温度が33℃未満である、請求項23に記載の方法。
- 乾燥温度が30℃未満である、請求項23に記載の方法。
- 乾燥温度が25℃未満である、請求項23に記載の方法。
- 前記組成物を≦15℃の露点を有する調整空気に接触させるステップを含んでなる、請求項23〜27の何れか一項に記載の方法。
- 前記露点が<10℃である、請求項28に記載の方法。
- 前記露点が<5℃である、請求項28に記載の方法。
- 前記組成物を、流動床乾燥機を使用して乾燥させるステップを含んでなる、請求項23〜30の何れか一項に記載の方法。
- 請求項1〜22の何れか一項に記載の組成物から、あるいは請求項23〜31の何れか一項に記載の方法によって得られるまたは得られ得る組成物から、前記LC−PUFAまたはLC−PUFAを含んでなる油を単離するステップを含んでなる、LC−PUFAまたはLC−PUFAを含んでなる油を得る方法。
- 請求項32に記載の方法に従ってLC−PUFAまたはLC−PUFAを含んでなる油を得るステップと、前記LC−PUFAまたは前記LC−PUFAを含んでなる油を食品に組み込むステップを含んでなる、食品を得る方法。
- 請求項32に記載の方法に従ってLC−PUFAまたはLC−PUFAを含んでなる油を得るステップと、前記LC−PUFAまたは前記LC−PUFAを含んでなる油を乳児用調製粉乳に組み込むステップを含んでなる、乳児用調製粉乳を得る方法。
